Summary Berberine absorption by patients with ischemic heart failure (IHF) after oral administration and the relationship between clinical effect and plasma berberine concentration are studied. Plasma samples were pretreated by chloroform extraction. Berberine was determined on a μBondapak column with acetonitrile-phosphoric acid mobile phase and UV detection. The limit for berberine in plasma was 8 ng mL−1 for an injection volume of 50 μL. Average berberine recovery was 96.5%. Results showed that improvements in symptoms were more significant for patients with plasma berberine concentration>0.1 mg L−1 than for those with<0.1 mg L−1. Plasma berberine monitoring may be helpful in the treatment of patients with IHF.  相似文献

A new procedure for spectrofluorimetric determination of free and total glycerol in biodiesel samples is presented. It is based on the oxidation of glycerol by periodate, forming formaldehyde, which reacts with acetylacetone, producing the luminescent 3,5-diacetyl-1,4-dihydrolutidine. A flow system with solenoid micro-pumps is proposed for solution handling. Free glycerol was extracted off-line from biodiesel samples with water, and total glycerol was converted to free glycerol by saponification with sodium ethylate under sonication. For free glycerol, a linear response was observed from 5 to 70 mg L−1 with a detection limit of 0.5 mg L−1, which corresponds to 2 mg kg−1 in biodiesel. The coefficient of variation was 0.9% (20 mg L−1, n = 10). For total glycerol, samples were diluted on-line, and the linear response range was 25 to 300 mg L−1. The detection limit was 1.4 mg L−1 (2.8 mg kg−1 in biodiesel) with a coefficient of variation of 1.4% (200 mg L−1, n = 10). The sampling rate was ca. 35 samples h−1 and the procedure was applied to determination of free and total glycerol in biodiesel samples from soybean, cottonseed, and castor beans.  相似文献

A multi-pumping flow system for the spectrophotometric determination of nitrite and nitrate is described. The determination of nitrite is based on the Griess-Ilosvay reaction. Nitrate can be determined after its on-line reduction to nitrite using hydrazine sulphate in alkaline medium. Calibration was linear up to 3 mg NO2 L−1 with a limit of detection (3sb/S) of 0.013 mg NO2 L−1 an injection throughput of 55 injections h−1 and a repeatability (RSD) of 0.5% for the direct determination of nitrite. Two calibration graphs within the ranges 0.039–7 mg NO3 L−1 and 0.026–5 mg NO2 L−1 were run for the determination of nitrate and nitrite under reducing conditions, respectively. A limit of detection of 0.039 mg NO3 L−1 was obtained. An injection throughput of 27 injections h−1 and an RSD lower than 1.5% were achieved. The method was successfully applied to the determination of nitrite and nitrate in water samples. A simple, selective, highly sensitive and accurate procedure for the determination of trace amounts of copper has been developed based on solid-phase spectrophotometry. Copper reacts with 5-(2-benzothiazolylazo)-8-hydroxyquinoline (BTAHQ) to give a complex with high molar absorptivity (3.17 × 107 L mol−1 cm−1, 3.07 × 108 L mol−1 cm−1, 1.22 × 109 L mol−1 cm−1, and 1.80 × 109 L mol−1 cm−1), fixed on a Dowex 1-X8 type anion-exchange resin for 10 mL, 100 mL, 500 mL, and 1000 mL, respectively. The absorbance at 667 nm and 800 nm packed in a 1.0 mm cell was measured directly. Calibration is linear over the range 0.2–3.7 μg L−1 with RSD of < 1.28 % (n = 10). The detection and quantification limits of the 500 mL sample method are 79 ng L−1 and 260 ng L−1 when using 60 mg of Dowex 1-X8. For a 1000 mL sample, the detection and quantification limits are 67 ng L−1 and 220 ng L−1 using 60 mg of the exchanger. Increasing the sample volume can enhance the sensitivity. The proposed method was applied to the determination of copper in different environmental water samples (tap, pit, spring, and river), food products (rice, corn flour, and tea), and mushrooms, using the standard addition technique.  相似文献

An efficient ion chromatographic (IC) method was developed for the simultaneous quantitative determination of Li+, Na+, NH4 +, K+, Cs+, Ca2+, Mg2+, Sr2+, Ba2+ and Be2+ in energy drinks, pharmaceutical and drinking water samples by non-suppressed conductometric detection. The separation of ten cations including ammonium was achieved using a cation-exchange column and low conductivity mobile phase. The mobile phase consisted of tartaric acid, dipicolinic acid and boric acid. The separation of the cations was completed in less than 18 min, with a flow rate of 1.2 mL min−1. The separation was not affected by the existence of cations Co2+, Cr3+, Cd2+, Cu2+, Bi3+, Ag+, Fe3+ and Zn2+ in concentrations up to 20 mg L−1. Using an injection volume of 20 μL the obtained detection limits were 0.003 mg L−1, 0.02 mg L−1, 0.01 mg L−1, 0.01 mg L−1, 0.10 mg L−1, 0.01 mg L−1, 0.02 mg L−1, 0.02 mg L−1, 0.003 mg L−1 and 0.1 mg L−1, for Li+, Na+, NH4+, K+, Cs+, Ca2+, Mg2+, Sr2+, Be2+ and Ba2+ respectively. The intra-day repeatability (RSD%, n=5) ranged from 1.1% to 4.8%, and the inter-day (n=5) between 1.8% and 5.4% respectively. The method was applied to the analysis of various bottled and tap water, pharmaceutical preparations and energy drinks commercially available.   相似文献

A simple, sensitive, selective, and low-cost method is proposed for rapidly determining nitric oxide (NO) in some rat tissues. Polymer monolith microextraction (PMME) using a poly(methacrylic acid–ethylene glycol dimethacrylate) (MAA-EGDMA) monolithic column was combined with derivatization of NO using 1,3,5,7-tetramethyl-8-(3′,4′-diaminophenyl)-difluoroboradiaza-s-indacene (TMDABODIPY), and this was used to analyze the derivatives of NO by high-performance liquid chromatography (HPLC) with fluorescence detection at λ ex/λ em = 498/507 nm. The baseline separation of TMDABODIPY and its NO derivative is performed under simple conditions in which a C18 column is used and eluted with 50 mmol L−1 ethanolamine and methanol. The conditions for the extraction of NO derivatives were optimized. The limit of detection of NO was 2 × 10−12 mol L−1 (S/N = 3). The linearity range of the method was 9 × 10−11−4.5 × 10−8 mol L−1. The interday and intraday relative standard deviations were less than 5%. The proposed method was successfully applied to the determination of NO levels in some rat tissue samples including heart, kidney, and liver with recoveries varying from 87.1 to 95.2%.  相似文献

Summary 15 glycol ethers can be extracted from water by solidphase microextraction with a carboxen-polydimethyl-siloxane and separated by GC a Carbowax column. Water containing 15 glycol ethers at concentrations 0.1–10 mg.L−1 is saturated at ambient temperature with NaCl. A carboxen-polydimethylsiloxane-coated fiber is then exposed to the liquid for 20 min and then automatically injected into a capillary GC injection port. Calibration curves were linear for different glycol ethers in the rang 0.1–10 mg.L−1 Detection limits of each component of the mixture of glycol ethers between 50–500 μg.L−1. The SPME method with direct immersion in water results in better sensivity than methods based on liquid-liquid extraction.  相似文献

Summary A method for the determination of 5-hydroxy-N-methyl-2-pyrrolidone (5-HNMP) and 2-hydroxy-N-methylsuccinimide (2-HMSI) in plasma was developed. 5-HNMP and 2-HMSI are metabolites to the widely used organic solvent N-methyl-2pyrrolidone (NMP). The 5-HNMP and 2-HMSI were purified from plasma by C8 solid phase extraction, derivatised by bistrimethylsilyl trifluoroacetamid, and analysed by gas chromatography with mass spectrometric detection. For 5-HNMP, the precision was 2–7 % (120 and 780 ng mL−1) and the detection limit was 6 ng mL−1 (m/z 98). For 2-HMSI, the precision was 2–9 % (160 and 1000 ng mL−1) and the detection limit was 4 ng mL−1 (m/z 144). The method is applicable for analysis of plasma samples from workers exposed to NMP.  相似文献

Summary A reversed-phase liquid chromatographic method with sodium dodecylsulphate-n-propanolwater as mobile phase has been used to separate and determine six water-soluble vitamins in twelve minutes. The analytical characteristics linear range, sensitivity, detection limits, and precision were evaluated. The lowest detection limits were those of nicotinic acid (not usually present in pharmaceutical products), 0.7 mgL−1, nicotinamide, 1.3 mg L−1, and pyridoxine, 1.4 mg L−1. When the method was applied to the determination of the vitamins in pharmaceutical samples the values found agreed with those on the labels.  相似文献

A combined method involving electrochemical oxidation of iodide to iodate at a platinum electrode followed by extraction in CCl4 of ionic associates of iodine-iodide complexes with brilliant green, formed in excess of iodide, was developed for the spectrophotometric quantification of iodide. The slope of the calibration curve yields a molar extinction coefficient of ɛ = 3·105 L mol−1cm−1. This method can be used for the quantification of iodide in the concentration range of 3·10−7 − 3·10−6 mol L−1 with a detection limit of 5·10−8 mol L−1. The interfering effect of other ions on the determination of the iodide concentration was also investigated. The method was successfully applied for the determination of iodide in real samples of NaCl and spring water. Relative standard deviation is 1–2%.  相似文献

A rapid and convenient method for the determination of furfural is presented that is based upon sequential perturbation of the Mn(II)-catalyzed B-Z oscillating system with different amounts of furfural using a continuous-flow stirred tank reactor (CSTR). When the sample was injected, the change in the amplitude and/or period was linearly proportional to the logarithm of the concentration of furfural over the range 3×10−8∼1×10−5 mol L−1. This method gave a detection limit of 3×10−9 mol L−1 under optimum conditions. Finally, the possible mechanism of furfural perturbation in the oscillating reaction is discussed. When the furfural was injected into the Mn(II)-catalyzed B-Z oscillating system, the change in the amplitude and/or period was linearly proportional to the logarithm of the concentration of furfural over the range 3×10−8~1×10−5 mol L−1, with a detection limit of 3×10−9 mol L−1 under optimum conditions.   相似文献

A novel flow injection procedure has been developed for the determination of tannic acid based on the inhibition of the chemiluminescences in luminol-H2O2-Manganese tetrasulfonatophthalocyanine (MnTSPc) system by tannic acid. The method is simple, rapid and sensitive with a detection limit of 8 × 10−10 mol·L−1 and a linear range of 7 × 10−9–5 × 10−6 mol·L−1. The relative standard deviation is 1.9% for eleven measurements of 5 × 10−7 mol·L−1 tannic acid. The method has been successfully applied to the determination of tannic acid in real Chinese gall and hop pellets samples.  相似文献

In this work, the hyphenation of the multisyringe flow injection analysis technique with a 100-cm-long pathlength liquid core waveguide has been accomplished. The Cl/Hg(SCN)2/Fe3+ reaction system for the spectrophotometric determination of chloride (Cl) in waters was used as chemical model. As a result, this classic analytical methodology has been improved, minimizing dramatically the consumption of reagents, in particular, that of the highly biotoxic chemical Hg(SCN)2. The proposed method features a linear dynamic range composed of two steps between (1) 0.2–2 and (2) 2–8 mg Cl L−1, thus extended applicability due to on-line sample dilution (up to 400 mg Cl L−1). It also presents improved limits of detection and quantification of 0.06 and 0.20 mg Cl L−1, respectively. The coefficient of variation and the injection throughput were 1.3% (n = 10, 2 mg Cl L−1) and 21 h−1. Furthermore, a very low consumption of reagents per Cl determination of 0.2 μg Hg(II) and 28 μg Fe3+ has been achieved. The method was successfully applied to the determination of Cl in different types of water samples. Finally, the proposed system is critically compared from a green analytical chemistry point of view against other flow systems for the same purpose.  相似文献

The use of malonic acid as a complexing agent has enabled a new method to be devised to allow the determination of magnesium to be made using miniaturised isotachophoresis. Using a leading electrolyte of 10 mmol L−1 caesium hydroxide and 2 mmol L−1 malonic acid at pH 5.1 gave the method a high specificity towards magnesium. Investigations using a poly(methyl methacrylate) chip device with an integrated conductivity detector showed that no interference from calcium, strontium, barium and sodium should occur. The method was found to be linear over the range of magnesium concentrations from 0.625 to 75 mg L−1 and the limit of detection was calculated to be 0.45 mg L−1. Separations were demonstrated with water samples but the procedure should also be applicable to more complex sample matrices such as inorganic explosive residues, blood or urine.  相似文献

A method is described for determination of residues of the insecticide Etofenprox in environmental samples. Anionic surfactant micelle-mediated extraction (coacervation extraction) was evaluated for isolation of Etofenprox before HPLC. The optimum conditions used for extraction included: 0.09 g sodium dodecanesulfonate (SDoS), 3.1 mL (3.3, for concentrations below 0.04 mg L−1) 12 mol L−1 HCl, 5 min vortex stirring, 5 min centrifugation at 4000 rpm, 2 h equilibration time. The limits of quantification (LOQ) and detection (LOD) were 0.01 and 0.004 mg L−1, respectively, and recoveries obtained from five real samples ranged from 94.33±2.48 to 100.13±2.71%. The precision of the method was good; relative standard deviations (RSD) were less than 7%.   相似文献

Summary Elevated plasma homocysteine is, a known risk factor in arteriosclerotic vascular disease. To measure homocysteine in a large number of samples, we have developed a rapid, simple, robust and inexpensive reversed-phase HPLC method for routine analysis. Mercaptopro-pionylglycine was used as the internal standard and an external calibration in plasma was performed. Improvement was achieved by the use of gradient elution (using a sodium acetate buffer and methanol) resulting in a higher number of samples analyzed per day. Plasma samples were reduced with tributylphosphine and the proteins were precipitated with perchloric acid before addition of internal standard. The analytes were derivatized by use of 7-fluorobenzofurazone-4-sulfonic acid ammonium salt. For calibration human plasma was spiked with nine different concentrations of homocysteine (range 2–50 μmol L−1). The inter-assay precision of replicate (n=29) analysis of the concentration of homocysteine in a sample of pooled plasma was 3.0%. The limit of detection, defined as three times the signal-to-noise ratio, was 0.25 μmol L−1. The linearity of the assay was confirmed for a plasma concentration range of 2–2000 μmol L−1. The variation of duplicate analyses of 842 plasma samples was 2.6±1.7%.  相似文献

A new immunosensor for the determination of procalcitonin was developed. A sandwich assay format was implemented on a polymethylmetacrylate optical biochip, opportunely shaped in order to obtain several flow channels and potentially suitable for point of care testing applications. The sandwich format makes use of two new rat monoclonal antibodies. The capture antibody was covalently immobilised on the surface of the plastic chip, and the detection antibody was labelled with DY647 dye. Different combinations of capture and detection antibodies were investigated, and particular attention was devoted in order to avoid the non-specific adsorption. A limit of detection of 0.088 mg L−1 was achieved within the working range of 0.28–50 mg L−1 in buffer samples. The assay was also implemented in human serum, and 0.2 and 0.7–25 mg L−1 were the attained limit of detection and working range, respectively.  相似文献

A molecularly imprinted polymer was developed and used for solid-phase extraction (MISPE) of the antihelmintic fenbendazole in beef liver samples. Detection of the analyte was accomplished using square wave voltammetry (SWV) at a cylindrical carbon fibre microelectrode (CFME). A mixture of MeOH/HAc (9:1) was employed both as eluent in the MISPE system and as working medium for electrochemical detection of fenbendazole. The limit of detection was 1.9 × 10−7 mol L−1 (57 μg L−1), which was appropriate for the determination of fenbendazole at the maximum residue level permitted by the European Commission (500 μg kg−1 in liver). Given that the SW voltammetric analysis could not be directly performed in the sample extract as a consequence of interference from some sample components, a sample clean-up with a MIP for selectively retaining fenbendazole was performed. The MIP was synthesized using a 1:8:22 template/methacrylic acid/ethylene glycol dimethacrylate ratio. A Britton–Robinson Buffer of pH 9.0 was selected for retaining fenbendazole in the MIP cartridges, and an eluent volume of 5.0 mL at a flow rate of 2.0 mL min−1 was chosen in the elution step. Cross-reactivity with the MIP was observed for other benzimidazoles. The synthesized MIP exhibited a good selectivity for benzimidazoles with respect to other veterinary drugs. The applicability of the MISPE-SWV method was tested with beef liver samples, spiked with fenbendazole at 5,000 and 500 μg kg−1. Results obtained for ten different liver samples yielded mean recoveries of (95 ± 12)% and (96 ± 11)% for the upper and lower concentration level, respectively.  相似文献

A survey of contamination of surface and drinking waters around Lake Maggiore in Northern Italy with polar anthropogenic environmental pollutants has been conducted. The target analytes were polar herbicides, pharmaceuticals (including antibiotics), steroid estrogens, perfluorooctanesulfonate (PFOS), perfluoroalkyl carboxylates (including perfluorooctanoate PFOA), nonylphenol and its carboxylates and ethoxylates (NPEO surfactants), and triclosan, a bactericide used in personal-care products. Analysis of water samples was performed by solid-phase extraction (SPE) then liquid chromatography–triple-quadrupole (tandem) mass spectrometry (LC–MS–MS). By extraction of 1-L water samples and concentration of the extract to 100 μL, method detection limits (MDLs) as low as 0.05–0.1 ng L−1 were achieved for most compounds. Lake-water samples from seven different locations in the Southern part of Lake Maggiore and eleven samples from different tributary rivers and creeks were investigated. Rain water was also analyzed to investigate atmospheric input of the contaminants. Compounds regularly detected at very low concentrations in the lake water included: caffeine (max. concentration 124 ng L−1), the herbicides terbutylazine (7 ng L−1), atrazine (5 ng L−1), simazine (16 ng L−1), diuron (11 ng L−1), and atrazine-desethyl (11 ng L−1), the pharmaceuticals carbamazepine (9 ng L−1), sulfamethoxazole (10 ng L−1), gemfibrozil (1.7 ng L−1), and benzafibrate (1.2 ng L−1), the surfactant metabolite nonylphenol (15 ng L−1), its carboxylates (NPE1C 120 ng L−1, NPE2C 7 ng L−1, NPE3C 15 ng L−1) and ethoxylates (NPE n Os, n = 3-17; 300 ng L−1), perfluorinated surfactants (PFOS 9 ng L−1, PFOA 3 ng L−1), and estrone (0.4 ng L−1). Levels of these compounds in drinking water produced from Lake Maggiore were almost identical with those found in the lake itself, revealing the poor performance of sand filtration and chlorination applied by the local waterworks.  相似文献

To evaluate the pharmacokinetics of a novel analogue of ginkgolide B, 10-O-dimethylaminoethylginkgolide B (XQ-1) in rat plasma in pre-clinical studies, a sensitive and specific liquid chromatographic method with electrospray ionization mass spectrometry detection (LC–ESI–MS) was developed and validated. After a simple extraction with ethyl acetate, XQ-1 was analyzed on a Shim-pack C18 column with a mobile phase of a mixture of 1 μmol L−1 ammonium acetate containing 0.02% formic acid and methanol (55:45, v/v) at a flowrate of 0.3 mL min−1. Detection was performed in selected ion monitoring (SIM) mode using target ions at [M + H]+ m/z 496.05 for XQ-1 and m/z 432.10 for the internal standard (lafutidine). Linearity was established for the concentration range from 2 to 1,000 ng mL−1 . The extraction recoveries ranged from 86.0 to 89.9% in plasma at concentrations of 5, 50, and 500 ng mL−1. The lower limit of quantification was 2 ng mL−1 with 100 μL plasma. The validated method was successfully applied to a pharmacokinetic study after intragastic administration of XQ-1 mesylate in rats at a dose of 20 mg kg−1.  相似文献
