在常规不锈钢色谱管中以甲基丙烯酸为功能单体,采用原位聚合法制备了(5S,11S)-特罗格尔碱(S-TB)的印迹整体柱.考察了流动相中添加不同量的醋酸和水对分离的影响,结合台阶梯度洗脱模式在S-TB整体柱上实现了对TB消旋体的快速分离.另外,以碱性单体2-二甲基乙基胺甲基丙烯酸酯(DAMA)为功能单体,在毛细管中采用原位聚合法制备了毛细管分子印迹整体柱,用于在毛细管电色谱(CEC)中对消旋体1,1'-联-2-萘酚(BNL)进行手性分离.结果表明,以DAMA为功能单体可以制备其他酸性模板的分子印迹聚合物,从而扩大了分子印迹聚合物(MIP)在CEC分离中的应用范围.  相似文献

在石英毛细管中原位聚合制备了4-氨基吡啶印迹聚合物毛细管整体柱,通过考察流动相中乙腈比例对4-氨基吡啶、2-氨基吡啶和硫脲在分子印迹聚合物毛细管整体柱、空白聚合物毛细管整体柱和硅烷化衍生的开管柱上迁移时间和分离情况的影响,研究了4-氨基吡啶分子印迹聚合物毛细管整体柱的CEC识别机理。发现有机添加剂的含量对印迹聚合物的印迹识别能力影响很大,甚至能改变混合物的流出顺序。根据随乙腈含量改变混合物迁移时间和流出顺序的变化规律,可以推测:随着乙腈含量的提高,色谱保留对迁移的影响越来越大;随着乙腈含量的降低,电泳对迁移的影响越来越大。  相似文献

本研究以烯丙基咪唑鎓β-环糊精(AI-β-CD)为功能单体,乙二醇二甲基丙烯酸酯(EDMA)为交联剂,采用原位聚合法制备了一种毛细管电色谱整体柱。优化制柱条件,通过红外光谱和扫描电镜对整体柱固定相的结构和形貌进行表征。以D,L-酪氨酸为分析物对整体柱性能进行评价。结果表明,所制备固定相成功键合到毛细管内,并形成错综复杂的网状结构。同时该整体柱具有较强的对映体分离能力及稳定性。应用毛细管电色谱与质谱联用技术对手性除草剂甲氧咪草烟对映体进行分离,分离度为2.25。  相似文献

以2,4-二氯苯氧乙酸分子为模板,甲基丙烯酸为单体,乙二醇二甲基丙烯酸酯为交联剂,甲苯和十二醇为混合致孔剂,采用热引发原位聚合法制备了作为高效液相色谱固定相的分子印迹整体柱.用红外光谱、扫描电镜、比表面积分析法对聚合物进行了表征.考察了模板分子在不同条件下合成的印迹整体柱及空白整体柱上容量因子的变化规律,同时探讨了流动相中甲醇的体积分数、pH值、流速对印迹整体柱分离性能的影响.结果表明,在优化的合成条件下制备的分子印迹整体柱可在15 min内分离2,4-二氯苯氧乙酸及其类似物苯氧乙酸,分离度为1.52.对柑桔提取液进行了分离测试,结果满意.  相似文献

本文采用原位聚合法制备了以四肽YPLG为模板的毛细管分子印迹整体柱,在毛细管电色谱模式下以模板分子和它的结构类似物YPGL为样品,对分子印迹聚合物的识别机理进行了研究。这两种四肽由于化学结构相似且等电点非常相近,普通的电色谱和毛细管电泳方法分离非常困难。但我们的实验表明,印迹整体柱对模板分子具有特异性识别能力,因此YPLG与YPGL之间的分离因子为1.73,分离度达3.72。实验中系统地研究了流动相中有机溶剂的含量、缓冲溶液的pH值、缓冲溶液的盐浓度以及柱温对四肽识别的影响。实验中我们观察到模板在印迹柱上具有非线性的Van’t Hoff行为,揭示可能存在多重保留机理。本研究结果表明,在毛细管电色谱模式下,分子印迹整体柱的分子识别主要决定于样品与印迹聚合物之间的氢键作用以及印迹孔穴的三维结构。  相似文献

以丙烯酰胺-β-环糊精和聚甲基丙烯酸缩水甘油酯为功能单体,乙二醇二甲基丙烯酸酯为交联剂,采用原位聚合法制备了聚丙烯酸酯-β-环糊精手性毛细管整体柱.通过红外和扫描电子显微镜表征了整体柱固定相的结构和形貌.并以苯丙氨酸为考察物考察了柱子的稳定性和重现性,柱效达到47701 plates/m,获得了良好的分离效果.在毛细管电色谱模式下,采用丙烯酰胺-β-环糊精整体柱首次拆分盐酸依替福林对映体,结果使盐酸依替福林对映体达到了基线分离.  相似文献

采用紫外光谱法选择丙烯酰胺为功能单体,以二苯甲酰-D-酒石酸(D-DBTA)为模板分子,丙烯酰胺为功能单体,二甲基丙烯酸乙二醇酯为交联剂,甲苯和十二醇为混合致孔剂,用原位分子印迹技术,合成了D-DBTA分子印迹整体聚合物。在优化色谱条件的基础上,25min内基本实现了DBTA消旋体的手性分离,分离度达1.25。对色谱分离过程中的热力学进行分析,在所考察的温度范围内,色谱分离存在两种不同的热力学过程,且两者与范特霍夫方程能够很好地吻合。在这两种热力学过程的转换温度处,分离因子α达最大值。  相似文献

A vanillin imprinted capillary monolithic column was synthesized by in situ polymerization reaction using ethylene-glycol dimethacrylate as cross-linking monomer and methacrylic acid as functional monomer. Under the optimum conditions of capillary electrochromatography, this molecularly imprinted polymer (MIP)-based column showed high selectivity and could recognize not only template molecule vanillin but also positional isomer o-vanillin from their structural analogues.  相似文献

Molecularly imprinted monolithic columns were prepared for chiral separation of tyrosine and its amino acid derivatives by in situ therm-initiated copolymerization of methacrylic acid, 4-vinylpyridine and ethylene glycol dimethacrylate. The enantiomers were rapidly separated on monolithic columns in less than 10 min by pressurized capillary electrochromatography (pCEC). The influences of several parameters such as the content of cross-linking monomer on the composition of the pre-polymerization mixture were systematically investigated. The influence of the pCEC conditions including the composition of the mobile phase was also optimized to obtain the good enantioseparation. It was found that in addition to molecularly imprinted recognition, chromatographic retention and electrophoretic migration play important roles in the retention and chiral recognition of molecularly imprinted polymer (MIP) columns. The cross-selectivity for similar amino acids and its derivatives were systematical investigated for understanding the recognition mechanism on the MIP monolithic columns. The results indicated that molecularly imprinted polymer recognizes the template molecule by its molecular shape defined binding cavity.  相似文献

A monolithic molecularly imprinted polymer with specific recognition ability for 4-hydroxybenzoic acid (4-HBA) was prepared by in situ photopolymerization, using methacrylic acid (MAA) as a functional monomer, ethylene glycol dimethacrylate (EDMA) as a cross-linking agent, toluene and isooctane as porogenic solvents and Irgacure 1800 as an initiator. Baseline separation of isomers of hydroxybenzoic acid was achieved in less than 8 min on this monolithic column using 4-HBA as template, but not on the blank polymer. Furthermore, some neutral compounds could also be baseline-separated on the imprinted polymer column in the mode of pressure-driven capillary electrochromatography.  相似文献

通过分子模拟研究模板分子与功能单体的相互作用,可以缩短优化时间,为选取合适的功能单体以及模板分子/功能单体比例提供依据.本研究以山奈酚为模板分子,通过分子模拟优化实验条件,确定以甲基丙烯酸(MAA)为最优的功能单体,山奈酚/MAA最佳比例为1∶4 (w/w).此外,以二苄基三硫代碳酸酯(DBTTC)为可逆加成-链断裂转移剂,乙二醇二甲基丙烯酸酯(EDMA)为交联剂,实现了仅需优化引发剂和可逆加成-断裂链转移聚合(RAFT)试剂即可制得性能优异的山奈酚分子印迹整体柱.此整体柱对山奈酚和相似物槲皮素的分离度为1.52,相对标准偏差为1.8%.实验结果表明,分子模拟计算简化了实验步骤,以DBTTC为RAFT试剂得到了具有更好形态和结构的分子印迹整体柱.  相似文献

Two molecular imprinting polymer (MIP) monolithic columns with (S)-(-)-1,1'-bi-2-naphthol and (R)-(+)-5,5',6,6',7,7',8,8'-octahydro-1,1'-bi-2-naphthol as the templating molecules, respectively, have been prepared by in situ polymerization using 4-vinylpyridine and ethylene dimethacrylate as functional monomer and cross-linker, respectively. The columns with good flow-through properties were obtained by changing the molar ratio of the functional monomer and the template molecule. The effects of mobile-phase composition on separation of enantiomers were systematically investigated. The results indicate that hydrophobic interaction in aqueous solution and hydrogen-bonding interaction in ACN between the enantiomers and polymers could play important roles in the retention and resolution. The effects of chromatographic conditions, such as flow rate, column temperature, sample loading, on the enantioseparation were also studied. Further, these two MIP columns show a cross-reactivity.  相似文献

A novel solid phase extraction (SPE) method for determination of tetracyclines (TCs) in milk and honey samples by molecularly imprinted monolithic column was developed. Using tetracycline (TC) as the template, methacrylic acid (MAA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the cross-linker, methanol as the solvent, cyclohexanol and dodecanol as the mixed porogenic solvents, a TC imprinted monolithic column was prepared by in situ molecular imprinting technique for the first time, and the optimal synthesis conditions and the selectivity of TC imprinted monolithic column were investigated. The interfering substances in food samples and TCs can be separated successfully on imprinted column. Molecularly imprinted solid phase extraction (MISPE) coupling with C18 column was used to determinate the TCs in milk and honey. The recoveries of this method for six tetracyclines antibiotics such as tetracycline (TC), oxytetracycline (OTC), minocycline (MINO), chlortetracycline (CTC), metacycline (MTC) and doxycycline (DTC) were investigated, and high recoveries of 73.3-90.6% from milk samples and 62.6-82.3% from honey samples were obtained. A method for determination of TCs at low concentration level in milk and honey samples was successfully developed by using the monolithic column as the precolumn for solid phase extraction of six TCs compounds.  相似文献

A method for direct determination of DL-tetrahydropalmatine (DL-THP) in Corydalis yanhusuo, a traditional Chinese herb, by L-THP imprinted monolithic precolumn on-line/off-line coupling with reversed-phase high performance liquid chromatography (RP-HPLC) was developed. The L-THP imprinted monolithic column has been prepared by in situ polymerization using methacrylic acid (MAA) and ethylene dimethacrylate (EDMA) as functional monomer and cross-linker, respectively. With the optimization of chromatographic conditions, such as mobile phase composition, flow rate, column temperature and sample loading, for the separation of enantiomer, DL-THP was base-line separated on the MIP. The imprinted monolithic column was used as a precolumn for fractionation of the C. yanhusuo extract. Both the non-retained and retained fractions were separated by RP-HPLC. Meanwhile, the D-THP and L-THP can be detected in the non-retained and retained fractions, respectively. Additionally, direct determination of L-THP using molecularly imprinted monolith on-line coupling with a reversed-phase column was acquired.  相似文献

In this work, we prepared a monolithic and surface initiated molecularly imprinted polymeric (MIP) column for HPLC and explored its application for template separation from plant extract. The silica beads (40-60 μm) were coupled with initiator on the surface and then packed in to a stainless steel HPLC column. The pre-polymerization mixture (the template, functional monomer and crosslinker were emodin, acrylamide and divinylbenzene, respectively) was injected into the column and polymerized by thermal initiation. The prepared MIP column exhibited excellent retention capability and large imprinted factor for template (the retention time and imprinted factor for emodin on MIP column were 16.26 min and 7.21, respectively). Moreover, the emodin-molecularly imprinted polymeric column could be applied to separate emodin from alcohol extract of Rheum palmatum L. at semi-preparative scale and 1.2 mg of emodin was obtained in 4 h.  相似文献

The recognition mechanism of molecularly imprinted polymer (MIP) in capillary electrochromatography (CEC) is complicated since it possesses a hybrid process, which comprises the features of chromatographic retention, electrophoretic migration and molecular imprinting. For an understanding of the molecular recognition of MIP in CEC, a monolithic MIP in a capillary with 1,1'-binaphthyl-2,2'-diamine (BNA) imprinting was prepared by in situ copolymerization of imprinted molecule, methacrylic acid and ethylene glycol dimethacrylate in porogenic solvent, a mixture of toluene-isooctane. Strong recognition ability and high column performance (theory plates was 43,000 plates/m) of BNA were achieved on this monolithic MIP in CEC mode. In addition, BNA and its structural analogue, 1,1'-bi-2, 2'-naphthol, differing in functional groups, were used as model compounds to study imprinting effect on the resultant BNA-imprinted monolithic column, a reference column without imprinting of BNA and a open capillary. The effects of organic modifier concentration, pH value of buffer, salt concentration of buffer and column temperature on the retention and recognition of two compounds were investigated. The results showed that the molecular recognition on MIP monolith in CEC mode mainly derived from imprinting cavities on BNA-imprinted polymer other than chromatographic retention and electrophoretic migration.  相似文献
