The techniques of chemical ligation have attracted great attention as an alternative to enzymatic joining of DNA ends. Here we introduce the photoligation of anthracene-modified ODN conjugates through anthracene cyclodimer formation. The effect of the positions and the kinds of single base mismatch on the template was evaluated using eight templates with one-base displacements. We found out that the yield of the ligation was affected by mispairing in a position-dependent manner. Such results would be attributed to the disruption of the local structure at the ligation site.  相似文献

3-Bromo boron dipyrromethene (3-bromo BODIPY) has been used as key synthon to prepare one ethynyl bridged and six ethynylphenyl bridged BODIPY-chromophore conjugates using mild Pd(0) coupling conditions. The chromophores possessing very distinct features, such as anthracene, BODIPY, terpyridine, porphyrin, Zn(II)porphyrin, 21,23-dithiaporphyrin and thiasapphyrin were connected at 3-position of boronboron-dipyrromethene dye by coupling of 3-bromo BODIPY with ethynyl or ethynylphenyl chromophore in toluene/triethylamine in the presence of catalytic amount of AsPh3/Pd2(dba)3 at 40 °C followed by column chromatographic purification. The spectral studies indicated that the interaction is stronger in ethynyl bridged BODIPY-chromophore conjugate compared to ethynylphenyl bridged BODIPY-chromophore conjugates. The steady-state fluorescence indicated that in ethynyl bridged BODIPY-anthracene conjugate, the BODIPY unit act as energy acceptor and showed a possibility of energy transfer from donor anthracene unit to acceptor BODIPY unit on selective excitation of anthracene unit. However, in ethynylphenyl bridged BODIPY-porphyrin conjugates, the BODIPY unit act as energy donor and exhibited a possibility of singlet-singlet energy transfer from BODIPY unit to porphyrin unit.  相似文献

Diblock and triblock dendron–polymer conjugates containing biodegradable polyester dendron blocks and polyethylene glycol (PEG) polymer were synthesized using the Diels–Alder “click” cycloaddition reaction. PEG polymers with furan‐protected maleimide functionality were synthesized and reacted with biodegradable polyester dendrons containing an anthracene moiety at their focal point. First through third generations of biodegradable polyester dendrons containing an anthracene unit at their focal point were synthesized using a divergent strategy. Efficient conjugation of the dendrons to polymers was demonstrated using 1HNMR and size exclusion chromatography. A two-step general approach to a new chiral auxiliary starting from levoglucosenone is reported. The compound is obtained by a [4+2] cycloaddition reaction with anthracene followed by a diastereoselective reduction of the C-2 keto function in high overall yield. The auxiliary has been used as chiral template in an asymmetric Diels-Alder reaction of the corresponding acrylic ester derivative with cyclopentadiene and shown to be efficient for asymmetric induction.  相似文献

Highly stereoselective Friedel-Crafts reactions have been performed using a chiral anthracene template to control the selectivity of the reaction. In the case of additions to fully substituted N-acyliminium ions, competitive elimination and condensation reactions were observed. Retro-Diels-Alder reaction of one of the reaction products led to a precursor that could be used for the construction of pyroglutamic acids bearing quaternary stereogenic centres.  相似文献

An efficient five-step synthetic method was developed to access a homologous series of spermidine-acridine and spermidine-anthracene conjugates. The derivatives were comprised of a spermidine fragment covalently tethered at its N4 position to either an acridine or anthracene nucleus via an aliphatic chain (e.g., spermidine-[aliphatic tether]-acridine). The distance separating the spermidine and aromatic nucleus was altered by using different tethers comprised of four or five methylene units, respectively. These ligands (2-5) were shown to inhibit human DNA topoisomerase-II (TOPO-II) activity at 10 microM. Enzymatic activity was assessed as the ability to unknot (decatenate) and cleave kinetoplast DNA (kDNA). Polyamine conjugation did not disrupt the ability of the acridine-spermidine conjugates 2 and 3 to inhibit TOPO-II activity as compared with the 9-aminoacridine and 9-(N-butyl)aminoacridine controls (at 10 microM). In general, the acridine derivatives (2 and 3) showed higher TOPO-II inhibitory activity than their anthracene counterparts (4 and 5). However, this trend was reversed in a whole cell assay with L1210 (murine leukemia) cells, wherein the anthracene analogues were more potent than their acridine counterparts. In this regard the qualitative enzyme-based assay did not predict the trends in the corresponding IC(50) values. Within either series insertion of an additional methylene unit did not significantly alter activity. While the appended spermidine unit did not disrupt TOPO II inhibition by the tethered DNA intercalator, it did provide an alternative mode of entry into the cell as demonstrated by spermidine protection assays. These results were compared with a spermine-intercalator analogue. Of all the conjugates tested the N(4)-(4-(9-aminoacridinyl)butyl)spermine hexahydrochloride (conjugate 16)resulted in the highest degree of L1210 cell rescue upon cotreatment of the cells with exogenous spermidine. It was concluded that the monoalkylated spermine motif present in 16 holds promise as a better vector than its N4 monoalkylated spermidine counterpart.  相似文献

Acylation of anthracene and oxalyl chloride in the presence of anhydrous AlCl3 was investigated. Pure 1,2-aceanthrylenedione, which is used as new functional macromolecule, was prepared by extraction and recrystallation. The structure of 1,2-aceanthrylenedione was identified by measure of melting point, GC/MS, FTIR and 1↑HNMR analyses. The influences of various parameters, such as solvent, dosage of catalyst, molar ratio of anthracene to oxalyl chloride, reaction temperature, and reaction time were studied. The anhydrous AlCl3 was found to catalyze the reaction with high yeild and good selectivity. When the reaction temperature is 303K, reaction time is 5h and molar ratio of anthracene to oxalyl chloride is 1:2, the yield and selectivity of 1,2-aceanthrylenedione is 83.8 % and 92.3% respectively.  相似文献

本文以鱼明胶为保护介质,在不同的鱼明胶 溴化银比值下,采用双注法制备AgBr/I纳米颗粒乳剂,以TEM观测颗粒形态,据此统计得出该乳剂中AgBr/I纳米颗粒平均粒径(d)及粒径分布系数(|±σ|/d).鱼明胶 溴化银比值在0.31g/mmol-0.62g/mmol范围内,颗粒平均粒径仅有微小变化,但粒径分布系数随该比值的增大而减小,表明颗粒均一性提高.采用微波光导法对该纳米尺度AgBr/I乳剂颗粒的光电子衰减过程进行测试,结果表明,纳米尺度Ag-Br/I乳剂颗粒的光电子衰减过程为双组分,即初始快衰减和随后慢衰减,其衰减过程皆符合一级动力学规律.该纳米尺度乳剂颗粒光电子寿命较常规乳剂颗粒明显缩短.  相似文献

Acylation reaction of anthracene with oxalyl chloride in the presence of [Emim]Cl-AlCl3 ionic liquid has been investigated. Pure 1,2-aceanthryenedione, which is used as intermediate of functional aromatic polymer material, was obtained by recrystalling the reaction mixture with aether and was determined by GC/MS, 1↑HNMR and FTIR analysis. The influences of various parameters, such as the contents of AlCl3 in [Emim]Cl-AlCl3, the amount of acylation agent, amount of [Emim]Cl-AlCl3, reaction temperature and reaction time were investigated. The optimum conditions were as follows: the molar fraction of AlCl3 in ionic liquid [x(AlCl3)] being 0.67, molar ratio of ionic liquid to anthracene being 2:1, molar ratio of oxalyl chloride to anthracene being 2:1, reaction temperature being 40℃ and reaction time being 6h. Under above conditions, the yield and selectivity of 1,2-aceanthrylenedione can reach 91.5% and 98.3% respectively. Further more, [Emim]Cl-AlCl3 ionic liquid, compared with metal halides such as AlCl3, was found to catalyze the reaction as a novel environmental friendly catalyst and solvent and can be reused.  相似文献

A new cyclophane featuring two opposite anthracene units linked in 9,10-positions has been synthesized thanks to the template effect of the Me4N(+) ion. It forms pseudorotaxane complexes with alkylviologen ions and undergoes a fast and reversible reaction with tetracyanoethylene. A quantitative analysis has been carried out of the formation of Diels-Alder adducts, whose distribution can be controlled by host-guest complexation. 分子印迹聚合物(MIP)在合成与催化方面的应用受到越来越多的关注.传统的本体聚合生成的聚合物往往具有活性中心的利用率低、活性中心不均一、底物难以到达等问题.由于纳米材料具有很高的比表面积,将MIP制备成纳米级别的材料将有可能解决这些问题.本研究的目的是在纳米材料上构建分子印迹微反应器(MIM).通过多壁碳纳米管(MWNTs)上羟基的酯化反应在碳纳米管表面枝接双键,以蒽和马来酸的加成反应产物为模板分子,甲基丙烯酸(MAA)为功能单体,乙二醇二甲基丙烯酸酯(EDMA)为交联剂,通过微波辐射聚合在碳纳米管表面枝接一层厚度约为30 nm 的分子印迹催化剂. 该催化剂对蒽和马来酸的加成反应具有明显的催化作用,反应开始后的180min内,催化反应速率是不加催化剂反应速率的1.77倍. 催化动力学遵守Michaelis-Menten方程,催化反应最大速率(vmax)为0.713 umolL-1S-1,米氏常数(KM)为17735.24 umolL-1.  相似文献

Generation-4 polyamidoamine (PAMAM) dendrimers were surface-functionalized with azides or alkynes and conjugated to one DNA strand. DNA-controlled self-assembly of alternating azide and alkyne dendrimers on a DNA template enabled the coupling of the dendrimers by the azide-alkyne "click" reaction to form covalently coupled dimers, trimers, and tetramers. Polymerization of the DNA-dendrimer conjugates was also demonstrated, as well as assembly in a circular structure on DNA origami and imaging by atomic force microscopy.  相似文献

Interactions of double-stranded nucleic acids with cell surface proteins, which are involved in binding and transport of extracellular nucleic acids, were studied by the photoaffinity modification with a binary system of oligonucleotide conjugates. The photoreactive double-stranded complex involved an oligonucleotide template and two complementary to adjacent sequences oligonucleotide conjugates. One conjugate contained a photoreagent, viz., 4-azido-2,3,5,6-tetrafluorobenzaldehyde N-(3-aminopropionyl)hydrazone, at the terminus located in proximity to the terminus of another conjugate containing the sensitizer, viz., 9-aminomethylanthracene. Binding of photoreagent and the sensitizer to a single-stranded template yields the photoreactive center. Upon irradiation with visible light (400—580 nm), this photoreactive double-stranded complex forms covalent cross-linkages with oligonucleotide-binding surface proteins of eucaryotic SPEV cells.  相似文献

We herein report the modular design and the synthesis of new molecular conjugates, which can combine a cell targeting function (ligand domain) with potential cytotoxic molecules (effector domain). The present approach utilizes a cyclic peptide template, Chemoselectively Addressable Template (CAT) as a key intermediate. These CAT molecules exhibit two independent and chemically addressable domains which permits the sequential and regioselective assembly of different ligand and/or effector domains. The attachment of various units to the template was achieved by the formation of iterative oxime bonds. The chemoselective oxime bonds were produced by the reaction of glyoxylyl aldehyde groups obtained from serine precursors. The process was further developed to prevent transoximation reactions. RAFT(c[-RGDfK-])4, a synthetic vector targeting the tumor-associated a alpha(V)beta3 integrin was prepared and coupled to either a cytotoxic peptide or oligonucleotide as an illustration of present approach. The potential application of this approach has been further demonstrated by the synthesis of high molecular weight compounds such as RAFT(c[-RGDfK-])16, a alpha(V)beta3-targeting ligand of high valency index.  相似文献

A practical and efficient method to synthesize iptycene quinones has been developed. As a result, a series of pentiptycene quinones 8-16 were conveniently synthesized by one-pot reaction of triptycene quinone 4 or 5 with anthracene 1 or its derivatives 2-3 in refluxing acetic acid in the presence of p-chloranil, followed by CAN oxidative demethylation. Similarly, a series of heptiptycene quinones 17-23 with U-shaped cavities were achieved with pentiptycene quinone 10 and triptycene diquinone 6 as precursors. Non-iptycene triquinones 24 with one tweezer-shaped cavity and 25 with two U-shaped cavities were synthesized by one-pot reactions of anthracene with pentiptycene triquinones 16a and 16b, respectively. Non-iptycene triquinone 26 with a dendritic structure was conveniently obtained by the reaction of anthracene with either pentiptycene diquinone 12 or triptycene triquinone 7. The structures of regioisomers 16a and 16b were determined by the single-crystal structure analysis of 16b. The structures of other regioisomers, including heptiptycene tetraquinones 19a/19b/19c and heptiptycene triquinones 23a/23b, were identified by comparative reactions.  相似文献

The synthesis and spectroscopic properties of new cyclam-type ligands 5,7-dimethyl-6-R-1,4,8,11-tetraazacyclotetradecane (L), where R is a pendant chromophore such as an anthracene derivative, are reported. These ligands were prepared according to a nickel(II) template procedure, and the X-ray crystal structures of several Ni(II) intermediates are described. Reaction of the free base ligands L with CrCl(3)x3THF resulted in facile formation of trans-[Cr(L)Cl(2)]Cl complexes, and the structures and spectroscopic characterizations of these complexes are also described. Examination of the photophysical properties of trans-[Cr(L)Cl(2)]Cl solutions at 77 K demonstrated the emission spectra to be dominated by phosphorescence from the ligand field doublet of the chromium(III) center. This also applies to the Cr(III) complex trans-[Cr(mac)Cl(2)]Cl, where mac is the anthracene derivative 5,7-dimethyl-6-anthracenylcyclam. Excitation into the pi-pi(*) states of the anthracene leads to marked quenching of the fluorescence from this chromophore and sensitized phosphorescence from the metal-centered doublet state.  相似文献

Instead of azides, which can be explosive and poorly soluble in nonpolar solvents, azabicycloheptadienes can be used to synthesize nitrides. The reaction of diiodide 1 with the lithium amide 3 provides the red nitride 2 in 60 % yield with loss of lithium iodide and anthracene. We report a general approach to bimodify gold nanoparticles (GNPs) with two different DNA strands via DNA template reaction. Two thioctic acid modified DNA strands, one at 5' end and one at 3' end, were attached to GNPs through bivalent thiol-gold bond. By sequence design, assemblies of 5 nm GNPs chains, 10 nm GNPs chains and alternative arrangement of 5 and 10 nm GNPs could be achieved. Gel electrophoresis, transmission electron microscope (TEM), UV-vis spectra were used to characterize the assemblies. It is believed that this new kind of bimodified GNPs with two different DNA strands at different ends would enrich the toolbox of DNA-GNP conjugates and provide diverse selectivity for further assembly.  相似文献

The anthracene cyclophane bis-anthracene (BA) can undergo a [4 + 4] photocycloaddition reaction that results in a photodimer with two cyclobutane rings. We find that the subsequent dissociation of the dimer, which involves the rupture of two carbon-carbon bonds, is strongly accelerated by the application of mild pressures. The reaction kinetics of the dimer dissociation in a Zeonex (polycycloolefin) polymer matrix were measured at various pressures and temperatures. Biexponential reaction kinetics were observed for all pressures, consistent with the presence of two different isomers of bis(anthracene). One of the rates showed a strong dependence on pressure, yielding a negative activation volume for the dissociation reaction of ΔV(++) = -16 ?(3). The 93 kJ/mol activation energy for the dissociation reaction at ambient pressure is lowered by more than an order of magnitude from 93 to 7 kJ/mol with the application of modest pressure (0.9 GPa). Both observations are consistent with a transition state that is stabilized at higher pressures, and a mechanism for this is proposed in terms of a two-step process where a flattening of the anthracene rings precedes rupture of the cyclobutane rings. The ability to catalyze covalent bond breakage in isolated small molecules using compressive forces may present opportunities for the development of materials that can be activated by acoustic shock or stress.  相似文献
