Reactions of 2-(allyloxymethyl)- and 2-[2-(vinyloxy)ethoxy]methyloxiranes with 2-propynol (~3 wt % of t-BuOK, 75–85°C, 5–10 h) lead to formation of new 1-organyloxy-3-(2-propynyloxy)propan-2-ols (yield 65–95%). On heating to 45–100°C in the presence of bases (KOH, t-BuOK), 1-allyloxy- and 1-[2-(vinyloxy)ethoxy]-3-(2-propynyloxy)propan-2-ols are transformed into the corresponding 2-vinyl-1,3-dioxolane, 6-methyl-2,3-dihydro-1,4-dioxine, 6-methylene-1,4-dioxane, and 2,3-dihydro-5H-1,4-dioxepine derivatives, whose yield and ratio strongly depend on the solvent nature, catalyst, and substituent at the hydroxy group. 2-Vinyl-1,3-dioxolane and 6-methyl-2,3-dihydro-1,4-dioxine derivatives are formed as the major products (yield 70–99%) in the presence of t-BuOK in aprotic media (toluene, THF, DMSO) or in the absence of a solvent as a result of prototropic isomerization followed by intramolecular heterocyclization. Intramolecular nucleophilic cyclization of 3-(2-propynyloxy)propan-2-ols to 6-methylene-1,4-dioxane is the predominant process in water in the presence of KOH. In all cases, the fraction of 2,3-dihydro-5H-1,4-dioxepine derivatives among the cyclization products ranges from 0 to 5% (KOH) or to 14% (t-BuOK).  相似文献

An investigation of the one-carbon homologation of some 1-tribromomethyl-isoquinoline and 2-tribromomethyl-quinoline derivatives was conducted. Under the influence of an aqueous solution of silver nitrate in the presence of a nucleophilic species (MeOH, H2O, EtNH2), these derivatives led to the respective expanded heterocycles, 2H-1- and 2H-3-benzazepin-2-one derivatives. A mechanism for this novel ring enlargement involving initial formation of an aziridinium, and its subsequent opening to form a stabilized benzylic carbocation, is proposed to explain the results.  相似文献

The preparation of both (2S)- and (2R)-1-monoricinolein derivatives has been developed to synthesize ferroelectric liquid crystals. Lipase-catalyzed hydrolysis of 2-protected-1,3-diricinoleins provided (2S)-1-monoricinolein derivatives with high diastereoselectivities, while transesterification of 2-protected glycerols with vinyl recinoleate gave (2R)-1-monoricinolein counterparts in good yields with high diastereoselectivities.  相似文献

Ethyl 2-amino-5-(2-aryl-2-oxoethylidene)-4-oxo-1H-4,5-dihydrofuran-3-carboxylates and 2-amino- 5-(2-aryl-2-oxoethylidene)-4-oxo-1H-4,5-dihydrofuran-3-carbonitriles reacted with alcohols in the presence of a catalytic amount of concentrated aqueous HCl to give the corresponding 5-alkoxy-2-amino-5-(2-aryl-2- oxoethyl)-4-oxo-1H-4,5-dihydrofuran-3-carboxylic acid derivatives. A probable reaction mechanism was proposed on the basis B3LYP/6-311G(d) quantum chemical calculations.  相似文献

A new protocol for the synthesis of 2-allylindole and 2-allylbenzofuran derivatives has been developed from readily accessible starting material, 2-((trimethylsilyl)ethynyl)arenes via Pd-catalysis. The presence of trimethylsilyl group in the alkyne is vital for this reaction. Stereoselectivity of 2-allylindole derivatives is controlled by the use of different nitrogen-protecting groups for 2-((trimethylsilyl)ethynyl)aniline. The CO2Me protection gives Z-isomers, whereas acetyl protection gives E-isomers.  相似文献

Diastereoselective lithiation of (S)-2-ferrocenyl-4-(1-methylethyl)oxazoline, followed by addition of N2O4, gave (S)-2-[(pS)-2-nitroferrocenyl]-4-(1-methylethyl)oxazoline which was subsequently converted into derivatives of (pS)-2-aminoferrocenecarboxylic acid. The corresponding (pR)-derivatives were obtained through use of a removable TMS blocking group. The 2-nitroferrocenyloxazolines produced in this work underwent facile photo-decomplexation to give 2-nitrocyclopentadienyliden-1,3-oxazolidenes.  相似文献

Two series of new adamantyl derivatives of polynitrogen heterocycles, 2-(adamantan-1-yl)-5-aryl- 1,3,4-oxadiazoles and 2-(adamantan-1-yl)-5-aryl-2H-tetrazoles, have been synthesized, and their structure has been determined by NMR spectroscopy, mass spectrometry, and X-ray analysis. Biological studies in vitro have revealed high inhibitory activity of some of the synthesized 2-(adamantan-1-yl)-5-aryl-2H-tetrazoles against H1N1 influenza A viruses in combination with a relatively low selectivity.  相似文献

A series of novel derivatives of the 1,3-tropolone (β-tropolone) system—2-(2-quinolyl)-5,6,7-trichloro-1,3-tropolones and 2-(2-quinolyl)-4,5,6,7-tetrachloro-1,3-tropolones have been prepared by the acid-catalyzed reaction of 2-methylquinolines with 3,4,5,6-tetrachloro-1,2-benzoquinone. The molecular structures of two compounds, 2-(4-chloro-6,8-dimethyl-5-nitro-2-quinolyl)-5,6,7-trichloro-1,3-tropolone 8 and 2-(4-chloro-7,8-dimethyl-5-nitro-2-quinolyl)-4,5,6,7-tetrachloro-1,3-tropolone 9, have been determined using X-ray crystallography. According to the performed DFT B3LYP/6-311++G∗∗ calculations the tautomeric (OH) and (NH) forms of β-tropolones 8 and 9 are nearly energy equivalent, the latter being more stabilized in polar media. Photolysis of 2-(2-quinolyl)-1,3-tropolones in heptane solution leads to the disrotatory electrocyclic rearrangement resulting in the formation of a mixture of E- and Z-isomers of 3-[2(1H)-quinolinylyden]-bicyclo[3.2.0]hept-6-en-2,4-dione derivatives.  相似文献

A series of novel 5-[(Z,2Z)-2-chloro-3-(4-nitrophenyl)-2-propenylidene]-thiazolidinones (Ciminalum–thiazolidinone hybrid molecules) have been synthesized. Anticancer activity screening toward the NCI60 cell lines panel, gastric cancer (AGS), human colon cancer (DLD-1), and breast cancer (MCF-7 and MDA-MB-231) cell lines allowed the identification of 3-{5-[(Z,2Z)-2-chloro-3-(4-nitrophenyl)-2-propenylidene]-4-oxo-2-thioxothiazolidin-3-yl}propanoic acid (2h) with the highest level of antimitotic activity with mean GI50/TGI values of 1.57/13.3 μM and a certain sensitivity profile against leukemia (MOLT-4, SR), colon cancer (SW-620), CNS cancer (SF-539), melanoma (SK-MEL-5), gastric cancer (AGS), human colon cancer (DLD-1), and breast cancers (MCF-7 and MDA-MB-231) cell lines. The hit compounds 2f, 2i, 2j, and 2h have been found to have low toxicity toward normal human blood lymphocytes and a fairly wide therapeutic range. The significant role of the 2-chloro-3-(4-nitrophenyl)prop-2-enylidene (Ciminalum) substituent in the 5 position and the substituent’s nature in the position 3 of core heterocycle in the anticancer cytotoxicity levels of 4-thiazolidinone derivatives have been established  相似文献

A green and highly efficient strategy for the preparation of bridged spirocyclic compounds via visible-light-induced cyclization of 2-(2-(arylethynyl)benzylidene)malononitrile derivatives with 2,6-di(tert-butyl)-4-methylphenol (BHT) at room temperature was developed. In trifluoromethanesulfonic acid, 2-nitromethylene-1-(ω-phenylalkyl)imidazolidine or 2-nitromethylene-1-(ω-phenylalkyl)hexahydropyrimidine derivatives undergo an intramolecular cyclization to afford (E)-hydroxyiminohydroazaazoniabenzocycloalkenes, in their trifluoromethanesulfonate salt form. The reaction probably occurres via the formation of an electrophilic transient hydroxynitrilium ion (or O-protonated nitrile oxide). Alkylation of 3-acetyl-5-(alkoxymethyl)tetrahydrofuran-2-ones gave 3-acetyl-5-(alkoxymethyl)-tetrahydrofuran-2-ones containing various substituents in position 3 of the heterocycle. Bromination of the latter afforded 3-(bromoacetyl) derivatives which reacted with various nucleophiles to produce derivatives containing benzimidazole, 2-amino-1,3-thiazole, 3-chloropyridine, and 3,5-dibromopyridine fragments together with the butanolide ring.  相似文献

A simple route to 1-R-3-(2-indolyl)-1-propanones has been elaborated based on recyclization of 2-(2-aminobenzyl)furan derivatives. Being a modification of the Reissert indole synthesis, our approach employs the furan ring as a source of carbonyl function. Hydrazones derived from substituted benzophenones and fluorenone reacted with 4-aryl-2-hydroxy-4-oxobut-2-enoic and 2-hydroxy-5,5-dimethyl-4-oxohex-2-enoic acids to give the corresponding 2-(2-ylidenehydrazino) derivatives which may be used as initial compounds for the synthesis of 3-hydrazono-3H-furan-2-ones. The obtained but-2-enoic and hex-2-enoic acid derivatives in solution may exist as Z- and E-isomeric enehydrazine tautomers or hydrazone tautomers with syn or anti orientation of substituents with respect to the double C=N bond.  相似文献
