Intramolecular charge transfer (ICT) reaction in a newly synthesized molecule, of 4-(1-morpholenyl) benzonitrile (M6C), in AOT/water/heptane reverse micelles at different pool sizes has been studied by using steady-state and time-resolved fluorescence emission spectroscopy. The pool size dependences of the reaction equilibrium constant and reaction rate have been explained in terms of the average polarity of the confined solvent pools estimated from the fluorescence emission Stokes shift of a nonreactive probe, coumarin 153, dissolved in these microemulsions. 6-Propionyl-2-(N,N-dimethyl)aminonaphtahalene, PRODAN, is widely used as a fluorescent molecular probe due to its significant Stokes shift in polar solvents. It is an aromatic compound with intramolecular charge-transfer (ICT) states which can be particularly useful as sensors. In this work, we performed absorption, steady-state, time-resolved fluorescence (TRES), and time-resolved area normalized emission (TRANES) spectroscopies on PRODAN dissolved in nonaqueous reverse micelles. The reverse micelles are composed of polar solvents/sodium 1,4-bis-2-ethylhexylsulfosuccinate (AOT)/n-heptane. Sequestered polar solvents included ethylene glycol (EG), propylene glycol (PG), glycerol (GY), formamide (FA), dimethylformamide (DMF), and dimethylacetamide (DMA). The experiments were performed with varying surfactant concentrations at a fixed molar ratio W(S) = [polar solvent]/[AOT]. In every reverse micelle studied, the results show that PRODAN undergoes a partition process between the external solvent and the reverse micelle interface. The partition constants, K(p), are quantified from the changes in the PRODAN emission and/or absorption spectra with the surfactant concentration. The K(p) values depend strongly on the encapsulated polar solvent and correlate quite well with the AOT reverse micelle interface's zones where PRODAN can exist and emits. Thus, the partition toward the reverse micelle interface is strongly favored in DMF and DMA containing micelles where the PRODAN emission comes only from an ICT state. For GY/AOT reverse micelles, the K(p) value is the lowest and only emission from the local excited (LE) state is observed. On the other hand, for EG/AOT, PG/AOT, and water/AOT reverse micelles, the K(p) values are practically the same and emission from both states (LE and ICT) is simultaneously detected. We show here that it is possible to control the PRODAN state emission by simply changing the properties of the AOT reverse micelle interfaces by choosing the appropriate polar solvent to make the reverse micelle media. Indeed, we present experimental evidence with the answer to the long time question about from which state does PRODAN emit, a process that can be controlled using the unique reverse micelle interfaces properties.  相似文献

The dramatic impact of differing environments on proton transfer dynamics of the photoacid HPTS prompted us to investigate these systems with two highly complementary methods: ultrafast time-resolved transient absorption and two-dimensional NMR spectroscopies. Both ultrafast time-resolved transient absorption spectroscopy and time-resolved anisotropy decays demonstrate the proton transfer dynamics depend intimately on the specific reverse micellar system. For w(0) = 10 reverse micelles formed with anionic AOT surfactant, the HPTS proton transfer dynamics are similar to dynamics in bulk aqueous solution, and the corresponding (1)H 2D NOESY NMR spectra display no cross peaks between HPTS and AOT consistent with the HPTS residing well hydrated by water in the interior of the reverse micelle water pool. In contrast, ultrafast transient absorption experiments show no evidence for HPTS photoinduced proton transfer reaction in reverse micelles formed with the cationic CTAB surfactant. In CTAB reverse micelles, clear cross peaks between HPTS and CTAB in the 2D NMR spectra show that HPTS embeds in the interface. These results indicate that the environment strongly impacts the proton transfer reaction and that complementary experimental techniques develop understanding of how location critically affects molecular responses.  相似文献

Catalytic performance of lignin peroxidase in a novel reverse micelle   总被引:1,自引:0,他引:1  
To enhance the catalytic activity of lignin peroxidase (LiP) in a reverse micelle, a synthesized two-tail nonionic surfactant N-gluconyl glutamic acid didecyl ester (GGDE) was used to formulate a novel reverse micelle. Based on the LiP catalyzed oxidation of veratryl alcohol (VA) in this novel GGDE/TritonX-100-cyclohexane-H(2)O reverse micelle, the effects of the size of the reverse micelle, the buffer pH, and the concentration of H(2)O(2) on the catalytic activity of LiP were investigated. Under the optimized conditions, the catalytic efficiency of LiP in the GGDE/TritonX-100 reverse micelle was 40 times higher than that in the AOT reverse micelle. The full expression of catalytic activity of LiP in this medium was mainly due to the lack of electrostatic interaction between LiP and the head group of GGDE and TritonX-100 and to the size fit between LiP and the inner water cavity of the reverse micelle.  相似文献

The effects of various salts and HClO4 on the configuration change of cobalt(II)-halide complexes in CHCl3/CTAC or CTAB/H2O reverse micelle systems were examined at 25 degrees C by means of spectrophotometry, where CTAC and CTAB represent cetyltrimethylammonium chloride and bromide, respectively. The formation of the [CoCl4]> or [CoBr4]2- species of the tetrahedral configuration from [Co(H2O)6]2+ of the octahedral configuration in the reverse micelles was greatly promoted not only by a decrease in the W value (W = [H2O]/[surfactant]), but also, at a constant W value (e.g., W = 2.0), by the addition of relatively low concentrations of salts or the acid (e.g., 4.0 mol dm(-3) in the aqueous phase or 4.0 x 10(-2) mol dm(-3) in the whole reverse micelle system). The effects of perchlorate salts increased as Na+ < or = Li+ approximately H+ < Sr2+ < Ca2+ < Mg2+. Non-metallic salts, various tetraalkylammonium (R4N+) salts at lower concentrations, gave minor effects. The enhanced effects of metal salts on the configuration change of the cobalt(II)-halide complexes were interpreted by a further distortion of the hydrogen-bonded structure of the water in a "water pool" in the presence of salts of even relatively low concentrations. A conformation change with increasing temperature was also attributed to a further distortion of the water structure. An almost completed formation of [CoBr4]2- as well as [CoCl4]2- was attained in the reverse micelles at a low W value of 0.69 containing LiClO4 or HClO4. A partial transfer of the [CoX4]2- species from a "water pool" into the CHCl3 phase by the addition of the metal salts may be suspected. An examination of cobalt(II)-bromide complexes in dichloromethane/CTAB/H2O at W = 1.3 - 5.55 justified all the arguments concerning the chloroform systems. The Raman spectra of D2O containing concentrated LiBr and LiClO4 have supplied conclusive evidence that the hydrogen-bonded structure of the bulk water is completely distorted by extremely concentrated salts.  相似文献

Fluorescence resonance energy transfer (FRET) from coumarin 480 (C480) to fluorescein 548 (F548) in a sodium dioctyl sulfosuccinate (AOT) reverse micelle is studied by picosecond and femtosecond emission spectroscopy. In bulk water, at the low concentration of the donor (C480) and the acceptor (F548), no FRET is observed. However, when the donor (C480) and the acceptor (F548) are confined in a AOT reverse micelle very fast FRET is observed. The time constants of FRET were obtained from the rise time of the emission of the acceptor (F548). In a AOT microemulsion, FRET is found to occur in multiple time scales--3, 200, and 2700 ps. The 3 ps component is assigned to FRET in the water pool of the reverse micelle with a donor-acceptor distance, 16 A. The 200 ps component corresponds to a donor-acceptor distance of 30 A and is ascribed to the negatively charged acceptor inside the water pool and the neutral donor inside the alkyl chains of AOT. The very long 2700 ps component may arise due to FRET from a donor outside the micelle to an acceptor inside the water pool and also from diffusion of the donor from bulk heptane to the reverse micelle. With increase in the excitation wavelength from 375 to 405 nm the relative contribution of the FRET due to C480 in the AOT reverse micelle (the 3 and 200 ps components) increases.  相似文献

The higher order structure of proteins solubilized in an bis(2-ethylhexyl) sulfosuccinate sodium (AOT) reverse micellar system was investigated. From circular dichroic (CD) measurement, CD spectra of cytochrome c, which is solubilized at the interface of reverse micelles, markedly changed on going from buffer solution to the reverse micellar solution, and the ellipticity values in the far- and near-UV regions decreased with decreasing the water content (W0: molar ratio of water to AOT), indicating that the secondary and tertiary structures of cytochrome c changed with the water content. The ellipticity of ribonuclease A, which is solubilized in the center of micellar water pool, in the near-UV region was dependent on W0 and became minimum when W0 of ca. 8 while the ellipticity in the far-UV region was almost constant, indicating that the tertiary structure of ribonuclease A was affected by the water content, but the secondary structure was conserved. The degree of curvature of the micellar interface appears to influence the protein structure because the reverse micelle size is linearly proportional to the W0 value. As evidence of this, when the micelle size was comparable to the protein's dimensions, the structures were more affected by the water content. Judging from the dependence of the factor influencing the protein structure on the protein species, the location of solubilized protein in reverse micelles is significantly related to whether the protein structure in the system is affected by the micellar interface. In the cases of cytochrome c and lysozyme, the ellipticity against W0 was dependent on the AOT concentration. In contrast, ribonuclease A gave very similar ellipticity values whatever the AOT concentration. In the n-hexane micellar system, cytochrome c exhibited lower ellipticity values and ribonuclease A in the lower W0 range (W0 < ca. 8) higher ellipticity values. These results indicated that the interaction between the protein and the micellar interface is a dominant factor influencing the protein structure in reverse micelles, and that it is governed by the location of solubilized proteins and the state of the micellar interface.  相似文献

A series of ester, p-N,N-dimethylaminobenzoates ((CH3)2NC6H4COOR, R=n-CnH2n+1, n=1,2,...8), were synthesized and their fluorescence spectra were recorded. Intramolecular charge transfer (CT) dual fluorescence was found in common organic solvent or in water with cetyltrimethylammonium bromide (CTAB) micelle. The fluorescence intensity ratio of CT band to normal band (ICT/ILE) was decreased dramatically with the increasing of carbon number of R group when the carbon number of R group were 1-3, but the dependence was not obvious when the carbon number of R group were 4-8. Oppositely, the ICT/ILE was increased with increasing of carbon number of R group in water with CTAB micelle.  相似文献

Electrochemical extraction of proteins by reverse micelle formation   总被引:1,自引:0,他引:1  
The transfer of proteins by the anionic surfactant bis(2-ethylhexyl) sulfosuccinate (AOT) at a polarized 1,2-dichloroethane/water (DCE/W) interface was investigated by means of ion-transfer voltammetry. When the tetrapentylammonium salt of AOT was added to the DCE phase, the facilitated transfer of certain proteins, including cytochrome c (Cyt c), ribonuclease A, and protamine, could be controlled electrochemically, and a well-defined anodic wave for the transfer was obtained. At low pH values (e.g., pH 3.4), the anodic wave was usually well-separated from the wave for the formation of protein-free (i.e., unfilled) reverse micelles. The anodic wave for the protein transfer was analyzed by applying the theory for facilitated transfer of ions by charged ligands and then supplying information regarding the number of AOT anions reacting with one protein molecule and the total charge carried by the protein transfer. However, controlled-potential electrolyses performed for the transfer of Cyt c, which is red, revealed that the protein-AOT complexes were unstable in DCE and liable to aggregate at the interface when the pH of the W phase was 3.4. At pH 7.0, when formation of unfilled reverse micelles occurred simultaneously, the protein-AOT complexes appeared to be stabilized, probably via fusion with unfilled reverse micelles.  相似文献

The intramolecular charge transfer (ICT) property of trans-ethyl p-(dimethylamino) cinnamate is used to probe the anionic, cationic, and nonionic micelles by steady-state and picosecond time-resolved fluorescence spectroscopy. The ICT fluorescence band intensity was found to increase with concomitant blue shift with addition of surfactants. All the experimental results suggest that the probe molecule resides in the micelle-water interface rather than going into the core. However, the penetration is more toward the micellar core in nonionic surfactants when compared with ionic micelles. The decrease in nonradiative decay constants in micellar environments indicate restricted motion of the probe toward the formation of ICT state. Critical micelle concentrations were determined from the sharp change in fluorescence intensity and effective dielectric constants of the micelle-water interface were calculated from the correlation diagram of 0,0 transition energy with polarity of the medium.  相似文献

Yeast alcohol dehydrogenase (YADH) showed substantial decrease in its catalytic activity due to the strong electrostatic interaction between the head groups of sodium bis(2-ethylhexyl) sulfosuccinate (AOT) and YADH in AOT reverse micelles. However, the catalytic activity of YADH in a nonionic reverse micellar interface (GGDE/TX-100) obtained from a functional nonionic surfactant N-gluconyl glutamic acid didecyl ester (GGDE) and Triton X-100 (TX-100) was higher than that in AOT reverse micelle under the respective optimum conditions. A comparison of the kinetic parameters showed that the turnover number kcat in GGDE/TX-100 reverse micelle was 1.4 times as large as that in AOT reverse micelle, but the Michaelis constants in AOT reverse micelle for ethanol KmB was twice and for coenzyme NAD+ KmA was 5 times higher than their counterparts in GGDE/TX-100 reverse micelle. For the conversion of ethanol, the smaller KmB and larger kcat in GGDE/TX-100 reverse micelle resulted in higher catalytic efficiency kcat/KmB. The stability of YADH in GGDE/TX-100 reverse micelle was also found to be better than that in AOT reverse micelle. They were mainly attributed to the absence of electric charge on the head groups of GGDE and TX-100 in the GGDE/TX-100 reverse micelle.   相似文献

Polypyrrole (PPy) nanotubes were readily fabricated through chemical oxidation polymerization in sodium bis(2-ethylhexyl) sulfosuccinate (AOT) reverse (water-in-oil) emulsions. The reverse cylindrical micelle phase was characterized, and the key factors affecting the formation of PPy nanotubes were systematically inspected. AOT reverse cylindrical micelles were prepared via a cooperative interaction between an aqueous FeCl3 solution and AOT in an apolar solvent. In the H2O/FeCl3/AOT/apolar solvent system, the aqueous FeCl3 solution played a role in increasing the ionic strength and decreasing the second critical micelle concentration of AOT. As a result, AOT reverse cylindrical micelles could be spontaneously formed in an apolar solvent. In addition, iron cations were adsorbed to the anionic AOT headgroups that were capable of extracting metal cations from the aqueous core. Under these conditions, the addition of pyrrole monomer resulted in the chemical oxidation polymerization of the corresponding monomer at the surface of AOT reverse cylindrical micelles, followed by the formation of tubular PPy nanostructures. In a typical composition (74.0 wt % hexane, 22.4 wt % AOT, and 3.6 wt % aqueous FeCl3 solution at 15 degrees C), the average diameter of PPy nanotubes was approximately 94 nm and their length was more than 2 mum. The PPy nanotube dimensions were affected by synthetic variables such as the weight ratio of aqueous FeCl3 solution/AOT, type of apolar solvent, and reaction temperature. Moreover, the relationship between the diameter and the conductivity of the nanotubes was investigated.  相似文献

The primary objective of the present study is to understand how the different nonionic surfactants modify the anisotropic interface of cationic water-in-oil (W/O) microemulsions and thus influences the catalytic efficiency of surface-active enzymes. Activity of Chromobacterium viscosum lipase (CV-lipase) was estimated in several mixed reverse micelles prepared from CTAB and four different nonionic surfactants, Brij-30, Brij-92, Tween-20, and Tween-80/water/isooctane/n-hexanol at different z ([cosurfactant]/[surfactants]) values, pH 6 (20 mM phosphate), 25 degrees C across a varying range of W0 ([water]/[surfactants]) using p-nitrophenyl-n-octanoate as the substrate. Lipase activity in mixed reverse micelles improved maximum up to approximately 200% with increasing content of non-ionic surfactants compared to that in CTAB probably due to the reduced positive charge density as well as plummeted n-hexanol (competitive inhibitor of lipase) content at the interfacial region of cationic W/O microemulsions. The highest activity of lipase was observed in CTAB (10 mM) + Brij-30 (40 mM)/isooctane/n-hexanol)/water system, k2 = 913 +/- 5 cm3 g-1 s-1. Interestingly, this observed activity is even higher than that obtained in sodium bis (2-ethyl-1-hexyl) sulfosuccinate (AOT)/n-heptane reverse micelles, the most popular W/O microemulsion in micellar enzymology. To ascertain the influence of non-ionic surfactants in improving the activity of surface-active enzymes is not limited to lipase only, we have also investigated the catalytic activity of Horseradish peroxidase (HRP) in different mixed W/O microemulsions. Here also following the similar trend as observed for lipase, HRP activity enhanced up to 2.5 fold with increasing concentration of nonionic surfactants. Finally, the enzyme activity was correlated with the change in the microenvironment of mixed reverse micelles by steady-state fluorescence study using 8-anilino-1-napthalenesulphonic acid (ANS) as probe.  相似文献

The mechanism of the solvent extraction of cytochrome c (Cyt c) via reverse micelle formation was studied from an electrochemical point of view. Potentiometric measurements showed that the Galvani potential difference of the oil/water (O/W) interface played a crucial role in the spontaneous extraction of Cyt c with bis(2-ethylhexyl)sulfosuccinate (AOT). However, the dependence of the extraction efficiency on the concentration of an aqueous electrolyte (KCl) could be explained not by the effect of the interfacial potential, but by the change in the interfacial tension (gamma). Electrocapillary measurements showed that the adsorption of AOT anions to the O/W interface resulted in a significant decrease of gamma in a higher potential range, where reverse micelles were formed. The bottom level of gamma in the higher potential range was increased with [KCl]. The lower extraction efficiency for higher [KCl]'s was elucidated by a "size exclusion effect". This was also supported by water-content measurements by the Karl Fisher method.  相似文献

The coupling reaction between cetylbromide (CB) and trimethylamine (TMA) to yield the surfactant cetyltrimethylammonium bromide (CTAB) is studied in the system chloroform/isooctane (2/1,v/v)/water in which CTAB forms reverse micelles. This system affords an endogenous micelle population growth, i.e., an increase of the concentration of the micelles due to appearance of the surfactant in situ. The reaction is studied in the presence of preexisting CTAB reverse micelles. The rate of CTAB formation is measured by NMR spectroscopy, and the endogenous micelle population growth is directly monitored by time-resolved fluorescence quenching analysis. Under our experimental conditions, a 100% yield of the chemical reaction brings about a fourfold increase in the population of the reverse micelles. Since the water concentration is constant during chemical reaction, the newly formed water pools are formed at the expense of the initial ones, which brings about a decrease of the average water pool radius during micellar growth. The implication of the endogenous micelle population growth as a model for biological systems is briefly discussed.  相似文献

Photoinduced disruption of a sodium bis(2-ethylhexyl) sulfosuccinate (AOT) reverse micelle is triggered by a Malachite Green leuconitrile derivative (MGL). UV irradiation of MGL solubilized in an AOT-water-chloroform mixture creates a cationic surfactant that interacts electrostatically with the anionic AOT. We investigated the disruption of the reverse micelle by using proton nuclear magnetic resonance spectroscopy and found that UV irradiation of MGL decreases the number of water molecules solubilized in the interior of the AOT reverse micelles. Furthermore, the photoinduced disruption of the reverse micelle is shown to release ribonuclease A, which is trapped in the water in the interior of the AOT reverse micelle. This photoinduced release may offer a desirable transport system of biopolymers.  相似文献

The nucleophilic aromatic substitution (S(N)Ar) reaction between 1-fluoro-2,4-dinitrobenzene and piperidine (PIP) were studied in two different reverse micellar interfaces: benzene/sodium 1,4-bis(2-ethylhexyl) sulfosuccinate (AOT)/water and benzene/benzyl-n-hexadecyl dimethylammonium chloride (BHDC)/water reverse micellar media. The kinetic profiles of the reactions were investigated as a function of variables such as surfactant and amine concentration and the amount of water dispersed in the reverse micelles, W0 = [H2O]/[surfactant]. In the AOT system at W0 = 0, no micellar effect was observed and the reaction takes place almost entirely in the benzene pseudophase, at every AOT and PIP concentration. At W0 = 10, a slight increment of the reaction rate was observed at low [PIP] with AOT concentration, probably due to the increase of micropolarity of the medium. However, at [PIP] > or = 0.07 M the reaction rates are always higher in pure benzene than in the micellar medium because the catalytic effect of the amine predominates in the organic solvent. In the BHDC system the reaction is faster in the micellar medium than in the pure solvent. Increasing the BHDC concentration accelerates the overall reaction, and the saturation of the micellar interface is never reached. In addition, the reaction is not base-catalyzed in this micellar medium. Thus, despite the partition of the reactants in both pseudophases the reactions effectively take place at the interface of the aggregates. The kinetic behavior can be quantitatively explained taking into account the distribution of the substrate and the nucleophile between the bulk solvent and the micelle interface. The results were used to evaluate the amine distribution constant between the micellar pseudophase and organic solvent and the second-order rate coefficient of S(N)Ar reaction in the interface. A mechanism to rationalize the kinetic results in both interfaces is proposed.  相似文献
