Two new lycodine‐type alkaloids, huperzinine N‐oxide ( 1 ) and 8,15‐dihydrohuperzinine ( 2 ) as well as five known compounds, huperzinine ( 3 ), huperzine B ( 4 ), huperzine D ( 5 ), N‐demethylhuperzinine ( 6 ), and β‐obscurine ( 7 ), were isolated from the club moss Lycopodium casuarinoides. The structures of 1 and 2 were elucidated by spectroscopic methods and chemical transformation. The absolute configuration of 1 was established by chemical correlation with 3 , and that of 2 was determined by its CD spectrum.  相似文献

Three new lycopodium alkaloids, huperserramines A–C ( 1 – 3 , resp.), along with 15 known ones, lycopodine‐6α,11α‐diol ( 4 ), lycoposerramine H ( 5 ), lycoposerramine I ( 6 ), lycopodine‐6α‐ol ( 7 ), lycoposerramine M ( 8 ), diphaladine A ( 9 ), lycoposerramine K ( 10 ), lycoposerramine W ( 11 ), huperzine M ( 12 ), luciduline ( 13 ), phlegmariuine N ( 14 ), huperzine A ( 15 ), huperzine B ( 16 ), lycodine ( 17 ), and lycoposerramine R ( 18 ), were isolated from the whole plant of Huperzia serrata. Their structures were established by spectroscopic methods, including 2D‐NMR and MS analyses. All the isolates were evaluated for their inhibitory effects on acetylcholinesterase (AChE) and α‐glucosidase. As a result, lycopodine‐6α,11α‐diol ( 4 ) exhibited more potent α‐glucosidase inhibitory activity (IC50 148±5.5 μM ) than the positive control acarbose (IC50 376.3±2.7 μM ).  相似文献

The newly isolated Lycopodium alkaloids huperzinine, N,N-dimethylhuperzine A and N-methylhuperzine B were studied by high-resolution and tandem mass Spectrometry. Differentiation of these subtly different isomers from each other and from the general class of Lycopodium alkaloids can be achieved by taking advantage of the structure-specific fragment ions formed upon electron ionization (El) or collisional activation of these alkaloids. β-Obscurine was identified as an impurity component in the N-methylhuperzine B sample and contributed to the El mass spectrum. The previously published El mass spectrum of N-methylhuperzine B is likely to be that of a mixture of N-methylhuperzine B and β-obscurine. The presence of the impurity can now be readily recognized on the basis of two characteristic fragment ions of m/z 215 and 187.  相似文献

A series of an ionic hydrogels composed of N,N‐diethylaminoethyl methacrylamide (DEAEMA), N‐vinyl‐2‐pyrrolidone (VP), and itaconic acid were synthesized by free‐radical cross‐linking copolymerization in water–ethanol mixture by using N,N‐methylenebis(acrylamide) as the cross‐linker, ammonium persulfate as the initiator, and N,N,N′,N′‐tetramethylenediamine as the activator. The swelling behaviors of these hydrogels were analyzed in buffer solutions at various pH. It was observed that the swelling behavior of cross‐linked ionic poly(N,N‐diethylaminoethyl methacrylamide‐coN‐vinyl‐2‐pyrrolidone) [P(DEAEMA/VP)] hydrogels at different pH agreed with the modified Flory–Rehner equation based on the affine network model and the ideal Donnan theory. The swelling process in buffer solutions at various pH was found to be Fickian‐type diffusion. The pH‐reversibility and on–off switching properties of the P(DEAEMA/VP) hydrogels may be considered as good candidate to design novel drug‐delivery system. © 2005 Wiley Periodicals, Inc. A rapid, simple and sensitive UHPLC‐MS/MS method was developed and validated for the simultaneous determination of brucine, strychnine and brucine N‐oxide in rat plasma using huperzine A as an internal standard (IS) after protein precipitation with methanol. The analytes were separated on a Purospher® STAR RP18 UHPLC column (2 µm, 2.1 × 100 mm) by gradient elution using a mobile phase composed of methanol and water (containing 0.1% formic acid) at a flow rate of 0.3 mL/min. Brucine, strychnine, brucine N‐oxide and IS were detected in positive ion multiple reaction monitoring mode by means of an electrospray ionization interface (m/z 395.2 → 324.1, m/z 335.2 → 184.1, m/z 411.2 → 394.2, m/z 243.1 → 226.1). The calibration curve was linear over the range of 1–500 ng/mL for brucine and strychnine and 0.2?50 ng/mL for brucine N‐oxide. The intra‐ and inter‐day precisions of these analytes were all within 15% and the accuracy ranged from 85 to 115%. The stability experiment indicated that the plasma samples at three concentration levels were stable under different conditions. The developed method was successfully applied for the first time to pharmacokinetic studies of brucine, strychnine and brucine N‐oxide following a single oral and intravenous administration of modified total alkaloid fraction in rats. A series of pyrimido[1,2‐a]benzimidazoles were obtained from aldehydes, 2‐aminobenzimidazole and ethyl acetoacetate in good‐to‐excellent yields by a simple, mild, and efficient procedure utilizing N,N,N′,N′‐tetrabromobenzene‐1,3‐disulfonamide (TBBDA) and poly(N‐bromo‐N‐ethylbenzene‐1,3‐disulfonamide) (PBBS) as catalysts.  相似文献

The reaction of N,N′‐diarylselenoureas 16 with phenacyl bromide in EtOH under reflux, followed by treatment with NH3, gave N,3‐diaryl‐4‐phenyl‐1,3‐selenazol‐2(3H)‐imines 13 in high yields (Scheme 2). A reaction mechanism via formation of the corresponding Se‐(benzoylmethyl)isoselenoureas 18 and subsequent cyclocondensation is proposed (Scheme 3). The N,N′‐diarylselenoureas 16 were conveniently prepared by the reaction of aryl isoselenocyanates 15 with 4‐substituted anilines. The structures of 13a and 13c were established by X‐ray crystallography.  相似文献

A kinetic and product study of the reaction of a series of α‐methyl‐substituted N‐methylpiperidines with thermally generated 1O2 in MeCN was carried out. It was found that as the number of α‐methyl groups (Me in α‐position relative to the N‐atom) increases, the rate of 1O2 quenching (physical plus chemical) slightly decreases. This finding shows that, with respect to the reaction rate, steric effects are much more important than electronic effects as the latter should have produced the opposite result. The opposite outcome was instead found for the chemical quenching that leads to the N‐demethylation products and N‐formyl derivatives. The same trend was observed for the ratio between N‐demethylation and formation of the N‐formyl derivatives (NH/NCHO ratio). All these results are consistent with the mechanism reported in Scheme 1 where an exciplex is first formed that by a H‐atom transfer process produces an α‐amino‐substituted C‐radical. The latter forms the product of N‐demethylation by one electron oxidation, or affords the N‐formyl derivative by radical coupling (Scheme 1). Similar results were obtained with N,N‐dimethylcyclohexanamine. However, this ‘acyclic’ amine exhibited behaviors quite distinct from those of the N‐methylpiperidines series, with respect to reaction rate, extent of chemical quenching, and NH/NCHO ratio.  相似文献

A variety of chiral N,N‐bidentate and N,N,N‐tridentate ligands based on the pyridine framework, namely C2‐symmetric dipyridylmethane and terpyridine, N‐(p‐toluensulfinyl)iminopyridines and two kinds of iminopyridines, has been assessed in the asymmetric copper(I)‐catalysed allylic oxidation of cyclic olefins. Catalytic activity and enantioselectivity were found to be highly dependent upon the framework of the ligands, which afforded cycloalkenyl benzoates in low to moderate yields and enantioselectivities. The best yields (up to 70%) and enantioselectivities (up to 53% enantiomeric excess) were obtained with an iminopyridine based on camphane and quinoline skeletons. In a search for new insect growth regulators with unusual biological properties and different activity spectrum, we thought that the preservation of the bioactive unit and the introduction of 2‐methyl‐3‐(triphenylgermanyl)propoxycarbonyl in Ntert‐butyl‐N,N′‐dibenzoylhydrazine would enhance their larvicidal activities to a significant degree. Therefore, we designed and synthesized N′‐tert‐butyl‐N′‐[2‐methyl‐3‐(triphenylgermanyl)propoxycarbonyl]‐N‐benzoylhydrazine and analogs by two procedures. These novel compounds were characterized by elemental analyses, IR, and 1H NMR. At the same time, Ntert‐butyl‐N‐substitutedbenzoylhydrazines were prepared by a new method, and some reactions involved were studied. The preliminary results indicate that some compounds have inhibitory effects against plant pathogenetic bacteria such as early blight of tomato. Copyright © 2002 John Wiley & Sons, Ltd.  相似文献

(S)‐N‐(3,5‐dinitrobenzoyl)leucine‐N‐phenyl‐N‐propylamine‐bonded silica was used as a chiral stationary phase for separation of a set of racemic π‐acidic and π‐basic α‐amino acid amides in electrolyteless ACN‐water eluents by CEC in the RP and polar organic (PO) modes. The effect of the amount of water in the ACN‐water eluent on chiral separation was examined. As water is added to ACN, retention was shortened but resolution and selectivity deteriorated severely. Retention, enantioselectivity, and resolution factors obtained in 100% ACN were compared with those in an n‐hexane‐isopropanol eluent with a small amount of water by normal phase (NP) CEC. Much shorter retention times with comparable enantioselectivities were observed with 100% ACN, demonstrating the advantage of separation on (S)‐N‐(DNB)leucine‐N‐phenyl‐N‐propylamine‐bonded silica in PO‐CEC over NP‐CEC.  相似文献

A convenient and efficient method for a one‐pot conversion of N‐alkylisatins to N‐alkylisatin O‐alkyloximes 7a – 7n as potential chemotherapeutic agents is described (Scheme) (isatin=1H‐indole‐2,3‐dione). In this method, the microwave‐assisted three‐component reaction of N‐alkylisatins 8 , hydroxylamine hydrochloride, and diverse alkyl halides in the presence of K2CO3 and Bu4NBr furnishes the corresponding N‐alkylisatin O‐alkyloximes under solvent‐free condition in short times (2–10 min) and good to excellent yields (62–83%). The O‐alkylation of in situ generated isatin oximes with alkyl halides was achieved regioselectively, and (Z)‐O‐alkyloximes were produced dominantly. PM3 Semi‐empirical quantum‐mechanic calculations were performed to rationalize the evidences, and the calculations indicated a lower heat of formation for the (Z)‐O‐alkyloximes.  相似文献

A new N‐phenylated amide (N‐phenylamide) unit containing aromatic diamine, N,N′‐bis(3‐aminobenzoyl)‐N,N′‐diphenyl‐1,4‐phenylenediamine, was prepared by the condensation of N,N′‐diphenyl‐1,4‐phenylenediamine with 3‐nitrobenzoyl chloride, followed by catalytic reduction. Two series of organosoluble aromatic poly(N‐phenylamide‐imide)s and poly(N‐phenylamide‐amide)s with inherent viscosities of 0.58–0.82 and 0.56–1.21 dL/g were prepared by a conventional two‐stage method and the direct phosphorylation polycondensation, respectively, from the diamine with various aromatic dianhydrides and aromatic dicarboxylic acids. All polyimides and polyamides are amorphous and readily soluble in many organic solvents such as N,N‐dimethylacetamide and N‐methyl‐2‐pyrrolidone. These polymers could be solution cast into transparent, tough, and flexible films with high tensile strengths. These polyimides and polyamides had glass‐transition temperatures in the ranges of 230–258 and 196–229 °C, respectively. Decomposition temperatures of the polyimides for 10% weight loss all occurred above 500 °C in both nitrogen and air atmospheres. © 2002 Wiley Periodicals, Inc. Poly(N‐acryloxysuccinimide) (PNAS) and poly(N‐acryloxysuccinimide‐coN‐vinylpyrrolidone) (P(NAS‐co‐NVP)) of adjustable molecular weights and narrow polydispersities were prepared by nitroxide‐mediated polymerization (NMP) in N,N‐dimethylformamide in the presence of free SG1 (Ntert‐butyl‐N‐1‐diethylphosphono‐(2,2‐dimethylpropyl) nitroxide), with MAMA‐SG1 (N‐(2‐methylpropyl)‐N‐(1‐diethylphosphono‐2,2‐dimethylpropyl)‐O‐(2‐carboxylprop‐2‐yl)hydroxylamine) alkoxyamine as initiator. The reactivity ratios of NAS and NVP were determined to be rNAS = 0.12 and rNVP = 0, indicating a strong alternating tendency for the P(NAS‐co‐NVP) copolymer. NAS/NVP copolymerization was then performed from a SG1‐functionalized poly(D ,L ‐lactide) (PLA‐SG1) macro‐alkoxyamine as initiator, leading to the corresponding PLA‐b‐P(NAS‐co‐NVP) block copolymer, with similar NAS and NVP reactivity ratios as mentioned above. The copolymer was used as a surface modifier for the PLA diafiltration and nanoprecipitation processes to achieve nanoparticles in the range of 450 and 150 nm, respectively. The presence of the functional/hydrophilic P(NAS‐co‐NVP) block, and particularly the N‐succinimidyl (NS) ester moieties at the particle surface, was evidenced by ethanolamine derivatization and zeta potential measurements. © 2011 Wiley Periodicals, Inc. The synthesis of a series of N‐glycosyl caboranylquinazolines is described. The condensation reaction of nitro‐acetylanthranilic acid with aminophenylcarborane gave 3‐[(o‐carboran‐1‐yl)phenyl]‐2‐methyl‐6‐nitroquinazolin‐4(3H)‐one 1 followed by reduction with Na2S to the corresponding 6‐amino‐3‐[(o‐carboran‐1‐yl)phenyl]‐2‐methylquinazolin‐4(3H)‐one 2 . Reaction of compound 2 with D‐glucose or D‐ribose in methanol in the presence of a catalytic amount of acetic acid affords boronated N‐glycosylaminoquinazolines namely: 2‐methyl‐3‐[4‐(o‐carboran‐1‐yl)phenyl]‐6‐[N‐β‐D‐glucopyranosyl)]aminoquinazolin‐4(3H)‐one 3 or 2‐methyl‐3‐[4‐(o‐carboran‐1‐yl)phenyl]‐6‐[N‐β‐D‐ribofuranosyl)]aminoquinazolin‐4(3H)‐one 4 , respectively. Degradation of the o‐caborane cage of compounds 3 and 4 yielded highly water‐soluble compounds of sodium 2‐methyl‐3‐[4‐( nido ‐undecarborate‐1‐yl)phenyl]‐6‐[N‐β‐D‐glucopyranosyl]aminoquinazolin‐4(3H)‐one 5 and sodium 2‐methyl‐3‐[4‐( nido ‐undecarborate‐1‐yl)phenyl]‐6‐[N‐β‐D‐ribofuranosyl)]aminoquinazolin‐4(3H)‐one 6 , respectively. The structures were established on the basis of elemental analysis, NMR, IR and mass spectrometry. The in vitro toxicity test using B16 melanoma cells showed that N‐glycosyl of nido ‐undecaboranylquinazolines ( 5 and 6 ), with higher water solubility, is not toxic at boron concentration of 3000 µg boron ml−1, whereas, N‐glycosyl of closo ‐carboranylquinazolines ( 3 and 4 ) has LD50 > 200 µg boron ml−1. A convenient method for the synthesis of 2,4,8‐trisubstituted 1,7‐naphthyridines 6 by the reaction of (E)‐4‐(1‐aryl‐2‐methoxyethenyl)‐3‐isocyanopyridines 4 , which could be easily prepared from commercially available 3‐aminopyridine via aroylation of lithium (4‐lithiopyridin‐3‐yl)pivalamide with N‐methoxy‐N‐methylbenzamides, with excess organolithiums has been developed.  相似文献

Terpolymers composed of Nn‐propylacrylamide (NPAAm), butyl methacrylate (BMA), and N,N‐diethylaminoethyl methacrylate (DEAEMA) were prepared in an attempt to investigate the temperature‐induced phase transition and its mechanism. Poly(NPAAm) showed the lower critical solution temperature (LCST) around 24°C in water. With the incorporation of DEAEMA with NPAAm, the LCST change was characterized by an initial increase. However, the LCST was shifted to the lower temperature at the later stage. This might be explained in terms of hydrophilic/hydrophobic contribution of DEAEMA to the LCST. The swelling behavior of copolymer gel in the various solvents and spin‐lattice relaxation time (T1) study by NMR strongly suggested the hydrophilic/hydrophobic contribution of DEAEMA to the LCST depending on the local environment. © 1999 John Wiley & Sons, Inc. New triphenylantimony(V) o‐amidophenolates (AP‐Me,Et)SbPh3 (1) and (AP‐Me,iPr)SbPh3 (2) with unsymmetrically substituted N‐aryl groups and (AP‐Et,Et)SbPh3 (3) with symmetrical N‐aryl group {AP‐R1,R2 is 4,6‐di‐tert‐butyl‐N‐[2‐alkyl(R1),6‐alkyl(R2)‐phenyl]‐o‐amidophenolate dianion} were synthesized and characterized in detail. Complexes were examined for dioxygen activity. The unsymmetrical complexes 1 and 2 were found to form different geometrical isomers (A and B) of spiroendoperoxides [L‐R1,R2(O2)]SbPh3 (4 and 5, respectively) with different dispositions of peroxide group and N‐aryl fragment (methyl and peroxide group are on the same side of the molecule in the less shielded isomer A, and on different sides in the more hindered isomer B). The isomer A prevails over isomer B, reflecting the possibility of steric control on the dioxygen‐binding reaction. Complex 3, where R1 = R2 = Et, formed the isomers 6A and 6B as 50:50. The ratio 4A:4B was 60:40 (for methyl‐ethyl containing complex 4) and it increased up to 80:20 for methyl‐isopropyl‐containing 5. Although the amide group of N,N‐diethyl‐2,4‐dimethoxybenzamide facilitates directed ortho‐metallation and subsequent allylation of the aromatic ring, it is not readily hydrolyzed prior to conversion into the title isochromenone. This article describes the use of 1‐(2‐allyl‐4,5‐dimethoxybenzoyl)‐4‐methylpiperazine and N,N‐diethyl‐2‐(2‐hydroxypropyl)‐4,6‐dimethoxybenzamide as alternative substrates for conversion into 6,8‐dimethoxy‐3‐methyl‐3,4‐dihydro‐1H‐isochromen‐1‐one.  相似文献
