Pd(II) and Pt(II) complexes with the anions of the model nucleobases 1-methylthymine (1-MethyH), 1-methyluracil (1-MeuraH), and 1-methylcytosine (1-MecytH) of the types [Pd(dmba)(mu-L)]2 [dmba = N,C-chelating 2-((dimethylamino)methyl)phenyl; L = 1-Methy, 1-Meura or 1-Mecyt] and [M(dmba)(L)(L')] [L = 1-Methy or 1-Meura; L' = PPh(3) (M = Pd or Pt), DMSO (M = Pt)] have been obtained. Palladium complexes of the types [Pd(C6F5)(N-N)(L)] [L = 1-Methy or 1-Meura; N-N = N,N,N',N'-tetramethylethylenediamine (tmeda), 2,2'-bipyridine (bpy), or 4,4'-dimethyl-2,2'-bipyridine (Me2bpy)] and [NBu4][Pd(C6F5)(1-Methy)2(H2O)] have also been prepared. The crystal structures of [Pd(dmba)(mu-1-Methy)]2, [Pd(dmba)(mu-1-Mecyt)]2.2CHCl3, [Pd(dmba)(1-Methy)(PPh3)].3CHCl3, [Pt(dmba)(1-Methy)(PPh3)], [Pd(tmeda)(C6F5)(1-Methy)], and [NBu4][Pd(C6F5)(1-Methy)2(H2O)].H2O have been established by X-ray diffraction. The DNA adduct formation of the new platinum complexes synthesized was followed by circular dichroism and electrophoretic mobility. Atomic force microscopy images of the modifications caused by the platinum complexes on plasmid DNA pBR322 were also obtained. Values of IC50 were also calculated for the new platinum complexes against the tumor cell line HL-60. All the new platinum complexes were more active than cisplatin (up to 20-fold in some cases).  相似文献

The structures of the ternary palladium(II) complexes of the formulations [Pd(Gly)(bpy)](+)Cl(-).4H(2)O (Gly=glycine; bpy=2,2'-bipyridine) (1), [Pd(Gly)(phen)](+)Cl(-).4H(2)O (2) (phen=1,10-phenanthroline) and {[Pd(Gly)(bpa)](+)Cl(-)}(2).6H(2)O (3) (bpa=2,2'-bipyridylamine) were determined. All complexes are positively charged and neutralized by the chloride anion located nearby the complexes. The central Pd(II) atoms of the complexes 1, 2 and 3 have a similar distorted square planar coordination geometry, in which each Pd(II) atom is coordinated to two N atoms of the bidentate heterocyclic ligand, and N and O atoms of the bidentate glycine ligand. The interaction of the complexes with calf thymus (CT) DNA was also studied using the fluorescence method. All complexes showed the inhibition of ethidium bromide binding to CT DNA, and the DNA-binding strengths were reflected as the relative order 2>1>3. The remarkable reduction of UV absorption intensity of 2 caused in the presence of DNA suggests the presence of pi-pi stacking interaction between the heterocyclic ring of the phen ligand and nucleobases. The intercalative DNA-binding of 2 is suggested by UV and CD measurements. DNA cleavage studies indicated that the cleavage of the plasmid supercoiled pBR322 DNA in the presence of H(2)O(2) and ascorbic acid could be enhanced by the complexes.  相似文献

Two bis(amido)ruthenium(IV) complexes, [Ru(IV)(bpy)(L-H)(2)](2+) and [Ru(IV)(L)(L-H)(2)](2+) (bpy = 2,2'-bipyridine, L = 2,3-diamino-2,3-dimethylbutane, L-H = (H(2)NCMe(2)CMe(2)NH)(-)), were prepared by chemical oxidation of [Ru(II)(bpy)(L)(2)](2+) and the reaction of [(n-Bu)(4)N][Ru(VI)NCl(4)] with L, respectively. The structures of [Ru(bpy)(L-H)(2)][ZnBr(4)].CH(3)CN and [Ru(L)(L-H)(2)]Cl(2).2H(2)O were determined by X-ray crystal analysis. [Ru(bpy)(L-H)(2)][ZnBr(4)].CH(3)CN crystallizes in the monoclinic space group P2(1)/n with a = 12.597(2) ?, b = 15.909(2) ?, c = 16.785(2) ?, beta = 91.74(1) degrees, and Z = 4. [Ru(L)(L-H)(2)]Cl(2).2H(2)O crystallizes in the tetragonal space group I4(1)/a with a = 31.892(6) ?, c = 10.819(3) ?, and Z = 16. In both complexes, the two Ru-N(amide) bonds are cis to each other with bond distances ranging from 1.835(7) to 1.856(7) ?. The N(amide)-Ru-N(amide) angles are about 110 degrees. The two Ru(IV) complexes are diamagnetic, and the chemical shifts of the amide protons occur at around 13 ppm. Both complexes display reversible metal-amide/metal-amine redox couples in aqueous solution with a pyrolytic graphite electrode. Depending on the pH of the media, reversible/quasireversible 1e(-)-2H(+) Ru(IV)-amide/Ru(III)-amine and 2e(-)-2H(+) Ru(IV)-amide/Ru(II)-amine redox couples have been observed. At pH = 1.0, the E degrees is 0.46 V for [Ru(IV)(bpy)(L-H)(2)](2+)/[Ru(III)(bpy)(L)(2)](3+) and 0.29 V vs SCE for [Ru(IV)(L)(L-H)(2)](2+)/[Ru(III)(L)(3)](3+). The difference in the E degrees values for the two Ru(IV)-amide complexes has been attributed to the fact that the chelating saturated diamine ligand is a better sigma-donor than 2,2'-bipyridine.  相似文献

Palladium and platinum complexes with HmtpO (where HmtpO=4,7-dihydro-5-methyl-7-oxo[1,2,4]triazolo[1,5-a]pyrimidine, an analogue of the natural occurring nucleobase hypoxanthine) of the types [M(dmba)(PPh3)(HmtpO)]ClO4[dmba=N,C-chelating 2-(dimethylaminomethyl)phenyl; M=Pd or Pt], [Pd(N-N)(C6F5)(HmtpO)]ClO4[N-N=2,2'-bipyridine (bpy), 4,4'-dimethyl-2,2'-bipyridine (Me2bpy), or N, N, N', N'-tetramethylethylenediamine (tmeda)] and cis-[M(C6F5)2(HmtpO)2] (M=Pd or Pt) (head-to-head atropisomer in the solid state) have been obtained. Pd(II) and Pt(II) complexes with the anion of HmtpO of the types [Pd(tmeda)(C6F5)(mtpO)], [Pd(dmba)(micro-mtpO)] 2, and [NBu4]2[M(C6F5)2(micro-mtpO)]2(M=Pd or Pt) have been prepared starting from the corresponding hydroxometal complexes. Complexes containing simultaneously both the neutral HmtpO ligand and the anionic mtpO of the type [NBu4][M(C6F5)2(HmtpO)(mtpO)] (M=Pd or Pt) have been also obtained. In these mtpO-HmtpO metal complexes, for the first time, prototropic exchange is observed between the two heterocyclic ligands. The crystal structures of [Pd(dmba)(PPh 3)(HmtpO)]+, cis-[Pt(C6F5)2(HmtpO)2].acetone, [Pd(C6F5)(tmeda)(mtpO)].2H2O, [Pd(dmba)(micro-mtpO)]2, [NBu4]2[Pd(C6F5)2(micro-mtpO)]2.CH2Cl2.toluene, [NBu4]2[Pt(C6F5)2(micro-mtpO)](2).0.5(toluene), and [NBu4][Pt(C6F5)2(mtpO)(HmtpO)] have been established by X-ray diffraction. Values of IC50 were calculated for the new platinum complexes cis-[Pt(C6F5)2(HmtpO)2] and [Pt(dmba)(PPh3)(HmtpO)]ClO4 against a panel of human tumor cell lines representative of ovarian (A2780 and A2780 cisR), lung (NCI-H460), and breast cancers (T47D). At 48 h incubation time, both complexes were about 8-fold more active than cisplatin in T47D and show very low resistance factors against an A2780 cell line, which has acquired resistance to cisplatin. The DNA adduct formation of cis-[Pt(C6F5)2(HmtpO)2] and [Pt(dmba)(PPh3)(HmtpO)]ClO4 was followed by circular dichroism and electrophoretic mobility. Eight new platinum(II)/palladium(II) complexes with 4-toluenesulfonyl-L-amino acid dianion and diimine/diamine ligands, [Pd(en)(Tsile)]·H2O (1), [Pd(bipy)(Tsile)] (2), [Pd(bipy)(Tsthr)]·0.5H2O (3), [Pd(phen)(Tsile)]·0.5H2O (4), [Pd(phen)(Tsthr)]·H2O (5), [Pd(bqu)(Tsthr)]·1.5H2O (6), [Pt(en)(Tsser)] (7), and [Pt(en)(Tsphe)]·H2O (8), have been synthesized and characterized by elemental analyses, 1H NMR and mass spectrometry. The crystal structure of 7 has been determined by X-ray diffraction. Cytotoxicities were tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and sulforhodamine B assays. The complexes exert cytotoxicity against HL-60, Bel-7402, BGC-823, and KB cell lines with 4 having the best cytotoxicity against HL-60, Bel-7402, and BGC-823 cell lines; the compounds are less cytotoxic than cisplatin.  相似文献

Reactions between the Os(VI)-nitrido complexes, [OsVI(L2)(Cl)3(N)] (L2 = 2,2'-bipyridine (bpy) ([1]), 4,4'-dimethyl-2,2'-bipyridine (Me2bpy), 1,10-phenanthroline (phen), and 4,7-diphenyl-1,10-phenanthroline (Ph2phen)), and bis-(triphenylphosphoranylidene)ammonium azide (PPNN3) in dry CH3CN at 60 degrees C under N2 give the corresponding Os(IV)-azidoimido complexes, [OsIV(L2)(Cl)3(NN3)]- (L2 = bpy = [2]-, L2 = Me2bpy = [3]-, L2 = phen = [4]-, and L2 = Ph2phen = [5]-) as their PPN+ salts. The formulation of the N42- ligand has been substantiated by 15N-labeling, IR, and 15N NMR measurements. Hydroxylation of [2]- at Nalpha with O<--NMe3.3H2O occurs to give the Os(IV)-azidohydroxoamido complex, [OsIV(bpy)(Cl)3(N(OH)N3)] ([6]), which, when deprotonated, undergoes dinitrogen elimination to give the Os(II)-dinitrogen oxide complex, [OsII(bpy)(Cl)3(N2O)]- ([7]-). The reactions of bidentate diimine ligands (L2) with cationic bis(diimine)[Ru(L)(L1)(CO)Cl]+ complexes (L, L1, L2 are dissimilar diimine ligands), in the presence of trimethylamine-N-oxide (Me3NO) as a decarbonylation reagent, lead to the formation of heteroleptic tris(diimine) ruthenium(II) complexes, [Ru(L)(L1)(L2)]2+. Typically isolated as hexafluorophosphate or perchlorate salts, these complexes were characterised by UV-visible, infrared and mass spectroscopy, cyclic voltammetry, microanalyses and NMR spectroscopy. Single crystal X-ray studies have elucidated the structures of K[Ru(bpy)(phen)(4,4'-Me(2)bpy)](PF(6))(3).1/2H(2)O, [Ru(bpy)(5,6-Me(2)phen)(Hdpa)](ClO(4))(2), [Ru(bpy)(phen)(5,6-Me(2)phen)](ClO(4))(2), [Ru(bpy)(5,6'-Me(2)phen)(4,4'-Me(2)bpy)](PF(6))(2).EtOH, [Ru(4,4'-Me(2)bpy)(phen)(Hdpa)](PF(6))(2).MeOH and [Ru(bpy)(4,4'-Me(2)bpy)(Hdpa)](ClO(4))(2).1/2Hdpa (where Hdpa is di(2-pyridyl)amine). A novel feature of the first complex is the presence of a dinuclear anionic adduct, [K(2)(PF(6))(6)](4-), in which the two potassium centres are bridged by two fluorides from different hexafluorophosphate ions forming a K(2)F(2) bridging unit and by two KFPFK bridging moieties.  相似文献

Two neutral complexes of formula [M(bpy)(cbdca)] [where M is palladium(II) (Pd(II)) or platinum(II) (Pt(II)), bpy is 2,2'-bipyridine and cbdca is anion of 1,1-cyclobutanedicarboxylic acid] have been synthesized. These water soluble complexes have been characterized by chemical analysis and conductivity measurements as well as 1H-NMR, ultraviolet-visible and infrared spectroscopy. In these complexes the ligand cbdca coordinates to Pt(II) or Pd(II) as bidentate with two oxygen atoms. They are nonelectrolyte in conductivity water. These complexes inhibit the growth of P388 lymphocytic leukemia cells and their targets are DNA. They invariably show ID50 values less than cisplatin. [Pt(bpy)(cbdca)] and [Pd(bpy)(cbdca)] have been interacted with calf thymus DNA and bind to DNA through coordinate covalent bond. In addition, the influence of binding of these complexes on the intensity of EtBr-DNA have been studied. They bind to DNA via a nonintercalating mode.  相似文献

The reaction of the functional diphosphine 1 [1 = 2-(bis(diphenylphosphino)methyl-oxazoline] with [PtCl(2)(NCPh)(2)] or [PdCl(2)(NCPh)(2)], in the presence of excess NEt(3), affords [Pt{(Ph(2)P)(2)C···C(···NCH(2)CH(2)O)}(2)] ([Pt(1(-H)-P,P)(2)], 3a) and [Pd{(Ph(2)P)(2)C···C(···NCH(2)CH(2)O)}(2)] ([Pd(1(-H)-P,P)(2)], 3b), respectively, in which 1(-H) is (oxazoline-2-yl)bis(diphenylphosphino)methanide. The reaction of 3b with 2 equiv of [AuCl(tht)] (tht = tetrahydrothiophene) afforded [Pd(1(-H)-P,N)(2)(AuCl)(2)] (4), as a result of the opening of the four-membered metal chelate since ligand 1(-H), which was P,P-chelating in 3b, behaves as a P,N-chelate toward the Pd(II) center in 4 and coordinates to Au(I) through the other P donor. In the absence of a base, the reaction of ligand 1 with [PtCl(2)(NCPh)(2)] in MeCN or CH(2)Cl(2) afforded the isomers [Pt{(Ph(2)P)(2)C═C(OCH(2)CH(2)NH)}(2)]Cl(2) ([Pt(1'-P,P)(2)]Cl(2) (5), 1' = 2-(bis(diphenylphosphino)methylene)-oxazolidine) and [Pt{(Ph(2)P)(2)C═C(OCH(2)CH(2)NH)}{Ph(2)PCH═C(OCH(2)CH(2)N(PPh(2))}]Cl(2) ([Pt(1'-P,P)(2'-P,P)]Cl(2) (6), 2' = (E)-3-(diphenylphosphino)-2-((diphenylphosphino)methylene)oxazolidine]. The P,P-chelating ligands in 5 result from a tautomeric shift of the C-H proton of 1 to the nitrogen atom, whereas the formation of one of the P,P-chelates in 6 involves a carbon to nitrogen phosphoryl migration. The reaction of 5 and 6 with a base occurred by deprotonation at the nitrogen to afford 3a and [Pt{(Ph(2)P)(2)C···C(···NCH(2)CH(2)O)}{Ph(2)PCH═COCH(2)CH(2)N(PPh(2))}]Cl ([Pt(1(-H)-P,P)(2'-P,P)]Cl (7)], respectively. In CH(2)Cl(2), an isomer of 3a, [Pt{Ph(2)P)(2)C···C(···NCH(2)CH(2)O)}{Ph(2)PC(PPh(2))═COCH(2)CH(2)N}] ([Pt(1(-H)-P,P)(1(-H)-P,N)] (8)), was obtained as a side product which contains ligand 1(-H) in two different coordination modes. Complexes 3b·4CH(2)Cl(2), 4·CHCl(3), 6·2.5CH(2)Cl(2), and 8·CH(2)Cl(2) have been structurally characterized by X-ray diffraction.  相似文献

The oxidation state of the chromium center in the following compounds has been probed using a combination of chromium K-edge X-ray absorption spectroscopy and density functional theory: [Cr(phen)(3)][PF(6)](2) (1), [Cr(phen)(3)][PF(6)](3) (2), [CrCl(2)((t)bpy)(2)] (3), [CrCl(2)(bpy)(2)]Cl(0.38)[PF(6)](0.62) (4), [Cr(TPP)(py)(2)] (5), [Cr((t)BuNC)(6)][PF(6)](2) (6), [CrCl(2)(dmpe)(2)] (7), and [Cr(Cp)(2)] (8), where phen is 1,10-phenanthroline, (t)bpy is 4,4'-di-tert-butyl-2,2'-bipyridine, and TPP(2-) is doubly deprotonated 5,10,15,20-tetraphenylporphyrin. The X-ray crystal structures of complexes 1, [Cr(phen)(3)][OTf](2) (1'), and 3 are reported. The X-ray absorption and computational data reveal that complexes 1-5 all contain a central Cr(III) ion (S(Cr) = (3)/(2)), whereas complexes 6-8 contain a central low-spin (S = 1) Cr(II) ion. Therefore, the electronic structures of 1-8 are best described as [Cr(III)(phen(?))(phen(0))(2)][PF(6)](2), [Cr(III)(phen(0))(3)][PF(6)](3), [Cr(III)Cl(2)((t)bpy(?))((t)bpy(0))], [Cr(III)Cl(2)(bpy(0))(2)]Cl(0.38)[PF(6)](0.62), [Cr(III)(TPP(3?-))(py)(2)], [Cr(II)((t)BuNC)(6)][PF(6)](2), [Cr(II)Cl(2)(dmpe)(2)], and [Cr(II)(Cp)(2)], respectively, where (L(0)) and (L(?))(-) (L = phen, (t)bpy, or bpy) are the diamagnetic neutral and one-electron-reduced radical monoanionic forms of L, and TPP(3?-) is the one-electron-reduced doublet form of diamagnetic TPP(2-). Following our previous results that have shown [Cr((t)bpy)(3)](2+) and [Cr(tpy)(2)](2+) (tpy = 2,2':6',2"-terpyridine) to contain a central Cr(III) ion, the current results further refine the scope of compounds that may be described as low-spin Cr(II) and reveal that this is a very rare oxidation state accessible only with ligands in the strong-field extreme of the spectrochemical series.  相似文献

The crystal structures of the series of four ternary complexes, [Pd(phen)(2,6-PDCA)].4H(2)O (1) (phen=1,10-phenanthroline; 2,6-PDCA=2,6-pyridinedicarboxylic acid), [Pd(bpy)(2,3-PDCA)].3H(2)O (2) (bpy=2,2'-bipyridineand; 2,3-PDCA=2,3-pyridinedicarboxylic acid) and [Pd(phen)(PHT)].2.5H(2)O (3) (PHT=o-phthalic acid ) and [Pd(bpy)(PHT)].1.5H(2)O (4), are determined and the coordination modes of palladium(II) ternary complexes are characterized. All complexes take the mononuclear Pd(II) complexes, in which central Pd(II) atom of each complex has a similar distorted square-planar four coordination geometry. In all complexes, the aromatic heterocyclic compounds, phen and bpy, behave as a bidentate N, N' ligand. In the complex 1 and 2, 2,6-PDCA and 2,3-PDCA behave as a bidentate N, O ligand, and in complex 3 and 4, PHT behaves as a bidentate O, O' ligand.  相似文献

Palladium and platinum complexes of the types cis-[M(C6F5)2(1-Mecyt)2] (M = Pd or Pt) and cis-[Pd(L')(C6F5)(1-Mecyt)2]ClO4 (L' = PPh(3) or t-BuNC) have also been prepared. The crystal structures of [Pd(bpzm)(C6F5)(1-Mecyt)]ClO4, [Pt(dmba)(DMSO)(1-Mecyt)]ClO4, cis-[Pd(C6F5)2(1-Mecyt)2], and cis-[Pd(t-BuNC)(C6F5)(1-Mecyt)2]ClO4 have been established by X-ray diffraction. There is extensive hydrogen bonding (N-H...O, C-H...F or C-H...O) in all the compounds. There are also intermolecular pi-pi interactions between pyrimidine rings of adjacent chains in [Pd(C6F5)2(1-Mecyt)2]. DNA adduct formation of the new complexes synthesized was followed by circular dichroism and electrophoretic mobility. Atomic force microscopy images of the modifications caused by the complexes on plasmid DNA pBR322 were also obtained. Values of IC(50) were also calculated for the new complexes against the tumor cell line HL-60. At a short incubation time (24 h) almost all new complexes were more active than cisplatin.  相似文献

The crystal structures of the series of three complexes, [Cu(Gly)(bpy)Cl].2H2O (1) (Gly=glycine; bpy=2,2'-bipyridine), [Cu(Gly)(phen)Cl]2.7H2O (2) (phen=1,10-phenanthroline), and [Cu(Gly)(bpa)(H2O)Cl] (3) (bpa=2,2'-bipyridylamine) were determined, and the coordination modes of Cu(II) ternary complexes were compared. The central Cu(II) atoms of complexes 1 and 3 have a similar distorted octahedral coordination geometry, while the Cu(II) atom of complex 2 has a distorted square pyramidal coordination. In all complexes, the aromatic heterocyclic compounds bpy, phen, and bpa, behave as a bidentate N,N' ligand, and Gly behaves as a bidentate N,O ligand. DNA-binding properties of the complexes to calf thymus (CT) DNA were studied by using the fluorescence method. Each of the complexes showed binding propensity to CT DNA with the relative order 2>3> or =1. DNA cleavage studies indicate that each of the complexes, especially 2, can cleave plasmid supercoiled pBR322 DNA in the presence of H2O2 and ascorbic acid with cleavage efficiency in the order 2>3 approximately 1. The degradation of the conformation of CT DNA by the complexes was also reflected in the decrease in the intensities of the characteristic CD bands with the relative order 2>3 approximately 1.  相似文献

Synthesis procedures are described for the new stable mixed ligand complexes, [Pd(Hpa)(pa)]Cl, [Pd(pa)(H2O)2]Cl, [Pd(pa)(en)]Cl, [Pd(pa)(bpy)]Cl, [Pd(pa)(phen)Cl], [Pd(pa)(pyq)Cl], cis-[MoO2(pa)2], [Ag(pa)(bpy)], [Ag(pa)(pyq)], trans-[UO2(pa)(pyq)](BPh4) and [ReO(PPh3)(pa)2]Cl (Hpa = 2-piperidine-carboxylic acid, en = ethylene diamine, bpy = 2,2′-bipyridyl, phen = 1,10-phenanthroline, pyq = 2(2′-pyridyl)quinoxaline). Their elemental analyses, conductance, thermal measurements, Raman, IR, electronic, 1H-n.m.r. and mass spectra have been measured and discussed. 2-Piperidine-carboxylic acid and its palladium complexes have been tested as growth inhibitors against Ehrlich ascites tumour cells (EAC) in Swiss albino mice.  相似文献

[Ru(bpy)(2)(Mebpy-COOH)](PF(6))(2).3H(2)O (1), [Ru(phen)(2)(Mebpy-COOH)](ClO(4))(2).5H(2)O (2), [Ru(dppz)(2)(Mebpy-COOH)]Cl(2).9H(2)O (3), and [Ru(bpy)(dppz)(Mebpy-COOH)](PF(6))(2).5H(2)O (4) (bpy = 2,2'-bipyridine, Mebpy-COOH = 4'-methyl-2,2'-bipyridine-4-carboxylic acid, phen = 1,10-phenanthroline, dppz = dipyrido[3,2,-a;2',3-c]phenazine) have been synthesized and characterized spectroscopically and by microanalysis. The [Ru(Mebpy-COOH)(CO)(2)Cl(2)].H(2)O intermediate was prepared by reaction of the monocarboxylic acid ligand, Mebpy-COOH, with [Ru(CO)(2)Cl(2)](n), and the product was then reacted with either bpy, phen, or dppz in the presence of an excess of trimethylamine-N-oxide (Me(3)NO), as the decarbonylation agent, to generate 1, 2, and 3, respectively. For compound 4, [Ru(bpy)(CO)Cl(2)](2) was reacted with Mebpy-COOH to yield [Ru(bpy)(Mebpy-COOH)(CO)Cl](PF(6)).H(2)O as a mixture of two main geometric isomers. Chemical decarbonylation in the presence of dppz gave 4 also as a mixture of two isomers. Electrochemical and spectrophotometric studies indicated that complexes 1 and 2 were present as a mixture of protonated and deprotonated forms in acetonitrile solution because of water of solvation in the isolated solid products. The X-ray crystal structure determination on crystals of [Ru(bpy)2(MebpyCOO)][Ru(bpy)(2)(MebpyCOOH)](3)(PF(6))(7), 1a, and [Ru(phen)(2)(MebpyCOO)](ClO(4)).6H(2)O, 2a, obtained from solutions of 1 and 2, respectively, revealed that 1a consisted of a mixture of protonated and deprotonated forms of the complex in a 1:3 ratio and that 2a consisted of the deprotonated derivative of 2. A distorted octahedral geometry for the Ru(II) centers was found for both complexes. Upon excitation at 450 nm, MeCN solutions of the protonated complexes 1-4 were found to exhibit emission bands in the 635-655 nm range, whereas the corresponding emission maxima of their deprotonated forms were observed at lower wavelengths. Protonation/deprotonation effects were also observed in the luminescence and electrochemical behavior of complexes 1-4. Comprehensive electrochemical studies in acetonitrile show that the ruthenium centers on 1, 2, 3, and 4 are oxidized from Ru(II) to Ru(III) with reversible potentials at 917, 929, 1052, and 1005 mV vs Fc(0/+) (Fc = ferrocene), respectively. Complexes 1 and 2 also exhibit an irreversible oxidation process in acetonitrile, and all compounds undergo ligand-based reduction processes.  相似文献

A series of mixed-ligand complexes [Cu(qui)(L)]NO(3)·xH(2)O (1-6), where Hqui = 2-phenyl-3-hydroxy-4(1H)-quinolinone, L = 2,2'-bipyridine (bpy) (1), 1,10-phenanthroline (phen) (2), bis(2-pyridyl)amine (ambpy) (3), 5-methyl-1,10-phenanthroline (mphen) (4), 5-nitro-1,10-phenanthroline (nphen) (5) and bathophenanthroline (bphen) (6), have been synthesized and fully characterized. The X-ray structures of [Cu(qui)(phen)]NO(3)·H(2)O (2) and [Cu(qui)(ambpy)]NO(3) (3a) show a slightly distorted square-planar geometry in the vicinity of the central copper(II) atom. An in vitro cytotoxicity study of the complexes found significant activity against human osteosarcoma (HOS) and human breast adenocarcinoma (MCF7) cell lines, with the best results for complex 6, where IC(50) equals to 2.1 ± 0.2 μM, and 2.2 ± 0.4 μM, respectively. The strong interactions of the complexes with calf thymus DNA (CT-DNA) and high ability to cleave pUC19 DNA plasmid were found. A correlation has been found between the in vitro cytotoxicity and DNA cleavage studies of the complexes.  相似文献

In search of potential anticancer drug candidates in ruthenium complexes, a series of mononuclear ruthenium complexes of the type [Ru(phen)(2)(nmit)]Cl(2) (Ru1), [Ru(bpy)(2)(nmit)]Cl(2) (Ru2), [Ru(phen)(2)(icpl)]Cl(2) (Ru3), Ru(bpy)(2)(icpl)]Cl(2) (Ru4) (phen=1,10-phenanthroline; bpy=2,2'-bipyridine; nmit=N-methyl-isatin-3-thiosemicarbazone, icpl=isatin-3-(4-Cl-phenyl)thiosemicarbazone) and [Ru(phen)(2)(aze)]Cl(2) (Ru5), [Ru(bpy)(2)(aze)]Cl(2) (Ru6) (aze=acetazolamide) and [Ru(phen)(2)(R-tsc)](ClO(4))(2) (R=methyl (Ru7), ethyl (Ru8), cyclohexyl (Ru9), 4-Cl-phenyl (10), 4-Br-phenyl (Ru11), and 4-EtO-phenyl (Ru12), tsc=thiosemicarbazone) were prepared and characterized by elemental analysis, FTIR, (1)H-NMR and FAB-MS. Effect of these complexes on the growth of a transplantable murine tumor cell line (Ehrlich Ascites Carcinoma) and their antibacterial activity were studied. In cancer study the effect of hematological profile of the tumor hosts have also been studied. In the cancer study, the complexes Ru1-Ru4, Ru10 and Ru11 have remarkably decreased the tumor volume and viable ascitic cell count as indicated by trypan blue dye exclusion test (p<0.05). Treatment with the ruthenium complexes prolonged the lifespan of Ehrlich Ascites Carcinoma (EAC) bearing mice. Tumor inhibition by the ruthenium chelates was followed by improvements in hemoglobin, RBC and WBC values. All the complexes showed antibacterial activity, except Ru5 and Ru6. Thus, the results suggest that these ruthenium complexes have significant antitumor property and antibacterial activity. The results also reflect that the drug does not adversely affect the hematological profiles as compared to that of cisplatin on the host.  相似文献

From the reaction mixture of [M(II)(bpy)Cl(2)], the ligand 2-anilino-4,6-di-tert-butylphenol, H[L(AP)], and 2 equiv of a base (NaOCH(3)) in CH(3)CN under anaerobic conditions were obtained the blue-green neutral complexes [M(II)(L(AP)-H)(bpy)] (M = Pd (1), Pt (2)). (L(AP)-H)(2)(-) represents the o-amidophenolato dianion, (L(AP))(1)(-) is the o-aminophenolate(1-), (L(ISQ))(1)(-) is its one-electron-oxidized, pi-radical o-iminobenzosemiquinonate(1-), and (L(IBQ))(0) is the neutral quinone. Complexes 1 and 2 can be oxidized by ferrocenium hexafluorophosphate, yielding the paramagnetic salts [M(II)(L(ISQ))(bpy)]PF(6) (S = (1)/(2)) (M = Pd (1a), Pt (2a)). The reaction of PtCl(2), 2 equiv of H[L(AP)], and 4 equiv of base in CH(3)CN in the presence of air yields diamagnetic [Pt(L(ISQ))(2)] (3), which is shown to possess an electronic structure that is best described as a singlet diradical. Complexes 1, 1a, 2, 2a, and 3 have been structurally characterized by X-ray crystallography at 100 K. It is clearly established that O,N-coordinated (L(AP)-H)(2)(-) ligands have a distinctly different structure than the corresponding O,N-coordinated (L(ISQ))(1)(-) radicals. It is therefore possible to unambiguously assign the protonation and oxidation level of o-aminophenol derived ligands in coordination compounds. All complexes have been investigated by cyclic voltammetry, spectroelectrochemistry, EPR, and UV-vis spectroscopy. Complexes 1 and 2 can be reversibly oxidized to the [M(II)(L(ISQ))(bpy)](+) and [M(II)(L(IBQ))(pby)](2+) mono- and dications, respectively, and reduced to the [M(L(AP)-H)(bpy(*))](-) anion, where (bpy(*))(1)(-) is the radical anion of 2,2'-bipyridine. Complex 3 exhibits four reversible one-electron-transfer waves (two oxidations and two reductions) which are all shown to be ligand centered. The EPR spectra of the one-electron-reduced species [Pt(L(AP)-H)(L(ISQ))](-) (S = (1)/(2)) and of the one-electron-oxidized species [Pt(L(ISQ))(L(IBQ))](+) (S = (1)/(2)) in CH(2)Cl(2) solutions have been recorded. To gain a better understanding of the electronic structure of 3 and its monooxidized and reduced forms, relativistic DFT calculations have been carried out. Magnetic coupling parameters and hyperfine couplings were calculated and found to be in very good agreement with experiment. It is shown that both the one-electron oxidation and reduction of 3 are ligand centered. A simple MO model is developed in order to understand the EPR properties of the monocation and monoanion of 3.  相似文献

The reactions of bidentate diimine ligands (L2) with binuclear [Ru(L1)(CO)Cl2]2 complexes [L1 not equal to L2 = 2,2'-bipyridine (bpy), 4,4'-dimethyl-2,2'-bipyridine (4,4'-Me2bpy), 5,5'-dimethyl-2,2'-bipyridine (5,5'-Me2bpy), 1,10-phenanthroline (phen), 4,7-dimethyl-1,10-phenanthroline (4,7-Me2phen), 5,6-dimethyl-1,10-phenanthroline (5,6-Me2phen), di(2-pyridyl)ketone (dpk), di(2-pyridyl)amine (dpa)] result in cleavage of the dichloride bridge and the formation of cationic [Ru(L1)(L2)(CO)Cl]+ complexes. In addition to spectroscopic characterization, the structures of the [Ru(bpy)(phen)(CO)Cl]+, [Ru(4,4'-Me2bpy)(5,6-Me2phen)(CO)Cl]+ (as two polymorphs), [Ru(4,4'-Me2bpy)(4,7-Me2phen)(CO)Cl]+, [Ru(bpy)(dpa)(CO)Cl]+, [Ru(5,5'-Me2bpy)(dpa)(CO)Cl]+, [Ru(bpy)(dpk)(CO)Cl]+, and [Ru(4,4'-Me2bpy)(dpk)(CO)Cl]+ cations were confirmed by single crystal X-ray diffraction studies. In each case, the structurally characterized complex had the carbonyl ligand trans to a nitrogen from the incoming diimine ligand, these complexes corresponding to the main isomers isolated from the reaction mixtures. The synthesis of [Ru(4,4'-Me2bpy)(5,6-Me2bpy)(CO)(NO3)]+ from [Ru(4,4'-Me2bpy)(5,6-Me2bpy)(CO)Cl]+ and AgNO3 demonstrates that exchange of the chloro ligand can be achieved.  相似文献
