We have prepared a number of isomeric red seaweed galactan-derivative sulfated oligosaccharides to determine whether there were diagnostic differences among the isomeric mass spectra obtained using ESI CID MS/MS (triple quadrupole instrument). Fragmentation of the single or multicharged molecular ions from di-, tetra-, and hexasaccharides indicated that the relative positioning of the sulfate groups and type of monosaccharide unit affect the rate of cleavage of the glycosidic bonds. We also performed a comparative [M-Na] fragmentation study of positional isomers of sulfated disaccharides that present all four monosulfation possibilities on the galactopyranosidic ring. In this case, negative-ion ESI CID MS/MS approach gave diagnostic product ions from cross-ring cleavages along with the same main B1 ion (from sulfated Galp), at m/z 241, for all isomers. The isomeric disaccharides were also submitted to increased spray energy conditions inducing in-source fragmentation; preformed B1 ions were then fragmented to give similar product ions as those found in [M-Na] analysis. Evaluation of the relative abundances mainly for cross-ring fragment ions at m/z 138, 139, 151, 153 allowed clear distinction among the members of the disaccharide series. The different ratios for m/z 151/153 ions were consistent with the predominance of m/z 153 being related to the cases when the bond involved in the cleavage process links a sulfated carbon. A quadrupole ion trap instrument (MSn analysis) was also utilized to compare the results obtained with the triple quadrupole instrument.  相似文献

A flowing afterglow-tandem mass spectrometer (FA-TMS) was used to study a series of C5 unsaturated alcohols and isoprene. The analytical procedure was validated through collision induced dissociation (CID) experiments on proton hydrates. In the FA, reagent H3O+ ions were used to chemically ionize the alcohols under study and isoprene. Chemical ionization (CI) by H3O+ is widely used, especially in PTR-MS instruments, and produces a main peak at m/z 69 for all studied compounds, implying the impossibility to distinguish them by a simple quadrupole mass filter. The CID of these ions at m/z 69 resulted in daughter ions with the same masses but with different intensities depending on the organic compound, the collision energy and the Ar target gas pressure in the collision cell. From these observations, pentenols were easily distinguished from methylbutenols and 3-methyl-3-buten-1-ol from the other compounds. CID experiments were also performed on the protonated alcohol, which is only a stable ion for 1-penten-3-ol, 2-methyl-3-buten-2-ol and 3-methyl-3-buten-1-ol, showing different CID patterns as a function of the collision energy. The coupling between a FA reactor and a TMS has proven to be a valuable approach to identify C5 unsaturated alcohols and isoprene.  相似文献

The ion-molecule reactions of dimethyl ether ions CH3OCH3 + and (CH3OCH3)H+, and four- to seven-membered ring lactams with methyl substituents in various positions were characterized by using a quadrupole ion trap mass spectrometer and a triple-quadrupole mass spectrometer. In both instruments, the lactams were protonated by dimethyl ether ions and formed various combinations of [M + 13] +, [M + 15] +, and [M + 45] + adduct ions, as well as unusual [M + 3] + and [M + 16] + adduct ions. An additional [M + 47] + adduct ion was formed in the conventional chemical ionization source of the triple-quadrupole mass spectrometer. The product ions were isolated and collisionally activated in the quadrupole ion trap to understand formation pathways, structures, and characteristic dissociation pathways. Sequential activation experiments were performed to elucidate fragment ion structures and stepwise dissociation sequences. Protonated lactams dissociate by loss of water, ammonia, or methylamine; ammonia and carbon monoxide; and water and ammonia or methylamine. The [M + 16] + products, which are identified as protonated lactone structures, are only formed by those lactams that do not have an N-methyl substituent. The ion-molecule reactions of dimethyl ether ions with lactams were compared with those of analogous amides and lactones.  相似文献

The sodium adduct of disodium salts of benzene dicarboxylic acids (m/z 233), when subjected to collision‐induced dissociation (CID), undergoes a facile loss of CO2 to produce an ion of m/z 189, which retains all the three sodium atoms of the precursor. The CID spectrum of this unusual m/z 189 ion shows significant peaks at m/z 167, 63 and 85. The enigmatic m/z 167 ion, which appeared to represent a loss of a 22‐Da neutral fragment from the precursor ion is in fact a fragment produced by the interaction of the m/z 189 ion with traces of water present in the collision gas. The change of the m/z 167 peak to 168, when D2O vapor was introduced to the collision gas of a Q‐ToF instrument, proved that such an intervention of water could occur even in collision cells of tandem‐in‐space mass spectrometers. The m/z 189 ion has such high affinity for water; it forms an ion/molecule complex even during the brief residence time of ions in collision cells of triple quadrupole instruments. The complex formed in this way then eliminates elements of NaOH to produce the ion observed at m/z 167. In an ion trap, the relative intensity of the m/z 167 peak increases with longer activation time even at the lowest possible collision energy setting. Similarly, the m/z 145 ion (which represents the sodium adduct of phenelenedisodium, formed by two consecutive losses of CO2 from the m/z 233 ion of meta‐ and para‐isomers) interacts with water to produce a fragment ion at m/z 123 for the sodium adduct of phenylsodium. Other uncommon ions that originate also from water/ion interactions are observed at m/z 85 and 63 for [Na3O]+ and [Na2OH]+, respectively. Tandem mass spectrometric experiments conducted with appropriately deuterium‐labeled compounds confirmed that the proton required for the formation of the [Na2OH]+ ion originates from traces of water present in the collision gas and not from the ring protons of the aromatic moiety. Collision-induced dissociation (CID) of deprotonated hexose-containing disaccharides (m/z 341) with 1–2, 1–4, and 1–6 linkages yields product ions at m/z 221, which have been identified as glycosyl-glycolaldehyde anions. From disaccharides with these linkages, CID of m/z 221 ions produces distinct fragmentation patterns that enable the stereochemistries and anomeric configurations of the non-reducing sugar units to be determined. However, only trace quantities of m/z 221 ions can be generated for 1–3 linkages in Paul or linear ion traps, preventing further CID analysis. Here we demonstrate that high intensities of m/z 221 ions can be built up in the linear ion trap (Q3) from beam-type CID of a series of 1–3 linked disaccharides conducted on a triple quadrupole/linear ion trap mass spectrometer. 18O-labeling at the carbonyl position of the reducing sugar allowed mass-discrimination of the “sidedness” of dissociation events to either side of the glycosidic linkage. Under relatively low energy beam-type CID and ion trap CID, an m/z 223 product ion containing 18O predominated. It was a structural isomer that fragmented quite differently than the glycosyl-glycolaldehydes and did not provide structural information about the non-reducing sugar. Under higher collision energy beam-type CID conditions, the formation of m/z 221 ions, which have the glycosyl-glycolaldehyde structures, were favored. Characteristic fragmentation patterns were observed for each m/z 221 ion from higher energy beam-type CID of 1–3 linked disaccharides and the stereochemistry of the non-reducing sugar, together with the anomeric configuration, were successfully identified both with and without 18O-labeling of the reducing sugar carbonyl group.  相似文献

The importance of the mass spectral product ion structure is highlighted in quantitative assays, which typically use multiple reaction monitoring (MRM), and in the discovery of novel metabolites. Estradiol is an important sex steroid whose quantitation and metabolite identification using tandem mass spectrometry has been widely employed in numerous clinical studies. Negative electrospray ionization tandem mass spectrometry of estradiol (E2) results in several product ions, including the abundant m/z 183 and 169. Although m/z 183 is one of the most abundant product ions used in many quantitative assays, the structure of m/z 183 has not been rigorously examined. We suggest a structure for m/z 183 and a mechanism of formation consistent with collision induced dissociation (CID) of E2 and several stable isotopes ([D4]-E2, [13C6]-E2, and [D1]-E2). An additional product ion from E2, namely m/z 169, has also been examined. MS3 experiments indicated that both m/z 183 and m/z 169 originate from only E2 [M – H] m/z 271. These ions, m/z 183 and m/z 169, were also present in the collision induced decomposition mass spectra of other prominent estrogens, estrone (E1) and estriol (E3), indicating that these two product ions could be used to elucidate the estrogenic origin of novel metabolites. We propose two fragmentation schemes to explain the CID data and suggest a structure of m/z 183 and m/z 169 consistent with several isotopic variants and high resolution mass spectrometric measurements.   相似文献

Glycoconjugates, such as chromophore-labeled disaccharides and permethylated glycosphingolipids (GSL) were used for comparison of triple quadrupole and double focusing mass spectrometers in analysis of product ions. A profound effect of collision energy was observed in the product ion spectra of ceramide ions (fragment ions of permethylated GSL): more product ions were observed from a double focusing mass spectrometer. Besides collision energy, the structure of the analyte had a significant effect on the formation of product ions. Despite the fact that masses of protonated molecular ions (MH+) of permethylated GSL are significantly larger than their ceramide fragments, the low-energy and high-energy product ion spectra of MH+ are, in general, similar. In a double focusing mass spectrometer of reversed geometry, more metastable ions were observed in the first field free region (FFR) than in the second FFR. The metastable ions observed in the second FFR were similar to those observed in low-energy collision-induced dissociation (CID). Although a double focusing mass spectrometer is superior to triple quadrupole instrument for detection of product ions, the poor resolution in either the selection of precursor ion or in the product ion spectra can be a serious problem in analysis of a mixture with similar masses.  相似文献

Low energy collision induced dissociation (CID) spectra were measured by a triple stage quadrupole mass spectrometer for the [MH]+ ions of diethyl and dimethyl esters of maleic, fumaric, citraconic and mesaconic acids. A very high degree of stereospecificity was observed for the geometrically isomeric diethyl esters. The cis esters give rise to very abundant [MH? EtOH]+ and [MH? EtOH? C2H4]+ ions, while the trans isomers exhibit very abundant [MH? C2H4]+ and [MH? 2 C2H4]+ ions. The highly stereospecific processes indicate that the double bond configuration is retained in the protonated species under the conditions of the experiment.  相似文献

The dissociation rates and energetics of the loss of halogen atoms from energy-selected halotoluene ions were investigated by photoelectron photoion coincidence (PEPICO) and collisional activation (CA) mass spectrometric experiments. Dissociation onsets, determined from the dissociation rates measured as a function of the internal energy of the parent ion, revealed the formation of three [C7H7]+ isomers, which were identified, on the basis of the CA data, as the tolyl, benzyl and tropylium ions. All of the ions investigated produced a mixture of isomeric ions. Only iodotoluene ions produced any tolyl product ions by a direct bond cleavage. The bromo- and chlorotoiuene ions produced mixtures of benzyl and tropyl ions. The observed two-component decay rates of the iodotoluene ions revealed the participation of a lower energy [C7H7I]+ ˙ isomer in the dissociation process. The identity of this isomer is not known but it probably does not have the cycloheptatriene ion structure because considerable kinetic energy was released in this dissociation.  相似文献

We describe a diagnostic ion that enables rapid semiquantitative evaluation of the degree of oxygen contamination in the collision gases used in tandem mass spectrometers. Upon collision-induced dissociation (CID), the m/z 359 positive ion generated from the analgesic etoricoxib undergoes a facile loss of a methyl sulfone radical [?SO2(CH3); 79-Da] to produce a distonic radical cation of m/z 280. The product-ion spectrum of this m/z 280 ion, recorded under low-energy activation on tandem-in-space QqQ or QqTof mass spectrometers using nitrogen from a generator as the collision gas, or tandem-in-time ion-trap (LCQ, LTQ) mass spectrometers using purified helium as the buffer gas, showed two unexpected peaks at m/z 312 and 295. This enigmatic m/z 312 ion, which bears a mass-to-charge ratio higher than that of the precursor ion, represented an addition of molecular oxygen (O2) to the precursor ion. The exceptional affinity of the m/z 280 radical cation towards oxygen was deployed to develop a method to determine the oxygen content in collision gases. Figure
