We have previously used the molecular‐imprinting method for the synthesis of artificial gel antibodies, highly selective for various proteins. In the present work, we have synthesized artificial gel antibodies against human albumin with the aim to develop a simple and rapid procedure to measure the concentration of this protein in samples of clinical interest. The procedure, based on the design of a standard curve (see the preceding paper), was applied on a quantitative analysis of albumin in human plasma and cerebrospinal fluid (CSF). We found that our technique permitted detection of albumin in these body fluids with high precision and that the concentration of this protein was significantly enhanced in CSF from patients with amyotrophic lateral sclerosis (ALS), compared to control samples. This finding is in agreement with results from earlier studies, which confirms the validity of our analysis technique and suggests that the barrier permeability may be affected in ALS, perhaps also for other proteins. No enhancement in plasma levels of albumin was seen in patients with ALS, but rather a decrease. The results further indicate that our approach might also apply well to other biomarkers for the actual neurological disease and other disorders.  相似文献

The ICH Q2(R1) (International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use) guideline for testing linearity in validation of analytical procedures suggests that “linearity should be evaluated by visual inspection of a plot of signals as a function of analyte concentration or content.” The EP6‐A guideline recommends more quantitative methods that compare straight‐line and higher‐order polynomial curve fits. In this paper, a new equivalence test is proposed to compare the quality of a straight‐line fit to that of a higher‐order polynomial. By using orthogonal polynomials and generalized pivotal quantity analysis, one may estimate the probability of equivalence between a straight line and a polynomial curve fit either in the assay signal space (the Y values) or in the concentration space (the X values). In the special case of the linear‐to‐quadratic polynomial comparison, an equivalence test may be constructed via a two one‐sided T test. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

The spectral displacement method in presence of methyl orange has been coupled to the continuous variation plot to allow the determination of the stoichiometries for inclusion compounds. Such a “competitive continuous variation plot” (CCV plot) is especially useful for the study of substrates which are not directly observable by spectroscopy or which are too poorly soluble to give observable signals. Moreover, when a mixture of complexes is observed, the position of the maximum in the competitive continuous variation plot gives information on the relative affinities of the complexes, which is not the case in the classical Job plot which also depends of the intrinsic spectral characteristics of each complex. The method is not restricted to β-cyclodextrin inclusion compounds, and it may be applied to any complexes if an appropriate competitive system is available.  相似文献

We demonstrated a unique optofluidic lab-on-a-chip device that can measure optically encoded forward scattering signals. From the design of the spatial pattern, we can measure the position and velocity of each cell in the flow and generate a 2-D cell distribution plot over the cross section of the channel. Moreover, we have demonstrated that the cell distribution is highly sensitive to its size and stiffness. The latter is an important biomarker for cell classification and our method offers a simple and unequivocal method to classify cells by their size and stiffness. We have proved the concept using live and fixed HeLa cells. Due to the stiffness and size difference of neutrophils compared to other types of white blood cells, we have demonstrated detection of neutrophils from other blood cells. Finally, we have performed the test using 5 μL of human blood. In a greatly simplified blood preparation process, skipping the usual steps of anticoagulation, centrifuge, antibody labelling or staining, filtering, etc., we have demonstrated that our device and detection principle can count neutrophils in whole human blood. Our system is compact, inexpensive and simple to fabricate and operate, having a commodity laser diode and a Si PIN photoreceiver as the main pieces of hardware. Although the results are still preliminary, the studies indicate that this optofluidic device holds promise to be a point-of-care and home care device to measure neutrophil concentration, which is the key indicator of the immune functions for cancer patients undergoing chemotherapy.  相似文献

How do we decide the stoichiometry of host–guest complexes?” This question has long been answered by the Job plot since its first report in 1928. However, as the Job plot was claimed to be misleading in 2016, the question became an open question again and called for renewed investigations. An information-theoretic approach, called Akaike's information criterion, is introduced in this study to select the best model of host–guest complexes, which can rank the models with weight of evidence. A few test cases with unique cylindrical hosts were examined to demonstrate the applicability of the information-theoretic method. Consequently, reasonable views over the thermodynamic behaviors of dumbbell-and-cylinder complexes were obtained. Akaike's information criterion can be a useful and superior alternative to statistical null hypothesis testing, which was proposed as a remedy in place of the Job plot.  相似文献

Abstract

Centrifugal Partition Chromatography (CPC) is a technique that uses two immiscible liquids. One liquid is used as a stationary phase, the second one as the mobile phase. Using the liquid systems methanol-hexane and octanol-water, the selectivity and efficiency of CPC apparatus were tested. It is shown that CPC can be used to determine partition coefficients or to purify compounds. The selectivity can be changed the same way as in liquid chromatography. The efficiency study has shown that the plate count presents a minimum value at a particular flow rate. By plotting the number of channels required to obtain one theoretical plate versus the flow rate, it is possible to obtain a “van Deemter-type” plot. Interestingly, these plots show maxima for CPC. These results are opposite to those found for liquid or gas chromatography which have minima. A flow change, from laminar to nonlaminar is thought to explain these results. Reynolds numbers were determined for all solvent systems. Efficiency in CPC was also found to be solute dependent: the larger the partition coefficient, the lower the efficiency.  相似文献

The interaction of bromothymol blue(BB) with human serum albumin(HSA) was studied by electrochemical techniques and a sensitive method for proteins assay was developed. When BB interacted with HSA, the voltammetric peak current value of BB decreased linearly with the concentration of HSA in a range of 1.0--40.0 mg/L, and the peak potential shifted negatively. Based on the results, a sensitive assay method for proteins, such as HSA, bovine serum albumin(BSA), and egg albumin etc. was established. This method was further applied to determining the HSA in healthy human blood samples, and the results are not significantly different from those obtained by the classic Coomassie Brilliant Blue G-250 spectrophotometic method. The detecting conditions of this method were optimized and the interaction mechanism was discussed. The results show that the electrochemical parameters(formal potential E^0, standard rate constant of the electrode reaction ks, parameter of kinetic nα) of BB have no obvious changes before and after the interaction, which indicate that BB can interact with HSA, forming an electrochemical non-active complex. The equilibrium constant(βs) and the binding ratio(m) for this complex were calculated. The m is 4 and βs is 1.41 × 10^19. This method is fast, simple, highly sensitive, and has good selectivity, which can be used in clinical measurements.  相似文献

Selective oxidation of amines using oxygen as terminal oxidant is an important area in green chemistry. In this work, we describe the use of graphite‐supported gold nanoparticles (AuNPs/C) to catalyze aerobic oxidation of cyclic and acyclic benzylic amines to the corresponding imines with moderate‐to‐excellent substrate conversions (43–100 %) and product yields (66–99 %) (19 examples). Oxidation of N‐substituted 1,2,3,4‐tetrahydroisoquinolines in the presence of aqueous NaHCO3 solution gave the corresponding amides in good yields (83–93 %) with high selectivity (up to amide/enamide=93:4) (6 examples). The same protocol can be applied to the synthesis of benzimidazoles from the reaction of o‐phenylenediamines with benzaldehydes under aerobic conditions (8 examples). By simple centrifugation, AuNPs/C can be recovered and reused for ten consecutive runs for the oxidation of dibenzylamine to N‐benzylidene(phenyl)methanamine without significant loss of catalytic activity and selectivity. This protocol “AuNPs/C+O2” can be scaled to the gram scale, and 8.9 g (84 % isolated yield) of 3,4‐dihydroisoquinoline can be obtained from the oxidation of 10 g 1,2,3,4‐tetrahydroisoquinoline in a one‐pot reaction. Based on the results of kinetic studies, radical traps experiment, and Hammett plot, a mechanism involving the hydrogen‐transfer reaction from amine to metal and oxidation of M‐H is proposed.  相似文献

Paper impregnated with Prussian blue sorbs cesium in low concentration to a different extent if varying initial concentrations of cesium are present. This dependence can be employed for the determination of submicrogram amounts of cesium. The selectivity and precision of the method are discussed. The mention of the word “crystal” invokes images of minerals, gems, and rocks, all of which are inevitably solid, hard, and durable entities with well‐defined smooth faces and straight edges. With the discovery in the first half of the 20th century that many molecular crystals are soft and can be deformed in a similar way as rubber or plastic, this perception is changing, and both the concept and formal definition of what a crystal is may require reinterpretation. The seemingly naïve question posed in the title of this Minireview does not have a simple answer. Here, we discuss how the effects of the elastic and plastic deformation of molecular crystals on the diffraction signature give primary evidence of their degree of crystallinity. In most cases, the definition of a crystal holds for both elastically and plastically deformed crystals and, unless there is significant or complete physical separation of the crystal during the deformation, they can safely be considered (deformed) single crystals with a high concentration of defects.  相似文献

A new method of detecting and diagnosing melanoma based on melanoma biomarker was developed and its feasibility demonstrated. The method is based on an electrochemical biosensor platform comprised of a special biochip and device, performing a multi‐channel amperometric detection of the enzymatic activity of tyrosinase, an enzyme biomarker of melanoma. The newly developed biosensor platform is able to electrochemically detect tyrosinase activity in fresh biopsy samples. This bioelectrochemical detection method is rapid, yielding results within minutes from biopsy removal. Using “as is” biopsy samples, without pretreatment, simplifies the process, saves time and reduces cost and labor dramatically. Using modern portable microelectronics provides an accurate biomarker expression measurement at the “point of care” increasing the accessibility of new bio‐chip technologies to the public.  相似文献

We describe the polarity of selectivity of a GC separation system in terms of Retention Index data. In a series-coupled capillary system having stationary phases of differing polarity even slight (independent!) carrier gas flow changes in one part of the capillary series result in a dramatic change of selectivity. “Dramatic” is a relative term! Using a simple electronically controlled flow changing device we found retention index changes of several hundred units for polar test compounds such as phenols on a SE30/Carbowax tandem. This means: The classical theoretical model for understanding retention (and selectivity) in chromatography must be corrected. We propose a very simple approach involving addition of the expression RESIDENCE TIME to the chromatographic vocabulary. Instead of using flow resistors, one can just add a pressure regulator to the coupling point. A powerful analytical concept is now in easy reach.  相似文献

Proteins play important roles in biological and cellular processes. The levels of proteins can be useful biomarkers for cellular events or disease diagnosis, thus the method for sensitive and selective detection of proteins is imperative to proteins express, study, and clinical diagnosis. Herein, we report a “signal-on” platform for the assay of protein based on binding-induced strategy and photoinduced electron transfer between Ag nanoclusters and split G-quadruplex-hemin complexes. By using biotin as the affinity ligand, this simple protocol could sensitively detect streptavidin with a detection limit down to 10 pM. With the use of an antibody as the affinity ligand, a method for homogeneous fluorescence detection of Prostate Specific Antigen (PSA) was also proposed with a detection limit of 10 pM. The one-step and wash-free assay showed good selectivity. Its high sensitivity, acceptable accuracy, and satisfactory versatility of analytes led to various applications in bioanalysis.  相似文献

Simple, sensitive, and selective detection of specific biopolymers is critical in a broad range of biomedical and technological areas. We present a design of turn-on near-infrared (NIR) fluorescent probes with intrinsically high signal-to-background ratio. The fluorescent signal generation mechanism is based on the aggregation/de-aggregation of phthalocyanine chromophores controlled by selective binding of small-molecule “anchor” groups to a specific binding site of a target biopolymer. As a proof-of-concept, we demonstrate a design of a sensor for EGFR tyrosine kinase—an important target in cancer research. The universality of the fluorescent signal generation mechanism, as well as the dependence of the response selectivity on the choice of the small-molecule “anchor” group, make it possible to use this approach to design reliable turn-on NIR fluorescent sensors for detecting specific protein targets present in the low-nanomolar concentration range.  相似文献

Previous studies on selective degradation of polyethylene single crystals with fuming nitric acid have been extended, both by using acid of lower concentration which gave better control over the degradation, and by resorting to ozone as an oxidizing agent which among others enabled the degradation temperature to be conveniently lowered. The molecular weight distribution was followed by gel-permeation chromatography in the course of degradation. Complete consistency between these different methods has been established, modifying some of the previous conclusions reached by this method. The principal feature which emerges is that we have a distribution of fold lengths. The largest straight fold stems can stretch across nearly the entire layer defined by the low-angle x-ray period, while there is a continuous distribution of shorter folds terminating deeper down in the crystals. The limiting depth at which the number of terminating folds becomes negligible can be identified and quantitatively assessed. The method of analysis is described, and individual data are discussed in detail. This picture of a fold surface layer containing essentially adjacently reentrant folds of uneven length agrees with quite recent results on other related chain-folded systems (annealed crystals, short chains, bulk structures) obtained in these laboratories and thus appears to be of general validity. The consequences of the model for our picture on polymer crystals in particular on the nature of the “amorphous” component, are discussed.  相似文献

Immunization with complex mixtures, like the human plasma resulted in the generation of cloned mAb libraries (PlasmaScan? and QuantiPlasma? libraries, with >1000 individual mAbs) reacting with a nonredundant set of antigenic epitopes. mAb proteomics refers to quasi‐hypothesis‐free profiling of plasma samples with nascent or cloned mAb libraries for the discovery of disease‐specific biomarkers. Once mAbs with biomarker potential have been identified, the next task is the determination of cognate antigens recognized by the respective mAbs. To determine the cognate protein antigen corresponding to each individual mAbs in the cloned mAb libraries, we have separated human plasma by consecutive steps of desalting and various chromatography procedures. The process resulted in 783 fractions, which we termed “Analyte Library” (AL). The AL represents the human plasma proteome in relatively low‐protein complexity fractions. Here, to determine the utility of the AL, we selected ten plasma proteins and checked for their presence in the fractions. Among the ten cases, the distribution of four selected plasma proteins matched expectations, as these proteins were present only in a few fractions corresponding to their physical, chemical, and biochemical properties. However, in six cases, we observed “smear” ‐like distribution or complete absence of the proteins, suggesting that protein–protein interactions or protein variants may alter the observed plasma distribution profiles. Nevertheless, we conclude that the AL is an efficient, high throughput tool to complement the mAb biomarker discovery process with cognate protein antigen identification for each mAbs.  相似文献

Labor intensive experiments are typically required to identify the causal disease variants from a list of disease associated variants in the genome. For designing such experiments, candidate variants are ranked by their strength of genetic association with the disease. However, the two commonly used measures of genetic association, the odds-ratio (OR) and p-value may rank variants in different order. To integrate these two measures into a single analysis, here we transfer the volcano plot methodology from gene expression analysis to genetic association studies. In its original setting, volcano plots are scatter plots of fold-change and t-test statistic (or −log of the p-value), with the latter being more sensitive to sample size. In genetic association studies, the OR and Pearson's chi-square statistic (or equivalently its square root, chi; or the standardized log(OR)) can be analogously used in a volcano plot, allowing for their visual inspection. Moreover, the geometric interpretation of these plots leads to an intuitive method for filtering results by a combination of both OR and chi-square statistic, which we term “regularized-chi”. This method selects associated markers by a smooth curve in the volcano plot instead of the right-angled lines which corresponds to independent cutoffs for OR and chi-square statistic. The regularized-chi incorporates relatively more signals from variants with lower minor-allele-frequencies than chi-square test statistic. As rare variants tend to have stronger functional effects, regularized-chi is better suited to the task of prioritization of candidate genes.  相似文献

Adding light stabilizers to polymeric materials can inhibit or delay the light aging effect and improve the light resistance of materials. 2-(2′-Hydroxy-5′-methylphenyl) benzotriazole (UV-P), as a typical benzotriazole ultraviolet absorber, is widely used in various polymer synthetic materials and products owning to its outstanding oil resistance, color change resistance and low volatility. Currently, it is of great theoretical and practical significance to develop an environmentally friendly method to produce UV-P. Here, we introduce ionic liquids, tetra-butyl ammonium hydroxide, into the palladium-based catalyst, design a “transfer hydrogenation site - alkaline site” duel active center system, and investigate the physical and chemical properties and possible mechanism of this bifunction catalyst system. Such heterogeneous catalytic transfer hydrogenation method can remain 100% conversion and 93.86% selectivity. This bifunction catalyst also shows an outstanding stability when it was used for ten times, proving a green and efficient transfer hydrogenation method for the synthesis of UV-P.  相似文献

Crown ether dyes with pendent anionic side-arms were synthesized for extractionspectrophotometry of alkali and alkaline earth metal ions. Dramatic changes in metal selectivity were obtained simply by changing the nature of the anionic side-arm on the same crown ether skeleton. A structure/metal selectivity relationship is discussed in detail in terms of “chelate” and “intramolecular ion-pair” formation. Small metal cations (high charge density) are preferred in the extraction by a crown ether reagent with a charge-localized anionic side-arm through the formation of a “chelate”. Large metal cations (low charge density) are preferred in the extraction by reagents with a charge-delocalized anionic side-arm through the formation of an “intramolecular ion-pair”. Steric restrictions imposed by the side-arm on the metal ion approaching the crown ether are also important factor in controlling the selectivity of these reagents.  相似文献

Abstract

Atomic absorption spectroscopy has been used to determine the concentration of lead in gasoline. The method employs the use of iodine to “level” the variation in response of different lead alkyls. It has been reported that this method is useless when applied to real gasolines, possibly because of the interference of iodine with olefins in the gasoline. The work performed here shows that olefins do not interfere with the lead analysis even when the concentration of olefins is 254 times that of iodine in the test gasoline.  相似文献
