While magnetic bead (MB)-based bioassays have been implemented in integrated devices, their handling on-chip is normally either not optimal—i.e. only trapping is achieved, with aggregation of the beads—or requires complex actuator systems. Herein, we describe a simple and low-cost magnetic actuator to trap and move MBs within a microfluidic chamber in order to enhance the mixing of a MB-based reaction. The magnetic actuator consists of a CD-shaped plastic unit with an arrangement of embedded magnets which, when rotating, generate the mixing. The magnetic actuator has been used to enhance the amplification reaction of an enzyme-linked fluorescence immunoassay to detect Escherichia coli O157:H7 whole cells, an enterohemorrhagic strain, which have caused several outbreaks in food and water samples. A 2.7-fold sensitivity enhancement was attained with a detection limit of 603?colony-forming units (CFU) /mL, when employing the magnetic actuator. Graphical Abstract

Colloidal crystal beads (CCBs) were fabricated by assembling monodisperse silica nanoparticles via a microfluidic device. The pore size of the CCBs was tuned by using different nanoparticles. The CCBs were then coated with cadmium telluride quantum dots and zinc(II) meso-tetraphenylporphyrin for the purpose of optical sensing. Ammonia causes the color of the sensor to change from green to red. The method has a dynamic range of 0–2500 ppm, good reversibility, and is not sensitive to humidity. The limit of detection is 7 ppm. The sensor has the advantage of a porous microcarrier structure and that pore sizes can be well controlled and thus can fulfill various demands in gas detection.

We explore the feasibility of conducting electron ionization (EI) in a radio-frequency (rf) ion source trap for mass spectrometry applications. Electrons are radially injected into a compact linear ion trap in the presence of a magnetic field used essentially to lengthen the path of the electrons in the trap. The device can either be used as a stand-alone mass spectrometer or can be coupled to a mass analyzer. The applied parallel magnetic field and the oscillating rf electric field produced by the trap give rise to a set of coupled Mathieu equations of motion. Via numerical simulations, electron trajectories are studied under varying intensities of the magnetic field in order to determine the conditions that enhance ion production. Likewise, the dynamic behavior of the ions are investigated in the proposed EI source trap and the fast Fourier transform FFT formalism is used to obtain the frequency spectrum from the numerical simulations to study the motional frequencies of the ions which include combinations of the low-frequency secular and the high-frequency micromotion with magnetron and cyclotron frequencies. The dependence of these motional frequencies on the trapping conditions is examined and particularly, the limits of applying a radial magnetic field to the EI ion trap are characterized.

A sensitive and selective electrochemical sensor for the determination of nitrobenzene (NB) was developed based on a carbon paste electrode (CPE) modified with a nanocomposite prepared from the conducting polymer poly(3,4-ethylenedioxythiophene) and carbon nanotubes. The modified CPE exhibits good conductivity, a large surface area, and excellent catalytic activity towards the electrochemical reduction of NB. Under optimal conditions, the modified CPE is capable of detecting NB in the 0.25 to 43 μM concentration range and with a detection limit at 83 nM. Moreover, the sensor is highly stable and reusable, and free of interferences by other commonly present nitro compounds. It was used to determine NB in wastewater samples.

We describe a fluorogenic assay for activated protein C (APC) by using magnetic beads modified with DNA aptamers, taking advantage of strong binding affinity of aptamer, facile magnetic separation, and signal amplification via an enzymatic reaction. APC is specifically captured from a sample by the DNA aptamers on magnetic beads, and the concentrated APC then catalyzes the conversion of a fluorogenic substrate of APC to a fluorescent product. Detection of APC is achieved by measuring the generated product. This method is simple, sensitive, and specific. APC can be detected at 0.4 pM concentration level in a sample volume of 250 μL, corresponding to 0.1 femtomole of APC, when 2-h enzymatic reaction is employed. The proteins thrombin, trypsin, proteinase K, chymotrypsin, and elastase do not interfere.

We have prepared a new voltammetric sensor for guanine and adenine. It is based on a glassy carbon electrode modified with a Langmuir-Blodgett film made from tetraoxocalix[2]arene[2]triazine. The direct electro-oxidation of adenine and guanine was investigated and the results indicat that in contrast to a bare glassy carbon electrode both guanine and adenine cause an increase in the oxidation peak currents along with a negative shift of the oxidation potentials. The electrode enables the simultaneous determination of guanine and adenine using square wave voltammetry. Analysis of acid denatured calf thymus DNA was carried out and the value of (G + C)/(A + T) was correctly found to be 0.75.

We report on the first application of terahertz metamaterials acting as transducers for chemical sensors based on conducting polymers. In our feasibility study aimed at sensing of gaseous hydrochloric and ammonia, a two-dimensional sensor metamaterial consisting of an array of split-ring resonators on the surface of undoped silicon wafer was prepared. The surface of the resonator was coated with a 150-μm layer of polyaniline. Binding of hydrogen chloride to polyaniline leads to distinct changes in the resonance frequency of the metamaterial. Measurements can be performed both in the reflection and transmission mode. A numerical simulation of the response revealed an increase of both the real and the imaginary components of the dielectric function of the polyaniline film. These changes are attributed to the transition from emaraldine base to emeraldine salt. The results demonstrate a new approach for formation of highly sensitive transducers for chemical sensors.

A novel quartz crystal microbalance (QCM) sensor has been developed for highly selective and sensitive detection of Pb2+ by exploiting the catalytic effect of Pb2+ ions on the leaching of gold nanoparticles from the surface of a QCM sensor. The use of self-assembled gold nanoparticles (AuNPs) strongly enlarges the size of the interface and thus amplifies the analytical response resulting from the loss of mass. This results in a very low detection limit for Pb2+ (30 nM). The high selectivity is demonstrated by studying the effect of potentially interfering ions both in the absence and presence of Pb2+ ions. This simple and well reproducible sensor was applied to the determination of lead in the spiked drinking water. This work provides a novel strategy for fabricating QCM sensors towards Pb2+ in real samples. Figure

We describe a highly sensitive and selective amperometric sensor for the determination of nitrite. A glassy carbon electrode was modified with a composite made from gold nanoparticles (AuNPs) and sulfonated graphene (SG). The modified electrode displays excellent electrocatalytic activity in terms of nitrite oxidation by giving much higher peak currents (at even lower oxidation overpotential) than those found for the bare electrode, the AuNPs-modified electrode, and the SG-modified electrode. The sensor has a linear response in the 10 μM to 3.96 mM concentration range, a very good detection sensitivity (45.44 μA mM?1), and a lower detection limit of 0.2 μM of nitrite. Most common ions and many environmental organic pollutants do not interfere. The sensor was successfully applied to the determination of nitrite in water samples, and the results were found to be consistent with the values obtained by spectrophotometry.

Graphene was prepared by electrochemical reduction of exfoliated graphite oxide at cathodic potentials, and used to fabricate a graphene-modified glassy carbon electrode (GCE) which was applied in a sensor for highly sensitive and selective voltammetric determination of hydroquinone (HQ). Compared to a bare (conventional) GCE, the redox peak current for HQ in pH 5.7 acetate buffer solution is significantly increased, indicating that graphene possesses electrocatalytic activity towards HQ. In addition, the peak-to-peak separation is significantly improved. The modified electrode enables sensing of HQ without interference by catechol or resorcinol. Under optimal conditions, the sensor exhibits excellent performance for detecting HQ with a detection limit of 0.8?μM, a reproducibility of 2.5% (expressed as the RSD), and a recoveries from 98.4 to 101.2%.

In this paper, we present an electrochemical DNA–protein interaction assay based on a combination of protein-specific immunoprecipitation at magnetic beads (MBIP) with application of oligonucleotide (ON) probes labeled with an electroactive oxoosmium complex (Os,bipy). We show that double-stranded ONs bearing a dT20 tail labeled with Os,bipy are specifically recognized by the tumor suppressor p53 protein according to the presence or absence of a specific binding site (p53CON) in the double-stranded segment. We demonstrate the applicability of the Os,bipy-labeled probes in titration as well as competition MBIP assays to evaluate p53 relative affinity to various sequence-specific or structurally distinct unlabeled DNA substrates upon modulation of the p53-DNA binding by monoclonal antibodies used for the immunoprecipitation. To detect the p53-bound osmium-labeled probes, we took advantage of a catalytic peak yielded by Os,bipy-modified DNA at the mercury-based electrodes, allowing facile determination of subnanogram quantities of the labeled oligonucleotides. Versatility of the electrochemical MBIP technique and its general applicability in studies of any DNA-binding protein is discussed. Figure

A cysteamine (CysAm) nanostructure was generated to act as an intermediate layer between gold electrode and carbon nanotubes. A bare gold electrode was placed in a solution of CysAm to create a self-assembled monolayer on its surface. The modified electrode was then incubated with a solution of multi-walled carbon nanotubes. Cyclic voltammetry and atomic force microscopy were used to characterize the modified electrode. The results indicated that the number of functionalized MWCNTs on the surface of the electrodes increased by enhancing incubation time.

We describe a sensitive sandwich immunoassay for alpha-fetoprotein (AFP). It is making use of gold nanoparticles (GNPs) and magnetic beads (MBs) as labels, and of resonance Rayleigh scattering for detection. Two antibodies were labeled with GNPs and MBs, respectively, and MB-antigen-GNP complexes were formed in the presence of antigens. The MB labels also serve as solid phase carriers that can be used to magnetically separate the immuno complex. The GNP labels are used as optical probes, and Rayleigh scattering was used to determine the concentration of free GNPs-antibody after separation of the MB-antigen-GNP complexes. The concentration of AFP is related to the intensity of light scattered by free GNPs in the 13.6 pM to 436 pM concentration range, and the limit of detection is 13.6 pM. The method was applied to the determination of AFP in sera of cancer patients, and the results agree well with those obtained by conventional ELISA.

A composite film made from a thiol-functionalized mesoporous molecular sieve and an ionic liquid is introduced for use in a voltammetric sensor for Cd(II). The electrode exhibits excellent sensitivity towards Cd(II) in showing a markedly increased stripping peak current. Following the optimization of the experimental parameters, a linear response is obtained in the concentration range from 29?nM to 0.87?mM of Cd(II). The detection limit is as low as 1.0?nM (at S/N?=?3) after an accumulation at ?1.1?V for 4?min. The method was successfully applied to determine Cd(II) in water samples. Features such as large electroactive area, fast electron transfer and low background current make this electrode a promising platform for fabricating reliable electrochemical sensors for various species, such as heavy metals and environmental pollutants.

A sensor for hydrogen peroxide is described that is based on an indium tin oxide electrode modified with Fe3O4 magnetic nanoparticles which act as a mimic for the enzyme peroxidase and greatly improve the analytical performance of the sensor. The amperometric current is linearly related to the concentration of H2O2 in the range from 0.2 mM to 2 mM, the regression equation is y?=?-0.5–1.82x, the correlation coefficient is 0.998 (n?=?3), and the detection limit is 0.01 mM (S/N?=?3). The sensor exhibits favorable selectivity and excellent stability.

DNA ligases are essential enzymes in all cells and have been proposed as targets for novel antibiotics. Efficient DNA ligase activity assays are thus required for applications in biomedical research. Here we present an enzyme-linked electrochemical assay based on two terminally tagged probes forming a nicked junction upon hybridization with a template DNA. Nicked DNA bearing a 5' biotin tag is immobilized on the surface of streptavidin-coated magnetic beads, and ligated product is detected via a 3' digoxigenin tag recognized by monoclonal antibody-alkaline phosphatase conjugate. Enzymatic conversion of napht-1-yl phosphate to napht-1-ol enables sensitive detection of the voltammetric signal on a pyrolytic graphite electrode. The technique was tested under optimal conditions and various situations limiting or precluding the ligation reaction (such as DNA substrates lacking 5′-phosphate or containing a base mismatch at the nick junction, or application of incompatible cofactor), and utilized for the analysis of the nick-joining activity of a range of recombinant Escherichia coli DNA ligase constructs. The novel technique provides a fast, versatile, specific, and sensitive electrochemical assay of DNA ligase activity.

We have developed a piezoelectric sensor for the determination of atrazine. It is based on the modification of a molecularly imprinted film of TiO2 that was placed on a quartz crystal via a surface sol?Cgel process. The resulting sensor exhibits high selectivity for atrazine, a re-usability that is better than that of other sensors, a response time of 3?min, a wider linear range (0.0005?C8?mM), and a lower detection limit (0.1???M). The analytical application of the atrazine sensor confirms the feasibility of atrazine determination.

We report on a new scheme for the determination of the activity of caspase-3 using a specific peptide labeled with N-(4-aminobutyl)-N-ethylisoluminol (ABEI) as a chemiluminescent (CL) probe and on the development of magnetic separation technology. Firstly, the ABEI-labeled and biotinylated peptide was prepared and conjugated to streptavidin-coated magnetic beads (MBs) to form f-MBs (functionalized magnetic beads). The f-MBs contain a site (DEVD, Asp-Glu-Val-Asp) that is cleaved by caspase-3. Upon cleavage, the terminal residue attached to ABEI can dissociate from the f-MBs and can be used for CL detection. CL intensity is linearly related to the concentration of caspase-3 in the range 1.0 to 600 ng mL?1, with a detection limit of 0.3 ng mL?1. The relative standard deviation of the assay is 3.6 % at a level of 50 ng mL?1 of caspase-3 (for n?=?11). The CL assay has been applied to the determination of caspase-3 in Jurkat cell extract with recoveries between 96.6 % and 106.1 % (n?=?5).

We present a microfluidic device, which enables single cells to be reliably trapped and cultivated while simultaneously being monitored by means of multifrequency electrical impedance spectroscopy (EIS) in the frequency range of 10 kHz–10 MHz. Polystyrene beads were employed to characterize the EIS performance inside the microfluidic device. The results demonstrate that EIS yields a low coefficient of variation in measuring the diameters of captured beads (~0.13 %). Budding yeast, Saccharomyces cerevisiae, was afterwards used as model organism. Single yeast cells were immobilized and measured by means of EIS. The bud growth was monitored through EIS at a temporal resolution of 1 min. The size increment of the bud, which is difficult to determine optically within a short time period, can be clearly detected through EIS signals. The impedance measurements also reflect the changes in position or motion of single yeast cells in the trap. By analyzing the multifrequency EIS data, cell motion could be qualitatively discerned from bud growth. The results demonstrate that single-cell EIS can be used to monitor cell growth, while also detecting potential cell motion in real-time and label-free approach, and that EIS constitutes a sensitive tool for dynamic single-cell analysis. The trapping or immobilization of individual cells at specific locations in microfluidic platforms is essential for single cell studies, especially those requiring cell stimulation and downstream analysis of cellular content. Selectivity for individual cell types is required when mixtures of cells are analyzed in heterogeneous and complex matrices, such as the selection of metastatic cells within blood samples. Here, we demonstrate a microfluidic device based on direct current (DC) insulator-based dielectrophoresis (iDEP) for selective trapping of single MCF-7 breast cancer cells from mixtures with both mammalian peripheral blood mononuclear cells (PBMC) as well MDA-MB-231 as a second breast cancer cell type. The microfluidic device has a teardrop iDEP design optimized for the selective capture of single cells based on their differential DEP behavior under DC conditions. Numerical simulations adapted to experimental device geometries and buffer conditions predicted the trapping condition in which the dielectrophoretic force overcomes electrokinetic forces for MCF-7 cells, whereas PBMCs were not trapped. Experimentally, selective trapping of viable MCF-7 cells in mixtures with PBMCs was demonstrated in good agreement with simulations. A similar approach was also executed to demonstrate the selective trapping of MCF-7 cells in a mixture with MDA-MB-231 cells, indicating the selectivity of the device for weakly invasive and highly invasive breast cancer cells. The DEP studies were complemented with cell viability tests indicating acceptable cell viability over the course of an iDEP trapping experiment.
