以二甲基硅油接枝端羟基聚氧乙烯(PDMS g PEO OH)为基材,用二环己基碳二亚胺(DCCI)作脱剂,研究了羟基(OH)与肝素上的羧基(—COOH)之间的脱水缩合反应,制备出肝素化的抗血栓材料PDMS g PEO Hep,并对其涂覆表面的肝素含量和体外抗凝血性能进行了初步评价.实验结果表明,肝素接枝的共聚物具有优良的抗凝血性能和一定的应用前景.  相似文献

A procedure for the preparation of new block copolymers composed of a hydrophobic block of polystyrene, a hydrophilic spacer-block of poly(ethylene oxide) and a bioactive block of heparin was investigated. Polystyrene with one amino group per chain was synthesized by free radical oligomerization of styrene in dimethylformamide, using 2-aminoethanethiol as a chain transfer agent. This amino group was used in the coupling reaction with amino-telechelic poly(ethylene oxide) to produce an AB type diblock copolymer with one amino group per polystyrene (PSt)–poly(ethylene oxide) (PEO) chain. The amino-semitelechelic oligo-styrene was converted into the isocyanate-semitelechelic oligo-styrene using toluene 2,4-diisocyanate and subsequent coupling with H2N–PEO–NH2 afforded AB type block copolymers with terminal amino groups. The coupling of PSt–PEO–NH2 with heparin was performed in a DMF–H2O mixture, first by activating the heparin carboxylic groups with EDC at pH 5.1–5.2 and subsequently reacting the activated carboxylic groups with the amino groups of the PSt–PEO–NH2 at pH 7.5. Depending on the molecular weights of the diblock copolymer used 25–29% w/w heparin was incorporated. These polymers will be further evaluated for their blood-compatibility.  相似文献

Polyurethane (PU) was modified using isocyanate chemistry to graft polyethylene oxide (PEO) of various molecular weights (range 300-4600). An antithrombin-heparin (ATH) covalent complex was subsequently attached to the free PEO chain ends, which had been functionalized with N-hydroxysuccinimide (NHS) groups. Surfaces were characterized by water contact angle and X-ray photoelectron spectroscopy (XPS) to confirm the modifications. Adsorption of fibrinogen from buffer was found to decrease by ~80% for the PEO-modified surfaces compared to the unmodified PU. The surfaces with ATH attached to the distal chain end of the grafted PEO were equally protein resistant, and when the data were normalized to the ATH surface density, PEO in the lower MW range showed greater protein resistance. Western blots of proteins eluted from the surfaces after plasma contact confirmed these trends. The uptake of ATH on the PEO-modified surfaces was greatest for the PEO of lower MW (300 and 600), and antithrombin binding from plasma (an indicator of heparin anticoagulant activity) was highest for these same surfaces. The PEO-ATH- and PEO-modified surfaces also showed low platelet adhesion from flowing whole blood. It is concluded that for the PEO-ATH surfaces, PEO in the low MW range, specifically MW 600, may be optimal for achieving an appropriate balance between resistance to nonspecific protein adsorption and the ability to take up ATH and bind antithrombin in subsequent blood contact.  相似文献

Constructing a solid electrolyte interface (SEI) is a highly effective approach to overcome the poor reversibility of lithium (Li) metal anodes. Herein, an adhesive and self‐healable supramolecular copolymer, comprising of pendant poly(ethylene oxide) (PEO) segments and ureido‐pyrimidinone (UPy) quadruple‐hydrogen‐bonding moieties, is developed as a protection layer of Li anode by a simple drop‐coating. The protection performance of in‐situ‐formed LiPEO–UPy SEI layer is significantly enhanced owing to the strong binding and improved stability arising from a spontaneous reaction between UPy groups and Li metal. An ultrathin (approximately 70 nm) LiPEO–UPy layer can contribute to stable and dendrite‐free cycling at a high areal capacity of 10 mAh cm?2 at 5 mA cm?2 for 1000 h. This coating together with the promising electrochemical performance offers a new strategy for the development of dendrite‐free metal anodes.  相似文献

We covalently immobilized poly(ethylene oxide) (PEO) chains onto a fluorinated ethylene propylene copolymer (FEP) surface. On the FEP surface, aldehyde groups were first deposited by plasma polymerization of acetaldehyde or acrolein. Then, amino‐PEO chains were immobilized through Schiff base formation, which was followed by reduction stabilization with sodium cyanoborohydride. The PEO‐grafted polymer surfaces thus prepared were characterized by X‐ray photoelectron spectroscopy (XPS), atomic force microscopy, contact‐angle measurements, and protein adsorption. The dramatic increase in the C O intensity of the high‐resolution XPS C 1s spectrum, together with an overall increase in oxygen content, indicated the successful attachment of PEO chains onto the acetaldehyde plasma surfaces. The amount of grafted PEO chains depended on the superfacial density of the plasma‐generated aldehyde groups. The grafted monoamino‐PEO chains formed a brushlike structure on the polymer surface, whereas the bisamino‐PEO chains predominately adopted a looplike conformation. The PEO surface had a regular morphology with greater roughness than the aldehyde surface underneath. Surface hydrophilicity increased with the grafting of PEO. Also, the bisamino‐PEO‐grafted surface had slightly higher surface hydrophilicity than its monoamino‐PEO counterpart. Novel poly(ether urethanes) containing diester groups in the side chains (PU) were synthesized from 4,4′-diphenylmethyl diisocyanate, polytetramethylene glycol, and diethyl bis(hydroxymethyl)malonate as a chain extender. The surface modification of the PU film was carried out by a hydrolysis reaction, poly(ethylene oxide) (PEO) grafting, and heparin immobilization, and the surface-modified PUs were then characterized by attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy, electron spectroscopy for chemical analysis (ESCA), and a contact angle goniometer. The concentration of carboxylic acid groups introduced on the PU surfaces as determined by the rhodamine interaction method was 61 nmol/cm2 when treated with 4N NaOH/methanol (1 : 2 v/v) for 30 min and subsequently with a citric acid–methanolic aqueous solution. The amounts of heparin coupled to the carboxyl groups on the PU surfaces and to the terminus amino groups on the PU-PEO were 0.92 and 0.84 μ g/cm2, respectively. There was almost no heparin released from the immobilized surface of a physiological solution for 100 h, thereby indicating the strong stability of immobilized heparin. © 1998 John Wiley & Sons, Inc. Surfaces coated with poly(ethylene oxide) containing nonionic polymers are of interest in medical applications due to, among other things, the low adsorption of proteins on such surfaces. In this paper we have studied the interfacial properties of surfaces coated with PEO by measuring the forces acting between two such surfaces in water and across a protein solution as well as between one such surface and a surface carrying adsorbed proteins. One type of surface coating was a graft copolymer of poly(ethylene imine) and poly(ethylene oxide) where the cationic poly(ethylene imine) group anchored the polymer to negatively charged mica surfaces. Three different ways to prepare this coating was used and compared. It was found that this coating was not stable in the presence of lysozyme, a small positively charged protein, when the PEO graft density was low. The other type of coating was obtained by adsorbing ethyl(hydroxyethyl)-cellulose onto hydrophobised mica surfaces. The driving force for adsorption is in this case the hydrophobic interaction between nonpolar segments of the polymer and the surface. The EHEC coating was stable in the presence of lysozyme and the interactions between adsorbed layers of lysozyme and EHEC coated surfaces are purely repulsive due to long-range steric forces.  相似文献

A reversible addition–fragmentation chain transfer (RAFT) polymerization technique was applied to graft polymerize brushes of poly(methyl methacrylate) (PMMA) and poly(poly(ethylene glycol) monomethacrylate) (PPEGMA) from poly(vinylidene fluoride) (PVDF) surfaces. PVDF surfaces were exposed to aqueous LiOH, followed by successive reductions with NaBH4 and DIBAL‐H to obtain hydroxyl functionality. Azo‐functionalities, as surface initiators for grafting, were immobilized on the PVDF surfaces by esterification of 4,4′‐azobis(4‐cyanopentanoic acid) and the surface hydroxyl groups. The chemical composition and surface topography of the graft‐functionalized PVDF surfaces were characterized by X‐ray photoelectron spectroscopy, attenuated total reflectance‐FTIR spectroscopy, and atomic force microscopy. Kinetics studies revealed a linear increase in the graft concentration of PMMA and PPEGMA with the reaction time, indicating that the chain growth from the surface was consistent with a “controlled” or “living” process. The living chain ends were used as the macroinitiator for the synthesis of diblock copolymer brushes. Superhydrophobic surfaces (SHS) find versatile applications as coatings due to their very high water‐repellency, self‐cleaning, and anti‐icing properties. This investigation describes the preparation of a SHS from surfactant‐free hybrid fluoropolymer latex. In this case, reversible addition‐fragmentation chain transfer (RAFT) polymerization was adopted to prepare a copolymer of 4‐vinyl pyridine (4VP) and vinyl triethoxysilane (VTES), where the pyridine units were quaternized to make the copolymer soluble in water. The copolymer was further used as a macro‐RAFT agent to polymerize 2,2,2‐trifluoroethyl methacrylate (TFEMA) in a surfactant‐free emulsion via polymerization‐induced self‐assembly (PISA). The macro‐RAFT agent contained a small amount of VTES as co‐monomer which was utilized to graft silica nanoparticles (SNPs) onto the P(TFEMA) spheres. The film prepared using the nanocomposite latex exhibited a nano‐structured surface as observed by SEM and AFM analyses. The objective of this work was to gain a better understanding of the mechanism of resistance to protein adsorption of surfaces grafted with poly(ethylene oxide) (PEO). A polyurethane-urea was used as a substrate to which PEO was grafted. Grafting was carried out by introducing isocyanate groups into the surface followed by reaction with amino-terminated PEO. Surfaces grafted with PEO of various chain lengths (PUU-NPEO) were prepared and characterized by water contact angle and X-ray photoelectron spectroscopy (XPS). XPS data indicated higher graft densities on the PUU-NPEO surfaces than on analogous surfaces prepared using hydroxy-PEO (PUU-OPEO) as reported previously [J.G. Archambault, J.L. Brash, Colloids Surf. B: Biointerf. 33 (2004) 111-120]. Protein adsorption experiments using radiolabeled myoglobin, concanavalin A, albumin, fibrinogen and ferritin as single proteins in buffer showed that adsorption was reduced on the PEO-grafted surfaces by up to 95% compared to the control. Adsorption decreased with increasing PEO chain length and reached a minimum at a PEO MW of 2000. Adsorption levels on surfaces with 5000 and 2000 MW grafts were similar. There was no clear effect of protein size on resistance to protein adsorption. Adsorption on the PUU-NPEO surfaces was significantly lower than on the corresponding PUU-OPEO surfaces, again suggesting higher graft densities on the former. Adsorption of fibrinogen from plasma was also greatly reduced on the grafted surfaces. From analysis (SDS-PAGE, immunoblotting) of the proteins eluted after plasma exposure, it was found that the grafted surfaces and the unmodified substrate adsorbed the same proteins in roughly the same proportions, suggesting that adsorption to the PEO surfaces occurs on patches of bare substrate. The PEO grafts did not apparently cause differential access to the substrate based on protein size.  相似文献

Biodegradable, amphiphilic, four‐armed poly(?‐caprolactone)‐block‐poly(ethylene oxide) (PCL‐b‐PEO) copolymers were synthesized by ring‐opening polymerization of ethylene oxide in the presence of four‐armed poly(?‐caprolactone) (PCL) with terminal OH groups with diethylzinc (ZnEt2) as a catalyst. The chemical structure of PCL‐b‐PEO copolymer was confirmed by 1H NMR and 13C NMR. The hydroxyl end groups of the four‐armed PCL were successfully substituted by PEO blocks in the copolymer. The monomodal profile of molecular weight distribution by gel permeation chromatography provided further evidence for the four‐armed architecture of the copolymer. Physicochemical properties of the four‐armed block copolymers differed from their starting four‐armed PCL precursor. The melting points were between those of PCL precursor and linear poly(ethylene glycol). The length of the outer PEO blocks exhibited an obvious effect on the crystallizability of the block copolymer. The degree of swelling of the four‐armed block copolymer increased with PEO length and PEO content. The micelle formation of the four‐armed block copolymer was examined by a fluorescent probe technique, and the existence of the critical micelle concentration (cmc) confirmed the amphiphilic nature of the resulting copolymer. The cmc value increased with increasing PEO length. The absolute cmc values were higher than those for linear amphiphilic block copolymers. © 2004 Wiley Periodicals, Inc. The graft polymer poly(ethylene oxide)‐g‐poly(?‐caprolactone)2 (PEO‐g‐PCL2) with modulated grafting sites was synthesized by the combination of ring‐opening polymerization (ROP) mechanism, efficient Williamson reaction, with thiol–ene addition reaction. First, the precursor of PEO‐Allyl‐PEO with two terminal hydroxyl groups and one middle allyl group was prepared by ROP of EO monomers. Then, the macroinitiator [PEO‐(OH)2‐PEO]s was synthesized by sequential Williamson reaction between terminal hydroxyl groups and thiol–ene addition reaction on pendant allyl groups. Finally, the graft polymer PEO‐g‐PCL2 was obtained by ROP of ?‐CL monomers using [PEO‐(OH)2‐PEO]s as macroinitiator. The target graft polymer and all intermediates were well characterized by the measurements of gel permeation chromatography, 1H NMR, and thermal gravimetric analysis. The crystallization behavior was investigated by the measurements of differential scanning calorimetry, wide‐angle X‐ray diffraction and polarized optical microscope. The results showed that when the PCL content of side chains reached 59.2%, the crystalline structure had been dominated by PCL part and the crystalline structure formed by PEO part can be almost neglected. © 2014 Wiley Periodicals, Inc. A polysiloxane–poly(methyl methacrylate) (PMMA) graft copolymer was prepared by hydrosilylation reaction between a Si? H containing polysiloxane and an allyl-terminated PMMA. The obtained graft copolymer was blended with PMMA homopolymer. The addition of only 0.01 wt % of graft copolymer was sufficient to make PMMA surfaces hydrophobic. In acetone–cyclohexane mixed solvent, the graft copolymer formed a polymeric micelle by the aggregation of PMMA branches.  相似文献

Amphiphilic dendritic–linear–dendritic triblock copolymers based on hydrophilic linear poly(ethylene oxide) (PEO) and hydrophobic dendritic carbosilane were synthesized with a divergent approach at the allyl end groups of diallyl‐terminated PEO. Their micellar characteristics in an aqueous phase were investigated with dynamic light scattering, fluorescence techniques, and transmission electron microscopy. The block copolymer with the dendritic moiety of a third generation could not be dispersed in water. The block copolymers with the first (PEO–D ‐Si‐1G) and second (PEO–D ‐Si‐2G) generations of dendritic carbosilane blocks formed micelles in an aqueous phase. The critical micelle concentrations of PEO–D ‐Si‐1G and PEO–D ‐Si‐2G, determined by a fluorescence technique, were 27 and 16 mg/L, respectively. The mean diameters of the micelles of PEO–D ‐Si‐1G and PEO–D ‐Si‐2G, measured by dynamic light scattering, were 170 and 190 nm, respectively, which suggests that the micelles had a multicore‐type structure. The partition equilibrium constants of pyrene in the micellar solution increased with the increasing size of the dendritic block (e.g., 7.68 × 104 for PEO–D ‐Si‐1G and 9.57 × 104 for PEO–D ‐Si‐2G). The steady‐state fluorescence anisotropy values (r) of 1,6‐diphenyl‐1,3,5‐hexatriene were 0.06 for PEO–D ‐Si‐1G and 0.09 for PEO–D ‐Si‐2G. The r values were lower than those of the linear polymeric amphiphiles, suggesting that the microviscosity of the dendritic micellar core was lower than that of the linear polymeric analogues. © 2001 John Wiley & Sons, Inc. This paper aims at reporting on the synthesis of a heterograft copolymer by combining the “grafting onto” process based on atom transfer radical addition (ATRA) and the “grafting from” process by atom transfer radical polymerization (ATRP). The statistical copolymerization of ε‐caprolactone (εCL) and α‐chloro‐ε‐caprolactone (αClεCL) was initiated by 2,2‐dibutyl‐2‐stanna‐1,3‐dioxepane (DSDOP), followed by ATRA of parts of the chlorinated units of poly(αClεCL‐co‐εCL) on the terminal double bond of α‐MeO,ω‐CH2?CH? CH2? CO2‐poly(ethylene oxide) (PEO). The amphiphilic poly(εCL‐g‐EO) graft copolymer collected at this stage forms micelles as supported by dynamic light scattering (DLS) and transmission electron microscopy (TEM). A novel stabilized hemocompatible multicomponent coating was engineered by consecutive alternating adsorption of two polysaccharides, alginate (Alg) and heparin (Hep), onto a Nitinol surface via electrostatic interaction in combination with photoreaction in situ. For this purpose, a photosensitive cross-linker, p-diazonium diphenyl amine polymer (PA), was used as an interlayer between alginate and heparin. The optical intensity of UV/vis spectra increased linearly with the number of layers, indicating the buildup of a multilayer structure and uniform coating. Photo-cross-linking resulted in higher stability without compromising its catalytic capacity to promote antithrombin III (ATIII)-mediated thrombin inactivation. Chromogenic assays for heparin activity proved definitively that anticoagulation activity really comes from surface-bound heparin in multilayer film, not from solution-phase free heparin that has leaked from multilayer film. The activated partial thromboplastin time (aPTT) assay showed that both (PA/Hep)8- and (PA/Alg/PA/Hep)4-coated Nitinol were less thrombogenic than the uncoated one. Yet, the latter was found to be more stable under a continuous shaken wash. In addition, (PA/Alg/PA/Hep)4 film exhibited lower surface roughness and higher hydrophilicity than (PA/Hep)8. As a result, hemolysis of (PA/Alg/PA/Hep)4 (0.34 +/- 0.064%) was lower than (PA/Hep)8 (0.52 +/- 0.241%). The naked Nitinol and (PA/Hep)8-coated Nitinol showed relatively strong platelet adhesion. On the contrary, no sign of any cellular matter was seen on the (PA/Alg/PA/Hep)4 surface. It is believed that the phenomenon of interlayer diffusion resulted in blended structures, hence, the enhanced wettability and antifouling properties after the incorporation of alginate layers. It is likely that the cooperative effect of alginate and heparin led to the excellent blood compatibility of the (PA/Alg/PA/Hep)4 coating. To simplify, there is greater advantage in utilizing cross-linked alginate/heparin surfaces rather than merely the heparin surface for improving blood- and tissue-compatible devices.  相似文献

The grafting of polyamide 6 (PA6) onto polystyrene (PS) can rely on the use of a copolymer of styrene (St) and 3‐isopropenyl‐α, α‐dimethylbenzene isocyanate (TMI), PS‐co‐TMI, to activate the polymerization of ε‐caprolactam (CL) in the presence of sodium ε‐caprolactam (NaCL) as an anionic catalyst. This article is aimed at answering the following key questions. First, do all the isocyanate moieties of the PS‐co‐TMI participate in the activation of the polymerization of CL? Second, what are the composition of the resulting polymer product and the structure of the resulting graft copolymer? The results show that the isocyanate moieties had all participated in the activation of the polymerization, implying that each isocyanate moiety has led to the formation of a PA6 graft. The as‐polymerized product was composed of a pure PS‐g‐PA6 graft copolymer, homo‐PA6, and unreacted CL. Moreover, when the composition of a PS‐co‐TMI/CL/NaCL system was fixed, the mass ratio between the PA6 grafts and PS backbone of the pure PS‐g‐PA6 graft copolymer was almost a constant and was almost independent of its molar mass. © 2008 Wiley Periodicals, Inc. Products of the radical dispersion copolymerization of methacryloyl‐terminated poly(ethylene oxide) (PEO) macromonomer and styrene were separated and characterized by size exclusion chromatography (SEC), full adsorption‐desorption (FAD)/SEC coupling and eluent gradient liquid adsorption chromatography (LAC). In dimethylformamide, which is a good solvent for PEO side chains but a poor solvent for polystyrene (PS), amphiphilic PS‐graft‐PEO copolymers formed aggregates, which were very stable at room temperature even upon substantial dilution. The aggregates disappeared at high temperature or in tetrahydrofuran (THF), which is a good solvent for both homopolymers and for PS‐graft‐PEO. FAD/SEC procedure allowed separation of homo‐PS from graft‐copolymer and determination of both its amount and molar mass. Effective molar mass of graft‐copolymer was estimated directly from the SEC calibration curve determined with PS standards. Presence of larger amount of the homo‐PS in the final graft‐copolymer products was also confirmed with LAC measurements. The results indicate that there are at least two or maybe three polymerization loci; namely the continuous phase, the particle surface layer and the particle core. The graft copolymers are produced mainly in the continuous phase while PS or copolymer rich in styrene units is formed mostly in the core of monomer‐swollen particles. © 2000 John Wiley & Sons, Inc. The thermodynamic properties of triblock copolymer of polystyrene–poly (ethylene oxide)–polystyrene (PS‐b‐PEO‐b‐PS) were investigated by means of inverse gas chromatography (IGC) using 15 different kinds of solvents as the probes. Some thermodynamic parameters, such as specific retention volume, molar heats of sorption, weight fraction activity coefficient, Flory‐Huggins interaction parameter, partial molar heats of mixing and solubility parameter were obtained to judge the interactions between PS‐b‐PEO‐b‐PS polymers and solvents and the solubility of the polymers in these solvents. It was found that increasing PEO content in PS‐b‐PEO‐b‐PS resulted in the increase in the solubility of PS‐b‐PEO‐b‐PS in alkanes and acetates solvents, but the solubility in alcohols had no change, and more PEO content in polymer caused a small decrease in the solubility parameter of PS‐b‐PEO‐b‐PS polymer, © 2007 Wiley Periodicals, Inc. This article describes the synthesis and characterization of new amphiphilic polymer conetworks containing hydrophilic poly(2,3‐dihydroxypropyl methacrylate) or poly(ethylene glycol) methacrylate (PEGMA) and hydrophobic polyisobutylene chains. This conetworks were prepared by a two‐step polymer synthesis. In the first step, a cationic copolymer of isobutylene and 3‐isopropenyl‐α,α‐dimethylbenzyl isocyanate (IDI) was prepared. The isocyanate groups of the IB‐IDI random copolymer were subsequently transformed in situ to methacrylate (MA) groups in reaction with 2‐hydroxyethyl methacrylate (HEMA). In the second step, the resulting MA‐multifunctional PIB‐based crosslinker, PIB(MA)n, with an average functionality of approximately four per chain, was copolymerized with 2,3‐dihydroxypropyl methacrylate or poly(ethylene glycol) methacrylate by radical mechanism in tetrahydrofuran giving rise to amphiphilic conetworks containing 11–60 mol % of DHPMA or 10–12 mol % of PEGMA. The synthesized conetworks were characterized with solid‐state 13C‐NMR spectroscopy and differential scanning calorimetry.
