Melamine is attracting much attention because of its toxicity. In the work discussed in this paper, microwave-assisted extraction in combination with ion-exchange high-performance liquid chromatography with diode-array detection was used for analysis of melamine in pet food. Trichloroacetic acid–N,N-dimethylformamide 10:1 was the best extractant, because of the strong polarity of melamine. Separation was performed on a 250 mm × 4.6 mm i.d., 5-μm particle, cation-exchange column; isocratic elution with a mixture of ammonium formate solution (pH 3.0) and acetonitrile was complete within 10 min. UV absorbance DAD detection was performed at 240 nm. Response was a linear function of melamine concentrations from 0.1 to 50 μg mL?1, with a detection limit of 1.0 mg kg?1. Intra-day and inter-day precision, as RSD, was <3% and the recovery of the assay was in the range 95.4–106.8%. In analysis of spiked pet food, the new method yielded satisfactory results. Because of its simplicity and accuracy this straightforward method is particularly suitable for routine melamine analysis.  相似文献

A new method involving matrix solid-phase dispersion (MSPD) extraction and UPLC in conjunction with photodiode array detection was developed for the rapid and simple determination of Sudan dyes in chili powder. Separation of Sudan I, Sudan II, Sudan III, and Sudan IV was achieved within 2 min on the 1.7 μm Acquity UPLC BEH C18 column by using gradient elution with a mobile phase consisting of acetonitrile–water at a flow rate of 0.5 mL min?1. Optimization of MSPD extraction parameters, such as type of solid sorbent and elution solvent were carried out. Optimal conditions selected for MSPD extraction were 0.25 g of sample, 0.5 g of silica gel as solid sorbent, and 7 mL of acetonitrile–methanol (9:1, v/v) as eluting solvent. Limits of detection ranged between 0.25 and 0.30 mg kg?1 depending on the dye involved. All analytes provided average recoveries from spiked (at 1, 1.5, and 2 mg kg?1) chili powder samples ranging from 81 to 106%. The method was applied to the analysis of chili powder samples obtained from different countries.  相似文献

Titanium was determined in nano-titanium(IV) oxide food packaging by microwave digestion with inductively coupled plasma atomic emission spectrometry (ICP-AES) and inductively coupled plasma mass spectrometry (ICP-MS). Microwave digestion was optimized using different acid combinations. Both spectrometry techniques showed good reproducibility, repeatability, and recovery. For ICP-AES, the limit of detection was 5.0 mg kg?1, the linear dynamic range was 100–5000 µ g L?1, the average recoveries for blank samples spiked with titanium were between 94.7% and 100.1%, and the relative standard deviations were from 2.1% to 7.1%. By ICP-MS, the limit of detection was 0.3 mg kg?1, the linear dynamic range was 0.5–200 µ g L?1, the recoveries were 88.4%–96.3%, and the relative standard deviations were 6.3%–7.4%. These results indicated that methods were effective for the determination of titanium in food packaging.  相似文献

Melamine attracts considerable attention because of its toxicity. The determination of melamine in seafood was performed by gas-chromatography–mass spectrometry, using an optimized version of a method adopted by the U.S. Food and Drug Administration. The melamine was extracted by closed-vessel microwave-assisted solvent extraction (MAE), as a valid alternative in sample preparation, to reduce analysis time and provide less ambiguous data. The procedure was optimized by means of experimental factorial design considering the three main variables that affect this process: microwave oven power, the maximum temperature inside the extraction tube, and the hold time. The recovery of melamine in spiked samples was used as the elemental response value of the design. Temperature and hold time had a more positive effect on the response than the microwave power. A significant positive interaction was observed between oven power and hold time. A temperature of 70°C and a hold time of 1 min gave a recovery of 92 ± 5% for a microwave power of 600 W. Under these conditions, the total microwave extraction time was approximately 2 minutes, a much shorter time compared to the ultrasonic bath, which required a total time of 40 min. The repeatability of the method was approximately 3%. The limits of quantification were 0.55 mg kg?1 for MAE and 1.9 mg kg?1 for the ultrasonic bath; the linearity was confirmed up to 10 mg kg?1. In conclusion, the MAE procedure was shown to be an excellent alternative to the official method.  相似文献

A facile and sensitive method utilizing capillary gas chromatography with nitrogen phosphorus detection (GC–NPD) has been developed and validated for simultaneous analysis of hexaconazole, myclobutanil, and tebuconazole, three broad-spectrum systemic fungicides, in apples and soil. Two samples were fortified with the three pesticides and subjected to ultrasonic extraction, followed by solid-phase extraction (SPE) to remove coextractives, before analysis by GC–NPD. SPE procedures were performed on PSA cartridges (500 mg, 3 mL), the analytes being eluted with n-hexane–acetone (9:1 v/v, 2 mL). Recovery of three pesticides from the fortified apple and soil samples ranged from 94.5 to 107.3% with relative standard deviations less than 9.7% at the three spike levels (0.01, 0.1, and 0.5 mg kg?1). Limits of quantification of the method for apple and soil were 0.01 mg kg?1, sufficiently below the maximum residue limits. Direct confirmation of the analytes in samples was achieved by gas chromatography–mass spectrometry (GC–MS).  相似文献

Solid-phase extraction (SPE) and reversed-phase liquid chromatography (RP-LC) have been used for simple, sensitive simultaneous analysis of cyromazine and melamine residues in liquid milk and eggs. The conditions used for SPE and LC were investigated and optimized. A combined cation-exchange–reversed-phase cartridge was used for clean-up, and an ODS (C18) column (150 mm × 4.6 mm i.d., 5-μm particles) with 62:38 (v/v) 5 mm sodium lauryl sulfate (pH 3.4)–acetonitrile as mobile phase was used for RP-LC. Under the optimum conditions the method limit of detection (LOD) for both cyromazine and melamine was 6.2 μg kg?1 for liquid milk samples, and 11.5 μg kg?1 for egg samples. Average recovery of cyromazine and melamine from milk samples was 90.3%, RSD 4.6–5.6%, and 99.6%, RSD 3.2–4.7%, respectively. Average recovery of cyromazine and melamine from egg samples was 85.3%, RSD 1.0–4.7%, and 89.6%, RSD 3.1–5.0%, respectively. The method enables detection of melamine and cyromazine at levels as low as 20.7 μg kg?1 in liquid milk and 38.3 μg kg?1 in egg.  相似文献

An analytical method for rapid screening and quantitative determination of ten antibiotics (chloramphenicol, thiamphenicol, tetracycline, oxytetracycline, chlortetracycline, metacycline, doxycycline, cefoperazone, ceftriaxone and cefaclor) residues in milk was developed using ultra performance liquid chromatography with photodiode array detector. After extraction with McIIvaine buffer + methanol (8 + 2), the extract was cleaned up with solid-phase extraction cartridge. The conditions of sample extraction, cleaning and separation were optimized. The average spiked recoveries of milk samples were 52.1–68.0, 70.1–81.0 and 76.2–101.0% at spiked levels of 0.1, 0.5, 2.5 μg g?1, respectively with precisions of 3.3–15.9%. The limits of detection and quantification were 0.003–0.022 and 0.01–0.08 μg g?1, respectively. The proposed method has been applied to the determination of antibiotics in actual milk samples with satisfactory results.  相似文献

Levels of nicotine higher than the maximum residue level were detected in edible mushrooms. Analyses of self-collected and purchased dried mushrooms were performed with a QuEChERS approach. A small amount (2.5 g) of dried sample matrix was mixed with 5 mL sodium hydroxide solution, 2.5 g of sodium sulfate/sodium chloride mixture (4:1) and 5 mL ethyl acetate. The organic phase was cleaned by using dispersive solid-phase extraction and analysed by GC–MS–MS. The linear range of the method was 0.01–10.00 mg kg?1 and the limit of detection 0.006 mg kg?1 on dry weight basis by using EURACHEM method.  相似文献

A sensitive matrix solid-phase dispersion (MSPD)/gas chromatography-electron capture detector (GC-ECD) method was developed to determine cypermethrin residues in three tissues (muscle, liver, and gill) of crucian carp. Cypermethrin was simultaneously extracted by MSPD and determined by GC-ECD. The main parameters affecting extraction yield and selectivity, such as the type of solid adsorbent material, choice of elution solvents and their volume, were investigated to obtain interference-free extracts and quantitative cypermethrin recovery. Fortified recoveries in muscle, liver, and gill samples ranged from 84.9% to 106.1%, and relative standard deviations were <8% with fortification levels of 0.05–1 mg kg?1. Detection limits were 1.4–2.1 μg kg?1, and quantitation limits were 5.8–7.8 μg kg?1. The proposed method was successfully applied to determination of cypermethrin in fish tissue samples.  相似文献

An extraction method based on matrix solid-phase dispersion was developed to determine carbofuran, pyrimethanil and tetraconazole in banana using gas chromatography–mass spectrometry. The best results were obtained using 2.0 g of banana, 1.0 g of silica as dispersant sorbent and n-hexane:ethyl acetate (1:4, v/v) as eluting solvent. The method was validated using banana samples fortified with pesticides at different concentration levels (0.05–2.0 mg kg?1). Average recoveries (four replicates) ranged from 68 to 111%, with relative standard deviations between 6.6 and 20.5%. Detection and quantification limits for banana ranged from 0.02 to 0.05 and 0.05 to 0.10 mg kg?1, respectively.  相似文献

An ion-pair high-performance liquid chromatography with ultraviolet detection method for the determination of cyromazine, melamine and its biodegradation products (ammeline, ammelide, cyanuric acid and biuret) was developed. C18 column was utilised to separate the six analytes with a mobile phase consisting of perchloric acid-ammonia solution and acetonitrile, under gradient elution and variable flow rate. The detection wavelengths were 205 nm for cyanuric acid and biuret and 222 nm for cyromazine, melamine, ammeline and ammelide. For analysis of sediment samples, the extraction solution containing acetonitrile, ammonia and water (80:10:10 by volume) was used to extract the analytes from sediment matrix. Using the extraction method for the spiked sediment sample, high linearity of matrix-matched standard curve could be obtained for the six analytes. The method detection limit was 0.1 μg g?1 for melamine and cyromazine, 0.2 μg g?1 for ammeline and ammelide, 1.2 μg g?1 for cyanuric acid and 1.0 μg g?1 for biuret in sediment matrix. The recoveries of these compounds were 70.1–98.3% and the relative standard deviations were 0.5–4.4%. Finally, the proposed method was successfully applied to the analysis of the sediment sample near the wastewater outlet of a melamine-producing factory.  相似文献

A reliable and sensitive method for determination of MCPA in soil by derivatization through p-toluenesulfonic acid and 1,3-dichloro-2-propanol followed by gas chromatographic detection under ECD mode has been established. After treatment with hydrochloric acid, the soil samples were directly extracted without any clean-up with dichloromethane by vortexing. After derivatization and liquid–liquid extraction, the product was subjected to GC analysis. Under optimized conditions, recovery of MCPA reached 87–91%; intra- and inter-day precision values were recorded in the range 3.4–6.4% and 7.4–8.7%, respectively. Excellent linear relationship was observed within 0.1–10 μg mL?1 (0.005–0.5 mg kg?1) with linear correlation coefficient (R) of 0.9997. The LOD and LOQ were 0.001 and 0.0026 mg kg?1, respectively, and the overall sensitivity for detection was found to be in the same range as with the conventional GC-MS technique.  相似文献

The fate of kresoxim-methyl was studied in a tobacco field ecosystem, and a simple and reliable method was developed for the determination of kresoxim-methyl in soil, green and cured tobacco leaves. Kresoxim-methyl residues were extracted from samples with petroleum ether, and determined by gas chromatography (GC) coupled with an electron capture detector (ECD). Kresoxim-methyl (30% suspension concentration) was applied at 150 g a.i. ha–1 (the recommended high dosage) and 225 g a.i. ha–1 (1.5 times the recommended high dosage) in the experimental fields in Huishui and Changsha in China. The limits of detection (LODs) and limits of quantification (LOQs) of kresoxim-methyl in green tobacco leaves, cured tobacco leaves and soil were 0.012 and 0.04 mg kg–1, 0.12 and 0.4 mg kg–1, and 0.0015 and 0.005 mg kg–1, respectively. The average recoveries were 84.5% to 95.7%, 79.8% to 94.3% and 83.3% to 93.8% with relative standard deviations (RSDs) less than 10% in green tobacco leaves at four spiked levels (0.04, 0.2, 2 and 8 mg kg–1), cured tobacco leaves at three spiked levels (0.4, 1 and 10 mg kg–1) and soil at three spiked levels (0.005, 0.05 and 0.5 mg kg–1), respectively. The results showed that the half-lives of kresoxim-methyl in green tobacco leaves and soil were 1.2–5.3 days and 6.7–10.4 days, respectively. At harvest, kresoxim-methyl residues in cured tobacco leaves samples collected 21 days after the last application at the recommended dosage were below 1.0 mg kg–1. These results could help establish appropriate application frequency and harvest intervals in the use of kresoxim-methyl on tobacco plants.  相似文献

This study reports on the development of a fast and efficient method based on headspace solid-phase microextraction (HS-SPME) coupled to gas chromatography–tandem mass spectrometry (GC–MS/MS) for simultaneous analysis of 128 volatile or semi-volatile pesticide residues belonging to nine classes of pesticides. The important factors related to HS-SPME performance were optimized; these factors include fiber types, water volume, ion strength, extraction temperature, and extraction time. The best extraction conditions include a PDMS/DVB fiber, and analytes were extracted at 90 °C for 60 min from 1 g of tea added to 5 mL of 0.2 g mL?1 NaCl solution. The methodology was validated using tea samples spiked with pesticides at three concentration levels (10, 50, and 100 μg kg?1). In green tea, oolong tea, black tea, and puer tea, 82.8, 88.3, 79.7, and 84.3% of the targeted pesticides meet recoveries ranging from 70 to 120% with a relative standard deviation of?≤?20%, respectively, when spiked at a level of 10 μg kg?1. Limits of quantification in this method for most of the pesticides were 1 or 5 μg kg?1, which are far below their maximum residue limits prescribed by EU. The optimized method was employed to analyze 30 commercial samples obtained from local markets; 17 pesticide residues were detected at concentrations of 2–452 μg kg?1. Chlorpyrifos was the most detected pesticide in 80% of the samples, and the highest concentration of dicofol (452 μg kg?1) was found in a puer tea. This is the first time to find that the optimized extraction temperature for pesticide residues is 90 °C, which is much higher than other reported HS-SPME extraction conditions in tea samples. This developed method could be used to screen over one hundred volatile or semi-volatile pesticide residues which belong to multiple classes in tea samples, and it is an accurate and reliable technique.  相似文献

A method for determination of chlormequat (CCC) residue in fruits by liquid chromatography?Ctandem mass spectrometry (LC?CMS?CMS) was developed. Residue of CCC was extracted from samples with methanol?Cwater (v/v, 1:1) containing 1.0% acetic acid, cleaned up by strong cationic exchange (SCX) cartridge, and then determined by LC?CMS?CMS. The method showed good linearity over the concentration range 0.002?C5.0 mg kg?1 with correlation coefficient above 0.997. The limit of detection (LOD) and limit of quantitation (LOQ) for CCC were 5 × 10?4 mg kg?1 (S/N = 3) and 0.002 mg kg?1 (S/N = 10), respectively. Recoveries for CCC at three spiked levels (0.025, 0.050, and 0.20 mg kg?1) were in the range 80?C102%. Estimation of measurement uncertainty was calculated for CCC at the level of 0.025 mg kg?1 in fruits. The results demonstrated that the uncertainty of recovery was the main contribution to the combined standard uncertainty. The relative combined standard uncertainties associated with the method ranged from 11 to 13%, depending on the sample matrices.  相似文献

The total metal concentration and bioaccessible concentration of Cr, Mn, Fe, Cu, Zn, Se in Momordica charantia, Asparagus racemosus, Terminalia arjuna and Syzyzium cumini were measured by instrumental neutron activation analysis and by inductively coupled plasma mass spectrometry analysis (ICP-MS). The bioaccessible concentrations were determined in the gastrointestinal digest obtained after treating dried powdered samples sequentially in gastric and intestinal fluid of porcine origin at physiological conditions. The bioaccessible concentration of Fe was in the range of 58–67 mg kg?1, Mn was 10.2–14.6 mg kg?1, Cu was 3.7–4.8 mg kg?1 and Zn was 10.6–18.4 mg kg?1, were within the safety limits set for vegetable food stuff set by Joint FAO/WHO. The bioaccessibility of Zn, an essential element, was high (40–50 %) in M. charantia and in S. cumini. In addition, the total metal contents and bioaccessible concentration of Ni, Se, Cd and Pb in these samples were measured by ICP-MS. The total Cd content in S. cumini (2.6 ± 0.2 mg kg?1) and its bioaccessible concentration (0.6 mg kg?1) were strikingly high as compared to the other samples. Though total Hg contents were determined by ICP-MS, but their bioaccessible concentrations were below the detection limit (0.036 mg kg?1).  相似文献

A robust and sensitive method was developed for the simultaneous analysis of metribuzin and quizalofop-p-ethyl residues in potato and soil, based on solid-phase extraction (SPE) coupled to capillary gas chromatography with electron capture detector (GC-ECD). Residues of two herbicides were extracted from potato and soil with acetone and methanol–water, followed by SPE to remove coextractives, before analysis by GC-ECD. SPE procedures were performed on Florisil cartridges (500 mg, 3 mL), the analytes from potato and soil matrix were eluted with petroleum ether-acetic ether (9:1 v/v, 5 mL) and petroleum ether-acetic ether (8:2 v/v, 2 mL), respectively. Limits of quantification of the method were 0.01 mg kg?1, and the mean recoveries ranged from 72.9 to 109.5% with relative standard deviation ranging from 0.7 to 9.2% at the three spike levels (0.01, 0.1, and 0.5 mg kg?1). The proposed method was successfully applied to the analysis of metribuzin and quizalofop-p-ethyl residues in potato and soil samples from an experimental field. A novel and simple method for determination of 15 organic nitrogen-containing pesticides in vegetables using matrix solid-phase dispersion (MSPD) coupled with gas chromatography tandem mass spectrometry (GC–MS–MS) has been developed. The efficiencies of different sorbents (florisil, silicone, neutral alumina) for the MSPD were compared. Mean recoveries of the method using neutral alumina varied from 73.26 to 111.83% with relative standard deviations of 0.79–15.33% in the concentration range of 0.01, 0.02 and 0.05 mg kg?1. The limits of detection were typically in the 0.0007–0.0320 mg kg?1 range, which were 10–100 times lower than the maximum residue levels established by the European Union. This method was applied to residue detection in vegetables, in which organic nitrogen-containing compounds were detected at low concentrations.  相似文献
