The catalytic olefination reaction of 2-nitrobenzaldehydes with CF3CCl3 afforded stereoselectively trifluoromethylated ortho-nitrostyrenes in up to 88% yield. The reaction of these alkenes with pyrrolidine permits preparation of α-CF3-β-(2-nitroaryl) enamines. Subsequent one pot reduction of nitro-group by Fe-AcOH-H2O system initiated intramolecular cyclization to afford 2-CF3-indoles. Target products can be prepared in up to 85% yields. Broad synthetic scope of the reaction was shown as well as some followed up transformations of 2- CF3-indole.  相似文献

The effect of fluorine as a substituent group on nucleophilic aromatic substitution is discussed, where a fluorine atom located ortho to the point of substitution may be of variable activating influence, whereas fluorine located para is slightly deactivating and meta is activating. A rationale of these effects is presented and evidence to support polar influences by ortho fluorine is advanced. The influence of CN, CF3, CF2H and CFH2 is also established by comparison of appropriate measured rate constants and compared with the activation effects of ring nitrogen.  相似文献

A broad survey of aromatic compounds with halogens positioned both ortho and para to activating groups was studied in SNAr reactions with piperidine. Regioselectivities varied with the substituent group and the polarity of the solvent. Many activating groups exhibited an overall bias toward ortho-substitution, and this led in nonpolar solvents to very high ortho selectivity. More polar solvents uniformly shifted the product ratio toward para substitution. Evidence is presented that argues for coordination via hydrogen bonding as a driver of much of the ortho selectivity observed. The data presented show ample evidence of the generality and synthetic utility of the ortho-directing ability of several common activating groups for this reaction type.  相似文献

The nucleophilic addition of organic anions to aromatic compounds with halogens positioned both ortho and para to activating groups was studied in a variety of solvents. Substrates showed strong preferences for ortho substitution in most cases. Evidence is presented for activating group-dependent coordination, which contributes to very high ortho-selectivity in nonpolar solvents. This also drives the overall reaction rate in these solvents, and is of close to the same magnitude of rate increase derived from polar solvents. para-Products are maximized by using crown ethers in protic solvents. Solvent effects overall are very different from corresponding reactions with amine nucleophiles due primarily to the different charges present in the transition states, and to solvation of the nucleophile.  相似文献

A practical synthesis of ortho-silyl-substituted phenol from ortho-bromophenyl silyl ethers without using RLi is described. Various ortho-bromophenyl silyl ethers are treated with commercially available Mg turnings, which are easy to handle in air, and transfer of the silyl group to the ortho-position occurs in good to high yields. Selective mono-magnesiation of 2,6-dibromophenyl silyl ether is observed even in the presence of excess Mg, and ortho-bromo-6-silylphenol is obtained as the predominant product. The obtained ortho-silyl-substituted phenol is formylated with (CH2O)n/MgCl2/Et3N, and then condensation with a diamine leads to a silyl-substituted salen-type ligand in a good yield. This scheme is suitable for the large scale synthesis of silyl-substituted salen-type ligands bearing imine groups.  相似文献

We report here ruthenium-catalyzed arylation of fluorinated aromatic ketones via ortho-selective carbon-fluorine bond cleavage. In the presence of trimethylvinylsilane and cesium fluoride, ortho carbon-fluoride bonds of aromatic ketones were phenylated by 5,5-dimethyl-2-phenyl-1,3,2-dioxaborinane using RuH2(CO)(PPh3)3 as a catalyst. Tandem C-F phenylation/C-H alkylation was observed for substrates bearing both one ortho hydrogen and one ortho fluorine atoms.  相似文献

The ortho-lithiation of a phenyl phosphorodiamidothioate derived from an enantiopure C2-symmetric diamine is studied. It is shown that the migration of the diaminophosphoryl group from sulfur to carbon, leading to an ortho-sulfanylated phenyl phosphonodiamidate, only occurs in the presence of an alkylating agent or a Lewis acid as BF3·Et2O. The influence of the chiral diaminophosphoryl group on the stereoselectivity of the oxidation of the ortho-sulfanyl or alkylsulfanyl group is also examined.  相似文献

The synthesis, structural characterization, and coordination behavior of ditopic ortho-hydroquinone-based bis(pyrazol-1-yl)methane ligands (ortho-(OH)2C6H3-4-CHpz2, ortho-(OH)2C6H3-4-CH(3-Phpz)2, and ortho-(OH)2C6H3-4-CH(3-tBupz)2) with pyrazole, 3-phenylpyrazole, and 3-tert-butylpyrazole as donors are described. The reaction of a soluble PdCl2-source with ortho-(OH)2C6H3-4-CHpz2 in acetonitrile yielded the related square-planar N,N-coordinated Pd(II) dichloride complex, whereas treatment of ortho-(OH)2C6H3-4-CH(3-Phpz)2 or ortho-(OH)2C6H3-4-CH(3-tBupz)2 with PdCl2 in acetonitrile resulted in degradation of these ligands. The Pd(II) complexes trans-(3-PhpzH)2PdCl2 and trans-(3-tBupzH)2PdCl2 were isolated and fully characterized including X-ray diffraction analyses.  相似文献

A mild and general strategy for the synthesis of 2-substituted 4-amino-1,2,4,5-tetrahydro-2-benzazepine-3-ones is described. The seven-membered lactam is prepared by intramolecular amide bond formation from the intermediate amino acid, which is obtained either by reductive alkylation of a variety of amines with N-Boc,N-Me-ortho-formyl-Phe and Phth-ortho-formyl-Phe, or by reductive amination of a variety of aldehydes with N-Boc-ortho-aminomethyl-Phe.  相似文献

The air and moisture stable triphenylphosphine adduct of cyclopalladated ferrocenylimine 2 has been successfully used in palladium-catalyzed Suzuki cross-coupling for the synthesis of ortho-substituted biaryls in air. In the presence of 0.05 mol% of 2 as catalyst and 3 equivalent of CsF as base in dioxane at 100 °C, ortho-substituted biaryls were synthesized with moderate to high yields in the reactions of 2-methoxy-1-naphthylboronic acid with aryl halides, and 14 new ortho-substituted biaryls were obtained and characterized.  相似文献

One-pot syntheses of (3-hydroxybenzo[b]thiophen-2-yl) aryl methanones from ortho-methylsulfanylaryl N,N-diethyl amides and of 1-(3-hydroxybenzo[b]thiophen-2-yl)ethanone and 1-(3-hydroxybenzo[b]thiophen-2-yl)propan-1-one via an anionic ortho-Fries rearrangement are described. Rate constants have been measured for the reactions of ammonia with various fluorinated pyridines and diazines in aqueous dioxan at 25°. From the results the activating effects of ring-nitrogen (relative to CH) and of trifluoromethyl (relative to -H) have been determined. Ring-nitrogen activates the system at points ortho-, meta- and para- to the point of substitution, in the ratios ortho- 6.2 × 104, meta- 8.5 × 102, and para- 2.3 × 105. Similarly a trifluoromethyl substituent is activating by a factor of 2.4 × 103ortho- and 4.5 × 103para- to the point of substitution.  相似文献

Aryl azoles are ubiquitous as bioactive compounds and their regioselective functionalization is of utmost synthetic importance. Here, we report the development of a toluene-soluble dialkylmagnesium base sBu2Mg. This new reagent allows mild and regioselective ortho-magnesiations of various N-arylated pyrazoles and 1,2,3-triazoles as well as arenes bearing oxazoline, phosphorodiamidate or amide directing groups. The resulting diarylmagnesium reagents were further functionalized either by Pd-catalyzed arylation or by trapping reactions with a broad range of electrophiles (aldehydes, ketones, allylic halides, acyl chlorides, Weinreb amides, aryl halides, hydroxylamine benzoates, terminal alkynes). Furthermore, several double ortho,ortho′-magnesiations were realized in the case of aryl oxazolines, N-aryl pyrazoles as well as 2-aryl-2H-1,2,3-triazoles by simply repeating the magnesiation/electrophile trapping sequence allowing the preparation of valuable 1,2,3-functionalized arenes.

A toluene solution of sBu2Mg allowed a regioselective ortho-magnesiation of aromatic and heterocyclic systems. A second ortho,ortho′-functionalization was achieved in the case of aryl oxazolines, N-aryl pyrazoles as well as N-aryl triazoles.  相似文献

ortho-Linked oxacalix[n]arene[n]hetarenes (n=2, 3) were prepared by one-step cyclooligomerization of catechol and meta-dichlorinated nitrogen containing heterocycles or via a two-step reaction process. Solid state structure of the ortho-linked oxacalix[n]arene[n]hetarenes (n=2, 3) has been determined by X-ray crystallography. 1,3-Alternate and 1,3,5-alternate conformations were found for ortho-linked oxacalix[2]arene[2]pyrazine (1) and oxacalix[3]arene[3]pyrazine (2), respectively. However, a core conformation with C3 symmetry was found for ortho-linked oxacalix[3]arene[3]pyrimidine (4), which is completely different from that of its isomer, compound 2.  相似文献

Addition of ethylene to the carbanions formed by the metallation of the lithium salts of di- and trimethylphenols by the strongly basic system, n-BuLi-LiK(OCH2CH2NMe2)2 provides a useful synthetic route to a range of alkylphenols. The ease of alkylation of the methyl groups decreases in the order ortho>meta>para while the inclusion of Mg(OCH2CH2OEt)2 in the catalyst restricts alkylation to the methyl groups ortho to the hydroxy group. Dialkylation occurs only at the ortho-methyl groups and only if the adjacent meta-position is unsubstituted. The potential of these products for the synthesis of sterically hindered ligands is outlined.  相似文献

The functionalisation of the title compounds via regioselective Directed ortho-Metallation (DoM) and cross-coupling reactions is studied. The compatibility of the 3,7-DiOxa-r-1-AzaBicyclo[3.3.0]Oct-c-5-ylmethoxy system (DOABO-CH2O) to typical reaction conditions is established. Its role as Directed ortho-Metallation Group (DoMG) is examined, including competition with classical DoMGs, chlorine and methoxy. The chelating ability of some functionalised terms such as DOABO-CH2O substituting chiral diarylmethanols and polyaza analogues of 2,6-terpyridine is discussed as intramolecular steric relationships determining configuration and aptitude to bind selectively transition metals, respectively.  相似文献

Ortholithiation of 4-cyanopyridine using 2,2,6,6-tetramethylpiperidide (LiTMP) and trapping the lithio intermediate with electrophiles represents an efficient and straightforward access to ortho-substituted-4-cyanopyridines. The cyano group can be used as an ortho-directing group and allows the preparation of 3-halogeno-4-cyanopyridines. Reactivity of 2- and 3-cyanopyridines is also investigated and seems to give similar results.  相似文献

The syntheses are reported of the ether-phosphine ligands: 2-(ortho-diphenylphosphinophenyl)-1,3-dioxolane (1a), 2-(ortho-diisopropylphosphinophenyl)-1,3-dioxolane (1b), 2-(ortho-diphenylphosphinophenyl)-1,3-dioxane (1c), 2-(ortho-diisopropylphosphinophenyl)-1,3-dioxane (1d). Their reaction with [(COD)RhCl]2 (COD: 1,5-cyclooctadiene) results in the formation of the mononuclear complexes: {chloro(COD)[2-(ortho-diphenylphosphinophenyl)-1,3-dioxolane]rhodium(I)} (2a), {chloro(COD)[2-(ortho-diisopropylphosphinophenyl)-1,3-dioxolane]rhodium(I)} (2b), {chloro(COD)[2-(ortho-diphenylphosphinophenyl)-1,3-dioxane]rhodium(I)} (2c), and {chloro(COD)[2-(ortho-diisopropylphosphinophenyl)-1,3-dioxane]rhodium(I)} (2d). The chloride ligands of compounds 2a and 2b were abstracted with TlPF6, with accompanied insertion of an acetal oxygen atom of the ligands 1a and 1b into the coordination sphere of the metal centre, producing {(COD)[η2-P,O-2-(ortho-diphenylphosphinophenyl)-1,3-dioxolane]rhodium(I)}PF6 (3a∗PF6) and {(COD)[η2-P,O-2-(ortho-diisopropylphosphinophenyl)-1,3-dioxolane]rhodium(I)}PF6 (3b∗PF6). In contrast the dioxane analogues of 3, 3c∗BF4 and 3d∗BF4, were formed by reacting the ligands 1c, 1d with [Rh(COD)2]BF4. The ligands 1 and the complexes 2 serve as model compounds for their via acetalation to a polyvinylalcohol resin bound analogues. The complexes synthesised were employed as pre-catalysts in the hydroformylation reaction of 1-octene.  相似文献

Condensations of o-halo-substituted benzenesulfonyl chlorides with 2-aminopyridines and amidines may give the corresponding 1,2,4-benzothiadiazine-1,1-dioxides under mild, non-catalytic conditions in nearly quantitative yields. The successful one-pot cyclization depends on three factors: (i) the nature of the o-halogen, (ii) the electronic character of the benzene ring substituent, and (iii) the steric load around the amidine unit. O-Fluorobenzenesulfonyl chlorides bearing methylcarboxyl- or nitro-group and o-chloro- and o-bromobenzenesulfonyl chlorides bearing nitro-group are reactive enough to give the desired 1,2,4-benzothiadiazine-1,1-dioxides in a one-pot base-promoted reaction. In all other cases, open-chain sulfonylated amidine intermediates are isolated. The latter are converted to the title compounds either in the presence of potassium carbonate or upon the addition of a copper(I) catalyst.  相似文献

Nitro-groups positioned ortho to thio-esters have been shown to engage in strong, non-bonded intramolecular interactions between the sulfur atom and one of the nitro-group oxygens. The effect of the sulfur-nitro group oxygen interaction on the chemical reactivity at the sulfur atom of 9-nitro-1-azaphenoxathiin is reported. Conditions which normally produce the sulfone exclusively have been shown to yield the sulfoxide with only a minimal quantity of the sulfone produced. Protracted periods of reflux are required to produce the sulfone from the sulfoxide.  相似文献
