The oxidation of selected clinically important neurotransmitter metabolites with acidic potassium permanganate in the presence of polyphosphates evokes chemiluminescence of sufficient intensity to enable the sensitive determination of these species. Limits of detection for 5-hydroxyindole-3-acetic acid (5-HIAA), vanilmandelic acid (VMA; α,4-dihydroxy-3-methoxybenzeneacetic acid), 4-hydroxy-3-methoxyphenylglycol (MHPG), homovanillic acid (HVA, 4-hydroxy-3-methoxyphenylacetic acid) and 3,4-dihydroxyphenylacetic acid (DOPAC) were between 5 × 10−9 and 4 × 10−8 M, using flow-injection analysis methodology. In addition, we demonstrate the rapid determination of homovanillic acid and 5-hydroxyindole-3-acetic acid in human urine - without the need for extraction procedures - using monolithic column chromatography with chemiluminescence detection.  相似文献

A new sensor for simultaneous determination of peroxyacetic acid and hydrogen peroxide using silver nanoparticles (Ag-NPs) as a chromogenic reagent is introduced. The silver nanoparticles have the catalytic ability for the decomposition of peroxyacetic acid and hydrogen peroxide; then the decomposition of them induces the degradation of silver nanoparticles. Hence, a remarkable change in the localized surface plasmon resonance absorbance strength could be observed. Spectra-kinetic approach and artificial neural network was applied for the simultaneous determination of peroxyacetic acid and hydrogen peroxide. Linear calibration graphs were obtained in the concentration range of (8.20 × 10−5 to 2.00 × 10−3 mol L−1) for peroxyacetic acid and (2.00 × 10−5 to 4.80 × 10−3 mol L−1) for hydrogen peroxide. The analytical performance of this sensor has been evaluated for the detection of simultaneous determination of peroxyacetic acid and hydrogen peroxide in real samples.  相似文献

Potentiometric FIA titrations were performed to determine enzyme activities of lipase type B from Candida antarctica, CAL-B. Two substrates, triacetin and tributyrin were hydrolyzed in phosphate buffer solutions, and the concentration change of the base component of the buffer was titrated in a carrier solution containing hydrochloric acid and potassium chloride. The system was calibrated with butyric acid and acetic acid, respectively. FIA titration peaks were evaluated with respect to peak height and peak area. Butyric acid and acetic acid could be titrated in the buffer solution from 3 × 10−3 mol L−1 to 0.1 mol L−1. The detection limit of enzyme activity was determined to be 0.07 U mL−1 (15 min reaction time) and the minimum activity was calculated to be 0.035 units corresponding to 35 nmol min−1. The specific activities of lipase B for the hydrolysis of tributyrin and triacetin were determined as 16 ± 2 U mg−1 and 2 ± 0.2 U mg−1 (per mg commercial lipase preparation), respectively.  相似文献

2-(Methylamino)nicotinic acid was readily prepared in high yield by reacting 2-chloronicotinic acid with 40% aq MeNH2 under microwave irradiation either at 120 °C for 2 h or at 140 °C for 1.5 h. Subsequently, we found that a range of 2-aminonicotinic acids could be obtained under microwave heating. The optimal reaction conditions involved the use of 3 equiv of amine, water as the solvent and heating at 200 °C for 2 h in the presence of diisopropylethylamine (3 equiv).  相似文献

The cross-coupling reaction of triarylbismuths with acid chlorides using a catalytic amount of PdCl2/PPh3 afforded the corresponding ketones in high yields. The reactions of aromatic and aliphatic acid chlorides occurred with atom efficiency, as 3 equiv of acid chlorides coupled effectively with 1 equiv of triarylbismuths to yield 3 equiv of the corresponding diaryl and alkyl aryl ketones.  相似文献

Molybdate was examined as a complex-forming additive to the CE background electrolytes (BGE) to affect the selectivity of separation of polyhydric phenols such as flavonoids (apigenin, hyperoside, luteolin, quercetin and rutin) and hydroxyphenylcarboxylic acids (ferulic, caffeic, p-coumaric and chlorogenic acid). Effects of the buffer concentrations and pH and the influence of molybdate concentration on the migration times of the analytes were investigated. In contrast to borate (which is a buffering and complex-forming agent generally used in CE at pH ≥9) molybdate forms more stable complexes with aromatic o-dihydroxy compounds and hence the complex-formation effect is observed at considerably lower pH. Model mixtures of cinnamic acid, ferulic acid, caffeic acid and 3-hydroxycinnamic acid were separated with 25 mM morpholinoethanesulfonic acid of pH 5.4 (adjusted with Tris) containing 0.15 mM sodium molybdate as the BGE (25 kV, silica capillary effective length 45 cm × 0.1 mm I.D., UV-vis detection at 280 nm). With 25 mM 2-hydroxy-3-[4-(2-hydroxyethyl)-1-piperazinyl]propanesulphonic acid/Tris of pH* 7.4 containing 2 mM sodium molybdate in aqueous 25% (v/v) methanol as the BGE mixtures of all the above mentioned flavonoids, p-coumaric acid and chlorogenic acid could be separated (the same capillary as above, UV-vis detection at 263 nm). The calibration curves (analyte peak area versus concentration) were rectilinear (r > 0.998) for ≈8-35 μg/ml of an analyte (with 1-nitroso-2-naphthol as internal standard). The limit of quantification values ranged between 1.1 mg l−1 for p-coumaric acid and 2.8 mg l−1 for quercetin. The CE method was employed for the assay of flavonoids in medicinal plant extracts. The R.S.D. values ranged between 0.9 and 4.7% (n = 3) when determining luteolin (0.08%) and apigenin (0.92%) in dry Matricaria recutita flowers and rutin (1.03%) and hyperoside (0.82%) in dry Hypericum perforatum haulm. An ascorbic acid sensor was fabricated via the drop-casting of dodecylbenzene sulphonic acid (DBSA)-doped polyaniline nanoparticles onto a screen-printed carbon-paste electrode. The modified electrode was characterised with respect to the numbers of drop cast layers, optimum potential and operating pH. The sensor was found to be optimal at neutral pH and at 0 V vs. Ag/AgCl. Under these conditions, the sensor showed good selectivity and sensitivity in that it did not respond to a range of common interferents such as dopamine, acetaminophen, uric acid and citric acid, but was capable of the detection of ascorbic acid at a sensitivity of 0.76 μA mM−1 or 10.75 μA mM−1 cm−2 across a range from 0.5 to 8 mM (r2 = 0.996, n = 6), and a limit of detection of 8.3 μM (S/N = 3). The sensor was compared to a range of other conducting polymer-based ascorbate sensors and found to be comparable or superior in terms of analytical performance.  相似文献

Diamagnetic ruthenium(II) complexes of the type [Ru(L)(CO)(B)(EPh3)] [where E = As, B = AsPh3; E = P, B = PPh3, py (or) pip and L = dibasic tridentate ligands dehydroacetic acid semicarbazone (abbreviated as dhasc) or dehydroacetic acid phenyl thiosemicarbazone (abbreviated as dhaptsc)] were synthesized from the reaction of [RuHCl(CO)(B)(EPh3)2] (where E = As, B = AsPh3; E = P, B = PPh3, py (or) pip) with different tridentate chelating ligands derived from dehydroacetic acid with semicarbazide or phenylthiosemicarbazide. All the complexes have been characterized by elemental analysis, FT-IR, UV–Vis and 1H NMR spectral methods. The coordination mode of the ligands and the geometry of the complexes were confirmed by single crystal X-ray crystallography of one of the complexes [Ru(dhaptsc)(CO)(PPh3)2] (5). All the complexes are redox active and are monitored by cyclic voltammetric technique. Further, the catalytic efficiency of one of the ruthenium complexes (5) was determined in the case of oxidation of primary and secondary alcohols into their corresponding aldehydes and ketones in the presence of N-methylmorpholine-N-oxide.  相似文献

The reaction of [RuHCl(CO)(B)(EPh3)2] (where E = As, B = AsPh3; E = P, B = PPh3, py, pip, or mor) and dehydroacetic acid thiosemicarbazone (abbreviated as H2dhatsc where H2 stands for the two dissociable protons) in benzene under reflux afford a series of new ruthenium(II) carbonyl complexes containing dehydroacetic acid thiosemicarbazone of general formula [Ru(dhatsc)(CO)(B)(EPh3)] (where E = As, B = AsPh3; E = P, B = PPh3, py, pip or mor; dhatsc = dibasic tridentate dehydroacetic acid thiosemicarbazone). All the complexes have been characterized by elemental analyses, FT-IR, UV-Vis, and 1H NMR spectral methods. The thiosemicarbazone of dehydroacetic acid behaves as dianionic tridentate O, N, S donor and coordinates to ruthenium via phenolic oxygen of dehydroacetic acid, the imine nitrogen of thiosemicarbazone and thiol sulfur. In chloroform solution, all the complexes exhibit metal-to-ligand charge transfer transitions (MLCT). The crystal structure of one of the complexes [Ru(dhatsc)(CO)(PPh3)2] (1) has been determined by single crystal X-ray diffraction which reveals the presence of a distorted octahedral geometry in the complexes. All the complexes exhibit an irreversible oxidation (RuIII/RuII) in the range 0.76-0.89 V and an irreversible reduction (RuII/RuI) in the range −0.87 to −0.97 V. Further, the free ligand and its ruthenium complexes have been screened for their antibacterial and antifungal activities. The complexes show better activity in inhibiting the growth of bacteria Staphylococcus aureus and Escherichia coli and fungus Candida albicans and Aspergillus niger. These results made it desirable to delineate a comparison between free ligand and its ruthenium complexes.  相似文献

An efficient methodology for the determination of carbohydrate content in soils, employing acid hydrolysis and subsequent capillary electrophoresis analysis (CE), is here described. Polysaccharides present in soil samples were hydrolyzed, at 100 °C during 4 h, to their monosaccharide form, by addition of 2 mol dm−3 trifluoroacetic acid (TFA) directly to soil. The resulting monosaccharides were then quantitatively derivatized with 4-aminobenzoic acid ethyl ester, via reductive amination with sodium cyanoborohydride, and separated by CE, coupled to an UV-vis diode array set at 300 nm. Separation electrolyte consisted of 5.0 × 10−2 mol dm−3 sodium tetraborate buffer (pH 10.2) and 6 × 10−3 mol dm−3 sodium dodecylsulphate. A 78 cm long capillary with an internal and external diameter of 75 and 375 μm, respectively, was used and separation performed at 16 °C, with an applied voltage of 30 kV. Quantification was undertaken using ribose as the internal standard. As an application example, carbohydrate composition (w/w) of a farmyard manure fertilized soil was found to vary between 0.0045 ± 0.0003% (glucose) and 0.0267 ± 0.0002% (arabinose) of the total soil content. Xylose, rhamnose, mannose, fucose and galactose content were also studied in the present work.  相似文献

A new method for uric acid (UA) determination based on the quenching of the cathodic ECL of the tris(2,2-bipyridine)ruthenium(II)–uricase system is described. The biosensor is based on a double-layer design containing first tris(2,2-bipyridine)ruthenium(II) (Ru(bpy)32+) electrochemically immobilized on graphite screen-printed cells and uricase in chitosan as a second layer. The uric acid biosensing is based on the ECL quenching produced by uric acid over the cathodic ECL caused by immobilized Ru(bpy)32+ in the presence of uricase. The use of a −1.1 V pulse for 1 s with a dwelling time of 10 s makes it possible to estimate the initial enzymatic rate, which is used as the analytical signal. The Stern–Volmer type calibration function shows a dynamic range from 1.0 × 10−5 to 1.0 × 10−3 M with a limit of detection of 3.1 × 10−6 M and an accuracy of 13.6% (1.0 × 10−4 M, n = 5) as relative standard deviation. Satisfactory results were obtained for urine samples, creating an affordable alternative for uric acid determination.  相似文献

Electrodeposition of Pt-Pb nanoparticles (PtPbNPs) to multi-walled carbon nanotubes (MWCNTs) resulted in a stable PtPbNP/MWCNT nanocomposite with high electrocatalytic activity to glucose oxidation in either neutral or alkaline medium. More importantly, the nanocomposite electrode with a slight modification exhibited high sensitivity, high selectivity, and low detection limit in amperometric glucose sensing at physiological neutral pH (poised at a negative potential). At +0.30 V in neutral solution, the nanocomposite electrode exhibited linearity up to 11 mM of glucose with a sensitivity of 17.8 μA cm−2 mM−1 and a detection limit of 1.8 μM (S/N = 3). Electroactive ascorbic acid (0.1 mM), uric acid (0.1 mM) and fructose (0.3 mM) invoked only 23%, 14% and 9%, respectively, of the current response obtained for 3 mM glucose. At −0.15 V in neutral solution, the electrode responded linearly to glucose up to 5 mM with a detection limit of 0.16 mM (S/N = 3) and detection sensitivity of ∼18 μA cm−2 mM−1. At this negative potential, ascorbic acid, uric acid, and fructose were not electroactive, therefore, not interfering with glucose sensing. Modification of the nanocomposite electrode with Nafion coating followed by electrodeposition of a second layer of PtPbNPs on the Nafion coated PtPbNP/MWCNT nanocomposite produced a glucose sensor (poised at −0.15 V) with a lower detection limit (7.0 μM at S/N = 3) and comparable sensitivity, selectivity and linearity compared to the PtPbNP/MWCNT nanocomposite. The Nafion coating lowered the detection limit by reducing the background noise, while the second layer of PtPbNPs restored the sensitivity to the level before Nafion coating.  相似文献

A new thermometric sensor, which is a transistor (OC71), has been introduced to follow thermometric titrations successfully to clear end points. The sensor was suitable in both normal and differential modes of titration. It is possible to titrate down to 1.32 μmol of HCl and 26.4 μmol of H3BO3in a final 20 ml solution with accuracy and precision of 1%, 2.2% and 1.4%, 2.2%, respectively. The sensor, in association with a pH glass electrode, was used for the determination of pK values of some well established weak acids such as, acetic acid (4.77), phosphoric acid (pK1 = 2.18, pK2 = 7.20 and pK3 = 12.32) as well as for a very weak acid of uncertain pK values H3BO3 (pK1 = 9.20, pK2 = 12.7 and pK3 = 13.80). The sensor was also examined for kinetic catalytic determination of iron(III) in water, milk and pharmaceuticals.  相似文献

A series of poly(aryl ether benzimidazole) copolymers bearing different aryl ether linkage contents were synthesized by condensation polymerization in polyphosphoric acid (PPA) by varying the feed ratio of 4,4′-dicarboxydiphenyl ether (DCPE) to terephthalic acid (TA). As the ether unit content in the copolymer increased, the solubility of the copolymer in PPA and N,N′-dimethylacetamide/LiCl improved. For example 3–7 wt.% DMAc solution containing 2 wt.% of LiCl could be prepared from the copolymers. XRD studies revealed that the incorporation of flexible aryl ether linkages increased the chain d-spacings of the polymer backbones and decreased the crystallinity of the copolymers. Still, these copolymers having ether linkages showed reasonably good thermal/mechanical stability and high proton conductivity. For example, the copolymer with 30 mol% ether linkage had a tensile strength of 43 MPa (at 26 °C and 40% relative humidity) at an acid doping level of 7.5 mol H3PO4 and a proton conductivity of 0.098 S cm−1 (at 180 °C and 0% relative humidity) at an acid doping level of 6.6 mol H3PO4.  相似文献

A simple and highly sensitive high-performance liquid chromatography (HPLC) for the determination of pipecolic acid (PA) in serum was developed. Pipecolic acid and nipecotic acid (internal standard (IS)) were derivatised with 4-(5,6-dimethoxy-2-phthalimidinyl)-2-methoxyphenylsulfonyl chloride (DMS-Cl) to produce fluorescent sulfonamides. The labelling reaction was carried out at 70 °C for 15 min at pH 9.0. The fluorescent derivatives were separated on a reversed-phase column (45 °C) with a stepwise elution using methanol/acetonitrile/10 mmol l−1 acetic acid (42:5:53) and methanol at a flow rate of 1.0 ml/min and detected at excitation and emission wavelengths of 316 and 403 nm, respectively. The labelling yield was 100.8%. The detection limit of pipecolic acid was 4 fmol at signal-to-noise ratio of 3. The within-day and day-to-day relative standard deviations were 3.3-8.1 and 1.4-6.4%, respectively. The concentration of pipecolic acid in normal human serum was 1.09±0.37 μmol l−1.  相似文献

A new, simple and sensitive spectrofluorimetric method for the determination of salicylic acid (λex = 315 nm, λem = 408 nm) using As(III) as a sensitizing reagent has been investigated by measuring the increase of fluorescence intensity of salicylic acid due to the complexation of As(III)-salicylic acid in presence of sodium dodecyl sulfate (SDS) 10−3 M. Under optimum conditions, a significant relationship was obtained between the fluorescence intensity and salicylic acid concentration. A linear calibration curve was obtained in the range 13.8-13812 μg l−1 with product-moment correlation coefficient (R) 0.99985 and detection limit 4.2 μg l−1. The R.S.D. is 2.35% (n = 5).The method was applied successfully to the determination of salicylic acid in human serum.  相似文献

The chemical interactions of 1-methyl-3-hydroxy-4-pyridinecarboxylic acid (1M3H4P) and 1-methyl-4-hydroxy-3-pyridinecarboxylic acid (1M4H3P) with Fe(III) were investigated in aqueous 0.6 m (Na)Cl at 25 °C by means of potentiometric titrations and UV–Vis spectrophotometry. A large number of complexes were formed in solution with stoichiometry FeLxHy, x = 1, 2, 3 and y = 0, −1, −2. In view of a possible application to Fe(III) chelation therapy, the efficiencies of the ligands to chelate the metal ion were evaluated in vitro at physiological pH. 1M3H4P and 1M4H3P show a lower complexation efficiency with Fe(III) than that of other available chelators, but higher than that of their non-methylated analogs 3H4P and 4H3P.  相似文献

A new method is proposed for the chemiluminescent determination of the pesticide 3-indolyl acetic acid by means of an flow injection analysis system. The chemiluminescence emission is obtained by oxidation of the analyte with Ce (IV) in nitric acid and presence of β-cyclodextrine.The continuous-flow method allows the determination of 159 samples h−1 of 3-indolyl acetic acid in an interval of concentrations over the range 0.5-15.0 mg l−1. The limit of detection was 0.1 μg l−1 and the R.S.D. (n, 17) at 2.0 mg l−1 of the pesticide level was 2.7%. The method was applied to water samples.  相似文献

A simple and rapid reversed-phase HPLC-UV method was developed for the determination of triterpenic acids in the crude extract of Prunellae Spica. Five triterpenic acids were extracted and isolated from P. Spica as marker compounds for use in the quality control of herbal medicines. Various solvent extraction techniques were evaluated, and the greatest efficiency was observed with sonication in 100% ethanol. Elemental compositions of the five marker compounds were determined by high-resolution mass spectroscopy. The dynamic range of the HPLC-UV method depended on the specific analyte, and acceptable quantitation was obtained between 10 and 250 μg mL−1 for oleanolic acid, between 10 and 300 μg mL−1 for ursolic acid, between 3 and 75 μg mL−1 for 2α,3α,24-trihydroxyolean-12en-28oic acid, between 5 and 100 μg mL−1 for euscaphic acid, and between 5 and 100 μg mL−1 for 2α,3α-dihydroxyurs-12en-28oic acid. The method was deemed satisfactory by inter- and intra-day validation and exhibited both high accuracy and precision (relative standard deviation <9.4%). Overall limits of quantitation and detection were approximately 0.5-2.5 μg mL−1 at a signal-to-noise ratio (S/N) of 3 and were about 3.0-10.0 μg mL−1 at a S/N of 10. In addition, principal component analysis (PCA) was performed on the analytical data of 15 different P. Spica samples in order to classify samples collected from different regions.  相似文献

In this paper, four organic acids constituents of Plateau alfalfa roots have been identified and detected by a novel capillary electrophoresis (CE) strategy which combined chitosan (CS) trapping and cetyltrimethyl ammonium bromide (CTAB) assisted sample stacking. Under the optimized condition, organic acids, i.e., aconitic acid, gallic acid, citric acid and l-malic acid were concentrated and separated within 3 min. Validation parameters of this method (such as detection limits, linearity and precision) were also investigated and the limit of detection (LOD) was 2.41-53.9 ng mL−1. Linearity was obtained over the magnitude range of 5-4000 ng mL−1 approximately for different organic acids and 3 × 102-1.5 × 104 folds enrichment was achieved. The method has been applied to the determination of organic acids in roots of normal grown Plateau alfalfa and stressing affected Plateau alfalfa. Satisfactory results and recoveries were obtained in the analysis without costly and complicated sample pretreatment.  相似文献
