The 4,4′,6,6′-tetrasubstituted 2,2′-alkylidenebis(phenols) 1 reacted with CISCOI to give spiro[1,3-benzoxathiepin-4(5H), 1′-cyclohexa[2,4]diene]-2,2′-diones 4 , together with cyclic carbonates 5 . The structures of the products were elucidated mainly by 13C-NMR and 1H-NMR spectroscopy.  相似文献

The three‐component reaction of thiazole (benzothiazole), dialkyl but‐2‐ynedioate, and isatinylidene malononitriles in toluene at 110–120°C in a sealed tube afforded a mixture of cis/trans‐isomers of functionalized diastereoisomeric spiro[indoline‐3,7′‐thiazolo[3,2‐a]pyridines] and spiro[benzo[4,5]thiazolo[3,2‐a]pyridine‐3,3′‐indolines] in good yields. Both cis‐isomers and trans‐isomers were successfully separated out and fully characterized with spectroscopy and single crystal determination. Under similar conditions, the three‐component reaction containing 2‐(1,3‐dioxo‐1H‐inden‐2(3H)‐ylidene)malononitrile resulted in spiro[indene‐2,7′‐thiazolo[3,2‐a]pyridine] derivatives.  相似文献

Previously unknown 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazoline]‐2,2′‐(7′H)‐diones and their N‐substituted analogues were obtained via reaction of 6‐R1‐3‐(2‐aminophenyl)‐1,2,4‐triazin‐5‐ones with isatin and its substituted derivatives. It was shown that alkylation of 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazolin]‐2,2′‐(7′H)‐diones by N‐R3‐chloroacetamides or chloroacetonitrile in the presence of а base proceeds by N‐1 atom of isatin fragment. The spectral properties (1H and 13C NMR spectra) of synthesized compounds were studied, and features of spectral patterns were discussed. The high‐effective anticonvulsant and radical scavenging agents among 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazolin]‐2,2′(7′H)‐diones and their N‐substituted derivatives were detected. It was shown that compounds 2.2 , 2.8 , and 3.1 exceed or compete the activity of the most widely used in modern neurology drug—lamotrigine on the pentylenetetrazole‐induced seizures model. The aforementioned fact may be considered as a reason for further profound study of synthesized compounds using other pathology models.  相似文献

An auto oxidation-rearrangement product 4 was isolated from a high dilution reaction of ninhydrin with 3,4,5-trimethoxyaniline in water. A general synthesis of this compound and its derivatives 4–6 was devised by oxidation of tetrahydroindeno[1,2-b]indol-10-ones 1–3 with sodium periodate to give isoindolo[2,1-a]-indole-6,11-diones 4–6 in good yield. Compounds 4–6 can be easily transformed into spiro[1H-isobenzofuran-1,2′-2H-indole]-3,3′-diones 8–10 , spiro[2H-indole-2,1′-1H-isoindole]-3,3′-diones 11–13 and isoindole[1,2-a:2′,1′-b]pyrimidine-5,15-diones 15, 16 in high yields. Analogous reactions were performed on 3-amino-5a, 10a-dihydroxybenzo[b]indeno[2,1-d]furan-10-one ( 17 ) to give a dibenzoxocintrione 18 , spiro-[benzofuran-2,1′-isobenzofuran]-3,3′-dione 19 and an isoindol-1-one 20 .  相似文献

The spiro[cyclohexane-1,2′-[2H]indene] derivatives 15a , b with molecular dimensions and nucleophilic functional groups similar to known steroid 5α-reductase inhibitors (e.g. 2 ) were synthesized. The spiro[cyclohexane-1,2′-[2H]indene]-1′(3′H),4-dione ( 5 ) was synthesized from 5-methoxyindan-l-one ( = 2,3-dihydro-5-methoxy-1H-inden-1-one). A Grignard reaction and a dehydration step led to the cyclohexene (±)- 7 which, upon a stereoselective hydrogenation catalyzed by Raney-Ni under mild conditions, gave 8a as a pure epimer. Further hydrogenation and hydrogenolysis of 8a over Pd/C at room temperature reduced the keto group to give pure 9a . Finally, the 5′-substituted derivatives 12a , 14a , and 15a were generated by deprotection and Heck-type reaction.  相似文献

A new spiroheterocyclic system spiro[9H-acridine-9,3′-[3H]indol]-2′(1′H)-one and related compounds have been prepared by the reaction of spiro[3H-indole-3,9′-[9H]xanthen]-2(1H)-ones with aromatic amine or ammonium acetate. The latter were prepared by heating fluorinated indole-2,3-diones with m-/p-cresols or α-naphthol in the presence of sulphuric acid at 230-240°. The synthesized compounds have been characterized on the basis of their elemental analyses, ir, nmr (1H, 13C, 19F) and mass spectral data.  相似文献

Diaminomethylene- and aminomethylthiomethylenehydrazones [2] of cyclic ketones 1–8 readily reacted with ethoxymethylenemalononitrile to give spiro[cycloalkane-1,2′-[1,2′,4′]triazolo[1,5′-c]pyrimidine-8′-carbonitrile] derivatives 12–19 through the electrocyclic reaction of the initially formed condensation products 26 in moderate to high yields. The spiro[cyclopentanetriazolopyrimidine] derivatives underwent ring-opening at the cycloalkane moiety upon heating in solution to give 2-alkyl-5-substituted-[1,2,4]triazolo[1,5-c]pyrimidine-8′-carbonitriles 20–23 . When an alkyl substituent was introduced into the cyclopentane ring, cleavage of the spiro compounds occurred preferentially at the cyclopentane moiety between the spiro carbon and the more branched one. In contrast, the cyclohexane ring, especially of spiro-5-amino-triazolopyrimidines 17 and 18 strongly resisted to ring-opening under similar conditions, but those of 5-methylthiotriazolopyrimidines 14 gave up to 17 percent of cleavage after prolonged heating in hot ethanol. 2-t-Butyl-5-methylthio-2,3-dihydro[1,2,4]triazolo[1,5-c]pyrimidine-8-carbonitrile 25 [R3 = C(CH3)3] was highly susceptible to the cleavage even at room temperature and produced the corresponding 2-unsubstituted triazolopyrimidine 24 with loss of the t-butyl group.  相似文献

The chemistry of the 2-aminospiro[indan-1,3′-pyrrolidine]system 2-Aminospir[indan-1,3′-pyrrolidines] 1 are easily synthesized from the α-hydroxyiminoketones 6 and 19 (Schemes 2 and 3, and 6 respectively). Removal of the N-acetyl group in the intermediate 8 of the trans series induces transposition to the 3a-aminomethyl-indano[2,1-b]pyrrolidine system. The configurations of all compounds have been determined by 1H-NMR. spectroscopy.  相似文献

Reaction of spiro[isobenzofuran-1(3H).9′(9H)-6′-(methylcyclohexylamino)-3′-methyl-2′-anilinoxanthene]-3-one ( 1 ), which is typical leuco fluoran dye, with N-bromosuccinimide and N-chlorosuccinimide leads to halogenated derivatives 2a–2c and 3 , respectively. Their structures were established by two-dimensional proton-proton (COSY) experiment and their thermal properties examined by means of DSC and compared with commercially available 1 .  相似文献

Intramolecular cycloaddition of [1,1′-binaphthyl]-2,2′-bis(allylamine) Unlike the 1,1′-binaphthyl-2,2′-bis(allylether) the corresponding [1,1′-binaphthyl]-2,2′-bis(allylamine) (1) upon heating to 230° in mesitylene undergoes thermal decomposition only. However, when 1 is heated in a mixture of 2-methylaminoethanol and water, besides 3 the policyclic ketones 4 and 5 are formed in isolated yields of 28 and 10%, respectively (Scheme 1). Intermediates are the imines corresponding to 4 and 5 which are hydrolysed under the reaction conditions rather than decomposed. The imines are formed by a intramolecular Diels-Alder reaction, in which the double bond of one N-allylgroup reacts with the naphthalene ring of the second half of the molecule. The policyclic ketones 4 and 5 are characterized as acetates 6 and 7 , respectively, and as the acetylated reduced products 11 , and 12 and 13 , respectively. The constitutions of all compounds are derived from spectroscopic data, chiefly from the 1H-NMR. spectra.  相似文献

A three‐component reaction of isatin, 3‐methyl‐1‐phenyl‐1H‐pyrazol‐5‐amine, and piperidine‐2,4‐dione was treated in ionic liquids catalyzed by TsOH and provided an efficient and green method for the synthesis of spiro[indoline‐3,4′‐pyrazolo[3, 4‐b][1,6]naphthyridine]‐2,5′(1′H)‐dione derivatives in high yields.  相似文献

A one‐step synthesis of ethyl 2,3‐dihydronaphtho[1,2‐b]furan‐2‐carboxylate and/or ethyl 4′‐oxospiro[cyclopropane‐1,1′(4′H)‐naphthalene]‐2′‐carboxylate derivatives 2 and 3 , respectively, from substituted naphthalen‐1‐ols and ethyl 2,3‐dibromopropanoate is described (Scheme 1). Compounds 2 were easily aromatized (Scheme 2). In the same way, 3,4‐dibromobutan‐2‐one afforded the corresponding 1‐(2,3‐dihydronaphtho[1,2‐b]furan‐2‐yl)ethanone and/or spiro derivatives 8 and 9 , respectively (Scheme 6). A mechanism for the formation of the dihydronaphtho[1,2‐b]furan ring and of the spiro compounds 3 is proposed (Schemes 3 and 4). The structures of spiro compounds 3a and 3f were established by X‐ray structural analysis. The reactivity of compound 3a was also briefly examined (Scheme 9).  相似文献

A variety of 3″,5″‐diaryl‐3″H,4′H‐dispiro[cyclohexane‐1,2′‐chromene‐3′,2″‐[1,3,4]thiadiazol]‐4′‐ones 3a‐c were synthesized regioselectively through the reaction of 4′H,5H‐trispiro[cyclohexane‐1,2′‐chromene‐3′,2″‐[1,3,4]oxadithiino[5,6‐c]chromene‐5″,1″′‐cyclohexan]‐4′‐one ( 1 ) with nitrilimines (generated in situ via triethylamine dehydrohalogenation of the corresponding hydrazonoyl chlorides 2a‐c ) in refluxing dry toluene. Single crystal X‐ray diffraction studies of 3a,b add support for the established structure. Similarly, 3′,5′‐diaryl‐2,2‐dimethyl‐3′H,4H‐spiro[chromene‐3,2′‐[1,3,4]thiadiazol]‐4‐ones 5a‐c were obtained in a regioselective manner through the reaction of 2,2,5′,5′‐tetramethyl‐4H,5′H‐spiro[chromene‐3,2′‐[1,3,4]oxadithiino[5,6‐c]chromen]‐4‐one ( 4a ) with nitrilimines under similar reaction conditions. On the other hand, reaction of 2,5′‐diethyl‐2,5′‐dimethyl‐4H,5′H‐spiro[chromene‐3,2′‐[1,3,4]oxadithiino‐[5,6‐c]chromen]‐4‐one ( 4b ) with nitrilimines in refluxing dry toluene afforded the corresponding 3′,5′‐diaryl‐2‐ethyl‐2‐methyl‐3′H,4H‐spiro[chromene‐3,2′‐[1,3,4]thiadiazol]‐4‐ones 5d‐f as two unisolable diastereoisomeric forms.  相似文献

Diaminomethylenehydrazones of cyclic ketones 1–5 reacted with ethyl N‐cyanoimidate (I) at room temperature or with bis(methylthio)methylenecyanamide (II) under brief heating to give directly the corresponding spiro[cycloalkane[1′,2′,4′]triazolo[1′,5′,‐a][1′,3′‐5′]triazine] derivatives 7–12 in moderate to high yields. Ring‐opening reaction of the spiro[cycloalkanetriazolotriazine] derivatives occurred at the cycloalkane moiety upon heating in solution to give 2‐alkyl‐5‐amino[1,2,4]triazolotriazines 13–16. Diaminomethylenehydrazones 17–19, of hindered acyclic ketones, gave 2‐methyl‐7‐methylthio[1,2,4]‐triazolo[1,5‐a][1,3,5]triazines 21–23 by the reaction with II as the main products with apparent loss of 2‐methylpropane from the potential precursor, 2‐tert‐butyl‐2‐methyl‐7‐methylthio[1,2,4]triazolo[1,5‐a]‐[1,3,5]triazines 20, in good yields. In general, bis(methylthio)methylenecyanamide II was found to be a favorable reagent to the one‐step synthesis of the spiro[cycloalkanetriazolotriazine] derivatives from the diaminomethylenehydrazones. The spectral data and structural assignments of the fused triazine products are discussed.  相似文献

The synthesis of the novel 2,4,6‐triaryl‐1‐(spiro[2H‐1‐benzopyran‐2,2′‐indoline]‐6‐yl)pyridiniumper‐chlorates 4 by reaction of 5 ‐nitrosalicylaldehydes 6 with 1,3,3‐trimethyl‐2‐methyleneindoline ( 7 ) to 6‐nitro‐spiro[2H‐1‐benzopyran‐2,2′‐indolines] 1 , their stannous chloride reduction to the 6‐amino derivatives 8 , followed by a 2,6‐[C5+N] ring transformation with 2,4,6‐triarylpyrylium perchlorates 9 , is reported. UV irradiation experiments in twenty solvents of different polarity prove their photochromic properties and show that the photochemically generated negative solvatochromic dyes 5 , formed by ring opening of the benzopyran moiety of 4 , are rather merocyanine than pyridinium phenolate betaine dyes.  相似文献

Derivatives of two new molecular structures, namely, 7,8-dihydro-6H,10H-[1,2,5]thiadiazolo[3′,4′:4,5]pyrimido[2,1-b][1,3]thiazin-10-one and 6,7-dihydro-9H-thiazolo[3,2-a][1,2,5]thiadiazolo[3,4-d][pyrimidin-9-one, and derivatives of N-substituted sulfamic acid, namely, (8-amino-3,4-dihydro-2H,6H-pyrimido[2,1-b][1,3]thiazin-6-on-7-yl)sulfamic acid and (7-amino-2,3-dihydro-5H-thiazolo[3,2-a]pyrimidin-5-on-6-yl)sulfamic acid, were separated out as by-products in the reduction reaction of 8-amino-3,4-dihydro-7-nitroso-2H,6H-pyrimido[2,1- b][1,3]thiazin-6-one and 7-amino-2,3-dihydro-6-nitroso-5H-thiazolo[3,2-a]pyrimidin-5-one derivatives, respectively, with sodium hydrosulfite. A mechanism of reaction, which hypothesizes the action of sodium hydrosulfite in an asymmetic form, is proposed. The results of single-crystal X-ray investigation on 7,8-dihydro-6H,10H-[1,2,5]thiadiazolo[3′,4′:4,5]pyrimido[2,1-b][1,3]thiazin-10-one (R = 0.032 for 863 reflections) and (8-amino-3,4-dihydro-2H,6H-pyrimido[2,1-b]- [1,3]thiazin-6-on-7-yl)sulfamic acid, sodium salt (R = 0.028 for 3507 reflections) are reported.  相似文献

Alkylation of cycloalkano[b]indolylmagnesium iodide with alkyl halides has been analyzed to prepare cycloalkano-3H-indoles, methyl and N,N-dimethylpropyl derivatives. Spiro[cycloalkane-1,2′-indolin-3′-one] derivatives appear as secondary products in this reaction. Selectivity and optimization of the reaction has been determined and a mechanism for the formation of the spiro derivative is proposed.  相似文献

A number of spiro[indoline-3,2′-thiazolidine]-2,4′-diones have been prepared via the cyclocondensation of isatin-3-imines with α-mercaptoacids. 1-Benzyl-5′,5′-dimethylspiro[indoline-3,2′-thiazolidine]-2,4′-diones were prepared by simultaneously refluxing 1-benzylisatin, α-mercaptoisobutyric acid and various anilines in toluene. 3′-(4-Chlorophenyl)-5,5′-dimethylspiro[indoline-3,2′-thiazolidine]-2,4′-dione was active in the P388 and the L1210 leukemia screen tests. A number of analogs of the active spiro compound have been prepared and submitted for antileukemic screening. Anticonvulsant screening results of related compounds are also included.  相似文献

2,2′-Bi-1H-imidazole, when protected with the [2-(trimethylsilyl)ethoxy]methyl (SEM) blocking group, on treatment with N-bromosuccinimide or N-chlorosuccinimide yields predominantly the monohalogenated derivatives 4a and 4b. 2-Amino-6,7-dihydrothieno[3′,2′:5,6]pyrido[2,3-rf]pyrimidin-4-one ( 1 ) was prepared in three steps from S-(3-butynyl)thiosemicarbazide hydroiodide ( 3 ) and diethyl ketomalonate. The featured step in this synthetic sequence was an intramolecular Diels-Alder reaction of the in situ generated 3-(3-butynylthio)-6-carboethoxy-5-chloro-1,2,4-triazine ( 9 ) to provide the key intermediate 5-carboethoxy-6-chloro-2,3-dihydrothieno-[2,3-b]pyridine ( 6 ). In the course of studies directed toward the preparation of 1 , thermolysis of 3-(3-butynyl-thio)-6-carboethoxy-1,2,4-triazin-5(2H)-one ( 2 ) was found to involve competitive intramolecular Diels-Alder and intramolecular coplanar cycloamination processes, providing the 2,3-dihydrothieno[2,3-b]pyridin-6(7H)-one ( 4 ) and the 1,3-thiazino[3,2-b]-1,2,4-triazin-3-one (5) derivatives, respectively.  相似文献
