Understanding biological membranes at physiological conditions requires comprehension of the interaction of lipid bilayers with sodium and potassium ions. These cations are adsorbed at palmitoyl-oleoyl-phosphatidylcholine (POPC) bilayers as indicated from previous studies. Here we compare the affinity of Na(+) and K(+) for POPC in molecular dynamics (MD) simulations with recent data from electrophoresis experiments and isothermal calorimetry (ITC) at neutral pH. NaCl and KCl were described using GROMOS or parameters matching solution activities on the basis of Kirkwood-Buff theory (KBFF), and K(+) was also described using parameters by Dang et al., all in conjunction with the Berger parameters for the lipids and the SPC water model. Apparent binding constants of GROMOS-Na(+) and KBFF-K(+) are the same within error and in good agreement with values from ITC. Although these force fields yield the same number of bound ions per number of lipids for Na(+) and K(+), they give a larger number of Na(+) ions per surface area compared to K(+), in agreement with the electrophoresis experiments, because Na(+) causes a stronger reduction in the area per lipid than K(+). The intrinsic binding constants, on the other hand, are reproduced by Dang-K(+) but overestimated by GROMOS-Na(+) and KBFF-K(+). That no ion force field reproduces the intrinsic and the apparent binding constant simultaneously arises from the fact that in MD simulations, implicitly meant to mimic neutral pH, pure PC is usually modeled with zero surface charge. In contrast, POPC at neutral conditions in experiment carries a low but significant negative surface charge and is uncharged only at acidic pH as indicated from electrophoretic mobilities. Implications for future simulation and experimental studies are discussed.  相似文献

In pure water, zwitterionic lipids form lamellar phases with an equilibrium water gap on the order of 2 to 3 nm as a result of the dominating van der Waals attraction between dipolar bilayers. Monovalent ions can swell those neutral lamellae by a small amount. Divalent ions can adsorb onto dipolar membranes and charge them. Using solution X-ray scattering, we studied how the structure of ions and zwitterionic lipids regulates the charge of dipolar membranes. We found that unlike monovalent ions that weakly interact with all of the examined dipolar membranes, divalent and trivalent ions adsorb onto membranes containing lipids with saturated tails, with an association constant on the order of ~10 M(-1). One double bond in the lipid tail is sufficient to prevent divalent ion adsorption. We suggest that this behavior is due to the relatively loose packing of lipids with unsaturated tails that increases the area per lipid headgroup, enabling their free rotation. Divalent ion adsorption links two lipids and limits their free rotation. The ion-dipole interaction gained by the adsorption of the ions onto unsaturated membranes is insufficient to compensate for the loss of headgroup free-rotational entropy. The ion-dipole interaction is stronger for cations with a higher valence. Nevertheless, polyamines behave as monovalent ions near dipolar interfaces in the sense that they interact weakly with the membrane surface, whereas in the bulk their behavior is similar to that of multivalent cations. Advanced data analysis and comparison with theory provide insight into the structure and interactions between ion-induced regulated charged interfaces. This study models biologically relevant interactions between cell membranes and various ions and the manner in which the lipid structure governs those interactions. The ability to monitor these interactions creates a tool for probing systems that are more complex and forms the basis for controlling the interactions between dipolar membranes and charged proteins or biopolymers for encapsulation and delivery applications.  相似文献

It is well-established that native plasma membranes are characterized by an asymmetric distribution of charged (anionic) lipids across the membrane. To clarify how the asymmetry can affect membrane electrostatics, we have performed extensive atomic-scale molecular dynamics simulations of asymmetric lipid membranes composed of zwitterionic (phosphatidylcholine (PC) or phosphatidylethanolamine (PE)) and anionic (phosphatidylserine (PS)) leaflets. It turns out that the asymmetry in transmembrane distribution of anionic lipids gives rise to a nonzero potential difference between the two sides of the membrane. This potential arises from the difference in surface charges of the two leaflets. The magnitude of the intrinsic membrane potential was found to be 238 mV and 198 mV for PS/PC and PS/PE membranes, respectively. Remarkably, this potential is of the same sign as the membrane potential in cells. Our findings, being in reasonable agreement with available experimental data, lend support to the idea that the transmembrane lipid asymmetry typical of most living cells contributes to the membrane potential.  相似文献

Though glycolipids are involved in a multitude of cellular functions, the understanding of their atom-scale properties in lipid membranes has remained very limited due to the lack of atomistic simulations. In this work, we employ extensive simulations to characterize one-component membranes comprised of glycoglycerolipids, focusing on two common glyco head groups, namely glucose and galactose. The properties of these two glycoglycerolipid bilayers are compared in a systematic manner with membranes consisting of phosphatidylcholine (PC) or phosphatidylethanolamine (PE) lipids, whose structures aside from the head group are identical with those of the two glycolipids. We find that the glycolipid systems are characterized by a substantial number of hydrogen bonds in the head group region, leading to membrane packing that is stronger than in a PC but less significant than that in a PE bilayer. The role played by the glyco head group is especially evident in the electrostatic membrane potential, which is particularly large in the glycolipid membranes. For the same reason, the interfacial forces near glycolipid bilayers are significantly different from those found in PC and PE bilayers, affecting, e.g., the ordering of water close to the membrane. These effects are particularly important for the case of galactose, an important component in thylacoids.  相似文献

By means of contact angle measurements with water and aqueous salt solutions, it is shown that plurivalent cations increase the hydrophobicity of negatively charged phospholipid vesicle membranes (consisting of phosphatidic acid, PA, or of phosphatidylserine, PS), but does not influence the hydrophobicity of neutral phospholipid membranes, (e.g., phosphatidylcholine, PC, at up to 200 mM of CaCl2). The hydrophobizing action of cations on PA and PS membranes is concomitant with the reduction in (negative) zeta potential with increasing cation concentrations. Trivalent cations, La3+, showed more effective in hydrophobizing negatively charged phospholipid membranes than divalent and monovalent cations. Except for hydrogen ions, monovalent cations do not show any appreciable hydrophobizing effect on lipid vesicle membranes at concentrations less than 1 M. The hydrophobizing effect on phospholipid membranes can also be used to explain the induction of lateral phase separation into patches of different phospholipids as well as cell fusion.  相似文献

Interactions between salt ions and lipid components of biological membranes are essential for the structure, stability, and functions of the membranes. The specific ionic composition of aqueous buffers inside and outside of the cell is known to differ considerably. To model such a situation we perform atomistic molecular-dynamics (MD) simulations of a single-component phosphatidylcholine lipid bilayer which separates two aqueous reservoirs with and without NaCl salt. To implement the difference in electrolyte composition near two membrane sides, a double bilayer setup (i.e., two bilayers in a simulation box) is employed. It turns out that monovalent salt, being in contact with one leaflet only, induces a pronounced asymmetry in the structural, electrostatic, and dynamical properties of bilayer leaflets after 50 ns of MD simulations. Binding of sodium ions to the carbonyl region of the leaflet which is in contact with salt results in the formation of "Na-lipids" complexes and, correspondingly, reduces mobility of lipids of this leaflet. In turn, attractive interactions of chloride ions (mainly located in the aqueous phase close to the water-lipid interface) with choline lipid groups lead to a substantial (more vertical) reorientation of postphatidylcholine headgroups of the leaflet adjoined to salt. The difference in headgroup orientation on two sides of a bilayer, being coupled with salt-induced reorientation of water dipoles, leads to a notable asymmetry in the charge-density profiles and electrostatic potentials of bilayer constitutes of the two leaflets. Although the overall charge density of the bilayer is found to be almost insensitive to the presence of salt, a slight asymmetry in the charge distribution between the two bilayer leaflets results in a nonzero potential difference of about 85 mV between the two water phases. Thus, a transmembrane potential of the order of the membrane potential in a cell can arise without ionic charge imbalance between two aqueous compartments.  相似文献

In order to elucidate the role of structural change of lipid membrane bilayer in the mode of action of local anesthetic, we studied the effects of local anesthetics, charged tetracaine and uncharged benzocaine, on ion permeability across various lipid planar bilayers (PC, mixed PC/PS (4/1, mol/mol); mixed PC/PE (1/1, mol/mol); mixed PC/SM (4/1, mol/mol)) under a constant applied voltage. The membrane conductances increased in the order of PC  PC/PS ≤ PC/SM  PC/PE. When the constant voltage of −100 or −70 mV was applied through the lipid bilayer membranes in the presence of positively charged tetracaine, the fluctuating current pulses with the large amplitude generated, but not appeared in the absence of tetracaine. The addition of uncharged benzocaine generated the fluctuating currents with the small amplitude. Both charged tetracaine and uncharged benzocaine facilitated electrophoretically the transport of small ions such as KCl in the buffer solution through the fluctuating pores in the lipid bilayer membranes formed by interaction with the local anesthetic under the negative applied membrane potential. The current pulses also contained actual transport of charged tetracaine together with the transport of the small ions. The amplitude and the duration time of the electrical current generated by adding the local anesthetics were dependent on the type of the lipid, the applied voltage and its voltage polarity.  相似文献

Simultaneous occupancy of the two binding sites in gramicidin A by monovalent cations is a well known property of this channel, but the energetic feasibility of this process in molecular dynamics simulations has not been established so far. Here the authors study the energetics of single and double ion occupancy in gramicidin A by constructing the potential of mean force for single and pair of cations. As representatives of small and large ions, they consider both Na+ and K+ ions in the calculations. Binding constants of ions are estimated from the free energy profiles. Comparisons with the experimental results indicate 3-4 kT discrepancy in the binding energies. They also study the coordination of the ions in their respective binding sites and the dynamic behavior of the channel water during the double ion binding process.  相似文献

以3-异氰酸丙基三乙氧基硅烷和对甲氧基苯胺为原料合成了一种可以自组装形成有机-无机杂化材料的化合物--3-(脲基-4-甲氧基苯基)丙基三乙氧基硅烷. 采用FT-IR, 1H NMR, DSC 和XRD 分析方法对该化合物的结构以及结晶性进行了表征. 将该化合物与聚乙烯醇(PVA)共混, 利用化合物的自组装性质构筑结构均一且致密无孔的离子通道杂化膜, 通过自制的膜运输实验装置测定膜对阳离子的传输性能并提出了相应的传输机制. SEM 照片显示, 自组装杂化膜致密无缺陷, 膜厚度为8 μm. 选择5 种阳离子进行运输实验测试, 结果表明, 自组装杂化离子通道膜对一价的碱金属离子Li+, Na+和K+有很好的传输功能, 这要归功于杂化材料中甲氧基苯基与碱金属阳离子形成的阳离子-π相互作用力. 碱金属阳离子在膜中的扩散过程可由溶解-扩散机制来解释, 结果显示, Li+, Na+和K+在杂化膜中传输的渗透率大小为: PNa+ > PK+ > PLi+ , 说明本研究中的的自组装杂化离子通道膜对Na+有优先选择性. 杂化离子通道膜对二价的Ca2+和Mg2+没有传输作用, 此结果给一二价阳离子的分离带来很好的研究思路.  相似文献

C-reactive protein (CRP) is an acute phase reactive protein, which has been shown to specifically bind to phosphorylcholine (PC) and phosphorylethanoamine (PE) moieties in the presence of calcium. In order to investigate the effect of steric hindrance on the specific binding of CRP to membranes, we designed and synthesized six phospholipids, each containing a long-arm spacer of 3, 6 or 8 atoms between the head group and hydrophobic tail. By mixing synthesized lipids and natural lipids the ligand-containing monolayers were prepared, which have PC or PE groups protruding out of the membrane surface. To characterize of the synthesized phospholipids, the thickness of the lipid monolayers was measured by surface plasmon resonance (SPR) technique, the phase behavior of the lipid monolayer at air/water interface was studied by pressure–area analysis, and the specific binding of rabbit C-reactive protein to the synthesized lipid containing membranes was studied by imaging ellipsometry.  相似文献

Entities such as ion distributions and forces between lipid membranes depend on effects due to the intervening salt solution that have not been recognized previously. These specific ion or Hofmeister effects influence membrane fusion. A typical illustrative example is this: measurements of forces between double-chained cationic bilayers adsorbed onto molecularly smooth mica surfaces across different 0.6-2 mM salt solutions have revealed a large degree of ion specificity [Pashley et al. J. Phys. Chem. 1986, 90, 1637]. This has been interpreted in terms of very specific anion "binding" to the adsorbed bilayers, as it would too for micelles and other self-assembled systems. However, we show here that inclusion of nonelectrostatic (NES) or ionic dispersion potentials acting between ions and the two surfaces explains such "ion binding". The observed Hofmeister sequence for the calculated pressure without any direct ion binding is given correctly. This demonstrates the importance of a source of ion specificity that has been ignored. It is due to ionic physisorption caused by attractive NES ionic dispersion potentials. There appear to be some far reaching consequences for interpretations of membrane intermolecular interactions in salt solutions.  相似文献

In the first paper (J. Phys. Chem. B, 2006, 110, 10878), effective ion-ion potentials in SPC/E water were obtained for Me-Me, Me-Cl-, and Cl(-)-Cl- pairs, where Me is Li+, Na+, K+, Mg2+, Ca2+, Sr2+, and Ba2+ cations. In this second part of the study of effective interionic potentials, ion-ion distribution functions obtained from implicit-water Monte Carlo simulations of electrolyte solution with these potentials have been explored. This analysis verifies the range of applicability of the primitive model of electrolyte. It is shown that this approximation can be applied to monovalent electrolyte solutions in a wide range of concentrations, whereas the nature of ion-ion interactions is notably different for 2:1 electrolytes. An improved model of ions is discussed. The model includes approximations of the ion hydration shell polarization and specific short-range ion-ion interaction. It allows approximation of the potential of mean force acting on ions in strong electric fields of highly charged macromolecules and bilayers.  相似文献

To study the change of interlayer structure of a Wyoming-type Na-montmorillonite as a result of the replacement of interlayer Na+ ions by cetylpyridinium (CP+) ions, a series of NPT Monte Carlo simulations of the clay mineral with different contents of CP+, Na+, Cl- ions and water in its interlayer space is carried out. In agreement with conclusions from experimental studies, the simulations show that the CP+ ions form monomolecular, bimolecular, and pseudotrimolecular layers with increasing interlayer contents. Calculated potential energies reveal that clay-organic interactions are stronger than organic-organic interactions in CP+-modified montmorillonite, which is in conformity with observations of earlier thermogravimetric experiments. The simulation results indicate that the pseudotrimolecular arrangement of CP+ ions is a prerequisite for the experimentally observed interlayer sorption of inorganic anions. Furthermore, in the interlayer space with a pseudotrimolecular layer, chloride ions favor the formation of pairs with inorganic rather than organic cations. On the basis of these findings and available experimental data, we propose that the interlayer sorption of inorganic anions from the pore space of an organically modified montmorillonite may occur not only in pairs with organic cations, as suggested earlier, but also in pairs with inorganic cations, which represents a so-far unconsidered and maybe more important mechanism of anion sorption on clay minerals.  相似文献

We report a solid-state (23)Na NMR study of monovalent cation (Li(+), Na(+), K(+), Rb(+), Cs(+) and NH(4) (+)) binding to double-stranded calf thymus DNA (CT DNA) at low relative humidity, ca 0-10%. Results from (23)Na--(31)P rotational echo double resonance (REDOR) NMR experiments firmly establish that, at low relative humidity, monovalent cations are directly bound to the phosphate group of CT DNA and are partially dehydrated. On the basis of solid-state (23)Na NMR titration experiments, we obtain quantitative thermodynamic parameters concerning the cation-binding affinity for the phosphate group of CT DNA. The free energy difference (DeltaG degrees ) between M(+) and Na(+) ions is as follows: Li(+) (-1.0 kcal mol(-1)), K(+) (7.2 kcal mol(-1)), NH(4) (+) (1.0 kcal mol(-1)), Rb(+) (4.5 kcal mol(-1)) and Cs(+) (1.5 kcal mol(-1)). These results suggest that, at low relative humidity, the binding affinity of monovalent cations for the phosphate group of CT DNA follows the order: Li(+) > Na(+) > NH(4) (+) > Cs(+) > Rb(+) > K(+). This sequence is drastically different from that observed for CT DNA in solution. This discrepancy is attributed to the different modes of cation binding in dry and wet states of DNA. In the wet state of DNA, cations are fully hydrated. Our results suggest that the free energy balance between direct cation-phosphate contact and dehydration interactions is important. The reported experimental results on relative ion-binding affinity for the DNA backbone may be used for testing theoretical treatment of cation-phosphate interactions in DNA.  相似文献

Fluidity and charging of supported bilayer lipid membranes (sBLMs) prepared from 1,2-dioleoyl-sn-glycero-3-phosphatidylcholine (DOPC) were studied by fluorescence recovery after photobleaching (FRAP) and microslit electrokinetic measurements at varying pH and ionic composition of the electrolyte. Measurements in neutral electrolytes (KCl, NaCl) revealed a strong correlation between the membrane fluidity and the membrane charging due to unsymmetrical water ion adsorption (OH(-) ? H(3)O(+)). The membrane fluidity significantly decreased below the isoelectric point of 3.9, suggesting a phase transition in the bilayer. The interactions of both chaotropic anions and strongly kosmotropic cations with the zwitterionic lipids were found to be related with nearly unhindered lipid mobility in the acidic pH range. While for the chaotropic anions the observed effect correlates with the increased negative net charge at low pH, no correlation was found between the changes in the membrane fluidity and charge in the presence of kosmotropic cations. We discuss the observed phenomena with respect to the interaction of the electrolyte ions with the lipid headgroup and the influence of this process on the headgroup orientation and hydration as well as on the lipid packaging.  相似文献

In an effort to use model fluid membranes for immunological studies, we compared the formation of planar phospholipid bilayers supported on silicon dioxide surfaces with and without incorporation of glycolipids as the antigen for in situ antibody binding. Dynamic light scattering measurements did not differentiate the hydrodynamic volumes of extruded small unilamellar vesicles (E-SUVs) containing physiologically relevant concentrations (0.5-5 mol%) of monosialoganglioside GM1 (GM1) from exclusive egg yolk L-alpha-phosphatidylcholine (egg PC) E-SUVs. However, quantifiable differences in deposition mass and dissipative energy loss emerged in the transformation of 5 mol% GM1/95 mol% egg PC E-SUVs to planar supported lipid bilayers (PSLBs) by vesicle fusion on thermally evaporated SiO2, as monitored by the quartz crystal microbalance with dissipation (QCM-D) technique. Compared to the 100 mol% egg PC bilayers on the same surface, E-SUVs containing 5 mol% GM1 reached a approximately 12% higher mass and a lower dissipative energy loss during bilayer transformation. PSLBs with 5 mol% GM1 are approximately 18% heavier than 100 mol% egg PC and approximately 11% smaller in projected area per lipid, indicating an increased rigidity and a tighter packing. Subsequent binding of polyclonal immunoglobulin G anti-GM1 to the PSLBs was performed in situ and showed specificity. The anti-GM1 to GM1 ratios at equilibrium were roughly proportional to the concentrations of anti-GM1 administered in the solution. Fluorescence recovery after photobleaching was utilized to verify the retained, albeit reduced lateral fluidity of the supported membranes. Five moles percentage of GM1 membranes (GM1 to PC ratio approximately 1:19) decorated with 1 mol% N-(Texas Red sulfonyl)-1,2-dihexadecanoyl-sn-glycerol-3-phosphoethanolamine (Texas Red DHPE) exhibited an approximately 16% lower diffusion coefficient of 1.32+/-0.06 microm2/s, compared to 1.58+/-0.04 microm2/s for egg PC membranes without GM1 (p<0.01). The changes in vesicle properties and membrane lateral fluidity are attributed to the interactions of GM1 with itself and GM1 with other membrane lipids. This system allows for molecules of interest such as GM1 to exist on a more biologically relevant surface than those used in conventional methods such as ELISA. Our analysis of rabbit serum antibodies binding to GM1 demonstrates this platform can be used to test for the presence of anti-lipid antibodies in serum.  相似文献

The present paper deals with the electrokinetic characterization of sepiolite. A series of systematic zeta potential measurements have been carried out to determine the isoelectric point (iep) and potential-determining ions (pdi), and the effect of mono-, di-, and trivalent electrolytes such as NaCl, KCl, LiCl, NaNO(3), NaCH(3)COO, MgCl(2), CaCl(2), BaCl(2), CoCl(2), CuCl(2), Pb(NO(3))(2), Na(2)CO(3), Na(2)SO(4), AlCl(3), FeCl(3), and Na(3)PO(4) on the zeta potential of sepiolite. Zeta potential has been calculated with the aid of Smoluchowski's equation. Sepiolite yields an isoelectric point at pH 6.6. The zeta potential for the sepiolite has ranged from +23.3 mV at pH approximately 2 to -22.4 mV at pH approximately 8 at 20 +/- 2 degrees C in water. The valency of the ions have proven to have a great influence on the electrokinetic behavior of the suspension. Monovalent cations were found to have a weak effect, while di- and trivalent cations made the zeta potential positive. Charge reversal was observed for divalent cations at 1 x 10(-2) M and for trivalent cations at 3 x 10(-4) M. As a result, it can be said that monovalent cations are indifferent ions when di- and trivalent cations are potential-determining ions.  相似文献

Host defense peptides (HDPs), part of the innate immune system, selectively target the membranes of bacterial cells over that of host cells. As a result, their antimicrobial properties have been under intense study. Their selectivity strongly depends on the chemical and mostly structural properties of the lipids that make up different cell membranes. The ability to synthesize HDP mimics has recently been demonstrated. To better understand how these HDP mimics interact with bilayer membranes, three homologous antimicrobial oligomers (AMOs) 1-3 with an m-phenylene ethynylene backbone and alkyl amine side chains were studied. Among them, AMO 1 is nonactive, AMO 2 is specifically active, and AMO 3 is nonspecifically active against bacteria over human red blood cells, a standard model for mammalian cells. The interactions of these three AMOs with liposomes having different lipid compositions are characterized in detail using a fluorescent dye leakage assay. AMO 2 and AMO 3 caused more leakage than AMO 1 from bacteria membrane mimic liposomes composed of PE/PG lipids. The use of E. coli lipid vesicles gave the same results. Further changes of the lipid compositions revealed that AMO 2 has selectively higher affinity toward PE/PG and E. coli lipids than PC, PC/PG or PC/PS lipids, the major components of mammalian cell membranes. In contrast, AMO 3 is devoid of this lipid selectivity and interacts with all liposomes with equal ease; AMO 1 remains inactive. These observations suggest that lipid type and structure are more important in determining membrane selectivity than lipid headgroup charges for this series of HDP mimics.  相似文献
