Sodium borohydride is stabilized on poly(n-butyl-4-vinylpyridinium) chloride, and it is used as an efficient and regenerable polymer-supported borohydride reagent for the reduction of a variety of carbonyl compounds, such as aldehydes, ketones, α,β-unsaturated carbonyl compounds, α-diketones, and acyloins.  相似文献

Lithium α-sulfinyl carbanions of 1-chloroalkyl p-tolyl sulfoxides were reacted with carbonyl compounds to afford adducts in high to quantitative yields. The adducts were treated with t-BuMgCl or LDA to give magnesium or lithium alkoxides, which were treated with i-PrMgCl or t-BuLi to afford the enolate with one-carbon elongation through β-oxido carbenoids. The enolate intermediates were found to be able to be trapped with electrophiles to give α,α-disubstituted carbonyl compounds in moderate to good yields. As a whole, this procedure offers a new and good method for synthesis of α,α-disubstituted carbonyl compounds from carbonyl compounds with one-carbon homologation in only two synthetic steps.  相似文献

The addition reaction of carbonyl compounds with lithium α-sulfinyl carbanions of 1-chloroalkyl p-tolyl sulfoxides gave adducts, α-chloro β-hydroxy sulfoxides, in high to quantitative yields. The adducts were first treated with a base to give alkoxides, which were treated with i-PrMgCl or t-BuLi to give β-oxido carbenoids via a sulfoxide-metal exchange reaction. The β-oxido carbenoid rearrangement then took place to afford the enolates with one-carbon elongation. The enolate intermediates were found to be able to be trapped with electrophiles such as aldehydes, ethyl chloroformate, benzoyl chloride, haloalkanes to give α,α-disubstituted carbonyl compounds in moderate to good yields. This method provides a new and efficient way for synthesis of α,α-disubstituted carbonyl compounds from carbonyl compounds with one-carbon homologation in only two synthetic operations.  相似文献

An efficient synthesis of biologically interesting α-CF3-trifluoroalanine derivatives bearing N-(diethoxyphosphoryl)difluoroacetyl moiety in good yields has been elaborated. The key substrate, α-TFM-imino ester, prepared from methyl trifluoropyruvate and (diethoxyphosphoryl)difluoroacetamide, followed by dehydration, was subjected to regiospecific reactions with various nucleophiles, including organometallic, π-donor compounds and sodium borohydride. These novel products may find a viable application in the modification of peptides or serve as potential drug candidates.  相似文献

In the presence of iron dust, diaryl disulfides react with α-bromo carbonyl compounds to obtain α-arylthio carbonyl compounds in good yield at 90 °C under an N2 atmosphere, using commercial dimethylformamide (DMF) as solvent. The possible reaction mechanism is that the disulfide is reduced by iron dust to give ArSFeSAr and then reacted with α-bromo carbonyl compounds to give product α-arylthio carbonyl compounds and FeBr2.  相似文献

α,α-Difluoro-α-phenylsulfanyl-α-trimethylsilylmethane (PhSCF2SiMe3) has been demonstrated as an α,α-difluoro-α-phenylsulfanylmethyl carbanion equivalent. gem-Difluorophenylsulfanylmethylation of carbonyl compounds has been successfully achieved by using PhSCF2SiMe3 in the presence of TBAF in THF. The adducts have been converted to the corresponding gem-difluoroalkenes by a novel pyrolytic and/or FVP elimination of the β-hydroxy-α-phenylsufinyl derivatives under reduced pressure.  相似文献

The elimination of benzotriazolyl group from N-(α-benzotriazol-1-ylalkyl)amides and N-(α-benzotriazol-1-ylalkyl)sulfonamides are readily realized with samarium diiodide as a reducing agent. The resulting intermediates undergo a dimerization or cross-coupling reaction with carbonyl compounds, thus affording the corresponding dimers or α-hydroxyalkylated sulfonamides in moderate yields.  相似文献

Abstract

Sodium borohydride adsorbed onto silica gel has now been utilized for on-line, pre-and post-analytical column chemical derivatizations via reductions of various organic carbonyl compounds. These on-line reactions have been performed using normal phase HPLC conditions, involving conventional silica gel packings, organic mobile phases, and commercially available HPLC equipment and instrumentation. This approach for on-line HPLC derivatizations has been evaluated for a large number of organic carbonyl compounds, at a variety of temperatures for aldehydes and ketones. The overall rates of such carbonyl reductions via sodium borohydride/silica are sufficiently different as a function of temperature of the reaction to permit for compound/class identifications. Analyte identification can be both qualitative and quantitative, even wherein an analyte co-elutes with a non-carbonyl compound. In-house prepared borohydride/silica gel reactors can be characterized via standard iodine titration procedures along with inductively coupled plasma (ICP) elemental boron analysis. These normal phase derivatization approaches have been applied to certain standard vitamins, and to cinnamaldehyde found in a commercial spice product. Heterodiene [4π+2π] cycloadditions of (S,S)-4,5-diaryl-2-methylene-1,3-dioxolanes 1 to a series of β-amido-α,β-unsaturated carbonyl compounds are diastereoselective (d.r.≥4:1). The products can be purified by trituration or crystallisation and hydrolysed with acid to generate the corresponding β-amido carbonyl compounds, the overall sequence effecting an auxiliary-based enantioselective conjugate addition of an acetate enolate, leading to β-aminoacid derivatives.  相似文献

Treatment of 1,2-epoxy-3-ketosteroids with tosylhydrazine and sodium borohydride in methanol lead to mixtures consisting of (i) ring A seco-acetylenic alcohols, (ii) methoxyhydrins formed by opening of the epoxide ring following the carbonyl reduction, and (iii) an epoxy-3,3-di- methylketal. For the corresponding 4-methylene derivative, the ring A fragmentation product was found to be the conjugated en-yne 8a. Reduction of these epoxyketones with sodium borohydride alone affords mixtures of cis- and trans epoxyalcohols; the cis derivatives were unaffected by heating with sodium borohydride in methanol, whereas the trans were converted into the corresponding diaxial methoxyhydrins.  相似文献

Summary Alkaline sodium borohydride has been employed as a reducing reagent for the titrimetric determination of compounds containing the carbonyl function. The sample is dissolved in methanol and is then reduced with alkaline sodium borohydride. After completion of the reaction, the excess sodium borohydride is back titrated against standardized hydrochloric acid solution using methyl red as indicator. The stoichiometry between the carbonyl function and sodium borohydride is 41.

The α-thiocyanation of carbonyl compounds is one of the most important processes in synthetic organic chemistry. These compounds are important precursors for the production of various biologically important heterocyclic compounds and other industrially important products. Ammonium thiocyanate (NH4SCN) is a key reagent for the production of such a class of thiocyanate intermediates. In addition to the inherent efficiency issues, there are also environmental concerns that need to be addressed. Therefore, in recent years considerable advances have been made for the synthesis of α-thiocyanation of carbonyl compounds with high selectivity and yield. A novel copper-catalyzed cross-dehydrogenative coupling reaction of α-amino carbonyl compounds with nitromethane to synthesis of (Z)-nitroalkene derivatives has been established. The first section deals with the preparation, scope, limits, mechanism of action and synthetic applications of chiral aluminohydrides, classified according to the chemical nature of the stereogenic modifier. For the first time, we report the synthesis of 2-substituted N-alkoxy pyrrole 3-carboxylate and furan 3-carboxylate via Rh-catalyzed [3+2] cycloaddition between α-diazo oxime ether or α-diazo carbonyl compounds with vinyl equivalents in a one-pot process. We have demonstrated ethyl vinyl ether as well as vinyl acetate as vinyl equivalents and both were found to give excellent yields. We have also demonstrated the synthesis of N-alkoxy dihydropyrrole derivatives by carrying out the reaction at low temperature.  相似文献

The synthesis of pyrazole derivative using α,β-unsaturated carbonyl compounds has attracted increasing attention of the synthetic organic chemist community. Interestingly, the simplicity of the synthetic method, high reactivity, and ease of incorporating diversity into the desired prototype have contributed a lot toward the exploration of α,β-unsaturated carbonyl compounds by various research groups. Due to the tremendous pharmacological significance of pyrazole derivatives, their synthesis has been one of the leading research frontiers in recent years. As prime examples, sildenafil, zometapin, Celebrex, and rimonabant have been successfully commercialized in the market to treat various life-challenging diseases. Considering the great profile of α,β-unsaturated carbonyl compound in the synthesis of biologically active pyrazole derivatives, this review incorporates contemporary literature (2011–2022) on the synthesis of pyrazole and its derivatives using α,β-unsaturated carbonyl compound as a starting precursor.  相似文献

The preparation and reactivity of some α,α-difluorophosphinates are investigated. Alkylation of H-phosphinates with LiHMDS and ClCF2H gives the corresponding α,α-difluorophosphinates in good yield. Deprotonation of these reagents with alkyllithium or LDA is then studied. Subtle electronic effects translate into significant differences in the deprotonation/alkylation of the two ‘Ciba-Geigy reagents’ (EtO)2CRP(O)(OEt)H (R=H, Me). On the other hand, attempted methylation of difluoromethyl-octyl-phosphinic acid butyl ester resulted in the exclusive alkylation of the octyl chain. Finally, reaction with carbonyl compounds results in the formation of 1,1-difluoro-2-phosphinoyl compounds.  相似文献
