The compounds [(η6p‐cymene)RuCl2(4‐nitroaniline)] and [(η6p‐cymene)RuCl2(2‐halogen‐4‐nitroaniline)] were synthesized and characterized by various means. The [(η6p‐cymene)RuCl2(4‐nitroaniline)] and [(η6p‐cymene)RuCl2(2‐fluoro‐4‐nitroaniline)] compounds were determined by X‐ray diffraction, appearing in a distorted piano‐stool type of arrangement with similar bond lengths and angles around the ruthenium. The compounds exhibited moderate to strong in vitro cytotoxicity against A549 and MCF‐7 human cancer cells. Substitution of heavy halogen atom on the ortho position of para‐nitroaniline weakened the cytotoxicity against both of MCF‐7 and A549, except the cases of fluorine substitution for hydrogen atom regarding A549 and bromine substitution for chlorine atom regarding MCF‐7, which showed minor deviation.  相似文献

A series of novel ruthenium(II)–cymene complexes ( 1 – 8 ) containing substituted pyridyl–thiazole ligands, [Ru(η6p‐cymene)(L)Cl]Cl (L = N,N‐chelating derivatives), have been synthesized and characterized using elemental analysis, infrared, 1H NMR and 13C NMR spectroscopies and mass spectrometry. All these complexes not only display marked cytotoxicity in vitro against three different human cancer cell lines (HeLa, A549 and MDA‐MB‐231), but also exhibit promising anti‐metastatic activity at sub‐cytotoxic concentrations. Cell cycle analysis shows that the ruthenium(II) complex‐induced growth inhibition was mainly caused by S‐phase cell cycle arrest. Further protein level analysis suggests that compound 5 may exert antitumor activity via a p53‐independent mechanism.  相似文献

A new class of half‐sandwich (η6p‐cymene) ruthenium(II) complexes supported by 2‐aminofluorene derivatives [Ru(η6p‐cymene)(Cl)(L)] ( L  = 2‐(((9H‐fluoren‐2‐yl)imino)methyl)phenol ( L 1 ), 2‐(((9H‐fluoren‐2‐yl)imino)methyl)‐3‐methoxyphenol ( L 2 ), 1‐(((9H‐fluoren‐2‐yl)imino)methyl)naphthalene‐2‐ol ( L 3 ) and N‐((1H‐pyrrol‐2‐yl)methylene)‐9H‐fluorene‐2‐amine ( L 4 )) were synthesized. All compounds were fully characterized by analytical and spectroscopic techniques (IR, UV–Vis, NMR) and also by mass spectrometry. The solid state molecular structures of the complexes [Ru(η6p‐cymene)(Cl)(L2)], [Ru(η6p‐cymene)(Cl)(L3)] and [Ru(η6p‐cymene)(Cl)(L4)] revealed that the 2‐aminofluorene and p‐cymene moieties coordinate to ruthenium(II) in a three‐legged piano‐stool geometry. The synthesized complexes were used as catalysts for the dehydrogenative coupling of benzyl alcohol with a range of amines (aliphatic, aromatic and heterocyclic). The reactions were carried out under thermal heating, ultrasound and microwave assistance, using solvent or solvent free conditions, and the catalytic performance was optimized regarding the solvent, the type of base, the catalyst loading and the temperature. Moderately high to very high isolated yields were obtained using [Ru(η6p‐cymene)(Cl)(L4)] at 1 mol%. In general, microwave irradiation produced better yields than the other two techniques irrespective of the nature of the substituents.  相似文献

This work describes the synthesis of a series of Ru(II)‐Arene (Arene=p‐cymene, benzene) complexes using different N‐substituted tetrazole ligands and their PTA analogues. All the complexes have been characterized thoroughly using different analytical techniques. Antiproliferative activity of the synthesized complexes against different cell lines indicates remarkable activity of certain complexes up to nanomolar level. In few cases introduction of water soluble PTA (PTA = 1,3,5‐ triaza‐7‐phospha‐tricyclo‐[3.3.1.1]decane) ligand induce significant cytotoxic activity in the ruthenium complex with respect to their chloro analogues, particularly against Jurkat and MCF‐7 cell lines. Interaction with different biomolecules and stability of the RAPTA complexes have been explored in pseudo‐pharmacological conditions.  相似文献

The reactions of two diaminotriazine ligands 2,4‐diamino‐6‐(2‐pyridyl)‐1,3,5‐triazine (2‐pydaT) and 6‐phenyl‐2,4‐diamino‐1,3,5‐triazine (PhdaT) with ruthenium–arene precursors led to a new family of ruthenium(II) compounds that were spectroscopically characterized. Four of the complexes were cationic, with the general formula [(η6‐arene)Ru(κ2N,N‐2‐pydaT)Cl]X (X=BF4, TsO; arene=p‐cymene: 1.BF4 , 1.TsO arene=benzene: 2.BF4 , 2.TsO ). The neutral cyclometalated complex [(η6p‐cymene)Ru(κ2C,N‐PhdaT*)Cl] ( 3 ) was also isolated. The structures of complexes 2.BF4 and 3.H2O were determined by X‐ray diffraction. Complex 1.BF4 underwent a partial reversible‐aquation process in water. UV/Vis and NMR spectroscopic measurements showed that the reaction was hindered by the addition of NaCl and was pH‐controlled in acidic solution. At pH 7.0 (sodium cacodylate) Ru–Cl complex 1.BF4 was the only species present in solution, even at low ionic strength. However, in alkaline medium (KOH), complex 1.BF4 underwent basic hydrolysis to afford a Ru–OH complex ( 5 ). Fluorimetric studies revealed that the interaction of complex 1.BF4 with DNA was not straightforward; instead, its main features were closely linked to ionic strength and to the [DNA]/complex ratio. The bifunctional complex 1.BF4 was capable of interacting concurrently through both its p‐cymene and 2‐pydaT groups. Cytotoxicity and genotoxicity studies showed that, contrary to the expected behavior, the complex species was biologically inactive; the formation of a Ru–OH complex could be responsible for such behavior.  相似文献

Half‐sandwiched ruthenium (II) arene complexes with piano stool‐like geometry with the general formula [(p‐cymene)RuClL1] and [(p‐cymene)RuClL2] [where L1 = (Z)‐N′‐((1,3‐diphenyl‐1H‐pyrazol‐4‐yl)methylene)furan‐2‐carbohydrazide and L2 = (Z)‐N′‐((1,3‐diphenyl‐1H‐pyrazol‐4‐yl)methylene)thiophene‐2‐carbohydrazide] were synthesized and characterized. The single crystal X‐ray data revealed that the complexes belong to the same crystal system (monoclinic) with octahedral geometry, where the ruthenium atom is surrounded by hydrazone ligand coordinated through ON atoms, one chloride labile co‐ligand and the remaining three coordination sites covered by an electron cloud of p‐cymene moiety. The interaction between the complexes and DNA/bovine serum albumin (BSA) was evaluated using absorption and emission titration methods showing intercalative modes of interaction. The DNA cleavage ability of the complexes was checked by agarose gel electrophoresis method exhibiting the destruction of DNA duplex arrangement. To understand the interaction between ruthenium complex and DNA/BSA molecule, molecular docking studies were performed. In vitro cytotoxicity of the complexes was examined by the 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay on human lung cancer cell line, A549, and found that at lower IC50, cell growth inhibition has occurred. Similarly, the IC50 values of the complexes treated with cancerous cell lines have produced a significant amount of lactase dehydrogenase and nitrite content in the culture medium, which were evaluated as apoptosis‐inducing factors, suggesting that the ruthenium (II) arene hydrazone complexes with pyrazole ligands have promising anticancer activities.  相似文献

Ruthenium(II) π‐coordination onto [28]hexaphyrins(1.1.1.1.1.1) has been accomplished. Reactions of bis‐AuIII and mono‐AuIII complexes of hexakis(pentafluorophenyl) [28]hexaphyrin with [RuCl2(p‐cymene)]2 in the presence of NaOAc gave the corresponding π‐ruthenium complexes, in which the [(p‐cymene)Ru]II fragment sat on the deprotonated side pyrrole. A similar reaction of the bis‐PdII [26]hexaphyrin complex afforded a triple‐decker complex, in which the two [(p‐cymene)Ru]II fragments sat on both sides of the center of the [26]hexaphyrin framework.  相似文献

Two mononuclear ruthenium complexes ( 1 and 2 ) with aroyl/acylthiourea as an ancillary ligand of type, [(η6p‐cymene)RuCl(L‐N,S)], where [ L1  = 2,4‐dichloro‐N‐(o‐tolylcarbamothioyl)benzamide] and L2  = N‐(phenylcarbamothioyl)cyclohexanecarboxamide] were synthesized and well characterized. The single crystal X‐ray diffraction studies revealed the coordination mode and the geometry of the complexes. The two complexes adopted general piano‐stool (three‐legged) geometry with a novel coordination mode of aroyl/acylthiourea through amide N (anionic) and thiocarbonyl S (neutral). This type of monobasic bidentate coordination of the aroyl/acylthiourea ligand was witnessed the first time around the metal ion. The coordination of the complexes was well explained through geometric parameters and frontier molecular orbital parameter values computed at the B3LYP/SDD level. The synthesized complexes were also screened for their antibacterial, antifungal, antioxidant and in vitro antiproliferative activities. Complexes exhibited good antimicrobial agents against various pathogens. The antioxidant activity of the complex 2 has shown most potent activity with IC50 value of 48.55 ± 1.7 μM compared to the reference drug. In addition, the in vitro antiproliferative activity of the complex 2 showed excellent activity against HepG‐2 cell line with the IC50 value of 24.30 ± 1.20 μM which is close to Doxorubicin standard drug.  相似文献

A novel diazadiphosphetidine ligand derived from the reaction of 2,4‐dichloro‐1,3‐dimethyl‐1,3,2,4‐diazadiphosphetidine‐2,4‐dioxide and 2,2′‐(ethane‐1,2‐diylbis[oxy])bis(ethan‐1‐amine) and its Ni(II), Cu(II), and Co(II) complexes have been synthesized, characterized by spectroscopic, elemental analyses, magnetic susceptibility, and conductivity methods, and screened for antimicrobial, DNA binding, and cleavage properties. Spectroscopic analysis and elemental analyses indicate the formula [M(H2L)Cl2] for the Cu(II), Co(II), Ni(II), and Zn(II) complexes and octahedral geometry for all the complexes. The non‐electrolytic nature of the complexes in dimethyl sulfoxide (DMSO) was confirmed by their molar conductance values, which are in the range 12.32–6.73 Ω?1 cm2 mol?1. Computational studies have been carried out at the density functional theory (DFT)‐B3LYP/6‐31G(d) level of theory on the structural and spectroscopic properties of diazadiphosphetidine H2L and its binuclear Cu(II), Co(II), Ni(II), and Zn(II) complexes. Six tautomers and geometrical isomers of the diazadiphosphetidine ligand were confirmed using semiempirical AM1 and DFT method from DMOL3 calculations. The copper complex had the best antibacterial activity against Staphylococcus aureus (ATCC 29213). DNA cleavage activities of the compounds, evaluated on pBR322 DNA by agarose gel electrophoresis in the presence and absence of an oxidant (H2O2) and a free‐radical scavenger (DMSO), indicated no activity for the ligand and moderate activity for the complexes, with the copper complex cleaving pBR322 DNA more efficiently in the presence of H2O2.  相似文献

The syntheses, structures, and chemotherapeutic activities of Ag(I)‐, Au(I)‐, and Ru(II)‐complexes ligated to a novel N‐heterocyclic carbene ligand, 2‐(4‐nitrophenyl)imidazo[1,5‐a]pyridin‐2‐ylidene ( 1 ), are described. The corresponding complexes, [Ag( 1 )2][PF6], [Au( 1 )2][PF6] ( 3 ), and [Ru( 1 )(p‐cymene)Cl][PF6] ( 4 ), were prepared using convenient transmetallation chemistry and characterized using a range of spectroscopic and analytical techniques. X‐ray crystallography revealed that complexes 2 and 3 adopted linear structures whereas 4 exhibited a prototypical “piano‐stool”‐like geometry; the structural assignments were further supported by DFT calculations. A series of in vitro studies revealed that while the aforementioned Ag(I), Au(I) and Ru(II) complexes exhibited significant cytotoxicities against the human colon adenocarcinoma (HCT 116), lung cancer (A549), and breast cancer (MCF7) cell lines, the Ru derivative was most prominent.  相似文献

Cationic (arene)ruthenium‐based tetranuclear complexes of the general formula [Ru4(η6‐p‐cymene)4(μ‐NN)2(μ‐OO∩OO)2]4+ were obtained from the dinuclear (arene)ruthenium complexes [Ru2(η6p‐cymene)2(μ‐OO∩OO)2Cl2] (p‐cymene=1‐methyl‐4‐(1‐methylethyl)benzene, OO∩OO=5,8‐dihydroxy‐1,4‐naphthoquinonato(2?), 9,10‐dihydroxy‐1,4‐anthraquinonato(2?), or 6,11‐dihydroxynaphthacene‐5,12‐dionato(2?)) by reaction with pyrazine or bipyridine linkers (NN=pyrazine, 4,4′‐bipyridine, 4,4′‐[(1E)‐ethene‐1,2‐diyl]bis[pyridine]) in the presence of silver trifluoromethanesulfonate (CF3SO3Ag) (Scheme). All complexes 4 – 12 were isolated in good yield as CF3SO salts, and characterized by NMR and IR spectroscopy. The host–guest properties of the metallarectangles incorporating 4,4′‐bipyridine and (4,4′‐[(1E)‐ethene‐1,2‐diyl]bis[pyridine] linkers were studied in solution by means of multiple NMR experiments (1D, ROESY, and DOSY). The largest metallarectangles 10 – 12 incorporating (4,4′‐[(1E)‐ethene‐1,2‐diyl]bis[pyridine] linkers are able to host an anthracene, pyrene, perylene, or coronene molecule in their cavity, while the medium‐size metallarectangles 7 – 9 incorporating 4,4′‐bipyridine linkers are only able to encapsulate anthracene. However, out‐of‐cavity interactions are observed between these 4,4′‐bipyridine‐containing rectangles and pyrene, perylene, or coronene. In contrast, the small pyrazine‐containing metallarectangles 4 – 6 show no interaction in solution with this series of planar aromatic molecules.  相似文献

Polyol Metal Complexes.471) Crystalline D ‐Mannose‐Copper Complexes from Fehling Solutions Blue, unstable crystals of K3[Cu5(β‐D ‐Manp)4H—13] · α‐D ‐Manp · 16.5 H2O ( 1 ), which contain a pentanuclear cupric complex of the reducing sugar D ‐mannose in its β‐pyranose form (β‐D ‐Manp), have been obtained from ice‐cold aqueous alkaline solutions. The homoleptic pentacuprate contains bridging mannopyranose ligands, which are charged 4— and 2.5—. Addition of ethylenediamine (en) to such Fehling solutions yields N, N′‐Bis(β‐D ‐mannopyranosyl)‐ethylenediamine (L) as a condensation product of the diamine and mannopyranose. Crystals of [(en)2Cu7(β‐D ‐Manp1, 2, 3, 4H—4)2(L2, 3, 4H—3)2] · 26.6 H2O ( 2 ) could be isolated. The heptanuclear cupric complex is a structural derivative of the homoleptic mannose complex.  相似文献

The Schiff base N,N′‐bis(salicylidene)‐1,5‐diamino‐3‐oxapentane (H2L) and its lanthanide(III) complexes, PrL(NO3)(DMF)(H2O) ( 1 ) and Ho2L2(NO3)2 · 2H2O ( 2 ), were synthesized and characterized by physicochemical and spectroscopic methods. Single crystal X‐ray structure analysis revealed that complex 1 is a discrete mononuclear species. The PrIII ion is nine‐coordinate, forming a distorted capped square antiprismatic arrangement. Complex 2 is a centrosymmetric dinuclear neutral entity in which the HoIII ion is eight‐coordinate with distorted square antiprismatic arrangement. The DNA‐binding properties of H2L and its LnIII complexes were investigated by spectrophotometric methods and viscosity measurements. The results suggest that the ligand H2L and its LnIII complexes both connect to DNA in a groove binding mode; the complexes bind more strongly to DNA than the ligand. Moreover, the antioxidant activities of the LnIII complexes were in vitro determined by superoxide and hydroxyl radical scavenging methods, which indicate that complexes 1 and 2 have OH · and O2– · radical scavenging activity.  相似文献

The dynamic kinetic resolution of 2‐aroyl‐1‐tetralones was achieved via asymmetric transfer hydrogenation using (S,S)‐RuCl(p‐cymene)TsDPEN (TsDPEN=N‐(tosyl)‐1,2‐diphenylethylenediamine) in formic acid/triethyl‐ amine (5:2, molar ratio), afforded the desired products in good yields (up to 85%) with diastereomeric ratio up to >99:1 and high enantiomeric excesses (up to >99%). The absolute configuration of major the product was confirmed by X‐ray crystal structure analysis.  相似文献

Three novel compounds, (η6‐p‐cymene)RuCl2(2‐fluoro‐5‐aminopyridine) (compound 1), (η6‐p‐cymene)RuCl2(5‐amino‐2‐chlorpyridine) (compound 2) and (η6‐p‐cymene)RuCl2(2‐bromo‐ 5‐aminopyridine) (compound 3), were synthesized and characterized. The compound 1 and 3 were determined by X‐ray diffraction, showing a distorted piano‐stool type of geometry with similar bond lengths and angles around the ruthenium. Compound 2 exhibited moderate in vitro activity against A549 and MCF‐7 human cancer cells, the other two lower activities. The UV–vis and fluorescent absorption titrations showed that three compounds binded with CT‐DNA in a minor groove. The intrinsic binding constants (Kb) were calculated to be 2.13(±0.03) × 105 M?1, 2.89(±0.03) × 105 M?1 and 2.45(±0.03) × 105 M?1 for compound 1, 2 and 3, respectively, by using UV–vis absorption titrations data. Among the three compound, the highest value of intrinsic binding constant of compound 2 was consistent with its highest cytoxicity against A549 and MCF‐7 human cancer cells in vitro.  相似文献

Tris‐o‐semiquinonato cobalt complexes react with a tetrapodal pyridine‐derived ligand to form dinuclear cobalt compounds of general formula (OMP)[CoQ2]2, where OMP = 2,2′‐(pyridine‐2,6‐diyl)bis(N1,N1,N3,N3‐tetramethylpropane‐1,3‐diamine), Q = mono‐ or dianion of 3,6‐di‐tert‐butyl‐o‐benzoquinone (complex 1 ) and it derivatives: 3,6‐di‐tert‐butyl‐4,5‐N,N′‐piperazino‐o‐benzoquinone (complex 2 ), and 3,6‐di‐tert‐butyl‐4‐Cl‐o‐benzoquinone (complex 3 ). Single crystal X‐ray crystallography of 1 and 3 indicates two bis‐quinonato cobalt units bound by an OMP ligand, which acts as a bridge. Each central cobalt atom is chelated by one N1,N1,N3,N3‐tetramethylpropane‐1,3‐diamine and two o‐quinonato fragments. The nitrogen atom of the pyridine ring is uncoordinated. All complexes were characterized by NIR‐IR and EPR spectroscopy, precise adiabatic vacuum calorimetry, and by variable‐temperature magnetic susceptibility measurements. All data indicate a reversible thermally driven redox‐isomeric (valence tautomeric) transformation in the solid state for all complexes.  相似文献

The syntheses of three mixed ligand chelate copper(II) complexes of the type [Cu(L)(acac)(H2O)]BPh4 where acac=acetyleacetonate; L=N,N‐dimethyl,N′‐benzylethane‐1,2‐diamine ( L1 ), N,N‐dimethyl, N′‐2‐methylbenzylethane‐1,2‐diamine ( L2 ) or N,N‐dimethyl,N′‐2‐chlorobenzylethane‐1,2‐diamine ( L3 ) are reported and characterized by elemental analyses, spectroscopic and molar conductance measurements. The X‐ray structure of complex 1 shows that the central copper atom is placed in a distorted square pyramidal geometry made by acac and diamine chelate in the base and a H2O molecule on the apex. The prepared complexes are fairly soluble in a large number of organic solvents and show positive solvatochromism. Calculations of SMLR (stepwise multiple linear regression) method was utilized to find the best model explaining the observed solvatochromic behavior and showed that among different solvent parameters, donor number (DN) is a dominant factor responsible for the shift in the d‐d absorption band of the complexes to the lower wavenumber with increasing its values. The importance of substituent effect in diamine ligand on the spectral and SMLR measurements is also discussed.  相似文献

Platinum antitumour agents, containing aromatic rings, which are used for targeting DNA in effective therapies for the treatment of cancer. We have synthesized the title metallocomplex with an aromatic ligand and determined its crystal structure. In many cases, complexes of platinum and other metals have a symmetrical structure. In contrast, the platinum(II) complex with pyridine and N‐(9‐anthracenylmethyl)‐1,2‐ethanediamine as ligands (systematic name: cis‐{N‐[(anthracen‐9‐yl)methyl]ethane‐1,2‐diamine‐κ2N ,N ′}bis(pyridine‐κN )platinum(II) dinitrate), [Pt(C5H5N)2(C17H18N2)](NO3)2, is asymmetric. Of the two pyridine ligands, only one is π‐stacked with anthracene, resulting in an asymmetric structure. Moreover, the angle of orientation of each pyridine ligand is variable. Further examination of the packing motif confirms an intermolecular edge‐to‐face interaction.  相似文献

The protonation and ZnII/CuII complexation constants of tripodal polyamine ligand N1‐(2‐aminoethyl)‐N1‐(1H‐imidazol‐4‐ylmethyl)‐ethane‐1,2‐diamine (HL) were determined by potentiometric titration. Three new compounds, i.e. [H3(HL)](ClO4)3 ( 5 ), [Zn(HL)Cl](ClO4) ( 6 ) and {[Zn(L)](ClO4)}n ( 7 ) were obtained by reactions of HL · 4HCl with Zn(ClO4)2 · 6H2O under different reaction pH, and they were compared with the corresponding CuII complexes reported previously. The results indicate that the reaction pH and metal ions have remarkable influence on the formation and structure of the complexes.  相似文献
