A sensitive and ultra-fast method utilizing the laser diode thermal desorption ion source using atmospheric pressure chemical ionization coupled to tandem mass spectrometry (LDTD-APCI-MS/MS) was developed for the quantitative analysis of BKM120, an investigational anticancer drug in human plasma. Samples originating from protein precipitation (PP) followed by salting-out assisted liquid-liquid extraction (SALLE) were spotted onto the LazWell? plate prior to their thermal desorption and detection by tandem mass spectrometry in positive mode. The validated method described in this paper presents a high absolute extraction recovery (>90 %) for BKM120 and its internal standard (ISTD) [D8]BKM120, with precision and accuracy meeting the acceptance criteria. Standard curves were linear over the range of 5.00 to 2000 ng mL?1 with a coefficient of determination (R 2) >0.995. The method specificity was demonstrated in six different batches of human plasma. Intra- and inter-run precision as well as accuracy within ±20 % at the lower limit of quantification (LLOQ) and ±15 % (other levels) were achieved during a three-run validation for quality control (QC) samples. The post-preparative stability on the LazWell? plate at room temperature was 72 h and a 200-fold dilution of spiked samples was demonstrated. The method was applied successfully to three clinical studies (n?=?847) and cross-checked with the validated LC-ESI-MS/MS reference method. The sample analysis run time was 10 s as compared to 4.5 min for the current validated LC-ESI-MS/MS method. The resultant data were in agreement with the results obtained using the validated reference LC-ESI-MS/MS assay and the same pharmacokinetic (PK) parameters were calculated for both analytical assays. This work demonstrates that LDTD-APCI-MS/MS is a reliable method for the ultra-fast quantitative analysis of BKM120 which can be used to speed-up and support its bioanalysis in the frame of the clinical trials.  相似文献

A complete characterization of the different physico-chemical properties of nanoparticles (NPs) is necessary for the evaluation of their impact on health and environment. Among these properties, the surface characterization of the nanomaterial is the least developed and in many cases limited to the measurement of surface composition and zetapotential. The biological surface adsorption index approach (BSAI) for characterization of surface adsorption properties of NPs has recently been introduced (Xia et al. Nat Nanotechnol 5:671–675, 2010; Xia et al. ACS Nano 5(11):9074–9081, 2011). The BSAI approach offers in principle the possibility to characterize the different interaction forces exerted between a NP's surface and an organic—and by extension biological—entity. The present work further develops the BSAI approach and optimizes a solid-phase microextraction gas chromatography–mass spectrometry (SPME/GC-MS) method which, as an outcome, gives a better-defined quantification of the adsorption properties on NPs. We investigated the various aspects of the SPME/GC-MS method, including kinetics of adsorption of probe compounds on SPME fiber, kinetic of adsorption of probe compounds on NP's surface, and optimization of NP's concentration. The optimized conditions were then tested on 33 probe compounds and on Au NPs (15 nm) and SiO2 NPs (50 nm). The procedure allowed the identification of three compounds adsorbed by silica NPs and nine compounds by Au NPs, with equilibrium times which varied between 30 min and 12 h. Adsorption coefficients of 4.66?±?0.23 and 4.44?±?0.26 were calculated for 1-methylnaphtalene and biphenyl, compared to literature values of 4.89 and 5.18, respectively. The results demonstrated that the detailed optimization of the SPME/GC-MS method under various conditions is a critical factor and a prerequisite to the application of the BSAI approach as a tool to characterize surface adsorption properties of NPs and therefore to draw any further conclusions on their potential impact on health. We report on the use of quercetin-functionalized gold nanoparticles (QC-AuNPs) as a colorimetric probe for the amino acids arginine (Arg), histidine (His) and lysine (Lys). The method is based on the aggregation of the QC-AuNPs that is caused by these amino acids and leads to a visually detectable color change from red to blue. The absorption maxima shift from 525 nm to 702, 693, and 745 nm, respectively. Aggregations are confirmed by dynamic light scattering (DLS) and transmission electron microscopic techniques (TEM). The effects of the QC concentration, temperature and reaction time for the preparation of QC-Au NPs were tested. Other amino acids do not interfere. Under the optimal conditions, linear relationships exist between the absorption ratios at 702/525 nm (for Arg), 693/525 nm (for His), and 745/525 nm (for Lys) over the concentrations ranges from 2.5–1,250 μM (Arg) and 1–1,000 μM (His and Lys), respectively. The respective limits of detection are 0.04, 0.03, and 0.02 μM. The method provides a useful tool for the rapid visual and instrumental determination of the three amino acids. Figure

Nonsteroidal anti-inflammatory drugs (NSAIDs) are the group of drugs having the therapeutic efficacy of analgesic and antipyretic. To detect health-threatening residues of NSAIDs, a fast and easy multiresidue method based on liquid chromatography tandem mass spectrometry (LC-MS/MS) was described. Ten NSAIDs were extracted from the tissues using 2 mL of acetonitrile and 0.1 mL of 2 mM ammonium formate in distilled water. After clean-up using C18 sorbent, it was evaporated under nitrogen, reconstituted with 1 mL distilled water and analyzed by LC-MS/MS. The method was validated based on guideline for residue testing laboratory. Furthermore, the method has also been applied successfully to detect ten NSAIDs from bovine, porcine, and chicken liver tissues. In a total of 315 liver samples tested, acetylic salicylic acid was detected from 28 porcine and 2 chicken liver tissues at levels of 13?~?576 and 50?~?53 ng/g, respectively. Subsequently, paracetamol was detected in 15 porcine liver tissues with a detection levels of 28?~?381 ng/g. Phenylbutazone and its metabolite, oxyphenylbutazone, were detected at 247 and 15 ng/g range in one of the bovine liver tissue, respectively.  相似文献

A novel direct method for the determination of EDTA in alkaline radioactive evaporator residue solution was developed and validated based on ion chromatography with suppressed conductimetric detection and anion exchange columns (A Supp 4, 4 mm × 250 mm and A Supp 5, 4 mm × 150 mm). The yttrium-EDTA complex resulted one single chromatographic peak in the eluent and allowed the correct determination of EDTA in an alkaline, high concentration radioactive waste water. Depending on coexisting substances, suitable eluent is 10.0 mM carbonate buffer/pH 10.6 or 10.75 (t R,Y–EDTA = 7.01 and 6.4 min, respectively). For 10.0 mM carbonate buffer/pH 10.6 and isocratic flow rate of 1.0 cm3/min, a linear calibration curve was obtained from 5 to 40 mg/dm3 (r > 0.999) EDTA. Good resolution was achieved from commonly coexisting anions (chloride, nitrite, nitrate, sulphate, phosphate, bromide and citrate). The developed simple ion chromatographic method was applied for the assay of EDTA in various radioactive alkaline solutions.  相似文献

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) has been a major approach for glycan analysis. However, the preferential cleavage of the sialic acid moiety by in- and post-source decay influences the determination of sialylated glycans by MALDI-MS. Many chemical derivatization methods were introduced to stabilize the sialylated glycan during MALDI-MS. Among current derivatization methods, methylamidation is a promising means for simultaneous analysis of natural sialylated glycans regardless of their sialic acid linkage types. Here, a novel derivatization method was developed, in which proteins were conjugated on the solid-phase support in order to stabilize the sialic acids by methylamidation and to reduce sample loss and contamination during the derivatization process. This derivatization strategy was used to investigate N-glycans from fetuin, a glycoprotein containing different types of complex N-glycans. The developed method was also applied to the N-glycan profiling of human serum from patients and healthy volunteers. Results were consistent with N-glycan profiling by HPLC-fluorescence detection. This new method provided a sensitive, simple, and robust approach for profiling glycan structures of complex samples.  相似文献

A highly porous silica-supported tungstophosphoric acid (PW) nanocluster was prepared for use in solid-phase microextraction (SPME) of polycyclic aromatic hydrocarbons (PAHs). The PWs represent a class of discrete transition metal-oxide nanoclusters and their structures resemble discrete fragments of metal-oxide structures of definite size and shape. Transition metal-oxide nanoclusters display large structural diversity, and their monodisperse sizes can be tuned from several Ångstroms up to 10 nm. The highly porous silica-supported tungstophosphoric acid nanocluster material is found to be capable of efficiently extracting PAHs from aqueous sample solutions. The nanomaterial was immobilized on a stainless steel wire for fabrication of the SPME fiber. Following thermal desorption, the PAHs were quantified by GC-MS. Analytical merits include limits of detection that range from 0.02 to 0.1 pg mL?1 and a dynamic range as wide as from 0.001 to 100 ng mL?1. Under optimum conditions, the repeatability for one fiber (n?=?3), expressed as the relative standard deviation, is between 4.3 % and 8.6 %. The method is simple, rapid, and inexpensive. The thermal stability of the fiber and the high relative recovery make this method superior to conventional methods of extraction.

We report herein the development of a highly sensitive colorimetric method for detection of d-Penicillamine using citrate-capped gold nanoparticles (AuNPs). This assay relies upon the distance-dependent of gold nanoparticles surface plasmon resonance band of gold nanoparticles. By replacing the thiol-containing chelator drug, d-Penicillamine, with citrate on the gold nanoparticles surface, a new peak appearing at a longer wavelength intensifies and shifts further to the red from the original peak position due to aggregation of gold nanoparticles which depends on ionic strength, gold nanoparticles and d-Penicillamine concentration. During this process, the plasmon band at 521 nm decreases gradually along with the formation of a new red-shifted band at 630 nm. The calibration curve which is derived from the ratio intensities of absorbance at longer wavelength (630 nm) to original wavelength (521 nm) displays a linear relation in the range of 5.0 × 10?6–3.0 × 10?4 M d-Penicillamine. Lower limit of detection for d-Penicillamine, at the signal-to-noise ratio of 3 (3σ), was 3.8 × 10?6 M. The developed methodology was successfully applied for the determination of d-Penicillamine in human urine and plasma.  相似文献

A stability-indicating ultra-high-performance liquid chromatography (UHPLC) method with a diode array detector was developed and validated for the determination of cis/trans isomers of perindopril l-arginine in bulk substance and pharmaceutical dosage form. The separation was achieved on a Poroshell 120 Hilic (4.6 × 150 mm, 2.7 µm) column using a mobile phase composed of acetonitrile–0.1 % formic acid (20:80 v/v) at a flow rate of 1 mL min?1. The injection volume was 5.0 µL and the wavelength of detection was controlled at 230 nm. The selectivity of the UHPLC-DAD method was confirmed by determining perindopril l-arginine in the presence of degradation products formed during acid–base hydrolysis and oxidation as well as degradation in the solid state, at an increased relative air humidity and in dry air. The method’s linearity was investigated in the ranges 0.40–1.40 µg mL?1 for isomer I and 0.40–2.40 µg mL?1 for isomer II of perindopril l-arginine. The UHPLC-DAD method met the precision and accuracy criteria for the determination of the isomers of perindopril l-arginine. The limits of detection and quantitation were 0.1503 and 0.4555 µg mL?1 for isomer I and 0.0356 and 0.1078 µg mL?1 for isomer II, respectively.  相似文献

A method for the multimeric high-performance liquid chromatography of plant sterols is proposed which permits the separation of compounds close in structure with similar chromatographic properties. The first stage includes the chemical modification of the sterols with the aid of a bis(p-nitrophenyl) phosphate group. The phosphotriesters formed as the result of the reaction are separated by normal-phase HPLC. The compounds isolated are treated with ammonia, as a result of which the sterol phosphotriesters are converted into the corresponding phosphodiesters. These phosphodiesters are then subjected to reversed-phase HPLC. The chromatographic separation of UV-absorbing sterol derivatives using several variants of HPLC substantially increases the resolving power of the method.  相似文献

We describe a new chemiluminescence (CL) system based on the oxidation of rhodamine B (RhoB) with alkaline potassium permanganate in the presence of gold nanoparticles (Au-NPs) and anionic detergent sodium dodecyl sulfate. Free RhoB is weakly chemiluminescent when oxidized with permanganate at alkaline pH values. However, a remarkably strong enhancement of CL is observed in the presence of Au-NPs, probably due to a strong interaction between RhoB and the NPs. The possible mechanism was studied via recording the CL emission. It is also found that the intensity of CL gradually decreases in the presence of cyanide due to its interaction with the Au-NPs. The relation between the decreased CL intensity and cyanide concentration was exploited to develop a method for the determination of cyanide in the 0.01–0.5 μM concentration range, with a detection limit of 2.8 nM. In this work, a novel ionic liquid (IL) chemically bonded sol–gel coating was prepared for stir bar sorptive extraction (SBSE) of nonsteroidal anti-inflammatory drugs (NSAIDs) followed by high-performance liquid chromatography-ultraviolet detection (HPLC-UV). By using γ-(methacryloxypropyl)trimethoxysilane (KH-570) as a bridging agent, 1-allylimidazolium tetrafluoroborate ([AIM][BF4]) was chemically bonded onto the bare stir bar, and the prepared IL-bonded sol–gel stir bar coating showed higher extraction efficiency and better adsorption/desorption kinetics for target NSAIDs over other polydimethylsiloxane (PDMS)-based or monolithic stir bar coatings. The mechanical strength and durability (chemical/thermal stability) of the prepared IL-bonded sol–gel coating were excellent. The influencing factors of SBSE, such as sample pH, salt effect, stirring rate, extraction time, desorption solvent, and desorption time, were optimized, and the analytical performance of the developed SBSE-HPLC-UV method was evaluated under the optimized conditions. The limits of detection (LODs) of the proposed method for three NSAIDs were in the range of 0.23–0.31 μg L?1, and the enrichment factors (EFs) were in the range of 51.6–56.3 (theoretical enrichment factor was 100). The reproducibility was also investigated at concentrations of 5, 20, and 100 μg L?1, and the relative standard deviations (RSDs) were found to be less than 9.5, 7.5, and 7.6 %, respectively. The heat of reaction and kinetics of curing of diglycidyl ether of bisphenol-A (DGEBA) type of epoxy resin with catalytic amounts of ethylmethylimidazole (EMI) have been studied by differential power-compensated calorimetry as a part of the program for the study of process monitoring for composite materials. The results were compared with those from 1∶1 and 1∶2 molar mixtures of DGEBA and EMI. A method of determination of heat of reaction from dynamic thermoanalytical instruments was given according to basic thermodynamic principles. The complicated mechanism, possibly involving initial ionic formation, has also been observed in other measurements, such as by time-domain dielectric spectroscopy. The behavior of commercially available DGEBA resin versus purified monomeric DGEBA were compared. The melting point of purified monomeric DGEBA crystals is 41.4 °C with a heat of fusion of 81 J/g. The melt of DGEBA is difficult to crystallize upon cooling. The glass transition of purified DGEBA monomer occurs around ?22 °C with aΔC p of 0.60 J/K/g.  相似文献

Methylisothiocyanate (MITC) is the main degradation product of metam sodium, a soil disinfectant widely used in agriculture, and is responsible for its disinfectant properties. Because MITC is highly toxic and volatile, metam sodium has to be applied in a manner that tries to reduce atmospheric emissions but still maintains adequate concentration of MITC in soil to ensure its disinfectant effect. Thus, monitoring of MITC concentrations in soil is required, and to this end sensitive, fast, and reliable analytical methods must be developed. In this work, a headspace solid-phase microextraction (HS-SPME) method was developed for MITC determination in water and soil samples using gas chromatography-tandem mass spectrometry (GC–MS–MS) with a triple-quadrupole analyzer. Two MS–MS transitions were acquired to ensure the reliable quantification and confirmation of the analyte. The method had linear behavior in the range tested (0.026–2.6 ng mL?1 in water, 1–100 ng g?1 in soil) with r 2 over 0.999. Detection limits were 0.017 ng mL?1 and 0.1 ng g?1 in water and soil, respectively. Recoveries for five replicates were in the range 76–92 %, and RSD was below 7 % at the two spiking levels tested for each matrix (0.1 and 1 ng mL?1 for water, 4 and 40 ng g?1 for soil). The potential of using multiple HS-SPME for analyzing soil samples was also investigated, and its feasibility for quantification of MITC evaluated. MicroRNAs (miRNAs), a class of small endogenous nonprotein-coding RNAs, regulate a wide range of biological processes, and their abnormal expressions are related to the growth and development of plants. Thus, a simple, rapid, and highly sensitive assay for miRNA detection is of great significance. In this work, a label-free and ultrasensitive assay for miRNA detection using protein cage nanoparticles has been developed. Apoferritin-encapsulated Cu nanoparticles (Cu-apoferritin) could be immobilized on the electrode through special reaction between amino and carboxyl. Hybridization event between the probe DNA and the target miRNA-159a is confirmed by electrochemical oxidation signal after Cu released into the detection buffer by adjusting the pH. This assay is highly selective and sensitive with a low detection limit of 3.5 fM. Moreover, the developed method can even discriminate single-base mismatched strand between the complementary targets. The effect of abscisic acid on the expression level of miRNA-159a in Arabidopsis thaliana seeds was also investigated.  相似文献

This study evaluates solid-phase micro-extraction (SPME) coupled with gas chromatography–mass spectrometry (GC–MS) to determine trace levels of bis-phenol A in water and leached from plastic containers. In our study, we used very thin composite membranes prepared in the laboratory. The extraction using headspace post-derivatization with bis(trimethylsilyl) trifluoroacetamide (BSTFA), containing 1 % trimethylchlorosilane (TMCS) vapor, following SPME was compared with extraction without derivatization. The SPME experimental procedures to extract bis-phenol A in water were optimized with a relatively polar polyacrylate (PA)-coated fiber, an extraction time of 50 min, and desorption at 300 °C for 2 min. Headspace derivatization following SPME was performed using 7 μL of BSTFA with 1 % TMCS at 65 °C for 30 s. The precision was 5.2 % without derivatization and 9.0 % headspace derivatization. The detection limit was determined to be at the nanogram per liter level. When SPME was used following headspace derivatization, the detection limit was one order of magnitude better than that achieved without derivatization. The results of this study reveal the adequacy of the SPME–GC–MS method for analyzing bisphenol A leached from plastic containers. The concentrations of bisphenol A leached from plastic containers into water ranged from 0.7 to 78.5 μg L?1.  相似文献

Free fatty acids (FFAs), major cellular metabolites, play an important role during tumor pathogenesis. Enhanced de novo fatty acid synthesis in tissues is a characteristic feature of cancer. Therefore, measurement of FFA concentration in biological samples is beneficial for cancer research and clinical diagnosis. Herein, a rapid, stable, and sensitive detection methodology was established to simultaneously quantify 22 FFAs using high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (HPLC/ESI–MS/MS). The HPLCMS/MS system was run in negative ion mode for 15 min using multiple reaction monitoring. The lipids were extracted from colon tissues of colon cancer patients and then injected into the HPLCMS/MS system for analysis. Colon samples were analyzed by inter-day repeatability and intra-day repeatability, with less than 5 % deviation for most fatty acids. This approach is successful to determine low picogram concentrations of each FFA molecule using milligrams of tissue, and provides a promising method for FFA microanalysis in clinical samples.  相似文献

The authors present an application of the PIXE (Particle Induced X-ray Emission) method: determination of trace element concentrations in uterine tissue after wet digestion by HNO3. A method for calibration of the system is described. Results of the analysis of a NBS reference material (National Bureau of Standards), SRM 1577 bovine liver, are given and possible sources of error are discussed. In the authors’ experience, the combination of wet digestion and PIXE provides a tool for reliable analysis of trace element concentrations in tissues.  相似文献

β-phase polyvinylidene fluoride (PVDF)–BaTiO3 nanocomposite samples have been prepared by solution mixing method. XRD data represent that the crystallinity of PVDF decreases with increase in loading level of BaTiO3 nanoparticles. DSC curve represents that the melting point of PVDF is lightly affected by loading concentration of BaTiO3. The morphology and microstructure of PVDF and PVDF embedded by BaTiO3 nanofillers were investigated by using inverted contrast microscopy (ICM) and scanning electron microscopy (SEM). FTIR interferrometry is proven that PVDF and BaTiO3 are not chemically interacting; therefore, interaction of BaTiO3 is van der Waals type of interaction. The thermally stimulated discharge current (TSDC) of PVDF and PVDF–BaTiO3 nanocomposites sample was characterized by single peak. The observed TSDC peak is discussed on the basis of dipolar and interfacial polarization.  相似文献
