Leaf waxes protect terrestrial plants from biotic and abiotic stresses and are important sedimentary biomarkers for terrestrial plants. Thus, understanding the production and ablation of leaf waxes is critical in plant physiology and for geochemical studies. However, there have been no accurate approaches to quantify leaf wax production at different time scales. In this study, we demonstrate a novel approach to study leaf wax regeneration by irrigating plants with a pulse of deuterium-enriched water, followed by measurements of leaf wax D/H ratios by gas chromatography/isotope-ratio mass spectrometry (GC/IRMS). We demonstrate the efficacy of this approach using the grass species Phleum pratense in a greenhouse environment. Using a binary isotope mass balance model, we are able to quantify the regeneration rates of the C(16), C(18) acids and leaf waxes (C(23)-C(31) n-alkanes; C(22)-C(30) n-acids) over a diurnal cycle. Our results show that within one day 33-47% of C(16) and C(18) acids are regenerated, and thus the recycling time for these compounds is 2-3 days. For C(22)-C(26) n-alkyl lipids, 7-21% are regenerated within one day and thus they require 5-16 days to recycle. In comparison, the recycling time for long-chain n-alkyl lipids (C(27)-C(31)) is as long as 71-128 days. Our approach can be applied to different plants at shorter or longer time scales by adjusting the degree of isotopic labeling, sampling intervals and the amount of irrigation water.  相似文献

Gas chromatography/mass spectrometry (GC/MS), with an ion trap mass analyzer, was used to examine the very-long-chain cuticular acid and certain non-acid wax constituents on the leaf sheath surface of Sorghum bicolor before and during 36 hours of light exposure. The mass spectra of the trimethylsilylated acids and alcohols did not match any of those published in searchable mass spectral libraries. The observed differences can be related to the interaction between water and the trimethylsilylated acids and alcohols. Understanding the observed mass spectra of the very-long-chain plant waxes is critical for studies that employ GC/MS with the ion trap mass analyzer to elucidate cuticular wax compositions on plants.  相似文献

The use of total internal reflection (TIR)-Raman spectroscopy to examine cuticular wax layers in vivo at the surface of barley leaves (Hordeum vulgare L. cultivar Pastoral) is demonstrated. The limited penetration depth (40 nm) of the evanescent wave compared to the thickness of the wax layer eliminates problems arising from fluorescence from underlying pigments. Epicuticular wax crystals are observed to be more crystalline than the amorphous wax layer, which is analysed after removal of the wax crystals by cellulose acetate stripping. Carotenoids are detected as trace species in the cuticular waxes.  相似文献

Antifungal bioassay-guided fractionation of Combretum nelsonii leaf extracts afforded two closely related triterpenes, asiatic acid and arjunolic acid. Antifungal activities of the mixture of asiatic acid and arjunolic acid were determined against five fungal animal pathogens. The minimum inhibitory concentrations of the mixture to the different pathogens varied from 0.2 to 1.6 microg mL(-1); Candida albicans (0.9 microg mL(-1)), Cryptococcus neoformans (0.4 microg mL(-1)), Aspergillus fumigatus (1.6 microg mL(-1)), Microsporum canis (0.2 microg mL(-1)) and Sporothrix schenckii (0.2 microg mL(-1)). Microsporum canis and S. schenckii were the most susceptible followed by C. neoformans. Aspergillus fumigatus was the most resistant. The R(f) value of the mixture of asiatic acid and arjunolic acid was 0.27 in CEF (chloroform : ethylacetate : formic acid), 0.09 (BEA; benzene : ethanol : ammonium hydroxide) and 0.55 (EMW; ethylacetate : methanol : water) which was active against all pathogens. In vitro cytotoxicity of mixture gave an LC(50) of 10.58 microg mL(-1) towards Vero monkey kidney cells.  相似文献

Laser desorption/ionization time-of-flight (LDI-TOF) mass spectrometry was applied for the direct analysis of cuticular waxes on intact plant tissues. Cuticular wax compounds were ionized by laser desorption in the presence of colloidal silver. Silver-adduct ions were detected on samples from Arabidopsis thaliana and from maize. Good spot-to-spot reproducibility indicated homogeneous coverage of the sample by the fine colloidal material. The results were consistent with GC-MS analyses of cuticular extracts, thus confirming the feasibility of direct analysis based on this protocol. Molecular masses of the adduct ions correspond well with the known composition of cuticular waxes. Moreover, LDI-TOF gave good estimates of the relative local abundances of a given compound. However, bias was found in cases where compounds with different ionization efficiencies were analyzed.

Three different sensitive and accurate spectroscopic procedures were developed for the determination of three angiotensin-converting enzyme inhibitors, namely, ramipril, enalapril maleate and fosinopril. The first two spectrophotometric (extractive and non-extractive) procedures were based on ternary complex formation with molybdenum(V) thiocyanate. The formed complex can be determined by extraction with chloroform measured at lambdamax 517 nm Beer's law was obeyed in the concentration range from (10--90 microg ml(-1)) for ramipril and fosinopril and (4--36 microg ml(-1)) for enalapril maleate with molar absorptivity 1.2x10(4), 2x10(4) and 3.4x10(4) l mol(-1) cm(-1), respectively, or by direct measurement after addition of benzalkonium chloride as surfactant and measuring the formed ternary complex at lambdamax 545 nm with a linear relationship in the concentration range from (8-7-2 microg ml(-1)), (3--27 microg ml(-1)) and (8--72 microg ml(-1)) for ramipril, enalapril maleate and fosinopril with molar absorptivity 1.5x10(4), 5x10(4) and 2.1x10(4) l mol(-1) cm(-1), respectively. The third procedure is atomic absorption measurement through the quantitative determination of molybdenum content of the complex. These methods hold their accuracy and precision well when applied to the determination of ramipril, enalapril maleate and fosinopril in their dosage forms.  相似文献

The mode specificity of proton-transfer dynamics in the ground electronic state (X (1)A(1)) of tropolone has been explored at near-rotational resolution by implementing a fully coherent variant of stimulated emission pumping within the framework of two-color resonant four-wave mixing spectroscopy. Three low-lying (E(vib) approximately 550-750 cm(-1)) vibrational features, assigned to nu(30)(a(1)), nu(32)(b(2)), and nu(31)nu(38)(a(1)), have been interrogated under ambient, bulk-gas conditions, with term energies determined for the symmetric and antisymmetric (tunneling) components of each enabling the attendant tunneling-induced bifurcations of 1.070(9), 0.61(3), and 0.07(2) cm(-1) to be extracted. The dependence of tunneling rate (or hydron migration efficiency) on vibrational motion is discussed in terms of corresponding atomic displacements and permutation-inversion symmetries for the tropolone skeleton.  相似文献

An on-site procedure was set up for direct gravimetric measurement of the mass of aerosol collected using high volume impactors (aerodynamic size cut point of 10 microm, PM10); this knowledge has hitherto been unavailable. Using a computerized microbalance in a clean chemistry laboratory, under controlled temperature (+/-0.5 degrees C) and relative humidity (+/-1%), continuous, long time filter mass measurements (hours) were carried out before and after exposure, after a 48 h minimun equilibration at the laboratory conditions. The effect of the electrostatic charge was exhausted in 30-60 min, after which stable measurements were obtained. Measurements of filters exposed for 7-11 days (1.13 m3 min(-1)) in a coastal site near Terra Nova Bay (December 2000 - February 2001), gave results for aerosol mass in the order of 10-20 mg (SD approximately 2 mg), corresponding to atmospheric concentrations of 0.52-1.27 microg m(-3). Data show a seasonal behaviour in the PM10 content with an increase during December - early January, followed by a net decrease. The above results compare well with estimates obtained from proxy data for the Antarctic Peninsula (0.30 microg m(-3)), the Ronne Ice Shelf (1.49 microg m(-3)), and the South Pole (0.18 microg m(-3), summer 1974-1975, and 0.37 microg m(-3), average summer seasons 1975-1976 and 1977-1978), and from direct gravimetric measurements recently obtained from medium volume samplers at McMurdo station (downwind 3.39 microg m(-3), upwind 4.15 microg m(-3)) and at King George Island (2.5 microg m(-3), summer, particle diameter <20 microm). This finding opens the way to the direct measurement of the chemical composition of the Antarctic aerosol and, in turn, to a better knowledge of the snow/air relationships as required for the reconstruction of the chemical composition of past atmospheres from deep ice core data.  相似文献

Autoclaving was used to manipulate nutrient utilization and availability. The objectives of this study were to characterize any changes of the functional groups mainly associated with lipid structure in flaxseed (Linum usitatissimum, cv. Vimy), that occurred on a molecular level during the treatment process using infrared Fourier transform molecular spectroscopy. The parameters included lipid CH(3) asymmetric (ca. 2959 cm(-1)), CH(2) asymmetric (ca. 2928 cm(-1)), CH(3) symmetric (ca. 2871 cm(-1)) and CH(2) symmetric (ca. 2954 cm(-1)) functional groups, lipid carbonyl CO ester group (ca. 1745 cm(-1)), lipid unsaturation group (CH attached to CC) (ca. 3010 cm(-1)) as well as their ratios. Hierarchical cluster analysis (CLA) and principal components analysis (PCA) were conducted to identify molecular spectral differences. Flaxseed samples were kept raw for the control or autoclaved in batches at 120°C for 20, 40 or 60 min for treatments 1, 2 and 3, respectively. Molecular spectral analysis of lipid functional group ratios showed a significant decrease (P<0.05) in the CH(2) asymmetric to CH(3) asymmetric stretching band peak intensity ratios for the flaxseed. There were linear and quadratic effects (P<0.05) of the treatment time from 0, 20, 40 and 60 min on the ratios of the CH(2) asymmetric to CH(3) asymmetric stretching vibration intensity. Autoclaving had no significant effect (P>0.05) on lipid carbonyl CO ester group and lipid unsaturation group (CH attached to CC) (with average spectral peak area intensities of 138.3 and 68.8 IR intensity units, respectively). Multivariate molecular spectral analyses, CLA and PCA, were unable to make distinctions between the different treatment original spectra at the CH(3) and CH(2) asymmetric and symmetric region (ca. 2988-2790 cm(-1)). The results indicated that autoclaving had an impact to the mid-infrared molecular spectrum of flaxseed to identify heat-induced changes in lipid conformation. A future study is needed to quantify the relationship between lipid molecular structure changes and functionality/availability.  相似文献

The anharmonic vibrational frequencies of FHF(-) were computed by the vibrational self-consistent-field, configuration-interaction, and second-order perturbation methods with a multiresolution composite potential energy surface generated by the electronic coupled-cluster method with various basis sets. Anharmonic vibrational averaging was performed for the bond length and nuclear magnetic resonance indirect spin-spin coupling constants, where the latter computed by the equation-of-motion coupled-cluster method. The calculations placed the vibrational frequencies at 580 (nu(1)), 1292 (nu(2)), 1313 (nu(3)), 1837 (nu(1) + nu(3)), and 1864 cm(-1) (nu(1) + nu(2)), the zero-point H-F bond length (r(0)) at 1.1539 A, the zero-point one-bond spin-spin coupling constant [(1)J(0)(HF)] at 124 Hz, and the bond dissociation energy (D(0)) at 43.3 kcal/mol. They agreed excellently with the corresponding experimental values: nu(1) = 583 cm(-1), nu(2) = 1286 cm(-1), nu(3) = 1331 cm(-1), nu(1) + nu(3) = 1849 cm(-1), nu(1) + nu(2) = 1858 cm(-1), r(0) = 1.1522 A, (1)J(0)(HF) = 124+/-3 Hz, and D(0) = 44.4+/-1.6 kcal/mol. The vibrationally averaged bond lengths matched closely the experimental values of five excited vibrational states, furnishing a highly dependable basis for correct band assignments. An adiabatic separation of high- (nu(3)) and low-frequency (nu(1)) stretching modes was examined and found to explain semiquantitatively the appearance of a nu(1) progression on nu(3). Our calculations predicted a value of 186 Hz for experimentally inaccessible (2)J(0)(FF).  相似文献

A flow-through optical absorption detector for HPLC was constructed using a novel deep-UV light-emitting diode as radiation source with a peak emission wavelength of 255 nm. For measuring the transmitted intensity (a property correlated to Transmittance) a special UV-sensitive photodiode was employed. Besides the power source, no optical or electronic components other than an inexpensive operational amplifier and a few passive components were necessary. The performance of the detector was tested with three substances, namely nitrobenzene, benzoic acid and methyl benzoate, which were separated by gradient elution using an acetonitrile/water mixture and tetrabutylammonium hydrogensulfate as pH-buffer. Calibration curves for concentrations between 1.6 microg.mL(-1) and 400 microg.mL(-1) (nitrobenzene) and 8 microg.mL(-1) and 2.5 mg.mL(-1) (benzoic acid and methyl benzoate) were determined and coefficients of determination, r(2), of 0.9945, 0.9972 and 0.9996 were obtained for quadratic curve fits for the 3 compounds respectively. Relative standard deviations (n = 7) for peak areas were determined as 0.35% (nitrobenzene, 80 microg.mL(-1)), 0.27% (benzoic acid, 400 microg.mL(-1)) and 0.83% (methyl benzoate, 200 microg.mL(-1)). The lower limits of detection were found to be 750 ng.mL(-1), 5.8 microg.mL(-1) and 12 microg.mL(-1) for nitrobenzene, benzoic acid and methyl benzoate respectively.  相似文献

Electronic spectra of the C3N radical have been observed for the first time in the near ultraviolet wavelength region by laser induced fluorescence (LIF) spectroscopy. Seventeen vibronic bands of the B 2Pii-X 2Sigma+ electronic transition system of C3N were identified in LIF spectra of products in a discharge of HC3N. The origin of the B 2Pii state was determined to be 27,929.985(1) cm(-1) from rovibrational analyses. It was found that observations of two types of 2Sigma vibronic levels, which have 2Sigma+ and 2Sigma+/- symmetries originated from excitations of the nu4 trans-bending mode (omega4=369.1(20) cm(-1)) with a large Renner-Teller (RT) interaction (epsilon4=-0.1549(50)), and the nu5 cis-bending mode (omega5=163.24(84) cm(-1)) with a small Renner-Teller interaction (epsilon5=-0.0503(68)), respectively. Vibronic levels, with excitations of the C-C stretching (omega3=869.7 cm(-1)) mode, were also identified. The spin-orbit interaction constant was determined to be Aso=-36.7(50) cm(-1) from the RT analysis. In dispersed fluorescence spectra from B 2Pii, vibrational structures of the low-lying electronically excited A 2Pii state were clearly observed with a strong progression due to the nu3' mode, together with those of the X 2Sigma+ state with weak intensities. The origin of A 2Pii, T0=1844(3) cm(-1), and the vibrational frequencies, omega3'=883(3) cm(-1) and omega5'=121(3) cm(-1) for A 2Pii, and omega3"=1054(3) cm(-1), omega4"=405(3) cm(-1), and omega5"=131(3) cm(-1) for X 2Sigma+, were determined. Time profiles of fluorescence from B 2Pii have short (50-200 ns) and long (>1 micros) decay components with quantum beats, indicating that there is a competition between radiative decay and the nonradiative internal conversion to vibrationally highly excited A 2Pii and X 2Sigma+.  相似文献

Stability indicating assays for determination of Donepezil Hydrochloride in presence of its oxidative degradate were developed and validated. The first three are spectrophotometric methods depending on using zero order (D(0)), first order (D(1)) and second order (D(2)) spectra. The absorbance was measured at 315 nm for (D(0)) while the amplitude was measured at 332.1nm for (D(1)) and 340 nm for (D(2)) using deionized water as a solvent. Donepezil Hydrochloride (I) can be determined in the presence of up to 70% of its oxidative degradate (II) using (D(0)), 80% using (D(1)) and 90% using (D(2)). The linearity range was found to be 8-56 microg ml(-1) for (D(0)), (D(1)) and (D(2)). These methods were applied for the analysis of I in both powder and tablet form. Also, a spectrofluorimetric method depending on measuring the native fluorescence of I in deionized water using lambda excitation 226 nm and lambda emission 391 nm is suggested. The linearity range was found to be 0.32-3.20 microg ml(-1) using this method, I was determined in the presence of up to 90% of II. The proposed method was applied for the analysis of I in tablet form as well as in human plasma. The last method depends on using TLC separation of I from its oxidative degradate II and I was then determined spectrodensitometrically. The mobile phase was methanol : chloroform : 25% ammonia (16 : 64 : 0.1 by volume). The linearity range was found to be 2-15 microg/spot. This method was applied to the analysis of I in both powder and tablet form using acetonitrile as a solvent.  相似文献

13C nuclear magnetic resonance spectroscopy (NMR) was applied to determine the different components of apolar and polar fractions which were isolated by column chromatography from the crude chloroform-soluble waxes of olive fruits (Olea europaea) Dritta cultivar.13C NMR enabled the determination in the wax apolar fraction, of aliphatic aldehydes, and of benzyl, alkyl and glyceryl esters. In particular, the fatty acid composition of alkyl esters, comprising saturated and unsaturated oleic and linoleic acids, was determined. Acyl chain composition and the chain composition of 1,3- and 2-glycerol positions were also determined for triacylglycerols of olive fruit waxes.Oleanolic and maslinic acids were confirmed to be the major components of wax polar fraction. Complete assignments of 13C NMR chemical shifts of oleanolic and maslinic acids as a mixture were achieved by using homonuclear correlation spectroscopy with gradient (g-COSY), attached proton test (APT), inverse-detected heteronuclear single-quantum coherence with gradient (g-HSQC), high-resolution heteronuclear correlation spectroscopy (HETCOR) for C-H directly attached pairs and C-H long-range-coupled experiments.  相似文献

The color varieties of spodumene (green spodumene, kunzite) from Nilaw mine (Nuristan, Afghanistan) have been investigated by microthermometry and Raman spectroscopy analyses. These minerals are rich in primary and secondary fluid inclusions. Measured values of temperature homogenization (T(h)) and pressure (P) for selected fluid-inclusion assemblages (I-IV) FIA in green spodumene and (I-II) FIA in kunzite ranges from 370 to 430°C, 1.16 to 1.44 kbar and 300 to 334°C, 0.81 to 1.12 kbar, respectively. The brine content and concentration varies from 4.3 to 6.6 wt.% eq. NaCl. Numerous and diverse mineral phases (quartz, feldspars, mica, beryl, zirconium, apatite, calcite, gypsum) present in this mineral as solid inclusions were studied by Raman microspectroscopy. Raman spectra of selected fluid, organic and solid inclusions were collected as line or rectangular maps and also depth profiles to study their size and contents. There appeared very interesting calcite (156, 283, 711 and 1085 cm(-1)), beryl (324, 397, 686, 1068 and 3610 cm(-1)), topaz (231, 285, 707, 780 and 910 cm(-1)) and spodumene (355, 707 and 1073 cm(-1)) inclusions accompanied by fluid and/or organic inclusions (liquid and gas hydrocarbons) with bands at 2350 cm(-1) (CO(2), N(2)), 2550 cm(-1) (H(2)S) and 2900 cm(-1) (C(2)H(6)-CH(3)). Some solid inclusions contain carbonaceous matter (D-band at ca. 1320 cm(-1) and/or G-band at ca. 1600 cm(-1)).  相似文献

Supercritical carbon dioxide (scCO2) can be used as a “green” selective solvent for extractions of wheat straw waxes. For the first time, the crude extraction yields of wheat straw waxes and the yields of a single high value group of components (wax esters) using various scCO2 conditions (305.15 to 373.15 K and 7.5 to 40 MPa) have been modelled using the Chrastil equation (Chrastil, 1982). The model accurately predicts both the crude yield and percentage of valuable wax esters within the extracts enabling maximum extraction efficiency. The key compounds within the waxes (fatty acids, sterols, fatty alcohols, wax esters, β-diketones and alkanes) have been identified and quantified. This study highlighted that, 14,16 hentriacontanedione was extracted at critical temperature and pressure in concentrations of 1000 μg.g−1 straw. This molecule could demonstrate significant potential as a natural chelate for metal recovery and also in the formation of super-hydrophobic coatings.  相似文献

The methods of stimulated emission pumping-hole filling spectroscopy (SEP-HFS) and stimulated emission pumping population transfer spectroscopy (SEP-PTS) were applied to the conformation-specific study of conformational isomerization in tryptamine [TRA, 3-(2-aminoethyl)indole]. These experimental methods employ stimulated emission pumping to selectively excite a fraction of the population of a single conformation of TRA to well-defined ground-state vibrational levels. This produces single conformations with well-defined internal energy, tunable over a range of energies from near the zero-point level to well above the lowest barriers to conformational isomerization. When the SEP step overcomes a barrier to isomerization, a fraction of the excited population isomerizes to form that product. By carrying out SEP excitation early in a supersonic expansion, these product molecules are subsequently cooled to their zero-point vibrational levels, where they can be detected downstream with a third tunable laser that probes the ground-state population of a particular product conformer via a unique ultraviolet transition using laser-induced fluorescence. The population transfer spectra (recorded by tuning the SEP dump laser while holding the pump and probe lasers fixed) exhibit sharp onsets that directly determine the energy thresholds for conformational isomerization in a given reactant-product conformer pair. In the absence of tunneling effects, the first observed transition in a given X-Y PTS constitutes an upper bound to the energy barrier to conformational isomerization, while the last transition not observed constitutes a lower bound. The bounds for isomerizing conformer A of tryptamine to B(688-748 cm(-1)), C(1)(860-1000 cm(-1)), C(2)(1219-1316 cm(-1)), D(1219-1282 cm(-1)), E(1219-1316 cm(-1)), and F(688-748 cm(-1)) are determined. In addition, thresholds for isomerizing from B to A(<1562 cm(-1)), B to F(562-688 cm(-1)), and out of C(2) to B(<747 cm(-1)) are also determined. The A-->B and B-->A transitions are used to place bounds on the relative energies of minima B relative to A, with B lying at least 126 cm(-1) above A. The corresponding barriers have been computed using both density functional and second-order many-body perturbation theory methods in order to establish the level of theory needed to reproduce experimental results. While most of the computed barriers match experiment well, the barriers for the A-F and B-F transitions are too high by almost a factor of 2. Possible reasons for this discrepancy are discussed.  相似文献

The first arsenic(V)-phthalocyanines, [As(tbpc)X(2)](+), where tbpc denotes tetra(tert-butyl)phthalocyaninate, C(48)H(48)N(8)(2-) and X = F, Cl, and Br) have been prepared through an appropriate oxidative addition process to a highly soluble arsenic(III) derivative, [As(tbpc)](+). Among them, [As(tbpc)F(2)](+) has been isolated as PF(6)(-) salt. Unlike conventional metal derivatives of phthalocyanines, they show a significantly red-shifted (by >1000 cm(-1)) Q-band and facile reduction of the macrocyclic ligand (redox potentials for [As(tbpc)F(2)](+) have been determined by cyclic voltammetry; 1.13 V vs ferricinium(+)/ferrocene (tbpc(-/2-)), -0.45 V (tbpc(2-/3-)), and -0.90 V (tbpc(3-/4-)), of which the values are anodically shifted by about 1 V) as compared to those of conventional phthalocyanines. Although the anomaly in their spectral and electrochemical properties is similar to that of the known antimony analogues, the arsenic-phthalocyanines have been found less stable.  相似文献

Time-resolved Fourier transform infrared emission spectroscopy was applied to the study of a pulsed discharge in a He/CH(4) mixture. The dynamics of the formation and decay of acetylene ν(3) (3289 cm(-1)), methane ν(3) (3019 cm(-1)) and ν(1) (2917 cm(-1)), the CH radical electronic ground state X(2)Π(r) (2309-2953 cm(-1)), C(2) Bernath electronic transition B(1)Δ(g)-A(1)Π(u) (3337-3606 cm(-1)), molecular hydrogen emission transitions 5g-4f and 2p-2s, atomic hydrogen, and atomic helium were monitored in the 1800-4000 cm(-1) region. The time profile of the rotational and vibrational temperature of the CH radical was obtained for a 30 μs time interval during and after the discharge pulse. A kinetic model was used for the study of the chemical dynamics of the formation and decay of the individual fragments. The results from the model were compared to the experimental emission spectra.  相似文献
