Gas chromatography coupled to high-resolution time-of flight mass spectrometry (GC-HR-TOF-MS) is a powerful analytical technique with excellent capabilities due to its high sensitivity in full-spectrum-acquisition mode together with its resolving power and accurate-mass measurements. These features make this technique very attractive in qualitative analysis, especially for wide-scope screening of a large number of organic contaminants and residues at trace levels.Full-spectrum MS allows data processing, in principle, of an unlimited number of compounds in samples, as no analyte-specific information is required before the injection. Also, as all data remain available, retrospective analysis is always possible without the need to reinject the sample - an important advantage of full-spectrum MS techniques. Despite these advantages, GC-HR-TOF-MS has rarely been applied, so we expect promising results in different applied fields in the years ahead.We discuss in detail the characteristics and the potential of GC-HR-TOF-MS. We describe different analytical strategies from wide-scope target screening to investigation of unknowns in biology, the environment and food safety. Recent instrumental developments (e.g., high-speed analog-to-digital converter or soft ionization sources) and advances in software for processing the huge amount of data available open up new prospects, making GC-TOF-MS one of the most promising techniques to investigate the presence of organic compounds in different fields.  相似文献

Experimental conditions for the analysis have been optimized, and the accuracy and relative standard deviation of the method has been determined. A multiclass method has been developed for screening, quantification and confirmation of organic micro-pollutants in water by gas chromatography coupled to mass spectrometry with a triple quadrupole analyzer. The work has been focused on the determination of more than 50 compounds belonging to different chemical families: 19 organochlorine and organophosphorus insecticides, 6 herbicides, 7 polychlorinated biphenyls, 16 polycyclic aromatics hydrocarbons, 2 brominated diphenyl ethers, and 3 octyl/nonyl phenols and pentachlorobenzene. Most of these analytes are included in the list of priority substances in the framework on European Water Policy.Analyte extraction was performed by solid phase extraction using C18 cartridges, and five isotopically labeled standards were added before extraction as surrogates. Analyses were performed by gas chromatography with tandem mass spectrometry (MS/MS) in electron impact mode. Accuracy and precision were evaluated by means of recovery experiments using water samples fortified at two concentration levels (25 and 250 ng L−1), with satisfactory results for most of analytes. The excellent selectivity and sensitivity reached in selected reaction monitoring mode allowed us satisfactory quantification and confirmation at levels as low as 25 ng L−1. Two MS/MS transitions were acquired for each analyte, using the Q/q intensity ratio as a confirmatory parameter. The method developed was applied to the analysis of surface, ground and wastewater samples collected from the Valencia Region (Spain).Analytical methodology using negative chemical ionization mode was also validated for the organochlorine compounds selected, showing a superior sensitivity and lower detection limits.  相似文献

A method for the speciation of silicon compounds in petroleum products was developed using gas chromatography coupled to mass spectrometry (GC-MS). Prior to analysis, several precautions about storage and conservation were applied for all samples. In spiked gasoline samples, limits of detection between 24 and 69 μg kg(-1) for cyclic siloxanes (D(4)-D(6)) and between 1 and 7 μg kg(-1) for other species were obtained. In this study, cyclic siloxanes (D(n)) and one ethoxysilane were quantified for the first time in petroleum products by a specific method based on response factor calculation to an internal standard. This method was applied to four samples of naphthas and gasolines obtained from a steam cracking process. Cyclic siloxanes were predominant in four investigated samples with concentrations ranging between 101 and 2204 μg kg(-1). Cyclic siloxane content decreased with an increase in their degree of polymerization. During a steam cracking process, silicon concentrations determined by GC-MS SIM (single ion monitoring) significantly increase. This trend was confirmed by ICP-OES (inductively coupled plasma optical emission spectroscopy) measurements but a difference on the total silicon content was observed, certainly highlighting the presence of unknown silicon species. GC-MS SIM method gives access to the chemical nature of the silicon species, which is crucial for the understanding of hydrotreatment catalyst poisoning in the oil and gas industry.  相似文献

This study presents the optimization and application of an analytical method based on the use of stir bar sorptive extraction (SBSE) gas chromatography coupled to mass spectrometry (GC–MS) for the ultra-trace analysis of POPs (Persistent Organic Pollutants) in Arctic ice. In a first step, the mass-spectrometry conditions were optimized to quantify 48 compounds (polycyclic aromatic hydrocarbons, brominated diphenyl ethers, chlorinated biphenyls, and organochlorinated pesticides) at the low pg/L level. In a second step, the performance of this analytical method was evaluated to determine POPs in Arctic cores collected during an oceanographic campaign. Using a calibration range from 1 to 1800 pg/L and by adjusting acquisition parameters, limits of detection at the 0.1–99 and 102–891 pg/L for organohalogenated compounds and polycyclic aromatic hydrocarbons, respectively, were obtained by extracting 200 mL of unfiltered ice water. α-hexachlorocyclohexane, DDTs, chlorinated biphenyl congeners 28, 101 and 118 and brominated diphenyl ethers congeners 47 and 99 were detected in ice cores at levels between 0.5 to 258 pg/L. We emphasise the advantages and disadvantages of in situ SBSE in comparison with traditional extraction techniques used to analyze POPs in ice.  相似文献

Summary Liquid chromatography-mass spectrometry (LC-MS) with atmospheric pressure chemical ionization (APCI), and gas chromatography-mass spectrometry (GC-MS) with electron impact ionization (EI), are compared for the determination of eight pesticides in oranges. Seven of the selected pesticides, chlorpyriphos, chlorpyriphos-methyl, imazalil, α and β-endosulfan, endosulfan sulphate and dicofol, are commonly determined by GC whereas one, thiabendazole, can only be directly determined by LC. Primary ions [M-H] or [M-Cl+O] are obtained using LC-APCI-MS in negative ionization (NI) mode. In contrast, a high degree of fragmentation is reported with GC-MS. Both techniques were applied to oranges, which had been previously extracted with ethyl acetate and anhydrous sodium sulphate. The data indicate equivalent detection limits that range from 0.01 to 0.1 mgkg−1 and similar degree of specificity. Mean recoveries ranged from 82% for α-endosulfant to 96% for imazalil, with relative standard deviation ranging from 7 to 17%.  相似文献

Thirteen polycyclic aromatic hydrocarbons have been determined in soy‐based nutraceutical products. First, an optimization of extraction procedure was performed, and a solid–liquid extraction assisted by sonication and a dilute and shoot procedure were compared, selecting the dilute and shoot approach for the extraction of target compounds, utilizing a mixture of acetone/n‐hexane (1:1 v/v) as extractant solvent. After this, a clean‐up step was needed bearing in mind the complexity of these matrices. Dispersive solid‐phase extraction, using a mixture of C18 and Zr‐Sep+ (25 mg/mL each) was used. The separation was achieved by gas chromatography and detection with triple quadrupole tandem mass spectrometry. For quantification purposes, matrix‐matched calibration was used. The validation was applied at three concentration levels (20, 100 and 250 μg/kg), obtaining recoveries between 70 and 120% and precision values equal to or lower than 23%. Limits of detection and quantification were below 8 and 20 μg/kg, respectively. The method was applied in 11 samples, detecting five polycyclic aromatic hydrocarbons at concentrations ranging from 4.1 to 18.5 μg/kg.  相似文献

In this work, a multiclass screening method for organic contaminants in natural and wastewater has been developed and validated for qualitative purposes, i.e. to ensure the reliable and sensitive identification of compounds detected in samples at a certain level of concentration. The screening is based on the use of GC-TOF MS, and the sample procedure involves solid phase extraction with C(18) cartridges. Around 150 organic contaminants from different chemical families were investigated, including PAHs, octyl/nonyl phenols, PCBs, PBDEs and a notable number of pesticides, such as insecticides (organochlorines, organophosphorus, carbamates and pyrethroids), herbicides (triazines and chloroacetanilides), fungicides and several relevant metabolites. Surface water, ground water and effluent wastewater were spiked with all target analytes at three concentration levels (0.02, 0.1 and 1 μg/L). Influent wastewater and raw leachate from a municipal solid waste treatment plant were spiked at two levels (0.1 and 1 μg/L). Up to five m/z ions were evaluated for every compound. The identification criterion was the presence of, at least, two m/z ions at the expected retention time, measured at their accurate mass, and the accomplishment of the Q/q(i) intensity ratio within specified tolerances. The vast majority of compounds investigated were correctly identified in the samples spiked at 1 μg/L. When analyte concentration was lowered down to 0.1 μg/L the identification was more problematic, especially in complex-matrix samples like influent wastewater. On the contrary, many contaminants could be properly identified at the lowest level 0.02 μg/L in cleaner matrices. The procedure was applied to the screening of water samples of different origin and matrix composition and allowed the detection of several target contaminants. A highly reliable identification could be carried out thanks to the sensitive full-spectrum acquisition at accurate mass, the high selectivity reached with the use of narrow-mass window extracted ion chromatograms, the low mass errors observed in the positive detections and the Q/q ratio accomplishment.  相似文献

In this work, we report the development and validation of an analytical method for the trace level determination of 14 selected (EU-directive) priority organic pollutants (namely, 1,2,3-trichlorobenzene (1,2,3-TCB), 1,2,4-trichlorobenzene, 1,3,5-trichlorobenzene, hexachloro-1,3-butadiene, pentachlorobenzene, hexachlorobenzene, alachlor, α-hexachloro-cyclohexane (α-HCH), β-HCH, γ-HCH (lindane), δ-HCH, tetra-brominated diphenyl ether (tetra-BDE), penta-brominated diphenyl ether and hepta-brominated diphenyl ether) in wastewater samples from 5 different sewage treatment plants (STPs) located in Spain. The proposed methodology is based on liquid-liquid extraction with n-hexane followed by identification and confirmation of the selected pollutants by gas chromatography high-resolution mass spectrometry in selected ion recording acquisition mode. Recovery studies performed with spiked wastewater samples at two different concentration levels (0.1 and 1 μg L−1) gave mean recoveries in the range 80-120% (except for trichlorobenzenes, ca. with 50%) with RSD values below 10% in most cases, thus confirming the usefulness of the proposed methodology for the analyses of this kind of complex samples. The obtained detection limits in effluent wastewater matrices were in the low nanogram per liter range, with values as low as 0.09 ng L−1 for tetra-BDE and 0.3 ng L−1 for hexachlorobenzene. Finally, the proposed methodology was successfully applied to a monitoring study intended to characterize wastewater effluents of 5 different sewage treatment plants with different major activities (Industrial, Coastal, Urban). Most of the compounds targeted were detected in the ng L−1 range at concentrations ranging from 0.19 ng L−1 to 135 ng L−1 (hexachlorobenzene).  相似文献

A detection method for 19 kinds of phthalic acid ester compounds analyzed by n‐hexane/ether/acetonitrile 1:7:8 v/v/v mixed solvent extraction, quick, easy, cheap, effective, rugged, and safe purification and internal standard method of quantitative gas chromatography with mass spectrometry was established. This method can effectively remove interfering materials, such as lipids, fatty acids, and pigments, from dairy products. The 19 kinds of phthalic acid ester compounds were within a 0.025–0.2 mg/kg range, the recovery rate was 65.2–125.7%, relative standard deviation was 7.9–15.4% (n = 6), and the limit of detection was 0.005–0.02 mg/kg. Concentrations of the 19 kinds of phthalic acid ester compounds ranged between 0.01 and 0.12 mg/kg in ten dairy materials and 20 dairy products. The established method is simple, rapid, accurate, and highly sensitive.  相似文献

The presence of toxins in harmful algal blooms (HABs) poses considerable concerns because of their potential adverse effects on ecological environments and human health. When marine HABs occur, efficient screening and identification of toxins in different kinds of HAB algae remains a challenge. In this study, the applicability of serial coupling of reversed-phase liquid chromatography (RPLC) and hydrophilic interaction chromatography (HILIC) combined with high resolution mass spectrometry (HR-MS) for the simultaneous screening and identification of various kinds of known lipophilic and hydrophilic toxins in HAB algae was investigated for the first time. Ultrasound-assisted extraction (UAE) was explored to extract both lipophilic and hydrophilic toxins in algae simultaneously. As in most cases, toxin standards were not available; therefore, an identification procedure based on accurate mass data and chromatographic behavior was proposed. According to this procedure, eight known lipophilic toxins and 11 hydrophilic toxins were successfully detected in a single injection, and the proposed method was validated. Satisfactory sensitivity, repeatability (RSD <14.87%) and recovery (89.4–105.8%) of the method were achieved. A major advantage of the proposed method is that it can almost detect members of all eight groups of marine algal toxins in a single run. Using this method, several known toxins in different marine toxigenic algae including Alexandrium tamarense, Alexandrium minutum and Prorocentrum lima were successfully observed and identified. This work demonstrates that RPLC/HILIC-HR-MS combined with an accurate mass list of known marine algal toxins may be used as a powerful tool for screening of different classes of known toxins in marine harmful algae.  相似文献

The characteristic fragmentation pattern of six reducing and two non reducing type disaccharides-(neohesperidose, acuminose, sambubiose, rutinose, vicianose, primverose, and two arabinosyl-inositols) has been described. These saccharides have not been previously identified by on-line chromatographic techniques. Unambiguous specific characteristics of the TMS (oxime)s such as mass distribution, syn/anti oximes ratios and elution order proved to be associated with their reducing or non reducing character, with their aldosyl property and with the position of their O-glycosidic linkages. The practical utility of the mass fragmentation study of these rare disaccharides was demonstrated, at the first time, by the simultaneous, on-line identification and quantification of the acuminose, vicianose, primverose and arabinosyl-inositol contents of tea leaves, from green and black tea blends of Indian and Chinese origin.  相似文献

Phthalates (PAEs) are ubiquitous toxic chemical compounds. During the last few years, some phthalate metabolites (MPAEs) have been proposed as appropriate biomarkers in human urine samples to determine PAE human intake and exposure. So, it is necessary to have fast, easy, robust and validated analytical methods to determine selected MPAEs in urine human samples. Two different instrumental methods based on gas (GC) and ultra-high performance liquid (UHPLC) chromatography coupled to mass spectrometry (MS) have been optimized, characterized and validated for the simultaneous determination of nine primary and secondary phthalate metabolites in urine samples. Both instrumental methods have similar sensitivity (detection limits ranged from 0.03 to 8.89 pg μL−1 and from 0.06 to 0.49 pg μL−1 in GC–MS and UHPLC–MS2, respectively), precision (repeatability, expressed as relative standard deviation, which was lower than 8.4% in both systems, except for 5OH-MEHP in the case of GC–MS) and accuracy. But some advantages of the UHPLC–MS2 method, such as more selectivity and lower time in the chromatographic runs (6.8 min vs. 28.5 min), have caused the UHPLC–MS2 method to be chosen to analyze the twenty one human urine samples from the general Spanish population. Regarding these samples, MEP showed the highest median concentration (68.6 μg L−1), followed by MiBP (23.3 μg L−1), 5cx-MEPP (22.5 μg L−1) and MBP (19.3 μg L−1). MMP (6.99 μg L−1), 5oxo-MEHP (6.15 μg L−1), 5OH-MEHP (5.30 μg L−1) and MEHP (4.40 μg L−1) showed intermediate levels. Finally, the lowest levels were found for MBzP (2.55 μg L−1). These data are within the same order of magnitude as those found in other similar populations.  相似文献

Yeast and yeast cultures are frequently used as additives in diets of dairy cows. Beneficial effects from the inclusion of yeast culture in diets for dairy mammals have been reported, and the aim of this study was to develop a comprehensive analytical method for the accurate mass identification of the ‘global’ metabolites in order to differentiate a variety of yeasts at varying growth stages (Diamond V XP, Yea-Sacc and Levucell). Microwave-assisted derivatization for metabolic profiling is demonstrated through the analysis of differing yeast samples developed for cattle feed, which include a wide range of metabolites of interest covering a large range of compound classes. Accurate identification of the components was undertaken using GC-oa-ToFMS (gas chromatography-orthogonal acceleration-time-of-flight mass spectrometry), followed by principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) for data reduction and biomarker discovery. Semi-quantification (fold changes in relative peak areas) was reported for metabolites identified as possible discriminative biomarkers (p-value <0.05, fold change >2), including d-ribose (four fold decrease), myo-inositol (five fold increase), l-phenylalanine (three fold increase), glucopyranoside (two fold increase), fructose (three fold increase) and threitol (three fold increase) respectively.  相似文献

An experimental method for the analysis of volatile organic compounds in polymers is described. The technique involves dynamic headspace sampling, collection, and concentration of the volatiles in a cold trap, followed by capillary column gas chromatography/mass spectometry. Flow switching is carried out by the Deans switching technique. Four technical polymers used as pharmaceutical packaging materials have been analyzed in order to demonstrate the method.  相似文献

A method for the analysis of chlorinated and brominated polycyclic aromatic hydrocarbon (Cl-/Br-PAHs) congeners in environmental samples, such as a soil extract, by comprehensive two-dimensional gas chromatography coupled to a high resolution time-of-flight mass spectrometry (GC×GC-HRTOF-MS) is described. The GC×GC-HRTOF-MS method allowed highly selective group type analysis in the two-dimensional (2D) mass chromatograms with a very narrow mass window (e.g. 0.02Da), accurate mass measurements for the full mass range (m/z 35-600) in GC×GC mode, and the calculation of the elemental composition for the detected Cl-/Br-PAH congeners in the real-world sample. Thirty Cl-/Br-PAHs including higher chlorinated 10 PAHs (e.g. penta, hexa and hepta substitution) and ClBr-PAHs (without analytical standards) were identified with high probability in the soil extract. To our knowledge, highly chlorinated PAHs, such as C(14)H(3)Cl(7) and C(16)H(3)Cl(7), and ClBr-PAHs, such as C(14)H(7)Cl(2)Br and C(16)H(8)ClBr, were found in the environmental samples for the first time. Other organohalogen compounds; e.g. polychlorinated biphenyls (PCBs), polychlorinated naphthalenes (PCNs), and polychlorinated dibenzofurans (PCDFs) were also detected. This technique provides exhaustive analysis and powerful identification for the unknown and unconfirmed Cl-/Br-PAH congeners in environmental samples.  相似文献

Simple and rapid gas chromatographic (GC) method employing a high-resolution time-of-flight mass analyzer that enables direct analysis (no derivatization) of acrylamide in various heat-processed foodstuffs has been developed and validated. Co-isolation of acrylamide precursors such as sugars and asparagine, constituting the risk of results overestimation due to additional formation of analyte in hot GC injector, is avoided by the extraction with n-propanol followed by solvent exchange to acetonitrile (MeCN). Introduction of a novel purification strategy, dispersive solid phase extraction, based on addition of primary-secondary amine (PSA) sorbent into deffated extract in MeCN, provides a significant reduction of some abundant matrix co-extracts (mainly free fatty acids). Isotope dilution technique (d3-acrylamide as an internal standard) is employed for compensation of potential target analyte losses and/or matrix-inducted chromatographic response enhancement. Limits of quantifications (LOQs) ranged between 15 and 40 μg kg−1 and recoveries were between 97 and 108% depending on the examined food matrix. The repeatability of measurements (expressed as relative standard deviation, R.S.D.) was as low as 1.9% for potato crisps containing acrylamide at a level of 1 mg kg−1. Slightly higher values (R.S.D. < 4.0%) were achieved for breakfast cereals and crisp bread with approximately 10 times lower content of this processing contaminant. Trueness of results generated by this new method was demonstrated via FAPAS® (Food Analysis Performance Assessment Scheme) interlaboratory proficiency tests.  相似文献
