Isotactic 6-armed star-shaped poly(vinyl alcohol) (PVA) with a narrow molecular weight distribution was successfully prepared by the living cationic polymerization of 6-armed star-shaped poly(tert-butyl vinyl ether) (PTBVE) and subsequent acidic ether cleavage. The PTBVE was synthesized using hexa(chloromethyl) melamine (HCMM) as a hexafunctional initiator and ZnI2 or ZnCl2 as an activator in toluene/MC (1/1 v/v) at −70 °C. A better living stability of PTBVE was obtained in the ZnCl2 activator system. The number average molecular weight and the polydispersity index of the 6-armed star-shaped PTBVE polymerized with ZnCl2 at −70 °C for 24 h were 156,000 g/mol and 1.47, respectively. The fraction of the mm sequence of the resulting PVA was 52%.  相似文献

We focus on the role of water in a protein-resistant poly(ethylene glycol) (PEG) layer. Using the combination of two experimental techniques, namely, the extended surface forces apparatus and the quartz crystal microbalance, we demonstrate that the water content inside these surface-grafted layers is over 80 vol % while the conformational space of the PEG chains is significantly modulated in water. Discrete and reversible film thickness transitions of 1.25 A size are shown to occur when the film is compressed, a finding that suggests a high degree of organization in the PEG/water complex. The results are discussed in terms of the excellent protein resistance properties of this type of surface.  相似文献

In the present research, the low water flux of polyphenylsulfone membranes was addressed, and a novel improvement in their water permeation and fouling resistance was achieved using polyethylene glycol (PEG) as the hydrophilic additive. Scanning electron microscopy and field emission scanning electron microscopy, atomic force microscopy, attenuated total reflection Fourier‐transform infrared spectroscopy, thermogravimetric analysis, and tensile test were applied for the investigation of membrane morphology, surface topography, surface chemical structure, thermal stability, and mechanical properties, respectively. Moreover, the relative hydrophilicity/hydrophobicity of the membranes was assessed via determination of membrane water uptake capacity and water contact angle. The membrane performance was studied and compared by determination of pure water flux and filtration of canned beans production wastewater as well as bovine serum albumin solution. The filtration results indicated a remarkable pure water flux and 100% turbidity rejection provided by the polyphenylsulfone/PEG 20 000 membrane. In addition, it was confirmed that the amount of residual PEG within the membrane was increased with increasing PEG molecular weight and concentration.  相似文献

The cationization of poly(ethylene glycol)s, PEG 4000 and PEG 6000, under matrix-assisted laser desorption/ionization conditions was studied by using different concentration ratios of the sodium ion, as the reference ion, and another alkali metal ion (Li(+), K(+), Rb(+), Cs(+)). A linear correlation was found between the intensity ratio of the sodiated PEGs and PEGs cationized with alkali metal ions versus the initial concentration ratio of sodium and alkali metal ions. The slopes of these straight lines are proposed as a novel selectivity ratio for the ionization process. The intensity distribution of the cationized PEGs was also investigated. It was found that the cationized oligomers follow Poisson statistics. The M(n) and M(w) values were also evaluated. An explanation for the observed effects is given.  相似文献

Two reaction schemes were developed to covalently graft poly(ethylene glycol) (PEG) chains on poly(ethylene-co-acrylic acid) (EAA) surfaces. The schemes involved surface grafting of linker molecules L-lysine or polypropyleneamine dendrimer (AM64), with subsequent covalent bonding of PEG chains to the linker molecules. NHS and EDC were used to activate the carboxylic acid groups of the EAA in the outermost region of the film, estimated to be 20 nm by ATR-FTIR spectroscopy. XPS demonstrated that the conversion of this activation step was almost 100% in the detected region. After activation, L-lysine or dendrimer was grafted onto the EAA surface, followed by PEG grafting. Combining the data from ATR-FTIR, XPS, and contact angle goniometry, it was found that the PEG chains were grafted on the surface of the EAA film and larger surface coverage was achieved when the dendrimer was used as the intermediate layer. This surface also had the lowest water contact angle.  相似文献

At high magnetic fields the 128.8 MHz (195)Pt NMR of all the species in the series [PtCl(n)(H(2)O)(6-n)](4-n) (n = 2-6) display unique (35/37)Cl isotope effects resulting in a unique 'fine-structure' of each individual resonance, which constitutes an unambiguous spectroscopic 'fingerprint' characteristic of the structure of the octahedral platinum(iv) complex, provided (195)Pt NMR are recorded at optimum magnetic field homogeneity and carefully controlled temperature (293 ± 0.1 K). The detailed (195)Pt resonance fine-structure observed experimentally can readily be accounted for by an isotopologue and isotopomer model for each complex, showing particularly noticeable differences between stereoisomer pairs such as the cis/trans- and fac/mer-complexes. Moreover partial isotopic (18)O enrichment of the coordinated water molecules in the series [Pt(35/37)Cl(n)(H(2)(16/18)O)(6-n)](n-2) (n = 2-6) confirms this model. This technique can thus be considered a novel, direct spectroscopic method of chemical speciation of appropriate platinum(iv) complexes in solution without reference to accurate chemical shifts of authentic members of such a series. These effects are interpreted qualitatively in terms of the high sensitivity of (195)Pt NMR shielding to very small and subtle Pt-(35/37)Cl and Pt-(16/18)OH(2) bond displacements. Preliminary work shows this also applied to the corresponding bromido-complexes.  相似文献

We prepared surface-grafted polystyrene (PS) beads with comb-like poly(ethylene glycol) (PEG) chains. To accomplish this, conventional gel-type PS beads (35-75 microm) were treated with ozone gas to introduce hydroperoxide groups onto the surface. Using these hydroperoxide groups, poly(methyl methacrylate) (PMMA, Mn= 22,000-25,000) was grafted onto the surface of the PS beads. The ester groups of the grafted PMMA were reduced to hydroxyl groups with lithium aluminum hydride (LAH). After adding ethylene oxide (EO) to the hydroxyl groups, we obtained the PS-sg-PEG beads, which had a rugged surface and a diameter of 80-150 microm. We could obtain several kinds of the PS-sg-PEG beads by controlling the chain lengths of the grafted PMMA and the molecular weights of the PEG chains. The grafted PEG layer was about 30-50 microm thick, which was verified from the cross-sectioned views of the fluorescamine-labeled beads. These fluorescence images proved that the beads possessed a pellicular structure. Furthermore, we found that the surface-grafted PEG chains had the characteristic property of reducing non-specific protein adsorption on the beads.  相似文献

Shortage or malfunction of pulmonary surfactant in alveolar space leads to a critical condition termed respiratory distress syndrome (RDS). Surfactant replacement therapy, the major method to treat RDS, is an expensive treatment. In this paper, the effect of poly(ethylene glycol) (PEG) to improve dynamic surface activity of a bovine lipid extract surfactant (BLES) was studied by axisymmetric drop shape analysis (ADSA) and a captive bubble method. The activity of BLES+PEG mixtures was compared to that of a natural surfactant containing surfactant proteins A and D. When PEG was added into BLES mixtures, the surface tension hysteresis of BLES films was minimized when the films were compressed by more than 50%. PEG also helps to quickly restore surfactant films after film collapse. Thus, as far as surface tension effects go, the findings suggest that PEG might be used as a substitute for surfactant-associated protein SP-A in therapeutic surfactant products, and might also be used to reduce the amount of BLES required in clinical applications.  相似文献

Novel poly(ethylene glycol) (PEG) and sulfonated PEG (PEG-SO3) acrylate copolymers have been prepared and characterized to apply as coating and blending materials for biomedical applications. The modified surfaces using acrylate copolymers demonstrated increased hydrophilicity, possibly due to the hypothesized reorientation of PEG/PEG-SO3 chains into water phase. All copolymer surfaces demonstrated less platelet adhesion than control. In addition, platelet adhesion on copolymer surfaces decreased as the chain length of PEG and sulfonated PEG in copolymers increases. All copolymer surfaces reduced bacterial adhesion significantly and the adhesion level differs depending on surfaces as well as media. The obtained results attest to the usefulness of these copolymers as a coating or additive material to improve the blood compatibility of blood contacting devices.  相似文献

Although poly(ethylene glycol) (PEG) has been widely used as an agent to induce cell aggregation and fusion, the physicochemical principles of its function are only becoming understood recently. PEG has an extremely high affinity for water. The PEG commonly used for these applications is in the molecular weight range of 8000 to 10 000. At low concentrations (0–15 wt.%), PEG in this molecular weight range tends to deplete from cell or lipid surfaces, creating an osmotic gradient which brings cells or lipid vesicles together. The depletion force is measured using a surface force apparatus. The corresponding reduction of surface viscosity is verified by shear viscosity measurements and by vesicle tumbling experiments. At higher concentrations (15–45 wt.%), the extremely high osmotic pressure generated by PEG compresses apposing surfaces of aggregated cells or vesicles to within limits where the membrane is no longer stable, and fusion occurs at point defects. A fusion lumen is formed with the help of cell swelling. If PEG is adsorbed or covalently link to the cell or vesicle surface, the surface force profile becomes entirely repulsive, and aggregation and fusion is inhibited. The repulsion is accountable by steric and electrostatic forces. Therefore, the fusogenic function of PEG can be explained quantitatively by colloidal stability theories.  相似文献

The interaction of lithium perfluoroheptanoate (LiPFHep) with poly(ethylene glycol) (PEG) of different molecular weights (300 < MW < 20 000 Da) was investigated in water at 298.15 and 308.15 K by the isothermal titration calorimetry (ITC). Density and sound velocity measurements were also performed at 288.15, 298.15, and 308.15 K, while viscosity and conductivity data were only collected at 298.15 K. The aggregation process of this surfactant on the PEG polymeric chain was found to be very similar to the process exhibited by the two homologous perfluorooctanoate and perfluorononanoate. Viscosity and ITC data indicated that the formation of polymer-surfactant complexes between PEG and LiPFHep also leads to a conformational change in the polymer. The aggregation of micelles of the lithium perfluoro surfactants on the PEG polymeric chain is characterized by a comparable thermodynamic stability, which results from a balance of enthalpy and entropy contributions, which both increase with the length of the surfactant hydrophobic chain.  相似文献

This work describes studying the permanent grafting of carboxylic acid end-functionalized poly(ethylene glycol) methyl ether (PEG) chains of different molecular weights from the melt onto a surface employing poly(glycidyl methacrylate) ultrathin film as an anchoring layer. The grafting led to the synthesis of the complete PEG brushes possessing exceptionally high grafting density. The maximum thickness of the attached PEG films was strongly dependent on the length of the polymer chains being grafted. The maximum grafting efficiency was close to the critical entanglement molecular weight region for PEG. All grafted PEG layers were in the "brush regime", since the distance between grafting sites for the layers was lower than the end-to-end distance for the anchored macromolecules. Scanning probe microscopy revealed that the grafting process led to complete PEG layers with surface smoothness on a nanometric scale. Practically all samples were partly or fully covered with crystalline domains that disappeared when samples were scanned under water. Due to the PEG hydrophilic nature, the surface with the grafted layer exhibited a low (up to 21 degrees ) water contact angle.  相似文献

A novel synthesis of poly(ethylene glycol) (PEG)-grafted poly(urethanes) (PURs) is described based on a precursor PUR containing free amino groups in the main chain. Three different poly(urethane) backbones were prepared: a homopoly(urethane) comprised of N-Bocdiethanolamine (BDA) and 4,4′-methylenebis(phenyl isocyanate) (MDI), a copoly(urethane) (COPUR) consisting of BDA, N-benzyldiethanolamine and MDI, and a poly(urethane urea) (PUU) that was prepared from BDA, MDI, and ethylenediamine as the chain extender. The Mn of these poly(urethanes) ranged from 32,000 to 72,000 g/mol. PEG (750, 1,900, and 5,000 g/mol) was grafted onto the boc-deprotected poly(urethanes) via the chloroformate. Films of the polymers were spin cast from dilute solutions, annealed, and the surfaces analyzed by goniometry. Water contact angle data indicates increasing PEG surface coverage of the poly(urethanes) with increasing PEG molecular weight. Reorientation of the polymer films is evidenced by contact angle hysteresis. Polymer thrombogenicity, which was studied using blood perfusion experiments, shows that COPUR-g-PEG5000 and PUU-g-PEG5000 exhibit very little platelet adhesion. © 1999 John Wiley & Sons, Inc. In this work we report the structural characteristics of bovine serum albumin/poly(ethylene glycol) lipid conjugate (BSA/PEG(2000)-PE) complexes under physiological conditions (37 degrees C and pH 7.4) for particular fractions of BSA to PEG-lipid concentration, c(BSA)/c(PEG)(2000)-PE. Ultraviolet fluorescence spectroscopy (UV) results shown that PEG(2000)-PE is associated to BSA, leading to protein unfolding for fixed c(BSA) = 0.01 wt % and variable c(PEG)(2000)-PE = 0.0015-0.6 wt %. Tryptophan groups on the BSA surface are in contact with the PEG-lipid at c(PEG)(2000)-PE = 0.0015 wt %, while they are exposed to water at c(PEG)(2000)-PE > 0.0015 wt %. Dynamic and static light scattering (DLS and SLS) and small-angle neutron scattering (SANS) point out the existence of individual BSA/PEG-lipid complexes in the system for fixed c(BSA) = 1 wt % and variable c(PEG)(2000)-PE = 0.15-2 wt %. DLS shows that there is only one BSA molecule per protein/PEG-lipid complex, while SLS shows that the PEG-lipid associates to the BSA without promoting aggregation between adjacent protein/polymer-lipid conjugate complexes. SANS was used to show that BSA/PEG(2000)-PE complexes adopt an oblate ellipsoidal shape. Partially unfolded BSA is contained in the core of the oblate ellipsoid, which is surrounded by an external shell containing the PEG(2000)-PE.  相似文献

We prepared pure and mixed monolayers of methoxy-terminated poly(ethylene glycol)s (m-PEG's) chemically attached to silica surfaces by using m-PEG silane coupling agents of three different molecular weights. These films were subsequently characterized in water by atomic force microscopy (AFM). Images of pure m-PEG monolayers showed the formation of polymer brushes on silica. Force curves between two modified surfaces suggested that an increase in the number of oxyethylene (OE) groups from 6 (PEG6 surface) to 43 (PEG43 surface) to 113 (PEG113 surface) decreased the flexibility of the m-PEG chains in the m-PEG brushes. Frictional force measurements also showed that the friction increased in the order PEG6 < PEG43 相似文献

Hyperbranching poly(allylamine) (PAAm) and poly(ethylene glycol) (PEG) on silicon and its effect on protein adhesion was investigated. Hyperbranching involves sequential grafting of polymers on a surface with one of the components having multiple reactive sites. In this research, PAAm provided multiple amines for grafting PEG diacrylate. Current methodologies for generating PEG surfaces include PEG-silane monolayers or polymerized PEG networks. Hyperbranching combines the nanoscale thickness of monolayers with the surface coverage afforded by polymerization. A multistep approach was used to generate the silicon-supported hyperbranched polymers. The silicon wafer surface was initially modified with a vinyl silane followed by oxidation of the terminal vinyl group to present an acid function. Carbodiimide activation of the surface carboxyl group allowed for coupling to PAAm amines to form the first polymer layer. The polymers were hyperbranched by grafting alternating PEG and PAAm layers to the surface using Michael addition chemistry. The alternating polymers were grafted up to six total layers. The substrates remained hydrophilic after each modification. Static contact angles for PAAm (32-44 degrees) and PEG (33-37 degrees) were characteristic of the corresponding individual polymer (30-50 degrees for allylamine, 34-42 degrees for PEG). Roughness values varied from approximately 1 to 8 nm, but had no apparent affect on protein adhesion. Modifications terminating with a PEG layer reduced bovine serum albumin adhesion to the surface by approximately 80% as determined by ELISA and radiolabel binding studies. The hyperbranched PAAm and PEG surfaces described in this paper are nanometer-scale, multilayer films capable of reducing protein adhesion.  相似文献

We study ternary mixtures of nonionic surfactants C(i)E(j) (i = 12; j = 5, 6, 8) and poly(ethylene glycol) (PEG) in water. For sufficiently large molecular mass of PEG (M >M(sep) approximately 600), we observe a lowering of phase separation temperature with an increase in polymer concentration. The value of M(sep) is consistent with the analysis based on depletion interactions between micelles induced by polymer chains. We also demonstrate that there is another critical molecular mass of PEG (M = M* approximately 2000) necessary to induce ordering in the surfactant-rich phase. This critical molecular mass follows from two requirements: (a) PEG has to reduce the separation temperature below a temperature of hexagonal-isotropic phase transition in a binary surfactant-water mixture and (b) the PEG radius of gyration has to be larger than the size of the water channels in the hexagonal phase.

Monodisperse magnetite nanoparticles modified with poly(ethylene glycol) (PEG) were synthesized using a silane functionalized PEG obtained by reacting 3-aminopropyl triethoxysilane with carboxylic acid-methoxy PEG (mPEG-COOH) using amide reactions. Transmission electron microscopy (TEM), dynamic light scattering (DLS), and zeta potential measurements show the particles are monodisperse (sigma(gv) approximately 0.2) and stable in water for pH of 3-9 and ionic strengths, up to 0.3 M NaCl. Thermogravimetric analysis coupled with TEM and DLS indicates formation of a dense graft layer on the particle surface. An analysis of the interparticle interaction energy indicates that the particles are stabilized by strong steric repulsions between PEG chains on their surface.  相似文献
