The natural dyes, and dye sources, in two seventeenth century Arraiolos carpets from the National Museum of Machado de Castro were analysed by high-performance liquid chromatography with UV–vis diode array detection (HPLC–DAD) and HPLC–mass spectrometry (LC–MS). Weld (Reseda luteola L.), indigo and spurge flax (Daphne gnidium L.) were found to be the dye sources, in agreement with original dyeing recipes collected during the nineteenth century. In order to fully characterize the plant sources, LC–MS conditions were optimized with plant extracts and the chromatographic separation and mass detection were enhanced. Extraction of the dyes, in the Arraiolos carpet samples, was performed using mild conditions that avoid glycoside decomposition. For the blues a dimethylformamide solution proved to be efficient for indigotin recovery. For all the other colours, an improved mild extraction method (with oxalic acid, methanol, acetone and water) was used, enabling to obtain the full dye source fingerprint, namely the flavonoid glycosides in the yellow dyes.  相似文献

A reversed-phase HPLC method has been developed for identification and quantitation of nine natural quinone dyes and applied to historical textile fibres. A Purospher RP18e column was used with a convex gradient of methanol in a mobile phase of 0.1 M aqueous citrate buffer (pH 2.5) and spectrophotometric diode-array detection at 270 nm. For identification of alizarin, purpurin and xanthopurpurin, occurring together in the madder plant, an isocratic method was used with a methanol-0.2 M acetate buffer (pH 4.3) (75:25) as the mobile phase. After an acid extraction of textile fibres and the analysis of the extracts, alizarin and purpurin were identified and quantitated in three fibres.  相似文献

The extraction and separation of dyes present on textile fibers offers the possibility of enhanced discrimination between forensic trace fiber evidence. An automated liquid sample handling workstation was programmed to deliver varying solvent combinations to acid-dyed nylon samples, and the resulting extracts were analyzed by an ultraviolet/visible microplate reader to evaluate extraction efficiencies at different experimental conditions. Combinatorial experiments using three-component mixture designs varied three solvents (water, pyridine, and aqueous ammonia) and were employed at different extraction temperatures for various extraction durations. The extraction efficiency as a function of the three solvents (pyridine/ammonia/water) was modeled and used to define optimum conditions for the extraction of three subclasses of acid dyes (anthraquinone, azo, and metal complex) from nylon fibers. The capillary electrophoresis analysis of acid dye extracts is demonstrated using an electrolyte solution of 15 mM ammonium acetate in acetonitrile/water (40:60, v/v) at pH 9.3. Excellent separations and discriminating diode array spectra are obtained even for dyes of similar color. High-performance liquid chromatography coupled with spectrophotometric and electrospray mass spectrometric detection (HPLC-UV-Vis-ESI MS) was used for characterization of natural dyes present in historical art works. The gradient program was developed for identification of 29 colorants of various polarities. Dual detection system (UV-Vis and ESI MS) allowed differentiation of all compounds, even if they were not completely separated. This enabled examination of more color compounds over a substantially shorter time in comparison with previously recommended methods. Moreover, for extraction of colorants from historical textiles a two-step sequential procedure was proposed, excluding evaporation used in earlier procedures. The developed method was successfully applied to identification of indigotin, carminic, kermesic, flavokermesic, dcII, dcIV, dcVII, and ellagic acids as well as luteolin, apigenin, and genistein in red, violet, and green fibers taken from three selected historical chasubles which belong to the collection of the Wawel Cathedral treasury (Cracow, Poland). Italian textiles from the fifteenth and sixteenth centuries, of which chasubles were made, were dyed with a limited number of dyestuffs, consistently used for all batches of fabrics. The obtained results also allowed confirmation of the structure of the so-called "dcII" component of cochineal as a C-glucose derivative of flavokermesic acid.  相似文献

The aim of this paper is to study the effect of the pH on the extraction of sinapic acid and its derivatives from mustard seed meal. Solutions of acidic pH (pH 2), basic pH (pH 12) and distilled water (uncontrolled pH ~ 4.5) were tested at different percentages of ethanol. The maximum extraction yield for sinapic acid (13.22 µmol/g of dry matter (DM)) was obtained with a buffered aqueous solution at pH 12. For ethyl sinapate, the maximum extraction yield reached 9.81 µmol/g DM with 70% ethanol/buffered aqueous solution at pH 12. The maximum extraction yield of sinapine (15.73 µmol/g DM) was achieved with 70% ethanol/buffered aqueous solution at pH 2. The antioxidant activity of each extract was assessed by DPPH assay; the results indicated that the extracts obtained at pH 12 and at low ethanol percentages (<50%) exhibit a higher antioxidant activity than extracts obtained at acidic conditions. Maximum antioxidant activity was reached at pH 12 with buffer solution (11.37 mg of Trolox Equivalent/g DM), which confirms that sinapic acid-rich fractions exhibit a higher antioxidant activity. Thus, to obtain rich antioxidant extracts, it is suggested to promote the presence of sinapic acid in the extracts.  相似文献

Textiles excavated from Scottish sites belonging now to the collections of the National Museums of Scotland, including seventeenth century textiles from peat bogs in the Scottish Highlands and Islands, were selected for analysis by high performance liquid chromatography with photodiode array detection (PDA HPLC) to detect whether any dyes remained and, if so, to identify their biological sources. Dye components were identified in 36 of the 81 samples analysed. Although it was not possible to identify the exact sources of the dyestuffs because of the wide-spread occurrence of these natural dyes components, the study has shown that textiles previously not thought to have been coloured had detectable traces of dye. Before the historical textiles were analyzed, an improved extraction procedure that combined the routine acid hydrolysis method with one using dimethylformamide (DMF) was applied. The DMF method enabled increased recovery of major flavonoid and anthraquinoid compounds, and very high efficiency of recovery of indigotin even in textiles with no colour visible, thereby complementing the acid hydrolysis method already in use. Extracts from historical thread samples were analysed by PDA HPLC using a reversed-phase gradient system comprising of a C18 column (150 mm x 4.6 mm i.d., 25 +/- 1 degrees C) with water, methanol and o-phosphoric acid at an eluent flow rate of 1.2 ml/min. A preliminary investigation to improve the detection limits further for a selection of natural dyes was made by comparing results from the 4.6mm internal diameter (i.d.) column with a narrow bore C18 column (2.1 mm i.d.). Pigments are among the most important components of historical paintings and textiles and their nature provides the unique character of color. They can be divided into two main groups: inorganic and organic, extracted from plants or animals. Their identification is a necessary stage in the conservation of art objects. Reversed-phase liquid chromatography with electrospray ionization mass spectrometry (ESI-MS) and UV/visible spectrophotometric methods were elaborated for the identification of indigoid (indigo, indirubin, isoindigo, isoindirubin) color components of natural dyestuffs and their natural or synthetic precursors (indican, isatin, indoxyl, 2-indolinone). ES-MS offers detection limits in the range 0.03-5.00 microg ml(-1) for the color compounds examined. The method developed made it possible to identify indigo and its isomers in genuine Indian indigo, indigo from woad and Tyrian Purple. It was applied to the identification of natural dyes on fiber from a 19th century Japanese tapestry, 'Cranes in the landscape'. A procedure based on freezing and grinding of a sample before the extraction of dyes from the textile was developed. The components of the extract obtained were identified after acidic hydrolysis as indigotin and methylene blue.  相似文献

A simple and rapid liquid chromatographic with diode-array UV-vis spectrophotometric detection (HPLC-DAD) method for identification of natural dyes has been developed. Chromatographic retention of carminic acid, indigotin, crocetin, gambogic acid, alizarin and purpurin has been studied. The mobile phase consisted of 40 mM SDS-10 mM phosphate buffer solution (pH 2.3)-0.1% TFA (eluent A) and acetonitrile (eluent B) using a programmed gradient (5% B to 95% B). Analyses were carried out on a Phenomenex, Luna 5u NH2 100(a) column (250 mm x 4.60 mm i.d., 5 microm particle) and the operating conditions were: 0.6 ml min(-1) flow rate, 20 microl volume injection and 35 degrees C column temperature. Extracts of samples of natural dyes taken from historical maps belonging to The Royal Chancellery Archives in Granada were successfully analyzed using the proposed method including a new technique for sampling.  相似文献

Organic colorants contained in 30 textiles (16th to early 20th century) from the monastery of Simonos Petra (Mount Athos) have been investigated using high-performance liquid chromatography equipped with diode-array detection and mass spectrometry (HPLC–DAD–MS). The components of natural dyes identified in samples treated by the standard HCl dyestuff extraction method were: alizarin, apigenin, butein, carminic acid, chrysoeriol, dcII, dcIV, dcVII, ellagic acid, emodin, fisetin, flavokermesic acid, fustin, genistein, haematein derivative (Hae′), indigotin, indirubin, isoliquiritigenin, isorhamnetin, kaempferide, kaempferol, kermesic acid, luteolin, naringenin, purpurin, quercetin, rhamnazin, rhamnetin, sulfuretin, and type B and type C compounds (last two are markers for Caesalpinia trees). Early, semi-synthetic dyes, for example indigo carmine, fuchsin components, and rhodamine B were identified in objects dated late 19th to early 20th century. A dyestuff extraction method which involves use of TFA, instead of HCl, was applied to selected historical samples, showing that the mild method enables efficient extraction of weld (Reseda luteola L.) and dyer’s broom (Genista tinctoria L.) glycosides. The marker compound (Hae′) for logwood (Haematoxylum campechianum L.) identification after treatment with HCl was investigated by liquid chromatography coupled to mass spectrometry (LC–MS) in negative electrospray ionization (LC–MS-ESI) mode. LC–MS in negative atmospheric pressure chemical ionization (LC–MS-APCI) mode was used, probably for the first time, to investigate cochineal (Dactylopius coccus Costa) samples. Positive electrospray ionization (LC–MS-ESI+) mode was used for identification of fuchsin components. Detailed HPLC–DAD studies were performed on young fustic (Cotinus coggygria Scop.) and Persian berries (Rhamnus trees).  相似文献

Nelumbo nucifera leaves are rich source of natural wax possessing super-hydrophobic properties. It provides protection to them from ecological turbulences and climatic wear and tear. In this study, various experiments have been conducted to observe the yield of extraction and the determination of various functional groups, which are present in natural wax, derived from Nelumbo nucifera leaves. The natural wax has been extracted from lotus leaves through non-polar (hexane) and polar (ethanol) solvent via different extraction methods. The superhydrophobic wax has been successfully extracted with hexane. Whereas, ethanol did not extract the water-repellent wax of lotus leaf. Considering the cumulative amount, i.e. (desired + undesired), the maceration shows the extraction of 2.9% (%w/w, through hexane) and 10.2% (%w/w, through ethanol), while it was found 2.5% (%w/w, cycle period 15 min) and 9.0% (%w/w, cycle period 26 min) respectively, in case of Soxhlet extraction technique. For this specific case of natural wax recovery from biomass (lotus leaf), the maceration (traditional method) resulted a little bit superior extraction yield in comparison to the Soxhlet extraction method for extraction of crude wax. In the case of non-polar solvent (hexane), an extraction yield of 1.97% (%w/w) through maceration method was observed while in the case of non-polar solvent (ethanol), an extraction yield of 1.62% (%w/w) through Soxhlet extraction was observed. The TLC analysis on both types of extracts was performed. For the detection of various hydrocarbon chains in the crude wax extracts, FTIR was also performed. Topography of wax surface and wax-coated waterproof fabric was compared through SEM.  相似文献

Natural dyestuffs used for painting or dyeing of textiles are complex mixtures of compounds of various chemical properties. Proper identification of the dye used by a painter and, even better, its origin is possible only when its compositional 'fingerprint' can be evaluated. For this reason gradient program for liquid chromatographic separation of 16 color compounds--components of natural blue dyes: elderberry, logwood and indigo--has been developed. Two detector systems were used simultaneously: UV-Vis spectrophotometry (at 280, 445, 520 and 600 nm) and ESI mass spectrometry (positive and negative SIM mode). It was found that fragmentation observed in ESI-MS is affected not only by ion source parameters, but also by chromatographic conditions, especially in case of the less stable substances: cyanidin glucosides, tannic acid, rutin and hematoxylin. Examination of characteristic dissociation pathways of the compounds under investigation after direct admission into ion source or after chromatographic separation allowed to select proper ions for SIM detection and to develop novel and efficient reversed phase high performance liquid chromatographic (RP-HPLC)-UV-Vis/ESI-MS method for the analysis of natural blue dyes. The procedure was successfully applied for identification of indigotin and carminic acid-main colorants extracted from a fiber taken from the blue-red 'Italian' tapestry (the collection of the National Museum in Warsaw, Poland).  相似文献

Spectrophotometric determinations of copper, nickel, cobalt, iron, and manganese, based on the chloroform extraction of the metal pyridine thiocyanates, have been investigated. Optimum conditions require the pH of the aqueous solution to be in the range about 5–8; tartaric acid is used to prevent precipitation of hydrous oxides. An excess of pyridine must be used because chloroform readily extracts pyridine from the aqueous solution. Results are improved by making the extraction from a solution of high ionic strength (2 or above), which is provided by magnesium nitrate. Perchlorate decreases the absorbance, but the effect is essentially constant over a perchlorate concentration range of 0.8 to 2M. Although the metal pyridine thiocyanates are extracted by benzene, substituted benzenes, and halogenated hydrocarbons, chloroform is superior to other solvents in extraction efficiency and in colour stability of the extracted species. An example is given of the simultaneous determination of copper, nickel, cobalt, and iron in the same solution. Anions that also form metal pyridine compounds must be absent.  相似文献

The efficiency of eight different procedures used for the extraction of natural dyes was evaluated using contemporary wool samples dyed with cochineal, madder, woad, weld, brazilwood and logwood. Comparison was made based on the LC-DAD peak areas of the natural dye's main components which had been extracted from the wool samples. Among the tested methods, an extraction procedure with Na(2)EDTA in water/DMF (1:1, v/v) proved to be the most suitable for the extraction of the studied dyes, which presented a wide range of chemical structures. The identification of the natural dyes used in the making of an eighteenth century Arraiolos carpet was possible using the Na(2)EDTA/DMF extraction of the wool embroidery samples and an LC-DAD-MS methodology. The effectiveness of the Na(2)EDTA/DMF extraction method was particularly observed in the extraction of weld dye components. Nine flavone derivatives previously identified in weld extracts could be identified in a single historical sample, confirming the use of this natural dye in the making of Arraiolos carpets. Indigo and brazilwood were also identified in the samples, and despite the fact that these natural dyes were referred in the historical recipes of Arraiolos dyeing, it is the first time that the use of brazilwood is confirmed. Mordant analysis by ICP-MS identified the widespread use of alum in the dyeing process, but in some samples with darker hues, high amounts of iron were found instead.  相似文献

Designed experiments based on a simplex mixture design were employed to explore the effects of three solvent components (water, formic acid, and aqueous acetic acid), extraction time, and extraction temperature for the automated microextraction of basic (cationic) dyes from acrylic fibers. Extractions were conducted by an automated liquid handling system, and dye extraction was evaluated using a UV/visible microplate reader. Highest extraction efficiency for two subclasses of basic dyes (methine and azo) from acrylic fibers was achieved with an extraction solvent containing 88% formic acid/12% water. Cationic dyes were analyzed by capillary electrophoresis using a 45 mM ammonium acetate buffer in acetonitrile–water at pH 4.7. The utility of microextraction combined with capillary electrophoresis–mass spectrometry for analysis of extracts from trace fibers was demonstrated by the detection and characterization of three basic dyes extracted from a 2-mm length of single acrylic fiber.  相似文献

A simple and rapid capillary electrophoretic method with UV detection (CE-UV) has been developed for the identification of five natural dyes namely, carmine, indigo, saffron, gamboge and Rubia tinctoria root. The separation was performed in a fused-silica capillary of 64.5 cm length and 50 microm id. The running buffer was 40 mM sodium tetraborate buffer solution (pH 9.25). The applied potential was 30 kV, the temperature was 25 degrees C and detections were performed at 196, 232, 252, 300 and 356 nm. The injections were under pressure of 50 mbar during 13 s. The method was applied to the identification of carminic acid, gambogic acid, crocetin, indigotin, alizarin and purpurin in the collection of drawings and maps at the Royal Chancellery Archives in Granada (Spain). The method was validated by using HPLC as a reference method.  相似文献

A simple and rapid HPLC method for determination of chlorogenic acid (5-O-caffeoylquinic acid) in mate tea extracts was developed and validated. The chromatography used isocratic elution with a mobile phase of aqueous 1.5% acetic acid-methanol (85:15, v/v). The flow rate was 0.8 mL/min and detection by UV at 325 nm. The method showed good selectivity, accuracy, repeatability and robustness, with detection limit of 0.26 mg/L and recovery of 97.76%. The developed method was applied for the determination of chlorogenic acid in mate tea extracts obtained by ethanol extraction and liquid carbon dioxide extraction with ethanol as co-solvent. Different ethanol concentrations were used (40, 50 and 60%, v/v) and liquid CO? extraction was performed at different pressures (50 and 100 bar) and constant temperature (27 ± 1 °C). Significant influence of extraction methods, conditions and solvent polarity on chlorogenic acid content, antioxidant activity and total phenolic and flavonoid content of mate tea extracts was established. The most efficient extraction solvent was liquid CO? with aqueous ethanol (40%) as co-solvent using an extraction pressure of 100 bar.  相似文献

Arsenic extraction in natural and spiked soil samples was studied using two main types of extractants. The extractants were phosphoric acid used alone and with the addition of reducing agents (ascorbic acid and hydroxylammonium hydrochloride). The stability of arsenite (As(III)), arsenate (As(V)), monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA) in the extracts over time and under different storing conditions was assessed by means of LC-HG-AFS. A protocol was devised to extract arsenic species from soils. The protocol assures the stability of the original sample species in the extract proposed by using an extraction solution of phosphoric acid with ascorbic acid and by purging the extracts after microwave extraction. The total As, Mn and Fe content in aqua regia and in phosphoric extracts was also measured by ICP-AES in order to perform the mass balance.  相似文献

A method for the analyses of pyridine in environmental samples is described. For soil samples a distillation procedure followed by an extraction, an acidic extraction or a Soxhlet extraction can be used. For water samples a distillation procedure followed by extraction can be employed. Deuterated pyridine is used as an internal standard and the extracts are analyzed by GC-MS. The recoveries of the methods are higher than 80%; the detection limits for pyridine are 0.01 mg/kg for soil samples and 0.2 g/l for water samples.  相似文献

Liquid chromatography (LC) combined with ultraviolet-visible (UV-Vis) and mass spectrometric (MS) detection was utilized to study the chemical components present in extracts of natural dyes originating from fiber samples obtained from Coptic textiles from Early Christian Art Collection of National Museum in Warsaw. Chromatographic retention, ionization, UV-Vis and mass spectra of twenty selected dye compounds of flavanoid-, anthraquinone- and indigo-types were studied. Most of the investigated compounds could be ionized by positive and negative ion electrospray ionization. Difficulties with the ionization by electrospray were experienced for indigotin and brominated indigotins, but these were ionized by atmospheric pressure chemical ionization. Mass spectrometric detection, utilizing different scanning modes of a triple quadrupole mass spectrometer, combined with the UV-Vis detection was demonstrated to be a powerful approach to detection and identification of dyes in the extracts of archeological textiles. Using this approach the following compounds were identified in the extracts of Coptic textiles: luteolin, apigenin, rhamnetin, kaempferol, alizarin, purpurin, xanthopurpurin, monochloroalizarin, indirubin, and so the type of dye that was utilized to dye the textiles could be identified. Detection capabilities for several dye-type analytes were compared for the UV-Vis and mass spectrometric detection. The signal-to-noise ratios obtained for luteolin, apigenin, and rhamnetin were higher for the MS detection for most of the examined sample extracts. Purpurin, alizarin, and indirubin showed similar signal-to-noise ratios for UV-Vis and mass spectrometric detection.  相似文献

Different methods for the extraction of Dactylopius coccus Costa, Rubia tinctorum L., Isatis tinctoria L., Reseda luteola L., Curcuma longa L. and Cotinus coggygria Scop. from wool fibres are investigated using high-performance liquid chromatography with diode array detector (HPLC-DAD). The efficiencies of five extraction methods which include the use of HCl (widely used extraction method), citric acid, oxalic acid, TFA and a combination of HCOOH and EDTA are compared on the basis of the (a) number, (b) relative quantities, measured as HPLC peak areas and (c) signal-to-noise ratios (S/N) of the compounds extracted from the wool substrates. Flavonoid glycosides and curcuminoids contained in R. luteola L. and C. longa L., respectively, according to liquid chromatography with mass spectrometry (LC-MS) identifications, are not detected after treating the fibres with HCl. All the other milder methods are successful in extracting these compounds. Experiments are performed using HPLC-DAD to compare the HPLC peak areas and the S/N of the following extracted compounds: indigotin, indirubin, curcumin, demethoxycurcumin, bisdemethoxycurcumin, fisetin, sulfuretin, luteolin, luteolin-7-O-glucoside, apigenin, carminic acid, alizarin, puruprin and rubiadin. It is shown that the TFA method provides overall the best results as it gives elevated extraction yields except for fisetin, luteolin, apigenin and luteolin-7-O-glucoside and highest S/N except for fisetin and luteolin-7-O-glucoside. It is noteworthy that treatment of the fibres with the typical HCl extraction method results overall in very low S/N. The TFA method is selected for further studies, as follows. First, it is applied on silk dyed samples and compared with the HCl method. The same relative differences of the TFA and HCl methods observed for the wool dyed samples are reported for the silk dyed samples too, except for rubiadin, luteolin and apigenin. Thus, in most cases, the nature of the substrate (wool or silk) appears to have negligible effects on the relative difference of the two extraction methods. Second, the selected TFA method is applied to treat wool and silk historical samples extracted from textiles of the Mamluk period, resulting in the identification of several colouring compounds. In all extraction methods mentioned above, DMSO is used to dissolve the dyes, after acid treatment.  相似文献
