A new stability-indicating high-performance liquid chromatographic method has been developed for simultaneous analysis of metformin hydrochloride (MET) and sitagliptin phosphate (SIT) in pharmaceutical dosage forms. Chromatographic separation was achieved on a C8 column. The mobile phase was methanol–water 45:55 % (v/v) containing 0.2 % (w/v) n-heptanesulfonic acid and 0.2 % (v/v) triethylamine; the pH was adjusted to 3.0 with orthophosphoric acid. The flow rate was 1 mL min?1 and the photodiode-array detection wavelength was 267 nm. The linear regression coefficients for metformin and sitagliptin were 0.9998 and 0.9996 in the concentration ranges 50–450, and 10–150 μg mL?1, respectively. The relative standard deviations for intra and inter-day precision were below 1.5 %. The drugs were subjected to a variety of stress conditions—acidic and basic hydrolysis, and oxidative, photolytic, neutral, and thermal degradation. The products obtained from photolytic degradation were similar to those from neutral hydrolytic degradation and different from produced by acidic and basic hydrolysis. The method resulted in detection of 15 degradation products (D1–D15); among these, the structures of D1, D3, D9, and D13 were identified. The respective mass balance for MET and SIT was found to be close to 97.60 and 99.12 %. The specificity of the method is suitable for a stability-indicating assay.  相似文献

Abstract

Polyvinyl chloride (PVC) membrane sensors for the determination of pioglitazone hydrochloride (PIO) and metformin hydrochloride (MET) were described by using the ion association complexes between these drugs with either sodium tetraphenyl-borate (TPB) or ammonium reineckate (RNC) counter ions. The performance characteristics of the sensors were evaluated according to IUPAC recommendations, reveal a fast, stable and linear response over the concentration range 3.162 × 10?5 ? 1 × 10?2 M for PIO and 1 × 10?3 ? 1 × 10?1 M for MET. The sensors are used for determination of PIO and MET in tablets and plasma. The developed method was found to be simple, accurate and precise when compared with the reported method.  相似文献

This paper describes development and validation of a high-performance liquid chromatographic method for simultaneous analysis of tramadol hydrochloride (TR) and aceclofenac (AC) in a tablet formulation. When the combination formulation was subjected to ICH-recommended stress conditions, adequate separation of TR, AC, and the degradation products formed was achieved on a C18 column with 65:35 (v/v) 0.01 M ammonium acetate buffer, pH 6.5—acetonitrile as mobile phase at a flow rate of 1 mL min?1. UV detection was performed at 270 nm. The method was validated for specificity, linearity, LOD and LOQ, precision, accuracy, and robustness. The method was specific against placebo interference and also during forced degradation. The linearity of the method was investigated in the concentration ranges 15–60 μg mL?1 (r = 0.9999) for TR and 40–160 μg mL?1 (r = 0.9999) for AC. Accuracy was between 98.87 and 99.32% for TR and between 98.81 and 99.49% for AC. Because degradation products were well separated from the parent compounds, the method was stability-indicating.  相似文献

Abstract

Three simple, sensitive, and highly accurate spectrophotometric methods have been developed for the determination of oxomemazine hydrochloride (OXO‐HCl) in bulk and in pharmaceutical formulations. These methods are based on the formation of yellow ion‐pair complexes between the examined drug and bromocresol green (BCG), congo red (CR), and methyl orange (MO) as reagents in universal buffer solution of pH 3.0, 5.5, and 3.5, respectively. The formed complexes were extracted with chloroform and measured at 413, 495, and 484 nm, respectively for the three systems. The best conditions of the reaction were studied and optimized. Beer's law was obeyed in the concentration ranges 2.0–18.0, 2.0–14.0, and 2.0–16.0 µg ml?1 with molar absorptivity of 4.1×104, 1.1×104, and 3.5×104 mol?1cm?1, for the BCG, CR, and MO methods, respectively. Sandell's sensitivity, correlation coefficient, detection, and quantification limits are also calculated. The proposed methods have been applied successfully for the analysis of the drug in pure and in its dosage forms. No interference was observed from common pharmaceutical excipients and additives. Statistical comparison of the results with those obtained by HPLC method shows excellent agreement and indicates no significant difference in accuracy and precision.  相似文献

Abstract

This article describes validated high-performance liquid chromatographic (HPLC) and high-performance thin-layer chromatographic (HPTLC) methods for simultaneous estimation of alprazolam (ALZ) and sertraline (SER) in pure powder and tablet formulation. The HPLC separation was achieved on a Nucleosil C18 column (150 mm long, 4.6 mm i.d., and 5-µm particle size) using acetonitrile and phosphate buffer (50 + 50 v/v), pH 5.5, as the mobile phase at a flow rate of 1.0 mL/min at ambient temperature. The HPTLC separation was achieved on an aluminum-backed layer of silica gel 60 F254 using acetone/toluene/ammonia (6.0:3.0:1.0, v/v/v) as the mobile phase. Quantification with the HPLC method was achieved with ultraviolet (UV) detection at 230 nm over the concentration range 3–18 µg/mL for both drugs with mean recovery of 101.86 ± 0.21 and 100.57 ± 0.31% for ALZ and SER, respectively. Quantification in HPTLC was achieved with UV detection at 230 nm over the concentration range of 400–1400 ng/spot for both drugs with mean recoveries of 101.32 ± 0.15 and 100.38 ± 0.51% for ALZ and SER, respectively. These methods are rapid, simple, precise, sensitive, and are applicable for the simultaneous determination of ALZ and SER in pure powder and formulations.  相似文献

A stability-indicating reversed-phase liquid chromatographic (RPLC) method has been established for analysis of ramipril (RAM) and moexipril hydrochloride (MOEX.HCl) in the presence of the degradation products generated in studies of forced decomposition. The drug substances were subjected to stress by hydrolysis (0.1 m NaOH and 0.1 m HCl), oxidation (30% H2O2), photolysis (254 nm), and thermal treatment (80 °C). The drugs were degraded under basic and acidic conditions and by thermal treatment but were stable under other stress conditions investigated. Successful separation of the drugs from the degradation products was achieved on a cyanopropyl column with 40:60 (v/v) aqueous 0.01 m ammonium acetate buffer (pH 6)–methanol as mobile phase at a flow rate of 1 mL min−1. Detection was by UV absorption at 210 nm. Response was a linear function of concentration over the range 5–50 μg mL−1 (r > 0.9995), with limits of detection and quantitation (LOD and LOQ) of 0.04 and 0.09 μg mL−1, respectively, for RAM and 0.014 and 0.32 μg mL−1, respectively, for moexipril. The method was validated for specificity, selectivity, solution stability, accuracy, and precision. Statistical analysis proved the method enabled reproducible and selective quantification of RAM and MOEX as the bulk drug and in pharmaceutical preparations. Because the method effectively separates the drugs from their degradation products, it can be used as stability-indicating.  相似文献

In the present study, a simple, rapid and precise liquid chromatographic method was developed and validated for the simultaneous determination of glimepiride and metformin in sustained release formulation. The separation was achieved on a Nucleosil 100-5SA column, 250 × 4.6 mm i.d., 5 μm using a mobile phase composed of 1.7 % ammonium dihydrogen phosphate buffer pH 3.0: acetonitrile (70:30 v/v).The instrumental settings were a flow rate of 1.0 ml min?1 and a detector wavelength of 230 nm. The retention time of glimepiride and metformin were 5.1 and 11.3, respectively. The developed method was validated in terms of specificity, sensitivity, linearity, range, accuracy, precision and ruggedness. The proposed method was successfully applied to the sustained release pharmaceutical dosage form for the simultaneous determination of glimepiride and metformin without any interference by excipients.  相似文献

A simple, sensitive, and precise thin layer chromatographic (TLC) method for simultaneous analysis of psychopharmacological drugs like amitriptyline HCl, trifluoperazine HCl, risperidone and alprazolam in their single dosage forms has been developed, validated, and used for determination of the compounds in commercial pharmaceutical products. The TLC separation was carried out on Merck TLC aluminium sheets of silica gel 60 F254 using carbon tetrachloride:acetone:triethylamine (8:2:0.3, v/v/v), as mobile phase. Densitometric measurements of their spots were achieved at 250 nm over the concentration range for amitriptyline HCl (50–1,200 ng spot−1), trifluoperazine HCl (50–1,200 ng spot−1), risperidone (100–2,400 ng spot−1) and alprazolam (25–600 ng spot−1). Limit of detection (LOD) for amitriptyline HCl (20 ng spot−1), trifluoperazine HCl (20 ng spot−1), risperidone (40 ng spot−1) and alprazolam (5 ng spot−1) was obtained. The study showed that TLC was sensitive and selective for determination of amitriptyline HCl, trifluoperazine HCl, risperidone and alprazolam using a single mobile phase. This proposed method is able for simultaneous determination of psychopharmacological drugs and also applicable for analysis of pharmaceutical formulations.

Two chromatographic methods have been described for the simultaneous determination of metronidazole (MET) and spiramycin (SPY) in their mixtures. The first method was based on a high performance thin layer chromatographic (HPTLC) separation of the two drugs followed by densitometric measurements of their spots at 240 nm. The separation was carried out on Merck TLC aluminum sheets of silica gel 60 F254 using methanol: chloroform (9:1, v/v) as a mobile phase. Analysis data was used for the linear regression line in the range of 1.0–2.0 and 0.8–2.0 μg band−1 for MET and SPY, respectively. The second method was based on a reversed-phase liquid chromatographic separation of the cited drugs on a C-18 column (5 μm, 250 × 4.6 mm, i.d.). The mobile phase consisted of a mixture of phosphate buffer of pH 2.4 and acetonitrile (70:30, v/v). The separation was carried out at ambient temperature with a flow rate of 1.0 mL min−1. Quantitation was achieved with UV detection at 232 nm based on peak area with linear calibration curves at concentration ranges 0.4–50.0 and 0.5–50.0 μg mL−1 for MET and SPY, respectively. The proposed chromatographic methods were successfully applied to the determination of the investigated drugs in pharmaceutical preparations. Both methods were validated in compliance with ICH guidelines; in terms of linearity, accuracy, precision, robustness, limits of detection and quantitation and other aspects of analytical validation.

A simple, fast, inexpensive capillary zone electrophoresis method for the separation and determination of tiapride hydrochloride and its two related impurities in pharmaceutical formulations has been developed and validated. The successful separation of these compounds was achieved in less than 3 min using a fused silica capillary and photodiode array detector at 218 nm. The best conditions were obtained using a 10 mM sodium tetraborate (pH 8.0) as the running buffer. The linear responses covered the ranges from 1.0 to 100 μg mL?1 (R = 0.9989) for tiapride hydrochloride. The detection (LOD) and quantitation limits (LOQ) for tiapride hydrochloride were 2.7 and 9.0 μg mL?1, respectively. The intra- and inter-day relative standard deviations for migration times and peak areas were less than 0.47 and 5.7%, respectively. The method was validated for the determination of tiapride hydrochloride in commercial tablets.  相似文献

Thin-layer chromatography, first derivative, ratio spectra derivative spectrophotometry and Vierordt's method have been developed for the simultaneous determination of paracetamol and drotaverine HCl. TLC densitometric method depends on the difference in Rf values using ethyl acetate:methanol:ammonia (100:1:5 v/v/v) as a mobile phase. The spots of the two drugs were scanned at 249 and 308 nm over concentration ranges of 60-1200 microg/ml and 20-400 microg/ml with mean percentage recovery 100.11%+/-1.91 and 100.15%+/-1.87, respectively. The first derivative spectrophotometric method deals with the measurements at zero-crossing points 259 and 325 nm with mean percentage recovery 99.25%+/-1.08 and 99.45%+/-1.14, respectively. The ratio spectra first derivative technique was used at 246 and 305 nm with mean percentage recovery 99.75%+/-1.93 and 99.08%+/-1.22, respectively. Beer's law for first derivative and ratio spectra derivative methods was obeyed in the concentration range 0.8-12.8 and 0.4-6.4 microg/ml of paracetamol and drotaverine HCl, respectively. Vierordt's method was applied to over come the overlapping of paracetamol and drotaverine HCl in zero-order spectra in concentration range 2-26 and 2-40 microg/ml respectively. The suggested methods were successfully applied for the analysis of the two drugs in laboratory prepared mixtures and their pharmaceutical formulation. The validity of the methods was assessed by applying the standard addition technique. The obtained results were statistically agreed with those obtained by the reported method.  相似文献

Cyclizine hydrochloride (CYC) and meclozine hydrochloride (MEC) are antihistaminic drugs generally co-formulated with pyridoxine hydrochloride (PYR) to treat nausea and vomiting in pregnancy. Several analytical techniques have been applied for the determination of CYC or MEC with PYR, but determination of CYC impurity; benzhydrol (BEH) or MEC impurity; or 4-chlorobenzophenone (BEP) has not been paid attention to. Therefore, micellar UPLC method is introduced for analysis of ternary mixtures containing PYR together with both CYC and BEH (mixture I) or MEC and BEP (mixture II). Chromatographic separation was achieved using a Hypersil gold C8 column (50 × 2.1 mm, 1.9 μm) using 0.01 M sodium dodecyl sulfate modified to pH 3.5 using phosphoric acid:acetonitrile (45:55 by volume) for mixture I and 0.1% sodium dodecyl sulfate, 0.1% sodium bicarbonate adjusted to pH 2.6 by phosphoric acid:acetonitrile (47:53 by volume) for mixture II as mobile phases. The separated peaks were detected at 230 and 245 nm for mixtures I and II, respectively. The adopted methods were validated in conformance with the International Conference on Harmonization (ICH) recommendations and were properly applied in commercial pharmaceutical formulation analysis. Comprehensive ecological comparison was achieved, confirming a higher ecological value of the presented methods compared to the earlier reported methods.  相似文献

Multivariate spectrophotometric calibration and liquid chromatographic (LC) methods were applied to the determination of 2 multicomponent mixtures containing diprophylline, guaiphenesin, methylparaben, and propylparaben (Mixture 1), or clobutinol, orciprenaline, saccharin sodium, and sodium benzoate (Mixture 2). For the multivariate spectrophotometric calibration methods, principal component regression (PCR) and partial least-squares regression (PLS-1), a calibration set of the mixtures consisting of the components of each mixture was prepared in 0.1 M HCl. Analytical figures of merit such as sensitivity, selectivity, limit of quantitation, and limit of detection were determined for both PLS-1 and PCR. The LC separation was achieved on a reversed-phase C18 analytical column by using isocratic elution with 20 mM potassium dihydrogen phosphate, pH 3.3-acetonitrile (55 + 45, v/v) as the mobile phase and UV detection at 260 and 220 nm for Mixture 1 and Mixture 2, respectively. The proposed methods were validated and successfully applied to the analysis of pharmaceutical formulations and laboratory-prepared mixtures containing the 2 multicomponent combinations.  相似文献

Abstract

A highly sensitive, spectrofluorometric method was developed for the determination of drotaverine HCl in pharmaceutical preparations. The proposed method is based on the measurement of the native fluorescence of the drug in 0.1 M H2SO4 at emission 465 nm after excitation at 295 nm. The relative fluorescence intensity-concentration plot was rectilinear over the range of 0.16–4 µg·ml?1, with good correlation (r = 0.9999) and a lower limit of detection (LOD) of 0.032 µg·ml?1. The proposed method was successfully applied for the determination of drotaverine HCl in tablets and ampoules with a recovery percentage of 100.42 ± 0.601,99.83 ± 0.82 and 99.43 ± 1.08, respectively, which were in accordance with those obtained by a compendial method.  相似文献

Abstract

A simple, reproducible, accurate, and effective spectrophotometric method was developed and validated for the quantitation of the antihistamine fexofenadine in capsules and coated tablets. Ethanol was used as solvent and the absorbance at the wavelength of 220 nm was employed to the quantitation of the drug. The method validation was fulfilled through the evaluation of the analytical parameters of linearity, precision, accuracy, limits of detection, and quantitation and specificity. The method was linear (r=0.9999) at concentrations ranging from 8.0 to 20.0 µg ml?1, precise (RSD intra‐day=0.29; 0.18; 0.39; RSD inter‐day=0.12 for capsules and RSD intra‐day=0.13; 0.16; 0.13; RSD inter‐day=0.13 for coated tablets), accurate (percentage recovery=99.97% for capsules and 100.51% for tablets), sensitive (limits of detection and quantitation of 0.10 and 0.29 µg ml?1, respectively) and specific. The method was compared to a high performance liquid chromatography (HPLC) method, which was previously developed to the same drug. The results showed no significant difference between the methods in fexofenadine hydrochloride quantitation.  相似文献

Two methods are described for simultaneous determination of amlodipine besylate and olmesartan medoxomil in formulation. The first method was based on the HPTLC separation of two drugs on Merck HPTLC aluminium sheets of silica gel 60 F254 using n-butanol: acetic acid: water (5:1:0.1, v/v/v) as the mobile phase. The second method was based on the HPLC separation of the two drugs on the RP-PerfectSil-100 ODS-3–C18 column from MZ-Analysetechnik GmbH, Germany and acetonitrile/0.03 M ammonium acetate buffer (pH = 3) in a ratio of 55:45 as the mobile phase. Both methods have been applied to formulation without interference of excipients of formulation.  相似文献

A simple and rapid LC method was developed and validated for simultaneous estimation of nebivolol and amlodipine in a bi-layer tablet formulation. Efficient chromatographic separation was achieved on (USP L10) Hypersil BDS cyano, 5 μm, 250 mm × 4.6 mm column with simple mobile phase composition delivered in isocratic mode. The method had requisite accuracy, selectivity, sensitivity, robustness and precision to assay nebivolol and amlodipine in pharmaceutical dosage form. Degradation products resulting from the stress studies did not interfere with the detection of nebivolol and amlodipine, these peaks remained pure and thus proved to be stability indicating. The mass balance of the stressed sample was in the range 99.0–100.2% for amlodipine and 99.3–100.3% for nebivolol.  相似文献

Abstract

Two simple, rapid, accurate, and economical analytical methods have been developed for the simultaneous estimation of Rabeprazole Sodium and Itopride Hydrochloride in combined capsule dosage form. First method is based on the determination of Q‐value and second method is based on simultaneous equation method. Rabeprazole Sodium has absorbance maxima at 284 nm and Itopride Hydrochloride has absorbance maxima at 258 nm in methanol AR. The absorption ratio (Q‐value) was determined at 266.6 nm (Iso‐bestic point) and 284 nm (λmax of Rabeprazole Sodium). Both the drugs obey Beer's law in the concentration ranges employed for these methods. Both the methods were found to be simple, rapid, accurate, and can be adopted in routine analysis of drugs in formulations. The accuracy and reproducibility of the proposed method was statistically validated by recovery studies  相似文献
