Abstract— The photochemical interaction between 8-methoxypsoralen (8-MOP) and the melanin precursorL–3,4-dihydroxyphenylalanine(dopaH2) has been studied using laser flash photolysis. Triplet excited 8-MOP was thus found to abstract electrons from dopaH2 ( k ∼ 2 × 109 dm3 mol-1 s-1) to form semireduced 8-MOP and semioxidised dopaH2.The technique of pulse radiolysis was used to establish separately the spectra of (a) the semi-reduced form of 8-MOP at pH 6.5 and (b) the semioxidised forms of dopaH2 at pH 6.5, 5.8, 4.6 and 3.3. The corresponding λmax and extinction coefficients found were: for 8-MOP at pH 6.5, λmax= 350 nm (= 9050 dm3 mol-1 cm-1); for dopa at pH 6.5, λmax= 305 nm (ε= 12000 dm3 mol-1 cm-1) and for dopaH at pH 3.3, λ= 305 nm (ε= 5900 dm3 mol-1 cm-1).  相似文献

Abstract— The extinction coefficient εT, of triplet benzophenone in benzene has been directly determined by absolute measurements of absorbed energy and triplet absorbance, Δ D 0T, under demonstrably linear conditions where incident excitation energy, E 0, and ground state absorbance, A 0, are both extrapolated to zero. The result, 7220 ± 320 M -1 cm-1 at 530 nm, validates and slightly corrects many measurements relative to benzophenone of triplet extinction coefficients made by the energy transfer technique, and of triplet yields obtained by the comparative method.
As E 0 and A 0 both decrease, Δ D 0T becomes proportional to their product. In this situation, the ratio R = (1/ A 0)(dΔ D 0T/d E 0) = (εT - εGT. Measurements of R , referred to benzophenone, give (εT - εGT for any substance, without necessity for absolute energy calibration.
Both absolute and relative laser flash measurements on zinc tetraphenyl porphyrin (εT - εG at 470 nm = 7.3 × 104 M -1 cm-1) give φT= 0.83 ± 0.04.  相似文献

Abstract— Transient absorption spectra produced by laser flash-photolysis of an aqueous solution of ephedrine have been measured under a variety of conditions. Ephedrine was found to photoionise via a biphotonic process. The apparent yield of photoionisation increases with lowering of pH, a value of 8.8 being found for the p K a associated with this change. The cation radical absorption spectrum has been determined using the techniques of both pulse radiolysis and laser flash photolysis. The extinction coefficient of the cation at 295 nm was determined to be 1.37 × 104 dm3 mol-1 cm-1 and 1.2 × 104 dm3 mol-1cm-1 by the two techniques, respectively, at pH 11. It is also shown that the rate constant for electron abstraction by the azide radical to form the ephedrine cation is controlled by protonation of the amine group in the side chain. The ephedrine anion radical spectrum and its extinction coefficient at 305 nm were also determined. The excited states responsible for photoionisation and photodegradation are discussed.  相似文献

Triplet formation quantum yields (Φτ) of psoralen in a set of 17 pure solvents ranging from n -hexane to water and in dioxane: water mixtures were obtained by nanosecond laser flash photolysis. The triplet yield increases with solvent polarity. The extremum values are 0.009 and 0.545 in n -hexane and water, respectively. Good correlations of the experimental Φτ values with empirical "polarity" scales (Dimroth/Reichardt's ET [30], Kamlet/Taft's solva-tochromic parameters β, and α, and Swains acity/basity AS/BS) were obtained: Ln(φT-1 - 1) = 8.86 - 0.143ET(30) Ln(φT-1 - 1) = 4.40 - 2.34τ - 1.70α Ln(φT-1 - 1) = 4.65 - 3.72As - 1.12Bs The results are discussed in terms of the sensitivity of psoralen triplet quantum yield to solvent polarity and hydrogen-bonding abilities.  相似文献

Abstract— Thymine and uracil triplet-triplet absorption spectra and triplet excited state lifetimes have been observed in acetonitrile and water by nanosecond laser flash spectroscopy. A study of triplet energy transfer from these pyrimidines to retino! has also allowed an estimation of the triplet extinction coefficient εTT of thymine and uracil. These εTT were then used to determine the triplet quantum yields ØT of both pyrimidines in acetonitrile and water.  相似文献

Abstract— Triplet-and singlet-related photoprocesses of pyrene-1-aldehyde (PA) in various solvents have been investigated in detail using 337.1 and 355 nm laser flash photolysis in conjunction with time-correlated determination of fluorescence lifetimes (τF) and steady-state photochemical and absorption-emission spectral measurements. In benzene, the lowest triplet of PA (43 < ET < 46 kcal/mol) has a lifetime of about 50 µs (τT) and displays the absorption maximum at 443 nm with a maximum extinction coefficient (εmax) of 21000 M -1cm-1; the corresponding ketyl radical has a sharp absorption maximum at 428 nm (εmax≥ 25000 M -1cm-1). The quantum yields (φT) of lowest triplet occupation are high in nonprotic solvents (0.6–0.8), decrease in protic solvents (alcohols) as the polarity of the latter is increased, and maintain a complementary relationship with the quantum yields (φF) of fluorescence. Quantum yields (φPC) of loss of PA due to photoreactions in some solvents have also been determined under conditions of steady irradiation at 366 nm; φPC is in the range 0.1–0.2 in electron-rich olefinic solvents such as cyclohexene and tetramethylethylene. These results concerning τF, τT, φF. φT and φPC as well as the effects of 1,2,4-trimethoxybenzene and 2,5-dimethyl-2,4-hexadiene as quenchers for fluorescence, triplet yield, and photochemistry are discussed in the light of possible state orders for PA in polar and nonpolar environments.  相似文献

Abstract— The triplet state characteristics (spectrum, lifetime and quantum yield) for four dye sensi tisers [methylene blue (MB), erythrosin (ER), haematoporphyrin (HP) and riboflavin (RF)] were determined in methanol by laser flash photolysis and singlet oxygen yields (0.60 to 0.48) from time-resolved measurements of the 1270 nm near infrared emission. The reaction of singlet oxygen with four long chain unsaturated phenyl esters [oleate (18: 1), linoleate (18: 2), linolenate (18: 3) and arachidonate (20: 4)] was followed quantitatively using the singlet oxygen luminescence technique and also, after continuous420–700 nm irradiation, by HPLC and other analysis of the isomeric product monohydroperoxides. The overall quantum yield of photooxidation (∼10-2) was shown to be consistent with the observed singlet oxygen quenching constants(2–12 times 104 dm3 mol-1 s-1) for the four esters studied and the singlet oxygen lifetime in methanol (τ∼ 9 μs). The isomer product distribution was interpreted in terms of a dual singlet oxygen and radical mechanism, the radical contribution increasing with sensitiser in the order ER = MB < HP ≪ RF, but also showing some dependence on substrate unsaturation. Evidence is presented for singlet oxygen quenching by MB and RF ( kO = 1.6 and 6.0 times 107 dm3 mol-1 s-1) and for the accelerated photobleaching of the dye sensitisers in the presence of the unsaturated esters.  相似文献

Abstract— Reactions of the triplet state of lumiflavin (3LF) in water adjusted at pH 7.2 were reexamined by means of a Xe-flash photolysis and a laser photolysis. Measurements of the decay of 3LF were made on solutions of LF ranging in the concentration from 4 to 61 times 10-6 mol/dm3. A one-electron reduced and a one-electron oxidized species of lumiflavin (LF- and LF+) were produced in the first decay stage of 3LF with a high efficiency (0.6 ± 0.1) in a bimolecular triplet-triplet reaction. The product radicals (LFH- and LF+) quench 3LF very efficiently (3 ± 0.8 × 109 mol-1dm3 s-1) compared with LF in the ground state (> 2 × 107 dm3 mol-1).  相似文献

Abstract— Nanosecond laser flash photolysis and pulse radiolysis have been used to generate and characterise the triplet state, and semioxidised and semireduced radicals of haematoporphyrin, and three 0 -acyl compounds derived from it (the monoacetate, the diacetate and the disuccinate).
After 347 nm irradiation in water containing 2% Triton X-100, haematoporphyrin forms the triplet state (φT= 0.92) and photoionises monophotonically (φI= 0.03). For the O -acyl derivatives, φT approaches unity and photoionisation is reduced. In acetone the triplet yield of all four compounds are close to unity. The difference and corrected spectra for the triplet species are presented and decay rates ( k 1˜104s-1) and oxygen quenching constants ( k Q˜1.5times109 M -1s-1) for the triplet state have been measured. The difference and corrected spectra for the semi-reduced species in methanol and semi-oxidised species in aqueous Triton X-100 are presented.
The photophysical characteristics in fluid solution of haematoporphyrin and its 0 -acyl derivatives are rather similar to those previously recorded for other photosensitising porphyrins.  相似文献

Abstract— A novel method for the determination of singlet oxygen reaction rate constants is described and applied to studies of cyclohexadiene in methanol and gelatins in H2O and D2O. The technique uses tris (2,2'-bipyridine) ruthenium(II) dication (Ru(bipy)32+) as both singlet oxygen sensitizer and in situ oxygen concentration monitor during irradiation of sealed samples. Because of the high efficiency with which the luminescence of Ru(bipy)32+* can be detected, and the fact that emission lifetimes are used, the method offers some advantages over those previously described. The advantages and disadvantages of the method are discussed. A rate constant of 2.1 (±0.3) x 106 mol-1 dm3 s-1 has been determined for the reaction of 1O2 with cyclohexadiene in methanol. For two different photographic gelatins the sums of reaction and quenching rate constants are 2.0 (±0.4) x 106 and 3.1 (±2.0) x 105 mol-1 dm3 s-1; for swine skin gelatin this value is 3.9 (±2.4) × 105 mol-1 dm3 s-1. Chemical reaction, rather than physical quenching, is the dominant reaction route for gelatins and 1O2.  相似文献

Abstract Triplet absorption spectra, extinction coefficients (ɛT), decay rates ( K 1), oxygen quenching rates (kq) and intersystem crossing yields (φT) for 3-carbethoxypsoralen (3-CPs). 8-methoxypsoralcn (8-MOP) and 5-methoxypsoralen (5-MOP) in methanol are reported. For 8-MOP and 3-CPs corresponding values are also reported with water as the solvent. Some photophysical data are also reported for 5-MOP in water, but ɛT and φT were not obtained.
The phosphorescence spectra for these furocoumarin derivatives in ethanol at 77 K are reported together with the corresponding lowest triplet energy and lifetime. The values of the various photophysical properties obtained are compared with values reported by previous workers.  相似文献

Abstract Laser flash photolysis has previously been used to study the nature of DHE via measurements of photophysical parameters which are dependent on the molecular weight of the system being studied. These results to date allow only a lower limit to be established for DHE which imply that in some environments such as detergents more than two porphyrin units are linked. We have now determined the triplet extinction coefficient of DHE by the pulse radiolysis technique via an energy transfer method which allows the triplet extinction of DHE to be estimated independent of the molecular weight. The combined techniques allow the actual molecular weight of DHE to be established at about 4200. Laser flash techniques have also now been used to determine, for a number of potential photodynamic sensitisers, the quantum yield of triplet state formation (θT) and, using the direct luminescence of singlet oxygen at 1270 nm, the quantum yield of singlet oxygen formation (θδ). For many of the porphyrins studied θδ is less than θT. For DHE itself there is a substantial increase in θδ in detergent compared to buffer. The θδ yields for a number of related systems including 'simple'systems such as haematoporphyrin, for linked porphyrin-chlorin systems, (including DHE in which the end porphyrin is reduced to a chlorin–DHEC), and for phthalocyanines are compared. For the DHEC the θδ is close to that of DHE itself which may imply that such chlorins could be of use in photodynamic therapy (PDT).  相似文献

Abstract— The triplet-triplet absorption spectra of 9-cis-, 13-cis- and all-fraw-retinal as well as the time-resolved resonance Raman spectra of the lowest electronically excited triplet states of 9-cis-, 11-cis, 13-ciy and all-trans-retinal in aromatic solvents at room temperature have been obtained under conditions ensuring the isomeric purity of the starting materials. The triplet states were produced by triplet energy transfer from a sensitizer in pulse-radiolysis experiments. The overall results suggest that the isomeric retinals form either different relaxed triplet species or different mixtures of relaxed triplet species. The possible implications about the size of the energy barriers separating the various triplet species are discussed. The resonance Raman spectra obtained by using either anthracene (ET= 177.7 kJ mol-1) or naphthalene (ET= 254.8 kJ mol-1) as sensitizers were virtually identical for the corresponding triplet states from each of the isomers 11-cis-, 13-a's- and all-tams-retinal, suggesting that the relaxed triplet species or the mixture of relaxed triplet species formed from each isomer is independent of the energy of the sensitizer.  相似文献

Abstract— –Photophysical properties of [26] porphyrin (26 P) were investigated in chloroform. The quantum yields of fluorescence, of S1→ T1 intersystem crossing and singlet oxygen formation were measured. The purity, stability, the strong absorption in the red (δmax= 783 nm; εmax= 28 000 M 1 cm-1) and the ability of singlet oxygen formation recommend 26 P as potential photosensitizer for tumor therapy.  相似文献

Abstract— The transient absorption spectra of the intermediates produced by the 355 nm laser excitation of gilvocarcin derivatives have been investigated in various solvents. The spectra consist of a triplet-triplet absorption in the visible region and a residual absorption observed between 340 and 700 nm due to a long-lived species, assigned to the radical cation. A broad-fast decaying band with a maximum at around 700 nm attributed to the solvated electron is also seen in solutions containing a low DMSO/water volume ratio and at 266 nm irradiation of a 50% methanol/water solvent mixture. The molar absorption coefficient of the triplet state of gilvocarcin V (GV) and gilvocarcin M (GM), determined by the energy transfer method, is independent of the solvent properties and has a value of 3.0 × 104/ M cm. The triplet decay rate constants for both drugs are between 1 and 5 × 104/s. A similar initial yield and triplet decay rate constant of GV were observed in the presence of 3.4 m M thymine. Thus, a quenching rate constant of the GV's triplet state by thymine is estimated to be lower than 106/Ms. The triplet quantum yields of both antibiotics determined by using the comparative method are higher in dimethylsulfoxide (DMSO) (0.18) than are those corresponding to 25% DMSO/water (0.06). The decrease in φT in the presence of water could be attributed to an enhanced internal conversion rate constant from the S1 state or to an increase in the photoionization yield. The similarity of the transient intermediates and their yields for GV and GM suggest that their photobiological differences are due to other factors such as DNA binding constants, preferential localization of the drugs in the cell or the enhanced reactivity of the vinyl group toward cellular components.  相似文献

Abstract— Transient absorption spectra produced by laser flash photolysis of an aqueous solution of 8-methoxypsoralen (8-MOP) have been studied. The biphotonic production of hydrated electrons and of the radical ions, 8-MOP + and 8-MOP- is reported. The hydrated electron was found to react with ground state 8-MOP with k ˜ 3 × 1010 M -1 s-1. In order to obtain a true triplet-triplet absorption spectrum. contributions from the radical ions were subtracted from the overall transient absorption. In addition, contributions from e-aq to the transient spectrum were removed by using N2O, low laser intensity to minimize photoionization or by measuring the transient O.D. after the electron has decayed. These three methods each produced the same triplet-triplet spectrum which differs in the red region from previously reported spectra.  相似文献

Abstract— Although the radiosensitising drugs metronidazole 1 and misonidazole 2 neither phosphoresce nor yield triplet-triplet absorption in the region 350–700 nm, it is, nevertheless, possible to determine their triplet state energies ET by kinetic quenching methods using laser flash photolysis, and also by a steady-state method due to Ullman depending on sensitisation of a colour reaction. Both approaches give acceptable concordance, with figures of 244±4 kJ mol-1 for both 1 and 2. These lead to estimates of triplet excited state one-electron reduction potentials for 1 and 2 in water of 2.04 and 2.14 V vs NHE, enabling an assessment of the possible role of excited 1 and 2 in radiosensitisation.  相似文献

Abstract— The P700 chlorophyll a -protein complex (CPI) isolated from green plants was oriented in aqueous solutions using pulsed electric fields of up to 6700 V cm-1. The electric linear dichroism spectrum is reported in the range of 400–720nm. Positive peaks in the linear dichroism Δ A = A I - A 1 (where AI and A1 are the absorbance components in which the polarizer orientation is parallel and perpendicular with respect to the electric field. respectively) are observed at 443 and 686 nm. The ΔA signal at 686 nm is discussed in terms of either a specialized chlorophyll form absorbing at 686 nm. or due to an exciton component absorbing at the same wavelength.  相似文献

Abstract— Laser flash photolysis studies of DNA-complexed ethidium bromide were undertaken. We have observed a singlet-singlet (S1-Sn) absorption process for DNA-complexed ethidium bromide. The observed lowest singlet excited state lifetime was 21 ± 2 ns. The molar difference extinction coefficient was measured to be 2.4 ± 0.4 × 103M-1 cm-1 at 370 nm. The assignment of this transition was confirmed by time resolved fluorescence measurements.  相似文献
