As part of a project studying the secondary metabolites extracted from the Chilean flora, we report herein three new β‐agarofuran sesquiterpenes, namely (1S,4S,5S,6R,7R,8R,9R,10S)‐6‐acetoxy‐4,9‐dihydroxy‐2,2,5a,9‐tetramethyloctahydro‐2H‐3,9a‐methanobenzo[b]oxepine‐5,10‐diyl bis(furan‐3‐carboxylate), C27H32O11, ( II ), (1S,4S,5S,6R,7R,9S,10S)‐6‐acetoxy‐9‐hydroxy‐2,2,5a,9‐tetramethyloctahydro‐2H‐3,9a‐methanobenzo[b]oxepine‐5,10‐diyl bis(furan‐3‐carboxylate), C27H32O10, ( III ), and (1S,4S,5S,6R,7R,9S,10S)‐6‐acetoxy‐10‐(benzoyloxy)‐9‐hydroxy‐2,2,5a,9‐tetramethyloctahydro‐2H‐3,9a‐methanobenzo[b]oxepin‐5‐yl furan‐3‐carboxylate, C29H34O9, ( IV ), obtained from the seeds of Maytenus boaria and closely associated with a recently published relative [Paz et al. (2017). Acta Cryst. C 73 , 451–457]. In the (isomorphic) structures of ( II ) and ( III ), the central decalin system is esterified with an acetate group at site 1 and furoate groups at sites 6 and 9, and differ at site 8, with an OH group in ( II ) and no substituent in ( III ). This position is also unsubstituted in ( IV ), with site 6 being occupied by a benzoate group. The chirality of the skeletons is described as 1S,4S,5S,6R,7R,8R,9R,10S in ( II ) and 1S,4S,5S,6R,7R,9S,10S in ( III ) and ( IV ), matching the chirality suggested by NMR studies. This difference in the chirality sequence among the title structures (in spite of the fact that the three skeletons are absolutely isostructural) is due to the differences in the environment of site 8, i.e. OH in ( II ) and H in ( III ) and ( IV ). This diversity in substitution, in turn, is responsible for the differences in the hydrogen‐bonding schemes, which is discussed.  相似文献

The crystal structures of salt 8 , which was prepared from (R)‐2‐methoxy‐2‐(2‐naphthyl)propanoic acid ((R)‐MβNP acid, (R)‐ 2 ) and (R)‐1‐phenylethylamine ((R)‐PEA, (R)‐ 6 ), and salt 9 , which was prepared from (R)‐2‐methoxy‐2‐(1‐naphthyl)propanoic acid ((R)‐MαNP acid, (R)‐ 1 ) and (R)‐1‐(p‐tolyl)ethylamine ((R)‐TEA, (R)‐ 7 ), were determined by X‐ray crystallography. The MβNP and MαNP anions formed ion‐pairs with the PEA and TEA cations, respectively, through a methoxy‐group‐assisted salt bridge and aromatic CH???π interactions. The networks of salt bridges formed 21 columns in both salts. Finally, (S)‐(2E,6E)‐(1‐2H1)farnesol ((S)‐ 13 ) was prepared from the reaction of (2E,6E)‐farnesal ( 11 ) with deuterated (R)‐BINAL‐H (i.e., (R)‐BINAL‐D). The enantiomeric excess of compound (S)‐ 13 was determined by NMR analysis of (S)‐MαNP ester 14 . The solution‐state structures of MαNP esters that were prepared from primary alcohols were also elucidated.  相似文献

Reaction of [IrCp*Cl2]2 with ferrocenylimines (Fc=NAr, Ar=Ph, p‐MeOC6H4) results in ferrocene C?H activation and the diastereoselective synthesis of half‐sandwich iridacycles of relative configuration Sp*,RIr*. Extension to (S)‐2‐ferrocenyl‐4‐(1‐methylethyl)oxazoline gave highly diastereoselective control over the new elements of planar chirality and metal‐based pseudo‐tetrahedral chirality, to give both neutral and cationic half‐sandwich iridacycles of absolute configuration Sc,Sp,RIr. Substitution reactions proceed with retention of configuration, with the planar chirality controlling the metal‐centred chirality through an iron–iridium interaction in the coordinatively unsaturated cationic intermediate.  相似文献

(2S,3S)‐2,6‐Dimethylheptane‐1,3‐diol, C9H20O2, (I), was synthesized from the ketone (R)‐4‐benzyl‐3‐[(2R,3S)‐3‐hydroxy‐2,6‐dimethylheptanoyl]‐1,3‐oxazolidin‐2‐one, C19H27NO4, (II), containing C atoms of known chirality. In both structures, strong hydrogen bonds between the hydroxy groups form tape motifs. The contribution from weaker C—H...O hydrogen bonds is much more evident in the structure of (II), which furthermore contains an example of a direct short Osp3...Csp2 contact that represents a usually unrecognized type of intermolecular interaction.  相似文献

Unlike the excellent (S)-enantioselective epoxidation of styrene performed by natural styrene monooxygenases (ee > 99%), the (R)-enantioselective epoxidation of styrene has not yet achieved a comparable efficiency using natural or engineered oxidative enzymes. This report describes the H2O2-dependent (R)-enantioselective epoxidation of unfunctionalized styrene and its derivatives by site-mutated variants of a unique non-natural P450BM3 peroxygenase, working in tandem with a dual-functional small molecule (DFSM). The observed (R)-enantiomeric excess (ee) of styrene epoxidation is up to 99% with a turnover number (TON) of 918 by the best enantioselective mutant F87A/T268I/L181Q, while the best active mutant F87A/T268I/V78A/A184L (with 98% ee) gave a catalytic TON of 4350, representing the best activity of a P450 peroxygenase towards styrene epoxidation to date. Following this approach, a set of styrene derivatives, such as o-, m-, p-chlorostyrenes and fluorostyrenes, could also be epoxidized with modest to very good TONs (362–3480) and high (R)-enantioselectivities (95–99% ee). The semi-preparative scale synthesis of (R)-styrene oxide performed at 0 °C with high conversion, maintaining enantioselectivity, and moderate isolated yields, further suggests the potential application of the current P450 enzymatic system in styrene epoxidation. This study indicates that the synergistic use of protein engineering and an exogenous DFSM constitutes an efficient strategy to control the enantioselectivity of styrene epoxidation, thus substantially expanding the chemical scope of P450 enzymes as useful bio-oxidative catalysts.

H2O2-dependent epoxidation of unfunctionalized styrenes is achieved with high (R)-enantioselectivity and moderate to excellent TONs by combining site-mutated variants of cytochrome P450BM3 monooxygenase and a dual-functional small molecule (DFSM).  相似文献

An optically active (S)‐α‐ethylleucine ((S)‐αEtLeu) as a chiral α‐ethylated α,α‐disubstituted α‐amino acid was synthesized by means of a chiral acetal auxiliary of (R,R)‐cyclohexane‐1,2‐diol. The chiral α‐ethylated α,α‐disubstituted amino acid (S)‐αEtLeu was introduced into the peptides constructed from 2‐aminoisobutyric acid (=dimethylglycine, Aib), and also into the peptide prepared from diethylglycine (Deg). The X‐ray crystallographic analysis revealed that both right‐handed (P) and left‐handed (M) 310‐helical structures exist in the solid state of CF3CO‐(Aib)2‐[(S)‐αEtLeu]‐(Aib)2‐OEt ( 14 ) and CF3CO‐[(S)‐αEtLeu]‐(Deg)4‐OEt ( 18 ), respectively. The IR, CD, and 1H‐NMR spectra indicated that the dominant conformation of pentapeptides 14 and CF3CO‐[(S)‐αEtLeu]‐(Aib)4‐OEt ( 16 ) in solution is a 310‐helical structure, and that of 18 in solution is a planar C5 conformation. The conformation of peptides was also studied by molecular‐mechanics calculations.  相似文献

The synthesis of 46 derivatives of (2R,3R,4S)‐2‐(aminomethyl)pyrrolidine‐3,4‐diol is reported (Scheme 1 and Fig. 3), and their inhibitory activities toward α‐mannosidases from jack bean (B) and almonds (A) are evaluated (Table). The most‐potent inhibitors are (2R,3R,4S)‐2‐{[([1,1′‐biphenyl]‐4‐ylmethyl)amino]methyl}pyrrolidine‐3,4‐diol ( 3fs ; IC50(B)=5 μM , Ki=2.5 μM ) and (2R,3R,4S)‐2‐{[(1R)‐2,3‐dihydro‐1H‐inden‐1‐ylamino]methyl}pyrrolidine‐3,4‐diol ( 3fu ; IC50(B)=17 μM , Ki=2.3 μM ). (2S,3R,4S)‐2‐(Aminomethyl)pyrrolidine‐3,4‐diol ( 6 , R?H) and the three 2‐(N‐alkylamino)methyl derivatives 6fh, 6fs , and 6f are prepared (Scheme 2) and found to inhibit also α‐mannosidases from jack bean and almonds (Table). The best inhibitor of these series is (2S,3R,4S)‐2‐{[(2‐thienylmethyl)amino]methyl}pyrrolidine‐3,4‐diol ( 6o ; IC50(B)=105 μM , Ki=40 μM ). As expected (see Fig. 4), diamines 3 with the configuration of α‐D ‐mannosides are better inhibitors of α‐mannosidases than their stereoisomers 6 with the configuration of β‐D ‐mannosides. The results show that an aromatic ring (benzyl, [1,1′‐biphenyl]‐4‐yl, 2‐thienyl) is essential for good inhibitory activity. If the C‐chain that separates the aromatic system from the 2‐(aminomethyl) substituent is longer than a methano group, the inhibitory activity decreases significantly (see Fig. 7). This study shows also that α‐mannosidases from jack bean and from almonds do not recognize substrate mimics that are bulky around the O‐glycosidic bond of the corresponding α‐D ‐mannopyranosides. These observations should be very useful in the design of better α‐mannosidase inhibitors.  相似文献

Asymmetric anionic homopolymerizations of N‐1‐naphthylmaleimide (1‐NMI) were performed with chiral ligand/organometal complexes to form optically active polymers. Poly(1‐NMI)s obtained with methylene‐bridged bisoxazoline derivatives (Rbox)‐diethylzinc (Et2Zn) complexes showed high specific optical rotations ([α]) from +152.3 to +191.4°. Circular dichroism spectra of the polymers exhibited a split Cotton effect in the UV absorption‐band region. According to the exciton chirality method, the absolute configuration of the polymer main chain was determined according to the following method: (+)‐poly[N‐substituted maleimides (RMI)] main chains can contain more (S,S)‐ than (R,R)‐configurations. The chiral compounds (R)‐ and (S)‐1‐benzoyl‐2,3,5,6‐tetrahydro‐3‐methyl‐2‐(1‐methylethyl)pyrimidin‐4(1H)‐one ((R)‐ and (S)‐ 1 ), derived from (R)‐ and (S)‐asparagine, respectively, were used as convenient starting materials for the preparation of the enantiomerically pure α‐alkylated (alkyl=Me, Et, Bn) α,β‐diamino acids (R)‐ and (S)‐ 11 – 13 . The chiral lithium enolates of (R)‐ and (S)‐ 1 were first alkylated, and the resulting diasteroisomeric products 5 – 7 were aminated with ‘di(tert‐butyl) azodicarboxylate’ (DBAD), giving rise to the diastereoisomerically pure (≥98%) compounds 8 – 10 . The target compounds (R)‐ and (S)‐ 11 – 13 could then be obtained in good yields and high purities by a hydrolysis/hydrogenolysis/hydrolysis sequence.  相似文献

The importance of β‐peptides lies in their ability to mimic the conformational behavior of α‐peptides, even with a much shorter chain length, and in their resistance to proteases. To investigate the effect of substitution of β‐peptides on their dominant fold, we have carried out a molecular‐dynamics (MD) simulation study of two tetrapeptides, Ac‐(2R,3S)‐β2,3hVal(αMe)‐(2S)‐β2hPhe‐(R)‐β3hLys‐(2R,3S)‐β2,3‐Ala(αMe)‐NH2, differing in the substitution at the Cα of Phe2 (pepF with F, and pepH with H). Three simulations, unrestrained (UNRES), using 3J‐coupling biasing with local elevation in combination with either instantaneous (INS) or time‐averaging (AVE) NOE distance restraining, were carried out for each peptide. In the unrestrained simulations, we find three (pepF) and two (pepH) NOE distance bound violations of maximally 0.22 nm that involve the terminal residues. The restrained simulations match both the NOE distance bounds and 3J‐values derived from experiment. The fluorinated peptide shows a slightly larger conformational variability than the non‐fluorinated one.  相似文献

The crystal structure of the title compound, C8H17NO2, consists of (R,R) and (S,S) enantiomeric pairs packed in adjacent double layers which are characterized by centrosymmetric hydrogen‐bonded dimers, generated via N—H...O and O—H...O interactions, respectively. Intermolecular interactions, related to acceptor and donor molecule chirality, link the achiral double layers into tubular columns, which consist of a staggered hydrophilic inner core surrounded by a hydrophobic cycloalkyl outer surface and extend in the [011] direction.  相似文献

The title enanti­omorphic compounds, C16H23NO4S, have been obtained in an enanti­omerically pure form by crystallization from a diastereomeric mixture either of (2S,4S)‐ and (2R,4S)‐ or of (2R,4R)‐ and (2S,4R)‐2‐tert‐butyl‐4‐methyl‐3‐(4‐tolyl­sulfon­yl)‐1,3‐oxazolidine‐4‐carbaldehyde. These mixtures were prepared by an aziridination rearrangement process starting with (S)‐ or (R)‐2‐tert‐butyl‐5‐methyl‐4H‐1,3‐dioxine. The crystal structures indicate an envelope conformation of the oxazolidine moiety for both compounds.  相似文献

The crystal structures of a pair of diastereomeric 1:2 salts of (R)‐ and (S)‐2‐methylpiperazine with (2S,3S)‐tartaric acid, namely (R)‐2‐methylpiperazinediium bis[hydrogen (2S,3S)‐tartrate] monohydrate, (I), and (S)‐2‐methylpiperazinediium bis[hydrogen (2S,3S)‐tartrate] monohydrate, (II), both C5H14N22+·2C4H5O6·H2O, each reveal the formation of well‐defined head‐to‐tail‐connected hydrogen tartrate chains; these chains are linked into a two‐dimensional sheet via intermolecular hydrogen bonds involving hydroxy groups and water molecules, resulting in a layer structure. The (R)‐2‐methylpiperazinediium ions lie between the hydrogen tartrate layers in the most stable equatorial conformation in (I), whereas in (II), these ions are in an unstable axial position inside the more interconnected layers and form a larger number of intermolecular hydrogen bonds than are observed in (I).  相似文献

Two pairs of enantiomeric compounds with formulas (S)‐ or (R)‐Co3(ppap)2(4,4′‐bpy)2(H2O)2 ? 4 H2O [(S)‐ 1 or (R)‐ 1 ], (S)‐ or (R)‐Co3(ppap)2(4,4′‐bpy)2(H2O)2 ? 3 H2O [(S)‐ or (R)‐ 2 ), and related racemic compound Co3(ppap)2(4,4′‐bpy)2(H2O)2 ? 4 H2O (rac‐ 3 ; 4,4′‐bpy=4,4′‐bipyridine, H3ppap=3‐phenyl‐2‐[(phosphonomethyl)amino]propanoic acid) are reported. Compounds 1 and rac‐ 3 show identical three‐dimensional framework structures, whereas compounds 2 have two‐dimensional layer structures. Compounds 1 and 2 are catenation isomers, formation of which is controlled solely by the pH of the reaction mixtures, whereas the formation of isomeric compounds 1 and rac‐ 3 is controlled purely by the chirality of the phosphonate ligand. The magnetic properties of fully dehydrated (S)‐ 1 , (S)‐ 2 , and rac‐ 3 are highly dependent on both structure and chirality.  相似文献

Starting from simple aromatic aldehydes and acetylfuran, (E)‐1‐(furan‐2‐yl)‐3‐arylprop‐2‐en‐1‐ones ( 2 ) were synthesized in high yields. Cyclopropanation of the C?C bond with trimethylsulfoxonium iodide (Me3SO+I?) furnished (furan‐2‐yl)(2‐arylcyclopropyl)methanones 3 in 90–97% yields. Selective conversion of cyclopropyl ketones to their (E)‐ and (Z)‐oxime ethers 5 and oxazaborolidine‐catalyzed stereoselective reduction of the C?N bond followed by separation of the formed diastereoisomers, furnished (2‐arylcyclopropyl)(furan‐2‐yl)methanamines 6 in optically pure form and high yield. Oxidation of the furan ring of (S,S,S)‐, (S,R,R)‐, (R,S,S)‐, and (R,R,R)‐ 6a afforded the four stereoisomers of α‐(2‐phenylcyclopropyl) glycine ( 1a ).  相似文献

New 2‐(aminomethyl)‐5‐(hydroxymethyl)pyrrolidine‐3,4‐diol derivatives were synthesized from (5S)‐5‐[(trityloxy)methyl]pyrrolidin‐2‐one ( 6 ) (Schemes 1 and 2) and their inhibitory activities toward 25 glycosidases assayed (Table). The influence of the configuration of the pyrrolidine ring on glycosidase inhibition was evaluated. (2R,3R,4S,5R)‐2‐[(benzylamino)methyl]‐5‐(hydroxymethyl)pyrrolidine‐3,4‐diol ((+)‐ 21 ) was found to be a good and selective inhibitor of α‐mannosidase from jack bean (Ki=1.2 μM ) and from almond (Ki=1.0 μM ). Selectivity was lost for the non‐benzylated derivative (2R,3R,4S,5R)‐2‐(aminomethyl)‐5‐(hydroxymethyl)pyrrolidine‐3,4‐diol ((+)‐ 22 ) which inhibited α‐galactosidases, β‐galactosidases, β‐glucosidases, and αN‐acetylgalactosaminidase as well.  相似文献

The title compound, C14H18INO, crystallizes as +sc/+sp/+sc 2‐iodoanilide molecules (and racemic opposites) and shows significant intermolecular I...O interactions in the solid state, forming dimeric pairs about centres of symmetry. Under asymmetric Heck conditions, the S enantiomer of the dihydroindol‐2‐one was obtained using (R)‐(+)‐2,2′‐bis(diphenylphosphino)‐1,1′‐binaphthyl [(R)‐BINAP], suggesting a mechanism that proceeds by oxidative addition to give the title (P) enantiomer of the compound and pro‐S coordination of the Re face of the alkene in a conformation similar to that defined crystallographically, except that rotation about the C—C bond of the butenyl group is required.  相似文献

P450 119 peroxygenase and its site‐directed mutants are discovered to catalyze the enantioselective epoxidation of methyl‐substituted styrenes. Two new site‐directed P450 119 mutants, namely T213Y and T213M, which were designed to improve the enantioselectivity and activity for the epoxidation of styrene and its methyl substituted derivatives, were studied. The T213M mutant is found to be the first engineered P450 peroxygenase that shows highly enantioselective epoxidation of cis‐β‐methylstyrenes, with up to 91 % ee. Molecular modeling studies provide insights into the different catalytic activity of the T213M mutant and the T213Y mutant in the epoxidation of cis‐β‐methylstyrene. The results of the calculations also contribute to a better understanding of the substrate specificity and configuration control for the regio‐ and stereoselective peroxygenation catalyzed by the T213M mutant.  相似文献

The crystal and molecular structures of (1S,3aR,7S,8S,8aR,8bR)‐(+)‐7,8‐Di‐tert‐butoxy‐1‐ph‐ enyloctahydro‐1H‐pyrrolo(1,‐b)‐1H‐phospholo(2,‐ d)isoxazole 1‐oxide ( III , hereafter) and (1R,3aS,7S, 8S,8aS,8bS)‐(+)‐7,8‐Di‐tert‐butoxy‐1‐phenyloctahyd‐ ro‐1H‐pyrrolo(1,2‐b)‐1H‐phospholo(2,3‐d)isoxazole 1‐ oxide ( IV , hereafter) have been determined. III crystallizes in space group P212121, and IV in P21 one. The conformational analysis of the puckered heteroatom three‐ring system shows the conformation of noticeable distorted envelope with puckering amplitude Q2 = 0.397 Å, the intermediate conformation between twisted and envelope with Q2 = 0.353 Å, and half‐chair conformation with Q2 = 0.451 Å, for phospholane, oxazolidine, and pyrrolidine rings of III , respectively. Rings in molecule of IV adopt conformations of envelopes with Q2 = 0.381 Å, Q2 = 0.367 Å, and Q2 = 0.363 Å, respectively, for the rings as described above. The molecules of III are assembled by intermolecular weak hydrogen bonds to the one‐dimensional chain along x‐axis. The structure of IV is built‐up of weak intermolecular hydrogen bonds to form a two‐dimensional hydrogen bond network. The differences in conformation between compounds III and IV cause changes in hydrogen bonding pattern, because in molecule IV there is no hydrogen cavern filled with three hydrogen bond donors, and one weak hydrogen bond has not enough strength to force such an arrangement as it is in III . © 2005 Wiley Periodicals, Inc. The stem bark CH2Cl2 extract of Ravensara crassifolia showed antifungal activity against the phytopathogenic fungus Cladosporium cucumerinum in a bioautographic TLC assay. Activity‐guided fractionation afforded two new α‐pyrones : (6S)‐5,6‐dihydro‐6‐[(2R)‐2‐hydroxy‐6‐phenylhexyl]‐2H‐pyran‐2‐one ( 1 ) and (6R)‐6‐[(4R,6R)‐4,6‐dihydroxy‐10‐phenyldec‐1‐enyl]‐5,6‐dihydro‐2H‐pyran‐2‐one ( 2 ). Their structures and absolute configurations were established by NMR spectroscopy, chemical methods, and CD spectroscopy. The antifungal activity against C. cucumerinum was determined for both compounds.  相似文献
