Ganglioside GM3 and its derivatives have many important biological functions. Using diethyl phosphite protected sialic acid as glycosyl donor and 3,2′,3′,4′-unprotected lactose as glycosyl acceptor, the sialic acid-containing trisaccharide was assembled with excellent anomeric stereoselectivity. The trisaccharide was further coupled with ceramide precursor to yield sphingosine 1. Based on this key intermediate, two different series of N-modified GM3 analogues with modifications on either the nitrogen of sialic acid residue or the nitrogen of ceramide moiety and GM3 itself were synthesized smoothly.  相似文献

Sialic acid linkages on N-glycans were distinguished by MALDI-TOF MS after two steps derivatization by dimethylamine and ammonium hydroxide. By using this method, more than 20 kinds of sialic acid with detailed linkage information were detected on A549 cells.  相似文献

The total synthesis of ganglioside GP3, which is found in the starfish Asterina pectinifera, has been accomplished through stereoselective and effective glycosylation reactions. The sialic acid embedded octasaccharide moiety of the target compound was constructed by [4+4] convergent coupling. A tetrasaccharyl donor and acceptor that contained internal sialic acid residues were synthesized with an orthogonally protected N‐Troc sialic acid donor as the key common synthetic unit, and they underwent highly stereoselective glycosidation. The resulting sialosides were subsequently transformed into reactive glycosyl acceptors. [4+4] coupling furnished the octasaccharide framework in 91 % yield as a single stereoisomer. Final conjugation of the octasaccharyl donor and glucosyl ceramide acceptor produced the protected target compound in high yield, which underwent global deprotection to successfully deliver ganglioside GP3.  相似文献

In this work, we report the spectral modulations of a intramolecular charge transfer (ICT) molecule ethyl ester of N,N-dimethylaminonaphthyl-(acrylic)-acid (EDMANA) when encapsulated in the water and N,N-dimethylformamide (DMF) solution of β-CD nanocavities. From the nature of the Benesi-Hildebrand (B-H) plots, the stoichiometry of the host guest inclusion complexes are found to be 1:1 in water β-CD solution and both 1:1 and 1:2 in DMF β-CD solution. The preferential location and difference in orientation of EDMANA molecule inside the β-CD cavity has been accessed by analysis of the effect of acid and metal cation Ni2+ on the spectral characteristics in both the media. In case of 1:1 complex, the polar donor group prefers to expose to bulk aqueous phase capable of binding with H+ and Ni2+ ions and the acceptor to the hydrophobic interior. On the other hand, the acceptor group remains exposed to the non-polar bulk phase and the donor group is orientated preferentially inside the non-polar core in 1:2 inclusion complexes.  相似文献

The synthesis of the putative prion-associated heparan sulfate tetrasaccharide containing two d-glucuronic acid units is reported. Key to the synthesis were the differentiation of the N-acetylated and N-unsubstituted glucosamine units, the high-yielding glucosylation at O-4 of an N-acetyl-d-glucosamine derivative and the α-selective glycosylation of the 4′-OH group of a β-d-GlcpA-(1→4)-d-GlcpNAc disaccharide building block with a disaccharide thioglycoside donor.  相似文献

The combination of the 5-N-tert-butoxycarbonyl (Boc) group of laurylthio sialoside and cyclopentyl methyl ether (CPME) as a solvent enhanced the reactivity and α-selectivity of the sialyl donor during sialylation. Selective deprotection of the N-Boc group of sialoside, including an acid-sensitive isopropylidene function, was successfully achieved by Yb(OTf)3-SiO2. Transformation of N,N-Ac,Boc into an N-acetylglycolyl group of sialoglycoside was easily performed via selective N-deacylation of the mixed Ac-N-Boc carbamate, subsequent Boc group removal, and acylation.  相似文献

Sialic acid (N-acetylneuraminic acid) is a carbohydrate that possess a nine carbon backbone, and it is often found at the termini of glycoconjugates in biological systems. Because of this prominence many syntheses have reported routes to sialic acid and many of its derivatives. Most of these compounds retain the endocyclic oxygen atom that becomes part of the ketal glycosidic linkage that joins sialic acid to the penultimate residue in the glycoconjugate. With respect to carba-sialic acid (replacement of the ring oxygen atom with a methylene group) a single synthesis has been reported (Ogawa et al. (Carbohydr. Res., 1995, 269, 53–78) in 30 steps and 0.5% yield. The current report details a robust synthesis of 6a-carba-α-d-sialic acid that involves 18 steps and give a 5% yield using d-quinic acid as the starting material.  相似文献

N-Methyl-, N-n-butyl-, N-t-butylperfluoroarylamines undergo nitrosation with nitrous acid to give the corresponding N-nitroso derivatives. Perfluoroaryl groups were selected from the benzene, indane, biphenyl, naphthalene and pyridine series. According to 1H and 19F NMR spectra, N-nitroso-N-methyl derivatives of polyfluoroarenes consist of E and Z isomers with the former prevailing. The more bulky n-butyl group promotes an increase in the formation of Z isomers. Only Z isomers have been obtained from N-t-butyl derivatives of perfluorinated 4-toluidine and 4-aminopyridine. The structure of the Z isomer of N-nitroso-N-methylperfluoro-4-toluidine is confirmed by X-ray data.  相似文献

The synthesis of four deuterated sialic acids and their 1,7-lactones has been performed in two ways, one based on sialic acid classical chemistry, and the other involving a direct exchange of the unlabeled acyl group of N-acetylneuraminic acid with a labeled one mediated by a perfluorinated amide. A ruthenium-catalyzed reduction of N-alkoxy- and N-hydroxyamides was found to afford corresponding amides in good to high yields. A simple RuCl3/Zn-Cu/alcohol system, without the addition of any other ligands, exhibited a high catalytic activity, and therefore the present reaction does not require a stoichiometric amount of metals or metal complexes as reductants. When β-substituted-α,β-unsaturated N-methoxyamides were employed as substrates, concurrent hydrogenation of the olefin moiety proceeded slowly with deprotection of the methoxy group. In the reduction of N-hydroxyamides, the alcoholic solvent was found to function as a hydrogen donor.  相似文献

Two histidinehydroxamic acid derivatives (N-methyl-histidinehydroxamic acid, N-Me-Hisha and Z-histidinehydroxamic acid, Z-Hisha) have been synthesized and their complexation with Cu2+-, Ni2+- and Zn2+-ions has been studied by using pH-potentiometric, UV–Vis, CD, 1H NMR, EPR and ESI-MS methods. Both of the two new derivatives contain one donor atom less compared to the histidinehydroxamic acid (Hisha). In the case of N-Me-Hisha the hydroxamate-N as donor is eliminated, while the coordination of the amino-N of Z-Hisha is not possible at all.  相似文献

In this paper, by introducing [1,2,5]thiadiazolo[3,4-c]pyridine (PT) as an auxiliary acceptor into the molecular design of organic sensitizers, we have synthesized four new dyes (PT1PT4) for dye-sensitized solar cells (DSSCs) with triphenylamine or N,N-diphenylthiophen-2-amine as the donor units and thiophene or benzene as the π-bridges, respectively. All the structures, optical and electrochemical properties were fully characterized. Nanocrystalline TiO2 dye-sensitized solar cells were also fabricated using these dyes. Among them, PT2-based DSSCs showed the highest overall conversion efficiency of 6.11% with Voc=668 mV, Jsc=12.61 mA cm−2 and a fill factor (FF)=0.74 after a chenodeoxycholic acid (CDCA) treatment under standard illumination condition (100 mW cm−2 simulated AM 1.5 solar light).  相似文献

The enthalpies of solution of N-acetyl-N′-methylglycinamide, N-acetyl-N′-methyl-l-α-alaninamide, N-acetyl-N′-methyl-l-α-leucinamide and N-acetyl-N′-methyl-l-α-serinamide have been measured in water and in aqueous urea solutions with molalities from 0.25 to 3.0 mol kg−1 at 298.15 K. From these data the standard dissolution enthalpies of amides in aqueous urea solutions have been determined. The results have been treated according to McMillan-Mayer's theory in order to obtain the enthalpic coefficients of the interactions between amino acid derivatives and urea molecules. The obtained parameters were compared with the hydrophobic scale for the amino acid side chains.  相似文献

Highly α‐selective sialylation of sialic acid N‐phenyltrifluoroacetimidate with various galactose and lactose acceptors has been achieved by introducing the C‐5 N‐phthalyl group on the donor. The “fixed dipole effect” of the N‐phthalyl group was proposed to explain the high reactivity and α‐selectivity. The microfluidic system was applied to the present α‐sialylation, which is amenable to large‐scale synthesis. The N‐phthalyl group was removed by treatment with methylhydrazine acetate, for which protocol can be readily applied to the synthesis of a variety of sialic acid‐containing oligosaccharides.   相似文献

Formation of peptide thioesters, based on an N to S acyl shift mediated by an auxiliary, N-4,5-dimethoxy-2-mercaptobenzyl (Dmmb) group, under acidic conditions, is described. The protected peptide was assembled on a hydroxymethylphenylacetamidomethyl resin via an N-Dmmb-amino acid residue according to standard Fmoc solid-phase peptide synthesis following treatment with trifluoroacetic acid. The peptide α-thioester was released from the resin by reaction with 2-mercaptoethanesulfonic acid in the presence of N,N-diisopropylethylamine.  相似文献

Simple and efficient photoacid generators (PAGs) for carboxylic and sulfonic acids based on N-acyl-N-phenylhydroxylamines have been demonstrated. Irradiation of o-carboxylates and thermally rearranged o-arenesulfonates of N-acyl-N-phenylhydroxylamines using UV light (≥254 nm) in aqueous methanolic solution resulted in efficient generation of carboxylic and sulfonic acids, respectively. The carboxylic acid generation ability of N-acyl-N-phenylhydroxylamines was found to be dependent on their N-acyl substituents. Further, polymer bearing o-arenesulfonates of N-acyl-N-phenylhydroxylamine was synthesized and demonstrated as PAG for sulfonic acids.  相似文献

A new class of organocatalysts is reported that incorporates an N-sulfinyl amide in place of the carboxylic acid of proline to serve as a hydrogen bond donor, chiral directing group, and solubilizing element. The successful application of this type of catalyst to the asymmetric aldol reaction of acetone and aryl aldehydes, for which proline performs poorly, is also described.  相似文献

The tetraruthenium cluster complex [Ru444-dmpu)(CO)10], H2dmpu = N,N′-bis(6-methylpyrid-2-yl)urea, has been prepared by treating [Ru3(CO)12] with H2dmpu in toluene at reflux temperature. An X-ray diffraction study has determined that this cluster has a butterfly metallic skeleton hold up by a doubly-deprotonated N,N′-bis(6-methylpyrid-2-yl)urea ligand (dmpu). This ligand has the pyridine N atoms attached to the wing-tip Ru atoms and the amido N atoms spanning Ru-Ru wing-edges, in such a way that the cluster has C2 symmetry. The donor atoms of doubly-deprotonated N,N′-dipyrid-2-ylureas seem to be appropriately arranged to hold butterfly tetranuclear clusters.  相似文献

Two pentasaccharides containing a common tetrasaccharide (lacto-N-neotetraose) core, and d-galactosamine and N-acetyl neuraminic acid in the non-reducing ends, respectively, corresponding to the lipooligosaccharides of Neisseria gonorrhoeae and Neisseria meningitidis were synthesized in a very concise manner from a common trisaccharide derivative using minimum number of steps. Thioglycosides and glycosyl trichloroacetimidate have been used as glycosyl donors for glycosylations and yields were excellent in every step.  相似文献

N-Benzylamides were debenzylated efficiently with 4 equiv. of p-TsOH in refluxing toluene. Good to quantitative yields of the desired primary amides were obtained within 2-4 h from a wide variety of N-2,4-dimethoxybenzylamides. N-4-Methoxylbenzyl amides and N-benzylamides were also debenzylated cleanly. In the case of N-2,4-dimethoxylbenzylamides, selective N-debenzylation was possible in the presence of N-Fmoc, N-t-BOC or N-trityl-protection. Protected amino acid amides survived these conditions without any detectable epimerization.  相似文献
