The present study was structured to investigate the anti-inflammatory potential of the extracts, fractions and compounds isolated from Carissa carandas (L.) roots. Bioassay guided fractionation of methanol extract based on inhibitory potential towards proinflammatory mediators (TNF-α, IL-1β and nitric oxide (NO)) led to the identification of stigmasterol (1), lupeol (2), oleanolic acid (3), carissone (4) and scopoletin (5) as potential anti-inflammatory agents. Carissone (4) (IC50 = 20.1 ± 2.69 μg/mL) and scopoletin (5) (IC50 = 24.6 ± 1.36 μg/mL) exhibited significant inhibition of NO production comparable to specific NO inhibitor (L-NAME; IC50 = 19.82 ± 1.64 μg/mL) without affecting the cell viability. Also, 4 and 5 at a concentration of 30 μM were found to inhibit 41.88–53.44% of TNF-α and IL-1β. To the best of our knowledge, this is the first report displaying the anti-inflammatory effects of C. carandas (L.) roots, partially mediated by inhibition of TNF-α, IL-1β and NO.  相似文献

In this study, the anti-tumour-promoting and thermal-induced protein denaturation inhibitory activities of β-sitosterol (1) and lupeol (2), isolated from Diospyros lotus L., were explored. Compound 1 showed a marked concentration-dependent inhibition against 12-O-tetradecanoylphorbol-13-acetate (20 ng/32 pmol)-induced Epstein–Barr virus early antigen activation in Raji cells with IC50 of 270 μg/ml, without significant toxicity (70% viability). Compound 2 showed significant anti-tumour-promoting effect with IC50 of 412 μg/ml, without significant toxicity (60% viability). In heat-induced protein denaturation assay, compound 1 exhibited a concentration-dependent attenuation with a maximum effect of 73.5% at 500 μg/ml with EC50 of 117 μg/ml, while compound 2 exhibited a maximum effect of 59.2% at 500 μg/ml with EC50 of 355 μg/ml. Moreover, in silico docking studies against the phosphoinositide 3-kinase enzyme also show the inhibitory potency of these compounds. In short, both the compounds exhibited a marked anti-tumour-promoting and potent inhibitory effect on thermal-induced protein denaturation.  相似文献

The phytochemical investigation of chloroform extract from Piper betle var. haldia, Piperaceae, leaves has resulted in the isolation of two new chemical constituents which were identified as 1-n-dodecanyloxy resorcinol (H1) and desmethylenesqualenyl deoxy-cepharadione-A (H4), on the basis of spectroscopic data 1D NMR (1H and 13C) and 2D NMR (1H-1H COSY and HMBC) as well as ESI-MS, FT-IR and HR-ESI-MS analyses. Compounds H1 and H4 showed excellent antioxidant DPPH free radical scavenging activity with IC50 values of 7.14 μg/mL and 8.08 μg/mL compared to ascorbic acid as a standard antioxidant drug with IC50 value of 2.52 μg/mL, respectively. Evaluation of cytotoxic activity against human hepatoma cell line (PLC-PRF-5) showed moderate effect with the GI50 values of 35.12 μg/mL for H1, 31.01 μg/mL for H4, compared to Doxorubicin® as a standard cytotoxic drug with GI50 value of 18.80 μg/mL.  相似文献

Ethanol extract of the aerial parts of Bergenia ligulata was subjected to solvent–solvent separation followed by various chromatographic techniques that lead to isolation of bergenine (1), p-hydroxybenzoyl bergenin (2), 11-O-galloylbergenin (3) and methyl gallate (4) as major constituents. Ethyl acetate fraction showed a dose-dependent urease inhibitory pattern with IC50 value of 54μg/mL. Structures of compounds 1 and 3 were established by XRD and 2, 4 by NMR. All these compounds were subjected to DPPH scavenging activity, reducing power assay and urease inhibitory activity. The EC50 7.45 ± 0.2 μg/mL and 5.39 ± 0.28 μg/mL values in terms of antioxidant and reducing power, respectively, were less for 3. Compounds 13 showed moderate to significant urease inhibitory potential with IC50 57.1 ± 0.7, IC50 48.4 ± 0.3 and 38.6 ± 1.5. Antioxidant activities and urease inhibitory potential were investigated and compound 3 was found to be the most active.  相似文献

The phytochemical study of ethyl acetate and n-butanol extracts of Pteranthus dichotomus Forssk. led to the isolation and identification of 11 compounds, including three glycolipids 13, one lignan 4, three flavonoids 57 and four phytosterols 811. Structures of the isolated compounds have been elucidated by analysis of 1D and 2D NMR data, and mass spectrometry EI-MS and ESI-MS and by comparison with literature data. Furthermore, the ethyl acetate and n-butanol extracts were examined for their antioxidant and antibacterial activities. The results showed that both extracts (PDAC and PDBU) had a moderate antioxidant activity (IC50 = 375.514 μg/mL and 691.333 μg/mL) respectively.  相似文献

A new methyl 2-(4-((2-hydroxy-3-methylbut-3-en-1-yl)oxy)phenyl) acetate 1, together with five known compounds 26, was isolated from the culture of the deep sea-derived fungus Aspergillus westerdijkiae SCSIO 05233. The new structure was determined by NMR (1H and 13C NMR, HSQC, HMBC and MS) and optical rotation analysis. Compound 5 displayed weak inhibitory activities towards K562 and promyelocytic HL-60 with IC50 values of 25.8 and 44.9 μM, and compound 6 showed strong antifouling activity with EC50 value 8.81 μg/mL.  相似文献

This work expands the phytochemical composition knowledge of Acanthus mollis and evaluates antioxidant and anti-inflammatory activities which could be related with its traditional uses. Extracts from leaves, obtained by sequential extraction, were screened using TLC and HPLC-PDA. The ethanol extract was the most active on DPPH assay (IC50 = 20.50 μg/mL) and inhibited nitric oxide (NO) production in RAW 264.7 macrophages (IC50 = 48.31 μg/mL). Significant amounts of cyclic hydroxamic and phenolic acids derivatives were detected. A lower antioxidant effect was verified for a fraction enriched with DIBOA derivatives (IC50 = 163.02 μg/mL), suggesting a higher contribution of phenolic compounds for this activity in ethanol extract. However, this fraction exhibited a higher inhibition of NO production (IC50 = 32.32 μg/mL), with absence of cytotoxicity. These results support the ethnomedical uses of this plant for diseases based on inflammatory processes. To our knowledge, it is the first report to the anti-inflammatory activity for DIBOA derivatives.  相似文献

Phytochemical investigation of the methanol extract of the aerial parts of Polygonum glabrum afforded one new natural product ( ? )-2-methoxy-2-butenolide-3-cinnamate (1) along with six known compounds, β-hydroxyfriedalanol (2), 3-hydroxy-5-methoxystilbene (3), ( ? ) pinocembrin (4), sitosterol-(6′-O-palmitoyl)-3-O-β-d-glucopyranoside (5), ( ? ) pinocembrin-5-methyl ether (6) and sitosterol-3-O-β-d-glucopyranoside (7). Compound 1 showed promising in vitro anti-HIV-1 activity against primary isolates HIV-1UG070 (X4, subtype D) and HIV-1VB59 (R5, subtype C) assayed using TZM-bl cell line with IC50 in the range of 15.68–22.43 μg/mL. The extract showed TI in the range of 19.19–27.37 with IC50 in the range of 10.90–15.55 μg/mL. Compounds 1, 3 and 4 exhibited in vitro anti-mycobacterium activity against Mycobacterium tuberculosis H37Ra with IC50 values of 1.43, 3.33 and 1.11 μg/mL in dormant phase and 2.27, 3.33 and 1.21 μg/mL in active phase, respectively. Compound 4 was found to be the most active antiproliferative with IC50 values of 1.88–11.00 μg/mL against THP-1, A549, Panc-1, HeLa and MCF7 cell lines.  相似文献

Osmanthus fragrans are well-known for their fragrance, but it is wasteful if to discard O. fragrans flower after extracting their essential oils. In this paper, we found that O. fragrans flower residues were rich in flavonoids. Six flavonoids and one phenylethanoid glycoside were isolated from the ethanol extract of O. fragrans flower residues, identified as quercetin (1), rutin (2), verbascoside (3), genistin (4), kaempferol (5), isorhamnetin (6) and naringin (7). In bioactivity study, kaempferol (IC50 = 1.43 μg/mL) showed the best anti-inflammatory activity. Isorhamnetin, quercetin, kaempferol, verbascoside and rutin (the values of IC50 were 18.30, 11.05, 16.88, 20.21 and 22.76 μg/mL, respectively) showed excellent DPPH free radical scavenging activity. Verbascoside performed relatively well at inhibiting the growth of both CT26 colonic carcinoma cells (IC50 = 46.87 μg/mL) and HepG2 hepatocarcinoma cells (IC50 = 30.58 μg/mL). In addition, quercetin and kaempferol showed strong anti-proliferation activity against HepG2 cells.  相似文献

The crude extracts of tubers of Aconitum spicatum (Bruhl) Stapf were investigated for in vitro antileishmanial activity against Leishmania major. The dichloromethane extract at pH 2.5 showed antileishmanial activity with IC50 value of 27.10 ± 0.0 μg/mL. Chromatographic purification of the dichloromethane extract led to isolation of three C-19 norditerpenoid alkaloids indaconitine (1), chasmaconitine (2) and ludaconitine (3). Compounds 3 and 2 showed antileishmanial activity with IC50 = 36.10 ± 3.4 and 56.30 ± 2.1 μg/mL, respectively. Compound 1 was less effective (IC50 > 100 μg/mL). The cytotoxicity of compounds 1, 2 and 3 studied against MCF7, HeLa and PC3 cancer cell lines and 3T3 normal fibroblast cell line did not show cytotoxicity at 30 μM.  相似文献

For the first time, eugenol, a natural bioactive allylphenol, was introduced into coordination with platinum(II) by replacement of ethylene from Zeise’s salt with eugenol (Eug). The obtained complex, K[PtCl3(Eug)] (1), was used as the key compound for preparation of the series of trans-[PtCl2(Eug)(Amine)] (2–11), [PtCl(Eug)(8-O-quinoline)] (12) and [PtCl(Eug)(2-O2C-quinoline)] (13). The synthesized complexes were characterized by elemental analyses, IR, 1H NMR, 13C NMR, HSQS, HMBC, NOESY, and MS spectra. In 113 eugenol coordinates with Pt(II) at ethylenic double bond of the allyl group, the donor N of the amines is in trans-position in comparison with the double bond. A display of the trans-effect on the chemical shift of 1H and 13C was remarked. Seven complexes were tested for cell in vitro cytotoxicity on human cancer cells. Complexes 3 and 12 exhibit high activities on Hep-G2 with IC50 = 3.12 and 5.29 μM; 12 gives high activity against KB, Lu and MCF-7 with IC50 = 0.43, 2.95 and 1.84 μM, respectively. Most of these IC50 are lower than those of cisplatin.  相似文献

A new glycoside, pinnatoside A (1), together with two known compounds (2 and 3), were isolated from the stems of Paullinia pinnata. Their structures were elucidated on the basis of extensive spectroscopic analysis and chemical methods. Compound 1 showed significant antibacterial activity with a minimum inhibitory concentration (MIC) value of 1.56 μg/mL against Escherichia coli, and 2 displayed significant antibacterial activity with a MIC value of 1.56 μg/mL against Enterobacter aerogenes and E. coli. Equally, compound 1 exhibited the best radical-scavenging activity (RSa50 = 25.07 ± 0.49 μg/mL).  相似文献

Naringin, as a component universal existing in the peel of some fruits or medicinal plants, was usually selected as the material to synthesise bioactive derivates since it was easy to gain with low cost. In present investigation, eight new acacetin-7-O-methyl ether Mannich base derivatives (18) were synthesised from naringin. The bioactivity evaluation revealed that most of them exhibited moderate or potent acetylcholinesterase (AChE) inhibitory activity. Among them, compound 7 (IC50 for AChE = 0.82 ± 0.08 μmol?L?1, IC50 for BuChE = 46.30 ± 3.26 μmol?L?1) showed a potent activity and high selectivity compared with the positive control Rivastigmine (IC50 for AChE = 10.54 ± 0.86 μmol?L?1, IC50 for BuChE = 0.26 ± 0.08 μmol?L?1). The kinetic study suggested that compound 7 bind to AChE with mix-type inhibitory profile. Molecular docking study revealed that compound 7 could combine both catalytic active site (CAS) and peripheral active site (PAS) of AChE with four points (Trp84, Trp279, Tyr70 and Phe330), while it could bind with BuChE via only His 20.  相似文献

As a part of ongoing investigation of Acokanthera oppositifolia (Lam.) Codd., four compounds were isolated from its seeds, a new compound; lup-20(29)-en-3β-O-(3′-β-hydroxy) palmitate (1), three known compounds; a triterpene; lupeol (2), a cardiac glycoside; acovenoside A (3) and a sterol; β-sitosterol (4). Their structures were investigated using 1D & 2D- 1H and 13CNMR spectroscopy. Antimicrobial potential of the compounds was evaluated against 10 microorganisms responsible for endocarditis. The minimum inhibitory concentration (MIC) of the compounds was determined using broth microdilution method. The new compound (1) evidenced significant antibacterial activity especially aganist Pseudomonas aeruginosa with (MIC 7.81 μg/ml). Lupeol (2) exhibited remarkable antimicrobial activity against Methicillin-resistant Staphylococcus aureus, Aspergillus fumigates and Candida albicans (MIC 3.9, 0.24 and 3.9 μg/ml, respectively). On the other hand, acovenoside A (3) inhibited the growth of Escherichia coli (MIC 0.98 μg/ml). We herein present the potential of A. oppositifolia as a cardioprotective agent against the microorganisms responsible for endocarditis.  相似文献

A new bibenzyl derivative (4), together with two glycosylated flavonoids (1 and 2), batatasin III (3) and the phenanthrene isohircinol (5) were isolated from the aerial parts of Gavilea lutea. Their structures were elucidated on the basis of spectroscopic studies including 1D and 2D NMR, UV, IR and HRESIMS. All isolated compounds were evaluated for their antifungal activity towards Candida albicans. The new compound 4 showed inhibitory activity with a MIQ of 50 μg. In addition, compound 4 exhibited a selective activity (IC50 = 2.3 μg/mL) against Leishmania donovani.  相似文献

Platinum(II) complexes, [Pt(Lx)X2] (16), where X = Br or I and Lx = 2,2′-bipyridine or 1,10-phenanthroline derivatives (5,5′-dimethyl-2,2′-bipyridine (5-Mebpy), 4,4′-dimethyl-2,2′-bipyridine (4-Mebpy), and 5-amino-1,10-phenanthroline (5-NH2phen)) were prepared. The complexes were characterized by the elemental analysis, mass spectrometry, infrared, and multinuclear (1H, 13C and 195Pt) 1-D and 2-D NMR spectroscopies, and by single-crystal X-ray analysis of [Pt(4-Mebpy)I2] (4). All the platinum(II) complexes (16) were evaluated for in vitro cytotoxicity against human cancer cell lines A2780 and A2780R, and against non-malignant MRC5 cell line. All the complexes were nontoxic up to the 50 μM concentration, although they were found to readily bind to calf-thymus DNA (CT-DNA), as determined by spectrophotometric titration (Kb ≈ 107 M?1) and ethidium bromide displacement assay.  相似文献

Two new isoprenylated benzofuran 3-ones, airlanggin A (1) and B (2) along with two known xanthones, ananixanthone (3) and trapezifolixanthone (4) were isolated from the stem bark of Calophyllum soulattri. Structures of all the compounds were elucidated using extensive spectroscopic methods, including UV, IR, HRESIMS, 1D and 2D NMR. Compounds 14 were evaluated for their cytotoxicity against P-388 cells, showing that compound 3 was the most active with IC50 0.68 μg/mL and compound 1 showed moderate activity with IC50 5.80 μg/mL.  相似文献

Abstract

The present work deals with the biological and phytochemical studies on Capparis decidua (Forssk) Edgew from Cholistan desert of Pakistan. Aerial and floral parts of C. decidua were collected and dried under shade. Powdered materials of each part of C. decidua were extracted with methanol separately, followed by phytochemical studies. Hexane fraction of aerial parts of the C. decidua obtained after solvent-solvent extraction was purified through repeated column chromatography by increasing order of polarity. Four compounds were purified and identified as simiarenol (1), lupeol (2), taraxerol (3) and β-sitosterol (4). Simiarenol and lupeol were isolated for the first time from genus Capparis. The structures of these compounds were established by comparing the spectroscopic data (1H NMR, 13C NMR, IR, UV & Mass spectrometry) reported in literature. The structure of 1 was further confirmed by XRD analysis. Anti-bacterial activities of crude methanolic extracts were determined against 13 bacterial strains (MIC 250-1000?μg/mL). α-Glucosidase and urease inhibitory activities of pure compounds were also determined. Compounds 1, 2 and 4 showed α-glucosidase inhibition with IC50 at 96.12?±?0.12, 65.28?±?0.13 and 128.14?±?0.17?μM, respectively.  相似文献

Acetone:chloroform (1:2) extract of the aerial parts of Euphorbia connata Boiss. (Euphorbiaceae) was investigated for its diterpenoids. This led to the isolation of one known and two new diterpenes, belonging to the pentahydroxy-13(17)-epoxy-8,10(18)-myrsinadiene and tetrahydroxy-5,6-epoxy-14-oxo-jatropha-11(E)-ene classes. The structures were elucidated based on 13C and 1H NMR as well as 2D NMR, IR and MS spectra and the cytotoxicity for compounds 13 were evaluated by using MTT assay against two human breast cancer cell lines. Myrsinane-type compounds – 3,7,14,15-tetraacetyl-5-propanoyl-13(17)-epoxy-8,10(18)-myrsinadiene (1) and 3,7,10,14,15-pentaacetyl-5-butanoyl-13,17-epoxy-8-myrsinene (2) – exhibited moderate inhibitory effects, with IC50 values of 24.53 ± 3.39 and 26.67 ± 1.41 μM against the MDA-MB cell line, and 37.73 ± 3.41 and 34.57 ± 2.12 μM against the MCF-7 cell line, respectively. Jatrophane-type diterpene – 5,6-epoxy-8,9,15-triacetyl-3-benzoyl-14-oxo-jatropha-11(E)-ene (3) – showed weak cytotoxicity, with IC50 values of 55.67 ± 7.09 μM against MDA-MB, and moderate cytotoxicity with IC50 values of 24.33 ± 3.21 μM against MCF-7 cell line.  相似文献

Phytochemical analysis of the leaves of Garcinia travancorica, a hitherto uninvestigated endemic species to the Western Ghats of south India, resulted in isolation and characterisation of the polyisoprenylated benzophenones 7-epi-nemorosone (1) and garcinol (2) along with biflavonoids GB-1a (3), GB-1 (4), GB-2 (5), morelloflavone (6) and morelloflavone-7″-O-β-d-glycoside or fukugiside (7). The compounds were identified using various spectroscopic techniques, mainly through NMR and MS. The methanol extract and the biflavonoids 3, 4, 5 and 7 showed potential in vitro antioxidant activities. The IC50 value of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of compound 7 was 8.34 ± 2.12 μg/mL, comparable to that of standard ascorbic acid (3.2 ± 0.50 μg/mL). In the superoxide radical scavenging assay, compound 7 gave IC50 value of 6.95 ± 1.33 μg/mL close to standard ascorbic acid with IC50 value of 5.8 ± 0.25 μg/mL. Validated HPTLC estimation revealed G. travancorica as a rich source of morelloflavone-7″-O-β-d-glycoside (7.12% dry wt. leaves).  相似文献
