Squaramide-substituted calix[4]pyrrole 2 was synthesized and its anion-binding properties were investigated. On the basis of UV spectroscopy, 1H NMR spectroscopic and computational DFT model analyses, compared with the parent calix[4]pyrrole 1, compound 2 shows significantly enhanced binding affinities for anions in CH2Cl2, and it was concluded that this increase in affinity is ascribed to participation of the squaramide unit in anion binding.  相似文献

A series of novel calix[4]arenocrowns 1a-c were efficiently synthesized by a one-pot reaction of calix[4]monohydroquinone diacetate 5 with ditosylate 6 and its analogues in the presence of sodium hydroxide. It was found that the calix[4]arenocrowns could form stable pseudorotaxane-type complexes 2a-c with paraquat, and further self-assemble into calix[4]areno[2]catenanes 3a-c with dicationic salt 8 and p-bis(bromomethyl)benzene.  相似文献

A novel series of 5H-pyridazino[4,5-b]indoles were designed and synthesized in order to find novel potent anticancer compounds.The structures were confirmed by ~1H NMR and MS.Their antiproliferative activities against two cancer cell lines were tested by the MTT method in vitro.Three of compounds (1e,1g,and 1h) exhibited potent antiproliferative activities,especially compound 1h (with IC_(50) values of 5.2μmol/L and 1.9μmol/L against Bel-7402 and HT-1080,respectively).The preliminary structure-activity relationships of 5H-pyridazino[4,5-b]indole derivatives were discussed.  相似文献

Two strategies for the design of new pillar[5]arene-based mechanically self-interlocked molecules (MSMs) are reported here. The first strategy is based on the construction of an intermediate pseudo[1]rotaxane followed by the desired bis-[1]rotaxane. The other one is based on the construction of the desired bis-[1]-rotaxane directly via a condensation reaction through host-guest interactions between a mono-functionalized pillar[5]arene and the axle. This compound has interesting self-assembly properties in methanol and some extended applications of this compound will be reported in the near future.  相似文献

Fluorinated ionomer p-perfluoro[1-(2-sulfonic)ethoxy]ethylated polyacrylonitrile-styrene (SFAS) (5) was synthesized via electron transfer reaction between polyacrylonitrile-styrene (AS) (1) and perfluoro-di[2-(2-fluorosulfonyl)ethoxy]propionyl peroxide (FAP) (2) and followed by alkali hydrolysis and acidification of p-perfluoro[1-(2-fluorosulfonyl)ethoxy]ethylated polyacrylonitrile-styrene (3). The microstructure of ionomer 5 was well characterized by FTIR and 19F NMR. Its desulfonation occurred above 197 °C was found by TGA, the degree of substitution (DS) and ion exchange capacity (IEC) determined by titration were well controlled through changing the molar ratio of 2:1. The proton exchange membranes made of ionomer 5 have water uptake from 13.4 to 135.3% and conductivity up to 10−2 S cm−1 at 25 °C.  相似文献

In this study, we reported the synthesis of three kinds of mono-functional pillar[5]arene derivatives PRI, PRII and R and their structures were studied by 1D and 2D NMR spectra and mass spectra. The 2D NMR spectra including 1H-13C HSQC, 1H-1H COSY and NOESY spectra indicated that PRI and PRII are both stable self-included pseudo[1]rotaxanes in CDCl3. These original structures are promising compounds for the design of pillar[5]-based [1]rotaxane. And the results showed that R could exist stable in CDCl3 and DMSO because of the coordination of N-H?O hydrogen bonding interaction and C-H?π interaction.  相似文献

A variety of new thiazolo[3,2- a ]pyridine derivatives 2a-h having 3-indolyl group were produced by refluxing 1a with different benzylidenemalononitrile derivatives. Reactivity of compound 4 toward some nitrogen nuclcophiles was investigated. Thus, the novel pyrazoles 6a , b were obtained when compound 4 was allowed to react with hydrazine and phenyl hydrazine in ethanol under reflux. On the other hand, pyrazolo[3',4':4,5]thiazolo[3,2- a ]-3-aza[1,8]naphthyridine 8 was formed by condensation of compound 4 with benzoyl hydrazine. Finally, condensed heterocyclic compounds containing pyran rings 9 and 10 were obtained by treatment compound 4 with active ethylene compounds.  相似文献

The wide-spreading SARS-CoV-2 virus has put the world into boiling water for more than a year, however pharmacological therapies to act effectively against coronavirus disease 2019 (COVID-19) remain elusive. Chloroquine (CQ), an antimalarial drug, was found to exhibit promising antiviral activity in vitro and in vivo at a high dosage, thus CQ was approved by the FDA for the emergency use authorization (EUA) in the fight against COVID-19 in the US, but later was revoked the EUA status due to the severe clinical toxicity. Herein, we show that supramolecular formulation of CQ by a macrocyclic host, curcurbit[7]uril (CB[7]), reduced its non-specific toxicity and improved its antiviral activity against coronavirus, working in synergy with CB[7]. CB[7] was found to form 1:1 host-guest complexes with CQ, with a binding constant of ∼104 L/mol. The CQ-CB[7] formulation decreased the cytotoxicity of CQ against Vero E6 and L-02 cell lines. In particular, the cytotoxicity of CQ (60 μmol/L) against both Vero E6 cell line and L-02 cell lines was completely inhibited in the presence of 300 μmol/L and 600 μmol/L CB[7], respectively. Furthermore, the CB[7] alone showed astonishing antiviral activity in SARS-CoV-2 infected Vero E6 cells and mouse hepatitis virus strain A59 (MHV-A59) infected N2A cells, and synergistically improved the antiviral activity of CQ-CB[7], suggesting that CB[7]-based CQ formulation has a great potential as a safe and effective antiviral agent against SARS-CoV-2 and other coronavirus.  相似文献

Photocycloaddition reactions of 6,6′-dimethyl-4,4′-[bis(methylenoxy)phenylene]-di-2-pyrones (4a-c) with benzophenone (2a) by mixing in the solid state (solid solution) afforded the corresponding oxetane derivatives (5a-c; 1:2 adducts) with high site- and regioselectivity across the C5-C6 and C5′-C6′ double bonds in 4 via the triplet excited state of benzophenone. The oxetane formation proceeded more effectively in the solid state than in solution. The reaction mechanism was inferred by MO methods to be initiated by electrostatic interaction between the C6 position of 4a-c and the carbonyl oxygen of 2a in their ground states. The solid-state interaction may be enhanced by the electron density at the carbonyl oxygen of the triplet 2a. The transition state (TS) analysis of the [2+2] cycloaddition reactions also suggested some triplet complexes and high regioselectivity. The hydrogen-bonding interaction between 2a and 4a-c and the triplet reaction mechanism were also explained by the IR analyses and the quenching experiments, respectively.  相似文献

Summary 6-Phenyl-1,3-dimethyl-2,4-dioxo-1,2,3,4,8,9-hexahydro-[1,3,5]-thiadiazepino-[3,2-f]-purine (5) was obtained by a three-step synthesis from 8-mercapto-1,3-dimethyl-3,7-dihydro-1H-purine-2,6-dione (1) and 2-(benzoylamino)-ethyl chloride (2)via 8-(benzoylaminoethylthio)-1,3-dimethyl-3,7-dihydro-1H-purine-2,6-dione (3) and its chloromido derivative4. The analogous 9-phenyl-1,3-dimethyl-2,4-dioxo-1,2,3,4,6,7-hexahydro-[1,3,6]-thiadiazepino-[3,2-f]-purine (7) was synthesized either from compound1 and N-(2-chloroethyl)-benzimido chloridevia N-(chloroethyl)-S-(1,3-dimethyl-2,4-dioxo-1,2,3,4-tetrahydro-7H-purin-8-yl)-benzothioimide (6), or alternatively from 7-(2-benzoylaminoethyl)-8-bromo-1,3-dimethyl-3,7-dihydro-1H-purine-2,6-dione (9), its 8-mercapto derivative10 and the corresponding chloroimido compound11 being the intermediates.Part of this paper was presented as a preliminary report at the Congress of Czech and Slovak Chemical Societies, Olomouc, Czech Republic, September 13–16, 1993  相似文献

An unexpected ring-contraction from benzo[b]pyrazino[1,2-d][1,4]thiazine-1,4-diones (6) to benzo[4,5]thiazolo[3,2-a]pyrazine-1,4-diones (7) has been developed. The preliminary mechanistic studies showed the transformation contained two independent steps: the first step is the formation of a Michael adduct upon the addition of the protic solvent, in the presence of base, to the C-2-C-3 double bond of compound 6, and the second step is a ring-contraction induced by oxygen via the migration of sulfur atom from C-2 to C-3 position. 以非经典叶酸拮抗剂2,4-二氨基-6-(4-甲基苯基)乙基吡啶并[3,2-d]嘧啶(wm-5b)及其侧链简化产物2,4-二氨基吡啶并[3,2-d]嘧啶为先导化合物, 选取具有抗肿瘤活性的基团, 通过微波法高效合成了2-位或4-位取代吡啶并嘧啶类非经典叶酸拮抗剂, 研究了2-位及4-位取代基对抗肿瘤活性的影响, 为非经典叶酸拮抗剂的设计合成提供了更多的理论依据. 目标化合物的结构均经核磁共振波谱(NMR)和质谱(MS)确证. 生物活性测定结果表明, 所有目标化合物均具有抗肿瘤活性, 其中, 6-(4-甲基苯基)乙基-4-氨基-2-(3-氯-4-氟苯基)氨基吡啶并[3,2-d]嘧啶(6b)对HL-60细胞的IC50=(4.09±0.48) μmol/L, 对A549细胞的IC50=(17.99±7.20) μmol/L, 而对HCT116细胞的IC50=(14.52±4.74) μmol/L; 部分目标化合物具有二氢叶酸还原酶抑制活性. 此外, 对部分目标化合物和先导物进行了二氢叶酸还原酶晶体结构的分子对接, 对活性结果和构效关系从分子水平上进行解释.  相似文献

Abstract

Benzothiophene -2- carbaldehyde 1 reacted with 2-cyanoethanethioamide 2 in 1:2 molar ratios to give the corresponding 6-amino-4-(benzo[b]thiophen-2-yl)-2-thioxo-1, 2-dihydropyridine-3,5-dicarbonitrile 6. The synthetic potentiality of compound 6 was investigated via its reaction with active halogen-containing reagents to afford the corresponding thieno[2,3-b]pyridine derivatives 11a,b, 14, 16, and 19. Also, compound 6 reacted with hydrazine hydrate to give the pyrazolo[3,4-b]pyridine derivative 21. Compound 21 condensed with 4-(2-thienyl)benzaldehyde to afford pyrazolo[3,4-b]pyridine derivative 23. Structural elucidation of all the newly synthesized heterocyclic compounds was based on elemental analyses, IR, 1H NMR, and mass spectra.

Except for the special case of calix[4]arene diethyl ether 1, the chlorine dioxide oxidation of dialkyl ethers 2-5 yielded only the corresponding calix[4]diquinone dialkyl ethers 8-11. Chlorine dioxide oxidation of calix[4]arene diethyl ether 1 produced two isomeric products 6 and 7, which were stable enough to be isolated by column chromatography. However, a slow conformational interconversion between isomeric pair 6 and 7 was observed at room temperature, and the equilibrium was reached after 400 h at 18 °C with an amount of 5:3 in favor of syn-isomer.  相似文献

Calix[4]arene 3, which contains two distal triazole groups on the lower rim and two distal o-methoxyphenylazo groups on the upper rim, was synthesized and found to be a specific and ratiometric sensor for Hg2+ in a polar protic solvent. A series of o-methoxyphenylazo derivatives (3, 4, 5, 7, and 9) were synthesized, which proved that the lower-rim triazoles and the hydroxyl azophenol(s) were the major ligands for metal ion binding. Though analogues 4 and 10 showed some sensitivity for Hg2+, compound 3 was the only ratiometric chemosensor for Hg2+ among the series of azocalix[4]arenes synthesized in this work. The formation of 3·Hg2+ complex was supported by UV/vis and NMR titration studies and Mass spectrometry. Based on the symmetrical features of NMR spectra of 3·Hg2+, the complex is believed to be symmetrical with respect to the calix[4]arene cavity. Furthermore, the complex was determined to be 1:1 binding stoichiometry by Job’s plot, and the association constant was determined to be 4.02×103 M−1 using Benesi-Hildebrand plot.  相似文献

2-Aminocyclopenta[d]pyrimidines 3a-c were achieved via a one-pot, three-component reactions of cyclopentanone 1, aromatic aldehyde and guanidine hydrochloride (1:2:1 molar ratio). Also, cyclization of 2,5-bis-(arylmethylidene)cyclopentanones 2 with guanidine hydrochloride (1:1 molar ratio) in methanol in the presence of sodiummethoxide afforded cyclopenta-[d]pyrimidines 3. Compound 3a has been shown to be a useful building block for the synthesis of some novel pyrimido[1,2-a]pyrimidines 5, 7 and 12. The structures of the newly synthesized compounds were confirmed on the basis of analytical and spectral data.  相似文献

MgO-MgAl2O4 nanocomposite was prepared from the co-precipitation of Mg(NO3)2 and Al(NO3)3 salts, characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDS) and fourier transform infrared spectroscopy (FTIR) techniques and evaluated in the synthesis of thirty five derivatives of benzo[4,5]thiazolo[3,2-a]chromeno[4,3-d]pyrimidin-6-ones (d1-d34) via the multi-component reaction of 4-hydroxycoumarins, aldehydes, and 2-aminobenzothiazole derivatives under solvent free condition. The catalytic activity of MgO-MgAl2O4 nanocomposite and the synthesis of the above mentioned compounds were investigated under thermal solvent free (times: 1.4–3 h; yields: 75–95%), ultrasonic irradiation (US) conditions (times: 1–2.5 h; yields: 69–97%) and using high-speed ball milling (HSBM) technique (times: 0.7–2.5 h; yields: 67–97%). In all cases, the products were obtained in excellent yields. Nuclear Magnetic Resonance (NMR) and MASS spectroscopy were used to characterize the structure of the desired product. The mechanism for the preparation of compounds d1-d34 was proposed and confirmed by 1H NMR investigations.  相似文献

The results of our studies show that liquid linear polysulfide polymers of an average molecular weight 4260–1030 g mol−1 can be obtained by the reaction of sodium tetrasulfide and 1,1′-[methylenebis(oxy)]-bis-[2-chloroethane] in the presence of 2.0–5.0 mol of hydrazine and at least 4.0 mol of sodium hydroxide/mol of hydrazine in a one-step procedure. The disulfide polymers in the form of sodium mercaptide are soluble in the supernatant liquid from which they are separated by acidification of the solution to a pH value of about 4. The average molecular weight and the content of sulfur in the obtained linear polymers were determined and their IR spectra were recorded. © 1997 John Wiley & Sons, Inc. Two series of thieno[2,3-d]pyrimidine derivatives were designed and synthesized, in which bioactive a-aminophosphonate subunits were introduced at the N3 position through an N–N bond connection.The in vitro cytotoxic activity of the novel compounds was tested against human esophageal carcinoma cells(EC109), human hepatocarcinoma cells(Hep G2), human gastric carcinoma cells(MGC-803),respectively, by the MTT method. The evaluation results revealed that compounds 6mb, 6mf, 6mg, 6nd and 6nh exerted the most potent inhibition against Hep G2, MGC-803 and EC109 cells, respectively. In particular, compound 6mg presented excellent inhibitory effect against Hep G2(91.2%) and MGC-803(94.4%) cells.  相似文献
