Saikosaponins are triterpene saponins derived from the roots of Bupleurum falcatum L. (Umbelliferae), which has been traditionally used to treat fever, inflammation, liver diseases, and nephritis. It is difficult to analyze saikosaponins using HPLC-UV due to the lack of chromophores. Therefore, evaporative light scattering detection (ELSD) is used as a valuable alternative to UV detection. More recently, a charged aerosol detection (CAD) method has been developed to improve the sensitivity and reproducibility of ELSD. In this study, we compared CAD and ELSD methods in the simultaneous analysis of 10 saikosaponins, including saikosaponins-A, -B1, -B2, -B3, -B4, -C, -D, -G, -H and -I. A mixture of the 10 saikosaponins was injected into the Ascentis® Express C18 column (100 mm × 4.6 mm, 2.7 μm) with gradient elution and detection with CAD and ELSD by splitting. We examined various factors that could affect the sensitivity of the detectors including various concentrations of additives, pH and flow rate of the mobile phase, purity of nitrogen gas and the CAD range. The sensitivity was determined based on the signal-to-noise ratio. The best sensitivity for CAD was achieved with 0.1 mM ammonium acetate at pH 4.0 in the mobile phase with a flow rate of 1.0 mL/min, and the CAD range at 100 pA, whereas that for ELSD was achieved with 0.01% acetic acid in the mobile phase with a flow rate at 0.8 mL/min. The purity of the nitrogen gas had only minor effects on the sensitivities of both detectors. Finally, the sensitivity for CAD was two to six times better than that of ELSD. Taken together, these results suggest that CAD provides a more sensitive analysis of the 10 saikosaponins than does ELSD.  相似文献

The paper presents a new HPLC method, with UV and MSn detection, for the determination of seven pesticides, including the sulfonylurea herbicides amidosulfuron, azimsulfuron, nicosulfuron, rimsulfuron, thifensulfuron methyl, tribenuron methyl, and the fungicide azoxystrobin characterised by a methoxyacrilate structure. The methodology consists of a preconcentration/SPE (solid phase extraction) step and HPLC-UV (240 nm detection wavelength)-MSn analysis. Under the optimised conditions and after a 1000/1 preconcentration factor, the limits of detection were lower than 14.5 ng L−1 for UV detection and lower than 8.1 ng L−1 for MS detection. The limits of quantification were lower than 48.3 ng L−1 in UV detection and than 26.9 ng L−1 in MSn detection. The analysis of two samples, spiked with a mixture of the pesticides at threshold level concentrations, gave more than 60% recovery.  相似文献

Based on TiO2-nanoparticles coating fabricated by a one-step anodization method on titanium wire substrate, a novel phenyl functionalized solid-phase microextraction (SPME) fiber coating was prepared by simple and rapid in situ chemical assembling technique between the fiber surface titanol groups and trichlorophenylsilane reaction. The as-fabricated fiber exhibited good extraction capability for some UV filters and was employed to determine the ultraviolet (UV) filters in combination with high performance liquid chromatography–UV detection (HPLC–UV). The main parameters affecting extraction performance were investigated and optimized. Under the optimized conditions, the developed method was applied to detect several UV filters at trace concentration levels with only 8 mL of sample volume. They were determined in the range from 0.005 to 25 μg L−1 with detection limits (S/N = 3) from 0.1 to 50 ng L−1. The relative standard deviations (RSDs) for single fiber repeatability varied from 4.6 to 6.5% (n = 5) and fiber-to-fiber reproducibility (n = 5) ranged from 5.5 to 9.1%. The linear ranges spanned two-four magnitudes with correlation coefficients above 0.9990. Five real water samples including four Yellow River water samples and one rain water sample were determined sensitively with good recoveries ranging from 86.2 to 105.5%. The functionalized fiber coating performed good reproducible manner, high mechanical strength, good stability and long service life. Moreover, this study proposed an efficient sample pretreatment method for the determination of UV filters from environmental water samples.  相似文献

A sensitive and precise high performance liquid chromatography (HPLC)-electrochemical detection (ECD) method has been developed for the simultaneous determination of four isoquinoline alkaloids including berberine, jatrorrhizine, coptisine and palmatine in Chinese medicine Coptis chinensis. The typical HPLC analysis was performed on WondaSil® C18-WR column (250 × 4.6 mm, 5 μm) with the mobile phase comprising 40 mM phosphate buffer (pH 7.0)–acetonitrile (40:60, v/v) at the flow rate of 0.8 mL min−1. The electrochemical detection employed a three electrode system with a bare glassy carbon electrode at +1.3 V versus the Ag/AgCl reference electrode. The limits of detection (LODs) of four alkaloids ranged from 0.01 to 0.03 μmol L−1 and the LOD of berberine was 80 times lower than LOD obtained by UV detection. The rat plasma samples were assayed after oral administration of the traditional Chinese medicine Coptis chinensis by the proposed HPLC-ECD method. The recoveries of this method were ranging from 88.0 to 116%, with the relative standard deviation lower than 3.1% for intra-day precision and 5.7% for inter-day precision. These results show that HPLC-ECD is a useful tool for the quality control of herbal medicine Coptis chinensis and also for pharmacokinetic studies.  相似文献

A capillary zone electrophoresis (CZE) method for analyzing 11 chelating agents [β-alaninediacetic acid (β-ADA), trans-1,2-diaminocyclohexane-N,N,N′,N′-tetraacetic acid (CDTA), diethylenetriaminepentaacetic acid (DTPA), ethylenediaminetetraacetic acid (EDTA), N-(2-hydroxyethyl)ethylenediamine-N,N′,N′-triacetic acid (HEDTA), N-(2-hydroxyethyl)iminodiacetic acid (HEIDA), iminodiacetic acid (IDA), methylglycinediacetic acid (MGDA), nitrilotriacetic acid (NTA), 1,3-diaminopropane-N,N,N′,N′-tetraacetic acid (PDTA) and triethylenetetraaminehexaacetic acid (TTHA)] as negatively charged copper(II) complexes has been established. Both conventional and alternative chelating agents were included in this study, because they are used side by side in industrial applications. In this study, didecyldimethylammonium bromide (DMDDAB) was successfully used as a flow reversal reagent for the first time in an aqueous CZE method based on phosphate BGE with UV spectrophotometric detection. In addition this new flow modifier was compared to common TTAB. Method development was done using a fused silica capillary (61 cm × 50 μm i.d.). The optimized BGE was a 105 mmol L−1 phosphate buffer with TTAB or DMDDAB in the concentration 0.5 mmol L−1 at pH 7.1. The measurements were done with −20 kV voltage using direct UV detection at 254 nm. In both CZE methods all 11 analyte zones were properly separated (resolutions ≥2.4), and the calibrations gave excellent correlation coefficients (≥0.998; linear range tested 0.5-2.0 mmol L−1). The limits of detection were ≤34 and ≤49 μmol L−1 with the method of DMDDAB and TTAB, respectively. A clear benefit of both methods was the short analysis time; all 11 complexes were detected in less than 6 and 5.5 min with the methods of TTAB and DMDDAB, respectively. The two methods were tested with dishwashing detergents and paper mill wastewater samples and proved to be suitable for practical use.  相似文献

A method for the determination of mercury via UV photoreduction has been investigated. Mercury vapor was generated by the reduction of mercury species in an acetic acid solution using UV radiation. Detection of the volatile mercury was accomplished by atomic absorption spectrometry. An optimized system was found to provide a detection limit (defined as the concentration giving a signal equal to three times the standard deviation of the blank) of 2.1 μg L−1 with a precision of 2.9% relative standard deviation (n = 8) for a 500 μg L−1 mercury standard. The effect of various metal ions on the mercury signal was investigated and the method validated with a NRCC certified dogfish liver material (DOLT-3) using the method of standard additions. A reaction pathway is hypothesized for UV photoreduction.  相似文献

In this work, a capillary electrophoresis (CE) procedure was developed for the simultaneous determination of a pharmaceutical drug and its counter-ion, namely labetalol hydrochloride. For this purpose, an uncoated fused-silica capillary, a low conductivity background electrolyte (BGE) and a capacitively coupled contactless conductivity detector (C4D) were employed. This detection system is highly sensitive and enables detection of inorganic as well as organic ions unlike with direct UV detection. Moreover, to be able to simultaneously analyze the cationic drug (labetalol+) and its anionic counter-ion (Cl) in the same electrophoretic run without the need of a coated capillary, a dual-opposite end injection was performed. In this technique, the sample is hydrodynamically injected into both ends of the capillary. This method is simple and easy to perform since the different injection steps are automated by the CE software.This novel CE-C4D procedure with dual-opposite end injection has been successfully validated and applied for the analysis of chloride content in an adrenergic antagonist (labetalol hydrochloride). Thus, the hereby developed method has been shown to enable fast (analysis time < 10 min), precise (repeatability of migration times < 0.7% and of corrected-peak areas < 3.3%; n = 6) and rugged analyses for the simultaneous determination of a pharmaceutical drug and its counter-ion.  相似文献

A novel method that combines two concentration techniques, off-line centrifuge microextraction (CME) and on-line sweeping, is used to determine trace steroids in urine by micellar electrokinetic chromatography (MEKC). The CME-sweeping-MEKC is a promising technique, which has a variety of merits such as high sensitivity, rapid operation, minimal cost, and high sample throughput capability. Using CME-sweeping, over a 500-fold increase in sensitivity could be obtained as compared with the normal hydrodynamic injection without sample stacking. The linear range was 0.05-1 μg mL−1 with the square of the correlation coefficients ranging from 0.9998 to 0.9999. Detection limits (S/N = 3) were 5-15 ng mL−1 using a photodiode array UV detection at a wavelength of 240 nm. The relative standard deviations were between 2.8% and 3.5% (n = 5, progesterone as the internal standard). The diode array UV spectrum used can distinguish the analytes from the interference in the complex sample matrix, which is useful in biological and clinical sample analysis.  相似文献

A novel method has been developed for simultaneous analysis for three pairs of trans- and cis-isomers of 2-glucosyloxycinnamic acids, along with their biogenic metabolites (three coumarin derivatives including scopoletin, scoparone and ayapin) in a Chinese medicinal herb Dendrobium thyrsiflorum by using high-performance liquid chromatography (HPLC)-photodiode array detection (DAD)-electrospray ionization (ESI)-tandem mass spectrometry (MS). The method was carried out by using a Polaris C18 column with a gradient solvent system of 0.5% acetic acid aqueous solution-acetonitrile. Seven target analytes including isodensifloside, isothyrsifloside, densifloside, thyrsifloside, scoparone, ayapin and scopoletin were exclusively identified by comparing their retention behaviors, UV and MS spectra with the authentic standards, and their contents in D. thyrsiflorum were simultaneous determined by employing UV detection at 342 nm. In addition, another pair of isomers of 2-glucosyloxycinnamic acids was putatively elucidated mainly based on the MS fragmentation. The method was validated and found to be satisfactorily linear, selective and robust. Recoveries ranged from 95.56 to 97.94% for all compounds at three different spiking levels. The limits of detection (LOD) and quantitation (LOQ) ranged, respectively, from 0.02 to 0.13 μg mL−1 and 0.07 to 0.39 μg mL−1 depending on various compounds. The established quality evaluation method was successfully used for evaluating the quality of D. thyrsiflorum samples of different organs and collections.  相似文献

Laser-excited fluorescence was used for the selective determination of camptothecin in samples containing anti-cancer camptothecin-analogs (irinotecan and topotecan). The selectivity of the method was based on the UV photochemical derivatization in basic solution which increased the analyte fluorescence (337/450 nm) and eliminated fluorescence from the two campthotecin-analogs. The influence of UV exposure time and sodium hydroxide concentration was studied using an experimental design. Limit of detection was 4 × 10−10 mol L−1 with linear fluorescence response up to 1 × 10−6 mol L−1. Average recoveries of camptothecin (added to the samples to simulate a contamination) were 92 ± 4 and 94 ± 6% (n = 3) respectively in irinotecan and topotecan based pharmaceuticals.  相似文献

A novel high-performance liquid chromatographic (HPLC) method using pre-column derivatization and UV detection at 275 nm for the determination of clarithromycin in rat plasma has been validated. Clarithromycin was extracted from plasma sample spiked with internal standard (erythromycin) under alkaline condition with ethyl ether and derivatizated with trimethylbromosilane. The analyses were run on a C18 column, maintained at 40 °C during elution, using a mobile phase comprised of potassium dihydrogen phosphate (50 mM, pH 6.8, contained 0.7% triethylamine), acetonitrile, and methanol (30:45:25, v/v/v). The standard calibration curve for clarithromycin was linear (r2 = 0.9998) over the concentration range of 0.1-10 μg ml−1 in rat plasma. The limit of detection (LOD) and limit of quantitation (LOQ) was 30 ng ml−1 and 0.1 μg ml−1 respectively. The intra- and inter-day assay variability range was 2.6-7.4% and 3.3-8.5%, respectively. This method has been successfully applied to a pharmacokinetic study of clarithromycin in rats.  相似文献

The method for simultaneous separation and determination of trace monoadenosine and diadenosine monophosphate (i.e. 2′-AMP, 3′-AMP, 5′-AMP and 3′-5′ ApA) in biomimicking prebiotic synthesis was developed using high-performance liquid chromatography (HPLC) with ultraviolet (UV) detection and electrospray ionization mass spectrometry (ESI-MS) identification. The separation was performed on a Supelco C18 column with a gradient elution (solvent A: 10 mM NH4Ac aqueous solution; solvent B: MeOH). The flow rate was set at 1.0 ml/min. The quantitative determination was achieved by HPLC with UV detection at 260 nm. The linearity ranged from 0.5 to 100 μg/ml for each nucleotide. The limits of detection (LODs) for the four nucleotides were less than 0.30 μg/ml. The recovery ranged from 95.2 to 100.7%. The intra-day relative standard deviations (RSDs) of the retention times were between 0.7 and 1.1%. Both full-scan ESI-MS and -MS2 for the four nucleotides under both positive and negative polarity were carried out and the possible cleavage pathways of them were depicted. The specific ions, [AMP + H]+ at m/z 348 and [ApA + H]+ at m/z 597, were chosen to characterize the four nucleotides in biomimicking prebiotic synthesis between N-(O,O-diisopropyl) phosphoryl amino acid (Dipp-aa) and adenosine. Using the proposed HPLC/UV/ESI-MS method, the concentration of 2′-AMP, 3′-AMP, 5′-AMP and 3′-5′ ApA in the biomimicking prebiotic synthesis samples were determined.  相似文献

The high-performance liquid chromatography (HPLC) coupled with on-line solid phase extraction (SPE) and ultraviolet (UV) detection was developed for determining andrographolide and dehydroandrographolide in rabbit plasma. Plasma samples (100 μL) were injected directly into a C18 SPE column and the biological matrix was washed out for 6 min using 15% aqueous methanol. By rotation of the switching valve, andrographolide and dehydroandrographolide were eluted in the back-flush mode and transferred to the analytical column by the chromatographic mobile phase consisted of methanol:acetonitrile (ACN):water (50:10:40; v/v). The UV detection was performed at 225 nm. The calibration curves showed excellent linear relationship (R ≥ 0.9993) over the concentration range of 0.05-5.0 μg mL−1. The within- and between-day precisions (R.S.D.) of two analytes were in the range of 1.2-6.5% and the accuracies were between 92.0% and 102.1%. Their recoveries were all greater than 94%. The limits of detection were 0.019 μg mL−1 for andrographolide and 0.022 μg mL−1 for dehydroandrographolide. This method was successfully applied to the plasma concentration-time curve study after oral administration of Andrographis paniculata Nees extract in rabbit.  相似文献

The simple, reliable and reproducible HPLC and extraction methods were developed for the analysis of tadalafil in pharmaceutical preparation. The column used was monolithic silica column, Chromolith Performance RP-18e (100 mm × 4.6 mm, i.d.). The mobile phase used was phosphate buffer (100 mM, pH 3.0)-acetonitrile (80:20, v/v) at the flow rate of 5 mL min−1 with UV detection at 230 nm at ambient temperature. Extraction of tadalafil from tablet was carried out using methanol. Linearity was observed in the concentration range from 100 to 5000 ng mL−1 for tadalafil with a correlation coefficient (R2) 0.9999 and 100 ng mL−1 as the limit of detection. The values of linearity range, correlation coefficient (R2) and limit of detection were 50-5000 ng mL−1, 0.9999-50 ng mL−1, respectively for sildenafil. Parameters of validation prove the precision of the method and its applicability for the determination of tadalafil in pharmaceutical tablet formulation. The method is suitable for high throughput analysis of the drug.  相似文献

According to the current demands of environmentally friendly analytical chemistry and with a view to achieving lower reagent consumption with improved analytical performance, an automatic methodology composed of a photoreactor and fluorimetric detection (λexc = 287 nm, λem = 378 nm) was developed. To this end, a sequential injection analysis (SIA) system was developed for indomethacin determination using ultra-violet (UV) light which promotes an increase in the fluorescence of indomethacin. This increase in sensitivity makes it possible to apply this methodology to a dissolution test and to determine indomethacin in pharmaceutical formulations.The calibration graph for indomethacin was linear between 4.10 × 10−6 and 9.00 × 10−5 mol L−1and the detection limit was 1.23 × 10−6 mol L−1. The method was proven to be reproducible with a R.S.D. < 5% and sampling rate of approximately 20 per hour. The potential effect of several compounds commonly used as excipients on analytical signals was studied and no interfering effect was observed. Statistical evaluation at the 95% confidence level showed good agreement between the results obtained for the pharmaceutical samples with both the SIA system and comparison batch procedures.  相似文献

A novel simple isocratic HPLC method with UV detection for the determination of three compounds in spray solution (active component clotrimazole and two degradation products imidazole and (2-chlorophenyl)diphenylmethanol) using ibuprofen as an internal standard was developed and validated. The complications with different acido-basic properties of the analysed compounds in HPLC separation - while clotrimazole has pKa 4.7, imidazole has pKa 6.9 compared to relatively more acidic (2-chlorophenyl)diphenylmethanol - were finally overcome using a 3.5 μm Zorbax® SB-Phenyl column (75 mm × 4.6 mm i.d., Agilent Technologies).The optimal mobile phase for separation of clotrimazole, degradation products imidazole and (2-chlorophenyl)diphenylmethanol and ibuprofen as internal standard consists of a mixture of acetonitrile and water (65:35, v/v) with pH* conditioned by phosphoric acid to 3.5. At a flow rate of 0.5 ml min−1 and detection at 210 nm, the total time of analysis was less than 6 min.The method was applied for routine analysis (batch analysis and stability tests) in commercial spray solution.  相似文献

The new method presented in this article achieved the goal of capturing Salmonella typhimurium via immunoreaction and rapid in situ detection of the CdSe/ZnS quantum dots (QDs) labeled S. typhimurium by self-assembly light-emitting diode-induced fluorescence detection (LIF) microsystem on a specially designed multichannel microfluidic chip. CdSe/ZnS QDs were used as fluorescent markers improving detection sensitivity. The microfluidic chip developed in this study was composed of 12 sample channels, 3 mixing zones, and 6 immune reaction zones, which also acted as fluorescence detection zones. QDs–IgG–primary antibody complexes were generated by mixing CdSe/ZnS QDs conjugated secondary antibody (QDs–IgG) and S. typhimurium antibody (primary antibody) in mixing zones. Then, the complexes went into immune reaction zones to label previously captured S. typhimurium in the sandwich mode. The capture rate of S. typhimurium in each detection zone was up to 70%. The enriched QDs-labeled S. typhimurium was detected using a self-assembly LIF microsystem. A good linear relationship was obtained in the range from 3.7 × 10 to 3.7 × 105 cfu mL−1 using the equation I = 0.1739 log (C) − 0.1889 with R2 = 0.9907, and the detection limit was down to 37 cfu mL−1. The proposed method of online immunolabeling with QDs for in situ fluorescence detection on the designed multichannel microfluidic chip had been successfully used to detect S. typhimurium in pork sample, and it has shown potential advantages in practice.  相似文献

The aim of this study was to develop a fast capillary electrophoresis method for the determination of propranolol in pharmaceutical preparations. In the method development the pH and constituents of the background electrolyte were selected using the effective mobility versus pH curves. Benzylamine was used as the internal standard. The background electrolyte was composed of 60 mmol L−1 tris(hydroxymethyl)aminomethane and 30 mmol L−1 2-hydroxyisobutyric acid, at pH 8.1. Separation was conducted in a fused-silica capillary (32 cm total length and 8.5 cm effective length, 50 μm I.D.) with a short-end injection configuration and direct UV detection at 214 nm. The run time was only 14 s. Three different strategies were studied in order to develop a fast CE method with low total analysis time for propranolol analysis: low flush time (Lflush) 35 runs/h, without flush (Wflush) 52 runs/h, and Invert (switched polarity) 45 runs/h. Since the three strategies developed are statistically equivalent, Wflush was selected due to the higher analytical frequency in comparison with the other methods. A few figures of merit of the proposed method include: good linearity (R2 > 0.9999); limit of detection of 0.5 mg L−1; inter-day precision better than 1.03% (n = 9) and recovery in the range of 95.1–104.5%.  相似文献

A new method for sensitive and fast screening of melamine (MEL) in milk products was developed with a low-cost disposable microfluidic device coupled with ultraviolet (UV) detection. This method avoided the need of sample pretreatment prior to the separation process, thus was simple and green. Due to the advantages of the device and fracture sampling technique, milk sample could be directly sampled through the fracture to achieve baseline separation from amino acids, and proteins in the sample did not interfere with the detection. Using 20 mM phosphate running buffer (pH 9.0), a sampling time of 3 s at +180 V and a separation voltage of +1800 V (240 V/cm), this method could detect MEL in milk within 75 s. At the detection wavelength of 202 nm, the linear range for MEL was from 1.0 to 100 μg mL−1 with a detection limit of 0.23 μg mL−1 (S/N = 3). The novel protocol had been successfully used to screen MEL in milk samples with recovery more than 82%. The environmentally friendly methodology for pretreatment-free sensitive screening of MEL provided promising applications in monitoring the quality of different foods.  相似文献

In the present study, a capillary electrophoresis method using high concentration of amylose solutions as separation medium has been developed with the aid of dimethyl sulfoxide (DMSO) as co-solvent. The best buffer conditions for primaquine, trihexyphenidyl (THP), sulconazole and cetirizine enantiomers were optimized as 20 mM sodium phosphate buffer with DMSO/water (40/60, v/v) as solvent at a pH of 3.0, containing 10% (w/v) amylose. Partial-filling and semi-permanent coating techniques were used considering the influences of DMSO on UV detection. High chiral resolution for THP enantiomers was obtained showing good chiral separation capacity of this method. The method showed good linearity (R2 > 0.998) over the concentration range of 0.50 and 2.00 mg L−1 for all the enantiomers. The detection limits for the tested enantiomers were in the range from 0.05 to 0.12 mg L−1. The linear calibration models were proven to be adequate for the experimental data by lack-of-fit test. The intra-assay precision, inter-day precision and accuracy were all evaluated to be acceptable. Separation and determination of THP enantiomers in rabbit blood were also carried out.  相似文献
