A new three step synthetic route to construct azocine ring system using anion chemistry and intramolecular Friedel-Craft reaction of an ester is presented. This method allows synthesis of azocine ring analogues in excellent overall yields. The designed strategy was applied for the synthesis of naturally occurring magallanesine.  相似文献

We published recently our results on a new and convergent synthesis of natural steroids. The strategy was based on a cycloaddition reaction of Nazarov reagents 2 and 5 with cyclohexenones 1 and 4. In this paper we report results that deal with the synthesis of two new bicyclic Nazarov reagents (13 and 19) and their cycloaddition with two cyclohexenones (1 and 4). These new results constitute an important improvement concerning the versatility of the strategy since tetracycles having the stereochemistry found in natural steroids are now available.  相似文献

A new strategy for the synthesis of oligopeptides was developed using an ionic liquid as a soluble support. The efficiency of this ionic liquid-phase approach was demonstrated by the synthesis of a bioactive pentapeptide, Leu(5)-enkephalin, in good yield and reasonable purity. The structures and purities of the reaction intermediates in each step were verified easily by routine spectroscopic analysis, and no chromatographic procedures were needed during the synthesis.  相似文献

Triquinanes and their oxygenated congeners, oxa- and dioxa-triquinanes, exhibit versatile biological activities in conjunction with synthetically challenging molecular architecture. Owing to these properties, several new strategies have been developed to accomplish the synthesis of these sesquiterpenes. Among the new strategies, cascade radical cyclization strategy has been broadly explored and well studied. Herein, we report our efforts in detail for the synthesis of dioxa-triquinanes using a domino enyne/RCM strategy as the key step. Carbohydrate based synthesis not only allows the use of inexpensive and optically pure starting materials, but also the furanose derivatives, which already possess one of the requisite dihydro-furan moieties in the desired dioxa-triquinane. The other two five-membered rings were constructed simultaneously by the cascade enyne/RCM reaction using the Grubbs' second-generation catalyst. During the course of our synthesis it was observed that the acetonide protection hinders the RCM reaction, after the initial enyne metathesis reaction. The reaction underwent smoothly under argon atmosphere, whilst use of ethylene atmosphere was found to hinder the formation of the tandem enyne/RCM product. A three-component reaction based on trapping of ammonium ylides with formaldehyde was first reported. This reaction offers a new strategy for the synthesis of α-aryl serine derivatives and can be extended to the preparation of α-aryl threonine derivatives. A stereocontrolled total synthesis of (+)-longicin, a representative of the class of mono-THF-acetogenins, is described. The strategy involves the utilization of d- and l-glutamic acids as chirons that correspond to two five-carbon segments harboring stereogenic centers at C4 and at C17 of the C(32) polyketide-derived natural product. The use of Grubbs' RCM reaction as a novel "chain elongation" strategy for the synthesis of acetogenin-type structures and a new protocol for butenolide incorporation are also described. An efficient and green one-pot synthesis route for tetrahydrochromene derivatives was developed. Lipase from Porcine pancreas (PPL) shows excellent catalytic activity and exerts good adaptability to different substrates in the reaction. All the reactions go smoothly and provide tetrahydrochromene derivatives with isolate yield up to 97% under room temperature. This lipase-catalyzed multistep conversion method has provided a new strategy to synthesize chromene derivatives and expanded the application of enzyme in organic synthesis.  相似文献

A tandem anodic coupling-Friedel-Crafts alkylation strategy has been used to rapidly complete the asymmetric synthesis of alliacol A. The anodic oxidation reaction allowed for the generation of a new bond between two nucleophiles. In the synthesis, the absolute stereochemistry of the final natural product is set relative to a methyl group that is incorporated early in the sequence using an asymmetric Michael reaction.  相似文献

A stereoselective synthesis of coronafacic acid, a natural component of the phytotoxin coronatin, was achieved using an intramolecular Diels-Alder reaction as the key step. The triene precursor bearing a substituted diene and a vinylketone as dienophile was synthesized and then tested in the thermal intramolecular cyclization. We have devised a new strategy to assemble the E,Z-diene through the stereoselective aldol reaction of an ester enolate followed by a stereoselective dehydration. Following the thermal cyclization, the corresponding hydrindanone thereby obtained with the desired relative stereochemistry could easily be converted into the natural product. The synthesis of the coronafacic acid was accomplished in six steps in 29% overall yield.  相似文献

[reaction--see text] A new strategy for the synthesis of 2,3-disubstituted cyclopentenones emerges from two key reactions-the ruthenium-catalyzed three-component coupling of an equivalent of HBr, an alkyne, and a vinyl ketone and the Ni-Cr Barbier type reaction. As a result, these important structures are readily accessed from an alkyne and a vinyl ketone (which derive directly from carboxylic acids). We here report on a new straightforward strategy for the synthesis of cyclic cholic acid–peptide conjugates. A solid-phase synthesis method is presented in which a selected anti-lysozyme CDR3 fragment, Asp-Ser-Thr-Ile-Tyr-Ala-Ser-Tyr-Tyr-Glu-Ser, is immobilized onto a steroidal cholic acid derived scaffold in order to yield a loop-like structure. Therefore, part of the desired sequence, that is, Ser-Tyr-Tyr-Glu-Ser, is introduced, at the C12 position of the scaffold. Subsequently, the remainder of the envisaged sequence is introduced at C3 via a Cu-catalyzed cyclo-addition reaction. Finally, amide bond formation delivers the desired cyclic peptidosteroid. This new synthetic strategy offers an easy and short access to cyclic peptidosteroids via convergent peptide ligation and macrocyclization.  相似文献
