A method was developed for the determination of the major storage lipids, wax ester and triglycerides, in the copepod Calanus finmarchicus. A variation of the Folch method was used to extract the lipid. The method was scaled down to enable the extraction of either pooled (-1 mg) or individual (approximately 200 microg) copepods. The major lipid classes were identified using TLC and quantified using HPLC coupled with evaporative light scattering detection. Analysis of laboratory reference materials indicated that this method underestimated the minor triglyceride component, but gave a good estimate of the major wax ester component. The fatty acid and fatty alcohol composition of the C. finmarchicus were determined following trans-esterification of the lipid extract in methanol. Fatty acids and fatty alcohols were initially identified by comparison with authentic standard and by mass spectroscopy. Using GC with flame ionisation detection the normalised area percentage of the fatty alcohols and fatty acid methyl esters was determined simultaneously in one run for either pooled or individual copepod samples. These methods were applied to C. finmarchicus collected from the Irminger Sea, North Atlantic in 2001 and 2002.  相似文献

A new infrared spectroscopic method suitable for determining total fatty alcohol and fatty acid ester concentrations in industrial oils has been developed. Oil samples were diluted with toluene (1:3 w/w), the toxicity and volatility of which are relatively low compared with more commonly used IR solvents, like carbon tetrachloride or carbon disulfide. Mixture standards were prepared from dodecanol, tetradecanol, octadecanol, methyl stearate and methyl palmitate. Some analytical and statistical tests were performed on the developed method. The recoveries and the repeatability of the method proved to be sufficient for the quantitative determination of fatty alcohol and fatty acid ester additives in industrial oils. Reproducibility testing in another laboratory also produced satisfactory results. The developed method also proved to be relatively quick and simple. This method was developed to satisfy industry’s need to determine the concentrations of these oil additives, and it has already been applied successfully in machinery oil analysis.  相似文献

Total lipids have been extracted from human serum with chloroform–methanol 2:1 (v/v) and separated into individual classes by TLC. After transesterification the fatty acid methyl esters were analyzed by capillary gas chromatography on an FFAP column. The quantitation of ω-3 fatty acids has been performed using internal and external standards. Internal lipid standards for each lipid class were carried throughout the entire analytical procedure. Under normal diet eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are incorporated into the lipid classes to different extents: cholesterol esters; EPA, 6.5 ± 1.9 γ/ml serum; DHA, 4.3 ± 1.9 μg/ml: phospholipids; EPA, 5.9 ± 2.7 μg/ml; DHA, 31.8 ± 8.1 μg/ml. Fish oil supplementation leads to a 4 to 6-fold rise in EPA and to an approximately 2-fold rise in DHA.  相似文献

A new and simple procedure has been developed that allows the direct transesterification of lipids, using aluminium chloride as a catalyst and methanol as the esterifying alcohol. The concentration of the salt and reaction conditions have been investigated for the different lipid classes. Comparative studies, performed with boron trifluoride-methanol, indicate that the same values are obtained when using either reagent. In addition, the method has been adapted for transesterification in the presence of silica gel and other adsorbents, thus allowing the preparation of fatty acid methyl or ethyl esters directly from samples previously fractionated by thin-layer chromatography. This new reagent is very stable and easy to handle, the fatty acids being generated in the same tube without further purification steps.  相似文献

Lipoxygenase derived monohydroxy polyunsaturated fatty acid methyl esters were separated by chiral high-performance liquid chromatography (HPLC) with a Chiralcel OD-H column in the normal-phase mode. Major lipoxygenase derivatives of linoleic, -linolenic and arachidonic acids are well resolved by this column, provided they have been individually purified. Our method allows an easy and rapid determination of lipoxygenases enantioselectivity. In all cases tested the R enantiomer is eluted first.  相似文献

A method using sequential supercritical fluid extraction (SFE) and enzymatic transesterification has been developed for the rapid determination of total nutritional fat content in meat samples. SFE conditions of 12.16 MPa and 50°C were utilized to extract lipid species from the sample matrix. The enzymatic transesterification of the lipids by methanol was catalyzed by an immobilized lipase isolated from Candida antarctica. Conversion of the triglycerides to fatty acid methyl esters was monitored by supercritical fluid chromatography, while the fatty acid content of the extract was determined by capillary gas chromatography (GC). Total fat, saturated fat and monounsaturated fat contents were calculated from the GC data and compared to values from traditional extraction and lipid determination methods. Both off-line SFE and automated SFE followed by on-line GC analysis using two different instruments were utilized in this study. The enzymatic-based SFE method gave comparable results to the organic solvent extraction-based method followed by conventional BF3-catalyzed esterification.  相似文献

Two methyl esters of fatty acids, namely octadecanoic acid methyl ester (methyl stearate) and hexadecanoic acid methyl ester (methyl palmitate), in addition to four cinnamyl alcohol derivatives, sinapyl alcohol, coniferyl alcohol, p-coumaryl alcohol and coniferyl alcohol 4-O-glucoside (coniferin), were isolated from callus cultures of Wedelia prostrata. The structure of coniferin was established by spectroscopic and chemical methods, while the other compounds were identified by gas chromatography-mass spectrometry and thin layer chromatography in comparison with standards.  相似文献

Measurement of contamination of marine and naval diesel fuels (arising from product mixing or adulteration) with biodiesel or fatty acid methyl esters can be problematic, especially at very low levels. A suitable solution for this task for trace amounts of individual fatty acid methyl esters with resolution and quantification can be achieved by using a multidimensional gas chromatographic approach with electron and chemical ionization mass spectrometric detection. A unique column set comprising a 100 m methyl‐siloxane nonpolar first dimension column and high‐temperature ionic liquid column in the second dimension enabled identification of individual fatty acid methyl esters at below the lowest concentrations required to be reported in a diesel fuel matrix. Detection limits for individual fatty acid methyl esters compounds ranged from 0.5 to 5.0 mg/L, with excellent linearity up to 5000 mg/L and repeatability of the method from 1.3 to 3.2%. The method was applied to the analysis of diesel fuel samples with suspected biodiesel contamination. Contamination at 568 mg/L was calculated for an unknown sample and interpretation of the results permitted the determination of a likely source of the contamination.  相似文献

An analytical method for the determination of fatty acid ethyl esters in raw spirits of different quality or produced from various raw materials has been developed and optimized. A combination of headspace solid phase microextraction (HS-SPME) as the extraction technique and gas chromatography with flame ionization detection (GC-FID) as the determination technique was utilized. HS-SPME conditions such as: type of the stationary phase of the fiber, ethanol content, sample volume, extraction temperature and time, salt addition and sample agitation were investigated to determine the most suitable conditions for the analysis of volatile fatty acid ethyl esters in raw spirits. The quantification method was an internal standardization using methyl octanoate as the internal standard. The method's detection limits (MDLs) for the individual ethyl esters ranged from 26.8 to 0.0470 μg L−1 20% EtOH. The feasibility of SPME for the quantitative analysis of fatty acid ethyl esters in raw spirits of different organoleptic quality was demonstrated. High precision and simple sample preparation enable the use of this method for routine investigations in both industrial and research laboratories.  相似文献

A new capillary high‐performance liquid chromatography method with atmospheric pressure chemical ionization mass spectrometry was developed for the analysis of fatty acid methyl esters and long‐chain alcohols. The chromatographic separation was achieved using a Zorbax SB‐C18 HPLC column (0.3 × 150 mm, 3.5 μm) with a mobile phase composed of acetonitrile and formic acid and delivered isocratically at a flow rate of 10 μL/min. The column temperature was programmed simply, using a common column oven. Good reproducibility of the temperature profile and retention times were achieved. The temperature programming during the isocratic high‐performance liquid chromatography run had a similar effect as a solvent gradient; it reduced retention times of later eluting analytes and improved their detection limits. Two atmospheric pressure chemical ionization sources of the mass spectrometry detector were compared: an enclosed conventional ion source and an in‐house made ion source with a glass microchip nebulizer. The enclosed source provided better detectability of saturated fatty acid methyl esters and made it possible to determine the double bond positions using acetonitrile‐related adducts, while the open chip‐based source provided better analytical figures of merit for unsaturated fatty acid methyl esters. Temperature‐programmed capillary high‐performance liquid chromatography is a promising method for analyzing neutral lipids in lipidomics and other applications.  相似文献

Primary fatty acid amides are a group of biologically highly active compounds which were already identified in nature. Here, these substances were determined in tallow and tallow fatty acid methyl esters for the first time. As tallow is growing in importance as an oleochemical feedstock for the soap manufacturing, the surfactant as well as the biodiesel industry, the amounts of primary fatty acid amides have to be considered. As these compounds are insoluble in tallow as well as in the corresponding product e.g. tallow fatty acid methyl esters, filter plugging can occur. For the quantification in these matrices a purification step and a LC-APCI-MS method were developed. Although quantification of these compounds can be performed by GC-MS, the presented approach omitted any derivatization and increased the sensitivity by two orders of magnitude. Internal standard calibration using heptadecanoic acid amide and validation of the method yielded a limit of detection of 18.5 fmol and recoveries for the tallow and fatty acid methyl ester matrices of 93% and 95%, respectively. A group of commercially available samples were investigated for their content of fatty acid amides resulting in an amount of up to 0.54%m/m (g per 100 g) in tallow and up to 0.16%m/m (g per 100 g) in fatty acid methyl esters.  相似文献

Summary Acid catalyzed hydrolysis of fatty alcohol sulfates under reflux condition or by microwave heating is compared. Microwave hydrolysis turned out to be faster and more convenient to use. The combination of reflux hydrolysis of fatty alcohol sulfates or alkyl polyglycosides and the simultaneous steam distillation of the generated fatty alcohols gives a very powerful sample preparation and enrichment method for fatty alcohol sulfates and alkyl polyglycosides. The preconcentrated fatty alcohols are derivatized and detected via fluorescence detection. The applicability of the method is demonstrated by the analysis of fatty alcohol sulfates in cosmetics and in river water as matrices spiked with approximately 4 ppb fatty alcohol sulfates.  相似文献

Separation of fatty acid methyl esters and dimethyl acetals from complex biological samples has been achieved by gas-liquid chromatography on a capillary column coated with free fatty acid phase. Response-correcting factors were determined, showing rather large variations with fatty acid length. Polyunsaturated fatty acid methyl esters were shown to have lower responses than saturated species, whereas dimethyl acetals and equivalent methyl esters were found to give similar responses. Total fatty acid and aldehyde compositions of human and simian erythrocytes were determined and compared, showing a somewhat higher level of linoleate and arachidonate, and a lower level of plasmalogens in simian erythrocytes.  相似文献

Summary Polyunsaturated fatty acids have been analysed as methyl esters by liquid chromatography on porous graphitic carbon and the results compared with those obtained on octadecyl bonded phases. Chromatographic behaviour on octadecyl bonded phases arises principally as a result of hydrophobic interactions with the bonded phase. Because the retention of analytes is greater on porous graphitic carbon than on octadecyl phases, organic mobile phases are required. When the number of double bonds is low (ca 1–3), the behaviour of porous graphitic carbon is similar to that of octadecyl bonded phases, but when this number increases stronger interactions with the flat surface of the graphite appear, resulting in new selectivity. These two ‘reversed-phase’ systems are considered complementary for separation of different fatty acid methyl esters. An additional advantage of porous graphitic carbon is that it enables isolation of hexadecartrienoic and hexadecadienoic acids, which are not available commercially.  相似文献

Summary The present study was undertaken in order to characterize then to purify fatty acids from marine phytoplankton. From a crude mixture of fatty acid methyl esters it was possible to isolate by countercurrent chromatography a mixture of four polyunsaturated fatty acid methyl ester identified as being hexadecatrienoic acid methyl ester, octadecatetraenoic acid methyl ester, eicosapentaenoic acid methyl ester and docosahexaenoic acid methyl ester by gas chromatography coupled with mass spectrometry in electron impact and in positive-ion chemical ionization mode. The four polyunsaturated fatty acids are in different ratios in mixtures from the two microorganisms:Skeletonema costatum andIsochrysis galbana.  相似文献

An aminopropyl-based solid-phase extraction technique was used for isolation of chlorinated fatty acids in lipids. A range of different chlorinated fatty acids was eluted in a small volume of solvent (4 ml) and the recoveries of the different species and isomers were quantitative. Only 1% of the vastly dominating unchlorinated fatty acid methyl esters were recovered in the fractions containing the chlorinated fatty acid methyl esters. This method makes it possible to isolate and detect > or = 1 microg of a chlorinated fatty acid methyl ester in 1 g of lipid.  相似文献

Simple and rapid quantitative determination of fatty-acid-based biofuels is greatly important for the study of genetic engineering progress for biofuels production by microalgae. Ideal biofuels produced from biological systems should be chemically similar to petroleum, like fatty-acid-based molecules including free fatty acids, fatty acid methyl esters, fatty acid ethyl esters, fatty alcohols and fatty alkanes. This study founded a gas chromatography-mass spectrometry (GC-MS) method for simultaneous quantification of seven free fatty acids, nine fatty acid methyl esters, five fatty acid ethyl esters, five fatty alcohols and three fatty alkanes produced by wild-type Synechocystis PCC 6803 and its genetically engineered strain. Data obtained from GC-MS analyses were quantified using internal standard peak area comparisons. The linearity, limit of detection (LOD) and precision (RSD) of the method were evaluated. The results demonstrated that fatty-acid-based biofuels can be directly determined by GC-MS without derivation. Therefore, rapid and reliable quantitative analysis of fatty-acid-based biofuels produced by wild-type and genetically engineered cyanobacteria can be achieved using the GC-MS method founded in this work.  相似文献

建立了孔石莼脂肪酸的气相色谱/质谱(GC/MS)测定方法。使用Folch法提取了孔石莼中的总脂,经过2 mol/L HCl-甲醇溶液的甲酯化处理后,采用GC/MS法对其脂肪酸组成进行了分离分析,同时结合有机质谱学规律,分别对饱和脂肪酸甲酯、单不饱和脂肪酸甲酯和多不饱和脂肪酸甲酯的裂解规律和质谱特征进行了分析归纳。通过质谱数据库检索和标准品对照,鉴定出孔石莼中的24种脂肪酸,其中9,12,15-十八碳三烯酸、4,7,10,13-十六碳四烯酸和6,9,2,15-十八碳四烯酸3种主要多不饱和脂肪酸占总脂肪酸含量的45.14%。通过对孔石莼中脂肪酸的分析,表明特征离子在脂肪酸甲酯尤其是多不饱和脂肪酸甲酯的定性方面具有很好的应用价值。  相似文献

The constituents of total lipids and neutral lipid classes, i.e., hydrocarbons, wax esters, sterol esters, triacylglycerols, free fatty acids, 1,3-diacylglycerols, 1,2-diacylglycerols, free sterols, alcohols, and monoacylglycerols of three species of Citrus namely Citrus sinensis, Citrus paradisi, Citrus aurantium were investigated by thin layer and gas chromatography. Palmitic, oleic and linoleic acids were the major components in all the lipids and lipid classes studied.  相似文献
