A cascade cyclization/nucleophilic aromatic substitution (SNAr) reaction of α,β-unsaturated N-tosylhydrazones with N-heteroaryl chlorides was developed for the synthesis of N-heteroaryl pyrazole derivatives. This one-pot reaction provided bi(heteroaryl) derivatives in good to excellent yields and with excellent regioselectivity. The procedure is operationally simple and applicable to large-scale synthesis.  相似文献

Alkyne-based regioselective functionalization is an important transformation method for the synthesis of a wide variety of organic products. In this work, a regioselective TBAI-mediated oxidative 5-exo-dig iodo-oxycyclization of 2-alkynylbenzamide is used for the synthesis of various isobenzofuran derivatives with excellent functional group tolerance and high reaction efficiency. We hypothesized that using water in a mixed solvent could change the reaction pathway and realize a high reaction regioselectivity. Furthermore, the application of the developed procedure was demonstrated by the synthesis of phthalazin-1(2H)-one and aryl-substituted isobenzofurans.  相似文献

An efficient copper-catalyzed multicomponent reaction was developed for the synthesis of (arylselanyl)- or (arylsulfenyl)-alkyl-1,2,3-triazolo-1,3,6-triazonines. The products were obtained in moderate to excellent yields via the reaction of o-phenylenediamine, 2-azidobenzaldehyde and different arylchalcogenyl alkynes using catalytic copper iodide in 1,4-dioxane at 100?°C. The reactions tolerated a range of substituents on the arylchalcogenyl alkynes and proved to be an efficient methodology for the combinatorial synthesis of new selenium or sulfur-containing triazonine derivatives.  相似文献

A novel organocatalytic multicomponent cyanovinylation of aldehydes was designed for the synthesis of conjugated cyanomethyl vinyl ethers. The reaction was implemented for the synthesis of a 3-substituted 3-(cyanomethoxy)acrylates, using aldehydes as substrates, acetone cyanohydrin as the cyanide anion source, and methyl propiolate as the source of the vinyl component. The multicomponent reaction is catalyzed by N-methyl morpholine (2.5 mol%) to deliver the 3-(cyanomethoxy)acrylates in excellent yields and with preponderance of the E-isomer. The multicomponent reaction manifold is highly tolerant to the structure and composition of the aldehyde (aliphatic, aromatic, heteroaromatics), and it is instrumentally simple (one batch, open atmospheres), economic (2.5 mol% catalyst, stoichiometric reagents), environmentally friendly (no toxic waste), and sustainable (easy scalability).  相似文献

The synthesis of the Boc derivative of a novel member of the cyclopropane-modified proline library, Boc-protected 5-azaspiro[2.4]heptane-6-carboxylic acid, is reported. The synthesis was performed in six steps starting from (2S,4R)-4-hydroxyproline using a modified Simmons-Smith reaction as the key step. The reaction conditions for all the steps were carefully selected to avoid racemization at the chiral centers in the intermediates and the final product.  相似文献

The first asymmetric total synthesis of MK8383 through the iron-mediated coupling reaction is described. In this total synthesis, the key step was clearly the stereoselective construction of the (Z)-trisubstituted side-chain alkene. Although this problem was seemingly easy to resolve, in fact it presented us an opportunity to evaluate the coupling reactions reported to date, and the iron-mediated coupling reaction was found to be the only effective method for the stereoselective construction of the (Z)-trisubstituted side-chain alkene.  相似文献

An enantioselective total synthesis of (−)-strychnine was accomplished through the use of the highly practical catalytic asymmetric Michael reaction (0.1 mol% of (R)-ALB, greater than kilogram scale, without chromatography, 91% yield and >99% ee), and a domino cyclization that simultaneously constructed the B- and D- rings of strychnine (>77% yield). Newly-developed reaction conditions for thionium ion cyclization, NaBH3CN reduction of the imine moiety in the presence of a Lewis acid to prevent the ring-opening reaction, and chemoselective reduction of the thioether (desulfurization) in the presence of exocyclic olefin were pivotal to complete the synthesis. The described chemistry paves the way for the synthesis of more advanced Strychnos alkaloids.  相似文献

The biocatalytic processes are greener and safer alternatives for synthesis of drug and drug intermediates. This study reports one such approach of chemo-enzymatic synthesis of propranolol, a β-adrenergic receptor blocker. A green synthetic route was employed for synthesis of propranolol using enzymatic kinetic resolution. Racemic mixture of secondary alcohol [1-chloro-3-(naphthalen-1-yloxy)propan-2-ol] (RS)-5 was synthesized chemically in two step reaction and further its enzymatic kinetic resolution was carried out by using lipase to synthesize enantiomeric forms (R)-5 and S-(6) of propranolol. The kinetic resolution of (RS)-5 involves the transesterification of secondary racemic alcohol in which vinyl acetate is used as acyl donor. Initially, we screened commercially available lipases for kinetic resolution and of all the screened lipases Addzyme 001 showed the best results. The several reaction parameters such as organic solvent, acyl donor, temperature, reaction time and enzyme concentration were optimized to improve rate of reaction and to achieve maximum enantioselectivity. Addzyme 001 at 40 mg shows the maximum conversion rate of 49% using cyclohexane as the organic solvent, vinyl acetate as the acyl donor and it was found that the reaction yield was higher for (R)-5 along with the (S)-6 (eep = 98%, ees = 97%) at 40 °C in 48 h. Further, the treatment of (R)-5 with isopropyl amine resulted into formation of (S)-propranolol eep = 98%, and overall yield 29% independently. To synthesize (R)-propranolol, S-(6) acetate was produced enzymatically and was further deacylated by chemical hydrolysis for the production of (R)-propranolol. This study reports newer approach for synthesis of (R) and (S) propranolol using chemoenzymatic route.  相似文献

A microwave assisted green protocol for the synthesis of 1,4-disubstituted triazolyl N-carboxamides was explored using basic alumina as solid support. The method allows domino Ullmann-type reaction, Click reaction and formation of ester or amide linkages in a single reaction vessel using Cu(phen)(PPh3)Br and CMPA as catalyst and basic alumina as solid support in high yield. The protocol did not require addition of any external ligands or base. The method was also found to be equally good for the synthesis of bis triazole adducts.  相似文献

Enantioselective preference in the asymmetric synthesis where cyclohexene oxide is transformed enantioselectively to chiral (S)- or (R)-2-cyclohexen-1-ol by the reaction with the appropriate chiral lithium amide reagent has been evaluated theoretically using the MM3 force field. The plausible possible structures for each precursor (reaction intermediate complex) leading to a (S)- or (R)-2-cyclohexen-1-ol have been optimized with the extended MM3 force field applicable to the lithium amide functional group, and the populations of their (S)- or (R)-reaction intermediate complexes at an ambient temperature (298 K) were calculated. The initial structure for evaluating the reaction intermediates of this asymmetric synthesis was constructed on the basis of the optimized ab initio transition state structure (MP2/6-31+G) comprising lithium amide LiNH2 and propene oxide. To the thus obtained transition state structure composed of LiNH2 and propene oxide, the other remaining Cartesian coordinates for the actual reaction intermediates composed of the chiral lithium amides and cyclohexene oxide were added to make the reaction intermediate structure. The conformational search for the reaction intermediate has been carried out by using the Stochastic search Algorithm, and the optimized geometries and their conformational energies (steric energies) have been calculated by the MM3 force field. The populations calculated from the conformational energies of the reaction intermediate leading to the (S)- or (R)-2-cyclohexen-1-ol were shown to be linearly well correlated with the experimentally reported enantiomer excess (% ee) values. The critical factors to control the enantioselectivity were investigated on the basis of the optimized structures of the reaction intermediate complexes. The MM3 force field approach was shown to be applicable to the theoretical evaluation of the enantioselectivity and be useful for designing a new functional chiral lithium amide reagent for the asymmetric synthesis.  相似文献

This work aims to investigate more sustainable reaction conditions for the synthesis of metallophthalocyanines. Anisole, glycerol and their mixtures have been investigated as reaction media for the tetramerization of phthalonitriles. Acetates of three divalent first-transition metal cations, Co(II), Cu(II) and Zn(II), were used and several bases were tested, depending on the chosen substrates and reaction conditions, with a view to making the whole process more sustainable while ensuring its scalability. Unsubstituted phthalocyanines were synthesized to analyze the behavior of the different metal ions in terms of reactivity in the new reaction media, resulting in a general Cu > Co > Zn trend, while the nonpolar tetra-tert-butyl substitution was investigated to evaluate the synthesis of soluble derivatives in the new conditions. Furthermore, the potassium hydroxide (KOH)-aided statistical synthesis of the unsymmetrical 9(10), 16(17), 23(24)-tri-tert-butyl-2-iodophthalocyaninato zinc(II), starting from 4-tert-butylphthalonitrile and 4-iodophthalonitrile in a glycerol/anisole mixture, proceeded with a satisfactory 26% yield. Our results provide insights into the investigation of new reaction environments and the understanding of their strengths and weaknesses, with a view to further increasing the sustainability of the synthesis of metallomacrocycles with high added value while lowering their production cost.  相似文献

The reaction proceeded smoothly using molecular oxygen as an oxidant, providing an efficient method for the synthesis of N-hydroxyimide esters. A novel and green approach for efficient and rapid synthesis of polyhydroquinoline derivatives via unsymmetric Hantzsch reaction using organocatalysts at room temperature was reported. The process is a simple, environmentally friendly, rapid, and high yielding reaction for the synthesis of polyhydroquinoline derivatives. SmI2-induced reaction of (E)- and (Z)-β-alkoxyvinyl sulfones onto an aldehyde function afforded 2,6-syn-2,3-trans- and 2,6-syn-2,3-cis-tetrahydropyran-3-ols, respectively, via stereoselective cyclization. A simple and efficient procedure for the preparation of silica-bonded propylpiperazine-N-sulfamic acid (SBPPSA) by the reaction of 3-piperazine-N-propylsilica (3-PNPS) and chlorosulfonic acid in chloroform is described. Silica-bonded propylpiperazine-N-sulfamic acid is employed as a recyclable catalyst for the synthesis of highly substituted imidazoles from the reaction of benzil, aromatic aldehydes, ammonium acetate and amines under solvent-free conditions. The heterogeneous catalyst was recycled for five runs upon the reaction of benzil, 4-methylbenzaldehyde, benzylamine, and ammonium acetate without losing its catalytic activity.  相似文献

First synthesis of C2-symmetric chiral O,N,N,O-tetradentate 2,2-bipyridyldiolpropane ligands is described. The Mukaiyama–Michael reaction was applied as an important reaction for the synthesis of 2,2-bipyridylpropane 9. Among the ligands synthesized, ligand 11 exhibits excellent chiral induction (up to 97% ee) in diethylzinc addition to various aldehydes. The use of additional Lewis acid such as Ti(OiPr)4 in diethylzinc addition reaction is not required for the present catalytic system.  相似文献

Copper(II) or tin(II) trifluoromethanesulfonate in combination with TMSCl effectively activates a C-H bond for the imino-ene reaction of N-tosylarylaldimines with α-methylstyrene. A wide variety of N-tosylarylaldimines were used to give homoallylamines in good to excellent yields under mild conditions. The imino-ene adduct was converted into a β-amino ketone. The synthesis of a 2,4-substituted pyrrolidine and a piperidine was also achieved from the imino-ene product via a Mitsunobu reaction and a Grubbs cyclization, respectively.  相似文献

A new procedure was developed for the synthesis of a broad range of ansa-zirconocenes containing bis(2-methyl-4-arylindenyl)dimethylsilane ligands. The method is based on the palladium-catalyzed reaction of halogen-substituted bis(indenyl)dimethylsilanes with various organozinc compounds. The aryl-substituted bridging ligands thus prepared serve as the starting compounds for the synthesis of ansa-zirconocenes, which can be used as components of promising catalysts for propylene polymerization.  相似文献

3-Amino-substituted 5-aminopyrazoles were found to be suitable substrates for the synthesis of new 4-aminopyrazolo[1,5-a][1,3,5]triazines (5-aza-9-deaza-adenines) when used in the one-pot, three-component reaction with cyanamide and triethyl orthoformate under microwave irradiation. The reaction proceeded selectively and its scope was demonstrated by the preparation of a library of 4-aminopyrazolo[1,5-a][1,3,5]triazines. Some structural aspects of the prepared compounds were investigated using dynamic NMR spectroscopy and X-ray crystallography. The operational simplicity, short reaction time, and good reproducibility are attractive features of the developed robust and practical approach for the synthesis of 7-amino-substituted 4-aminopyrazolo[1,5-a][1,3,5]triazines.  相似文献
