3‐(ω′‐Alkenyl)‐substituted 5,6‐dihydro‐1H‐pyridin‐2‐ones 2 – 4 were prepared as photocycloaddition precursors either by cross‐coupling from 3‐iodo‐5,6‐dihydro‐1H‐pyridin‐2‐one ( 8 ) or—more favorably—from the corresponding α‐(ω′‐alkenyl)‐substituted δ‐valerolactams 9 – 11 by a selenylation/elimination sequence (56–62 % overall yield). 3‐(ω′‐Alkenyloxy)‐substituted 5,6‐dihydro‐1H‐pyridin‐2‐ones 5 and 6 were accessible in 43 and 37 % overall yield from 3‐diazopiperidin‐2‐one ( 15 ) by an α,α‐chloroselenylation reaction at the 3‐position followed by nucleophilic displacement of a chloride ion with an ω‐alkenolate and oxidative elimination of selenoxide. Upon irradiation at λ=254 nm, the precursor compounds underwent a clean intramolecular [2+2] photocycloaddition reaction. Substrates 2 and 5 , tethered by a two‐atom chain, exclusively delivered the respective crossed products 19 and 20 , and substrates 3 , 5 , and 6 , tethered by longer chains, gave the straight products 21 – 23 . The completely regio‐ and diastereoselective photocycloaddition reactions proceeded in 63–83 % yield. Irradiation in the presence of the chiral templates (?)‐ 1 and (+)‐ 31 at ?75 °C in toluene rendered the reactions enantioselective with selectivities varying between 40 and 85 % ee. Truncated template rac‐ 31 was prepared as a noranalogue of the well‐established template 1 in eight steps and 56 % yield from the Kemp triacid ( 24 ). Subsequent resolution delivered the enantiomerically pure templates (?)‐ 31 and (+)‐ 31 . The outcome of the reactions is compared to the results achieved with 4‐substituted 5,6‐dihydro‐1H‐pyridin‐2‐ones and quinolones.  相似文献

Reported herein is an example of highly regio‐, diastereo‐ and enantioselective Cu(I)‐catalyzed intermolecular [3+2] cycloaddition reaction of α‐substituted iminoesters with α‐trifluoromethyl α,β‐unsaturated esters. This novel strategy provided a facile access to pyrrolidines with two skipped (aza)quaternary stereocenters including a CF3 all‐carbon quaternary stereocenter. A broad substrate scope was observed and high yields (up to 94%) with excellent diastereoselectivity (up to >20 : 1 d.r.) and enantioselectivity (up to 98% ee) were obtained.  相似文献

(E)‐α,β‐Unsaturated pyrazoleamides undergo facile dienolization to furnish copper(I)‐(1Z,3Z)‐dienolates as the major in the presence of a copper(I)‐(R)‐DTBM‐SEGPHOS catalyst and Et3N, which react with aldimines to afford syn‐vinylogous products as the major diastereoisomers in high regio‐ and enantioselectivities. In some cases, the diastereoselectivity is low, possibly due to the low ratio of copper(I)‐(1Z,3Z)‐dienolates to copper(I)‐(1Z,3E)‐dienolates. (Z)‐Allylcopper(I) species is proposed as effective intermediates, which may form an equilibrium with copper(I)‐(1Z,3Z)‐dienolates. Interestingly, the present methodology is a nice complement to our previous report, in which (E)‐β,γ‐unsaturated pyrazoleamides were employed as the prenucleophiles in the copper(I)‐catalyzed asymmetric vinylogous Mannich‐Type reaction and anti‐vinylogous products were obtained. In the previous reaction, copper(I)‐ (1Z,3E)‐dienolates were generated through α‐deprotonation, which might form an equilibrium with (E)‐allylcopper(I) species. Therefore, it is realized in the presence of a copper(I) catalyst that (E)‐α,β‐unsaturated pyrazoleamides lead to syn‐products and (E)‐β,γ‐unsaturated pyrazoleamides lead to anti‐products. Finally, by use of (E)‐β,γ‐unsaturated pyrazoleamide, (E)‐α,β‐unsaturated pyrazoleamide, (R)‐DTBM‐SEGPHOS, and (S)‐DTBM‐SEGPHOS, the stereodivergent synthesis of all four stereoisomers is successfully carried out. Then by following a three‐step reaction sequence, all four stereoisomers of N‐Boc‐2‐Ph‐3‐Me‐piperidine are synthesized in good yields, which potentially serve as common structure units in pharmaceutically active compounds.  相似文献

A method for catalytic regio‐ and enantioselective synthesis of trifluoromethyl‐substituted and aryl‐, heteroaryl‐, alkenyl‐, and alkynyl‐substituted homoallylic α‐tertiary NH2‐amines is introduced. Easy‐to‐synthesize and robust N‐silyl ketimines are converted to NH‐ketimines in situ, which then react with a Z‐allyl boronate. Transformations are promoted by a readily accessible l ‐threonine‐derived aminophenol‐based boryl catalyst, affording the desired products in up to 91 % yield, >98:2 α:γ selectivity, >98:2 Z:E selectivity, and >99:1 enantiomeric ratio. A commercially available aminophenol may be used, and allyl boronates, which may contain an alkyl‐, a chloro‐, or a bromo‐substituted Z‐alkene, can either be purchased or prepared by catalytic stereoretentive cross‐metathesis. What is more, Z‐trisubstituted allyl boronates may be used. Various chemo‐, regio‐, and diastereoselective transformations of the α‐tertiary homoallylic NH2‐amine products highlight the utility of the approach; this includes diastereo‐ and regioselective epoxide formation/trichloroacetic acid cleavage to generate differentiated diol derivatives.  相似文献

By application of substoichiometric amounts (50 mol %) of a chiral Lewis acid, the intramolecular [2+2] photocycloaddition of the title compounds was achieved with high enantioselectivity (up to 94 % ee). Upon cleavage of the cyclobutane ring the resulting tricyclic products underwent ring‐expansion reactions under acidic conditions and formed anellated seven‐ or eight‐membered‐ring systems without racemization. The ring expansion could be combined with a diastereoselective reduction (triethylsilane) or allylation (allyltrimethylsilane) upon BF3 catalysis (48–87 % yield).  相似文献

A highly regio‐, diastereo‐ and enantioselective Michael addition–alkylation reaction between α‐substituted cyano ketones and (Z)‐bromonitrostyrenes has been realized by using a chiral N,N′‐dioxide as organocatalyst. A variety of substrates performed well in this reaction, and the corresponding multifunctionalized chiral 2,3‐dihydrofurans were obtained in up to 95 % yield with 95:5 dr and 93 % ee.  相似文献

Two [2+2] photocycloaddition routes were evaluated as possible ways to access the tricyclic core structure found in the terpene monoglycoside lactiflorin. While the first route via γ‐substituted cyclopentenones was quickly discarded, the reactions of racemic (5R*)‐3‐benzyloxy‐5‐but‐3′‐enyl‐4‐methoxycarbonylfuran‐2(5 H)‐ones proceeded in high yields and with perfect diastereoselectivity. However, it turned out that the regioselectivity was strongly dependent on the substitution pattern within the but‐3′‐enyl chain, which connects the terminal olefinic double bond to the photoexcited butenolide chromophor. If the chain was unsubstituted or if a tert‐butyldimethylsilyloxy group was placed at the 2′ position in a syn‐relationship to the existing stereogenic center (5R*,2′S*), the crossed product prevailed with regioselectivities of 89:11 to 69:31. If the tert‐butyldimethylsilyloxy group was positioned at 2′ in an anti‐relationship to the existing stereogenic center (5R*,2′R*), the desired straight products were obtained in regioselectivities of 74:24 to 55:45 (61–83 % yield). Following this route, the aglycon part of lactiflorin was obtained by an intramolecular [2+2] photocycloaddition and a subsequent hydrogenolysis in 53 % yield. Its further conversion into the natural product after glycosylation included a methyl addition to the lactone carbonyl group, which was optimized to give the desired key intermediate in a yield of 70 %. The further conversion to lactiflorin was achieved in four steps and with an overall yield of 49 %.  相似文献

Annulenoid Tetrathiafulvalenes: 5,16‐Bis(1,3‐benzodithiol‐2‐ylidene)‐5,16‐dihydrotetraepoxy‐ and 5,16‐Bis(1,3‐benzodithiol‐2‐ylidene)‐5,16‐dihydrotetraepithio[22]annulenes(2.1.2.1) The title compounds are among the first tetrathiafulvalenes with annulene spacers, here with tetraepoxy‐[22]annulene(2.1.2.1) (see 3a ), tetraepithio[22]annulene(2.1.2.1) (see 3b ), and diepithiodiepoxy[22]annulene(2.1.2.1) (see 23 ) units. The annulenoid tetrathiafulvalenes 3a and 3b are prepared by cyclizing McMurry coupling of the 5,5′‐(1,3‐benzodithiol‐2‐ylidenemethylene)bis[furan‐ or thiophene‐2‐carbaldehydes] ( 8a or 8b , resp.) or by Wittig reaction of (1,3‐benzodithiol‐2‐yl)tributylphosphonium tetrafluoroborate ( 13b ) with tetraepoxy[22]annulene(2.1.2.1)‐1,12‐dione 20 (formation of 3a ) or diepithiodiepoxy[22]annulene(2.1.2.1)‐1,12‐dione 22 (formation of 23 ). The annulenoide tetrathiafulvalene 3a is obtained as a mixture of the isomers (E,E)‐ and (Z,Z)‐ 3a . At 130°, (Z,Z)‐ 3a rearranges quantitatively into the (E,E)‐isomer. Isomer (E,E)‐ 3a is a dynamic molecule, where the (E)‐ethene‐1,2‐diyl bridges rotate around the adjacent σ‐bonds. The tetraepithioannulene derivative 3b as well as 23 only exist in the (Z,Z)‐configuration. The oxidation of (E,E/Z,Z)‐ 3a with Br2 yields the annulene‐bridged tetrathiafulvalene dication (E,E)‐ 3a Ox, while with 4,5‐dichloro‐3,6‐dioxocyclohexa‐1,4‐diene‐1,2‐dicarbonitrile (DDQ) obviously only the radical cation 3a Sem is formed, which belongs to the class of cyanine‐like violenes. The annulenoide tetrathiafulvalenes 3b and 23 , which exist only in the (Z,Z)‐configuration, obviously for steric reasons, cannot be oxidized by DDQ. Electrochemical studies are in agreement with these results.  相似文献

Parallel and practical methods for the preparation of both (E)‐ and (Z)‐β‐aryl1‐β‐aryl2‐α,β‐unsaturated esters 1 and (E)‐ and (Z)‐α‐aryl1‐β‐aryl2‐α,β‐unsaturated esters 2 are described. These methods involve accessible, robust, stereocomplementary N‐methylimidazole (NMI)‐mediated enol tosylations (14 examples, 70–99 % yield), as well as stereoretentive Suzuki–Miyaura cross‐couplings (36 examples, 64–99 % yield). The highlighted feature of the present protocol is the use of parallel and stereocomplementary approaches to obtain highly (E)‐ and (Z)‐pure products 1 and 2 by utilizing sequential enol tosylations and cross‐coupling reactions. An expeditious and parallel synthesis of (E)‐ and (Z)‐zimelidine ( 3 ), which is a highly representative selective serotonin reuptake inhibitor (SSRI), was performed by utilizing the present methods.  相似文献

The intermolecular cyclopropanation of styrene with ethyl diazo(triethylsilyl)acetate ( 1a ) proceeds at room temperature in the presence of chiral RhII carboxylate catalysts derived from imide‐protected amino acids and affords mixtures of trans‐ and cis‐cyclopropane derivatives 2a in up to 72% yield but with modest enantioselectivities (<54%) (Scheme 1 and Table 1). Protiodesilylation of a diastereoisomer mixture 2a with Bu4NF is accompanied by epimerization at C(1) (→ 3 ). The intramolecular cyclopropanation of allyl diazo(triethylsilyl)acetate ( 8a ), in turn, affords optically active 3‐oxabicyclo[3.1.0]hexan‐2‐one ( 9a ) with yields of up to 85% and 56% ee (Scheme 3 and Table 2). Similarly, the (2Z)‐pent‐2‐enyl derivative 8d reacts to 9d in up to 77% yield and 38% ee (Scheme 3 and Table 3). In contrast, the diazo decomposition of (2E)‐3‐phenylprop‐2‐enyl and 2‐methylprop‐2‐en‐1‐yl diazo(triethyl‐silyl)acetates ( 8b and 8c , resp.) is unsatisfactory and gives very poor yields of substituted 3‐oxabicyclo[3.1.0]hexan‐2‐ones 9b and 9c , respectively (Table 3).  相似文献

(E)-α-Stannylvinyl phenyl(or p-tolyl)sulfones underwent an iododestannylation reaction to afford (E)-α-iodovinyl phenyl(or p-tolyl)sulfones 1, which reacted with (E)-alkenylzirconium(IV) complexes 2 produced in situ by hydrozirconation of terminal alkynes in the presence of a Pd(PPh3)4 catalyst to afford stereoselectively (1Z,3E)-2- phenyl(or p-tolyl)sulfonyl-substituted 1,3-dienes 3 in good yields.  相似文献

Asymmetric intramolecular direct hydroarylation of α‐ketoamides gives various types of optically active 3‐substituted 3‐hydroxy‐2‐oxindoles in high yields with complete regioselectivity and high enantioselectivities (84–98 % ee). A direct catalytic asymmetric aldol‐type reaction of 3‐substituted‐2‐oxindoles with glyoxal derivatives and ethyl trifluoropyruvate, catalyzed by a chiral N,N′‐dioxide–Sc(OTf)3 (Tf=trifluoromethanesulfonyl) complex, has been developed that tolerates a wide range of substrates. The reaction proceeds in good yields and excellent enantioselectivities (up to 93 % yield, 99:1 diastereomeric ratio (dr), and >99 % enantiomeric excess (ee)) under mild conditions, to deliver 3‐(α‐hydroxy‐β‐carbonyl) oxindoles with vicinal quaternary–tertiary or quaternary–quaternary stereocenters. Even with 1 mol % catalyst loading or on scaleup (10 mmol of starting material), maintenance of ee was observed, which showed the potential value of the catalyst system. In studies probing the reaction mechanism, a positive nonlinear effect was observed and ScIII‐based enolate intermediates were detected by using ESIMS. On the basis of the experimental results and previous reports, a possible catalytic cycle was assumed.  相似文献

A Pd(dba)2–P(OEt)3 combination allowed the silastannation of arylacetylenes, 1‐hexyne or propargyl alcohols with tributyl(trimethylsilyl)stannane to take place at room temperature, producing (Z)‐2‐silyl‐1‐stannyl‐1‐substituted ethenes in high yields. Novel silyl(stannyl)ethenes were fully characterized by 1H‐, 13C‐, 29Si‐ and 119Sn‐NMR as well as infrared and mass analyses. Treatment of a series of (Z)‐1‐aryl‐2‐silyl‐1‐stannylethenes and (Z)‐1‐(3‐pyridyl)‐2‐silyl‐1‐stannylethene with hydrochloric acid or hydroiodic acid in the presence of tetraethylammonium chloride (TEACl) or tetrabutylammonium iodide (TBAI) led to the exclusive formation of (E)‐trimethyl(2‐arylethenyl)silanes with high stereoselectivity. A similar reaction of (Z)‐1‐(2‐anisyl)‐2‐silyl‐1‐stannylethene also produced E‐type trimethyl[2‐(2‐anisyl)ethenyl]silane, while (Z)‐trimethyl [2‐(2‐pyridyl)ethenyl]silane was produced exclusively from (Z)‐1‐(2‐pyridyl)‐2‐silyl‐1‐stannylethene. Protodestannylation of (Z)‐1‐[hydroxy(phenyl)methyl]‐2‐silyl‐1‐stannylethene with trifluoroacetic acid took place via the β‐elimination of hydroxystannane, providing trimethyl(3‐phenylpropa‐1,2‐dienyl)silane quite easily. 4‐(Acylamino)‐5‐nitrosopyrimidines react either by a reductive condensation to provide 8‐substituted guanines, or by a Diels–Alder cycloaddition, or an ene reaction, to provide 6‐substituted pteridinones, depending on the nature of the acyl group and the reaction conditions. Experimental details are provided for the transformation of (acylamino)‐nitrosopyrimidines to 8‐substituted guanines, and the scope of the reaction is further demonstrated by transforming the trifluoro acetamide 25 to the 8‐(trifluoromethyl)guanine ( 27 ), and the N,Nbis(nitrosopyrimidinyl)‐dicarboxamide 29 to the (R,R)‐1,2di(guan‐8‐yl)ethane‐1,2‐diol ( 32 ). An intramolecular Diels–Alder reaction of the N‐sorbyl (=N‐hexa‐2,4‐dienoyl) nitrosopyrimidine 10 , followed by a spontaneous elimination to cleave the N,O bond of the initial cycloaddition product provided the pteridinones 14 or 15 , characterized by a (Z)‐ or (E)‐3‐hydroxyprop‐1‐enyl group at C(6). Treatment of 10 with Ph3P led to the C(8)‐penta‐1,3‐dienyl‐guanine 18 . The ene reaction of the N‐crotonyl (=N‐but‐2‐enoyl) nitrosopyrimidine 19 provided the 6‐vinyl‐pteridinone 20a that dimerized readily to 21a , while treatment of 19 with Ph3P led in high yield to 8‐(prop‐1‐enyl)guanine ( 23 ). The structure of the dimer 21 was established by X‐ray analysis of its bis(N,N‐dimethylformamidine) derivative 21b . The crystal structure of the nitroso amide 10 is characterized by two molecules in the centrosymmetric unit cell. Intermolecular H‐bonds connect the amino group to the amide carbonyl and to N(1). The crystalline bis(purine) 30 forms a left‐handed helix with four molecules per turn and a pitch of 30.2 Å.  相似文献

Twelve compounds unknown in the literature N‐(E)‐2‐stilbenyloxymethylenecarbonyl substituted hydrazones of 2‐, 3‐ and 4‐pyridinecarboxaldehydes, as well as methyl‐3‐pyridylketone have been prepared. The stereochemical behavior of these compounds in dimethyl‐d6 sulfoxide solution has been studied by 1H NMR technique. The E geometrical isomers and cis/trans amide conformers have been found for N‐substituted hydrazones 1–12. EI induced mass spectral fragmentation of these compounds were also investigated. The data obtained create the basis for distinguishing isomers.  相似文献

The reaction of the 4‐hydroxyquinoline‐3‐carboxylate 6 with pentaerythritol tribromide gave the 1,1′‐(2‐methylenepropane‐1,3‐diyl)di(4‐quinolone‐3‐carboxylate) 11 , whose reaction with bromine afforded the 1,1′‐(2‐bromo‐2‐bromomethylpropane‐1,3‐diyl)di(4‐quinolone‐3‐carboxylate) 12 . Compound 12 was transformed into the (Z)‐1,1′‐(2‐acetoxymethylpropene‐1,3‐diyl)di(4‐quinolone‐3‐carboxylate) 13 or (E)‐1,1′‐[2‐(imidazol‐1‐ylmethyl)propene‐1,3‐diyl]di(4‐quinolone‐3‐carboxylate) 14 . Hydrolysis of the dimer (Z)‐ 13 or (E)‐ 14 with potassium hydroxide provided the (E)‐1,1′‐(2‐hydroxymethylpropene‐1,3‐diyl)di(4‐quinolone‐3‐carboxylic acid) 15 or (Z)‐1,1′‐[2‐(imidazol‐1‐ylmethyl)propene‐1,3‐diyl]di(4‐quinolone‐3‐carboxylic acid) 16 , respectively. The nuclear Overhauser effect (NOE) spectral data supported that those hydrolysis resulted in the geometrical conversion of (Z)‐ 13 into (E)‐ 15 or (E)‐ 14 into (Z)‐ 16 .  相似文献

Kinetic measurements for the thermal rearrangement of 2,2‐diphenyl‐1‐[(E)‐styryl]cyclopropane ( 22a ) to 3,4,4‐triphenylcyclopent‐1‐ene ( 23a ) in decalin furnished ΔH =31.0±1.2 kcal mol?1 and ΔS =?6.0±2.6 e.u. The lowering of ΔH by 20 kcal mol?1, compared with the rearrangement of the vinylcyclopropane parent, is ascribed to the stabilization of a transition structure (TS) with allylic diradical character. The racemization of (+)‐(S)‐ 22a proceeds with ΔH =28.2±0.8 kcal mol?1 and ΔS =?5±2 e.u., and is at 150° 106 times faster than the rearrangement. Seven further 1‐(2‐arylethenyl)‐2,2‐diphenylcyclopropanes 22 , (E)‐ and (Z)‐isomers, were synthesized and characterized. The (E)‐compounds showed only modest substituent influence in their krac (at 119.4°) and kisom (at 159.3°) values. The lack of solvent dependence of rate opposes charge separation in the TS, but a linear relation of log krac with log p.r.f., i.e., partial rate factors of radical phenylations of ArH, agrees with a diradical TS. The ring‐opening of the preponderant s‐trans‐conformation of 22 gives rise to the 1‐exo‐phenylallyl radical 26 that bears the diphenylethyl radical in 3‐exo‐position, and is responsible for racemization. The 1‐exo‐3‐endo‐substituted allylic diradical 27 arises from the minor s‐gauche‐conformation of 22 and is capable of closing the three‐ or the five‐membered ring, 22 or 23 , respectively. The discussion centers on the question whether the allylic diradical is an intermediate or merely a TS. Quantum‐chemical calculations by Houk et al. (1997) for the parent vinylcyclopropane reveal the lack of an intermediate. Can the conjugation of the allylic diradical with three Ph groups carve the well of an intermediate?  相似文献

The lithium salts of the Me3Si‐ as well as Me3Si‐ and Me2SiF‐substituted Cyclotrisilazanes I and II react with tert‐butylacylchloride under ring contraction and formation of the cyclodisilazane‐silylester, Me3SiN(SiMe2–N)2SiMe2–O–CO–CMe3 ( 1 ). The lithium salt of the fluorodi‐methylsilyl‐substituted cyclotrisilazan III forms with benzoylchloride primarily in the analogous reaction the carboxy‐silyl‐amide, Me2SiF(N–SiMe2)2SiMe2–NH–CO–C6H5+ ( 2 ), which can be converted with III and benzoylchloride into the cyclodisilazane‐silylester, Me2SiF(NSiMe2)2SiMe2–O–CO–C6H5, ( 3 ). A silylester substituted six‐membered disila‐oxadiazine ( 4 ) is the result of the reaction of the lithiated cyclotrisilazane, (Me2SiNH)2, (Me2SiNLi) with tert‐butyl‐acylchloride. The reaction includes anionic ring contraction and can be rationilized by a process analogous to keto‐enol‐tautomerism. Dilithiated octamethyl‐cyclotetrasilazane, (Me2SiNHMe2SiNLi)2, reacts with tert‐butyl‐acylchloride or benzoylchloride in a molar ratio 1:2 to yield symmetrically acylestersubstituted cyclodisilazanes, (RCO–O–SiMe2–NSiMe2)2, R = C6H5 ( 5 ), CMe3 ( 6 ). The reaction mechanisms are discussed and the crystal structures of 2 and 6 are reported.  相似文献

Desymmetrization of the divinyl carbinol 1,4‐pentadien‐3‐ol was accomplished by the asymmetric 1,3‐dipolar cycloaddition of azomethine imines based on a magnesium‐mediated, multinucleating chiral reaction system utilizing diisopropyl (R,R)‐tartrate as the chiral auxiliary. The corresponding optically active trans‐pyrazolidines, each with three contiguous stereogenic centers, were obtained with excellent regio‐, diastereo‐, and enantioselectivity, with results as high as 99 % ee. This reaction was shown to be applicable to both aryl‐ and alkyl‐substituted azomethine imines. The use of a catalytic amount of diisopropyl (R,R)‐tartrate was also effective when accompanied by the addition of MgBr2.  相似文献
