Short and highly efficient stereoselective syntheses provide machaeriols and cannabinoids in a divergent approach starting from a common precursor, commercially available (S)‐perillic acid. Key features of the novel strategy are a stereospecific palladium‐catalyzed decarboxylative arylation and a one‐pot sequence comprising a stereoselective hydroboration followed by oxidation or reduction of the corresponding intermediary boranes. The divergent approach is convincingly demonstrated by the five‐step syntheses of (+)‐machaeriol B, (+)‐machaeriol D, and related analogues, and the four‐step synthesis of (+)‐Δ8‐THC and an analogue.  相似文献

A qualitative and quantitative analytical method was developed for the simultaneous determination of Δ9‐tetrahydrocannabinol (THC), 11‐hydroxy‐Δ9‐tetrahydrocannabinol (11‐OH‐THC) and l1‐nor‐9‐carboxy‐Δ9‐tetrahydrocannabinol (THC‐COOH) in whole blood. The samples were prepared by solid‐phase extraction followed by ultra‐performance liquid chromatography/tandem mass spectrometry (UPLC/MS/MS) analysis using positive ion electrospray ionization and multiple reaction monitoring. The chromatographic separation was performed with an Acquity UPLC® HSS T3 (50 × 2.1 mm i.d., 1.8 µm) reversed‐phase column using a methanol/2 mM ammonium formate (formic acid 0.1%) gradient in a total run time of 9.5 min. MS/MS detection was achieved with two precursor‐product ion transitions per substance. The method was fully validated, including selectivity and capacity of identification, according to the identification criteria (two transitions per substance, signal‐to‐noise ratio, relative retention time and ion ratio) without the presence of interferences, limit of detection (0.2 µg/L for THC and 0.5 µg/L for 11‐OH‐THC and THC‐COOH), limit of quantitation (0.5 µg/L for all cannabinoids), recovery (53–115%), carryover, matrix effect (34‐43%), linearity (0.5‐100 µg/L), intra‐assay precision (CV < 10% for the relative peak area ratios and <0.1% for the relative retention time), inter‐assay accuracy (mean relative error <10%) and precision (CV <11%). In Cannabis sativa, Δ9‐Tetrahydrocannabinolic acid‐A (Δ9‐THCA‐A) is the non‐psychoactive precursor of Δ9‐tetrahydrocannabinol (Δ9‐THC). In fresh plant material, about 90% of the total Δ9‐THC is available as Δ9‐THCA‐A. When heated (smoked or baked), Δ9‐THCA‐A is only partially converted to Δ9‐THC and therefore, Δ9‐THCA‐A can be detected in serum and urine of cannabis consumers. The aim of the presented study was to identify the metabolites of Δ9‐THCA‐A and to examine particularly whether oral intake of Δ9‐THCA‐A leads to in vivo formation of Δ9‐THC in a rat model. After oral application of pure Δ9‐THCA‐A to rats (15 mg/kg body mass), urine samples were collected and metabolites were isolated and identified by liquid chromatography‐mass spectrometry (LC‐MS), liquid chromatography‐tandem mass spectrometry (LC‐MS/MS) and high resolution LC‐MS using time of flight‐mass spectrometry (TOF‐MS) for accurate mass measurement. For detection of Δ9‐THC and its metabolites, urine extracts were analyzed by gas chromatography‐mass spectrometry (GC‐MS). The identified metabolites show that Δ9‐THCA‐A undergoes a hydroxylation in position 11 to 11‐hydroxy‐Δ9‐tetrahydrocannabinolic acid‐A (11‐OH‐Δ9‐THCA‐A), which is further oxidized via the intermediate aldehyde 11‐oxo‐Δ9‐THCA‐A to 11‐nor‐9‐carboxy‐Δ9‐tetrahydrocannabinolic acid‐A (Δ9‐THCA‐A‐COOH). Glucuronides of the parent compound and both main metabolites were identified in the rat urine as well. Furthermore, Δ9‐THCA‐A undergoes hydroxylation in position 8 to 8‐alpha‐ and 8‐beta‐hydroxy‐Δ9‐tetrahydrocannabinolic acid‐A, respectively, (8α‐Hydroxy‐Δ9‐THCA‐A and 8β‐Hydroxy‐Δ9‐THCA‐A, respectively) followed by dehydration. Both monohydroxylated metabolites were further oxidized to their bishydroxylated forms. Several glucuronidation conjugates of these metabolites were identified. The crystal and molecular structures of four stereoisomers of tapentadol hydrochloride [systematic name: 3‐(3‐hydroxyphenyl)‐N,N,2‐trimethylpentan‐1‐aminium chloride], C14H24NO+·Cl, a novel analgesic agent, have been determined by X‐ray crystal structure analysis. Resolution of the isomers was carried out by reverse‐phase and chiral high‐performance liquid chromatographic (HPLC) methods. Stereoisomers (I) and (II) crystallize in the monoclinic space group P21, each with two tapentadol cations and two chloride anions in the asymmetric unit, while stereoisomers (III) and (IV) crystallize in the orthorhombic space group P212121, with one tapentadol cation and one chloride anion in the asymmetric unit. The absolute configurations of the four enantiomers were determined unambiguously by X‐ray crystallography. The crystal structures reveal the stereochemistries at the 3‐ethyl and 2‐methyl groups to be R,R, S,S, S,R and R,S in stereoisomers (I)–(IV), respectively. The ethyl and aminopropyl groups adopt different orientations with respect to the phenol ring for (I) and (IV). In all four structures, the chloride ions take part in N—H...Cl and O—H...Cl hydrogen bonds with the tapentadol molecules, resulting in one‐dimensional helical chains in the crystal packing in each case.  相似文献

The title compound, C10H18NO3S2, which finds application as a spin label, has triclinic (P) symmetry at 100 (2) K with two independent molecules in the asymmetric unit. Both molecules are very similar with respect to bond lengths and angles, but molecule 2 shows disordering of its side chain. The pyrroline rings differ slightly with respect to the position of the NO group, which in both cases are sterically shielded by the surrounding methyl groups. The crystal structure of the title compound represents the first example of a 2,2,5,5‐tetramethyl‐1‐oxyl‐Δ3‐pyrroline derivative with a side chain at the double bond which is linked to it through an sp3‐hybridized C atom. In the solid state, the side chain adopts a conformation with the methyl group above/below the pyrroline ring and a H atom directed towards a C atom of the double bond. The disordered side chain of molecule 2 represents a second conformation with low potential energy. Both molecules exhibit planar chirality, but in the solid state both pairs of stereoisomers are present. These four stereoisomers are stacked one behind the other in four different columns, denoted A, A′, B and B′, the angle between the vectors of the N—O bonds in columns A and B being 80.38 (8)°.  相似文献

The total synthesis and stereochemical structural elucidation of JBIR‐39, containing four nonproteinogenic piperazic acid (Piz) residues, is reported. The synthesis includes Sc(OTf)3‐catalyzed acylation of a Piz(γ‐OTBS) derivative with piperazic acid chloride, providing the desired Piz‐Piz(γ‐OTBS) dipeptide in high yield without epimerization. After assembling two additional Piz moieties and (S)‐isoleucic acid at the N‐terminus, amidation with the (R)‐α‐methylserine ester at the C‐terminus, and deprotection afforded the desired (2R,8S)‐hexapeptide, which is the assumed structure of JBIR‐39. Although the spectral data of the (2R,8S)‐hexapeptide was not identical to JBIR‐39, further synthesis of three stereoisomers confirmed the stereochemical structure of JBIR‐39 to be (2S,6S,8S,11R,16S,21R,26S,27S).  相似文献

The solid and solution structures of a new optically active aminopyridine compound, 2‐[(1S)‐(+)‐10‐camphorsulfonamino]‐6‐aminopyridine [(S)‐csaap], 1 , are reported. Crystal data: space group P21, a = 8.9729 (5), b = 10.9447 (6), c = 36.693 (2) Å, β = 96.435 (1)°, V = 3580.8 (3) Å3, Z = 8, R1 = 0.0673 and wR2 = 0.1600 with I > 2σ(I). This chiral compound shows an unprecedented cocrystallization of four stereoisomers, which are characterized by X‐ray crystallography and NMR spectroscopy.  相似文献

A novel aqueous in situ derivatization procedure with propyl chloroformate (PCF) for the simultaneous, quantitative analysis of Δ9‐tetrahydrocannabinol (THC), 11‐hydroxy‐Δ9‐tetrahydrocannabinol (OH‐THC) and 11‐nor‐Δ9‐tetrahydrocannabinol‐carboxylic acid (THC‐COOH) in human blood and urine is proposed. Unlike current methods based on the silylating agent [N,Obis(trimethylsilyl)trifluoroacetamide] added in an anhydrous environment, this new proposed method allows the addition of the derivatizing agent (propyl chloroformate, PCF) directly to the deproteinized blood and recovery of the derivatives by liquid–liquid extraction. This novel method can be also used for hydrolyzed urine samples. It is faster than the traditional method involving a derivatization with trimethyloxonium tetrafluoroborate. The analytes are separated, detected and quantified by gas chromatography–mass spectrometry in selected ion monitoring mode (SIM). The method was validated in terms of selectivity, capacity of identification, limits of detection (LOD) and quantification (LOQ), carryover, linearity, intra‐assay precision, inter‐assay precision and accuracy. The LOD and LOQ in hydrolyzed urine were 0.5 and 1.3 ng/mL for THC and 1.2 and 2.6 ng/mL for THC‐COOH, respectively. In blood, the LOD and LOQ were 0.2 and 0.5 ng/mL for THC, 0.2 and 0.6 ng/mL for OH‐THC, and 0.9 and 2.4 ng/mL for THC‐COOH, respectively. This method was applied to 35 urine samples and 50 blood samples resulting to be equivalent to the previously used ones with the advantage of a simpler method and faster sample processing time. We believe that this method will be a more convenient option for the routine analysis of cannabinoids in toxicological and forensic laboratories.  相似文献

Sodium bis[2‐(3′,6′,9′‐trioxadecyl)‐1,2‐dicarba‐closo‐dodecaborane‐1‐carboxylato]triphenylstannate, [(CH3OCH2CH2OCH2CH2OCH2CH2)‐1,2‐C2B10H10‐9‐COO)2SnPh3]? Na+, compound 1, was synthesized by the 1:1 condensation of triphenyltin(IV) hydroxide with 2‐(3′,6′,9′‐trioxadecyl)‐1,2‐dicarba‐closo‐dodecaborane‐1‐carboxylic acid and crystallized in the presence of sodium bicarbonate. Its structure was determined by spectroscopy, elemental analysis and X‐ray diffraction. The structure of 1 consists of trigonal bipyramidal [Sn(Ph)3(L)2]? anions and Na+ cations coordinated by oxygen atoms of polyoxaalkyl chains of different stannate anions, forming cation–anion chains elongated along the c axis. Compound 1 is significantly more active in vitro against seven tumour cell lines of human origin than 5‐fluorouracil, cis‐platin, carboplatin, and previously reported organotin carboranecarboxylates, but is less active than organotin polyoxaalkylcarboxylates. A simple, isocratic high performance liquid chromatography system (HPLC) with electrochemical detection (EC) was used to study Cannabis constituents. Several Cannabis extracts and reference standards were examined. A total of eleven constituents were separated, and three major cannabinoids; Δ8, Δ9 tetrahydrocannabinol (THC) and cannabidiol (CBD) were identified. A linear relationship was established for the quantitation of the halucinogenic constituent Δ9 THC. Minor contaminants in Δ9 THC reference standard, which were not detected by gas chromatography (GC) were detected for the first time. The detection limits of Δ9 THC and related cannabinoids were in the low nanogram range (2–20 ng).  相似文献

In the title compound, [Cu(C14H18O4)(H2O)]n, each CuII atom bonds to four O atoms of four adamantanediacetate (ada) ligands in equatorial positions and an O atom from a water mol­ecule in the apical position. Two adjacent CuII atoms form a paddle‐wheel unit with four ada ligands. The distance between the two Cu atoms is 2.5977 (3) Å. A crystallographic inversion center is located at the center of the Cu–Cu core. Each Cu2(ada)4 paddle‐wheel further bonds to four adjacent identical paddle‐wheel units, generating a two‐dimensional layered structure of Cu(ada)(H2O) with a 44 topology.  相似文献

The treatment of a β3‐amino acid methyl ester with 2.2 equiv. of lithium diisopropylamide (LDA), followed by reaction with 5 equiv. of N‐fluorobenzenesulfonimide (NFSI) at ?78° for 2.5 h and then 2 h at 0°, gives syn‐fluorination with high diastereoisomeric excess (de). The de and yield in these reactions are somewhat influenced by both the size of the amino acid side chain and the nature of the amine protecting group. In particular, fluorination of N‐Boc‐protected β3‐homophenylalanine, β3‐homoleucine, β3‐homovaline, and β3‐homoalanine methyl esters, 5 and 9 – 11 , respectively, all proceeded with high de (>86% of the syn‐isomer). However, fluorination of N‐Boc‐protected β3‐homophenylglycine methyl ester ( 16 ) occurred with a significantly reduced de. The use of a Cbz or Bz amine‐protecting group (see 3 and 15 ) did not improve the de of fluorination. However, an N‐Ac protecting group (see 17 ) gave a reduced de of 26%. Thus, a large N‐protecting group should be employed in order to maximize selectivity for the syn‐isomer in these fluorination reactions.  相似文献

The four stereoisomers of the novel title compounds were prepared by oxidative cyclization of their enantiomerically pure diarylheptanoid precursors by means of the straightforward biomimetic approach presented in the preceding article. The isocentrolobines are the methoxy regioisomers of the natural (+)‐ and (−)‐centrolobines and were characterized for the first time. The synthetic procedure established the absolute configurations and the unambiguous correlation with the chiroptical data. The spectroscopic and the chiroptical data of the isocentrolobines are highly similar to those of the natural products. The single diagnostic parameter that would allow a immediate assignment in the presence of only one of the isomers is the higher melting point (ca. 50°) of the cis‐configured isocentrolobines.  相似文献

3(2‐pyridinylmethylene)‐5‐aryl‐2(3H)‐furanones and 3(3‐pyridinylmethylene)‐5‐aryl‐2(3H)‐furanones were prepared as a mixture of (E) and (Z) stereoisomers by condensing pyridine‐2‐carboxaldehyde and pyridine‐3‐carboxaldehyde with 3‐aroylpropionic acids. The reaction of the furanones 6 and 7 with anhydrous aluminium chloride in benzene led to the formation of 4,4‐diaryl‐1‐(2‐pyridinyl)but‐1,3‐diene ( 8 ) and 4,4‐diaryl‐1‐(3‐pyridinyl)but‐1,3‐diene ( 9 ) as mixtures of geometrical (E,E‐ and E,Z‐) stereoisomers via an intermolecular alkylation mode. When the reaction was carried out in tetrachloroethane as a solvent, the reaction of 6 gave 5‐arylquinoline‐7‐carboxylic acid via intramolecular alkylation mode. The one‐ and two‐dimensional polymorphic cadmium polycarboxylate coordination polymers, catena‐poly[bis[μ2‐2‐(2‐methyl‐1H‐benzimidazol‐1‐yl)acetato‐κ3N3:O,O′]cadmium(II)], [Cd(C10H9N2O2)2]n, and poly[bis[μ2‐2‐(2‐methyl‐1H‐benzimidazol‐1‐yl)acetato‐κ3N3:O,O′]cadmium(II)], also [Cd(C10H9N2O2)2]n, were prepared under solvothermal conditions. In each structure, each CdII atom is coordinated by four O atoms and two N atoms from four different ligands. In the former structure, two crystallographically independent CdII atoms are located on twofold symmetry axes and doubly bridged in a μ2N:O,O′‐mode by the ligands into correspondingly independent chains that run in the [100] and [010] directions. Chains containing crystallographically related CdII atoms are linked into sheets viaπ–π stacking interactions. Sheets containing one of the distinct types of CdII atom are stacked perpendicular to [001] and alternate with sheets containing the other type of CdII atom. The second complex is a two‐dimensional homometallic CdII (4,4) net structure in which each CdII atom is singly bridged to four neighbouring CdII atoms by four ligands also acting in a μ2N:O,O′‐mode. A square‐grid network results and the three‐dimensional supramolecular framework is completed by π–π stacking interactions between the aromatic ring systems.  相似文献

Conformationally constrained bicyclic derivatives of the potent and selective κ-opioid receptor agonist 2-(3,4-dichlorophenyl)-N-methyl-N-[(1S)-1-Phenyl-2-(1-Pyrrolidinyl)-ethyl]acetamide(3,ICI-199,441)were designed to explore the effect of the conformational restriction and stereochemistry of the pharmacophoric ethylenediamine incorporated into the pyrrolidine on the affinity and κ-selectivity.A facile enantiosipecitfic synthetic route was extablished to afford all four stereoisomers starting from readily available amino acids through mild cyclization and amide formation.  相似文献

The crystal structure of the title compound, trans‐[PtCl2(C16H23P)2], has been determined at 100 K. The Pt atom is located on a twofold axis and adopts a distorted square‐planar coordination geometry. The structure is only the second example of a coordination complex containing a derivative of the 4,8‐dimethyl‐2‐phosphabicyclo[3.3.1]nonane (Lim) phosphine ligand family. The ligand contains four chiral C atoms, with the stereochemistry at three of these fixed during synthesis, therefore resulting in two possible ligand stereoisomers. The compound crystallizes in the chiral space group P43212 but is racemic, comprising an equimolar mixture of both stereoisomers disordered on a single ligand site. The effective cone angles for both isomers are the same at 146°.  相似文献

Intramolecular S‐alkylation in a series of ω‐haloalkyl‐substituted thiophosphorylacetonitriles 5–7 presents an effective synthetic route to the hitherto unknown 3‐cyano‐2‐oxo‐1,2‐thiaphospholanes 14 and thiaphosphinanes 15 . The compounds were obtained as a mixture of cis‐ and trans‐isomers that were resolved to individual stereoisomers in most cases. For some of them, X‐ray diffraction analysis has been performed. It was shown that 31P NMR spectroscopy can be used to assign the stereochemistry of 3‐cyano‐2‐oxo‐1,2‐thiaphosphacyclanes. © 2002 John Wiley & Sons, Inc. The synthesis of 2‐ethynyl‐9‐substituted carbazole and 3‐ethynyl‐9‐substituted carbazole monomers containing first‐generation chiral and achiral dendritic (i.e., minidendritic) substituents, 2‐ethynyl‐9‐[3,4,5‐tris(dodecan‐1‐yloxy)benzyl]carbazole (2ECz), 3‐ethynyl‐9‐[3,4,5‐tris(dodecan‐1‐yloxy)benzyl]carbazole (3ECz), 2‐ethynyl‐9‐{3,4,5‐tris[(S)‐2‐methylbutan‐1‐yloxy]benzyl}carbazole (2ECz*), and 3‐ethynyl‐9‐{3,4,5‐tris[(S)‐2‐methylbutan‐1‐yloxy]benzyl}carbazole (3ECz*), is presented. All monomers were polymerized and copolymerized by stereospecific polymerization to produce cis‐transoidal soluble stereoisomers. A structural analysis of poly(2ECz), poly(2ECz*), poly(3ECz), poly(3ECz*), poly(2ECz*‐co‐2ECz), and poly(3ECz*‐co‐3ECz) by a combination of techniques, including 1H NMR, ultraviolet–visible, and circular dichroism spectroscopy, thermal optical polarized microscopy, and X‐ray diffraction experiments, demonstrated that these polymers had a helical conformation that produced cylindrical macromolecules exhibiting chiral and achiral nematic phases.
