We successfully synthesized poly(l ‐lactide)‐b‐poly (methyl methacrylate) diblock copolymers at ambient temperature by combining ultraviolet light‐induced copper‐catalyzed ATRP and organo‐catalyzed ring‐opening polymerization (ROP) in one‐pot. The polymerization processes were carried out by three routes: one‐pot simultaneous ATRP and ROP, one‐pot sequential ATRP followed by ROP, and one‐pot sequential ROP followed by ATRP. The structure of the block copolymers is confirmed by nuclear magnetic resonance and gel permeation chromatography, which suggests that the polymerization method is facile and attractive for preparing block copolymers. © 2018 Wiley Periodicals, Inc. One‐step synthesis of block‐graft copolymers by reversible addition‐fragmentation chain transfer (RAFT) and ring‐opening polymerization (ROP) by using a novel initiator was reported. Block‐graft copolymers were synthesized in one‐step by simultaneous RAFT polymerization of n‐butylmethacrylate (nBMA) and ROP of ε‐caprolacton (CL) in the presence of a novel macroinitiator (RAFT‐ROP agent). For this purpose, first epichlorohydrin (EPCH) was polymerized by using H2SO4 via cationic ring‐opening mechanism. And then a novel RAFT‐ROP agent was synthesized by the reaction of potassium ethyl xanthogenate and polyepichlorohydrin (poly‐EPCH). By using the RAFT‐ROP agent, poly[CL‐b‐EPCH‐b‐CL‐(g‐nBMA)] block‐graft copolymers were synthesized. The principal parameters such as monomer concentration, initiator concentration, and polymerization time that affect the one‐step polymerization reaction were evaluated. The block lengths of the block‐graft copolymers were calculated by using 1H‐nuclear magnetic resonance (1H NMR) spectrum. The block length could be adjusted by varying the monomer and initiator concentrations. The characterization of the products was achieved using 1H NMR, Fourier‐transform infrared spectroscopy, gel‐permeation chromatography, thermogravimetric analysis, differential scanning calorimetry, elemental analysis, and fractional precipitation (γ) techniques. © 2013 Wiley Periodicals, Inc. The synthesis of polypeptide‐containing block copolymers combining N‐carboxyanhydride (NCA) ring‐opening polymerization and atom transfer radical polymerization (ATRP) was investigated. An amide initiator comprising an amine function for the NCA polymerization and an activated bromide for ATRP was used. Well‐defined polypeptide macroinitiators were obtained from γ‐benzyl‐L ‐glutamate NCA, O‐benzyl‐serine NCA, and N‐benzyloxy‐L ‐lysine. Subsequent ATRP macroinitiation from the polypeptides resulted in higher than expected molecular weights. Analysis of the reaction products and model reactions confirmed that this is due to the high frequency of termination reactions by disproportionation in the initial phase of the ATRP, which is inherent in the amide initiator structure. In some cases selective precipitation could be applied to remove unreacted macroinitiator to yield well‐defined block copolymers. © 2009 Wiley Periodicals, Inc. A range of well‐defined block copolymers were synthesized using 4‐cyano‐4‐(dodecylsulfanylthiocarbonyl)sulfanylpentanol (CDP) as a dual initiator for reversible addition‐fragmentation chain transfer (RAFT) polymerization and ring‐opening polymerization (ROP) in a one‐step process. Styrene, (meth)acrylate, and acrylamide monomers were polymerized in a controlled manner for one block composed of vinyl monomers, and δ‐valerolactone (VL), ε‐caprolactone (CL), trimethylene carbonate (TMC), and L ‐lactide (LA) were used for the other block composed of cyclic monomers. Diphenyl phosphate was used as a catalyst for the ROP of VL, CL, and TMC, and 4‐dimethyamino pyridine for the ROP of LA. These catalysts did not interfere with RAFT polymerization and the synthesis of various block copolymers proceeded in a controlled manner. CDP was found to be a very useful dual initiator for a one‐step synthesis of various block copolymers by a combination of RAFT polymerization and ROP. © 2012 Wiley Periodicals, Inc. Amphiphilic, biocompatible poly(N‐vinylpyrrolidone)‐b‐poly(l ‐lactide) (PVP‐b‐PLLA) block polymers were synthesized at 60 °C using a hydroxyl‐functionalized N,N‐diphenyldithiocarbamate reversible addition–fragmentation chain transfer (RAFT) agent, 2‐hydroxyethyl 2‐(N,N‐diphenylcarbamothioylthio)propanoate (HDPCP), as a dual initiator for RAFT polymerization and ring‐opening polymerization (ROP) in a one‐step procedure. 4‐Dimethylamino pyridine was used as the ROP catalyst for l ‐lactide. The two polymerization reactions proceeded in a controlled manner, but their polymerization rates were affected by the other polymerization process. This one‐step procedure is believed to be the most convenient method for synthesizing PVP‐b‐PLLA block copolymers. Reported here is the first aqueous ring‐opening polymerization (ROP) of N‐carboxyanhydrides (NCAs) using α‐amino‐poly(ethylene oxide) as a macroinitiator to protect the NCA monomers from hydrolysis through spontaneous in situ self‐assembly (ISA). This ROPISA process affords well‐defined amphiphilic diblock copolymers that simultaneously form original needle‐like nanoparticles.  相似文献

A novel amphiphilic biodegradable triblock copolymer (PGL‐PLA‐PGL) with polylactide (PLA) as hydrophobic middle block and poly(glutamic acid) (PGL) as hydrophilic lateral blocks was successfully synthesized by ring‐opening polymerization (ROP) of L ‐lactide (LA) and N‐carboxy anhydride (NCA) consecutively and by subsequent catalytic hydrogenation. The results of cell experiment of PGL‐PLA‐PGL suggested that PGL could improve biocompatibility of polyester obviously. The copolymer could form micelles of spindly shape easily in aqueous solution. The pendant carboxyl groups of the triblock copolymer were further activated with N‐hydroxysuccinimide and combined with a cell‐adhesive peptide GRGDSY. Incorporation of the oligopeptide further enhanced the hydrophilicity and led to formation of spherical micelles. PGL‐PLA‐PGL showed better cell adhesion and spreading ability than pure PLA and the GRGDSY‐containing copolymer exhibited even further improvement in cell adhesion and spreading ability, indicating that the copolymer could find a promising application in drug delivery or tissue engineering. © 2007 Wiley Periodicals, Inc. New copolymers of amino acid and urethane (PAU), in which a polyurethane segment was combined with poly(γ‐methyl‐L‐glutamate) (PMLG) of various contents, were synthesized by the copolymerization of the polyurethane prepolymer (UPP) having isocyanate groups at both terminals of the chain and γ‐methyl‐L ‐glutamate‐N‐carboxyanhydride (NCA) to improve the elastic recovery and adhesion of PMLG for application of PMLG to synthetic leather. The copolymerization of the UPP with NCA was carried out by applying the reactivity of isocyanate and the polymerization mechanism of NCA using the primary amine and tertiary amine as initiators. Infrared (IR) and 13C‐NMR spectra of these PAUs as well as the chemical analysis of the PAU intermediates showed that the PAUs would have a multiblock–triblock structure: namely, the PAUs consisted of the block copolymer segments of urethane and a small amount of PMLG at the center of the copolymer chain and most of the PMLG at both terminals of the copolymer chain. The elastic recovery and adhesion of these PAUs were significantly larger than those of the PMLG with the maintenance of a good sense of touch, which was a unique asset of PMLG. Furthermore, it was found that the PAUs had intermediary moisture permeability between that of PMLG and polyurethane. © 1999 John Wiley & Sons, Inc. Atom transfer radical polymerization (ATRP) and ring‐opening polymerization (ROP) were combined to synthesize poly(?‐caprolactone‐co‐octadecyl methacrylate‐co‐dimethylaminoethyl methacrylate) copolymers possessing a triblock or random block structure. Various synthetic pathways (sequential or simultaneous approaches) were investigated for the synthesis of both copolymers. For the preparation of these copolymers, an initiator with dual functionality for ATRP/anionic ring‐opening polymerization, 2‐hydroxyethyl 2‐bromoisobutyrate, was used. Copolymers were prepared with good structural control and low polydispersities (weight‐average molecular weight/number‐average molecular weight < 1.2), but one limitation was identified: the dimethylaminoethyl methacrylate (DMAEMA) block had to be synthesized after the ?‐caprolactone block. ROP could not proceed in the presence of DMAEMA because the complexation of the amine groups in poly(dimethylaminoethyl methacrylate) deactivated tin(II) hexanoate, which was used as a catalyst for ROP. © 2005 Wiley Periodicals, Inc. Dendron‐like poly(ε‐benzyloxycarbonyl‐L ‐lysine)/linear poly(ethylene oxide) block copolymers (i.e., Dm‐PZLys‐b‐PEO, m = 0 and 3; Dm are the propargyl focal point poly(amido amine) dendrons having 2m primary amine groups) were for the first time synthesized by combining ring‐opening polymerization (ROP) of ε‐benzyloxycarbonyl‐L ‐lysine N‐carboxyanhydride (Z‐Lys‐NCA) and click chemistry, where Dm‐PZLys homopolypeptides were click conjugated with azide‐terminated PEO. Their molecular structures and physical properties were characterized in detail by FTIR, 1H NMR, gel permeation chromatography, differential scanning calorimetry, polarized optical microscopy, and wide angle X‐ray diffraction. Both homopolypeptides and copolymers presented a liquid crystalline phase transition for PZLys block, and the transition was irreversible. Moreover, the degree of crystallinity of PEO block within linear copolymers decreased from 96.2% to 20.4% with increasing PZLys composition, whereas that within dendritic copolymers decreased to zero. The secondary conformation of PZLys progressively changed from β‐sheet to α‐helix with increasing the chain length. These copolymers self‐assembled into spherical nanoparticles in aqueous solution, and the anticancer drug doxorubicin‐loaded nanoparticles gave a similar morphology compared with their blank counterparts. The drug‐loaded nanoparticles showed a triphasic drug‐release profile at aqueous pH 7.4 or 5.5 and 37 °C and sustained a longer drug‐release period for about 2 months. © 2012 Wiley Periodicals, Inc. Di‐stereoblock polylactides (di‐sb‐PLA: PLLA‐b‐PDLA) having high molecular weight (Mn > 100 kDa) were successfully synthesized by two‐step ring‐opening polymerization (ROP) of L ‐ and D ‐lactides using tin(2‐ethylhexanoate) as a catalyst. By optimizing the polymerization conditions, the block sequences were well regulated at non‐equivalent feed ratios of PLLA and PDLA. This synthetic method consisted of three stages: (1) polymerization of either L ‐ or D ‐lactide to obtain a PLLA or PDLA prepolymer with a molecular weight less than 50 kDa, (2) purification of the obtained prepolymer to remove residual lactide, and (3) polymerization of the enantiomeric lactide in the presence of the purified prepolymer. Their 13C and 31P NMR spectra of the resultant di‐sb‐PLAs strongly supported their di‐stereo block structure. These di‐sb‐PLAs, having weight‐average molecular weights higher than 150 kDa, were fabricated into polymer films by solution casting and showed exclusive stereocomplexation. The thermomechanical analysis of the films revealed that their heat deformation temperature was limited probably because of their low crystallinity owing to the non‐equivalent PLLA/PDLA ratio. The blend systems of the di‐sb‐PLAs having complementary stereo‐sequences (the one with a long PLLA block and the other with long PDLA block) were also prepared and characterized to enhance the sc crystallinity. © 2010 Wiley Periodicals, Inc. Among three cyclopentadienyl titanium complexes studied, CpTiCl2(OEt), containing a 5% excess CpTiCl3, has proven to be a very efficient catalyst for the ring‐opening polymerization (ROP) of L ‐lactide (LLA) in toluene at 130 °C. Kinetic studies revealed that the polymerization yield (up to 100%) and the molecular weight increase linearly with time, leading to well‐defined PLLA with narrow molecular weight distributions (Mw/Mn ≤ 1.1). Based on the above results, PS‐b‐PLLA, PI‐b‐PLLA, PEO‐b‐PLLA block copolymers, and a PS‐b‐PI‐b‐PLLA triblock terpolymer were synthesized. The synthetic strategy involved: (a) the preparation of OH‐end‐functionalized homopolymers or diblock copolymers by anionic polymerization, (b) the reaction of the OH‐functionalized polymers with CpTiCl3 to give the corresponding Ti‐macrocatalyst, and (c) the ROP of LLA to afford the final block copolymers. PMMA‐g‐PLLA [PMMA: poly(methyl methacrylate)] was also synthesized by: (a) the reaction of CpTiCl3 with 2‐hydroxy ethyl methacrylate, HEMA, to give the Ti‐HEMA‐catalyst, (b) the ROP of LLA to afford a PLLA methacrylic‐macromonomer, and (c) the copolymerization (conventional and ATRP) of the macromonomer with MMA to afford the final graft copolymer. Ring opening polymerization (ROP) of N‐carboxy anhydride (NCA) amino acids presents a rapid way to synthesize high molecular weight polypeptides with different amino acid compositions. The compositional and functional versatility of polypeptides make these materials an attractive choice for biomaterials. The functional performance of polypeptide materials is equally linked to their conformation which is determined by the amino acid sequence in the polymer chains. Here, the interplay between composition and conformation of synthetic polypeptides obtained by NCA polymerization was explored. Various copolypeptides from Glu(Bzl) and Ser(Bzl) were prepared to investigate how polypeptide composition affected the conformation of the resulting copolymer. Polymerization kinetics indicated that the copolymerization of Glu(Bzl) and Ser(Bzl) preferentially yielded alternating copolymers. Both the polydispersity and the conformation of the polypeptides were dependent on the Ser(Bzl) content in the polymer, demonstrating that polypeptide functionalities could be tuned directly by altering the relative amounts of amino acids in the chain. This work presents the first step toward an improved understanding and control over polypeptide conformation through modulating the amino acid composition of the material. Understanding this sequence–functionality relationship is essential to advancing the use of ROP as a technique to design smart polypeptide based materials with specific functions. © 2016 The Authors. The synthesis and characterization of a photocleavable block copolymer containing an ortho‐nitrobenzyl (ONB) linker between poly(methyl methacrylate) and poly(d ‐lactide) blocks is presented here. The block copolymers were synthesized via atom transfer radical polymerization (ATRP) of MMA followed by ring‐opening polymerization (ROP) of d ‐Lactide and ROP of d ‐lactide followed by ATRP of MMA from a difunctional photoresponsive ONB initiator, respectively. The challenges and limitations during synthesis of the photocleavable block copolymers using the difunctional photoresponsive ONB initiator are discussed. The photocleavage of the copolymers occurs under mild conditions by simple irradiation with 302 nm wavelength UV light (Relative intensity at 7.6 cm: 1500 μW/cm2) for several hours. © 2013 Wiley Periodicals, Inc. A simple, one‐step procedure has been developed for the preparation of bifunctional initiators capable of polymerizing monomers suitable for atom‐transfer radical polymerization (ATRP) and ring‐opening polymerization (ROP). These bifunctional initiators were employed for making narrow disperse poly(styrene) macroinitiators, which were subsequently used for the ROP of various lactides to yield poly(styrene‐block‐lactide) copolymers. Thermogravimetric analysis (TGA) of these block copolymers are interesting in that it shows a two‐step degradation curve with the first step corresponding to the degradation of poly(lactide) segment and the second step associated with the poly(styrene) segment of the block copolymer. This nature of the block copolymer makes it possible to estimate the block copolymer content by TGA in addition to the 1H NMR spectroscopic analysis. Thus, this study for the first time highlights the possibility of making porous materials by thermal means which are otherwise obtained by base hydrolysis. The bifunctional initiators were prepared by the esterification of 3‐hydroxy, 4‐hydroxy, and 3,5‐dihydroxy benzyl alcohols with α‐bromoisobutyryl bromide and 2‐bromobutyryl bromide. A mixture of products was obtained, which were purified by column chromatography. The esterified benzyl alcohols were employed in the polymerization of styrene under copper (Cu)‐catalyzed ATRP conditions to yield macroinitiators with low polydispersity. These macroinitiators were subsequently used in the ROP of L ‐, DL ‐, and mixture of lactides. The formation of block copolymers was confirmed by gel permeation chromatography (GPC), spectroscopic and thermal characterizations. The molecular weight of the block copolymers was always higher than the macroinitiator, and the GPC chromatogram was symmetrical indicating the uniform initiation of ROP by the macroinitiators. © 2007 Wiley Periodicals, Inc. The recently introduced procedure of quantitatively switching thiocarbonyl thio capped (RAFT) polymers into hydroxyl terminated species was employed to generate narrow polydispersity (PDI ≈ 1.2) sulfur‐free poly(styrene)‐block‐poly(ε‐caprolactone) polymers (26,000 ≤ Mn/g·mol?1 < 45,000). The ring‐opening polymerization (ROP) of ε‐caprolactone (ε‐CL) was conducted under organocatalysis employing 1,5,7‐triazabicyclo[4.4.0]dec‐5‐ene (TBD). The obtained block copolymers were thoroughly analyzed via size exclusion chromatography (SEC), NMR, as well as liquid adsorption chromatography under critical conditions coupled to SEC (LACCC‐SEC) to evidence the block copolymer structure and the efficiency of the synthetic process. The current contribution demonstrates that the RAFT process can serve as a methodology for the generation of sulfur‐free block copolymers via an efficient end group switch. © 2010 Wiley Periodicals, Inc. A monomode microwave reactor was used for the synthesis of designed star‐shaped polymers, which were based on dipentaerythritol with six crystallizable arms of poly(ε‐caprolactone)‐b‐poly(L ‐lactide) (PCL‐b‐PLLA) copolymer via a two‐step ring‐opening polymerization (ROP). The effects of irradiation conditions on the molecular weight were studied. Microwave heating accelerated the ROP of CL and LLA, compared with the conventional heating method. The resultant hexa‐armed polymers were fully characterized by means of FTIR, 1H NMR spectrum, and GPC. The investigation of thermal properties and crystalline behaviors indicated that the crystalline behaviors of polymers were largely depended on the macromolecular architecture and the length of the block chains. © 2010 Wiley Periodicals, Inc. Aliphatic polyesters have many applications in the biomedical field due to their properties and facile degradation. They are commonly synthesized via ring opening polymerization (ROP) with metal‐based catalysts, but as high temperatures are needed and the products contain metal, organocatalysts are now widely adopted to polymerize them at room temperature while also ensuring short reaction times. Here, 1,7,7‐triazabicyclo[4.4.0]‐dec‐5‐ene is used to polymerize less reactive but renewably‐derived lactones, namely δ‐decalactone and δ‐dodecalactone. These monomers were chosen in the attempt of creating renewable and highly lipophilic materials for drug delivery applications as alternatives to the more traditional, but non‐renewable δ‐valerolactone and ?‐caprolactone. A combination of ROP and living radical polymerization Reversible Addition‐Fragmentation Chain Transfer is proposed here to synthesize grafted block copolymers. They are able to self‐assemble in water, forming micelles where the lipophilic polyester core is able to entrap a lipophilic drug, thus making the system a good candidate for drug delivery. © 2017 Wiley Periodicals, Inc. A controlled metal‐free synthetic methodology toward well‐defined thermoresponsive polypeptides by decreasing the reaction temperature to 0 °C has been developed. Good control over the molecular weight in the polymerization of a trithiocarbonate‐functionalized N‐carboxyanhydride (MES‐l ‐Glu‐NCA) monomer was obtained using n‐hexylamine as the initiator at 0 °C. It yielded homopolypeptide macro‐transfer agent (PMESLG) with narrow molecular weight distribution (PDI < 1.3) and controllable chain length. Detailed 1H NMR and MALDI‐TOF‐MS analysis clearly confirmed that frequently occurring side‐reactions was absent at 0 °C, and the polymerization was controlled. The resultant PMESLG was applied to mediate the reversible addition‐fragmentation chain transfer (RAFT) polymerization of oligo‐ethylene‐glycol acrylate (OEGA) for the metal‐free synthesis of thermoresponsive polypeptides. These thermoresponsive polypeptides have well‐controlled molecular weight, adopted regular α‐helical conformation, and exhibited a lower critical solution temperature between 23 °C and 55 °C. To the best of our knowledge, there are very few reports about the synthesis of well‐defined thermoresponsive graft polypeptides via NCA polymerization and RAFT. The surface modification of hydroxyapatite (HA) nanoparticles by the ring opening polymerization (ROP) of γ‐benzyl‐L ‐glutamate N‐carboxyanhydride (BLG‐NCA) was proposed to prepare the poly(γ‐benzyl‐L ‐glutamate) (PBLG)‐grafted HA nanoparticles (PBLG‐g‐HA) for the first time. HA nanoparticles were firstly treated by 3‐aminopropylthriethoxysilane (APS) and then the terminal amino groups of the modified HA particles initiated the ROP of BLG‐NCA to obtain PBLG‐g‐HA. The process was monitored by XPS and FT‐IR. The surface grafting amounts of PBLG on HA ranging from 12.1 to 43.1% were characterized by thermal gravimetric analysis (TGA). The powder X‐ray diffraction (XRD) analysis confirmed that the ROP only underwent on the surface of HA nanoparticles without changing its bulk properties. The SEM measurement showed that the PBLG‐g‐HA hybrid could form an interpenetrating net structure in the self‐assembly process. The PBLG‐g‐HA hybrid could maintain higher colloid stability than the pure HA nanoparticles. The in vitro cell cultures suggested the cell adhesion ability of PBLG‐g‐HA was much higher than that of pure HA.
