Manganese is extracted on-line from solid seafood samples by a simple continuous ultrasound-assisted extraction system (CUES). This system is connected to an on-line manifold, which permits the flow-injection flame atomic absorption spectrometric determination of manganese. Optimisation of the continuous leaching procedure is performed by an experimental design. The proposed method allows the determination of manganese with a relative standard deviation of 0.9% for a sample containing 23.4 μg g−1 manganese (dry mass). The detection limit is 0.4 μg g−1 (dry mass) for 30 mg of sample and the sample throughput is ca. 60 samples per hour. Accurate results are obtained by measuring TORT-1 certified reference material. The procedure is finally applied to mussel, tuna, sardine and clams samples.  相似文献

A fast and simple flow-through optosensor was designed and characterized for the direct screening of four tetracycline (TCC) antibiotics (tetracycline, oxytetracycline, chlortetracycline and doxycycline) in water and bovine milk samples. The proposed optosensor provides rapid binary yes/no overall responses, being appropriate for the screening of this family of antibiotics above or below a pre-set concentration threshold.The experimental set-up is based on a flow-injection manifold coupled on-line to a phosphorescence detector. Aliquots of the samples are pretreated with Eu(III) to form room temperature phosphorescent metal chelates and injected in the flow manifold. Those chelates are then on-line retained on a conventional flow-cell (packed with polymeric Amberlite XAD-4 particles) which is placed inside the cell holder of the phosphorimeter. After the emission is registered, the antibiotic-metal complexes are eluted from the packed resin with 1 M HCl (for milk samples a second regeneration step, using methanol, should be performed). A sample throughput of about 20 samples per hour was obtained. Optimum experimental conditions include a pH 9, a Eu(III) concentration of 2 × 10−4 M and 8 mM sodium sulphite as chemical deoxygenant. The phosphorescence emitted by the europium-TCC complexes was measured at 394 and 617 nm for excitation and emission wavelengths, respectively.The unreliability region, given by the probability of false positives and false negatives, respectively (set at 5% in both cases) was in the range between 0.2 and 11.6 nМ for detection of tetracyclines in water samples (at a cut-off level of 4 nM) and in the range between 165 and 238 nM for detection of tetracyclines in milk (cut-off level fixed at the normative EU level of 200 nM). Finally, the applicability of the proposed screening optosensor was tested for the reliable control of tetracyclines in contaminated and uncontaminated water and milk samples.  相似文献

A continuous ultrasound-assisted extraction system connected to a flow injection manifold has been used for the on-line determination of zinc in meat samples by flame atomic absorption spectrometry. An experimental design was used for the optimisation of the continuous manifold. This flow injection methodology allowed a sampling frequency of ca. 80 samples per hour with a relative standard deviation for the whole procedure of 0.3% (for a sample containing 163.6 μg g−1 Zn). The detection limit was 0.6 μg g−1 for a sample amount of 5 mg. Accurate results were obtained by measuring certified reference materials (BCR-186 (pig kidney) and BCR-184 (bovine muscle)). The analytical procedure was applied to different real meat samples with satisfactory results.  相似文献

This paper presents the development of a new flow-injection system combined with solid-surface fluorescence detection for the determination of the widely used fungicide thiabendazole. Nylon powder was probed as a novel solid support for building the optosensor. The method is based on the on-line immobilization of thiabendazole onto nylon in a continuous flow system, followed by the measurement of its native fluorescence. Aqueous samples are directly injected in a water carrier, resulting in a very simple and economical method. The analytical figures of merit obtained using 1500 μL of sample and 75% methanol (v/v) as eluting solution were: linear calibration range from 8 to 120 ng mL−1 (the lowest value corresponds to the quantitation limit), relative standard deviation, 0.9% (n = 5) at a level of 64 ng mL−1, limit of detection calculated according to 1995 IUPAC recommendations is to 2.8 ng mL−1, and sampling rate of 14 samples h−1. The potential interference from other agrochemicals, metal ions and common anions, and the viability of determining thiabendazole in real water samples were also evaluated.  相似文献

A simple, fast chemiluminescence (CL) flow-injection (FI) method based on the reaction of luminol with KMnO4 in alkaline medium has been described for the direct determination of carbofuran. The method is based on the enhancing effect in the emission light from the oxidation of luminol produced in presence of carbofuran. The optimisation of instrumental and chemical variables influencing the CL response of the method has been carried out by applying experimental design, using the proposed flow-injection manifold. Under the optimal conditions, the CL intensity was linear for a carbofuran concentration over the range of 0.06-0.5 μg ml−1, with a detection limit of 0.02 μg ml−1. The method has been successfully applied to the determination of carbofuran residues in spiked water and lettuce samples.  相似文献

Milk samples can be efficiently digested using a focused microwave oven, however the conventional procedure of addition of concentrated acids to the liquid sample leads to digestates with elevated acidity and residual carbon concentrations. In this work a focused microwave oven was applied for acid digestion of bovine milk samples using a conventional and an alternative procedure based on gradual sample addition to hot and concentrated acids. A two-level 23 full factorial design experiment with eight runs was carried out to evaluate the optimum experimental conditions for reducing both the residual carbon and the final acidity of digestates. The three studied parameters were: temperature of the digestion medium for sample addition, addition of sulfuric acid before the sample or during the first step, and number of aliquots of the sample gradually added. The best conditions were attained by adding small aliquots of milk (ten-fold a volume of 0.5 ml added during 5.0 min) to a digestion mixture containing 3.0 ml nitric acid plus 1.0 ml sulfuric acid heated at 105 °C. It was demonstrated that the digestion efficiency of the alternative procedure was better than the conventional procedure, i.e. 98 and 80%, respectively. The alternative procedure was applied for determination of Ba, Ca, Cu, K, Mg, Na, P, and Zn in whole and non-fat bovine milk. The accuracy was proved using two certified reference materials (whole and non-fat milk powder).  相似文献

This paper describes a spectrophotometric flow-injection method for the determination of an antiretroviral drug (zidovudine) in plasma samples. The main goal of this study is the development of feasible analytical method to monitor the plasmatic drug levels in a rapid and simple way as an alternative to chromatographic procedures. The flow-injection system proposed consists of a two-channel manifold with the injection of the sample into an acid carrier and on-line generation of a pH-gradient. The corresponding data are monitored over time using a diode array spectrophotometer. The discrimination of zidovudine species from plasma components is accomplished through chemometric data analysis based on the zidovudine features on both spectral and time domains. A pretreatment procedure consisting of liquid-liquid extraction is used for sample clean-up. However, despite the pretreatment, noticeable amounts of unknown substances acting as interferences are still present in the extracts. As relevant analytical parameters, the analyte recovery in the extraction process is 101% at 5.3 μg mL−1 and the detection limit is 0.013 μg mL−1. Multivariate curve resolution with alternating least squares is used to recover the analyte profiles for its further determination. As a result, the quantification of zidovudine in plasma can be accomplished even in the presence of plasma components with overall prediction error below 3%.  相似文献

A highly sensitive mechanized method has been developed for the determination of mercury in milk by atomic fluorescence spectrometry (AFS). Samples were sonicated for 10 min in an ultrasound water bath in the presence of 8% (v/v) aqua regia, 2% (v/v) antifoam A and 1% (m/v) hydroxilamine hydrochloride, and after that, they were treated with 8 mmol l−1 KBr and 1.6 mmol l−1 KBrO3 in an hydrochloric medium. Atomic fluorescence measurements were made by multicommutation, which provides a fast alternative in quality control analysis, due to the easy treatment of a large number of samples (approximately 70 h−1), and is an environmentally friendly procedure, which involves a waste generation of only 94.5 ml h−1 as compared with the 605 ml h−1 obtained by using continuous AFS measurements. The limit of detection found was 0.011 ng g−1 Hg in the original sample. The method provided a relative standard deviation of 3.4% for five independent analysis of a sample containing 0.30 ng g−1 Hg. To validate the accuracy of the method, a certified reference material NIST-1459 (non-fat milk powder) containing 0.3±0.2 ng g−1 Hg was analysed and a value of 0.27±0.06 ng g−1 Hg was found. A comparison made between data found by the developed procedure and those obtained by microwave-assisted digestion and continuous AFS measurements evidenced a good comparability between these two strategies. Results obtained for commercially available milk samples varied between 0.09 and 0.61 ng g−1 Hg depending on the type of sample and its origin. The confluence of the analytical waste with a 6 mol l−1 NaOH allowed us to reduce the waste generation in a working session from 1 l to 5 g solid residue with a matrix of Fe(OH)3 which contributes to the deactivation of traces of heavy metals presents in the samples that does not form volatile hydrides.  相似文献

An automated flow injection (FI) manifold based on iterative change of the flow direction has been designed to carry out continuous liquid-liquid extraction without phase separation and hydrolysis both with ultrasound-assistance. The dynamic approach has been applied to suppositories from which paracetamol has been extracted in this way into an aqueous phase and hydrolysed prior to reaction with o-cresol in the alkaline medium used as extractant. The three linked reactions, extraction-hydrolysis-derivatisation, cause displacement of the balance to completeness. The strategic location of the photometric flow-cell in the flow-injection manifold enables monitoring of the overall process and the obtaining of a characteristic multipeak recording. The influence of ultrasounds on the different steps was investigated. The indophenol blue dye formed was monitored at 620 nm. The limits of detection (LOD) and quantification (LOQ) of the method are 0.38 and 0.64 μg ml−1, respectively, with a linear range from 0.64 to 50 μg ml−1; a within-laboratory reproducibility between 2.07 and 4.66% and repeatability from 2.01 to 4.63%, both expressed as relative standard deviation. The results obtained with the proposed method are in excellent agreement with those provided by the official method, but with a shorter analysis time, lower sample and reagent consumption and less analyst involvement.  相似文献

In the present work, thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS) was employed for Cu and Zn determination in bovine milk and fruit juice samples without any pretreatment. TS-FF-AAS system was optimized and a sample volume of 300 μl was injected into the carrier stream (0.014 mol l−1 HNO3 at a flow rate of 0.4 ml min−1), and it was introduced into a hot Ni tube. The detection limits obtained for Cu and Zn in aqueous solution were 2.2 and 0.91 μg l−1, respectively, and 3.2 μg l−1 for Cu in a medium containing water-soluble tertiary amines. The relative standard deviations varied from 2.7 to 4.2% (n=12). Sample preparation was carried out by simple dilution in water or in water-soluble tertiary amines medium. Accuracy was checked by performing addition-recovery experiments as well as by using reference materials (whole milk powder, non-fat milk powder, and infant formula). Recoveries varied from 97.7 to 105.3% for Cu and Zn. All results obtained for reference materials were in agreement with certified values at a 95% confidence level.  相似文献

Continuous-flow (CF) and flow-injection (FI) analysis using the fluoride ion-selective electrode (FISE) as detector have been investigated. The measurements were performed in a home-made cell under appropriate flow conditions (2.86 or 3.45 ml min−1, 0.2 ml samples, 10−6 M sodium fluoride). The calibration graph was obtained by plotting the signal height versus concentration of iron in the range of Fe(III) concentration from 10−5 to 10−1 M in acetate buffer (pH 2.8 or 3.4). In all described procedures, the range of linear response extends to the Fe(III) concentration from 1×10−3 to 1×10−1 M, with detection limit 9×10−5 M. The effect of double-line, two-line flow manifold and CF was investigated and discussed.  相似文献

The present work evaluates the influence of vessel cooling simultaneously to microwave-assisted digestion performed in a closed system with diluted HNO3 under O2 pressure. The effect of outside air flow-rates (60–190 m3 h−1) used for cooling of digestion vessels was evaluated. An improvement in digestion efficiency caused by the reduction of HNO3 partial pressure was observed when using higher air flow-rate (190 m3 h−1), decreasing the residual carbon content for whole milk powder from 21.7 to 9.3% (lowest and highest air flow-rate, respectively). The use of high air flow-rate outside the digestion vessel resulted in a higher temperature gradient between liquid and gas phases inside the digestion vessel and improved the efficiency of sample digestion. Since a more pronounced temperature gradient was obtained, it contributed for increasing the condensation rate and thus allowed a reduction in the HNO3 partial pressure of the digestion vessel, which improved the regeneration of HNO3. An air flow-rate of 190 m3 h−1 was selected for digestion of animal fat, bovine liver, ground soybean, non fat milk powder, oregano leaves, potato starch and whole milk powder samples, and a standard reference material of apple leaves (NIST 1515), bovine liver (NIST 1577) and whole milk powder (NIST 8435) for further metals determination by inductively coupled plasma atomic emission spectroscopy (ICP-OES). Results were in agreement with certified values and no interferences caused by matrix effects during the determination step were observed.  相似文献

A gel-based non-instrumental immuno-affinity assay was developed for the rapid screening of chloramphenicol (CAP) in food samples with the limit of detection (LOD) of 1 μg L−1. The immuno-affinity test column (IATC) consisted of a test layer containing anti-CAP antibody coupled gel, and a control layer with anti-HRP antibody coupled gel. Based on the direct competitive immuno-reaction and the horseradish peroxidase enzymatic reaction, the test results could be evaluated visually. Basically, blue color development represented the negative results, while the absence of color development represented the positive results. In this study, CAP spiked samples of raw milk, pasteurized milk, UHT milk, skimmed milk powder, acacia honey, date honey, fish and shrimp were tested. Little or none sample pretreatment was required for this assay. The whole procedure was completed within 10 min. In conclusion, the gel-based immuno-affinity test is a simple, rapid, and promising on-site screening method for CAP residues in food samples, with no instrumental requirement.  相似文献

This paper reports a flow-injection analysis (FIA) of glucose not using enzyme based on the electrocatalytic oxidation of glucose at a nickel electrode. The electrocatalytic mechanism and quantificational method of glucose have been investigated. The current intensity of the electrocatalytic oxidation to glucose at the potential of 550 mV is proportional to the concentration of glucose over the range of 0.10-2.50 mmol l−1, with a 0.04 mmol l−1 detection limit (S/N = 3) and a correlation coefficient of 0.9991. The relative standard deviation (R.S.D.) is less than 4.3% (n = 5) for the determination of practical serum samples. The biologic compounds probably existed in the sample, such as ascorbic acid, uric acid, dopamine and epinephrine, do not disturb the determination of glucose. The result is satisfactory for the determination of glucose in human serum sample as comparison to that from the routine hexokinase method.  相似文献

A single automatic method for continuous flow determination of β-naphthol based on the enhancement of its native fluorescence once the analyte was transitorily retained on-line on a solid support (QAE A-25 resin) is reported. So, a flow-through optosensor was developed using a flow-injection analysis system with solid phase fluorimetric transduction. KCl (0.15 mol l−1) at pH 12.0 was used as carrier solution. To obtain the optimum fluorescence signal the wavelengths chosen were 245 nm (excitation) and 420 nm (emission). The response of the sensor was directly proportional to the sample volume injected in the studied range 40-1500 μl. Approximately one higher order of magnitude is achieved in sensitivity when 1500 μl are used with respect to the use of 40 μl of sample. The sensor was calibrated for three different injection volumes: 40, 600 and 1500 μl, responding linearly in the measuring range of 2-60, 0.5-15 and 0.2-5 μg l−1 with detection limits of 0.5, 0.09 and 0.05 μg l−1, respectively. The relative standard deviation for ten independent determination is 0.6% (40 μl), 0.9% (600 μl) and 2.3% (1500 μl). A recovery study was performed onto three different spiked water samples at concentration levels from 1 to 2.5 μg l−1 and the recovery percentage from the experimental data ranged between 101±2 and 105±5.  相似文献

A new method for sensitive and fast screening of melamine (MEL) in milk products was developed with a low-cost disposable microfluidic device coupled with ultraviolet (UV) detection. This method avoided the need of sample pretreatment prior to the separation process, thus was simple and green. Due to the advantages of the device and fracture sampling technique, milk sample could be directly sampled through the fracture to achieve baseline separation from amino acids, and proteins in the sample did not interfere with the detection. Using 20 mM phosphate running buffer (pH 9.0), a sampling time of 3 s at +180 V and a separation voltage of +1800 V (240 V/cm), this method could detect MEL in milk within 75 s. At the detection wavelength of 202 nm, the linear range for MEL was from 1.0 to 100 μg mL−1 with a detection limit of 0.23 μg mL−1 (S/N = 3). The novel protocol had been successfully used to screen MEL in milk samples with recovery more than 82%. The environmentally friendly methodology for pretreatment-free sensitive screening of MEL provided promising applications in monitoring the quality of different foods.  相似文献

The use of membranes for on-line separations in flow-through dialyser as a part of a flow system is extremely useful for automated samples preparation. In this paper a method to couple the dialysis and the UV detection on-line of Vitamin B12 from milk is proposed. Firstly, the milk samples were pre-treated with trichloroacetic and centrifuged (to eliminate proteins and fats) and later, using a dialyser coupled a flow-continuous manifold was possible dialyse the Vitamin B12, which was monitored spectrophotometrically at 361 nm. On the other hand, the milk samples were also dialysed on-line but without the acid treatment and the results were compared. The influence of various parameters, including the pump speed for both the donor and acceptor stream, dialysis time, donor and acceptor loop volume on dialysis efficiency was studied. The method was applied to different kinds of milk (skimmed and semi-skimmed milk, evaporated milk, lactose free milk and liquid and powder whole milk). The relative standard deviation (R.S.D.) of the proposed method was of 0.45% and the obtained dialysis percentage was of 5.8%. The proposed method very easy permit a pre-treatment of the sample, quick and on-line with the detection. The dialysis process permitted the pass of vitamin and avoided the pass of other analytes as proteins in the case of the milk samples without acid treatment.  相似文献

This paper reports the development of a new strategy for low-level determination of copper in water samples by using a flow-injection system coupled to solid-phase extraction (SPE) using flame atomic absorption spectrometry (F AAS) as detector. In order to preconcentrate copper from samples, a minicolumn packed with a styrene-divinylbenzene resin functionalized with (S)-2-[hydroxy-bis-(4-vinyl-phenyl)-methyl]-pyrrolidine-1-carboxylic acid ethyl ester was used and the synthesis procedure is described. System operation is based on the on-line retention of Cu(II) ions at pH 9.0 ± 0.2 in a such minicolumn with posterior analyte elution with 2 mol l−1 HCl directly to the F AAS nebulizer. The influence of several chemical (sample pH, buffer concentration, HCl eluent concentration and effect of the ionic strength) and flow (sample and eluent flow rates and preconcentration time) variables that could affect the performance of this system were investigated as well as the possible interferents. At optimized conditions, for 2 min of preconcentration time (13.2 ml of sample volume), the system achieved a detection limit of 1.1 μg l−1, a R.S.D. 1% at 20 μg g l−1 and an analytical throughput of 25 h−1, whereas for 4 min of preconcentration time (26.4 ml of sample volume), a detection limit of 0.93 μg l−1, a R.S.D. 5.3% at 5 μg l−1 and a sampling frequency of 13 h−1 were reported.  相似文献

Fabrication of a macro segmented flow analysis (MSFA) system based on reconfiguration of the manifold by adjustment of the sample/reagent ratio, has been found to produce a sensitive method for orthophosphate analysis based on colorimetric detection at 880 nm. Optimization of sample tube length, reaction temperature and molybdate concentration in the carrier solutions has been carried out. The larger sample tube internal diameter led to the combined advantages of better sensitivities, wider working range and higher sample throughput over most existing methods. Using the optimized conditions of 50.0 cm sample tube length (1.6 mm i.d.), 37.0 °C reaction temperature and 0.0113 M molybdate concentration in the carrier solution, the calibration model for orthophosphate standard solutions was found to be linear (y = 0.04895x + 0.003561; correlation coefficient, r2=0.9970) over the working range 0.01-2.00 mg l−1 orthophosphate. The volume of the sample injected was 1.396 ml at a flow rate of 6.0 ml min−1. The sample throughput of this MSFA method was 40 samples per an hour, with a detection limit of 4.0 μg l−1, and %R.S.D.’s below 5%. The MSFA method was successfully applied to analysis of water and wastewater samples.  相似文献

To address people's concerns of exogenous oxytocin (OT) administration to lactating bovines, a study was undertaken to (a) establish an enzyme immunoassay (EIA) for OT determination in milk, (b) quantify OT in milk of cows administered OT, and (c) study influence of pasteurization on OT stability in milk. A sensitive EIA validated according to the criteria of European Union—Decision 2002/657/EC was developed for OT in skim milk in an analytical range of 10-250 pg mL−1 with a decision limit (CCα) of 30 pg mL−1 and detection capability (CCβ) of 41.5 pg mL−1. Milk samples collected from cows (n = 38) administered either 25 or 50 IU OT prior to milking were investigated for the presence of OT. There was no significant difference among both groups with the mean concentrations of OT being 15.8 and 14.9 pg mL−1 for cows subjected to 25 and 50 IU OT administration, respectively. The OT levels in skim milk of control cows (n = 30; untreated) were basal (around 10 pg mL−1). All the analyzed milk samples were below the CCα value of 30 pg mL−1. Pasteurization of OT spiked milk samples at different temperature and sample holding conditions reduced the immunological activity of OT to 43% at 110 °C. However, no further decline occurred in the immunological activity with increased pasteurization temperature and time. It was concluded that the milk OT concentrations after OT administrations were minimal and below the assay decision limit. However, OT was quite stable to pasteurization in OT spiked milk.  相似文献
