传统中药五味子具有多种药理活性。20世纪70年代以来,我国临床发现五味子能改善慢性肝炎病人的肝功能异常,在此基础上,我所化学家和药理学家对五味子分离得到的20余种联苯环辛烯类木酯素成分和其药理作用进行了大量研究。五味子的药理活性主要有:保肝,抗氧化,诱导肝药酶,增强解毒功能,促进蛋白质、糖原合成,克服肿瘤耐药性,增强对抗药癌的敏感性以及对中枢神经系统的镇静作用。在这些研究基础上,合成了保肝作用最强的五味子丙素结构类似物,相继研制出两种抗肝炎新药联苯双酯(DDB)和双环醇(bicyclol)。本文对五味子木脂素类成分的结构与药理活性关系以及两种新药的创制做一综述。  相似文献

满山香[Schisandra propinqua(Wall.)Hook.f.et Thoms]为五味子科植物,云南民间代五味子药用^[1],由于从五味子科植物中已分离到不少保肝降酶,抗艾滋病毒,抗癌和PAF拮抗等活性成分,其研究受到重视^[2-4],主含满山香根和提取物的复方注射液曾在云南省几所医院临床用于治疗肺癌,但其研究仅分离鉴定了两个三萜 酸([2],为进一步寻找有效成分,我们对满山香茎藤进行了研究,从中分得8种联苯环辛二烯木脂素,经波谱和分析鉴定它们的结构为:acetylgomisin R(1) angeloylgomisin R(2),gomisinA(3),gomisin B(4),gomisinN(5),gomisinO(6),6-O-benzoylgomosin O(7)和Schisantherin A(8),1为新化合物,2系首次从五味子属植物中分得,其它均为首次从满山香中分得。  相似文献

采用密度泛函B3LYP方法在6-31G基组水平上对五味子素A、B及五味子丙素3种五味子提取物进行了优化计算,并从平衡几何构型、前线分子轨道、净电荷分布等方面对计算结果做了比较.计算结果表明分子中的二氧五环对分子的药物活性具有较大影响.随着分子中二氧五环数目的增加,分子中联苯环扭转角减小,前线轨道能级和能级差都减小,联苯环上正电荷增加,由此可判断3种分子活性顺序应为五味子丙素>五味子素B>五味子素A.  相似文献

采用液相色谱-电喷雾质谱联用(HPLC-ESI-MSn)技术, 对北五味子与南五味子中木脂素类成分进行了系统研究. 通过HPLC-ESI-MS技术, 获得了相应化合物的保留时间、紫外光谱和分子量等信息, 利用电喷雾多级串联质谱技术(ESI-MSn), 获得了相应化合物的结构信息. 研究结果表明, 北五味子与南五味子的主要木脂素成分除5个共有成分外其它成分差异较大, 并且其共有成分含量差别较大. 在此基础上, 建立了简便、快速的北五味子与南五味子药材分析鉴定的新方法.  相似文献

分别在ESI( )、ESI(-)和APCI( )3种电离检测模式下对绵马贯众抗禽流感病毒活性部位进行了液相色谱分离及离子阱质谱检测,共有21种组分能在2种或3种检测模式下被同时检测到.比较分析3种检测模式下得到的总离子流色谱图及各色谱峰的多级质谱图,并参考相关文献,推测活性部位中含有15种间苯三酚衍生物,是绵马贯众抗AIV活性部位的主要成分.  相似文献

建立了用高效液相色谱法测定五味子和含五味子的制剂中五味子乙素的流动相体系。用Shim-pack VP-ODS (250 mm×4.6 mm,5 μm)色谱柱,在柱温30 ℃、检测波长285 nm、流速1.0 mL/min的条件下,用不同的流动相组成及其不同比例研究了五味子提取液中3种木脂素(五味子乙素、安五脂素和五味子甲素)的保留时间及其分离情况,从中选择测定五味子乙素的最佳流动相体系。结果表明,在甲醇-水、甲醇-冰醋酸-水的流动相体系中,安五脂素和五味子乙素很难完全分离;而在乙腈-甲醇-水和乙腈-冰醋酸-水流动相体系中,3种木脂素可很好地分离。以乙腈-甲醇-水(体积比为17:58:25)为流动相测定五味子和护肝片中3种木脂素的含量,相对标准偏差(n4)为0.95%~5.8%,平均加标回收率为94.50%~105.6%。将该流动相体系用于供试品中3种木脂素含量的测定,分离效果好,结果令人满意。  相似文献

从四川宜宾产翼梗五味子的果实中分得七个四氢萘木脂素, 其中三个鉴定为: 恩施辛enshicine, 表恩施辛epienshicine和schisandrone, 其余四个均属新化合物, 命名为五脂素wnlignan A1, 五脂素A2, 表五脂素epiwnlignan A1和epischisandrone. 它们的结构(包括绝对构型)由光谱分析和化学转化为(+)-dimethylguaiacine和(-)-dimethylisoguaiacine而阐明. 四个新化合物皆有不同程度的体外抗癌活性.  相似文献

This investigation has resulted in the isolation of a new phenolic glycoside (1), along with seven known compounds. 建立了用荧光法快速鉴别五味子和部分含五味子制剂中五味子是属于北五味子还是南五味子的方法,探明了南北五味子荧光光谱差异的主要原因。研究了五味子的五种活性成分五味子醇甲、五味子酯甲、五味子甲素、五味子乙素和安五脂素的荧光性质和五味子以及护肝片乙醇提取液的荧光光谱,并用高效液相色谱测定了供试品五种活性成分的含量。研究表明:发光效率按安五脂素、五味子乙素、五味子甲素和五味子醇甲依次减弱,五味子酯甲不具有荧光性;北五味子的主要荧光物质为五味子乙素,而南五味子的主要荧光物质是安五脂素,它们是导致南北五味子荧光光谱差异的主要原因。用荧光法鉴别南北五味子具有简单、快速和灵敏的特点  相似文献

Wuweizi (Fructus Schisandrae) is classified in traditional Chinese medicine as a superior drug, and has been used for thousands of years. Modern pharmacological research has demonstrated that most of the biological actions and pharmacological effects of Wuweizi can be attributed to its lignan constituents, particularly the dibenzocyclooctadiene-type lignans, which can lower the serum glutamate-pyruvate transaminase (SGPT) level, inhibit platelet aggregation, and show antioxidative, calcium antagonism, antitumor-promoting, and anti-HIV (human immunodeficiency virus) effects. The dried ripe fruits of both Schisandra chinensis and Schisandra sphenanthera have long been used as Wuweizi, although their chemical constituents and contents of the bioactive components are quite different. Since 2000, they have been accepted as two different crude drugs, Bei-Wuweizi and Nan-Wuweizi, respectively, by the Chinese Pharmacopoeia. In order to provide a useful reference for good quality control of Wuweizi, many studies on the chemical constituents, pharmacological effects, identification and quality control methods of the two drugs have been reported in the literature and are summarized herein. Particular attention was given to the different methodologies developed for the qualitative and quantitative analysis of the major bioactive lignans. In our opinion, thin-layer chromatography (TLC) is the most simple and convenient method for identification of these two crude drugs, and high-performance liquid chromatography with UV detection (HPLC-UV) is the preferred method for quantitative analysis based on the bioactive lignans. Some newly developed methods, particularly hyphenated chromatographic-analytical techniques, are effective in determination of the lignans that occur in low content and those difficult to be fully separated with HPLC.  相似文献

The essential oil of Schisandra Chinensis(Turc z. ) Baill was extracted and analyzed. The essential oilwas obtained in a 1.8% (mL/100 g material) yield. Forty eight compounds were isolated and characterizedfrom the oil. Schisandra chinensis is a traditional Chinese medicine, which has played an important role in the field of medicine and food. In this study, ultra-high-performance liquid chromatography quadrupole-orbitrap-mass spectrometry was used to rapidly classify and identify the chemical compositions. Note that 32, 28, and 30 kinds of compounds were successfully identified from northern Schisandra chinensis, vinegar-processed Schisandra chinensis, and wine-processed Schisandra chinensis, respectively. The cleavage patterns of various components including lignans, organic acids, flavonoids, and terpenoids were summarized, and the effects of different processing methods on Schisandra chinensis were analyzed through chemical composition. This method realized the rapid classification and identification of raw Schisandra chinensis and two different processed products, and provided references for improving the traditional processing methods, strengthening quality control, and ensuring safe clinical application.  相似文献

One novel, highly oxygenated nortriterpenoid, schintrilactone C ( 1 ), and four known compounds, 2 – 5 , were isolated from the rattan of Schisandra sphenanthera. Their structures were determined by analysis of 1D‐ and 2D‐NMR spectroscopic data. Schintrilactone C is the third example of wuweiziartane‐type nortriterpenoids, bearing a modified five‐membered D ring, a δ‐lactone E ring, and a spirocyclic moiety in the side chain at C(13).  相似文献

通过超高效液相色谱法,对不同生长特征的五味子样品中5种主要的木脂素成分:五味子醇甲、五味子醇乙、五味子酯甲、五味子甲素和五味子乙素进行测定,同时考察其抗氧化活性。 对所得到的5种木脂素成分的含量及抗氧化数据进行聚类分析和主成分分析,并且通过逐步判别分析建立判别函数,从而筛选五味子优良品种。 主成分分析和聚类结果基本一致,均将样品分为3大类,逐步判别分析的正确率高达90%。  相似文献
