The gadolinium(iii) complex of S-SSSS-NO(2)BnDOTMA exhibits water exchange kinetics that are optimal for use in high relaxivity or targeted contrast agents. However, the synthesis of this ligand is hampered by the steric encumbrance imparted upon the cyclen ring by the nitrobenzyl substituent. A relatively simple modification has been used to enable the synthesis of larger quantities of a bifunctional ligand that retains similar fast water exchange properties. The gadolinium complex of S-SSS-NO(2)BnDO3MA-1A is shown to retain the rapid water exchange kinetics characteristic of a twisted square antiprismatic (TSAP) coordination geometry (tau(M)= 6 +/- 0.4 ns).  相似文献

We conducted relaxometric and water exchange studies of the cationic [Gd((S,S,S,S)-THP)(H2O)]3+ complex (THP 1,4,7,10-tetrakis(2-hydroxy-propyl)-1,4,7,10-tetraazacyclododecane). While the NMRD profiles obtained are typical for DOTA-like complexes (DOTA = 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetate), variable-temperature 7O NMR investigations revealed a relatively high water exchange rate (k(298)(ex) = 1.89 x 10(7) s(-1)). These results differ from those reported for other cationic tetraamide macrocyclic Gd(III) complexes, which exhibit characteristically low exchange rates. Since the low exchange rates are attributed partially to the geometry of the M isomer (square antiprismatic) in the tetraamide derivatives, the atypical water exchange rate observed in [Gd((S,S,S,S)-THP-(H2O)]3+ may result from a twisted square antiprismatic structure in this complex and from the relatively high steric strain at the water coordination site as a result of the presence of methyl groups at the alpha-position with respect to the Gd(III)-bound O atoms of THP.  相似文献

We report the synthesis and characterization of the novel ligand H(5)EPTPA-C(16) ((hydroxymethylhexadecanoyl ester)ethylenepropylenetriaminepentaacetic acid). This ligand was designed to chelate the Gd(III) ion in a kinetically and thermodynamically stable way while ensuring an increased water exchange rate (kappa(ex)) on the Gd(III) complex owing to steric compression around the water-binding site. The attachment of a palmitic ester unit to the pendant hydroxymethyl group on the ethylenediamine bridge yields an amphiphilic conjugate that forms micelles with a long tumbling time (tau(R)) in aqueous solution. The critical micelle concentration (cmc = 0.34 mM) of the amphiphilic [Gd(eptpa-C(16))(H(2)O)](2-) chelate was determined by variable-concentration proton relaxivity measurements. A global analysis of the data obtained in variable-temperature and multiple-field (17)O NMR and (1)H NMRD measurements allowed for the determination of parameters governing relaxivity for [Gd(eptpa-C(16))(H(2)O)](2-); this is the first time that paramagnetic micelles with optimized water exchange have been investigated. The water exchange rate was found to be kappa(298)(ex) = 1.7 x 10(8) s(-1), very similar to that previously reported for the nitrobenzyl derivative [Gd(eptpa-bz-NO(2))(H(2)O)](2-) kappa(298)(ex) = 1.5 x 10(8) s(-1)). The rotational dynamics of the micelles were analysed by using the Lipari-Szabo approach. The micelles formed in aqueous solution show considerable flexibility, with a local rotational correlation time of tau(298)(l0) = 330 ps for the Gd(III) segments, which is much shorter than the global rotational correlation time of the supramolecular aggregates, tau(298)(g0) = 2100 ps. This internal flexibility of the micelles is responsible for the limited increase of the proton relaxivity observed on micelle formation (r(1) = 22.59 mM(-1) s(-1) for the micelles versus 9.11 mM(-1) s(-1) for the monomer chelate (20 MHz; 25 degrees C)).  相似文献

In the objective of optimizing water exchange rate on stable, nine-coordinate, monohydrated Gd(III) poly(amino carboxylate) complexes, we have prepared monopropionate derivatives of DOTA4- (DO3A-Nprop4-) and DTPA5- (DTTA-Nprop5-). A novel ligand, EPTPA-BAA(3-), the bisamylamide derivative of ethylenepropylenetriamine-pentaacetate (EPTPA5-) was also synthesized. A variable temperature 17O NMR study has been performed on their Gd(III) complexes, which, for [Gd(DTTA-Nprop)(H2O)]2- and [Gd(EPTPA-BAA)(H2O)] has been combined with multiple field EPR and NMRD measurements. The water exchange rates, k(ex)(298), are 8.0 x 10(7) s(-1), 6.1 x 10(7) s(-1) and 5.7 x 10(7) s(-1) for [Gd(DTTA-Nprop)(H2O)]2-, [Gd(DO3A-Nprop)(H2O)]- and [Gd(EPTPA-BAA)(H2O)], respectively, all in the narrow optimal range to attain maximum proton relaxivities, provided the other parameters (electronic relaxation and rotation) are also optimized. The substitution of an acetate with a propionate arm in DTPA5- or DOTA4- induces increased steric compression around the water binding site and thus leads to an accelerated water exchange on the Gd(III) complex. The k(ex) values on the propionate complexes are, however, lower than those obtained for [Gd(EPTPA)(H2O)]2- and [Gd(TRITA)(H2O)]- which contain one additional CH(2) unit in the amine backbone as compared to the parent [Gd(DTPA)(H2O)]2- and [Gd(DOTA)(H2O)]-. In addition to their optimal water exchange rate, [Gd(DTTA-Nprop)(H2O)]2- has, and [Gd(DO3A-Nprop)(H2O)]- is expected to have sufficient thermodynamic stability. These properties together make them prime candidates for the development of high relaxivity, macromolecular MRI contrast agents.  相似文献

Detailed analyses of the solution structure and exchange dynamics of two sets of homologous mono-amide triacetate lanthanide complexes (Ln = Eu, Gd) of cyclen have been undertaken. The complex [LnL1], bearing an N-linked CH2CH2NHCO-pyridyl moiety, forms mono-aqua (q = 1) species in solution and the Gd complex undergoes rapid water exchange (k(ex)= 11 x 10(7) s(-1), 298 K) as a result of the steric destabilisation of the Ln-water binding interaction. The homologous complex with a C-3 spacing chain, [LnL2], forms a q = 0 species.  相似文献

The effects of surface water pretreatment on membrane fouling and the influence of these different fouling types on the rejection of 21 neutral, positively and negatively charged pharmaceuticals were investigated for two nanofiltration membranes. Untreated surface water was compared with surface water, pretreated with a fluidized anionic ion exchange and surface water, pretreated with ultrafiltration. Fouling the nanofiltration membranes with anionic ion exchange resin effluent, resulted in the deposition of a mainly colloidal fouling layer, with a rough morphology. Fouling the nanofiltration membranes with ultrafiltration permeate, resulted in the deposition of a smooth fouling layer, containing mainly natural organic matter. The fouling layer on the nanofiltration membranes, caused by the filtration of untreated surface water, was a combination of both colloids and natural organic matter.Rejection of pharmaceuticals varied the most for the membranes, fouled with the anionic ion exchange effluent, and variations in rejection were caused by a combination of cake-enhanced concentration polarisation and electrostatic (charge) effects. For the membranes, fouled with the other two water types, variations in rejection were smaller and were caused by a combination of steric and electrostatic effects.Changes in membrane surface hydrophobicity due to fouling, changed the extent of partitioning and thus the rejection of hydrophobic, as well as hydrophilic pharmaceuticals.  相似文献

The mechanism by which naphthenic acids stabilize water/oil interfaces has received extensive attention because of its industrial relevancy. In this work, we employed a molecular dynamics simulation to study its molecular origin. Two models were adopted, wherein naphthenic acid coverage of water/n-heptane interfaces, both spherical and flat, was hypothesized, respectively. It was found that the coalescence of two water clusters is entirely attributed to the diffusional motion of the components involved which requires the initial departure of the naphthenic acid molecules from the interface so that a water bridge can form. The naphthenic acids not only act as a steric barrier but also reduce the mobility of the water and n-heptane molecules making the formation of the water bridge rather difficult. In fact, our results show that the coalescence of two water clusters fully covered by naphthenic acid molecules is a low-probability event even at evaluated temperatures. In addition, the results from the flat interface models suggest that the emulsion stability is weakly dependent on the molecular weight of the naphthenic acids utilized. Order parameter calculations reveal liquid-crystal-like ordering of naphthenic acids at the water/n-heptane interfaces. All these observations are consistent with the corresponding experimental observations. The present work also suggests that the mobility of naphthenic acids is considerably enhanced with more n-heptane molecules present outside a water droplet. However, in such a case, coalescence could not occur as the water clusters are far apart from each other.  相似文献

The NH tautomerism of five Mg-free chlorophyll a and b derivatives 2-6 was studied utilizing NMR spectroscopy and molecular modeling. The results from the dynamic NMR measurements of the chlorins revealed that substituent effects contribute crucially to the free energy of activation (DeltaG(double dagger)) in the NH tautomeric processes. An intermediate tautomer for the total tautomeric NH exchange in a chlorin was observed for the first time, when the (1)H NMR spectra of chlorin e(6) TME (3) and rhodin g(7) TME (4) (TME = trimethyl ester) were measured at lower temperatures. The lower energy barriers (DeltaG(1)(double dagger)) obtained for the formation of the intermediate tautomers of 3 and 4, assigned to the N(22)-H, N(24)-H trans-tautomer, were 10.8 and 10.6 kcal/mol, respectively. The energy barrier (DeltaG(2)(double dagger) value) for the total tautomeric NH exchange in the five chlorins was found to vary from 13.6 kcal/mol to values higher than 18 kcal/mol. The lowest DeltaG(2)(double dagger) value (13.6 kcal/mol) was obtained for rhodochlorin XV dimethyl ester (2), which was the only chlorophyll derivative lacking the C(15) substituent. In the case of chlorins 4 and 5, the steric crowding around the methoxycarbonylmethyl group at C(15) raised the DeltaG(2)(double dagger) activation free-energy to 17.1 kcal/mol. However, the highest energy barrier with DeltaG(2)(double dagger) > 18 kcal/mol was observed for the NH exchange of pyropheophorbide a methyl ester (6), possessing the macrocycle rigidifying isocyclic ring E. Our results demonstrate that the steric strain, arising either from the steric crowding around the bulky substituent at C(15) or the macrocycle rigidifying isocyclic ring E, slows down the NH tautomeric process. We suggest that deformations in the chlorin skeleton are closely connected to the NH tautomeric exchange and that the exchange occurs by a stepwise proton-transfer mechanism via a hydrogen bridge.  相似文献

The stability and water exchange dynamics of gadolinium (GdIII) complexes are critical characteristics that determine their effectiveness as contrast agents for magnetic resonance imaging (MRI). A new heteropodal GdIII chelate, [Gd-TREN-bis(6-Me-HOPO)-(TAM-TRI)(H2O)2] (Gd-2), is presented which is based on a hydroxypyridinate (HOPO)-terephthalamide (TAM) ligand design. Thermodynamic equilibrium constants for the acid-base properties and the GdIII complexation strength of TREN-bis(6-Me-HOPO)-(TAM-TRI) (2) were measured by potentiometric and spectrophotometric titration techniques, respectively. The pGd of 2 is 20.6 (pH 7.4, 25 degrees C, I = 0.1 M), indicating that Gd-2 is of more than sufficient thermodynamic stability for in vivo MRI applications. The water exchange rate of Gd-2 (kex = 5.3(+/-0.6) x 107 s-1) was determined by variable temperature 17O NMR and is in the fast exchange regime - ideal for MRI. Variable pressure 17O NMR was used to determine the volume of activation (DeltaV) of Gd-2. DeltaV for Gd-2 is -5 cm3 mol-1, indicative of an interchange associative (Ia) water exchange mechanism. The results reported herein are important as they provide insight into the factors influencing high stability and fast water exchange in the HOPO series of complexes, potentially future clinical contrast agents.  相似文献

On the basis of structural considerations in the inner sphere of nine-coordinate, monohydrated Gd(III) poly(aminocarboxylate) complexes, we succeeded in accelerating the water exchange by inducing steric compression around the water binding site. We modified the common DTPA(5-) ligand (DTPA=(diethylenetriamine-N,N,N',N",N"-pentaacetic acid) by replacing one (EPTPA(5-)) or two (DPTPA(5-)) ethylene bridges of the backbone by propylene bridges, or one coordinating acetate by a propionate arm (DTTA-prop(5-)). The ligand EPTPA(5-) was additionally functionalized with a nitrobenzyl linker group (EPTPA-bz-NO(2) (5-)) to allow for coupling of the chelate to macromolecules. The water exchange rate, determined from a combined variable-temperature (17)O NMR and EPR study, is two orders of magnitude higher on [Gd(eptpa-bz-NO(2))(H(2)O)](2-) and [Gd(eptpa)(H(2)O)](2-) than on [Gd(dtpa)(H(2)O)](2-) (k(ex)298=150x10(6), 330x10(6), and 3.3x10(6) s(-1), respectively). This is optimal for attaining maximum proton relaxivities for Gd(III)-based, macrocyclic MRI contrast agents. The activation volume of the water exchange, measured by variable-pressure (17)O NMR spectroscopy, evidences a dissociative interchange mechanism for [Gd(eptpa)(H(2)O)](2-) (DeltaV(not equal sign)=(+6.6+/-1.0) cm(3) mol(-1)). In contrast to [Gd(eptpa)(H(2)O)](2-), an interchange mechanism is proved for the macrocyclic [Gd(trita)(H(2)O)](-) (DeltaV (not equal sign)=(-1.5+/-1.0) cm(3) mol(-1)), which has one more CH(2) group in the macrocycle than the commercial MRI contrast agent [Gd(dota)(H(2)O)](-), and for which the elongation of the amine backbone also resulted in a remarkably fast water exchange. When one acetate of DTPA(5-) is substituted by a propionate, the water exchange rate on the Gd(III) complex increases by a factor of 10 (k(ex)298=31x10(6) s(-1)). The [Gd(dptpa)](2-) chelate has no inner-sphere water molecule. The protonation constants of the EPTPA-bz-NO(2) (5-) and DPTPA(5-) ligands and the stability constants of their complexes with Gd(III), Zn(II), Cu(II) and Ca(II) were determined by pH potentiometry. Although the thermodynamic stability of [Gd(eptpa-bz-NO(2))(H(2)O)](2-) is reduced to a slight extent in comparison with [Gd(dtpa)(H(2)O)](2-), it is stable enough to be used in medical diagnostics as an MRI contrast agent. Therefore both this chelate and [Gd(trita)(H(2)O)](-) are potential building blocks for the development of high-relaxivity macromolecular agents.  相似文献

The effect of silver ions on the water dissociation of bipolar membranes was first investigated in this paper. To do this, the bipolar membranes were prepared by immersing the anion exchange layers in an AgNO3 solution and then coating a solution of sulfonated polyphenylene oxide (SPPO) on the anion exchange layers. XPS and AES observations indicated that silver at the intermediate layer was in the form of AgCl. The experimental results proved that AgCl has an excellent catalytic function for water dissociation in terms of I-V curves, and the quantity of AgCl played an important role in the behavior of a bipolar membrane. The bipolar membranes with gelatin and the gelatin doped with silver as a catalytic layer were also prepared in the same way, and their I-V behavior and the water dissociation pilot tests were also investigated. The experimental results showed that in the case of gelatin alone, the voltage drop increased slightly at high gelatin concentrations, due mainly to the steric effect and electrostatic interaction, but decreased at low gelatin concentrations due to the hydrophilicity. However, when gelatin was doped with AgCl, the bipolar membranes have an appreciable improvement in both stability and catalytic function, in comparison with those prepared from silver or gelatin.  相似文献

Access to the fast exchange kinetics of labile protein hydrogens in solution is provided by exchange broadening of the water 1H NMR line. We analyzed the chemical shift modulation contribution of labile hydrogens in bovine pancreatic trypsin inhibitor (BPTI) to the transverse 1H spin relaxation rate, R2, of the bulk solvent. Both the experimental pH dependence and the CPMG dispersion of R2 could be quantitatively accounted for on the basis of known chemical shifts, exchange rates, and ionization constants for BPTI. This analysis provided, for the first time, the hydrogen exchange rate constants for Lys and Arg side chains in a protein and pointed to an internal catalysis of the N-terminal amino protons in BPTI by a salt bridge. The method can be used for mapping the hydrogen exchange rates in protein solutions and biomaterials, which may be important for the control of relaxation-weighted contrast in biological MRI.  相似文献

The authors developed a rigorous framework to model nanofiltration (NF) membrane selectivity at high feed water recoveries and verify it experimentally. The phenomenological model and the Donnan steric partitioning pore model (DSPM) were incorporated into a differential element approach for predicting removal of a variety of solutes from single salt solutions and natural water by NF membranes up to 90% feed water recovery in the temperature range 5-41 degrees C. In this approach, the entire membrane ensemble was divided into numerous sub-elements analogous to real-world full-scale NF installations, where concentrate (or reject) from one element feeds into the next element. Fundamental membrane properties (average pore radius, surface charge density, and ratio of thickness to porosity) and the reflection coefficient and permeability coefficient were first independently obtained for each solute-membrane-temperature combination using separate low recovery experiments with negligible concentration polarization and later used as model inputs to calculate solute removal in a purely predictive fashion for 5-90% recovery. This modeling approach accurately predicted removals from single salt solutions of NaCl and MgSO(4) as well as natural organic matter, disinfection by-product precursors, and several ions from pretreated Lake Houston water in a wide range of operating conditions demonstrating its use to simulate NF permeate water quality under real-world conditions of high feed water recovery.  相似文献

Isotopic exchange between oxygens of water and mu-O bridges in the di-mu-O dimanganese complexes, [(mes-terpy)2Mn2(III/IV)(mu-O)2(H2O)2](NO3)3 (1, mes-terpy = 4'-mesityl-2,2':6',2' '-terpyridine) and [(phen)4Mn2III/IV(mu-O)2](ClO4)3 (2, phen = 1,10-phenanthroline), has been investigated by a study of the kinetics of exchange. The data provide evidence for distinct mechanisms of exchange in 1 and 2 and suggest that these differences arise due to the presence and absence of terminal water-binding sites in 1 and 2, respectively. Exchange of oxygen atoms between water and mu-O bridges must involve the elementary steps of bridge protonation, deprotonation, opening, and closing. On the basis of the existing literature on these reactions in oxo-bridged metal complexes and our present data, we propose pathways of exchange in 1 and 2. The mechanism proposed for 1 involves an initial fast protonation of an oxo-bridge by water coordinated to Mn(IV), followed by a slow opening of the protonated bridge as proposed earlier for an analogous complex on the basis of DFT calculations. The mechanism proposed for 2 involves initial dissociation of phen, followed by coordination of water at the vacated sites, as observed for rearrangement of 2 to a trinuclear complex. The subsequent steps are proposed to be analogous to those for 1. Our results are discussed in the context of data on 18O-labeled water isotope exchange in photosystem II and provide support for the existence of fully protonated terminal waters bound to Mn in the O2-evolving complex of photosystem II.  相似文献

Water and acetic acid eliminations from 1- and 2-indan derivatives have been investigated. Deuterium labeling, high resolution peak matching and the metastable peak analysis capabilities of a high resolution triple analyzer (E B E) mass spectrometer were employed to examine the eliminations. These experiments showed that water was lost from 1-indanol via 1,2 and 1,3 processes. These results contrast with those obtained for 1-tetralol, which specifically eliminates water in a 1,4 process involving the benzylic hydrogens. Water elimination from 2-indanol is preceded by a slow hydroxyl-benzylic hydrogen exchange and proceeds specifically 1,2. Water losses from both 1- and 2-indanol are characterized by large kinetic energy releases. Acetic acid elimination is shown to occur specifically 1,3 from 1-acetoxyindan and 1,2 from 2-acetoxyindan.  相似文献

Isotopic H/D exchange between coadsorbed acetone and water on the TiO2(110) surface was examined using temperature programmed desorption (TPD) as a function of coverage and two surface pretreatments (O2 oxidation and mild vacuum reduction). Coadsorbed acetone and water interact repulsively on reduced TiO2(110) on the basis of results from the companion paper to this study, with water exerting a greater influence in destabilizing acetone and acetone having only a nominal influence on water. Despite the repulsive interaction between these coadsorbates, about 0.02 monolayers (ML) of a 1 ML d6-acetone on the reduced surface (vacuum annealed at 850 K to a surface oxygen vacancy population of 7%) exhibits H/D exchange with coadsorbed water, with the exchange occurring exclusively in the high-temperature region of the d6-acetone TPD spectrum at approximately 340 K. The effect was confirmed with combinations of d0-acetone and D2O. The extent of exchange decreased on the reduced surface for water coverages above approximately 0.3 ML due to the ability of water to displace coadsorbed acetone from first layer sites to the multilayer. In contrast, the extent of exchange increased by a factor of 3 when surface oxygen vacancies were pre-oxidized with O2 prior to coadsorption. In this case, there was no evidence for the negative influence of high water coverages on the extent of H/D exchange. Comparison of the TPD spectra from the exchange products (either d1- or d5-acetone depending on the coadsorption pairing) suggests that, in addition to the 340 K exchange process seen on the reduced surface, a second exchange process was observed on the oxidized surface at approximately 390 K. In both cases (oxidized and reduced), desorption of the H/D exchange products appeared to be reaction limited and to involve the influence of OH/OD groups (or water formed during recombinative desorption of OH/OD groups) instead of molecularly adsorbed water. The 340 K exchange process is assigned to reaction at step sites, and the 390 K exchange process is attributed to the influence of oxygen adatoms deposited during surface oxidation. The H/D exchange mechanism likely involves an enolate or propenol surface intermediate formed transiently during the desorption of oxygen-stabilized acetone molecules.  相似文献

This paper investigates the effect of polyvinyl alcohol (PVA) as the intermediate layer of bipolar membranes on water dissociation. The bipolar membranes are prepared by coating a solution of sulfonated polyphenylene oxide (SPPO) on the anion exchange layers, which had been immersed in PVA aqueous solutions with different concentrations in advance. The experimental results show that the effect of PVA on water dissociation is significantly affected by its concentration: at low concentration range PVA solution catalyzes water dissociation and at high concentration range PVA solution shows a retardant effect. The phenomenon is different from the situation where polyethylene glycol (PEG) is used as the intermediate layer. Based on the adsorption data and the measurements of X-ray photoelectron spectroscopy and electrochemical impedance spectroscopy, it is found that (1) the effect of PVA concentration on water dissociation can be explained by the competition and compromise of two effects--the catalytic and hydrophilic effect of the PVA molecule and the steric effect of the enlargement of junction thickness; (2) the reason that at high concentration range PVA and PEG show different effects on water dissociation is that the junction thickness is enlarged excessively by PVA, which is based on the intrinsic characteristics of PVA molecules.  相似文献
