Effect of ultrasound on the yield and physiochemical properties of hemicelluloses from wheat straw using 0.5 M NaOH in 60% aqueous methanol has been comparatively studied. The results showed that applying sonication for 5, 10, 15, 20, 25, 30, and 35 min resulted in an increasing yield of hemicelluloses by 2.9%, 3.9%, 6.0%, 8.6%, 8.6%, 8.6%, and 9.2% of the original hemicelluloses, respectively, in comparison with the experiment performed without ultrasonic assistance. The hemicelluloses isolated by ultrasound-assisted extraction, showed a slightly lower molecular weight, associated lignin, and thermal stability during the first stage of decomposition, but slightly more linear as compared to the hemicelluloses obtained by classical method without ultrasonic irradiation. No substantial differences in the main structure features between the hemicellulosic preparations isolated by classical or ultrasound-assisted extractions were found.  相似文献

Bamboo was submitted to ultrasound-assisted extraction in aqueous ethanol to evaluate the effect of ultrasonic irradiation on the dissolution of lignin. In this case, the dewaxed bamboo culms were subjected to ball milling for 48 h, and then were suspended in 95% ethanol followed by ultrasonic irradiations for varied times at 20 °C to obtain ethanol-soluble fractions. The structural and thermal properties of the ethanol-soluble fractions were comparatively investigated by chemical analysis including alkaline nitrobenzene oxidation, bound carbohydrate determination, FT-IR spectra, HSQC spectra, TG, and DTA. The results showed that the yields of the ethanol-soluble fractions were between 4.29% and 4.76% for the fractions prepared with ultrasonic irradiation time ranging from 5 to 55 min, as compared to 4.02% for the fraction prepared without ultrasonic irradiation. It was found that the lignin content of the fraction increased with the increase of the ultrasonic irradiation time. There was a slight increase of the molecular weight of the lignin with the increase of the ultrasonic irradiation time. Alkaline nitrobenzene oxidation coupled with HSQC analysis indicated that the lignin in the fractions was mainly composed of G S H type units as well as minor amounts of ferulic acids. In addition, the fraction prepared with ultrasonic irradiation exhibited a slightly higher thermal stability as compared to the fraction prepared without ultrasonic irradiation.  相似文献

Low intensity ultrasound can produce various effects on biological materials, such as stimulating enzyme activity, cell growth, biosynthesis, etc., which may improve the efficiency of enhanced biological phosphorus removal (EBPR). We adopt total phosphorus (TP) and dehydrogenase activity (DHA) as indicators to confirm the feasibility of applying low intensity ultrasound in EBPR. Single-factor experiments and orthogonal test were conducted in batch anaerobic/oxic (A/O) process simulation to study the influence of ultrasonic intensity and exposure time in the EBPR process. The results showed that the optimal ultrasonic parameters were 0.2 W/cm2 and 10 min under which condition the TP concentration in the effluent was 35–50% lower than that of the control (without ultrasonic irradiation). Changes of sludge activities after ultrasonic irradiation were examined. The improvement of sludge activity by ultrasound took 4 h after irradiation to reach the peak level, when an increase above 50% of DHA has been achieved by ultrasonic irradiation, and the enhancing effects induced by ultrasound disappeared in 16 h after irradiation. A tentative mechanism of biological phosphorus removal enhancement stimulated by ultrasound was discussed based on these phenomena.  相似文献

The ultrasound assisted three phase partitioning (UATPP) is a novel bioseparation method for separation and purification of biomolecules. In the present work, UATPP was investigated for the first time for purification of serratiopeptidase from Serratia marcescens NRRL B 23112. Effect of various process parameters such as ammonium sulphate saturation, t-butanol to crude extract ratio, pH, ultrasonic frequency, ultrasonic intensity, duty cycle and irradiation time were evaluated and optimized. The optimized conditions were found to be as follows: ammonium sulphate saturation 30% (w/v), pH 7.0, t-butanol to crude ratio 1:1 (v/v), ultrasound frequency 25 kHz, ultrasound intensity 0.05 W/cm2, duty cycle 20% and irradiation time 5 min. The maximum purity and recovery obtained from UATPP was 9.4-fold and 96% respectively as compared to the three phase partitioning (TPP) (4.2-fold and 83%). Also the process time for UATPP was significantly reduced to 5 min from 1 h as compared to TPP. The results indicate that, UATPP is an efficient technique for the purification of serratiopeptidase with maximum purity, recovery and reduced processing time.  相似文献

Delignification of sawdust was studied using ultrasound assisted alkali peroxide approach using longitudinal horn for the first time and the efficacy compared with more commonly used configurations of ultrasonic reactors. Comparison with the conventional approach based on stirring has also been presented to establish the process intensification benefits. Effect of different operating parameters such as sodium carbonate concentration (0.1, 0.15, 0.2, 0.25 M), hydrogen peroxide concentration (0.2, 0.4, 0.6, 0.8, 1 M) and biomass loading (2, 4, 6, 8, 10 wt%), on the efficacy of lignin extraction has been investigated for different ultrasonic reactors. The optimum conditions for probe type ultrasonic horn were established as 150 W, 50% duty cycle and 80% amplitude with optimum process conditions as Na2CO3 concentration as 0.2 M, H2O2 concentration as 1 M, biomass loading of 10 wt% and operating time of 70 min. Longitudinal horn resulted in best efficacy (both in terms of yield and energy requirements) followed by ultrasonic horn and ultrasonic bath whereas the conventional approach was least effective. The obtained lignin was also analyzed using different characterization techniques. The presence of peaks at wavelength range of 875–817, 1123–1110, and at 1599 cm−1 for the extracted sample confirmed the presence of lignin. Increase in the crystallinity index of the processed sample (maximum for longitudinal horn) also confirmed the lignin removal as lignin is amorphous in nature. Overall it has been concluded that ultrasound can be effectively used for delignification with longitudinal horn as best configuration.  相似文献

In this research work, dextranase has immobilized onto calcium alginate beads using a novel ultrasound method. The process of immobilization of the enzyme was carried out in a one-step ultrasound process. Effects of ultrasound conditions on loading efficiency and immobilization yield of the enzyme onto calcium alginate beads were investigated. Furthermore, the activity of the free and immobilized enzymes prepared with and without ultrasound treatment, as a function of pH, temperature, recyclability and enzyme kinetic parameters, was compared. The maximum loading efficiency and the immobilization yield were observed when the immobilized dextranase was prepared with an ultrasonic irradiation at 25 kHz, 40 W for 15 min, under which the loading efficiency and the immobilization yield increased by 27.21% and 18.77%, respectively, compared with the immobilized enzymes prepared without ultrasonic irradiation. On the other hand, immobilized enzyme prepared with ultrasonic irradiation showed Vmax and KM value higher than that for the immobilized enzyme prepared without ultrasonic irradiation, likewise, both the catalytic and specificity constants of immobilized enzyme prepared with ultrasonic irradiation were higher than that for immobilized enzyme prepared without ultrasound, indicating that, this new ultrasonic method improved the catalytic kinetics activity of immobilized dextranase at all the reaction conditions studied. Compared with immobilized enzyme prepared without ultrasound treatment, the immobilized enzymes prepared with ultrasound irradiation exhibited: a higher pH optimum, optimal reaction temperature, activation energy, and thermal stability, as well as, a higher recyclability, which, illustrating the effectiveness of the sonochemical method. To the best of our knowledge, this is the first report on the effect of ultrasound treatments on the immobilization of dextranase.  相似文献

An ultrasonic-assisted separation of alkali chloride (LiCl, NaCl, and KCl) salts have been carried out using of an hydrophobic ionic liquid membrane (ILM). The ionic liquid 1-butyl-3-methylimidazolium hexafluorophosphate and tributyl phosphate mixture have been used as ILM. An ultrasonic probe with different frequencies (25, 100, and 250) kHz have been applied as source of ultrasound generator with different times of sonication (2, 5, and 10) min in three phases system containing feed, ILM, and receiver in osmotic U-shaped tube. Also, 250, 500, and 1000 ppm of the feed (alkali chloride) concentration have been used to separate. The frequency of 250 kHz with higher sonication time provides optimum condition for separation of LiCl with lower feed concentration. The thermodynamic properties such as density and speed of sound and the related thermodynamic properties have been calculated to optimize ILM composition (xIL = 0.45) for ultrasound-separation.  相似文献

Ultrasound-assisted approach has been investigated for delignification so as to develop green and sustainable technology. Combination of NaOH with ultrasound has been applied with detailed study into effect of various parameters such as time (operating range of 15–90 min), alkali concentration (0.25 M−2.5 M), solvent loading (1:15–1:30 w/v), temperature (50–90 ˚C), power (40–140 W) and duty cycle (40–70 %) at fixed frequency of 20 kHz. The optimized operating conditions established for the ultrasonic horn were 1 M as the NaOH concentration, 1 h as treatment time, 70˚C as the operating temperature, 1:20 as the biomass loading ratio, 100 W as the ultrasonic power and 70% duty cycle yielding 67.30% as the delignification extent. Comparative study performed using conventional and ultrasonic bath assisted alkaline treatment revealed lower delignification as 48.09% and 61.55% respectively. The biomass samples were characterized by SEM, XRD, FTIR and BET techniques to establish the role of ultrasound during the treatment. The morphological changes based on the ultrasound treatment demonstrated by SEM were favorable for enhanced delignification and also the crystallinity index was more in the case of ultrasound treated material than that obtained by conventional method. Specific surface area and pore size determinations based on BET analysis also confirmed beneficial role of ultrasound. The overall results clearly demonstrated the intensification obtained due to the use of ultrasonic reactors.  相似文献

The process time in chrome tannage in leather making, using an elastic compression cycle followed by irradiation by high-intensity ultrasound, is quite short lasting only a few minutes, compared with a process time of several hours in modern chrome tannage. After ultrasonic irradiation, samples were basified in 17 h in chrome liquor at a pH of 4.0 and the shrinkage temperature was measured. The determination of the efficiency for the chrome liquor penetrating into the hides can be based on the steepness of the shrinkage temperature-processing time curve. An approximate value of 20 degrees C min(-1) can be evaluated for the initial slope of the curve when elastic compression and high-intensity ultrasonic irradiation is used, and a processing time of 2 min is required in chrome liquor (plus 17 h basification and 24 h storage time) to obtain leather stable to boiling. Usually, hides are kept in chrome liquor for 2 h.  相似文献

In the present study a bleaching process of a kaolinite was carried out using thiourea as the leachant agent in the iron removal process, in the absence and presence of ultrasound. The effect of thiourea was investigated together with other factors, such as thiourea concentration, temperature, treatment time, and ultrasonic parameters. The optimum conditions for the maximum whiteness of 89% with ultrasound were determined as follows: reaction temperature, 20 °C; ultrasound frequency, 80 kHz; ultrasound power, 500 W; thiourea concentration, 0.4 wt.%; pH, 3.0; reaction time, 20 min. The assistance of ultrasound led to a remarkable acceleration for the iron leaching process, and dramatic reduction in the concentration of leach reagent, irradiation time, and reaction temperature, when compared with the conventional bleaching method using thiourea in the absence of ultrasound.  相似文献
