Several derivatives have been synthesized from chrysin, diosmetin, apigenin, and luteolin, which were isolated from diverse natural plants. The α-glucosidase inhibitory activity of these compounds was evaluated. The glucosidase inhibitory activity of all derivatives (IC50 〈 24.396 μmol]L) was higher compared with that of the reference drug, acarbose (IC50=563.601 ±40.492μmol/L), and 1- deoxynojirimycin (IC50 = 226.912± 12.573 μmol/L). O3',O7-Hexyl diosmetin (IC50 = 2.406 ± 0.101μmol/L) was the most potent inhibitor identified. These compounds showed a higher inhibitory ability compared with their precursors except the luteolin derivatives. In general, the inhibitory activity of the synthetic derivatives was enhanced with long alkyl chains at positions 3', 4' and 7 of the flavonoid.  相似文献

A series of novel 1,2,3-benzotriazin-4-one derivatives were designed,synthesized and their inhibitory activities against leulcotriene A_4 hydrolase aminopeptidase in vitro were evaluated.Many compounds showed moderate to good activities at the concentration of 10 μmol/L.Among them,compound Ⅳ-16 exhibited the highest inhibitory activity up to 80.6% with an IC_(50) of 1.30 ± 0.20 μmol/L The compound Ⅳ-16 was also tested the proliferation inhibitory activities in THP1 human AML cell line and its binding model with LTA_4H enzyme by molecular docking was studied.It indicated that 1,2,3-benzotriazin-4-one was a promising scaffold for further study.The relationship between structure and inhibitory activity was also preliminarily discussed.  相似文献

Lithocarpinols A(1) and B(2), a pair of tenellone diastereoisomers with novel fused skeleton were isolated from the deep-sea derived fungus Phomopsis lithocarpus FS508. Their structures were elucidated by comprehensive spectroscopic analyses, X-ray diffraction and quantum molecular calculation. Their plausible biogenetic pathway featured an intriguing carbonyl-ene cyclization. Lithocarpinol A exhibited moderate inhibitory effect against HepG-2 and A549 tumor cell lines with IC_(50) values of 9.4 μmol/L and10.9 μmol/L,respectively.  相似文献

Novel purine-2,6-diamine derivatives were designed and synthesized as cyclin-dependent kinase (CDK) inhibi- tors. According to the preliminary biological evaluation, most of the compounds show good inhibitory activities in CDK1 enzyme assay and potent antiproliferative activities in some tumor cell lines. Nine 5-aryl-2-methyloxazole derivatives were synthesized via gold-catalyzed alkyne oxidation. All of the compounds have been screened for their antiproliferative activities against MCF-7 cell (human breast carcinoma), A549 cell (human lung carcinoma) and Hela cell (human cervical carcinoma) lines in vitro. The results revealed that compounds 1b, 1c and 1d exhibited strong inhibitory activities against the MCF-7 cell lines (with ICso values of 4.6, 9.7 and 2.2 μmol/L, respectively).  相似文献

Twelve S-substituted 7-mercapto-4-methylcoumarin analogs were synthesized and evaluated for the inhibition to HBV in HepG2 2.2.1.5 cell. Among them, ten compounds exhibited potent inhibition to HBsAg and/or HBeAg with the IC50 values of sub μmol/L level. The ICso of anti-HBsAg activities of 5c and 5j reached 0.01 μmol/L respectively which were 16 fold more potent than that of 3TC. Compounds 3, 5e, 5g, 5h and 5i showed admirable inhibitory activity to both HBsAg and HBeAg. The bioassay results indicated the S-substituted 7-mercapto-4-methylcoumarin analogs merit attention as novel anti-HBV agents.  相似文献

Bis-thiobarbiturate derivatives 1–15 have been synthesized, characterized by1 HNMR and EI-MS and screened for urease inhibition. All compounds showed various degree of urease inhibitory activity with IC_(50) values ranging 7.45 0.12 74.24 0.81 mmol/L while the standard thiourea behaved normally(IC_(50) = 21.10 0.12). Compounds 1(IC_(50) = 7.45 0.12 μmol/L), 9(IC_(50) = 18.17 1.03 μmol/L) and 13(IC_(50) = 8.61 0.45 μmol/L) showed excellent urease inhibitory activity in the series. Molecular modeling studies were performed to understand the binding site with the bimetallic nickel center of the enzyme.Structure-activity relationship has also been established for these compounds. This study identified bisthiobarbiturate as a novel class of urease inhibitors.  相似文献

A novel series of ethyl 5-hydroxy-4-substituted aminomethyl-2-sulfinylmethyl-1H-indole-3-carboxylates 8a―8j and 11e―11f was synthesized and evaluated in HepG2.2.15 cells for their anti-hepatitits B virus(HBV) acti-vity and cytotoxicity.Among them,six compounds showed more potent inhibitory activity than lamivudine.Compound 8e exhibited the most significant anti-HBV activity with an IC50 value of 1.62 μmol/L,which was 33-times more potent than the reference drug lamivudine(IC50=54.78 μmol/L).  相似文献

Nine ethyl N-hydroxy-N-(substituted)phenyloxamates(2a—2i), based on the structure of the KARI inhibitor IpOHA, were synthesized. Their structures were established on the bases of 1H NMR, IR, ESI-MS and elemental analyses. Among the nine compounds, four show in vitro inhibitory activity on rice KARI, with 2c and 2g[Ki values of (35.2±4.9) and (49.4±6.3) μmol/L] having similar activity to the precursor of IpOHA(A, Ki=34.1±6.7 μmol/L). The results will be helpful to further molecular design of more potent KARI inhibitors.  相似文献

One new indoloditerpene,6,7-dehydropaxilline(1),one new indole-diketopiperazine,spirotryprostatin F(2),and one new 13-membered macrolide,N-demethylmelearoride A(3),as well as 8 known alkaloids(4-11),were isolated from the solid cultures of the endophytic fungus,Penicillium brefeidianum.The structures and absolute configurations of compounds 1-3 were elucidated by comprehensive spectroscopic analysis and the circular dichroism(CD) exciton chirality method.All the metablites were evaluated for their cytotoxic activites against three human tumor cell lines.Compound 2 exhibited cytotoxicities against HepG2 and MDA-MB-231 cells with IC_(50) values of 14.1 μmol/L and 35.5μmol/L,respectively.Compound 3 showed moderate activity against HepG2 cells with IC_(50) values of 36.6 μmol/L.  相似文献

A series of naphthyridinone derivatives based on la(a precursor of Voreloxin) were designed and synthesized.Seven compounds having 70%inhibition against HL60 at 30 μmol/L were further evaluated for their in vitro antitumor activity by SRB assay.Results reveal that thiazol-2-yl and 3-aminomethyl-4-benzyloxyimino-3-methylpyrrolidin-l-yl groups are optimal at the N-1 and C-7positions of naphthyridinone core,respectively.10 j exhibits broad-spectrum activity(IC_(50):0.5-6.25 μmol/L) against all of the tested cell lines including Etoposide- and/or la-resistant ones,and is 1.3-fold to 100-fold more potent than the two references against eight of these cell lines.  相似文献

The miniature endplate currents (MEPC's) were recorded at the neuromuscular junction of Xenopus laevis embryo neuron-muscle co-cultured cells. These MEPC's were due to the spontaneous release of acetylcholine from the nerve terminal. After perfusion with glutamate (10 μmol/L), both frequency and amplitude of the MEPC's increased. After washing away of glutamate, this effect persisted. We named this phenomena "Long-Term Facilitation".GABA (20 μmol/L) on the other hand had an inhibitory effect on both frequency and amplitude of the MEPC's. After washing away of GABA, the MEPC frequency and amplitude increased. We named this effect "Post-Potentiation". Local perfusion experiments furthermore indicated that the effect of glutamate was restricted to the neuromuscular junction, the effect of GABA was restricted to the soma.  相似文献

SARS CoV 3CL^pro is known to be a promising target for development of therapeutic agents against the severe acute respiratory syndrome (SARS). A quinolinone compound 1 was selected via virtual screening, and it was syn- thetized and tested for enzymatic inhibition in vitro. Compound 1 showed potent inhibitory activity (ICs0= 0.44 μmol/L) toward SARS CoV 3CL^pro. Further work on a series of quinolinone derivatives resulted in the discovery of the most potent compound 23, inhibiting SARS CoV 3CL^pro with an IC50 of 36.86 μmol/L. Bistachybotrysins D and E (1 and 2), one stereoisomeric pair of phenylspirodrimane dimers, were isolated from Stachybotrys chartarum CGMCC 3.5365. They represent novel phenylspirodrimane dimers with a central [6,5,6]-tricyclic carbon scaffold containing a cyclopentanone core. The structures of 1 and 2 were elucidated through extensive spectroscopic data analysis, and their absolute configurations were characterized by calculated electronic circular dichroism (ECD). Compounds 1 and 2 displayed potent cytotoxic activity against the four human tumor cell lines HCT116, BGC823, Daoy and HepG2 with IC_ (50)values ranging from 6.7 μmol/L to 11.6 μmol/L. 75%aqueous ethanol extract from the whole herbs of Selaginella doederleinii was isolated,and two new apigenin derivatives,doederflavones A(1) and B(2),together with ten known compounds(3-12) were characterized.Their structures were assigned by extensive spectroscopic methods including 1D/2D NMR and HR-ESIMS.Compounds 1-6 bear an aryl substituent at the C-8 or C-6 positions in ring A of apigenin skeleton.Compounds 1 and 2 were evaluated for their in vitro cytotoxicity against four human cancer cell lines A549,MCF-7,SMMC-7721,and LoVo,both of which exhibited significant cytotoxicity against A549 with IC_(50) values of 0.82 μmol/L and 1.32 μmol/L,respectively.  相似文献

The application of p-aminophenol as a suitable mediator, as a sensitive and selective voltammetric sensor for the determination of hydrazine using square wave voltammetric method were described. The modified multiwall carbon nanotubes paste electrode exhibited a good electrocatalytic activity for the oxidation of hydrazine at pH = 7.0. The catalytic oxidation peak currents showed a linear dependence of the peaks current to the hydrazine concentrations in the range of 0.5–175 μmol/L with a correlation coefficient of 0.9975. The detection limit (S/N = 3) was estimated to be 0.3 μmol/L of hydrazine. The relative standard deviations for 0.7 and 5.0 μmol/L hydrazine were 1.7 and 1.1%, respectively. The modified electrode showed good sensitivity and selectivity. The diffusion coefficient (D = 9.5 × 10–4 cm2/s) and the kinetic parameters such as the electron transfer coefficient (α = 0.7) of hydrazine at the surface of the modified electrode were determined using electrochemical approaches. The electrode was successfully applied for the determination of hydrazine in real samples with satisfactory results.  相似文献

New isoconessimine derivatives were synthesized from conessine (1) and evaluated as acetylcholinesterase (ACHE) inhibitors. The derivatives were prepared via two reaction steps, N-demethylation and nuc]eophilic substi- tution. All of the synthesized derivatives exhibited more potential anti-acetylcholinesterase activities than conessine (1) (IC50=16μmol·L^-1) and isoconessimine (2) (IC50〉300 μmol·L ^-1). Compound 7b (3fl-[methyl-[2-(4-nitro- phenoxy)ethyl]amino]con-5-enine) showed the most potent inhibitory activity with an IC50 of 110 μmol/L which is close to that of reference compound huperzine A (IC50= 70 μmol/L). The mode of AChE inhibition by 7h was re- versible and non-competitive. In addition, molecular modeling was performed to explore the binding mode of in- hibitor 7b at the active site of AChE and the results showed that 7b could be docked into the acetylcholinesterase active site and compound 7h had hydrophobic interactions with Trp279 and Leu282.  相似文献
