1H-and13C-NMR. spectra of methyl esters of the diastereomeric (±)-dihydropalustramic acids; structural correlation of (?)-dihydropalustramic acid with the alkaloid palustrin Unambiguous assignment of cis-trans configuration at the piperidine ring of the four diastereomeric methyl esters of the (±)-dihydropalustramic acids is based on the interpretation of 1H- and 13C-NMR. data. (?)-Dihydropalustramic acid, the important degradation product of the alkaloid palustrin is shown to be threo-cis-[6-(1-hydroxypropyl)-2-piperidyl]acetic acid. The same conclusion holds for palustrin and dihydropalustrin. Therefore, palustrin is threo-cis-17-(1-hydroxy-propyl)-1,5,10-triazabicyclo[11.4.0]heptadec-15-en-11-one.  相似文献

Photochemical Reaction of Optically Active 2-(1′-Methylallyl)anilines with Methanol It is shown that (?)-(S)-2-(1′-methylallyl)aniline ((?)-(S)- 4 ) on irradiation in methanol yields (?)-(2S, 3R)-2, 3-dimethylindoline ((?)-trans- 8 ), (?)-(1′R, 2′R)-2-(2′-methoxy-1′-methylpropyl)aniline ((?)-erythro- 9 ) as well as racemic (1′RS, 2′SR)-2-(2′-methoxy-1′-methylpropyl) aniline ((±)-threo- 9 ) in 27.1, 36.4 and 15.7% yield, respectively (see Scheme 3). By deamination and chemical correlation with (+)-(2R, 3R)-3-phenyl-2-butanol ((+)-erythro- 13 ; see Scheme 4) it was found that (?)-erythro- 9 has the same absolute configuration and optical purity as the starting material (?)-(S)- 4 . Comparable results are obtained when (?)-(S)-N-methyl-2-(1′-methylallyl)aniline ((?)-(S)- 7 ) is irradiated in methanol, i.e. the optically active indoline (+)-trans- 10 and the methanol addition product (?)-erythro- 11 along with its racemic threo-isomer are formed (cf. Scheme 3). These findings demonstrate that the methanol addition products arise from stereospecific, methanol-induced ring opening of intermediate, chiral trans, -(→(?)-erythro-compounds) and achiral cis-spiro [2.5]octa-4,6-dien-8-imines (→(±)-threo-compounds; see Schemes 1 and 2).  相似文献

Synthesis of Optically Active Natural Carotenoids and Structurally Related Compounds. VIII. Synthesis of (3S,3′S)-7,8,7′,8′-Tetradehydroastaxanthin and (3S,3′S)-7,8-Didehydroastaxanthin (Asterinic Acid) The synthesis of all-trans-(3S,3′S)-3,3′-dihydroxy-7,8, 7′,8′-tetradehydro-β, β-carotene-4,4′-dione ( 1 ), of all-trans-(3S,3′S)-3,3′-dihydroxy-7, 8-didehydro-β,β-carotene-4,4′-dione ( 2 ) (asterinic acid = mixture of 1 and 2 ), and of their 9,9′-di-cis- and 9-cis-isomers is reported starting from (4′S)(2E)-5-(4′-hydroxy-2′, 6′,6′-trimethyl-3′-oxo-l′-cyclohexenyl)-3-methyl-2-penten-4-ynal ( 8 ). The absolute configuration (3S,3′S) for both components 1 and 2 of asterinic acid ex Asterias rubens is confirmed on the basis of spectroscopic and direct comparison.  相似文献

Synthesis of Enantiomerically Pure Mimulaxanthin and of Its (9Z,9′Z)- and (15Z)Isomers We present the details of a synthesis of optically active, enantiomerically pure stereoisomers of mimulaxanthin (=(3s,5R,6R,3′S,5′R,6′R)-6,7,6′,7′-tetradehydro-5,6,5′,6′-tetrahydro-β,β-carotin-3,5,3′,5′-tetrol) either as free alcohols 1a and 24a or as their crystalline (t-Bu)Me2Si ethers 1b and 24b . Grasshopper ketone 2a , a presumed synthon, unexpectedly showed a very sluggish reaction with Wittig-Horner reagents. Upon heating with the ylide of ester phosphonates, an addition across the allenic bond occurred. On the contrary, a slow but normal 1,2-addition took place with the ylide from (cyanomethyl)phosphonate but, unexpectedly, with concomitant inversion at the chiral axis. So a mixture of(6R,6S,9E,9Z)-isomers 6 – 9 was produced {(Scheme 1). However, a fast and very clean 1,2-addition occurred with the ethynyl ketone 12 to yield the esters 13 and 14 (Scheme 2). DIBAH reduction of the separated stereoisomers gave the allenic alcohols 15 and 16 in high yield. Mild oxidation to the aldehydes 17 and 18 followed by their condensation with the acetylenic C10-bis-ylide 19 led to the stereoisomeric 15,15′-didehydromimulaxanthins 20 and 22 , respectively (Schemes 3 and 4). Mimulaxanthins 1 and 24 were prepared by partial hydrogenation of 20 and 22 followed by a thermal (Z/E)-isomerization. As expected, the mimulaxanthins exhibit very weak CD curves, obviously caused by the allenic bond that insulates the chiral centers in the end group from the chromophor. On the contrary, some of the C15-allenic synthons showed not only fairly strong CD effects but also a split CD curve which, in our interpretation, results from an exciton coupling between the allene and the C(9)?C(10) bond. We postulate a rotation around the C(8)? C(9) bond, presumably caused by an intramolecular H-bond in 16 or by a dipol interaction between the polarized double bonds in 6 , 7 , 8 , and 17 .  相似文献

Carotenoids with 7-Oxabicyclo[2,2.1]heptyl End Groups. Attempted Synthesis of Cycloviolaxanthin ( = (3S,5R,6S,3′S,5′R,6′R)-3,6:3′,6′- Diepoxy-5,6,5′,6′-tetrahydro-β,β-carotin-5,5′-diol) Starting from our recently described synthon (+)- 24 , the enantiomerically pure 3,6:4,5:3′,6′:4′,5′-tetraepoxy-4,5,4′,5′-tetrahydro-ε,ε-carotene ( 34 ) and its 15,15′-didehydro analogue 32 were synthesized in eleven and nine steps, respectively (Scheme 4). Chiroptical data show, in contrast to the parent ε,ε-carotene, a very weak interaction between the chiral centers at C(5), C(5′), C(6), C(6′), and the polyene system. Diisobutylaluminium hydride reduction of 32 lead rather than to the expected 15,15′-didehydro analogue 35 of Cycloviolaxanthin ( 8 ), to the polyenyne 36 (Scheme 5). We explain this reaction by an oxirane rearrangement leading to a cyclopropyl ether followed by a fragmentation to an aldehyd on the one side and an enol ether on the other (Scheme 6). This complex rearrangement includes a shift of the whole polyenyne chain from C(6), C(6′) to C(5), C(5′) of the original molecule.  相似文献

The Stereoselectivity of the α-Alkylation of (+)-(1R, 2S)-cis-Ethyl-2-hydroxy-cyclohexanecarboxylate In continuation of our work on the stereoselectivity of the α-alkylation of β-hydroxyesters [1] [2], we studied this reaction with the title compound (+)- 2 . The latter was prepared through reduction of 1 with baker's yeast. Alkylation of the dianion of (+)- 2 furnished (?)- 4 in 72% chemical yield (Scheme 1) and with a stereoselectivity of 95%. Analogously, (?)- 7 was prepared with similar yields. Oxidation of (?)- 4 and (?)- 7 respectively furnished the ketones (?)- 6 (Scheme 3) and (?)- 8 (Scheme 4) respectively, each with about 76% enantiomeric excess (NMR.). It is noteworthy that yeast reduction of rac- 6 (Scheme 3) is completely enantioselective with respect to substrate and product and gives optically pure (?)- 4 in 10% yield, which was converted into optically pure (?)- 6 (Scheme 3). The alkylation of the dianionic intermediate shows a higher stereoselectivity (95%) from the pseudoequatorial side than that of 1-acetyl- or 1-cyano-4-t-butyl-cyclohexane (71% and 85%) [9] or that of ethyl 2-methyl-cyclohexanecarboxylate (82%). The stereochemical outcome of the above alkylation is comparable with that found in open chain examples [1] [2]. Finally (+)-(1R, 2S)- 2 was also alkylated with Wichterle's reagent to give (?)-(1S, 2S)- 9 in 64% yield. The latter was transformed into (?)-(S)- 10 and further into (?)-(S)- 11 (Scheme 5). (?)-(S)- 10 and (?)-(S)- 11 showed an e.e. of 76–78% (see also [11]). Comparison of these results with those in [11] confirmed our former stereochemical assignment concerning the alkylation step.  相似文献

Syntheses of the four stereoisomeric dihydropalustramic acids ([6-(1-hydroxypropyl)-2-piperidyl]acetic acids) (?)-Dihydropalustramic acid, a key product in the structure elucidation of the alkaloid palustrin, has been assigned the threo-cis structure 20 by comparison with the four stereoisomeric (±)-dihydropalustramic acids (threo-cis, threo-trans, erythro-cis, erythro-trans). The latter were synthesized by a new route to α, α′-di-substituted piperidines of this type. Ring closure to the piperidine ring with simultaneous stereospecific formation of the hydroxylated side chain has been achieved by reaction of the stereoisomeric methylesters of 7,8-epoxy-2-decenoic acids with benzylamine. Assignment of the configuration at the piperidine ring is based on careful comparison of the H-NMR. spectra of the N-benzylpiperidines and with the help of lanthanide shift reagents.  相似文献

Carotenoids mit 7-Oxabicyclo[2.2.1]heptyl-End Groups. Synthesis of (2S,5R,6S,2′S,5′R,6′S)-2,5:2′5′-Diepoxy-5,6,5′,6′-tetrahydro-β,β-carotene Mukayama's ester 6 (methyl (1S,2R,5S)-2,5-epoxy-2,6,6-trimethylcyclohexane-1-carboxylate) was transformed in a few conventional steps into the title compound 14 . Its CD curve was found to be significantly different from that of the analogous 3,6-epoxide, a fact we tentatively lake as an indication of a (weak) electronic interaction between the ring O-atom and the π-orbitals of the polyene chain.  相似文献

(?)-(R)-4,4,4,4′,4′,4′-Hexafluorovaline hydrochloride ((R)- 5 ) of 98% ee is prepared from β,β-bis(trifluoromethyl)acrylic acid (= benzyl 4,4,4-trifluoro-3-(trifluoromethyl)but-2-enoate; 1 ) in 4 steps with an overall yield of 9.6%. Key step is the separation of the TsOH salts of the diastereoisomers obtained by anti-Michael addition of (+)-(R)-1-phenylethylamine ( 2 ) to 1 (→ (R,R)- 3 ). In contrast to the published (S)-chirality, the X-ray structure analysis of (R,S)- 6 reveals, that (R)-chirality has to be assigned to the levorotatory (?)-4,4,4,4′,4′,4′-hexafluorovaline hydrochloride.  相似文献

We describe the stereoselective synthesis of (2′S)‐2′‐deoxy‐2′‐C‐methyladenosine ( 12 ) and (2′S)‐2′‐deoxy‐2′‐C‐methylinosine ( 14 ) as well as their corresponding cyanoethyl phosphoramidites 16 and 19 from 6‐O‐(2,6‐dichlorophenyl)inosine as starting material. The methyl group at the 2′‐position was introduced via a Wittig reaction (→ 3 , Scheme 1) followed by a stereoselective oxidation with OsO4 (→ 4 , Scheme 2). The primary‐alcohol moiety of 4 was tosylated (→ 5 ) and regioselectively reduced with NaBH4 (→ 6 ). Subsequent reduction of the 2′‐alcohol moiety with Bu3SnH yielded stereoselectively the corresponding (2′S)‐2′‐deoxy‐2′‐C‐methylnucleoside (→ 8a ).  相似文献

Synthesis of (+)-(5S, 6S)-Azafrin Methyl Ester; Absolute Configuration of Aeginetic Acid and of Further Vicinal Apocarotenediols We describe the synthesis of a series of optically active vicinal apo-β-carotenediols. Thus, starting from (+)-(5S, 6S)-5,6-dihydroxy-5,6-dihydro-β-ionone ( 2 ) we have prepared the (Z/E)-isomeric (+)-C15-esters 7 and 8 , the (+)-retinoic derivatives 14 , 15 , 18 , 19 and (+)-methyl azafrinate ( 22 ), the enantiomer of the naturally occur-ring compound (s. Scheme 1). Our synthesis also establishes the absolute configura-tion of aeginetic acid ( 24 ), aeginetoside ( 25 ) and aeginetin ( 26 ), compounds isolated from the root parasite Aeginetia indica by Indian and Japanese workers (s. Scheme 2). The presented synthesis of optically active methyl azafrinate confirms our previous assignment [14] of the absolute configuration of azafrin ( 1a ), which was based on degradative evidence.  相似文献

Their chiroptical properties are compared with those of saturated twistane ((+)- and (?)- 5 ) as well as with those of the unsaturated and saturated 2, 7-dioxatwistane analogs (+)- and (?)- 9 , and (+)- and (?)- 10 , respectively, which also are compounds of known absolute configurations.  相似文献

Michael addition of carbothioates. Application to the synthesis of (±)-jasmine ketolactone It is shown that the lithium enolate of S-t-butyl thioacetate adds to 2-cyclopentenone in the β-position and that fluoride ions catalyze the 1, 4-addition of the trimethylsilyl enol ether of S-t-butyl thioacetate ( 5 ) to 2-cyclopentenone ( 4 ) to give 6 . These novel versions of the Michael addition have been applied to a synthesis of jasmonoid compounds. Cleavage of the trimethylsilyl enol ether in 6 with tetrabutylammonium fluoride produced the corresponding ketone enolate which could be trapped in situ by alkylation with 1-bromo-5-(2′-tetrahydropyranoxy)-2-pentyne ( 7 ) to form 8 . Removal of the alcohol protecting group in 8 , followed by partial hydrogenation of the triple bond over Lindlar palladium and mercury ion promoted hydrolysis of the carbothioate moiety in 9 , led to 5′-hydroxy jasmonic acid ( 10 , Scheme 3). 10 was converted into the S-(2-pyridyl) carbothioate and cyclized in dilute benzene solution under the influence of silver ion to give (±)-jasmine ketolactone ( 1 , Scheme 4), a component of the essential oil of Jasminum grandiflorum, in 72% yield. Similarly, methyljasmonate ( 2 , Scheme 2) was obtained from 6 by the reaction with 1-bromo-2-pentyne and tetrabutylammonium fluoride followed by methanolysis and partial hydrogenation of the triple bond.  相似文献

The synthesis of sarcinaxanthin ((2R,6R,2′R,6′R)- 1 ), a symmetrical C50-carotenoid with two γ-end groups, isolated from Sarcina lutea and from Cellulomonas biazotea as major pigment, was based on the strategy C20 + C10 + C20 = C50 using camphoric acid as starting material for the C20-end group 3. The key step of the synthesis is a ring enlargement of the cyclopentane derivative 10 with 2,4,4,6-tetrabromocyclohexa-2,5-dien-1-one (TBCO) to give the cyclohexane derivative 11 (Scheme 1). The spectroscopic data of the synthetic compound are in full agreement with the data of the isolated product and give the final proof for the (2R,6R,2′R,6′R) chirality of natural sarcinaxanthin.  相似文献

Following a known procedure, a mixture of (?)-(2S,3R)- and (+)-(2R,3R)-2,3-epoxy-citronellols ( 5 ) was prepared from (?)-(R)-linalool ( 3 ) via epoxy alcohol 4 and then reduced to (?)-(R)-3-hydroxy-citronellol ( 6 ). Sensitized photooxygenation of (?)-(R)-diol 6 led in part to (?)-(R)-triol 8 which was cyclodehydrated by dilute acid to a mixture of diastereoisomeric tetrahydropyran-4-ols 9 and 10 . Dehydration of hydroxy ethers 9 and 10 afforded (?)-(S)-nerol oxide ( 11 ) and (+)-(R)-nerol oxide ( 12 ), respectively, with an optical purity of 91%. Nerol oxide isolated from Bulgarian rose oil (0.038%) proved to be racemic. These results shed some light on the formation of nerol oxide in plants.  相似文献

Enantiospecific Synthesis of (+)-(2R)- and (?)-(2S)-6-Ethyl-3,4-dihydro-2-methyl-4-oxo-2H-pyran-5-carboxylic Acid The two enantiomers (?)-(2S)- and (+)-(2R)-6-ethyl-3,4-dihydro-2-methyl-4-oxo-2H-pyran-5-carboxylic acid ((S)- and (R)- 7 ) have been synthesized from (+)-(3S) and (?)-(3R)-3-hydroxybutanoates, respectively (Scheme 1). By reduction and decarboxylation, the tetrahydro-2H-pyranols (2R, 4R, 6S)- and (2S, 4S, 6R)- 13 , respectively, were obtained with an enantiomeric excess of ≥ 93%.  相似文献

We describe the synthesis of (5′S)‐5′‐C‐butylthymidine ( 5a ), of the (5′S)‐5′‐C‐butyl‐ and the (5′S)‐5′‐C‐isopentyl derivatives 16a and 16b of 2′‐deoxy‐5‐methylcytidine, as well as of the corresponding cyanoethyl phosphoramidites 9a , b and 14a , b , respectively. Starting from thymidin‐5′‐al 1 , the alkyl chain at C(5′) is introduced via Wittig chemistry to selectively yield the (Z)‐olefin derivatives 3a and 3b (Scheme 2). The secondary OH function at C(5′) is then introduced by epoxidation followed by regioselective reduction of the epoxy derivatives 4a and 4b with diisobutylaluminium hydride. In the latter step, a kinetic resolution of the diastereoisomer mixture 4a and 4b occurs, yielding the alkylated nucleoside 2a and 2b , respectively, with (5′S)‐configuration in high diastereoisomer purity (de=94%). The corresponding 2′‐deoxy‐5‐methylcytidine derivatives are obtained from the protected 5′‐alkylated thymidine derivatives 7a and 7b via known base interconversion processes in excellent yields (Scheme 3). Application of the same strategy to the purine nucleoside 2′‐deoxyadenine to obtain 5′‐C‐butyl‐2′‐deoxyadenosine 25 proved to be difficult due to the sensitivity of the purine base to hydride‐based reducing agents (Scheme 4).  相似文献
