We have investigated gas‐phase fragmentation reactions of protonated benzofuran neolignans (BNs) and dihydrobenzofuran neolignans (DBNs) by accurate‐mass electrospray ionization tandem and multiple‐stage (MSn) mass spectrometry combined with thermochemical data estimated by Computational Chemistry. Most of the protonated compounds fragment into product ions B ([M + H–MeOH]+), C ([ B –MeOH]+), D ([ C –CO]+), and E ([ D –CO]+) upon collision‐induced dissociation (CID). However, we identified a series of diagnostic ions and associated them with specific structural features. In the case of compounds displaying an acetoxy group at C‐4, product ion C produces diagnostic ions K ([ C –C2H2O]+), L ([ K –CO]+), and P ([ L –CO]+). Formation of product ions H ([ D –H2O]+) and M ([ H –CO]+) is associated with the hydroxyl group at C‐3 and C‐3′, whereas product ions N ([ D –MeOH]+) and O ([ N –MeOH]+) indicate a methoxyl group at the same positions. Finally, product ions F ([ A –C2H2O]+), Q ([ A –C3H6O2]+), I ([ A –C6H6O]+), and J ([ I –MeOH]+) for DBNs and product ion G ([ B –C2H2O]+) for BNs diagnose a saturated bond between C‐7′ and C‐8′. We used these structure‐fragmentation relationships in combination with deuterium exchange experiments, MSn data, and Computational Chemistry to elucidate the gas‐phase fragmentation pathways of these compounds. These results could help to elucidate DBN and BN metabolites in in vivo and in vitro studies on the basis of electrospray ionization ESI‐CID‐MS/MS data only.  相似文献

Mass spectrometry offers an arsenal of tools for diverse proteomic investigations. This perspective article reviews some of the recent developments in the field of coupling laser‐induced dissociation with mass spectrometry (LID‐MS). Strategies involving labelling with a chromophore to induce specific photo‐absorption properties are considered, with a focus on specific amino acid derivatization. Some of the opportunities and challenges of LID‐MS after targeted labelling for increasing specificity in complex sample analysis are discussed.  相似文献

Glycosylation is one of the most common posttranslational modifications (PTMs) of proteins, the characterization of which is commonly achieved through proteomic protocol, involving trypsin digestion followed by liquid chromatography/tandem mass spectrometry (LC/MS/MS). However, it is often not possible to characterize all glycopeptides in a complex sample because of the high complexity of glycoproteomic samples, and the relative lower abundances of glycopeptides in comparison to the unmodified peptides. We present here a targeted MS/MS analysis approach, which utilizes a previously developed computational tool, GlyPID, to guide multiple experiments, thus permitting a complete characterization of all N‐glycosylation sites of glycoproteins present in a complex sample. We have tested our approach using model glycoproteins analyzed by high‐resolution LTQ‐FT MS. The results demonstrate a potential use of our method for a high‐throughput characterization of complex mixtures of glycosylated proteins. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Representative compounds with a 1,3‐dihydroxybenzene substructure belonging to different important polyphenol classes (stilbenes, flavones, isoflavones, flavonols, flavanones, flavanols, phloroglucinols, anthraquinones and bisanthraquinones) were investigated based on detailed high‐resolution tandem mass spectrometry measurements with an Orbitrap system under negative ion electrospray conditions. The mass spectral behaviour of these compound classes was compared among each other not only with respect to previously described losses of CO, CH2CO and C3O2 but also concerning the loss of CO2 and successive specific fragmentations. Furthermore, some unusual fragmentations such as the loss of a methyl radical during mass spectral decomposition are discussed. The obtained results demonstrate both similarities and differences in their mass spectral fragmentation under MSn conditions, allowing a characterization of the corresponding compound type. Mass spectrometric fragmentation data of neo‐clerodane diterpenes are almost inexistent but they can prove helpful for the qualitative and quantitative analysis of these compounds as well as for the identification of unknown compounds belonging to this class of plant secondary metabolites. [M–H] ions of nine neo‐clerodane diterpenes (1–9), recently isolated from Teucrium chamaedrys, were generated by electrospray ionization and were fragmented in the collision cell of a Triple Quadrupole (TQ) and of a Quadrupole Ion Trap (QIT) mass spectrometer. The deprotonated neo‐clerodane glucosides, chamaedryoside A and B (1, 2), readily lost the sugar residue to give, as their main fragmentation channel, the neo‐clerodane ions, I and II, which were structurally characterized by TQ and QIT MS. The collision‐activated dissociation (CAD) mass spectra of I and II and of deprotonated neo‐clerodanes 3–9 allowed us to reach some general conclusions on the fragmentation pathways of this class of compounds. For example, teuflin and its OH derivatives, teucrin A, teuflidin and 6‐β‐hydroxyteucridin, showed a characteristic fragmentation pattern involving the loss of 94 Da and 124 Da from the lactone moiety, whereas a loss of 44 Da was observed for teucrin E, and of 58 Da for teucrin F and G. In addition, several compound‐specific fragmentations were observed and can be proposed for the identification of individual compounds. The systematic approach allowed us to hypothesize the mechanisms of the most important collision‐activated dissociation/isomerization channels. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

This work reports the use of two‐dimensional (2D) liquid chromatography system coupled with a tandem mass spectrometry for the quantification of ampicillin in bovine milk. A restrict access media column (RAM‐BSA C8, 50 × 2.1 mm, Luna, 10 μm, 100 Å) was used in the first dimension in order to exclude macromolecules, while an ACQUITY UPLC BEH C18 (50 × 2.1 mm, 1.7 μm) column was used in the second dimension. Three different channels of selected reaction monitoring (SRM) were used: 350 > 106 m/z, 350 > 160 m/z, and 350 > 192 m/z. The first transition was used for the quantification (higher intensity), and latter two for confirmation. The developed method is simple and requires a total analysis time of only 14 min/sample. The sample treatment involved only a centrifugation step for 20 min. The validated method has been successfully applied to monitor AMP residues in raw milk samples. To our knowledge, this is the first study to report the use of ultrahigh‐performance liquid chromatography (UHPLC) in 2D configuration.  相似文献

Psoralea Corylifolia L. is a traditional Chinese medicine with many beneficial effects in medical therapies. Bakuchiol was the main active ingredient of Psoralea Corylifolia L., used for the treatment of various diseases and also as a natural food additive. A specific and reliable ultra‐high performance liquid chromatography–tandem mass spectrometry has been developed and fully validated for the quantification of bakuchiol in rat plasma. Chromatographic separation of bakuchiol and an internal standard, daidzein, was achieved on a Hypersil Gold C18 column with gradient elution that consisted of methanol and water at a flow rate of 0.2 mL/min. The compounds were detected at negative ionization mode using mass transition m/z 255.2 → 172.0 and 252.9 → 132.0 for bakuchiol and daidzein, respectively. Good linearity was obtained over the range of 2–1000 ng/mL and the lower limit of quantification was 2 ng/mL. The intra‐ and inter‐day accuracies ranged from 91.1 to 105.7% and precisions (relative standard deviations) were within 9.3%. Bakuchiol was found to be stable under three freeze–thaw cycles, short‐term temperature, post‐preparative and long‐term temperature conditions. The method was applied to a pharmacokinetic study of bakuchiol intravenously administered to rats at a dose of 5 mg/kg. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

A sensitive analytical method has been developed and validated for the quantification of L‐ergothioneine in human plasma and erythrocytes by liquid chromatography‐tandem mass spectrometry. A commercially available isotope‐labeled L‐ergothioneine‐d9 is used as the internal standard. A simple protein precipitation with acetonitrile is utilized for bio‐sample preparation prior to analysis. Chromatographic separation of L‐ergothioneine is conducted using gradient elution on Alltime C18 (150 mm × 2.1 mm, 5 µ). The run time is 6 min at a constant flow rate of 0.45 ml/min. The mass spectrometer is operated under a positive electrospray ionization condition with multiple reaction monitoring mode. The mass transitions of L‐ergothioneine and L‐ergothioneine‐d9 are m/z 230 > 127 and m/z 239 > 127, respectively. Excellent linearity [coefficient of determination (r2) ≥ 0.9998] can be achieved for L‐ergothioneine quantification at the ranges of 10 to 10 000 ng/ml, with the intra‐day and inter‐day precisions at 0.9–3.9% and 1.3–5.7%, respectively, and the accuracies for all quality control samples between 94.5 and 101.0%. This validated analytical method is suitable for pharmacokinetic monitoring of L‐ergothioneine in human and erythrocytes. Based on the determination of bio‐samples from five healthy subjects, the mean concentrations of L‐ergothioneine in plasma and erythrocytes are 107.4 ± 20.5 ng/ml and 1285.0 ± 1363.0 ng/ml, respectively.
