用反相离子对高效液相色谱法初步研究了铝与儿茶酚胺的相互作用。结果显示,去甲肾上腺素的峰高随加入铝的增加而线性下降,铝与去甲肾上腺素的配住比为1:1。该项研究为进一步活体测定铝提供了一条新思路。  相似文献

A method developed using the combination of HPLC and AAS enabling both qualitative and quantitative study of the protein binding and speciation of trace metals in biological fluids at clinical relevant levels is reviewed. The whole system was made metal-free by using polymer-based columns, column holders and tubing and by the insertion of a silica-based scavenger column placed immediately before the injection valve to selectively retain any trace of aluminum and iron originating from buffer solutions and recipients. ETAAS instrumental conditions were carefully selected to eliminate interferences secondary to the salt gradient elution. Particular attention was paid to the choice of HPLC columns. Protein recoveries varied between 95 and 105% and trace metal recoveries were close to 100%. Intra-assay and inter-assay CVs of the HPLC/AAS hybrid technique were below 10%. Because of its high sensitivity, the method can be used at clinically relevant concentrations and was applied successfully to study (i) the interaction between iron and aluminum for binding to transferrin, (ii) the influence of citrate on the transferrin binding of aluminum, (iii) the speciation of silicon in the serum of dialysis patients and (iv) the toxicity of mercury compounds in cell cultures.  相似文献

Indole alkaloid of Cathuranthus roseus was purified by applying aluminum oxide column chromatography and an HPLC method was established to determine it. Applying aluminum oxide column chromatography produced a high purified monomer of alkaloids; the extracts of indole alkaloid were then loaded on the neutral aluminum oxide column chromatography and washed with an organic solvent (a mixture of chloroform, ether, and petroleum ether). Effect of purification was the best when the proportion of chloroform, ether, and petroleum ether was 10:10:1. To determine the content of vindoline, vinblastine, and cathuranthine exactly, the mobile phase of HPLC was water (containing 0.005 mol·L?1 ammonium hydrogen phosphate), methanol (containing 0.67% triethylamine), and acetonitrile, the proportion of each was nonlinear.  相似文献

Boron, at sub-ppm levels, in U3O8 powder and aluminum metal, was determined using complex formation and dynamically modified reversed-phase high-performance liquid chromatography (RP-HPLC). Curcumin was used for complexing boron extracted with 2-ethyl-1,3-hexane diol (EHD). Separation of complex from excess reagent and thereafter its determination using the online diode array detector (DAD) was carried out by HPLC. Calibration curve was found to be linear for boron amounts in the sample ranging from 0.02 microg to 0.5 microg. Precision of about 10% was achieved for B determination in samples containing less than 1 ppmw of boron. The values obtained by HPLC were in good agreement with the data available from other analytical techniques. The precision in the data obtained by HPLC was much better compared to that reported by other techniques. The present hyphenated methodology of HPLC and complex formation reaction is interesting because of cost performance, simplicity, versatility and availability when compared to other spectroscopic techniques like ICP-MS and ICP-AES.  相似文献

采用高效液相色谱同时检测水产品中孔雀石绿、结晶紫及无色孔雀石绿和无色结晶紫的残留量,样品经提取、净化处理后所得残渣用乙腈溶解后,通过采用C_(18)色谱柱,以乙腈(A)和pH3.0的0.02 mol·L~(-1)磷酸二氢钾缓冲溶液(B)按不同比例混合进行梯度淋洗,实现孔雀石绿、结晶紫及其代谢物的分离。用自制的二氧化铅柱氧化无色孔雀石绿及无色结晶紫。在588 nm波长处,测定4种物质的质量浓度在0.3~6.0 mg·L~(-1)范围内与其峰面积呈线性关系,相对标准偏差(n=6)小于2.5%,检出限(3S/N)小于1.9μg·kg~(-1),分析时间20 min。以凤尾鱼罐头为基体进行回收试验,方法的回收率在71.5%~88.6%范围。  相似文献

Summary The reversed-phase ion-pair liquid chromatographic behaviour of several metal complexes with 5-sulphoquinoline-8-ol (HQS) on an octadecyl-modified silica column was investigated by using a fluorescence detector. The aluminum complex gave two peaks on the chromatograms when eluents containing phosphate buffer were used. The effects of the pH of the eluent, HQS concentration, the nature of anions in the eluent, and the column temperature on the retention behaviour of the aluminum complex are discussed on the basis of the equilibria and the kinetics of the complexation of the aluminum ion with HQS and anions in the eluent. The equilibrium constant for the interconversion reaction between the two Al(III)-HQS species which correspond to the two peaks observed and the second stepwise formation constant for the Zn(II)-HQS complex were evaluated from the HPLC data obtained.  相似文献

The latest developments and in particular important synthetic aspects for the preparation of modern HPLC supports are reviewed. In this context, the chemistry of inorganic supports based on silica, zirconia, titania or aluminum oxide as well as of organic supports based on poly(styrene-divinylbenzene), acrylates, methacrylates and other, more specialized polymers is covered. Special consideration is given to modern approaches such as sol-gel technology, molecular imprinting, perfusion chromatography, the preparation of monolithic separation media as well as to organic HPLC supports prepared by new polymer technologies such as ring-opening metathesis polymerization. A method for HPLC/FTIR is discussed in which the effluent from a microbore HPLC is continuously deposited onto and eliminated from the surface of a circular rotating germanium crystal that has been aluminum coated on its opposite side. After deposition, the germanium disk is again rotated, this time in the sample compartment of an FTIR spectrometer while reflectance-absorbance infrared spectra are continuously collected. The novel germanium-aluminum deposition surface allows collection of reflectance-absorbance spectra that are free of the degrading effects of superposition phenomena characteristic of reflectance-absorbance spectra obtained on metal surfaces. Furthermore, germanium is impervious to aqueous solvent mixtures and, therefore, allows for the direct deposition of reversed phase separations, including those requiring acid modified mobile phases.  相似文献

A novel and reliable high-performance liquid chromatographic method for trace short-chain organic acids measurement in wheat root exudates under aluminum stress has been optimized and validated. The chromatographic separation of the short-chain organic acids (citric, oxalic, malonic, succinic, tartaric, malic, and acetic acids) was achieved with Bio-rad Aminex HPX-87H cation exchange resin column. These seven organic acids were identified and quantified in 25 min. Well-shaped peaks were obtained for trace organic acids using dilute sulfuric acid as mobile phase. Under optimum conditions, Bio-rad Aminex HPX-87H column showed distinct advantages of the ability to well separate different short-chain organic acids (especially for tartaric and malic acids, as well as malonic and acetic acids) in wheat root exudates under aluminum stress, and offered accurate and precise results for the analysis of these organic acids. This HPLC method can efficiently eliminate the aluminum’s interference and is quite suitable to the trace detection of trace organic acids in wheat root exudates under aluminum stress.

In High Performance Flow Atomic Spectrometry (HPF-AS), the aerosol is generated by injecting the sample solution into the gas mixing chamber with the aid of an HPLC pump (Hydraulic High Pressure Nebulization, HHPN, working pressure about 200 bar). Using an additional analytical HPLC column, between sample injection valve and nebulization nozzle, the elements under investigation can be separated from interfering elemental traces or major components (High Performance/Pressure Flow System, HPF). The elements to be determined and the interfering substances reach the flame at different times. This means, for example, that traces of Mg in solutions containing aluminum can be determined without interference. Compared to pneumatic nebulization, there is a 20 fold increase in the detection power when determining calcium in a saturated NaCl solution.
