Reducibly degradable hydrogels of poly(N‐isopropylacrylamide) (PNIPAM) and poly(N,N‐dimethylaminoethyl methacrylate) (PDMAEMA) were synthesized by the combination of reversible addition‐fragmentation chain transfer (RAFT) polymerization and click chemistry. The alkyne‐pending copolymer of PNIPAM or PDMAEMA was obtained through RAFT copolymerization of propargyl acrylate with NIPAM or DMAEMA. Bis‐2‐azidyl‐isobutyrylamide of cystamine (AIBCy) was used as the crosslinking reagent to prepare reducibly degradable hydrogels by click chemistry. The hydrogels exhibited temperature or pH stimulus‐responsive behavior in water, with rapid response, high swelling ratio, and reproducible swelling/shrinkage cycles. The loading and release of ceftriaxone sodium proved the feasibility of the hydrogels as the stimulus‐responsive drug delivery system. Furthermore, the presence of disulfide linkage in AIBCy favored the degradation of hydrogels in the reductive environment. © 2010 Wiley Periodicals, Inc. Macroporous, temperature‐sensitive poly(N‐isopropylacrylamide) (PNIPAAm) hydrogels were synthesized with poly(ethylene glycol)s (PEGs; molecular weight = 2000–6000) as the pore‐forming agents. The influence of the molecular weight and PEG content on the responsive kinetics of these macroporous hydrogels was investigated. The PEG‐modified PNIPAAm hydrogels were characterized by the swelling ratio, deswelling–reswelling kinetics, Fourier transform infrared, and differential scanning calorimetry. The morphology of these hydrogels was analyzed with scanning electron microscopy. The prepared macroporous hydrogels exhibited some unique properties in comparison with the gels with low molecular weight PEGs (molecular weight < 2000) as the pore‐forming agents. In addition, a preliminary study on the controlled release of bovine serum albumin from these macroporous hydrogels was carried out. © 2002 Wiley Periodicals, Inc. The strategy for in situ chemical gelation of poly(N‐isopropylacrylamide‐co‐hydroxylethyl methacrylate) [P(NIPAAm‐co‐HEMA)]‐based polymers was demonstrated. Two types of new P(NIPAAm‐co‐HEMA) derivatives with alkyne and azide pendant groups, respectively, were prepared. When the solutions of the two derivatives were mixed together, a crosslinking reaction, a type of Huisgen's 1,3‐dipolar azide‐alkyne cycloaddition, in the presence of Cu(I) catalyst occurs. The morphology, equilibrium swelling ratio, swelling kinetics, and temperature response kinetics of the in situ gelated hydrogels were studied. In comparison with the conventional PNIPAAm hydrogel, because of the spatial hindrance of polymeric chains, the resulted hydrogels had a macroporous structure as well as a fast shrinking rate. The strategy described here presents a potential alternative to the traditional synthesis techniques for the in situ formation of thermoresponsive hydrogels. © 2008 Wiley Periodicals, Inc. A consecutive radical addition‐coupling reaction induced by spin‐trapping agent is applied to produce degradable multisegmented polymer using α,ω‐dibromo polymer as a precursor. The macroradical generated by single electron transfer process catalyzed by Cu/PMDETA from α,ω‐dibromo polymer can be efficiently captured by 2‐methyl‐2‐nitrosopropane (MNP), which results in nitroxide radical. The in situ formed nitroxide radical immediately undergoes cross‐coupling reaction with polymeric radical, generating block polymer bridged with alkoxyamine moiety. The consecutive radical addition‐coupling reaction generates multisegmented polymer via step‐growth mechanism. Different multisegmented polymers have been prepared from α,ω‐dibromo‐PS, PtBA, and PtBA‐PS‐PtBA. The block number of multisegmented polymers can be tailored by varying the feed ratio of α,ω‐dibromo precursor to MNP. The multisegmented polymer can be degraded in the presence of hydrogen atom donor or air, and the molecular weight distribution transformed back into shape of its original precursor as it is conjugated by alkoxyamine moieties. © 2010 Wiley Periodicals, Inc. A facile synthetic route for the preparation of dicationic ethylene glycol based‐ionic liquids (ILs) via the azide/alkyne “click” reaction is presented. The copper(I) catalyzed, microwave‐assisted azide/alkyne “click” reaction between diazido‐ethylene glycols and the corresponding alkyne containing IL‐head group enables a simple preparation of different sets of poly(ethylene glycol)‐based ILs. Beside tetra‐ and hexa(ethylene glycol)‐based ILs, also oligomeric (Mn = 400 g/mol) and polymeric ILs (Mn up to 1550 g/mol) could be prepared in good yield and with full conversion of the ionic head group. The prepared ILs were extensively characterized via NMR spectroscopy and ESI‐time‐of‐flight (TOF) mass spectroscopy, revealing the formation of multiply charged ions in the negative mode. Dual temperature‐ and pH‐sensitive hydrogels composed of N‐isopropylacrylamide (NIPAM) and 2‐acrylamido‐2‐methyl‐propanosulfonic acid (AMPS) were prepared by free‐radical crosslinking copolymerization in aqueous solution at 22 °C. The mole percent of AMPS in the comonomer feed was varied between 0.0 and 7.5, while the crosslinker ratio was fixed at 5.0/100. The effect of AMPS content on thermo‐ and pH‐ induced phase transitions as well as equilibrium swelling/deswelling, interior morphology and network structure was investigated. The volume phase transition temperature (VPT‐T) was determined by both swelling/deswelling measurements and differential scanning calorimetry (DSC) technique. In addition, the volume phase transition pH (VPT‐pH) was detected from the derivative of the curves of the swelling ratio (dQv/dpH) versus pH. The polymer‐solvent interaction parameter (χ) and the average molecular mass between crosslinks ( ) of hydrogels were calculated from swelling ratios in buffer solutions at various pHs. The enthalpy (ΔH) and entropy (ΔS) changes appearing in the χ parameter of hydrogels were also determined by using the modified Flory–Rehner equation. The negative values for ΔH and ΔS indicated that the hydrogels had a negative temperature‐sensitive property in water, that is, swelling at a lower temperature and shrinking at a higher temperature. It was observed that the experimental swelling data of hydrogels at different temperature agreed with the modified Flory‐Rehner approach based on the affine network model. © 2008 Wiley Periodicals, Inc. In this work, a series of biodegradable and pH‐responsive hydrogels based on polyphosphoester and poly(acrylic acid) are presented. A novel biodegradable macrocrosslinker α‐methacryloyloxyethyl ω‐acryloyl poly(ethyl ethylene phosphate) (HEMA‐PEOP‐Ac) was synthesized by first ring‐opening polymerization of the cyclic monomer 2‐ethoxy‐2‐oxo‐1,3,2‐dioxaphospholane using HEMA as the initiator and Sn(Oct)2 as catalyst, and subsequent conversion of hydroxyl into vinyl group. The hydrogels were then fabricated by the copolymerization of the macromonomer with acrylic acid, and their swelling/deswelling and degradation behaviors were investigated. The results demonstrated that the crosslinking density and pH values of media strongly influenced both the swelling ratio and the degradation rate of the hydrogels. The rheological properties of these hydrogels were also studied from which the storage modulus (G′) showed clear dependence on the crosslinking density. MTT and “live/dead” assay showed that these hydrogels were compatible to fibroblast cells, not exhibiting apparent cytotoxicity even at high concentrations. Moreover, in vitro bovine serum albumin release from these hydrogels was also investigated, and it could be found that the release profiles showed a burst effect followed by a continuous release phase, and the release rate was inversely proportional to the crosslinking density of hydrogels. © 2010 Wiley Periodicals, Inc. A new strategy is developed to prepare both α,ω‐dithiol and α,ω‐divinyl linear telechelic polythiolether oligomers by visible light induced thiol‐ene chemistry in the presence of a fac‐Ir(ppy)3 photoredox catalyst. Polythiolether oligomers of well‐defined end groups and controlled molecular weights have been successfully synthesized at varying monomer molar ratios of 1,4‐benzenedimethanethiol (BDMT) to diethylene glycol divinyl ether (DEGVE). 1H NMR and MALDI‐TOF MS analyses demonstrate that as‐prepared polythiolethers possess high end‐group fidelity, which is further supported by the successful polyaddition of polythiolethers bearing α,ω‐dithiol and α,ω‐divinyl groups. For example, with the α,ω‐dithiol‐ (Mn = 1900 g mol?1, PDI = 1.25) and α,ω‐divinyl‐terminated (Mn = 2000 g mol?1, PDI = 1.29) polythiolethers as macromonomers, the molecular weight of resulting polythiolether is up to 7700 g mol?1 with PDI as 1.67. The reactivity of the terminal thiol group is further confirmed by the addition reaction with N‐(1‐pyrenyl)maleimide. UV‐vis spectra and fluorescene measurements suggest that fac‐Ir(ppy)3 undergo a redox quenching process reacted with BDMT to generate thiyl free radicals. Temperature‐responsive hydrogels are one of the most widely studied types of stimuli‐responsive hydrogel systems. Their ability to transition between their swollen and collapsed states makes them attractive for controlled drug delivery, microfluidic devices, and biosensor applications. Recent work has shown that poly(ethylene glycol) (PEG) methacrylate polymers are temperature‐responsive and exhibit a wide range of lower critical solution temperatures based on the length of ethylene glycol units in the macromer chain. The addition of iron oxide nanoparticles into the hydrogel matrix can provide the ability to remotely heat the gels upon exposure to an alternating magnetic field (AMF). In this work, diethylene glycol (n = 2) methyl ether methacrylate and PEG (n = 4.5) methyl ether methacrylate copolymers were polymerized into hydrogels with 5 mol % PEG 600 (n = 13.6) dimethacrylate as the crosslinker along with 5 wt % iron oxide nanoparticles. Volumetric swelling studies were completed from 22 to 80 °C and confirmed the temperature‐responsive nature of the hydrogel systems. The ability of the gels to collapse in response to rapid temperature changes when exposed to an AMF was demonstrated showing their potential use in biomedical applications such as controlled drug delivery and hyperthermia therapy. © 2010 Wiley Periodicals, Inc. In this article, novel smart hydrogels based on biodegradable pH sensitive poly(L ‐glutamic acid‐g‐2‐hydroxylethyl methacrylate) (PGH) chains and temperature‐sensitive hydroxypropylcellulose‐g‐acrylic acid (HPC‐g‐AA) segments were designed and synthesized. The influence of pH and temperature on the equilibrium swelling ratios of the hydrogels was discussed. The optical transmittance of the hydrogels was also changed as a function of temperature, which reflecting that the HPC‐g‐AA part of the hydrogels became hydrophobic at the temperature above the lower critical solution temperature (LCST). At the same time, the LCST of the hydrogels had a visible pH‐dependent behavior. Scanning electron microscopic analysis revealed the morphology of the hydrogels before and after enzymatic degradation. The biodegradation rate of the hydrogels was directly related to the PGH content and the pH value. The in vitro release of bovine serum albumin from the hydrogels were investigated. The release profiles indicated that both the HPC‐g‐AA and PGH contents played important roles in the drug release behaviors. New hyperbranched hydrophobic cross‐linkers with peripheral azide groups were synthesized as follows: First, star‐shaped polycaprolactones (sPCL) were synthesized by ring‐opening polymerization of caprolactone in the presence of pentaerythritol and tin (II) octoate. In the next step, sequential acrylation, Micheal addition, tosylation, and azidation by acryloyl chloride, diethanol amine, tosyl chloride, and sodium azide were respectively exploited to synthesize azide‐functionalized hyperbranched star‐shaped polycaprolactones which were named sPCL‐acrylate‐diethanolamine‐azide (sPCL‐AC‐DEA‐N3) and sPCL‐acrylate‐diethanolamine‐acrylate‐diethanolamine‐azide (sPCL‐AC‐DEA‐AC‐N3). All steps were thoroughly characterized by FT‐IR and 1H NMR spectroscopy. The GPC analysis showed that the molecular weight of sPCL increased after two azide functionalizations. Amphiphilic hydrogels based on sPCL‐AC‐DEA‐N3 (Mn = 8130 g/mol) and sPCL‐AC‐DEA‐AC‐N3 (Mn = 10112 g/mol) with linear alkyne‐terminated polyethylene glycols (PEG) (Mn = 2000, 4000, and 6000 g/mol) were synthesized through click coupling between azide and alkyne groups. In both hydrogels, the swelling ratio increased by increasing the molecular weight of PEG. The obtained results showed that the branching of the cross‐linker, significantly affected the swelling ratio of hydrogels. For instance, the swelling ratio of sPCL‐AC‐DEA‐AC‐N3 and PEG‐6000 (Q = 900) was higher than sPCL‐AC‐DEA‐N3 and PEG‐6000 (Q = 600). Despite the high cross‐linking density of sPCL‐AC‐DEA‐AC‐DEA‐N3–based hydrogels, the amount of released theophylline was higher than sPCL‐AC‐DEA‐N3–based hydrogels, due to the high content of PEG in these hydrogels.  相似文献

A new class of temperature and pH dual‐responsive and injectable supramolecular hydrogel was developed, which was formed from block copolymer poly(ethylene glycol)‐block‐poly[(2‐dimethylamino)ethyl methacrylate] (PEG‐b‐PDMAEMA) and α‐cyclodextrin (α‐CD) inclusion complexes (ICs). The PEG‐b‐PDMAEMA diblock copolymers with different ratio of ethylene glycol (EG) to (2‐dimethylamino)ethyl methacrylate (DMAEMA) (102:46 and 102:96, respectively) were prepared by atom transfer radical polymerization (ATRP). 1H NMR measurement indicated that the ratio of EG unit to α‐CD in the resulted ICs was higher than 2:1. Thermal analysis showed that thermal stability of ICs was improved. The rheology studies showed that the hydrogels were temperature and pH sensitive. Moreover, the hydrogels were thixotropic and reversible. The self‐assembly morphologies of the ICs in different pH and ionic strength environment were studied by transmission electron microscopy. Sodium dithionite in the presence of NaHCO3 in water acts as a single‐electron‐transfer agent and facilitates the single‐electron‐transfer/degenerative‐chain‐transfer mediated living radical polymerization (SET–DTLRP) of acrylates initiated with iodoform at room temperature. The resulting α,ω‐di(iodo)polyacrylates can be used as macroinitiators for the SET–DTLRP of other acrylates. Ultrahigh‐molar‐mass poly(tert‐butyl acrylate) can be synthesized via the SET–DTLRP of tert‐butyl acrylate and has a very low weight‐average molecular weight/number‐average molecular weight ratio of 1.15. © 2005 Wiley Periodicals, Inc. To synthesize the novel molecular‐ and pH‐stimulus‐responsive hydrogel, we prepared poly(ethylene glycol)‐based hydrogel containing ionic groups. We evaluated the fundamental swelling/shrinking properties of the hydrogels synthesized by various conditions. Decreasing the molecular weight of a crosslinker provided the increasing of the equilibrium swelling ratio. Also, the equilibrium swelling ratio was changed by the introduction of functional ionic monomers and its compositions. Furthermore, the swelling/shrinking behaviors of the hydrogels were affected by the environmental condition of aqueous solution, in fact the hydrogels were considerably shrunk (to one‐fifth volume) using a di‐ionic solute in the aqueous solution through the ionic interactions between the hydrogel and the solutes. Additionally, the specific shrinking to diamine compounds was also observed in response to pH change. These results clearly show the swelling/shrinking responsibility of the hydrogels toward the molecular recognitions and its pH conditions. © 2013 Wiley Periodicals, Inc. J. Polym. Fabrication and functionalization of hydrogels from well‐defined dendron‐polymer‐dendron conjugates is accomplished using sequential radical thiol‐ene “click” reactions. The dendron‐polymer conjugates were synthesized using an azide‐alkyne “click” reaction of alkene‐containing polyester dendrons bearing an alkyne group at their focal point with linear poly(ethylene glycol)‐bisazides. Thiol‐ene “click” reaction was used for crosslinking these alkene functionalized dendron‐polymer conjugates using a tetrathiol‐based crosslinker to provide clear and transparent hydrogels. Hydrogels with residual alkene groups at crosslinking sites were obtained by tuning the alkene‐thiol stoichiometry. The residual alkene groups allow efficient postfunctionalization of these hydrogel matrices with thiol‐containing molecules via a subsequent radical thiol‐ene reaction. The photochemical nature of radical thiol‐ene reaction was exploited to fabricate micropatterned hydrogels. Tunability of functionalization of these hydrogels, by varying dendron generation and polymer chain length was demonstrated by conjugation of a thiol‐containing fluorescent dye. © 2015 Wiley Periodicals, Inc. Poly(N‐vinyl‐pyrrolidone) (PVP) hydrogel has been considered as a very interesting and promising thermosensitive material. The most vital shortcoming of PVP hydrogel as thermosensitive material is that it does not exhibit thermosensitivity under usual conditions. In this work, semi‐interpenetrating polymer network (semi‐IPN) hydrogels based on PVP and carboxymethylcellulose (CMC) were prepared. The volume phase transition temperature (VPTT) of the hydrogels was determined by swelling behavior and differential scanning calorimetry (DSC). The results showed that the VPTT was significantly dependent on CMC content and the pH of the swelling medium. The amount of CMC in the semi‐IPN hydrogels was 0.050, 0.075, and 0.100 g, the VPTT in buffer solution of pH 1.2 was 29.9 °C, 27.5 °C and 24.5 °C, respectively. In addition, the VPTT occurred in buffer solution of pH 1.2, but did not appear in alkaline medium. Bovine serum albumin (BSA) as a model drug was loaded and the in vitro release studies were carried out in different buffer solutions and at different temperatures. The results of this study suggest that PVP/CMC semi‐IPN hydrogels could serve as potential candidates for protein drug delivery in the intestine. © 2010 Wiley Periodicals, Inc. A pH and reduction dual‐stimuli‐responsive PEGDA/PAMAM injectable network hydrogel containing “acetals” as pH‐sensitive groups and “disulfides” as reducible linkages was designed and synthesized via aza‐Michael addition reaction between PAMAM and PEGDA diacrylates. The pore size and swelling ratio of hydrogels was varied from 14 ± 3 to 19 ± 4 μm and 214 ± 13 to 300 ± 19 μm, respectively, with varying ethylene glycol repeating units in diacrylates. The swelling ratio of PEGDA/PAMAM network hydrogel increased with increase in the molecular weight of PEG and with decrease in pH. The presence of different cationizable amino‐functionalities in PEGDA/PAMAM network hydrogel helped to enhance the swelling ability of hydrogel under the acidic conditions. The continuous increase in metabolically active live HeLa cells with time in MTT assay implied biocompatibility/noncytotoxicity of the synthesized PEGDA/PAMAM injectable network hydrogel. Furthermore, the prepared PEGDA/PAMAM hydrogel showed higher degradation at lower pH and at higher concentration of DTT. The burst release of doxorubicin from PEGDA/PAMAM hydrogel under the environment of the lower pH and in presence of DTT compared to the release at normal physiological pH and in absence of DTT suggested the potential ability of this model hydrogel system for targeted and selective anticancer drug release at tumor tissues. © 2018 Wiley Periodicals, Inc. A novel one‐step approach is reported to prepare thermosensitive hydrogels simply by using hydroxypropyl‐β‐cyclodextrin (HP‐β‐CD)/glycidyl methacrylate (GMA)/N‐isopropylacrylamide (NIPAM) system. From GMA and HP‐β‐CD, HP‐β‐CD/GMA inclusion complex was prepared and identified with NMR, FTIR, and UV‐vis spectroscopies. GMA in the form of HP‐β‐CD/GMA complex was copolymerized with NIPAM in water with K2S2O8 as initiator, yielding hydrogels designated as poly(NIPAM‐CD‐GMA). The inclusion of CD in the hydrogels was confirmed by FTIR spectroscopy. The contents of CD and GMA placed considerable influence on the swelling ratio and temperature‐sensitivity of the produced hydrogels. The hydrogels bearing CD moieties showed higher swelling ratio and temperature‐sensitivity when compared with that without CD. The porous structure of the hydrogels containing CD was observed in the SEM images. Relevant mechanism of the ring‐opening reaction of epoxide groups in GMA, the subsequent crosslinking reactions and the formation of hydrogels containing CD moieties were proposed. © 2008 Wiley Periodicals, Inc. A novel series of temperature‐ and pH‐sensitive hydrogels based on poly(2‐ethyl‐2‐oxazoline) and three‐arm poly(D,L ‐lactide) were synthesized via photocopolymerization. For the creation of polymeric networks, two types of macromers terminated with methacrylate groups were prepared: poly(2‐ethyl‐2‐oxazoline) dimethacrylate and three‐arm poly(D,L ‐lactide) trimethacrylate. The chemical structures were analyzed with 1H NMR and Fourier transform infrared techniques. The thermal behaviors, morphologies, and swelling properties were measured for the characterization of the polymeric networks. All the poly(2‐ethyl‐2‐oxazoline)/three‐arm poly(D,L ‐lactide)hydrogels provided high water retention capacity and exhibited reversible swelling–shrinking behavior in response to temperature and pH variations. The hydrogels with higher poly(2‐ethyl‐2‐oxazoline) dimethacrylate contents were more effective in raising the swelling ratio and temperature and pH sensitivity. However, higher contents of three‐arm poly(D,L ‐lactide) trimethacrylate produced larger particles and pore sizes in the hydrogels. This study effectively proves that this unique combination of water swellability and biodegradability provides hydrogels with a much wider range of applications in biomedical fields. © 2002 Wiley Periodicals, Inc. Interpenetrating polymer network (IPN) hydrogels have been fabricated through a facile one‐pot approach from tetra/bifunctional telechelic macromonomers with epoxy, amine, azide, and alkyne groups by orthogonal double click reactions: epoxy‐amine reaction and copper‐catalyzed azide‐alkyne cycloaddition. Both the crosslinked networks are simultaneously constructed in water from the biocompatible poly (ethylene glycol)‐based macromonomers. The crosslinking density of each network was finely tuned by the macromonomer structure, permitting control of network molecular weights between crosslinks of the final gels. Compared to corresponding single network gels, the IPN gels containing both tightly and loosely crosslinked networks exhibited superior mechanical properties with shear moduli above 15 kPa and fracture stresses over 40 MPa. The synthetic versatility of this one‐pot approach will further establish design principles for the next generation of robust hydrogel materials. © 2016 Wiley Periodicals, Inc. J. Polym.
