作为一种新型的辐射诱变源, 重离子束在辐射诱变育种中的优越性已经显现。 在此基础上综述了重离子束用于微生物诱变育种的基本原理、 独特优点、 所取得的成果及研究进展, 并对其在新型生物能源开发中的潜力进行了展望。As a new radiation source, heavy ion beams have demonstrated the outstanding advantage in mutation breeding. Based on this background, the basic principal and unique peculiarity of heavy ion beams, the achievement and the progress in the research of microbial mutation breeding are reviewed in the paper. The potential application of heavy ion beams to new biological energy is also prospected.  相似文献

载能重离子与高能中子在材料中能够产生相似的级联碰撞损伤,加之重离子具有大的离位损伤截面和在材料样品中低的感生放射性,载能重离子束成为模拟先进核能装置内部结构材料辐照损伤的重要手段。HIRFL能区的重离子在结构材料中的射程一般远大于晶粒尺寸,因此能够产生材料体损伤,借助小样品技术可以获得材料力学性能变化(尤其辐照脆化)的有用信息,为探讨材料辐照损伤微结构和宏观力学性能变化的关联提供了重要条件。本文简要介绍了近年来我们基于HIRFL高能离子束开展的聚变堆候选材料辐照损伤的研究,包括低活化钢的辐照脆化行为、氧化物弥散强化(ODS)铁素体钢的结构优化对于抗辐照性能的影响、不同载能粒子辐照条件下铁素体/马氏体钢的辐照肿胀数据的关联,以及高能重离子辐照的钨材料中氢同位素的滞留行为。研究表明,结合特殊的测试技术及数据分析方法,高能重离子可作为核能结构材料辐照损伤研究及评估的有效手段。Because of the similarity in cascade damage structure in materials produced by energetic heavy ions and by fast neutrons, and the high displacement rate and low induced radioactivity of samples by heavy ions, heavy ion beam becomes an important tool to simulate radiation damage by energetic neutrons in materials in advanced nuclear energy systems. The ranges of heavy ions provided by HIRFL (Heavy Ion Research Facility in Lanzhou) are generally much larger than the mean dimensions of grains in alloys candidate to advanced nuclear reactors, and is capable of producing radiation damage in bulk scale. It therefore makes possible the evaluation of change of mechanical properties including the radiation induced embrittlement from the irradiated specimens by using miniaturized specimen techniques. In the present paper, we provide an introduction of our recent studies of radiation damage of materials candidate to future fusion reactors by utilizing heavy ion beams in HIRFL.The studies include issues as follows:ductility loss of RAFM steels causes by high-energy Ne ions, impact of oxide dispersoids on the radiation resistance of ODS ferritic steels, correlation of void swelling of ferritic/martensitic steels under different particle irradiation, and behavior of deuterium retention in tungsten under irradiation with high-energy heavy ions. The results show that high-energy heavy ions can be used as a tool to efficiently investigate or evaluate radiation damage in structure materials if combined with some special test techniques and data analysis.  相似文献

综述了离子束辐照拟南芥种子生物学效应、 生长发育变化、 向重力性等研究的最新进展。 阐释了离子束辐照拟南芥染色体DNA碱基变化、 DNA断裂或损伤、 染色体重组、 突变遗传性等分子机理。 探讨了离子束介导外源基因转化拟南芥的有效性和机理。 同时展望了辐照拟南芥分子机理研究中的辐射原初效应传递、 信号转导等其他机理研究及重离子辐射生物学效应的应用前景。 Newly research progresses were summarized in effect of ion beams on seed surface, biological effect, growth, development, gravitropism and so on. Furthermore, mutation molecular mechanism of Arabidopsis thaliana was discussed, for example, alteration of DNA bases, DNA damage, chromosomal recombination, characteristics of mutant transmissibility, etc. Meanwhile, the achievements of transferring extraneous gene to Arabidopsis thaliana by ion beams were reviewed in the paper. 考察不同剂量重离子束对北里链霉菌的致死及诱变效应, 可确定最有利于筛选高产菌株的重离子束辐照剂量。 利用不同剂量的重离子束辐照北里链霉菌孢子, 统计了存活率、 致死率、 正负突变率。 结果发现, 在5 Gy重离子辐照时北里链霉菌出现较高致死率, 其后随剂量升高, 致死率变化较平缓。 各辐照剂量下正负突变率相比较, 40 Gy时正突变率最高, 负突变率相对较低, 存活率为0.92%。 因此确定40 Gy是对北里链霉菌高产菌株筛选最有利的辐照剂量。 To define the optimum dose of heavy ion beams for selecting high productive strains, we should study mortality and mutation effects of Streptomyces kitasatoensis irradiated by heavy ion beams in diffe rent doses. In this research, spores of Streptomyces kitasatoensis were irradiated by heavy ion beams with different doses. And survival rate, mortality rate, positive mutation and negative mutation were analyzed statistically. The results showed that high mortality rate appeared from 5 Gy and then the mortality rate curve became gently. Compared the positive and negative mutations in different doses, highest positive mutation was obtained in 40 Gy, while the negative mutation was lower in this dose, and the survival rate was 0.92%. So we defined that optimum dose of heavy ions radiation for Streptomyces kitasatoensis selection was 40 Gy in this experiment.  相似文献

核辐照应用于中药领域的研究得到人们广泛关注。综述了核辐照在中草药的栽培、育种和消毒等方面的应用, 指出辐照诱变技术与生物技术相结合将为提高细胞突变率和加快中草药遗传改良开辟广阔前景。The application of nuclear irradiation in the field of traditional Chinese medicine has received much attention. In this paper we reviewed the application of nuclear radiation on the cultivation, breeding and disinfection of traditional Chinese medicine, and pointed out that the combination of radiation induced mutagenesis and biological technology would promise broad prospects for increasing the cellular mutation rate and speeding up the genetic improvement of traditional Chinese medicine.  相似文献

定向诱导基因组局部突变技术(Targeting Induced Local Lesions IN Genomes, 简称TILLINC)是一种全新的、高通量和低成本反向遗传学研究方法。近年来, 随着突变筛选技术的革新, TILLING技术平台日趋多元化, 使得TILLING技术的操作更为简单﹑快速, 并广泛应用于作物育种研究领域。简要介绍了TILLING技术平台的最新发展动态, 并初步探讨了将辐射诱变处理与TILLING高通量筛选相结合在诱变育种中的应用前景。To investigate the M1 biological effects of heavy ions irradiation on Lycopersicon esculentum Mill., its seeds were irradiated by 12C6+heavy ions (80 MeV/u) with the dosages of 30, 60, 90, 120 and 160 Gy respectively . The results showed that with doses increased gradually, germination rate and seedling rate of Lycopersicon esculentum Mill. were decreased, and the latter was lower than the former, mainly due to the inhibition of root growth. The irradiation increased the content of MDA and proline evidently, showing irradiation could damage biomembrane, and also decreased the activities of POD and SOD with distinct inhibition pattern. However, the low dose and high dose irradiation promoted APX activity, illustrating APX was induced to protect irradiation injury. In brief, exposure to 12C6+ heavy ions had obvious injury effects on the seeds of Lycopersicon esculentum Mill.. Heavy ions irradiation damaged biomembrane, inhibited activities of enzymes, and finally inhibited the growth of the first generation of these seeds.  相似文献

在基于被动式束流配送系统的分层适形重离子治疗当中,需要利用微型脊形过滤器(mini ridge filter,mini-RF)将单能重离子束Bragg峰展宽为峰区近似高斯分布的微小展宽峰(mini spread-out Bragg peak,mini-SOBP),从而达到减少照射分层数及照射时间的目的。采用较宽的mini-SOBP可以有效减少照射分层数,但会增大展宽Bragg峰(SOBP)远端剂量跌落距离,增加对靶区后方正常组织或危及器官的辐照剂量。这一问题可以通过双mini-SOBP的组合照射方法来解决。使用2种mini-RF对单能重离子束Bragg峰略微展宽得到2种半高宽(FWHM)且剂量分布近似高斯分布的mini-SOBP,通过基于放射生物学模型的剂量优化,证实了在SOBP平顶区按生物有效剂量均匀和物理吸收剂量均匀的展宽情况下,双mini-SOBP组合照射方法均可以在减少照射分层数的同时较大幅度地减小SOBP远端剂量跌落距离。In layer-stacking conformal heavy-ion therapy based on passive beam delivery system,it is necessary to minimize the layer numbers and reduce irradiation time for layer-stacking conformal heavy-ion therapy.Gaussian shaped mini spread-out Bragg peaks (mini-SOBP) were generated by mini ridge filters (mini-RF) for monoenergetic heavy ion beams.It is effective to minimize the layer number by using mini-SOBPs with the bigger full width at the half maximum (FWHM),but in this way the distal dose fall-off distance of a spread-out Bragg peak (SOBP) will be enlarged,increasing the radiation damage to normal tissue or organ at risk behind the target volume.This issue could be solved by using mini-SOBPs based combinatorial irradiation method.In this study,Gaussian shaped mini-SOBPs with two different FWHMs were generated by two different mini-RFs for monoenergetic heavy ion beams.Based on radiobiological model calculations and dose optimizations,the mini-SOBPs based combinatorial irradiation method was confirmed to reduce the distal dose fall-off distances of SOBPs while minimizing the layer numbers for layer-stacking conformal heavy-ion therapy.  相似文献

应用大剂量重离子束12C6+ 对菌株H3001 进行二次辐照选育,对初选获得的高产柠檬酸菌株进行摇瓶发酵试验及10~ 100 L 中试发酵罐试验,采用酸碱中和法测定发酵液中柠檬酸的含量。结果表明:当二次重离子12C6+剂量为857.8 Gy 时,致死率和正突变率达到最大值,分别为94.5% 和8%。通过摇瓶发酵试验,最终获得一株高产柠檬酸菌株hw317,控制该菌株发酵周期为60 h,柠檬酸酸度能达到19.2±0.2%。Heavy 12C6+ ion beams in various high doses were employed to irradiate H3001 strain for screening Aspergillus niger strain for hyper citric acid production. Three high-yield strains were obtained after shaker fermentation test. Among the three strains, the strain hw317 was implemented shaker fermentation for stability test and 10~100 L pilot fermentation tank for citric acid productive maximization. Acid-base neutralization method was applied to determinate the content of citric acid in fermented liquid. The results showed that: when the secondary heavy ion 12C6+ dose was 857.8 Gy, both of the fatality rate (94.5%) and the positive mutation rate (8%) were highest. Through the shaker fermentation tests and 10 ~ 100 L pilot fermentation test, one strain hw317 was screened and obtained for hyper citric acid production. Consequently, the final citric acid acidity can reach up to 19.2±0.2% with controlling fermentation cycle for 60 h.  相似文献

在2000—2005年将杂交技术和重离子辐射相结合, 采用55 MeV/u的 40Ar17+离子对马铃薯杂交种子和微型薯进行了不同剂量的贯穿辐射或离子注入处理。 结果表明, 对马铃薯微型薯进行重离子辐射能显著提高块茎的产量。 而杂交技术和重离子辐射相结合是一种更高效的育种方式, 不仅能提高块茎的增产幅度, 还能改善其加工品质。 微型薯重离子辐射的最佳剂量为60 Gy, 杂交种子辐射的适宜剂量范围在60—120 Gy之间。 通过对辐射后代的多年选育, 获得了几个产量显著高于对照、 品质优良的马铃薯新品系。 The effect of 55 MeV/u 40Ar17+ irradiation with different doses on hybrid seeds and micro tubers of potatoes were studied from year 2000 to 2005. The results showed that the yields of potatoes irradiated by heavy ions grew significantly. This new mutation method combining cross breeding technique with heavy ion irradiation is more effective, which could not only increase the yield of potatoes, but also improve their quality. The optimal mutagenic dose was 60 Gy to micro tubers of potatoes, 60—120 Gy to hybrid seeds, respectively. Finally, several new lines which have higher yield and better qualities were obtained through multi generation screening from the offspring of irradiated potatoes.  相似文献

用高能(500MeV)Ne离子束对GaAs和InP进行了辐照,用MonteCarlo模拟、正电子湮没谱学以及红外光谱研究了辐照产生的缺陷特性.结果表明,在未辐照的样品中存在单空位,经辐照后,可在样品中产生单空位;当剂量较大时,还会形成双空位甚至尺寸较大的空洞.红外光谱测量发现,在辐照后的GaAs样品中有非晶区形成.此外,辐照在样品中还产生了反位缺陷GaAs和InP以及受主杂质ZnIn.对经1014ions/cm2剂量辐照的InP进行了光学实验,在辐照后的InP材料中发现了亚稳态中心. Both GaAs and InP were irradiated by high energy (500 MeV) Ne ions. The Monte Carlo simulation, positron annihilation and IR spectroscopy were used to study the radiation induced defects. The result showed that monovacancies existed in as grown samples, but more monovacancies were introduced, after Ne ions irradiation, and with increasing radiation dose, divacancies were formed, and eventually large voids were observed.
