Au‐Fe3O4 nanoparticles were widely used as nanoplatforms for biologic applications through readily further functionalization. Dopamine (DA)‐coated superparamagnetic iron oxide (SPIO) nanoparticles (DA@Fe3O4) have been successfully synthesized using a one‐step process by modified coprecipitation method. Then 2–3 nm gold nanoparticles were easily conjugated to DA@Fe3O4 nanoparticles by the electrostatic force between gold nanoparticles and amino groups of dopamine to afford water‐soluble Au‐Fe3O4 hybrid nanoparticles. A detailed investigation by dynamic light scatting (DLS), transmission electron microscopy (TEM), fourier transform infrared (FT‐IR) and X‐ray diffraction (XRD) were performed in order to characterize the physicochemical properties of the hybrid nanoparticles. The hybrid nanoparticles were easily functionalized with a targeted small peptide A54 (AGKGTPSLETTP) and fluorescence probe fluorescein isothiocyanate (FITC) for liver cancer cell BEL‐7402 imaging. This simple approach to prepare hybrid nanoparticles provides a facile nanoplatform for muti‐functional derivations and may be extended to the immobilization of other metals or bimolecular on SPIO surface.  相似文献

Functionalised MCM‐41 mesoporous silica nanoparticles were used as carriers of GdIII complexes for the development of nanosized magnetic resonance imaging contrast agents. Three GdIII complexes based on the 1,4,7,10‐tetraazacyclododecane scaffold (DOTA; monoamide‐, DOTA‐ and DO3A‐like complexes) with distinct structural and magnetic properties were anchored on the silica nanoparticles functionalised with NH2 groups. The interaction between GdIII chelates and surface functional groups markedly influenced the relaxometric properties of the hybrid materials, and were deeply modified passing from ionic ? NH3+ to neutral amides. A complete study of the structural, textural and surface properties together with a full 1H relaxometric characterisation of these hybrid materials before and after surface modification was carried out. Particularly for the anionic complex 2 attached to MCM‐41, an impressive increase in relaxivity (r1p) was observed (from 20.3 to 37.8 mM ?1 s?1, 86.2 % enhancement at 20 MHz and 310 K), mainly due to a threefold faster water exchange rate after acetylation of the surface ? NH3+ ions. This high r1p value, coupled with the large molar amount of grafted 2 onto the silica nanoparticles gives rise to a value of relaxivity per particle of 29 500 mM ?1 s?1, which possibly allows it to be used in molecular imaging procedures. Smaller changes were observed for the hybrid materials based on neutral 1 and 3 complexes. In fact, whereas 1 shows a weak interaction with the surface and acetylation induced only some decrease of the local rotation, complex 3 appears to be involved in a strong interaction with surface silanols. This results in the displacement of a coordinated water molecule and in a decrease of the accessibility of the solvent to the metal centre, which is unaffected by the modification of ammonium ions to neutral amides.  相似文献

Lanthanide‐doped upconversion nanoparticles (UCNPs) have attracted considerable attention for their application in biomedicine. Here, silica‐coated NaGdF4:Yb,Er/NaGdF4 nanoparticles with a tetrasubstituted carboxy aluminum phthalocyanine (AlC4Pc) photosensitizer covalently incorporated inside the silica shells were prepared and applied in the photodynamic therapy (PDT) and magnetic resonance imaging (MRI) of cancer cells. These UCNP@SiO2(AlC4Pc) nanoparticles were uniform in size, stable against photosensitizer leaching, and highly efficient in photogenerating cytotoxic singlet oxygen under near‐infrared (NIR) light. In vitro studies indicated that these nanoparticles could effectively kill cancer cells upon NIR irradiation. Moreover, the nanoparticles also demonstrated good MR contrast, both in aqueous solution and inside cells. This is the first time that NaGdF4:Yb,Er/NaGdF4 upconversion‐nanocrystal‐based multifunctional nanomaterials have been synthesized and applied in PDT. Our results show that these multifunctional nanoparticles are very promising for applications in versatile imaging diagnosis and as a therapy tool in biomedical engineering.  相似文献

Chemodynamic therapy (CDT) utilizes iron‐initiated Fenton chemistry to destroy tumor cells by converting endogenous H2O2 into the highly toxic hydroxyl radical (.OH). There is a paucity of Fenton‐like metal‐based CDT agents. Intracellular glutathione (GSH) with .OH scavenging ability greatly reduces CDT efficacy. A self‐reinforcing CDT nanoagent based on MnO2 is reported that has both Fenton‐like Mn2+ delivery and GSH depletion properties. In the presence of HCO3?, which is abundant in the physiological medium, Mn2+ exerts Fenton‐like activity to generate .OH from H2O2. Upon uptake of MnO2‐coated mesoporous silica nanoparticles (MS@MnO2 NPs) by cancer cells, the MnO2 shell undergoes a redox reaction with GSH to form glutathione disulfide and Mn2+, resulting in GSH depletion‐enhanced CDT. This, together with the GSH‐activated MRI contrast effect and dissociation of MnO2, allows MS@MnO2 NPs to achieve MRI‐monitored chemo–chemodynamic combination therapy.  相似文献

Nonporous monodispersed silica spheres of 1.3 μm were coated with gold nanoparticles (AuNPs) and subsequently coated with n‐octadecanethiol. By transmission electron microscopy analysis, the average diameter of the AuNPs on the silica spheres was determined to be 12 nm. The chromatographic and electrochromatographic properties of self‐assembled n‐octadecanethiol AuNP‐coated silica microspheres (C18‐AuNPs‐SiO2) were investigated using a group of nonpolar PAHs. The stationary phase appears to display a characteristic reversed‐phase behavior. Higher separation efficiency and shorter separation times were obtained using pressurized CEC (pCEC) compared with capillary LC (CLC). A maximum column efficiency of about 2.5×105 plates per meter and less than 18 min separation time for benzene were obtained in pCEC while only 66 507 plates per meter and an analysis time of nearly 100 min were observed in CLC mode. A chemical stability test of the C18‐AuNPs‐SiO2 stationary phase under extremely high and low pH conditions demonstrated that it is stable at pH 12 and 1 for at least 60 h. The results confirm that C18‐AuNPs‐SiO2 possesses a high rigidity to withstand high packing pressures and can be used as a good stationary phase for CLC and pCEC.  相似文献

We present a facile strategy to prepare the molecularly imprinted polymers layer on the surface of Fe3O4 nanoparticles with core‐shell structure via sol–gel condensation for recognition and enrichment of triclosan. The Fe3O4 nanoparticles were first synthesized by a solvothermal method. Then, template triclosan was self‐assembled with the functional monomer 3‐aminopropyltriethoxysilane on the silica‐coated Fe3O4 nanoparticles in the presence of ethanol and water. Finally, the molecularly imprinted polymers were formed on the surface of silica‐coated Fe3O4 nanoparticles to obtain the product. The morphology, magnetic susceptibility, adsorption, and recognition property of magnetic molecularly imprinted polymers were characterized using transmission electron microscopy, Fourier transform infrared spectroscopy, X‐ray diffractometry, vibrating sample magnetometry, and re‐binding experiments. The magnetic molecularly imprinted polymers showed binding sites with good accessibility, fast adsorption rate, and high adsorption capacity (218.34 μg/g) to triclosan. The selectivity of magnetic molecularly imprinted polymers was evaluated by the rebinding capability of triclosan and two other structural analogues (phenol and p‐chlorophenol) in a mixed solution and good selectivity with an imprinting factor of 2.46 was obtained. The application of triclosan removal in environmental samples was demonstrated.  相似文献

In this communication, we report the synthesis of small‐sized (<10 nm), water‐soluble, magnetic nanoparticles (MNPs) coated with polyhedral oligomeric silsesquioxanes (POSS), which contain either polyethylene glycol (PEG) or octa(tetramethylammonium) (OctaTMA) as functional groups. The POSS‐coated MNPs exhibit superparamagnetic behavior with saturation magnetic moments (51–53 emu g?1) comparable to silica‐coated MNPs. They also provide good colloidal stability at different pH and salt concentrations, and low cytotoxicity to MCF‐7 human breast epithelial cells. The relaxivity data and magnetic resonance (MR) phantom images demonstrate the potential application of these MNPs in bioimaging.  相似文献

An on‐column preconcentration technique, pH‐mediated acid stacking, was used in this study to improve the sensitivity of MEKC‐UV analysis of IgG in human serum. Various parameters affecting pH‐mediated acid stacking were optimized systematically. To eliminate the matrix interferences of human serum and to combine the sample pretreatment procedure with the detection methodology, silica‐coated Fe3O4 magnetic nanoparticles modified with N‐(2‐aminoethyl)‐3‐aminopropyl‐trimethoxysilane were prepared and employed as solid phase extraction adsorbent to remove the abundant HSA from human serum. HSA was quantitatively removed by silica‐coated Fe3O4 magnetic nanoparticles modified with N‐(2‐aminoethyl)‐3‐aminopropyl‐trimethoxysilanes without retaining IgG at pH 9.3. Under the optimum conditions, the sensitivity of IgG was improved 40.3‐fold using a 100 s electrokinetic injection as compared with a 6 s hydrodynamic injection. The detection limit of IgG was found to be 0.1 mg/L, and the proposed method was successfully applied for the determination of IgG in human serum with satisfactory results.  相似文献

Multimodal imaging and simultaneous therapy is highly desirable because it can provide complementary information from each imaging modality for accurate diagnosis and, at the same time, afford an imaging‐guided focused tumor therapy. In this study, indocyanine green (ICG), a near‐infrared (NIR) imaging agent and perfect NIR light absorber for laser‐mediated photothermal therapy, was successfully incorporated into superparamagnetic Fe3O4@mSiO2 core–shell nanoparticles to combine the merit of NIR/magnetic resonance (MR) bimodal imaging properties with NIR photothermal therapy. The resultant nanoparticles were homogenously coated with poly(allylamine hydrochloride) (PAH) to make the surface of the composite nanoparticles positively charged, which would enhance cellular uptake driven by electrostatic interactions between the positive surface of the nanoparticles and the negative surface of the cancer cell. A high biocompatibility of the achieved nanoparticles was demonstrated by using a cell cytotoxicity assay. Moreover, confocal laser scanning microscopy (CLSM) observations indicated excellent NIR fluorescent imaging properties of the ICG‐loaded nanoparticles. The relatively high r2 value (171.6 mM ?1 s?1) of the nanoparticles implies its excellent capability as a contrast agent for MRI. More importantly, the ICG‐loaded nanoparticles showed perfect NIR photothermal therapy properties, thus indicating their potential for simultaneous cancer diagnosis as highly effective NIR/MR bimodal imaging probes and for NIR photothermal therapy of cancerous cells.  相似文献

Graphene oxide (GO) has been considered as a promising stationary phase for chromatographic separation. However, the very strong adsorption of the analytes on the GO surface lead to the severe peak tailing, which in turn resulting in decreased separation performance. In this work, GO and silica nanoparticles hybrid nanostructures (GO/SiO2 NPs@column) were coated onto the capillary inner wall by passing the mixture of GO and silica sol through the capillary column. The successful of coating of GO/SiO2 NPs onto the capillary wall was confirmed by SEM and electroosmotic flow mobilities test. By partially covering the GO surface with silica nanoparticles, the peak tailing was decreased greatly while the unique high shape selectivity arises from the surface of remained GO was kept. Consequently, compared with the column modified with GO (GO@column), the column modified with GO and silica nanoparticles through layer‐by‐layer method (GO‐SiO2 NPs@column), or the column modified with silica nanoparticles (SiO2 NPs@column), GO/SiO2 NPs@column possessed highest resolutions. The GO/SiO2 NPs@column was applied to separate egg white and both acidic and basic proteins as well as three glycoisoforms of ovalbumin were separated in a single run within 36 min. The intra‐day, inter‐day, and column‐to‐column reproducibilities were evaluated by calculating the RSDs of the retention of naphthalene and biphenyl in open‐tubular capillary electrochromatography. The RSD values were found to be less than 7.1%.  相似文献

The selected‐control preparation of uniform core–shell and yolk–shell architectures, which combine the multiple functions of a superparamagnetic iron oxide (SPIO) core and europium‐doped yttrium oxide (Y2O3:Eu) shell in a single material with tunable fluorescence and magnetic properties, has been successfully achieved by controlling the heat‐treatment conditions. Furthermore, the shell thickness and interior cavity of SPIO@Y2O3:Eu core–shell and yolk–shell nanostructures can be precisely tuned. Importantly, as‐prepared SPIO@Y2O3:Eu yolk–shell nanocapsules (NCs) modified with amino groups as cancer‐cell fluorescence imaging agents are also demonstrated. To the best of our knowledge, this is the first report on the selected‐control fabrication of uniform SPIO@Y2O3:Eu core–shell nanoparticles and yolk–shell NCs. The combined magnetic manipulation and optical monitoring of magnetic–fluorescent SPIO@Y2O3:Eu yolk–shell NCs will open up many exciting opportunities in dual imaging for targeted delivery and thermal therapy.  相似文献

Iron oxide (Fe3O4) magnetic nanoparticles as movable cores were used to synthesize yolk–shell nanoparticles with pH‐responsive shell composed of ethylene glycol dimethacrylate (EGDMA)‐crosslinked poly(acrylic acid) (PAA) via two different routes. In the first more common route, Fe3O4 nanoparticles were coated with silica layer via the Stöber process to yield Fe3O4@SiO2 core–shell nanoparticles, subsequently used as seeds in the distillation precipitation copolymerization of AA and EGDMA to yield Fe3O4@SiO2@P(AA‐EGDMA). The silica layer was selectively removed through alkali etching to yield Fe3O4@air@P(AA‐EGDMA). In the second route, Fe3O4 nanoparticles without any stabilization were used as seeds in the distillation precipitation copolymerization of AA and EGDMA to yield Fe3O4@P(AA‐EGDMA) core–shell nanoparticles. The nanoparticles were subsequently dispersed in acidic medium of pH = 2. Yolk–shell Fe3O4@air@P(AA‐EGDMA) nanoparticles were formed through deswelling of crosslinked PAA because of protonation of carboxyl groups at low pH values. Various techniques were utilized to investigate the characteristics of the synthesized core–shell nanoparticles. Formation of yolk–shell nanostructure was observed for both synthesis routes, namely etching of silica layer and deswelling approaches, from vibrating sample magnetometry and transmission electron microscopy results. Both types of nanoparticles showed pH‐responsive behaviour, i.e. decrease in absorption with increase in pH, as examined using UV–visible spectroscopy.  相似文献

Porous metal‐organic frameworks (MOFs) loading metal nanoparticles to form a composite photocatalyst demonstrated unique advantages. Modification of the electron donating group on the aromatic linkers of MOFs could increase the absorption range of light, thereby increasing the photocatalytic activity. In this study, we prepared a composite photocatalyst using a stable NH2‐functionalized MOF (UiO‐66‐NH2) to load semiconductor Ag/AgBr nanoparticles, and the resultant composites have intense optical absorption throughout visible light range. The greatly enhanced optical absorption and the unique hetero‐junction between Ag/AgBr and UiO‐66‐NH2 render efficient separation and utilization of photogenerated electron‐hole pairs. Therefore, Ag/AgBr@UiO‐66‐NH2 showed much more excellent photocatalytic activity, compared with unmodified UiO‐66 loading Ag/AgBr (Ag/AgBr@UiO‐66) and reported AgX@MOF catalysts. Moreover, the composite photocatalysts showed excellent stability during cycling experiment.  相似文献

In this study, porous sandwich structure Fe3O4 nanoparticles coated by polyhedral oligomeric silsesquioxanes and β‐cyclodextrin were prepared by surface polymerization and were used as the magnetic solid phase extraction adsorbent for the extraction and determination of carbaryl and carbofuran. The Fe3O4 nanoparticles coated with polyhedral oligomeric silsesquioxanes and β‐cyclodextrin were characterized by Fourier transform infrared spectroscopy, X‐ray diffraction, thermogravimetric analysis, vibrating sample magnetometry, and scanning electron microscopy. After optimizing the extraction conditions, a method that combined magnetic solid phase extraction with high‐performance liquid chromatography was developed for the determination of carbaryl and carbofuran in apple. The method exhibited a good linearity in the range of 2–400 μg/kg for carbaryl and carbofuran (R= 0.9995), respectively. The limits of detection were 0.5 μg/kg of carbaryl and 0.7 μg/kg for carbofuran in apple, respectively. Extraction recoveries ranged from 94.2 to 103.1% with the preconcentration factor of 300 and the relative standard deviations were less than 5.9%. These results indicated that the method combined magnetic solid phase extraction with high‐performance liquid chromatography and was promising for the determination of carbaryl and carbofuran at trace amounts.  相似文献

Reported herein is a study of the unusual 3′–3′ 1,4‐GG interstrand cross‐link (IXL) formation in duplex DNA by a series of polynuclear platinum anticancer complexes. To examine the effect of possible preassociation through charge and hydrogen‐bonding effects the closely related compounds [{trans‐PtCl(NH3)2}2(μ‐trans‐Pt(NH3)2{NH2(CH2)6NH2}2)]4+ (BBR3464, 1 ), [{trans‐PtCl(NH3)2}2(μ‐NH2(CH2)6NH2)]2+ (BBR3005, 2 ), [{trans‐PtCl(NH3)2}2(μ‐H2N(CH2)3NH2(CH2)4)]3+ (BBR3571, 3 ) and [{trans‐PtCl(NH3)2}2{μ‐H2N(CH2)3‐N(COCF3)(CH2)4}]2+ (BBR3571‐COCF3, 4 ) were studied. Two different molecular biology approaches were used to investigate the effect of DNA template upon IXL formation in synthetic 20‐base‐pair duplexes. In the “hybridisation directed” method the monofunctionally adducted top strands were hybridised with their complementary 5′‐end labelled strands; after 24 h the efficiency of interstrand cross‐linking in the 5′–5′ direction was slightly higher than in the 3′–3′ direction. The second method involved “postsynthetic modification” of the intact duplex; significantly less cross‐linking was observed, but again a slight preference for the 5′–5′ duplex was present. 2D [1H, 15N] HSQC NMR spectroscopy studies of the reaction of [15N]‐ 1 with the sequence 5′‐d{TATACATGTATA}2 allowed direct comparison of the stepwise formation of the 3′–3′ IXL with the previously studied 5′–5′ IXL on the analogous sequence 5′‐d(ATATGTACATAT)2. Whereas the preassociation and aquation steps were similar, differences were evident at the monofunctional binding step. The reaction did not yield a single distinct 3′–3′ 1,4‐GG IXL, but numerous cross‐linked adducts formed. Similar results were found for the reaction with the dinuclear [15N]‐ 2 . Molecular dynamics simulations for the 3′–3′ IXLs formed by both 1 and 2 showed a highly distorted structure with evident fraying of the end base pairs and considerable widening of the minor groove.  相似文献

We report the preparation of supported palladium(II) acetylacetonate, Pd(acac)2, coordinated by pendant acac groups, by reacting palladium acetate with acac‐functionalized doubly silica‐coated magnetic nanoparticles. The solid support consists of an amorphous silica‐coated (as magnetite protecting layer) magnetite core and a mesoporous silica shell. The magnetically separable palladium nanocatalyst is active for Suzuki cross‐coupling reaction of acyl halides with boronic acids. The catalyst is simply isolated from the reaction mixture that allows fast and efficient isolation of product and catalyst compared to traditional methods that generally make use of time‐ and solvent‐consuming procedures. Fe3O4 superparamagnetic nanoparticles with various functionalities were synthesized using a chemical co-precipitation method and used to demonstrate their analytical applications for protein separation of protein and metal ion extraction. The chemically inert silica layer coated with tetraethoxysilane (TEOS) protected the Fe3O4 core from a chemical attack and allowed the nanoparticles to be well dispersed in an aqueous solution. Particularly, the beads were resistant to an acidic solution with a pH ≥ 3. The amino (− NH2) groups were covalently bonded to the silica coated Fe3O4, and then the carboxyl (− COOH) groups were functionalized to the nanoparticle surface through the reaction of − NH2 and glutaric anhydride. The synthesized magnetic nanoparticles (MNP) were characterized using FT-IR, FE-TEM, XRD, and SQUID. The presence of functional groups on the nano beads was confirmed using molecular fluorescence spectrometry. For the presence of the amino (− NH2) groups, FITC was tagged and monitored using an excitation laser with a wavelength of 473 nm and a fluorescence emission of 518 nm. Biotin was immobilized on the MNP and the fluorescent of FITC tagged on avidin was monitored to identify the carboxyl (− COOH) group.The proteins of Cytochrome C (12,000 Da), Rnase B (15,000 Da), and Myoglobin (17,000 Da) were separated using the MNP functionalized with the carboxyl (− COOH) group and identified using MALDI-TOF-MS. Amino benzyl EDTA (ethylenediaminetetraacetic acid) was immobilized on the MNP for metal–EDTA complexation to use the synthesized magnetic particles to extract metal ions for environmental and clinical application. Cu, Cd, Co, and Pb ions were extracted from ∼ 10 ng/mL solutions in the batch-type procedure and the extraction efficiency was > 90% at a pH of 4.  相似文献

A strategy has been developed for the synthesis, characterization and catalysis of magnetic Fe3O4/P(GMA‐EGDMA)‐NH2/HPG‐COOH‐Pd core‐shell structure supported catalyst. The P(GMA‐EGDMA) polymer layer was coated on the surface of hollow magnetic Fe3O4 microspheres through the effect of KH570. The core‐shell magnetic Fe3O4/P(GMA‐EGDMA) modified by ‐NH2 could be grafted with HPG. Then, the hyperbranched glycidyl (HPG) with terminal ‐OH were modified by ‐COOH and adsorbed Pd nanoparticles. The hyperbranched polymer layer not only protected the Fe3O4 magnetic core from acid–base substrate corrosion, but also provided a number of functional groups as binding sites for Pd nanoparticles. The prepared catalyst was characterized by UV–vis, TEM, SEM, FTIR, TGA, ICP‐OES, BET, XRD, DLS and VSM. The catalytic tests showed that the magnetic Fe3O4/P(GMA‐EGDMA)‐NH2/HPG‐COOH‐Pd catalyst had excellent catalytic performance and retained 86% catalytic efficiency after 8 consecutive cycles.  相似文献

A novel magnetic rhodium catalyst was prepared through immobilizing Wilkinson's catalyst on the surface of silica‐coated iron oxide nanoparticles. After (thio)diphenylphosphine (─S&─PPh2) was modified on the surface of the silica‐coated iron oxide nanoparticles, tris(triphenylphosphine)rhodium(I) chloride was employed to synthesize the Rh(Cl)(PPh3)2(Ph2P&─S&─) complex, affording a rhodium loading of 0.16 mmol g−1. The Rh(I) organometallic magnetic nanoparticles form a novel class of heterogeneous catalyst which is particularly suitable for the practice of organic synthesis. The prepared system exhibits high catalytic efficiency in Suzuki–Miyaura and Miyaura–Michael reactions in ethanol–water solution. High yield, low reaction times, use of green solvents and non‐toxicity of the catalyst are the main merits of this protocol. Also, magnetic separation is an environmentally friendly alternative for the recovery of the catalyst, since it minimizes energy and catalyst loss by preventing mass loss and oxidation. The produced catalyst was characterized using a variety of techniques.  相似文献

The versatile application of nanoparticles in integrating imaging and therapy has aroused extensive research interest in precision medicine. Of the various nanoparticles that have been studied, CuS has shown great potential in the construction of multifunctional agents, owing to its excellent photothermal heating properties. Herein, we report a facile one‐pot biomineralization approach for the preparation of versatile bovine‐serum‐albumin‐conjugated CuS/Gd2O3 hybrid nanoparticles (BSA?CuS/Gd2O3 HNPs), which simultaneously possessed strong longitudinal relaxivity, an outstanding photothermal effect, high drug‐loading capacity, and pH/temperature‐responsive drug release. The versatile nanoparticles were used for magnetic resonance imaging (MRI) and antitumor photothermal chemotherapy, both in vitro and in vivo. In vivo MRI showed that the BSA?CuS/Gd2O3 HNPs had a long circulation time and effective passive tumor‐uptake ability. More importantly, combined in vitro and in vivo therapy demonstrated that drug‐loaded BSA?CuS/Gd2O3 HNPs offered outstanding synergistic therapeutic efficacy for tumor inhibition.  相似文献
