7-(Diethylamino)-3-[4-[iodoacetyl)amino)phenyl]-4-methylcoumarin (DCIA) and 4-(bromomethyl)-7-methoxycoumarin have been evaluated as fluoropyrimidine-derivatizing agents to be detected using peroxyoxalate chemiluminescence with high-performance liquid chromatography. The derivatization procedure required only one step. No chemiluminescence was observed from the bromo derivatives, and the detection limits of fluoropyrimidine compounds derivatized with the iodo compound and detected with peroxyoxalate chemiluminescence were in the low femtomole range.  相似文献

An overview of liquid phase chemiluminescence (CL) processes is presented and the potential for CL detection in liquid chromatography (LC) is discussed, with particular reference to the luminol and peroxyoxalate reactions. Post column ion displacement from a solid phase reagent [a cation exchange resin in the copper(II) form] followed by catalysis of the luminol reaction is used for the quantification of mixtures of weak acids after separation by ion-exclusion chromatography. Polyaromatic hydrocarbons (PAHs) released during the combustion of biomass fuels are separated by reversed-phase chromatography and quantified by their sensitizing effect on the peroxyoxalate reaction. This procedure is also used for the determination of carboxylic acids in non-aqueous media after selective pre-column derivatization with a fluorescent label (9-anthracenemethanol).  相似文献

Electron-capture gas--liquid chromatographic and reversed-phase high-performance liquid chromatographic assays are described for the quantitation of the compound, 9-chloro-7-(2-chlorophenyl)-5H-pyrimido [5,4-d] [2]-benzazepine, [I], a member of the benzazepine class of compounds undergoing clinical evaluation as anxiolytic agents. Studies on the biotransformation of [I] in the rat and dog showed that the compound was metabolized mainly by hydroxylation to yield the 5-hydroxy compound, [II], 9-chloro-7-(2-chlorophenyl)-5H-pyrimido [5,4-d] [2]-benzazepin-5-ol (major metabolite), along with the formation of lesser amounts of the N-oxide, [III], 9-chloro-7-(2-chlorophenyl)-5H-pyrimido [5,4-d] [2]-benzazepine 3-oxide, and the phenolic analogue, [IV], 3-chloro-4-(9-chloro-5H pyrimido-[5,4-d] [2] benzazepin-7-yl)phenol. This report describes the quantitation of [I] and [II] (major metabolite) in plasma using the above analytical techniques, both in preclinical studies in the dog and in clinical pharmacokinetic studies in man.  相似文献

[reaction: see text] A single-step process for the preparation of 2-amino-7-chlorothiazolo[5,4-d]pyrimidines, 2, was achieved by the reaction of the commercially available 4,6-dichloro-5-aminopyrimidine 1 with isothiocyanates. This mild reaction accommodates a variety of functionalized isothiocyanates and proceeds in good to excellent yields. The determination of seven sulfonamides by means of HPLC with chemiluminescence detection is proposed for the first time. The analytes are derivatized with fluorescamine, separated and subsequently they participate in the post-column chemiluminescence (CL) peroxyoxalate system using imidazole as a catalyst. Among the different peroxyoxalates tested, bis[4-nitro-2-(3,6,9-trioxadecyloxycarbonyl)phenyl] oxalate provides higher sensitivities and stabilities, avoiding precipitation problems. A rigorous optimization of the significant variables by means of experimental designs has been developed in order to reconcile the chromatographic conditions with the CL reaction. The method provides detection limits in the low microgl(-1) range and has been satisfactorily applied to the analysis of spiked raw milk samples.  相似文献

Summary Despite the impressive limits of detection and inherent selectivity afforded by peroxyoxalate chemiluminescence (POCL) detection, efficient coupling of POCL to capillary electrophoresis (CE) remains limited by the relatively slow kinetics of the reactions that drive imidazole-catalyzed chemiluminescence. Moreover, oxalate esters, used in POCL, are sparingly soluble in polar solvents and hydrolyze rapidly, presenting an additional challenge with respect to detection following aqueous phase separations. In this paper, a novel method for coupling an ultrafast POCL reaction to CE is presented. Post separation electrokinetic delivery of the POCL reagent bis(2,4,6-trichlorophenyl)oxalate (TCPO) was accomplished using a commercially available micro tee. Electrokinetic addition of TCPO allowed for precise control of the ratio of TCPO to the chemiluminescence (CL) reagents 1,2,2,6,6-pentamethylpiperidine (PMP) and 1,2,4-triazole (triazole), spiked into the running buffer. This novel method for CL reagent delivery avoided the problems and costs associated with using pressure or mechanical pumps to deliver reagents post separation. Use of this dual-component system (PMP and triazole) resulted in intense CL with half-lives of less than 2 seconds. Optimum conditions for CE-POCL detection were investigated using stopped-flow kinetics. The detection limit for 3-aminofluoranthene, following separation by CE, was<0.95 nM.  相似文献

Indolo[6,5-d]benzo[b]thiophene, indolo[4,5-d]benzo[b]thiophene, indolo[5,6-d]benzo-[b]thiophene, and indolo[5,4-d]benzo[b]thiophene were obtained from the 2- and 3-dibenzothienylhydrazones by means of the Fischer reaction. Substituted 4-[(3-cyanopyridin-2-yl)thiomethyl]coumarins have been synthesized by the alkylation of 2-mercapto-4,6-dimethylnicotinonitrile with 4-chloromethylcoumarins. Substituted 4-(3-amino-thieno[2,3-b]pyridin-2-yl)coumarins have been obtained by a subsequent intramolecular condensation of the methylene and cyano groups. 6H-Chromeno[4,3-d]thieno[3,2-b:5,4-b']dipyridin-6-ones were isolated as a result of the interaction of these compounds with aldehydes, and their aminomethyl derivatives were synthesized.  相似文献

[1-(15)N]-Labeled 4,6-dimethyl-4H-[1,2,5]oxadiazolo[3,4-d]pyrimidine-5,7-dione 1-oxide (1-(15)N1) was easily prepared by nitration of commercially available 6-amino-1,3-dimethyl-1H-pyrimidine-2,4-dione using 15N-enriched nitric acid followed by an intramolecular oxidative cyclization with iodosylbenzene diacetate under mild conditions. On the basis of the experimental results using 1-(15)N1, the formation of 8-phenyltheophylline (3), the 1,3-dimethylalloxazines (4: n = 0, 1), and 1,3,7,9-tetramethyl-1H,9H-pyrimido[5,4-g]pteridine-2,4,6,8-tetraone++ + (5) in the thermal reaction of the N-oxide 1 with benzylamine, aniline, or piperidine, and the generation of NO or NO-related species in the reaction with N-acetylcysteamine were reasonably explained by considering the initial attack of the employed nucleophiles on the 3a-position of 1.  相似文献

Novel 1,6-methanocycloundeca[b]pyrimido[5,4-d]pyrrole-12,14-dione derivatives 6a,b and 7a-c were synthesized in moderate to good yields by the enamine alkylation and dehydrating condensation reactions of 11-chloro-3,8-methano[11]annulenone (8) with 6-amino-3-substituted uracil derivatives and subsequent elimination of HCl. The (1)H NMR spectra clarified that compounds 6a,b and 7a-c are aromatic molecules having a diatropic pi-system, which is suggested by the chemical shifts of the bridge methylene protons and peripheral protons. The electrochemical reduction exhibited more positive reduction potentials as compared with those of the vinylogous compounds of cyclohepta[b]pyrimido[5,4-d]pyrrole derivatives. In a search for the oxidizing function of 6a,b and 7a-c, compounds 6a and 7b were demonstrated to oxidize benzylamines, cyclohexylamine, and benzyl alcohol to give the corresponding carbonyl compounds in more than 100% yield under aerobic and photoirradiation conditions, while only benzylamine was oxidized under aerobic and thermal conditions at 100 degrees C. Thus, compounds 6a and 7b oxidize amines and alcohols in an autorecyling process, and the efficiency is higher under photoirradiation as compared with the thermal process, suggesting the oxidation reaction probably proceeds via electron transfer from amine and alcohol to the excited and ground states of compounds 6a and 7b.  相似文献

The reactivities of new heterocyclic systems, viz., indolo [5,6-d]- and indolo[5,4-d]benzo[b]furans, in several electrophilic-substitution reactions (the Mannich and Vilsmeier reactions and acylation) were studied. The behaviour of 19 fluorescent compounds of various types in peroxyoxalate chemiluminescence reactions was studied in terms of the relation of their excitation efficiency to their singlet excitation energy and oxidation potential. Compounds having low singlet excitation energy and low oxidation potential were excited effectively. As a result of the study, 3-aminoperylene was selected as a fluorophore for derivatization of simple car?ylic acids. The derivatives were separated by reversed-phase microbore h.p.l.c. and were detected by a peroxyoxalate chemiluminescence reaction detector. The detection limit was 0.1 fmol.  相似文献

The title compound N,N'-bistosyl-1H,4H-quinoxaline-2,3-dione (TsQD) provides peroxyoxalate chemiluminescence (PO-CL) when reacted with hydrogen peroxide in the presence of fluorophores. The chemiluminescence (CL) efficiency of TsQD was superior to that of other related compounds such as bis(2,4,6-trichlorophenyl) oxalate (TCPO), a typical oxalate for the peroxyoxalate PO-CL, under an aqueous condition. Factors affecting the PO-CL efficiency are discussed from the viewpoint of the structures of the substrates and the electronic nature of the fluorophores. A linear correlation of the logarithmic values evaluated from the CL quantum yields with the oxidation potentials of the aromatic fluorophores supports the involvement of the chemically initiated electron exchange luminescence (CIEEL) mechanism in both TsQD- and TCPO-CL systems. Also, an excellent Hammett relationship was derived from the correlation between the sigma(+) values and the relative singlet excitation yields in TsQD-CL enhanced by a series of fluorescent para,para'-disubstituted distyrylbenzenes.  相似文献

We developed an immune microanalysis system incorporating chemiluminescence detection, where the peroxyoxalate chemiluminescence (CL) detection using bis[4-nitro-2-(3,6,9-trioxadecyloxycarbonyl)phenyl]oxalate (TDPO)-hydrogen peroxide (H2O2)-fluorescein isothiocianate (FITC) reaction was newly adopted. The analysis system performed the following three processes on a microchip: immune reaction for high selectivity, electrophoresis for formation and transportation of the sample plug, and CL detection. The immune reaction was carried out using an antibody-immobilized glass bead. The glass bead was placed in one of the reservoirs in the microchip along with antigen (analyte) and a known amount of FITC-labeled antigen to set up a competitive immune reaction. The reactant after the immune reaction was fed electrophoretically into the intersection, resulting in a sample plug. The sample plug was then moved into another reservoir containing TDPO-H2O2 acetonitrile solution. At this point, CL detection was performed. The system described here was capable of determining human serum albumin or immunosuppressive acidic protein as a cancer marker in human serum.  相似文献

We report the use of solution-processed thin-film organic photodiodes for microscale chemiluminescence. The active layer of the photodiodes comprised a 1 : 1 blend by weight of the conjugated polymer poly(3-hexylthiophene) [P3HT] and [6,6]-phenyl-C(61)-butyric acid-methylester [PCBM]--a soluble derivative of C(60). The devices had an active area of 1 mm x 1 mm, and a broad-band response from 350 to 700 nm, with an external quantum efficiency of more than 50% between 450 and 550 nm. The photodiodes have a simple layered structure that permits facile integration with planar chip-based systems. To evaluate the suitability of the organic devices as integrated detectors for microscale chemiluminescence, a peroxyoxalate based chemiluminescence reaction (PO-CL) was monitored within a poly(dimethyl-siloxane) (PDMS) microfluidic device. Quantitation of hydrogen peroxide indicated excellent linearity and yielded a detection limit of 10 microM, comparable with previously reported results using micromachined silicon microfluidic chips with integrated silicon photodiodes. The combination of organic photodiodes with PDMS microfluidic chips offers a means of creating compact, sensitive and potentially low-cost microscale CL devices with wide-ranging applications in chemical and biological analysis and clinical diagnostics.  相似文献

The bis(carbodiimides) 4, obtained from bis-aza-Wittig reactions of bis(iminophosphorane) 3 with 2 equiv of aromatic isocyanates, were reacted with secondary amine to give symmetrically substituted 2,7-diaminothieno[2,3-d:5,4-d']dipyrimidine-4,5(3H,6H)-dione 6 in the presence of a catalytic amount of EtO(-)Na(+). Reactions of 4 with phenols or ROH in the presence of a catalytic amount of potassium carbonate or RO(-)Na(+) gave symmetrically substituted 2,7-diaryl(alkyl)oxythieno[2,3-d:5,4-d']dipyrimidine-4,5(3H,6H)-diones 6 in satisfactory yields. However, iminophosphoranes 9 were obtained via reaction of bis(iminophosphorane) 3 with 1 equiv of aromatic isocyanate and subsequent reaction with an amine in the presence of a catalytic amount of EtO(-)Na(+). Further reaction of iminophosphoranes 9 with aromatic isocyanates and various nucleophile generated unsymmetrically substituted thieno[2,3-d:5,4-d']dipyrimidine-4,5(3H,6H)-diones 12 in good yields.  相似文献

A high performance liquid chromatographic assay of methamphetamine (MP) and its related compounds, i.e. ephedrine (EP), norephedrine (NE), p-hydroxymethamphetamine (p-HMP), p-hydroxyamphetamine (p-HAP) and amphetamine (AP), with peroxyoxalate chemiluminescence detection has been developed. 4-(N,N-Dimethylaminosulphonyl)-7-fluoro-2,1,3-benzoxadiazole (DBD-F) was used as a fluorescent labeling reagent. A mixture of hydrogen peroxide and bis[4-nitro-2-(3,6,9-trioxadecyloxycarbonyl)phenyl] oxalate in acetonitrile was used as a postcolumn chemiluminogenic reagent. DBD derivatives of MP and its related compounds were separated by a gradient elution with acetonitrile and 0.01 M imidazole buffer (pH 7.0) within 65 min. The detection limits (S/N = 3) for the proposed method for MP, AP, EP, NE, p-HMP and p-HAP were 27, 100, 40, 133, 25 and 133 fmol on column, respectively. The recoveries of these compounds with normal urine samples were 87.4-106.4%. The method was successfully applied to the assay of MP and its metabolites in urine samples from MP addicts. A good linear correlation for the resulted amounts of MP or AP between the proposed method and gas chromatography was obtained (r = 0.993 for MP or 0.991 for AP).  相似文献

Synthesis and spectral study of two new cyano-substituted fluorescamine as the fluorescent probes for amino acid detection have been carried out comparing with the original fluorescamine. Of the three compounds, the derivative with a cyano group at the meta-position on the 4-phenyl group was found to be superior to the original one in the reactivity toward some amino acids as well as the fluorescence intensity of the adducts. The fluorescent amino acid adducts were also applied to the peroxyoxalate chemiluminescence system as the fluorophores, in which the derivative described above was found to be more effective also in chemiluminescence than the original one.  相似文献

By reaction of 4-substituted 5-amino-6-mercaptopyrimidines with 5-bromo-2,2-dimethyl-4,6-dioxo-1,3-dioxane, we have obtained 4′-substituted 5-(5-amino-6-pyrimidylthio)-2,2-dimethyl-4,6-dioxo-1,3-dioxanes. We have studied diazotization of these compounds by isoamyl nitrite. In the case of 4′-methoxy- and 4′-dimethylamino-substituted derivatives, we have obtained derivatives of novel heterocyclic systems: pyrimido[5,4-e][1,3,4]thiadiazine and pyrimido[5,4-e][1,3,4]thiadiazine-7-spiro-5′-1,3-dioxane, and in the case of the 4′-isopropylamino-substituted derivative we obtained 4-isopropyl-7-(2,2-dimethyl-4,6-dioxo-1,3-dioxan-5-ylidene)-1,2,3-triazolo[5,4-d]pyrimidin-7-ylidene.__________Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 613–623, April, 2005.  相似文献

The synthesis and the binding affinity for the putative adenosine receptor antagonist 6-methyl-7-[1,2,3]triazol-2-yl-1,6-dihydrobenzo[1,2-d;3,4-d']diimidazole (10) and 5-oxazol-2-yl-1H-pyrazolo[4,3-b]pyridin-3-ylamine (16) are reported. The title compounds were prepared from commercially available 1-chloro-2,4-dinitrobenzene (1) and 2-chloro-6-methoxy-3nitropyridine (11), respectively, but proved devoid of affinity for the adenosine A1 and A2A receptors.  相似文献
