A series of novel N‐(substituted benzyl)‐3,5‐bis(benzylidene)‐4‐piperidones 5a – 5o were synthesized with substituted benzylamines as raw materials via a series of Michael addition, Dieckmann condensation, hydrolysis decarboxylation and aldol condensation. The structures were confirmed by 1H NMR, IR, MS techniques and elemental analysis. Assay‐based antiproliferative activity study using leukemic cell lines K562 revealed that most of the title compounds have high effectiveness in inhibiting leukemia K562 cells proliferation, among which the compounds 5g (IC50=7.81 µg·mL−1), 5k (IC50=6.35 µg·mL−1), 5l (IC50=7.20 µg·mL−1), and 5o (IC50=5.79 µg·mL−1) have better inhibition activities than standard 5‐fluorouracil (IC50=8.56 µg·mL−1).  相似文献

目的:建立刺激胰岛素分泌的新型降糖药物(-)-2 (S)-苄基-4-酮-4-(顺式-全氢化异吲哚-2-基)丁酸钙对映体的HPLC拆分方法。方法:采用Sumichiral OA-3300手性柱(250 × 4.6 mm I.D., 5 μm), 柱温35℃,以0.05 mol·L-1醋酸铵的甲醇溶液为流动相,检测波长为210 nm。结果:本品两对映体在22分钟内实现良好分离,分离度达3以上,S-异构体分别在0.028 ~ 5.6 μg mL-1和0.03 ~ 6.0 μg mL-1范围内线性关系良好,回归方程分别为:Y=1.32×103x-2.54 (r=0.9997)和Y=1.15×103x-1.78 (r=0.9998),最低检测限分别为0.15 ng和0.10 ng,方法精密度RSD低于1.0% (n=5)。结论:建立的对映体分离方法可用于本品光学异构体的质量控制。  相似文献

A new energetic material, 4,5‐diacetoxyl‐2‐(dinitromethylene)‐imidazolidine (DADNI), was synthesized by the reaction of 4,5‐dihydroxyl‐2‐(dinitromethylene)‐imidazolidine (DDNI) and acetic anhydride, and characterized by single crystal X‐ray diffraction. Crystal data for DADNI are monoclinic, space group C2/c, a=15.9167(3) Å, b=8.6816(4) Å, c=8.5209(3) Å, β=103.294(9)°, V=1145.9(3) Å3, Z=4, µ=0.150 mm−1, F(000)=600, Dc=1.682 g·cm−3, R1=0.0565 and wR2=0.1649. Thermal decomposition behavior of DADNI was studied and an intensely exothermic process was observed. The kinetic equation of the decomposition reaction is: dα/dT=(1016.64/β)×4α3/4exp(−1.582×105/RT). The critical temperature of thermal explosion is 163.76°C. The specific heat capacity of DADNI was studied with micro‐DSC method and theoretical calculation method. The molar heat capacity is 343.30 J·mol−1·K−1 at 298.15 K. The adiabatic time‐to‐explosion of DADNI was calculated to be 87.7 s.  相似文献

Guanidimium‐4,4‐azo‐1‐hydro‐1,2,4‐triazol‐5‐one (GZTO·H2O) was synthesized from 4‐amino‐1,2,4‐triazol‐5‐one as a starting material by two‐step including oxidation coupling using acid KMnO4 and reaction with (NH2)2CNH·HNO3 (GN) in KOH solution. The single crystal of the title compound was obtained by slow evaporation method at room temperature, and its structure was firstly determined with X‐ray single‐crystal diffractometer. It is a orthorhombic crystal, space group Pbca with cell dimensions of a=1.0459(2) nm, b=1.3584(3) nm, c=1.6103(3) nm, α=90.00(10)°, β=90.00(11)°, γ=90.00(11)°, V=2.2878(8) nm3, Z=8, Dc=1.587 g·cm−3, F(000)=1136, µ=0.132 mm−1, R1=0.0455, wR2=0.1397. The thermal behavior of GZTO·H2O was studied under a non‐isothermal condition by DSC‐TGA method, and its thermal decomposition process can be divided into three stages, and the first stage is an intense exothermic decomposition process. The second stage and the third stage are slow exothermic decomposition processes. The critical temperature of thermal explosion is 237.74°C.  相似文献

Two new complexes: [Cu(TBZ)(bipy)Cl]Cl·H2O ( 1 ) and [Cu(TBZ)(phen)Cl]Cl·H2O ( 2 ) [TBZ=2‐(4′‐thiazolyl)‐ benzimidazole, phen=1,10‐phenanthroline and bipy=2,2′‐bipyridine] have been synthesized and characterized by elemental analysis, molar conductivity, IR, and UV‐vis methods. Complex 2 , structurally characterized by single‐crystal X‐ray crystallography, crystallizes in the monoclinic space group P21/c in a unit cell of a=0.85257(12) nm, b=2.5358(4) nm, c=1.15151(13) nm, β=118.721(8)°, V=2.183.2(5) nm3, Z=4, Dc=1.624 g·cm−3, µ=1.367 mm−1. The complexes, free ligands and chloride copper(II) salt were each tested for their ability to inhibit the growth of two gram‐positive (B. subtilis and S. aureus) and two gram‐negative (Salmonella and E. coli) bacteria. The complexes showed good antibacterial activities against the microorganisms. The interaction between the complexes and calf thymus DNA in aqueous solution was investigated adopting electronic absorption spectroscopy, fluorescence spectroscopy, viscosity measurements and cyclic voltammetry. Results suggest that the two complexes can bind to DNA by intercalative mode. In addition, the result of agarose gel electrophoresis suggested that the complexes can cleave the plasmid DNA at physiological pH and room temperature. Mechanistic studies with different inhibiting reagents reveal that hydroxyl radicals, and a singlet oxygen‐like copper‐oxo species are all involved in the DNA scission process mediated by the complexes.  相似文献

On the basis of the principle of combination of active groups, a series of novel N‐(4‐([2,2′:5′,2′′‐terthiophen]‐5‐yl)‐2‐methylbut‐3‐yn‐2‐yl) benzamide derivatives were designed, synthesized and systematically evaluated for their antiviral activity against tobacco mosaic virus (TMV). The bioassay results showed that most of these compounds displayed good anti‐TMV activity, and some of them exhibited higher antiviral activity than commercial Ningnanmycin. Especially, compound 8e with excellent anti‐TMV activity (inactivation activity, 92.3%/500 µg·mL?1; curative activity, 85.7%/500 µg·mL?1 and protection activity, 64.7%/500 µg·mL?1) emerged as a potential inhibitor of plant virus TMV. Quantitative structure‐activity relationship studies proved that the van der Waals volume (V) and electronic parameter (∑(∑σo+σp) and ∑σm) for the substituent R1 were very important for antiviral activities in this class of compounds.  相似文献

A simple reversed‐phase high performance liquid chromatography (RP‐HPLC) method was developed to determine the level of caffeine and theophylline in human plasma samples. The sample clean‐up step involved the on‐line solid‐phase extraction (SPE) of the analytes from plasma samples into a weak cation monolithic column using a column switching system. Separation was performed on a C18 column (5 µm, 150 mm×4.6 mm) with ultraviolet detection at 274 nm. The mobile phase consisted of methanol‐water (32/68, V/V) under isocratic conditions at a flow rate of 0.6 mL·min−1. The measured concentration of caffeine and theophylline showed a good linear relationship over the concentrations range, 0.1–80.0 µg·mL−1. The absolute recoveries ranged from 77.10% to 85.39%, and the inter‐day and intra‐day relative standard deviations (RSD) were all less than 5%. This method avoids a tedious pretreatment and provides an economic, repeatable and effective method for assaying trace drugs in biological samples.  相似文献

On the line of a previous work on the spectral properties of some of heteroaryl chalcone, the effect of medium acidity and photoreactivity of 3‐(4‐dimethylamino‐phenyl)‐1‐(2,5‐dimethyl‐thiophen‐3‐yl)‐propenone (DDTP) has been investigated in dimethylformamide and in chloromethane solvents such as methylenechloride, chloroform and carbon tetrachloride. The dye solution (ca. 5×10−4 mol·L−1 in DMF) gives a good laser emission in the range 470–560 nm with emission maximum at 515 nm upon pumping by nitrogen laser (λex=337.1 nm). The laser parameters such as gain coefficient (α), emission cross section (δe) and half life energy (E1/2) at maximum laser emission are also determined.  相似文献

A short ionic liquids (ILs)‐based monolithic cartridge was prepared and used as the selective extraction sorbent. After the material was evaluated by field emission‐scanning electron microscopy (FE‐SEM), a new approach for the extraction and determination of quercetin and myricetin from Chamaecyparis obtusa (C. obtusa) by using ILs‐based, monolithic cartridge system was developed. Chromatographic analysis was conducted on a C18 column with UV detection at 372 nm, an eluting solution consisting of acetonitrile‐water (25/75,V/V) as the mobile phase, and a flow rate of 0.7 mL·min−1. A good linear relationship was demonstrated when the concentrations of quercetin and myricetin were in the range of 0.5–100.0 µg·mL−1. The recoveries ranged from 101.6% to 104.6% and the inter‐ and intra‐day relative standard deviations (RSD) were less than 5.0%. This method effectively removed the impurities and avoided tedious pretreatment. It provided a fast, economic and effective method for assaying trace drugs from natural plants.  相似文献

4,4′‐Bis(3‐N‐methoxyformyl thioureido)‐diphenyloxide was prepared via reaction of 4,4′‐diaminodiphenyl alter with potassium sulfocyanate and ethyl chloroacetate in ethyl acetate. The single crystal of the title compound was cultured by slow evaporation method at room temperature. The crystal structure was determined with X‐ray diffractometer. It is a monoclinic crystal, space group C2/c with a=0.95911(19) nm, b=0.75922(15) nm, c=2.7161(5) nm, α=90°, β=97.675 (3) °, γ=90°, V=1.9601(7) nm3, Z=4, Dc=1.472 g·cm−3, F(000) =904, µ=0.311 cm−1, R1=0.0367, wR2=0.1408. The specific heat capacity of the title compound was determined with continuous Cp mode of mircocalorimeter. The thermal behavior of the title compound was studied under a non‐isothermal condition by DSC method.  相似文献

本文首次合成标题化合物N-(4-甲基苯甲酰氨基)-N’-[5-(2-三氟甲基苯基)-2-呋喃甲酰硫脲。化合物(C21H16F3N3O3S, Mr = 447.43)单晶经测定为单斜晶体,空间群为P -1。在晶体中,存在一些分子内和分子间的相互作用,分子间还有C—H···π 的相互作用,这可能导致晶体更稳定的原因。目标产物的结构经IR, H NMR和元素分析测定确证。初步生物活性测试表明,部分化合物对棉花枯萎病、黄瓜灰霉病、苹果轮纹病和棉花炭疽病有较好的选择性杀菌活性;部分目标化合物有较好的除草活性。  相似文献

The β‐sitosterol imprinted polymer was prepared for selective extraction and analysis of β‐sitosterol from Oldenlandia diffusa (O. diffusa) followed by HPLC with UV detection. The imprinted polymers show high affinity and selectivity to β‐sitosterol. Using this molecularly imprinted polymer (MIP) cartridge as solid‐phase extraction (SPE) material, the interferences could be quickly washed out and β‐sitosterol was selectively retained and enriched. HPLC analysis method was used to evaluate the characteristics of this MIP material. At the condition of mobile phase composed of MeOH/H2O/H3PO4 (99/1/0.1, V/V/V, pH=6.0) and the flow rate of 1.0 mL·min?1, a good linear relationship was demonstrated when the concentrations of β‐sitosterol were in the range of 0.50–100.0 µg·mL?1. The recoveries ranged from 75.3% to 86.5% and the inter‐day and intra‐day relative standard deviations were less than 5%. This developed HPLC method was proved to be acceptable for extraction of β‐sitosterol from O. diffusa.  相似文献

A novel amphiphilic silica‐based monolithic column having surface‐bound octanoyl‐aminopropyl moieties was successfully prepared by a one‐step in situ derivatization process. As expected, the amphiphilic monolithic column exhibited RP chromatographic behavior toward non‐polar solutes (e.g., alkyl benzenes) with high column performance. As the pH of the buffer inside the column increases, the EOF changed from −2.65×10−8m2 V−1s−1 at pH 3.0 to 1.20×10−8 m2 V−1s−1 at pH 8.0 with the reversion of EOF at about pH 6.4. Using acidic mobile phase, five aromatic acids can be efficiently separated in less than 6 min under co‐EOF conditions. For basic compounds, symmetrical peaks were obtained due to the existence of hydrophilic acyl amide group, which can effectively minimize the adsorption of the positively charged basic analyte to the silica‐based surface of the capillary column.  相似文献

Twenty nine novel N‐4‐methyl‐1,2,3‐thiadiazole‐5‐carbonyl‐N′‐phenyl ureas were designed and synthesized, and their structures were confirmed by proton nuclear magnetic resonance (1H NMR), infra red spectroscopy (IR) and high‐resolution mass spectroscopy (HRMS). Compounds V‐9 , V‐11 , V‐12 , V‐15 , V‐19 , V‐21 , V‐22 and V‐24 exhibit excellent activity against Culex pipiens pallens. Compounds V‐12 and V‐22 present good insecticidal activity against Plutella xylostella L. Their median lethal concentrations (LC50) are 164.15 and 89.69 mg·L?1, respectively. Compound V‐11 also has potential wide spectrum of fungicide activity. Its median effective concentrations (EC50) detected from 3.82 µg·mL?1 against Physalospora piricola to 31.60 µg·mL?1 against Cercospora arachidicola. Compounds V‐15 and V‐24 show outstanding induction activities as same as positive controls TDL and ningnanmycin, furthermore V‐24 has the highest induction activity of 41.85%±4.43%. To elucidate the structure activity relationship in these compounds, a 3D‐QSAR model has been built. The established model showed a reliable predicting ability with q2 values of 0.643 and r2 values of 0.982.  相似文献

The electrochemical, UV/Vis–NIR absorption, and emission‐spectroscopic features of (TBA+)( 1 ) and the corresponding neutral complex 1 were investigated (TBA+=tetrabutylammonium; 1 =[AuIII(Pyr,H‐edt)2]; Pyr,H‐edt2−=pyren‐1‐yl‐ethylene‐1,2‐dithiolato). The intense electrochromic NIR absorption (λmax=1432 nm; ε=13000 M −1 cm−1 in CH2Cl2) and the potential‐controlled visible emission in the range 400–500 nm, the energy of which depends on the charge of the complex, were interpreted on the grounds of time‐dependent DFT calculations carried out on the cis and trans isomers of 1 , 1 , and 1 2−. In addition, to evaluate the nonlinear optical properties of 1 x (x=0, 1), first static hyperpolarizability values βtot were calculated (βtot=78×10−30 and 212×10−30 esu for the cis isomer of 1 and 1 , respectively) and compared to those of differently substituted [Au(Ar,H‐edt)2]x gold dithiolenes [Ar=naphth‐2‐yl ( 2 ), phenyl ( 3 ); x=0, 1].  相似文献

A facile,rapid and sensitive spectrophotometric method was developed for the determination of carbaryl in itsformulations,water and grain(rice and wheat)samples with newly synthesised reagent.The proposed method wasbased on the alkaline hydrolysis of carbaryl pesticide and thus resulted 1-naphthol was coupled with diazotised4,4'-methylene-bis-m-nitroaniline in basic medium(pH 9)to give red colored product having λ_(max)480 nm for dia-zonium method(DM)or 1-naphthol reacts with 4,4'-methylene-bis-m-nitroaniline in the presence of oxidising agentpotassium dichromate(K_2Cr_2O_7)to give red colored product having λ_(max)510 nm for oxidation method(OM).Theformation of colored derivatives with the coupling agent is instantaneous and stable for 48 h(DM)and 32 h(OM)respectively.Beer's law was obeyed in the concentration range of 0.2—10.0 μg·mL~(-1)for DM and 0.2—0,150μg·mL~(-1)for OM.The proposed methods are sensitive,easy to operate and permitted for the determination of car-baryl with detection limits of 0.028 μg·mL~(-1)for DM and 0.024 μg·mL~(-1)for OM respectively.The experimentalresults indicate that the procedure can eliminate the fundamental interferences caused by other pesticides andnon-target ions,which made the methods more sensitive and selective.The method was applicable to the determina-tion of carbaryl residue in water and food grain samples up to μg level.  相似文献

The banned addition of psychiatric drugs such as phenothiazines to animal feed and foodstuffs increases the risk of human organ lesion. Phenothiazines usually exhibit weak native fluorescence and can be oxidized to strongly fluorescent compounds. In this study, a novel, sensitive and convenient method of HPLC‐fluorescence detection based on post‐column on‐line oxidizing with lead dioxide solid‐phase reactor has been developed for simultaneous determination of three banned psychotropic drugs, promethazine, chlorpromazine and thioridazine. Three compounds were successfully separated on an Agilent TC‐C18 column with mobile phase of acetonitrile (A) and water (B), both containing 0.5% (v/v) formic acid. A gradient elution was programmed and fluorimetric detection was performed at λex/λem of 332/373 nm for promethazine, 340/380 nm for chlorpromazine and 352/432 nm for thioridazine. The calibration graphs gave good linearity over the concentration ranges of 30.0–4976.4 µg/L for promethazine, 2.0–2153.2 µg/L for chlorpromazine, and 15.0–3088.0 µg/L for thioridazine, and correlation coefficients (r) were ≥0.995. The method was applied to the determination of phenothiazines in pig feed and pig tissue, and the average spiked recoveries were in the range 69.1–115.4%. [(n-Bu)2Sn[{4-ClC6H4C(O)NHO}2] (DBDCT) 是课题组自主设计合成的一种新型芳香异羟肟酸二丁基锡化合物(已获国际国内发明专利),有较高的体内和体外抗肿瘤活性,小鼠急毒实验揭示其具有较低的毒性作用,初步动物实验提示DBDCT还具有升高白细胞的功能,在肿瘤化疗治疗中将产生重要的影响。本文首次建立了HPLC法测定化合物在血浆中的动力学参数。用甲醇直接沉淀血浆蛋白,乙酰苯胺为内标, Diamonsil ODS(4.6 mm × 200 mm, 5 μm)色谱柱,甲醇:0.5%三氟乙酸水溶液(30:70,pH 3.0,v/v)为流动相,检测波长238 nm。方法在0.1~25 µg·mLl-1范围内线性关系良好(r = 0.9992),定量限和检测限分别为50 和10 ng·mL-1。该方法用来测定单次静脉注射不同剂量(2,5,12mg·kg-1) DBDCT后大鼠体内的浓度-时间曲线,并采用3p97软件对动力学参数和房室模型进行估计,结果表明DBDCT在大鼠体内的动力学符合二室模型,方法简便快速,专属性好,其动力学研究中的应用为制剂的质量控制和临床前动物合理用药以及临床研究提供了实验基础。  相似文献

A cadmium complex bis(benzyltriethylammonium) bis(1,3‐dithiole‐2‐thione‐4,5‐dithiolato)‐cadmium(II) ((TEBA)2[Cd(DMIT)2]) has been synthesized and its crystal structure has been determined by means of X‐ray single‐crystal diffraction. The central cadmium(II) ion coordinates with two DMIT, which constructed a distorted tetrahedron environment. Its third‐order nonlinear optical properties have been studied using Z‐scan technique with 20 ps pulses at wavelength 1064 nm. Its third‐order nonlinear susceptibility χ(3) value was determined to be 1.24 × 10−19 m2 V−2, the figure of merit, χ(3)/α0, was estimated to be 2.64 × 10−20 m3 V−2. An alternating narrow bandgap conjugated copolymer (PICZ‐DTBT, Eg = 1.83 eV) derived from 5,11‐di(9‐heptadecanyl)indolo[3,2‐b]carbazole and 4,7‐di(thieno[3,2‐b]thien‐2‐yl)‐2,1,3‐benzothiadiazole (DTBT), was prepared by the palladium‐catalyzed Suzuki coupling reaction. The resultant polymer absorbs light from 350–690 nm, exhibits two absorbance peaks at around 420 and 570 nm and has good solution processibility and thermal stability. The highest occupied molecular orbital (HOMO) energy level and lowest unoccupied molecular orbital (LUMO) level of the copolymer determined by cyclic voltammetry were about −5.18 and −3.35 eV, respectively. Prototype bulk heterojunction photovoltaic cells from solid‐state composite films based on PICZ‐DTBT and [6,6]‐phenyl‐C71 butyric acid methyl ester (PC71BM), show power conversion efficiencies up to 2.4% under 80 mW · cm−2 illumination (AM1.5) with an open‐circuit voltage of Voc = 0.75 V, a short current density of Jsc = 6.02 mA · cm−2, and a fill factor of 42%. This indicates that the copolymer PICZ‐DTBT is a viable electron donor material for polymeric solar cells.
