Derivatization with N-methyl-N-(tert-butyldimethylsilyl)trifluoroacetamide (MTBSTFA) was used for gas chromatography–mass spectrometry (GC–MS) analysis of citrulline and ornithine. Aqueous 50 μl aliquots at 1 and 10 mM concentrations were dried and derivatized separately, and 70 eV electron ionization or CH4 positive chemical ionization were used. Ornithine produced a single GC peak. Physiological citrulline concentrations produced GC artifact peaks for the ornithine derivative, and a compound consistent with elimination of a water molecule from the tri-tert-butyldimethylsilyl (TBDMS) citrulline derivative. A third GC peak obtained using 10 mM citrulline concentrations gave a mass spectrum consistent with a mixture of true tri- and tetra-TBDMS citrulline. Analyses of 13C-ureido-labeled citrulline confirmed the presence of the true TBDMS citrulline derivatives produced from 10 mM samples and provided evidence that the TBDMS ornithine artifact results from loss of TBDMS isocyanate from tetra-TBDMS citrulline. Linear-programmed temperature GC retention index data relative to n-alkanes are reported for observed GC peaks.  相似文献

Optical biosensor arrays for rapidly determining the glucose concentrations in a large number of beverage and blood samples were developed by immobilizing glucose oxidase (GOD) on oxygen sensor layer. Glucose oxidase was first encapsulated in silica based gels through sol-gel approach and then immobilized on 96-well microarrays integrated with oxygen sensing film at the bottom. The oxygen sensing film was made of an organically modified silica film (ORMOSIL) doped with tris(4,7-diphenyl-1,10-phenanthroline) ruthenium dichloride (Ru(dpp)3Cl2). The oxidation reaction of glucose by glucose oxidase could be monitored through fluorescence intensity enhancement due to the oxygen consumption in the reaction. The luminescence changing rate evaluated by the dynamic transient method (DTM) was correlated with the glucose concentration with the wide linear range from 0.1 to 5.0 mM (Y = 13.28X − 0.128, R = 0.9968) and low detection limit (0.06 mM). The effects of pH and coexisting ions were systemically studied. The results showed that the optical biosensor arrays worked under a wide range of pH value, and normal interfering species such as Na+, K+, Cl, PO43−, and ascorbic acid did not cause apparent interference on the measurement. The activity of glucose oxidase was mostly retained even after 2-month storage, indicating their long-term stability.  相似文献

A new automated method for the determination of glucose in honey is proposed. The method is based on multicommutated flow analysis (MCFA) and employs an immobilized glucose oxidase reactor and spectrophotometric detection at 505 nm of the red quinoneimine formed (Trinder's method).The calibration curve obeyed a second order equation in the range 0-0.14 g L−1 (h = −2.2199 C2 + 1.3741C + 0.0077, r2 = 0.9991, where h is the peak height (absorbance) and C the concentration in g L−1). The method was validated analyzing eight commercial samples, both by the AOAC 954.11 and 977.20 official methods. According to Student's t-test of mean values, at the confidence level of 95% the results obtained with the proposed method were in agreement with those obtained by the official methods. Precision (sr(%), n = 10) was 3% and the sampling frequency of the system was 20 samples h−1.  相似文献

The determination of glutamate pyruvate activity (GPT) is of particular clinical importance. A portable GPT sensor for both diagnostic and home-care purpose is highly expected. A highly sensitive and stable l-glutamate sensor was fabricated for the rapid detection of the GPT activity in serum. The sensor is composed of immobilized l-glutamate oxidase in a photo-crosslinkable polymer (PVA-SbQ) membrane on a palladium-deposited screen-printed carbon electrode. The sensor exhibited high sensitivity (detection limit of 50 nM for monosodium glutamate), remarkable long-term stability in storage (5 months in the dry dark state and 1 month in buffer solution) and good reproducibility (R.S.D.=2.6%, n=100). The electrode-to-electrode reproducibility was found to depend on the composition of the polymeric matrix. The optimal substrate composition for the detection of GPT activity was 1 mM α-ketoglutarate and 100 mM l-alanine. The GPT activity in serum can be determined within 3 min. The response of the sensor to GPT activity is linear over the range of 8-250 U/l. Good correlation between the sensor and the Sigma GPT assay kit was achieved (R2=0.9958). The sensor is potentially applicable to a home-care purpose when a portable measuring device is adapted.  相似文献

In this study, a flow injection analysis (FIA) system using a cartridge of immobilized isocitrate lyase (ICL) and isocitrate dehydrogenase (ICDH) was developed to monitor the concentrations of succinic acid in biotechnological processes. The ICL and ICDH immobilized on VA-Epoxy Biosynth E3-carrier had a good operational lifetime (up to 24 h) and storage stability (up to 30 days). The FIA system with the immobilized ICL/ICDH cartridge was characterized with respect to the factors affecting the activity of the immobilized enzymes, such as pH of carrier solution, temperature, sample matrix, etc. Optimal pH value of the immobilized enzymes was slightly shifted in the alkaline range, i.e. 9.0. Some components such as 10 g l−1 lactose, 3 g l−1 malate and 3 g l−1 oxaloacetate in sample solution had significant activating effects (more than 10%) on the response of the FIA system. But the activity of the immobilized ICL and ICDH was not largely influenced by some components like imidazole (1 mM), sodium azide (10 mM) and semicarbazide (2 g l−1) added to carrier buffer solution. The FIA system with an enzyme cartridge was applied to on-line monitor the concentrations of succinic acid in a continuously stirred reactor and a fermentation process of immobilized Escherichia coli, and showed good sensitivity and reliability of the FIA system developed in this work.  相似文献

A flow-injection system for the quantification of pyruvate based on the coupled reactions of pyruvate decarboxylase (PDC) and aldehyde dehydrogenase (AlDH) was conceived and optimized. A co-immobilized PDC and AlDH reactor was introduced into the flow line. Sample and reagent (NAD+) were injected into the flow line by an open sandwich method and the increase of NADH produced by the immobilized-enzyme reactor was monitored fluorometrically at 455 nm (excitation at 340 nm). Linear relationships between the responses and concentrations of pyruvate were observed in the ranges of 2.0 × 10−5 to 1.5 × 10−3 M at the flow rate of 1.0 ml min−1 and 5.0 × 10−6 to 1.0 × 10−3 M at the flow rate of 0.5 ml min−1. The relative standard deviation for 10 successive injections was 0.95% at the 1.0 mM level. This FIA system for pyruvate was applied to the measurement of acetate, citrate and l-lactate.  相似文献

We designed and prepared a micro biosensing system consisting of a flow through system with a sub-micro liter injection valve and a sub-micro liter volume bioreactor. An electrochemical detector was combined with the reactor for immediate detections. The volumes of the reactor and the sample loop for the injection were 850 nl and 320 nl, respectively. This paper described about the characteristics of the sensing system in the case of histamine detection for food analysis. Histamine oxidase from KAIT-B-007 was prepared by using a gene recombination technique and they were immobilized with chitosan beads (? = 70-105 μm). The detection less than one minute after injection made possible fast analysis for histamine. The biosensing system also showed a high performance for histamine detection in wide range of 1 μM-1 mM. In addition, we practically measured histamine content in raw tuna stored at room temperature and 35 °C up to 96 h. As a result of the comparison between our sensing system and HPLC method, there was good agreement. These results show that our microfluidic biosensing system has the potential to assist miniaturization with small sample volume and short determination time for a sequential food analysis.  相似文献

A sequential injection analysis (SIA) system was developed to monitor the concentration of l-cysteine in biological processes on-line. It is based on the redox reaction of l-cysteine with iron(III) in the presence of 1,10-phenanthroline (phen) and the detection of the red-iron(II)-phen complex with a spectrophotometry. The system was fully automated using software written in the LabVIEW™ development environment. A number of system variables such as the flow rate of the carrier buffer solution, the volume ratio of the sample to the reagents, and the reaction coil length, etc., were evaluated to increase the sensitivity and performance of the SIA system. Under partially optimized operating conditions the performance of the SIA system was linear up to a concentration of l-cysteine of 1 mM (R2 = 0.998) with a detection limit of 0.005 mM and a sample frequency of 15 hr−1. The SIA system was employed to monitor the concentration of l-cysteine on-line in a continuously stirred reactor and a fermentation process of Saccharomyces cerevisiae. The on-line monitored data were in good agreement with the off-line data measured by a HPLC with a fluorescence detector (n = 15, R2 = 09899).  相似文献

A novel enzyme reactor with co-immobilization of β-galactosidase and glucose oxidase in calcium alginate fiber (CAF) and amine modified nanosized mesoporous silica (AMNMS) was prepared which incorporate the adsorption and catalysis of AMNMS with the cage effect of the polymer to increase catalytic activity and stability of immobilized enzyme. The enzyme reactor was applied to prepare a chemiluminescence (CL) flow-through biosensor for determination of lactose combined with a novel luminol-diperiodatonickelate (DPN) CL system we reported. It shows that the CL flow-through biosensor possesses long lifetime, high stability, high catalytic activity and sensitivity. The relative CL intensity was linear with the lactose concentration in the range of 8 × 10−8-4 × 10−6 g mL−1 with the detection limit of 2.7 × 10−8 g mL−1 (3σ). It has been successfully applied to the determination of lactose in milk.  相似文献

A novel method for the determination of ethanol in tequila based on the immobilized enzyme fluorescence capillary analysis (IE-EFCA) has been proposed. Alcohol dehydrogenase (ADH) was immobilized in inner surface of a capillary and an immobilized enzyme capillary bioreactor (IE-ECBR) was formed. After nicotinamide adenine dinucleotide (NAD+) as an oxidizer is mixed with alcohol sample solution, it was sucked into the IE-ECBR. The fluorescence intensity of the mixed solution in the IE-ECBR was detected at λex = 350 nm and λem = 459 nm. The experimental conditions are as follows: The reaction time is 20 min; temperature is 40 °C; the concentrations of phosphate buffer solution (pH 7.5) and NAD+ are 0.1 mol L−1 and 5 mmol L−1, respectively; immobilization concentration of ADH is 10 U L−1. The determination range of ethanol is 2.0-15.0 g L−1 (F = 10.44C + 6.6002, r > 0.9958); its detection limit is 1.11 g L−1; and relative standard deviation is 1.9%. IE-EFCA method is applicable for the determination of the samples containing alcohol in medicine, industry and environment.  相似文献

A spectrometric method was investigated to measure the activities of recombinant human cyclic nucleotide phosphodiesterase 4 (PDE4), based on the use of malachite green (MLG) to quantify phosphate released from adenosine-5′-monophosphate (AMP) by the action of calf intestinal alkaline phosphatase (CIAP). Glycerol at 2% stabilized the complex between MLG and phosphomolybdate, whose absorbance at 630 nm was proportional to phosphate concentrations with resistance to common substances in PDE4 reaction mixtures except papaverine. CIAP had the Michaelis-Menten constant (Km) of (12.0 ± 2.1) μM (n = 3) for AMP at pH 7.4, and was resistant to EDTA below 0.20 mM. By the coupled end-point assay at 30.0 U L−1 CIAP with reaction durations within 30 min, the rates to release phosphate in PDE4 reaction mixtures containing 10.0 mM MgCl2 and 0.10 mM EDTA linearly responded to the amounts of PDE4 over wide ranges. Meanwhile, Km of PDE4 was (8.8 ± 0.2) μM (n = 2), zinc ion inhibited PDE4 and rolipram had the inhibition constant about 10 nM. These results supported that by the coupled end-point assay, this method was promising to screen of PDE inhibitors that had no interference with the MLG assay of phosphate.  相似文献

Electrochemical paper-based analytical devices (ePADs) with integrated plasma isolation for determination of glucose from whole blood samples have been developed. A dumbbell shaped ePAD containing two blood separation zones (VF2 membranes) with a middle detection zone was fabricated using the wax dipping method. The dumbbell shaped device was designed to separate plasma while generating homogeneous flow to the middle detection zone of the ePAD. The proposed ePADs work with whole blood samples with 24–60% hematocrit without dilution, and the plasma was completely separated within 4 min. Glucose in isolated plasma separated was detected using glucose oxidase immobilized on the middle of the paper device. The hydrogen peroxide generated from the reaction between glucose and the enzyme pass through to a Prussian blue modified screen printed electrode (PB-SPEs). The currents measured using chronoamperometry at the optimal detection potential for H2O2 (−0.1 V versus Ag/AgCl reference electrode) were proportional to glucose concentrations in the whole blood. The linear range for glucose assay was in the range 0–33.1 mM (r2 = 0.987). The coefficients of variation (CVs) of currents were 6.5%, 9.0% and 8.0% when assay whole blood sample containing glucose concentration at 3.4, 6.3, and 15.6 mM, respectively. Because each sample displayed intra-individual variation of electrochemical signal, glucose assay in whole blood samples were measured using the standard addition method. Results demonstrate that the ePAD glucose assay was not significantly different from the spectrophotometric method (p = 0.376, paired sample t-test, n = 10).  相似文献

Tyrosinase from a plant source Amorphophallus companulatus was immobilized on eggshell membrane using glutaraldehyde. Among the three different approaches used for immobilization, activation of eggshell membrane by glutaraldehyde followed by enzyme adsorption on activated support could stabilize the enzyme tyrosinase and was found to be effective. Km and Vmax values for dopamine hydrochloride calculated from Lineweaver-Burk plot were 0.67 mM and 0.08 mM min−1, respectively. Studies on effect of pH showed retention of more than 90% activity over a pH range 5.0-6.5. Membrane bound enzyme exhibited consistent activity in the temperature range 20-45 °C. Shelf life of immobilized tyrosinase system was found to be more than 6 months when stored in phosphate buffer at 4 °C. An electrochemical biosensor for dopamine was developed by mounting the tyrosinase immobilized eggshell membrane on the surface of glassy carbon electrode. Dopamine concentrations were determined by the direct reduction of biocatalytically liberated quinone species at −0.19 V versus Ag/AgCl (3 M KCl). Linearity was observed within the range of 50-250 μM with a detection limit of 25 μM.  相似文献

A single and sensitive spectrophotometric method for chloride ions determination based on a commuted flow system with a 100 cm optical path flow cell and a solid-phase reactor containing immobilized silver chloranilate was proposed. This procedure exploited the AgCl formation in the solid-phase reactor leading the chloranilate ions, monitored spectrophotometrically at 530 nm. The analytical signals were 75-fold higher and the sensitivity was 12-fold than that achieved with a 1 cm flow cell, allowed a chloride determination in the 0.5-100 mg l−1 range. The R.S.D. was 1.1% (n = 20) with a sample throughput of 80 h−1 and a waste generated of ca. 100 ng of chloranilate ions per determination. Four samples of natural waters from São Carlos and Araraquara cities were evaluated using the proposed method. Results agreed with the obtained by a reference method at the 95% confidence level.  相似文献

A long-life capillary enzyme bioreactor was developed that determines glucose concentrations with high sensitivity and better stability than previous systems. The bioreactor was constructed by immobilizing glucose oxidase (GOx) onto the inner surface of a 0.53 mm i.d. fused-silica capillary that was part of a continuous-flow system. In the presence of oxygen, GOx converts glucose to gluconic acid and hydrogen peroxide (H2O2). Hydrogen peroxide detection was accomplished using an amperometric electrochemical detector. The integration of this capillary reactor into a flow-injection (FIA) system offered a larger surface-to-volume ratio, reduced band-broadening effects, and reduced reagent consumption compared to packed column in FIA or other settings. To obtain operational (at ambient temp) and storage (at 4 °C) stability for 20 weeks, the glucose biosensing system was prepared using an optimal GOx concentration (200 mg/mL). This exhibited an FIA peak response of 7 min and a detection limit of 10 μM (S/N = 3) with excellent reproducibility (coefficient of variation, CV < 0.75%). It also had a linear working range from 101 to 104 μM. The enzyme activity in this proposed capillary enzyme reactor was well maintained for 20 weeks. Furthermore, 20 serum samples were analyzed using this system, and these correlated favorably (correlation coefficient, r2 = 0.935) with results for the same samples obtained using a routine clinical method. The resulting biosensing system exhibited characteristics that make it suitable for in vivo application.  相似文献

A flow amperometric enzymatic biosensor for the determination of glucose was constructed. The biosensor consists of a flow reactor based on porous silver solid amalgam (AgSA) and a flow tubular detector based on compact AgSA. The preparation of the sensor and the determination of glucose occurred in three steps. First, a self-assembled monolayer of 11-mercaptoundecanoic acid (MUA) was formed at the porous surface of the reactor. Second, enzyme glucose oxidase (GOx) was covalently immobilized at MUA-layer using N-ethyl-N′-(3-dimethylaminopropyl) carboimide and N-hydroxysuccinimide chemistry. Finally, a decrease of oxygen concentration (directly proportional to the concentration of glucose) during enzymatic reaction was amperometrically measured on the tubular detector under flow injection conditions. The following parameters of glucose determination were optimized with respect to amperometric response: composition of the mobile phase, its concentration, the potential of detection and the flow rate. The calibration curve of glucose was linear in the concentration range of 0.02–0.80 mmol L−1 with detection limit of 0.01 mmol L−1. The content of glucose in the sample of honey was determined as 35.5 ± 1.0 mass % (number of the repeated measurements n = 7; standard deviation SD = 1.2%; relative standard deviation RSD = 3.2%) which corresponds well with the declared values. The tested biosensor proved good long-term stability (77% of the current response of glucose was retained after 35 days).  相似文献

A flow injection method for monitoring xylitol was developed using xylitol oxidase (XYO) immobilized on a VA-Epoxy Biosynth E3-support. The immobilized XYO cartridge had a good operational lifetime (up to 24 h) and storage stability (up to 1 month). The XYO-FIA system with an oxygen electrode was investigated systematically regarding the factors that can affect enzyme activity, such as pH, reaction temperature, carrier solution and sample matrix. In order to attain high activity of the immobilized XYO, potassium phosphate solution (1 M) with 0.5 g l−1 Triton X-100 adjusted to pH 8.5 was used as the carrier solution. Sample matrix effects on the immobilized XYO were also investigated. High concentrations of some components (arabinose, 20 g l−1; xylose, 30 g l−1; NaCl, 30 g l−1) in the sample had significant inhibitory effects on the response of the XYO-FIA system. The performance of the XYO-FIA system was tested by using different sample injection volumes (75-250 μl) and carrier flow rates (0.7-2.0 ml min−1).  相似文献

A three-layer poly(dimethylsiloxane) (PDMS)/glass microfluidic system for performing on-chip solid-phase enzymatic reaction and chemiluminescence (CL) reaction was used for the determination of glucose as a model analyte. A novel method for the immobilization of controlled-pore-glass based reactive particles on PDMS microreactor beds was developed, producing an on-chip solid-phase reactor that featured large reactive surface and low flow impedance. Efficient mixing of reagent/sample/carrier streams was achieved by incorporating chaotic mixer structures in the microfluidic channels. A conventional sequential injection (SI) system was adapted for direct coupling with the microfluidic system, and combined with hydrostatic delivery of reagents to achieve efficient and reproducible sample introduction at 10 μl levels. A detection limit of 10 μM glucose (3σ), and a precision of 3.1% RSD (n=7, 0.2 mM glucose) were obtained using the SI-microfluidic-CL system integrated with a glucose oxidase (GOD) reactor. Carryover was <5% at a throughput of 20 samples/h.  相似文献

A new nanomaterial was prepared by grafting a layer of sulfonated polyaniline network (SPAN-NW) on to the surface of multi-walled carbon nanotube (MWNT) and effectively utilized for immobilization of an enzyme and for the fabrication of a biosensor. SPAN-NW was formed on the surface of MWNT by polymerizing a mixture of diphenyl amine 4-sulfonic acid (DPASA), 4-vinyl aniline (VA) and 2-acrylamido-2-methyl-1-propane sulfonic acid (APASA) in the presence of amine functionalized MWNT (MWNT-NH2). The MWNT-g-SPAN-NW was immobilized with glucose oxidase (GOx) to fabricate the SPAN-NW/GOx biosensor. MWNT-g-SPAN-NW/GOx electrode showed direct electron transfer (DET) for GOx with a fast heterogeneous electron transfer rate constant (ks) of 4.11 s− 1. The amperometric current response of MWNT-g-SPAN-NW/GOx biosensor shows linearity up to 9 mM of glucose, with a correlation coefficient of 0.99 and a detection limit of 0.11 μM (S/N = 3). At a low applied potential of − 0.1 V, MWNT-g-SPAN-NW/GOx electrode possesses high sensitivity (4.34 μA mM− 1) and reproducibility towards glucose.  相似文献

Lactic acidemia is commonly associated with severe diseases in pediatric patients. Quantitation of blood lactate and pyruvate is important for the diagnosis and clinical management. A liquid chromatography–tandem mass spectrometry (LC–MS/MS) method using dried blood spots (DBS) was developed and could be used for simultaneous quantification of blood lactate and pyruvate. The applicability of the developed method was tested and confirmed by the regression analysis between LC–MS/MS method and enzymatic assay. Lactate and pyruvate were extracted from DBS obtained from 580 full-term, 120 pre-term infants (gestations ranging from 24 to 36 weeks), and 65 patients with suspected lactic acidemia, with methanolic internal standard (IS) solutions of sodium l-lactate-13C3 and pyruvate-13C3. An API-2000 LC–MS/MS system with multiple reaction monitoring (MRM) mode was applied. The within-run and between-run precisions (CV%) were determined and the results were 1.9% and 3.9% for lactate (n = 20) and 5.7% and 7.3% for pyruvate (n = 20). The linearity of lactate (r = 0.9986) and pyruvate (r = 0.9973) based on the IS was excellent. The parameter r squared (r2) of linear regression between LC–MS/MS method and enzymatic assay was 0.9405 for lactate and 0.9447 for pyruvate, respectively, and the agreement between these methods was consistent and acceptable. The stability of lactate and pyruvate on DBS was also confirmed. The LC–MS/MS method we developed is a specific, sensitive, and reproducible method for measuring blood lactate and pyruvate concentrations. The use of DBS in this method makes it particularly attractive for pediatric patients.  相似文献
